BackgroundSmartphone apps for heart rate variability (HRV) measurement have been extensively developed in the last decade. However, ultra–short-term HRV recordings taken by wearable devices have not been examined. ObjectiveThe aims of this study were the following: (1) to compare the validity and reliability of ultra–short-term and short-term HRV time-domain and frequency-domain variables in a novel smartphone app, Pulse Express Pro (PEP), and (2) to determine the agreement of HRV assessments between an electrocardiogram (ECG) and PEP. MethodsIn total, 60 healthy adults were recruited to participate in this study (mean age 22.3 years [SD 3.0 years], mean height 168.4 cm [SD 8.0 cm], mean body weight 64.2 kg [SD 11.5 kg]). A 5-minute resting HRV measurement was recorded via ECG and PEP in a sitting position. Standard deviation of normal R-R interval (SDNN), root mean square of successive R-R interval (RMSSD), proportion of NN50 divided by the total number of RR intervals (pNN50), normalized very-low–frequency power (nVLF), normalized low-frequency power (nLF), and normalized high-frequency power (nHF) were analyzed within 9 time segments of HRV recordings: 0-1 minute, 1-2 minutes, 2-3 minutes, 3-4 minutes, 4-5 minutes, 0-2 minutes, 0-3 minutes, 0-4 minutes, and 0-5 minutes (standard). Standardized differences (ES), intraclass correlation coefficients (ICC), and the Spearman product-moment correlation were used to compare the validity and reliability of each time segment to the standard measurement (0-5 minutes). Limits of agreement were assessed by using Bland-Altman plot analysis. ResultsCompared to standard measures in both ECG and PEP, pNN50, SDNN, and RMSSD variables showed trivial ES (0.8). The nVLF, nLF, and nHF demonstrated a variation of ES (from trivial to small effects, 0.01-0.40), ICC (from moderate to nearly perfect, 0.39-0.96), and Spearman correlation coefficient values (from moderate to nearly perfect, 0.40-0.96). Furthermore, the Bland-Altman plots showed relatively narrow values of mean difference between the ECG and PEP after consecutive 1-minute recordings for SDNN, RMSSD, and pNN50. Acceptable limits of agreement were found after consecutive 3-minute recordings for nLF and nHF. ConclusionsUsing the PEP app to facilitate a 1-minute ultra–short-term recording is suggested for time-domain HRV indices (SDNN, RMSSD, and pNN50) to interpret autonomic functions during stabilization. When using frequency-domain HRV indices (nLF and nHF) via the PEP app, a recording of at least 3 minutes is needed for accurate measurement.

ObjectivePhysical exercise may increase brain volume and cortical thickness in late adulthood. However, few studies have examined the possibility for exercise to influence brain morphology in women treated for breast cancer. We conducted a nested sub-study within a randomized clinical trial to examine whether 6 months of moderate-intensity aerobic exercise in postmenopausal women with early-stage breast cancer influences brain morphology.MethodsWe included twenty-eight postmenopausal women newly diagnosed with Stage 0-IIIa breast cancer (M age = 62.96 ± 5.40) who were randomized to either 45–60 min of supervised aerobic exercise 3 days/week (n = 16) or usual care (n = 12). Before beginning aromatase inhibitor aromatase inhibitor therapy, and the exercise intervention, and again at 6-month follow-up, volumetric and cortical thickness measures were derived from magnetic resonance imaging scans.ResultsThere were no significant intervention effects on brain volume and cortical thickness. However, greater average exercise intensity (%) during the intervention was associated with greater post-intervention cortical volume, mean cortical thickness, precentral gyrus thickness, and superior parietal thickness (all p < 0.05). Finally, total supervised exercise time was associated with higher precentral gyrus thickness after the intervention (p = 0.042, R2 = 0.263).ConclusionThe exercise intervention did not significantly affect brain volumes and cortical thickness compared to the control group. However, positive associations were found between exercise intensity and brain morphology changes after the 6-month intervention, indicating that exercise may reduce the vulnerability of the brain to the deleterious effects of breast cancer and its treatment.

Abstract Senile plaque, composed of amyloid β protein (Aβ) aggregates, is a critical pathological feature in Alzheimer's disease (AD), leading to cognitive dysfunction. However, how Aβ aggregates exert age‐dependent toxicity and temporal cognitive dysfunction in APP/PS1 mice remains incompletely understood. In this study, we investigated AD pathogenesis and dynamic alterations in lysosomal pathways within the hippocampus of age‐gradient male mice using transcriptome sequencing, molecular biology assays, and histopathological analyses. We observed high levels of β‐amyloid precursor protein (APP) protein expression in the hippocampus at an early stage and age‐dependent Aβ deposition. Transcriptome sequencing revealed the enrichment of differential genes related to the lysosome pathway. Furthermore, the protein expression of ATP6V0d2 and CTSD associated with lysosomal functions exhibited dynamic changes with age, increasing in the early stage and decreasing later. Similar age‐dependent patterns were observed for the endosome function, autophagy pathway, and SGK1/FOXO3a pathway. Nissl and Golgi staining in the hippocampal region showed age‐dependent neuronal loss and synaptic damage, respectively. These findings clearly define the age‐gradient changes in the autophagy–lysosome system, the endosome/lysosome system, and the SGK1/FOXO3a pathway in the hippocampus of APP/PS1 mice, providing new perspectives and clues for understanding the possible mechanisms of AD, especially the transition from compensatory to decompensated state.

Objective To investigate the effects of mindfulness-based intervention on cognitive reappraisal, positive attentional bias and depression in army aviation maintenance personnel. Methods A total of 44 officers and soldiers of an army aviation brigade were selected by cluster sampling and then divided into intervention group (n=22) and control group (n=22).The intervention group received 2 hours'mindfulness intervention course once a week for 8 consecutive weeks, while the control group were given mental health education at same frequency and duration.Then the participants were assessed with self-compiled demographic scales, Five Facet Mindfulness Questionnaire (FFMQ), Emotion Regulation Questionnaire (ERQ), Attention to Positive and Negative Information Scale (APNIS), and Self-rating Depression Scale (SDS) before intervention (T1), at the end of the intervention (T2), and 1 month after the intervention (T3), respectively. Results There were no significant differences in the demographic data and psychological indicators between the 2 groups before training.After intervention, the scores of mindfulness level, cognitive reappraisal, positive attention bias and depression were significantly different between the 2 groups in terms of measurement time and group interaction (P < 0.05).The scores of mindfulness level and the cognitive reappraisal scores were greatly higher at T2 and T3 than T1(P < 0.01, P < 0.05), and the positive attention bias score at T2 was also notably higher than that of T1(P < 0.05) in the intervention group.In addition, no significant difference was observed in the depression score at the 3 time points in the intervention group, while in the control group, the score was obviously higher at T3 than T2 and T1(P < 0.05, P < 0.01). Conclusion Mindfulness-based intervention significantly improves the level of mindfulness, cognitive reappraisal and positive attentional bias, and relieves the impact of military training and management on depression of the army aviation officers and soldiers to a certain extent.

Background: Alcohol abuse, a prevalent global health issue, is associated with the onset of cognitive impairment and neurodegeneration. Actin filaments (F-actin) and microtubules (MTs) polymerized from monomeric globular actin (G-actin) and tubulin form the structural basis of the neuronal cytoskeleton. Precise regulation of the assembly and disassembly of these cytoskeletal proteins, and their dynamic balance, play a pivotal role in regulating neuronal morphology and function. Nevertheless, the effect of prolonged alcohol exposure on cytoskeleton dynamics is not fully understood. This study investigates the chronic effects of alcohol on cognitive ability, neuronal morphology and cytoskeleton dynamics in the mouse hippocampus. Methods: Mice were provided ad libitum access to 5% (v/v) alcohol in drinking water and were intragastrically administered 30% (v/v, 6.0 g/kg/day) alcohol for six weeks during adulthood. Cognitive functions were then evaluated using the Y maze, novel object recognition and Morris water maze tests. Hippocampal histomorphology was assessed through hematoxylin-eosin (HE) and Nissl staining. The polymerized and depolymerized states of actin cytoskeleton and microtubules were separated using two commercial assay kits and quantified by Western blot analysis. Results: Mice chronically exposed to alcohol exhibited significant deficits in spatial and recognition memory as evidenced by behavioral tests. Histological analysis revealed notable hippocampal damage and neuronal loss. Decreased ratios of F-actin/G-actin and MT/tubulin, along with reduced levels of polymerized F-actin and MTs, were found in the hippocampus of alcohol-treated mice. Conclusions: Our findings suggest that chronic alcohol consumption disrupted the assembly of the actin cytoskeleton and MTs in the hippocampus, potentially contributing to the cognitive deficits and pathological injury induced by chronic alcohol intoxication.

Introduction Physical activity (PA) has beneficial effects on brain health and cardiovascular disease (CVD) risk. Yet, we know little about whether PA-induced changes to physiological mediators of CVD risk influence brain health and whether benefits to brain health may also explain PA-induced improvements to CVD risk. This study combines neurobiological and peripheral physiological methods in the context of a randomised clinical trial to better understand the links between exercise, brain health and CVD risk.Methods and analysis In this 12-month trial, 130 healthy individuals between the ages of 26 and 58 will be randomly assigned to either: (1) moderate-intensity aerobic PA for 150 min/week or (2) a health information control group. Cardiovascular, neuroimaging and PA measurements will occur for both groups before and after the intervention. Primary outcomes include changes in (1) brain structural areas (ie, hippocampal volume); (2) systolic blood pressure (SBP) responses to functional MRI cognitive stressor tasks and (3) heart rate variability. The main secondary outcomes include changes in (1) brain activity, resting state connectivity, cortical thickness and cortical volume; (2) daily life SBP stress reactivity; (3) negative and positive affect; (4) baroreflex sensitivity; (5) pulse wave velocity; (6) endothelial function and (7) daily life positive and negative affect. Our results are expected to have both mechanistic and public health implications regarding brain–body interactions in the context of cardiovascular health.Ethics and dissemination Ethical approval has been obtained from the University of Pittsburgh Institutional Review Board (IRB ID: 19020218). This study will comply with the NIH Data Sharing Policy and Policy on the Dissemination of NIH-Funded Clinical Trial Information and the Clinical Trials Registration and Results Information Submission rule.Trial registration number NCT03841669.

Air-core inductors are key components in power converters and measurement equipment. To optimally design inductors, their self-resonant frequency (SRF) as a result of inherent stray capacitance should be accurately estimated. This paper presents optimal design procedures for air-core inductors, considering constraints such as SRF and inductor volume. To this end, a methodology is proposed to estimate the variability of stray capacitances, accounting for various uncertainty factors. Specifically, for single-layer air-core inductors, an empirical expression is adopted and experimentally verified to provide better predictions compared to the commonly used physical-based approach. For multi-layer air-core inductors, an enhanced analytical approach based on the energy-conservation method is proposed, which considers the impact of five influence factors. Two key factors (the proximity effect and the variation of turn length) are investigated by FEM simulations and incorporated into the enhanced models. The other three factors (the effective permittivity of the wire insulation coating, the winding tightness, and the electric field path assumption) are taken into account as uncertainty factors, leading to statistical estimates of the stray capacitance and SRF. Finally, two samples are fabricated for single- and multi-layer air-core inductors, which agree well with the design specifications, proving the effectiveness of the proposed design methodology.

Abstract Background Abnormally expressed in diverse cancers, circZFR has been correlated with clinical outcomes of cancer patients. Aim of this meta-analysis was to elucidate the prognostic role of circZFR in multiple human malignancies. Methods Literature retrieval was conducted by systematically searching on Pubmed, Web of Science, and the Cochrane Library up to December 2nd, 2021. Hazard ratios (HRs) or odds ratios (ORs) with 95% confidence intervals (CIs) were pooled to evaluate the association between circZFR expression and overall survival (OS). The reliability of the pooled results was assessed through sensitivity analysis and the publication bias was measured by Begg’s and Egger’s test. Results A total of seventeen studies comprising 1098 Chinese patients were enrolled in this meta-analysis. Results demonstrated that high circZFR expression was correlated with an unfavorable OS (HR = 2.14, 95% CI 1.74, 2.64). High circZFR expression predicted larger tumor size (OR = 2.79, 95% CI 1.52, 5.12), advanced clinical stage (OR = 3.38, 95% CI 1.49, 7.65), tendentiousness of lymph node metastasis (LNM) (OR = 3.08, 95% CI 2.01, 4.71), and malignant grade (OR = 3.18, 95% CI 1.09, 9.30), but not related to age, gender, and distant metastasis (DM). Conclusions High circZFR expression was associated with unfavorable OS and clinicopathologic parameters including tumor size, clinical stage, LNM, and histology grade, implicating a promising prognostic factor in cancers.

Objective To explore the mechanism of career adaptability on life satisfaction in cadets and investigate the mediating role of self-efficacy and hope. Methods From January to February 2021, a questionnaire survey was conducted among 759 cadets in a military university using the convenient sampling method. Evaluation tools include Chinese Career Adaptability Scale (CCAS), General Self Efficacy Scale (GSES), Herth Hope Inventory (HHI), and the with Life Scale (SWLS). Results ① Career adaptability, self-efficacy, hope and life satisfaction are positively correlated (r=0.48~0.71, P < 0.01); ② Career adaptability affected life satisfaction mainly through 3 mediation ways. The mediation effect of self-efficacy accounted for 15.25%, the mediation effect of hope for 12.34%, and the chain mediation effect of self-efficacy and hope for 12.30%. Conclusion The career adaptability of cadets can not only directly affect life satisfaction, but also indirectly affect life satisfaction through self-efficacy and hope.

Open and sustainable development is the theme that underpins a country's high-quality economic development. This study uses GMM regression, mediation effect test to conduct empirical tests based on the panel data of China's industrial sectors from 2003 to 2015 to analyze the internal mechanism of the impact of intermediate product imports on China's industrial pollution emissions. The results show that (1) Intermediate product imports can significantly promote the emission reduction of industrial wastes, including wastewater, waste gas and solid waste. (2) Considering the differences in the level of pollution intensity, this paper classified the sample and found the impact is heterogeneous that for the heavily, moderately, lightly polluted industries, intermediate product imports have different negative impacts on their pollution emissions. (3) Intermediate products imports reduce industrial pollution emissions through import competition effect, variety effect and technology spillover effect, and all of them play a partial mediating role.

Denosumab is a monoclonal antibody that targets and inhibits the osteoclast activating factor receptor activator for nuclear factor-κB ligand (RANKL). It has been widely used in the treatment of osteoporosis, giant cell tumors of bone, and in the prevention of malignant skeletal-related events (SREs). We collected the research results and related MeSH terms of denosumab from 2011 to 2021 through the Web of Science and PubMed, respectively. The literature was visualized and analyzed by CiteSpace and bibliometric online analysis platforms. The MeSH terms were biclustered using the Bibliographic Co-Occurrence Analysis System (BICOMB) and graph clustering toolkit (gCLUTO). The results show that the number of denosumab-related annual publications had increased from 51 to 215, with the United States leading and Amgen Inc. being the most influential in the past 10 years. Articles published in the Journal of Bone and Mineral Research had the highest total citations. Three scholars from Shinshu University in Matsumoto, Yukio Nakamura, Takako Suzuki, and Hiroyuki Kato, joined the field relatively late but produced the most. The clinical comparison and combination of denosumab with other drugs in the treatment of osteoporosis was the most significant focus of research. Drug withdrawal rebound and management strategies have gained more attention and controversy recently. MeSH analysis revealed eight major categories of research hotspots. Among them, exploring the multiple roles of the RANK-RANKL-OPG system in tumor progression, metastasis, and other diseases is the potential direction of future mechanism research. It is a valuable surgical topic to optimize the perioperative drug administration strategy for internal spinal fixation and orthopedic prosthesis implantation. Taken together, the advantages of denosumab were broad and cost-effective. However, there were still problems such as osteonecrosis of the jaw, severe hypocalcemia, a high recurrence rate of giant cells in the treatment of bone and individual sarcoidosis, and atypical femoral fractures, which need to be adequately solved.

Abstract Osteosarcoma is a common and highly malignant bone tumor among children, adolescents and young adults. However, the underlying molecular mechanisms remain largely unexplored. LncRNAs are transcripts with no or limited protein-coding capacity in human genomes, and have been demonstrated to play crucial functions in initiation, progression, therapeutic resistance, recurrence and metastasis of tumor. Considerable studies revealed a dysregulated lncRNA expression pattern in osteosarcoma, which may act as oncogenes or suppressors to regulate osteosarcoma progression. Wnt signaling pathway is an important cascade in tumorigenesis by modulation of pleiotropic biological functions including cell proliferation, apoptosis, differentiation, stemness, genetic stability and chemoresistance. Hyperactivation or deficiency of key effectors in Wnt cascade is a common event in many osteosarcoma patients. Recently, increasing evidences have suggested that lncRNAs could interplay with component of Wnt pathway, and thereby contribute to osteosarcoma onset, progression and dissemination. In this review, we briefly summarize Wnt signaling-related lncRNAs in osteosarcoma progression, aiming to gain insights into their underlying crosstalk as well as clinical application in osteosarcoma therapeutic modalities.

Abstract Background Pleomorphic liposarcoma is the least common but most aggressive subtype of liposarcoma. Very few studies have presented data on pleomorphic liposarcoma specifically, often including a limited number of cases and short-term follow-up. As a result, the survivorship and prognostic characteristics of this tumor remain incompletely identified. Study design and setting Cross-sectional analysis of the Surveillance Epidemiology and End Results database (1996–2015). Results Overall survival for the entire series was 54% (95% confidence interval [CI], 49–58%) and 40% (95% CI, 35–45%) at 5 and 10 years, respectively. Disease-specific survival for the entire series was 60% (95% CI, 56–65%) and 53% (95% CI, 48–58%) at 5 and 10 years, respectively. Patients who survived 10 years or more were more likely to die of events unrelated to pleomorphic liposarcoma. Univariate and multivariate analysis demonstrated that not receiving cancer-directed surgery was an independent poor prognostic factor. Older age (≥ 65 years old) was associated with worse overall survival but not disease-specific survival. Tumor stage and radiotherapy showed different impact on survival depending on tumor size. In comparison to localized staged tumors, regional stage only predicts poor survival in patients with tumor size less than 5 cm, while distant stage is an independent worse prognosis factor. Radiotherapy only benefits patients with tumor size larger than 10 cm. These results were confirmed in competing risk analysis. Conclusion Survival rates of patients with pleomorphic liposarcoma has not changed over the past 20 years. Patients with distant stage have poor prognosis; regional stage indicates worse survival in patients with tumor size less than 5 cm. Receiving surgery could prolong the survival, while radiotherapy only benefits patients with large tumor size (> 10 cm). Older age is associated with poor overall survival but not disease-specific survival. Routine patient surveillance following initial diagnosis should at least be 10 years for pleomorphic liposarcoma.

Objectives: To investigate factors associated with the duration of viral shedding in patients with COVID-19, outside of Wuhan. Methods: In this retrospective cohort study, patients with laboratory-confirmed COVID-19 in Changsha, China were included. Clinical characteristics, laboratory findings, treatment, and outcome were retrieved. Univariate and multivariate analyses were performed to explore potential factors. Results: Overall, 147 patients with COVID-19 were included. The median duration of viral shedding (the number of days from symptoms onset until the successive negative detection of SARS-CoV-2 RNA) was 17 days (interquartile range [IQR], 12–21). Multivariate Logistic regression analysis indicated that the highest temperature at admission (odds ratio [OR], 5.200; 95% confidence interval [CI]: 1.190−22.726; p = 0.028), time from symptom onset to admission (OR, 1.740; 95% CI: 1.296−2.337; p < 0.001) and hospital length of stay (OR, 1.604; 95% CI: 1.262−2.040; p < 0.001) were risk factors for prolonged duration of viral shedding. Conclusions: This study, with a relatively large sample size, focused on the duration of viral shedding and related factors in patients with COVID-19, outside of Wuhan, China. Potential risk factors were identified and should be taken into consideration for the strategy of quarantining infected patients.

Abstract Introdction Previous studies have argued that people tend to isolate themselves from negative information. This tendency is modulated by the individual's role in social interaction, that is, as an initiative actor (e.g., “I hit Tom”) or a passive recipient (e.g., “Paul hits me”). Depressed patients tend to focus on negative aspects of themselves and cope with situations passively. It is still an open question how the actor/recipient role affects the behavioral and neural responses to self in depression. Methods The present study adopted functional magnetic resonance imaging (fMRI) technology to investigate behavioral and neural responses to self (as an actor/recipient) in depressed patients and the matched healthy controls when attributing negative events. Results Compared with healthy controls, depressed patients showed more self‐attribution for negative events. Depressed patients showed increased brain activity in the dorsal medial prefrontal cortex (dmPFC) subsystem of the default mode network (DMN) when they played recipient role in self‐related negative events. Activity of the dmPFC subsystem was negatively correlated with depressed patients’ self‐attribution for negative events in recipient condition. While decreased brain activity in the medial temporal lobe (MTL) subsystem was observed in depressed patients when they played the actor or recipient role in self‐related negative events. Activity of the MTL subsystem was negatively correlated with depressed patients’ reaction time when they played recipient role in selfrelated negative events. Conclusion These results implicated that depressed patients manifested the negative self‐view. Actor/recipient role affected their activation patterns in the DMN which were different from the healthy controls. The correlation between the abnormal brain activations of the DMN and the behavioral performances might manifest more easily when depressed patients played recipient role in negative events.

G3-powerline communication (G3-PLC) is a robust communication protocol originally developed for smart metering in low-voltage power distribution networks. Modeling G3-PLC modems is an essential task to investigate electromagnetic compatibility (EMC) issues related to the coexistence of the PLC signal with the high-frequency noise affecting low-voltage networks, mainly due to the presence of power converters and non-linear loads. Since detailed information on the modem internal architecture is usually not available to the end-user, this work investigates the possibility of developing behavioral (black-box) models of G3-PLC modems, whose parameters can be estimated starting from measurements carried out at the modem output ports. To this end, suitable test benches are set up and used for model-parameter extraction as well as for validation purposes. Experiments have proven that an equivalent representation involving non-ideal voltage sources (i.e., in terms of extended Thevenin/Norton equivalent circuits) is no longer feasible for the transmitting modem, since the presence of a closed-loop control system invalidates the linearity assumption. Hence, while the receiving modem is still modeled through an impedance matrix (since it behaves as a linear device), an alternative representation is proposed for the transmitting modem, which resorts to the use of two ideal voltage sources in accordance with the substitution theorem. Experimental results prove that the proposed modeling strategy leads to satisfactory predictions of the currents propagating on the PLC system in the frequency interval of interest. Hence, it could be used in combination with high-frequency models of the other components in the network to investigate EMC and the coexistence of the PLC signal with the high-frequency noise generated by power converters.

Aiming at the limitations of a single High Resolution Range Profile (HRRP) in recognition, this paper proposes a Time step Correlation-based Feature Fusion (TCFF) method. This method calculates the covariance matrix at the time step of the two features extracted by the two channels, and assigns different weights to the time step according to the strength of the covariance correlation for feature fusion. The experimental results on the simulated ship target HRRP dataset show that the feature fusion method can achieve better recognition performance than the single channel model. Compared with simple feature fusion such as element addition and element contacting, it can also achieve better recognition results.

The concept of multituned thirteen-tower umbrella antenna is developed in this study to address the problem that increasing the power capacity of a single-tuned VLF transmitting antenna leads to a decrease in self-resonance. The calculation method of electrical parameters such as input resistance and radiation efficiency is theoretically analyzed based on the radiation resistance of different antenna units in a thirteen-tower umbrella antenna, and the electrical performance of the single-tuned and multituned thirteen-tower umbrella antennas is simulated and compared with the measured results of the Cutler antenna. Under the same antenna structure parameters and input power, the antenna radiation efficiency of the multituned thirteen-tower umbrella antenna is basically the same as that of the single tuned one. However, the multituned antenna has lower input current and higher self-resonance, which can effectively improve the antenna’s safety and broaden its operating frequency range.

Due to the widespread integration of renewable energy sources connected to AC and DC power systems by means of power electronics converters, electromagnetic noise propagates along lines, and metallic earth-return structures. EMI filters are commonly used to mitigate the common mode and differential mode noise at the interface of distribution lines, and their suppression characteristics are usually assessed in standard test setups, the impedances of which are, however, scarcely representative of real-world applications. In this paper, an online, inductively coupled impedance measurement method is proposed. A sensitivity analysis to highlight the benefits of the proposed setup and experimental verification is performed. The proposed method enables non-intrusive impedance measurement in energized systems, including power converters. These measures, in turn, allow the evaluation of modal insertion losses of EMI filters in real-world operating conditions. The three-phase example considered in this study shows significant deviations from manufacturer specifications, thus justifying the need for more advanced estimation techniques.

Abstract Background Fibroblast growth factor (FGF) and tumor growth factor-β (TGFβ) have emerged as pivotal regulators during the progression of osteosarcoma (OS). LHX9 is one crucial transcription factor controlled by FGF, however, its function in OS has not been investigated yet. Methods The expression of LHX9, FRS2, BMP4, TGF-beta R1, SMAD2, beta-catenin and metastasis-related proteins was measured by real-time quantitative PCR (RT-qPCR) and Western blot. CCK-8 assay and colony formation assay were employed to determine the proliferation of OS cells, while scratch wound healing assay and transwell assay were used to evaluate their migration and invasion, respectively. In vivo tumor growth and metastasis were determined by subcutaneous or intravenous injection of OS cells into nude mice. Results LHX9 expression was evidently up-regulated in OS tumor tissues and cell lines. Knockdown of LHX9 impaired the proliferation, migration, invasion and metastasis of OS cells. Mechanistically, LHX9 silencing led to the down-regulation of BMP-4, β-catenin and metastasis-related proteins, which was also observed in beta-catenin knockdown OS cells. By contrast, FRS2 knockdown conduced to the up-regulation of LHX9, BMP4, β-catenin and TGF-βR1, while TGF-beta inhibition repressed the expression of LHX9 and metastasis-related proteins. Additionally, let-7c modulates LHX9 and metastasis-related proteins by suppressing TGF-beta R1 expression on transcriptional level. Conclusions This study revealed LHX9 was essential for the proliferation, migration, invasion, and metastasis of OS cells via FGF and TGF-β/β-catenin signaling pathways.

Supernumerary teats are commonly found in sheep. To investigate the expression levels of hormone receptors in supernumerary teats of Hu sheep, mammary tissue samples were collected from two groups of Hu sheep, four- and two-teat sheep, during adolescent (A), pregnancy (P), lactation (L), and non-pregnancy (N). Using hematoxylin and eosin staining, we found that mammary alveoli were denser in the four-teat sheep than in the two-teat sheep during P and L. Immunohistochemistry and Western blotting were used to detect the expression patterns and relative protein expression levels of the estrogen receptor 1 (ESR1), prolactin receptor (PRLR), growth hormone receptor (GHR), and insulin-like growth factor 1 receptor (IGF1R) in Hu sheep mammary glands during the four periods. All four receptors were mainly expressed in mammary epithelial cells and adipose cells. Furthermore, the expression levels of PRLR and GHR in the four-teat sheep were significantly higher than those in the two-teat sheep during P and L. Our data suggest that four-teat sheep have more developed mammary gland tissue compared with two-teat sheep.

The traditional High-Resolution Range Profile (HRRP) target recognition method has difficulty automatically extracting target deep features, and has low recognition accuracy under low training samples. To solve these problems, a ship recognition method is proposed based on the lightweight Transformer model. The model enhances the representation of key features by embedding Recurrent Neural Networks (RNN) into Transformer’s encoder. The Group Linear Transformations (GLTs) are introduced into Transformer to reduce the number of parameters in the model, and stable features are extracted through linear intergroup dimensional transformations. The adaptive gradient clipping algorithm is combined with the Stochastic Gradient Descent (SGD) optimizer to allow the gradient to change dynamically with the training process and to improve the training speed and generalization ability of the model. Experimental results on the simulated dataset show that multi-layer model stacking can effectively extract deep features of targets and raise recognition accuracy. At the same time, the lightweight Transformer model can maintain good recognition performance with low parameters and low training samples.

Power Line Communication (PLC) technologies are being used in many applications and offer the advantage of utilizing existing power cables for both power and data transmission, thus minimizing cost and complexity. Nevertheless, PLC technology requires further investigation to solve possible co-existence issues. Indeed, recent studies confirmed that alternative modulation schemes such as Random Pulse Width Modulation (RPWM), applied to switching-mode power converters to minimize conducted emissions, detrimentally interfere with the PLC system. This paper presents an experimental test campaign aimed at investigating the effects of RPWM on the G3-PLC system, with the final goal of understanding the conditions under which RPWM schemes can be considered as an effective alternative to conventional Pulse Width Modulation (PWM) in applications involving PLC systems. In details, the effects of different RPWM parameters such as switching frequency, modulation index, and Random Number Update Rate (RNUR) on the G3-PLC is investigated. In addition, different RPWM schemes such as Random Frequency Modulation (RFM) and Random Pulse Position Modulation (RPPM) are compared in terms of performance so as to highlight which RPWM is best suited to assure coexistence with PLC systems. The impact of RPWM on the communication channel is evaluated in terms of Frame Error Rate (FER), Channel Capacity, and Channel Capacity Loss metrics. Experimental results confirmed that randomly modulated converters with switching frequencies near the G3-PLC bandwidth cause more significant disturbance and possible coexistence issues than the switching frequencies out of this range. Results also show that the modulation index and the RNUR of RPWM have a direct effect on the communication channel. Moreover, a trade-off between Electromagnetic Interference (EMI) reduction and coexistence issues is observed: RFM, which is very effective for EMI reduction, is found to be very disruptive for G3-PLC, compared to alternative random modulation techniques such as RPPM.

RNA modifications have emerged as key regulators of transcript expression in diverse physiological and pathological processes. As one of the most prevalent types of RNA modifications, N6-methyladenosine (m6A) has become the highlight in modulation of various diseases through interfering RNA splicing, translation, nuclear export, and decay. In many cases, the detailed functions of m6A in cellular processes and diseases remain unclear. Notably, recent studies have determined the relationship between m6A modification and musculoskeletal disorders containing osteosarcoma, osteoarthritis, rheumatoid arthritis, osteoporosis, etc. Herein, this review comprehensively summarizes the recent advances of m6A modification in pathogenesis and progression of musculoskeletal diseases. Specifically, the underlying molecular mechanisms, detection technologies, regulatory functions, clinical implications, and future perspectives of m6A in musculoskeletal disorders are discussed, with the aim to provide a novel insight into their association.

Heat pumps coupled with thermal energy storage (TES) systems are seen as a promising technology for load management that can be used to shift peak loads to off-peak hours. Most of the existing model predictive control (MPC) studies on tariff-based load shifting deploying hot water tanks use simplified tank models. In this study, an MPC framework that accounts for transient thermal behavior (i.e., mixing and stratification) by applying energy (EMPC) and exergy (XMPC) analysis is proposed. A case study for an office building equipped with an air handling unit (AHU) revealed that the MPC strategy had a high load-shifting capacity: over 80% of the energy consumption took place during off-peak hours when there was an electricity surplus in the grid. An analysis of a typical day showed that the XMPC method was able to provide more appropriate stratification within the TES for all load characteristics. An annual exergy analysis demonstrated that, during cold months, energy degradation in the TES is mainly caused by exergy destruction due to irreversibility, while, during the transition to milder months, exergy loss dominates. Compared to the EMPC approach, the XMPC strategy achieves additional reductions of 18% in annual electricity consumption, 13% in operating costs, and almost 17% in emissions.

Electromagnetic interference (EMI) from renewable power systems to the grid attracts more attention especially in the low-frequency range, due to the low switching frequency of high-power inverters. It is significantly important to derive EMI models of power inverters as well as to develop strategies to suppress the related conducted emissions. In this work, black-box modelling is applied to a three-phase inverter system, by implementing an alternative procedure to identify the parameters describing the active part of the model. Besides, two limitations of black-box modelling are investigated. The first regards the need for the system to satisfy the linear and time-invariant (LTI) assumption. The influence of this assumption on prediction accuracy is analysed with reference to the zero, positive and negative sequence decomposition. It is showing that predictions for the positive/negative sequence are highly influenced by this assumption, unlike those for the zero sequence. The second limitation is related to the possible variation of the mains impedance which is not satisfactorily stabilized at a low frequency outside the operating frequency range of standard line impedance stabilization networks.

The aim of this study was to explore possible changes in whole brain gray matter volume (GMV) after spinal cord injury (SCI) using voxel-based morphometry (VBM), and to study their associations with the injury duration, severity, and clinical variables. In total, 21 patients with SCI (10 with complete and 11 with incomplete SCI) and 21 age- and sex-matched healthy controls (HCs) were recruited. The 3D high-resolution T1-weighted structural images of all subjects were obtained using a 3.0 Tesla MRI system. Disease duration and American Spinal Injury Association (ASIA) Scale scores were also obtained from each patient. Voxel-based morphometry analysis was carried out to investigate the differences in GMV between patients with SCI and HCs, and between the SCI sub-groups. Associations between GMV and clinical variables were also analyzed. Compared with HCs, patients with SCI showed significant GMV decrease in the dorsal anterior cingulate cortex, bilateral anterior insular cortex, bilateral orbital frontal cortex (OFC), and right superior temporal gyrus. No significant difference in GMV in these areas was found either between the complete and incomplete SCI sub-groups, or between the sub-acute (duration 1 year) sub-groups. Finally, the GMV of the right OFC was correlated with the clinical motor scores of left extremities in not only all SCI patients, but especially the CSCI subgroup. In the sub-acute subgroup, we found a significant positive correlation between the dACC GMV and the total clinical motor scores, and a significant negative correlation between right OFC GMV and the injury duration. These findings indicate that SCI can cause remote atrophy of brain gray matter, especially in the salient network. In general, the duration and severity of SCI may be not associated with the degree of brain atrophy in total SCI patients, but there may be associations between them in subgroups.

Economic development usually leads to increased energy consumption, which in turn will result in an increase in carbon emissions. To break the relationship between economic development and carbon emissions, scholars have turned their attention to the phenomenon of decoupling. In this paper, we studied the decoupling relationship between carbon emissions and economic growth of the equipment manufacturing industry in China from 2000 to 2014. We adapted the LMDI decomposition method, and we used the Tapio decoupling evaluation model to analyze our data. We found that the decoupling relationship between carbon emissions and economic growth of China’s equipment manufacturing industry is weak, which indicates the industry is experiencing faster economic growth than carbon emission growth. We found the economic output is the factor that has the strongest influence on the industry’s carbon emission, and energy consumption intensity has the strongest relationship with the decoupling of economic growth and carbon emission. The indicators of the industry’s decoupling-effort are all less than 1.0, which indicates that the industry is in the state of weak decoupling, and we also observed an annual decreasing trend in the industry’s indicators. Toward the end of this paper, we used the Grey forecasting model to predict the decoupling relationship between carbon emission and economic growth for 2015–2024, and we discussed the implications of our research.

We propose a fast data relay (FDR) mechanism to enhance existing CGRA (coarse-grained reconfigurable architecture). FDR can not only provide multicycle data transmission in concurrent with computations but also convert resource-demanding inter-processing-element global data accesses into local data accesses to avoid communication congestion. We also propose the supporting compiler techniques that can efficiently utilize the FDR feature to achieve higher performance for a variety of applications. Our results on FDR-based CGRA are compared with two other works in this field: ADRES and RCP. Experimental results for various multimedia applications show that FDR combined with the new compiler deliver up to 29% and 21% higher performance than ADRES and RCP, respectively.

Abstract Background Brazilin, isolated from the heartwood of Caesalpinia sappan L., has been shown to possess multiple pharmacological properties. Methods In this study, platelet aggregation, flow cytometry, immunoblotting analysis, and electron spin resonance (ESR) spectrometry were used to investigate the effects of brazilin on platelet activation ex vivo. Moreover, fluorescein sodium-induced platelet thrombi of mesenteric microvessels was also used in in vivo study. Results We demonstrated that relatively low concentrations of brazilin (1 to 10 μM) potentiated platelet aggregation induced by collagen (0.1 μg/ml) in washed human platelets. Higher concentrations of brazilin (20 to 50 μM) directly triggered platelet aggregation. Brazilin-mediated platelet aggregation was slightly inhibited by ATP (an antagonist of ADP). It was not inhibited by yohimbine (an antagonist of epinephrine), by SCH79797 (an antagonist of thrombin protease-activated receptor [PAR] 1), or by tcY-NH2 (an antagonist of PAR 4). Brazilin did not significantly affect FITC-triflavin binding to the integrin αIIbβ3 in platelet suspensions. Pretreatment of the platelets with caffeic acid phenethyl ester (an antagonist of collagen receptors) or JAQ1 and Sam.G4 monoclonal antibodies raised against collagen receptor glycoprotein VI and integrin α2β1, respectively, abolished platelet aggregation stimulated by collagen or brazilin. The immunoblotting analysis showed that brazilin stimulated the phosphorylation of phospholipase C (PLC)γ2 and Lyn, which were significantly attenuated in the presence of JAQ1 and Sam.G4. In addition, brazilin did not significantly trigger hydroxyl radical formation in ESR analysis. An in vivo mouse study showed that brazilin treatment (2 and 4 mg/kg) significantly shortened the occlusion time for platelet plug formation in mesenteric venules. Conclusion To the best of our knowledge, this study provides the first evidence that brazilin acts a novel collagen receptor agonist. Brazilin is a plant-based natural product, may offer therapeutic potential as intended anti-thrombotic agents for targeting of collagen receptors or to be used a useful tool for the study of detailed mechanisms in collagen receptors-mediated platelet activation.

Abstract Background Functional tricuspid regurgitation (TR) occurs in patients with rheumatic mitral valve disease even after mitral valve surgery. The aim of this study was to analyze surgical results of TR after previous successful mitral valve surgery. Methods From September 1996 to September 2008, 45 patients with TR after previous mitral valve replacement underwent second operation for TR. In those, 43 patients (95.6%) had right heart failure symptoms (edema of lower extremities, ascites, hepatic congestion, etc.) and 40 patients (88.9%) had atrial fibrillation. Twenty-six patients (57.8%) were in New York Heart Association (NYHA) functional class III, and 19 (42.2%) in class IV. Previous operations included: 41 for mechanical mitral valve replacement (91.1%), 4 for bioprosthetic mitral valve replacement (8.9%), and 7 for tricuspid annuloplasty (15.6%). Results The tricuspid valves were repaired with Kay's (7 cases, 15.6%) or De Vega technique (4 cases, 8.9%). Tricuspid valve replacement was performed in 34 cases (75.6%). One patient (2.2%) died. Postoperative low cardiac output (LCO) occurred in 5 patients and treated successfully. Postoperative echocardiography showed obvious reduction of right atrium and ventricle. The anterioposterior diameter of the right ventricle decreased to 25.5 ± 7.1 mm from 33.7 ± 6.2 mm preoperatively (P < 0. 05). Conclusion TR after mitral valve replacement in rheumatic heart disease is a serious clinical problem. If it occurs or progresses late after mitral valve surgery, tricuspid valve annuloplasty or replacement may be performed with satisfactory results. Due to the serious consequence of untreated TR, aggressive treatment of existing TR during mitral valve surgery is recommended.

ObjectiveAge-related hippocampal atrophy is associated with memory loss in older adults, and certain hippocampal subfields are more vulnerable to age-related atrophy than others. Cardiorespiratory fitness (CRF) may be an important protective factor for preserving hippocampal volume, but little is known about how CRF relates to the volume of specific hippocampal subfields, and whether associations between CRF and hippocampal subfield volumes are related to episodic memory performance. To address these gaps, the current study evaluates the associations among baseline CRF, hippocampal subfield volumes, and episodic memory performance in cognitively unimpaired older adults from the Investigating Gains in Neurocognition Trial of Exercise (IGNITE) (NCT02875301).MethodsParticipants (N = 601, ages 65–80, 72% female) completed assessments including a graded exercise test measuring peak oxygen comsumption (VO2peak) to assess CRF, cognitive testing, and high-resolution magnetic resonance imaging of the hippocampus processed with Automated Segmentation of Hippocampal Subfields (ASHS). Separate linear regression models examined whether CRF was associated with hippocampal subfield volumes and whether those assocations were moderated by age or sex. Mediation models examined whether hippocampal volumes statistically mediated the relationship between CRF and episodic memory performance. Covariates included age, sex, years of education, body mass index, estimated intracranial volume, and study site.ResultsHigher CRF was significantly associated with greater total left (B = 5.82, p = 0.039) and total right (B = 7.64, p = 0.006) hippocampal volume, as well as greater left CA2 (B = 0.14, p = 0.022) and dentate gyrus (DG; B = 2.34, p = 0.031) volume, and greater right CA1 (B = 3.99, p = 0.011), CA2 (B = 0.15, p = 0.002), and subiculum (B = 1.56, p = 0.004) volume. Sex significantly moderated left DG volume (B = −4.26, p = 0.017), such that the association was positive and significant only for males. Total left hippocampal volume [indirect effect = 0.002, 95% CI (0.0002, 0.00), p = 0.027] and right subiculum volume [indirect effect = 0.002, 95% CI (0.0007, 0.01), p = 0.006] statistically mediated the relationship between CRF and episodic memory performance.DiscussionWhile higher CRF was significantly associated with greater total hippocampal volume, CRF was not associated with all underlying subfield volumes. Our results further demonstrate the relevance of the associations between CRF and hippocampal volume for episodic memory performance. Finally, our results suggest that the regionally-specific effects of aging and Alzheimer’s disease on hippocampal subfields could be mitigated by maintaining higher CRF in older adulthood.

The study was focussed on the inhibitory mechanism associated with the ability of an endophytic bacterium - Bacillus altitudinis Q7, obtained from Ginkgo biloba, to inhibit the growth of Alternaria alternata in vitro. A fungus, identified as Alternaria alternata was isolated from the rotten part of an apple. One of the endophytes was identified as Bacillus altitudinis Q7. Effect of Bacillus altitudinis Q7 on Alternaria alternata was characterized by measuring membrane permeability and lipid peroxidation. Antioxidant enzymes in Alternaria alternata were determined before and after exposure to Bacillus altitudinis Q7. Results indicated that cell membranes in Alternaria alternata were damaged, lipid peroxidation was enhanced, and superoxide dismutase, peroxidase, and polyphenol oxidase were significantly inhibited. Content of lipopeptides produced by Bacillus altitudinis Q7 was higher than other metabolites. The FTIR spectra revealed that the prepared antibacterial lipopeptides is a cyclic lipopeptides. The effect of lipopeptides produced by Bacillus altitudinis Q7 at 2 mg/mL against Alternaria alternata in vitro reached 83.2%. The lipopeptide-treated group showed the same trend as Q7 fermentation broth in inhibition mode of action. Moreover, the utilization of proteins and sugars during culturing of Alternaria alternata decreased when adding lipopeptides to the growth medium. Bacillus altitudinis Q7 from Ginkgo biloba significantly inhibited the growth of Alternaria alternata and is a very promising biocontrol strain against Alternaria alternata. The study of endophytic bacteria and its metabolites is being done to identify biological control alternatives. The results provide a theoretical basis for understanding their antagonistic properties.

Objective To investigate the prevalence and risk factors of anxiety and depressive symptoms among individuals isolated during coronavirus disease 2019 (COVID-19) pandemic, and provide suggestions for psychological self-regulation and interventions for these individuals. Method Self-rating Anxiety Scale, Self-rating Depression Scale, Insomnia Severity Index and Chinese Perceived Stress Scale were used to survey 2 322 individuals who were isolated in quarantine facilities or in home quarantine for COVID-19 from January 31 to February 27, 2020. Results Of the 2 322 individuals surveyed, 344 (14.81%) were found to have depressive symptoms, 103 (4.44%) had anxiety symptoms, and 82 (3.53%) had both depressive and anxiety symptoms during quarantine. The scores for anxiety and depression of the quarantined individuals were significantly higher than the national norms of China (t=35.26, P < 0.01; t=43.25, P < 0.01). Multivariate logistic regression analysis showed that the anxiety symptoms of the quarantined individuals were closely associated with their education level (β=-2.84, P < 0.01), severity of insomnia (β=0.11, P < 0.01), perceived tension (β=0.20, P < 0.01) and depression (β=0.19, P < 0.01); the depressive symptoms were positively correlated with their perceived loss of control (β=0.18, P < 0.01) and anxiety (β=0.32, P < 0.01). Conclusion The levels of anxiety and depression among the quarantined individuals for COVID-19 are higher than the national norms. The main risk factors for anxiety symptoms include education level, severity of insomnia, perceived tension and depression, while perceived loss of control and anxiety are the major risk factors for the depressive symptoms in these individuals.

Soft tissue sarcoma (STS) represents an uncommon and heterogenous group of malignancies, and poses substantial therapeutic challenges. Pyroptosis has been demonstrated to be related with tumor progression and prognosis. Nevertheless, no studies exist that delineated the role of pyroptosis-related genes (PRGs) in STS. In the present study, we comprehensively and systematically analyzed the gene expression profiles of PRGs in STS. The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) databases were utilized to identify differentially expressed PRGs. In total, 34 PRGs were aberrantly expressed between STS and normal tissues. Several PRGs were validated with RT-qPCR. Consensus clustering analysis based on PRGs was conducted to divide STS patients into two clusters, and significant survival difference was observed between two distinct clusters (p = 0.019). Differentially expressed genes (DEGs) were identified between pyroptosis-related clusters. Based on the least absolute shrinkage and selection operator (LASSO) COX regression analysis, the pyroptosis-related gene signature with five key DEGs was constructed. The high pyroptosis-related risk score group of TCGA cohort was characterized by poorer prognosis (p < 0.001), with immune infiltration and function significantly decreased. For external validation, STS patients from Gene Expression Omnibus (GEO) were grouped according to the same cut-off point. The survival difference between two risk groups of GEO cohort was also significant (p < 0.001). With the combination of clinical characteristics, pyroptosis-related risk score was identified to serve as an independent prognostic factor for STS patients. In conclusion, this study provided a comprehensive overview of PRGs in STS and the potential role in prognosis, which could be an important direction for future studies.

Abstract The organic passivated carbon nanotube (CNT)/silicon (Si) solar cell is a new type of low‐cost, high‐efficiency solar cell, with challenges concerning the stability of the organic layer used for passivation. In this work, the stability of the organic layer is studied with respect to the internal and external (humidity) water content and additionally long‐term stability for low moisture environments. It is found that the organic passivated CNT/Si complex interface is not stable, despite both the organic passivation layer and CNTs being stable on their own and is due to the CNTs providing an additional path for water molecules to the interface. With the use of a simple encapsulation, a record power conversion efficiency of 22% is achieved and a stable photovoltaic performance is demonstrated. This work provides a new direction for the development of high‐performance/low‐cost photovoltaics in the future and will stimulate the use of nanotubes materials for solar cells applications.

Deconstruction is an essential step of conversion of polysaccharides, and polysaccharide-degrading enzymes play a key role in this process. Although there is recent progress in the identification of these enzymes, the diversity and phylogenetic distribution of these enzymes in marine microorganisms remain largely unknown, hindering our understanding of the ecological roles of marine microorganisms in the ocean carbon cycle. Here, we studied the phylogenetic distribution of nine types of polysaccharide-degrading enzymes in marine bacterial genomes. First, we manually compiled a reference sequence database containing 961 experimentally verified enzymes. With this reference database, we annotated 9,335 enzyme sequences from 2,182 high-quality marine bacterial genomes, revealing extended distribution for six enzymes at the phylum level and for all nine enzymes at lower taxonomic levels. Next, phylogenetic analyses revealed intra-clade diversity in the encoding potentials and phylogenetic conservation of a few enzymes at the genus level. Lastly, our analyses revealed correlations between enzymes, with alginate lyases demonstrating the most extensive correlations with others. Intriguingly, chitinases showed negative correlations with cellulases, alginate lyases, and agarases in a few genera. This result suggested that intra-genus lifestyle differentiation occurred many times in marine bacteria and that the utilization of polysaccharides may act as an important driver in the recent ecological differentiation of a few lineages. This study expanded our knowledge of the phylogenetic distribution of polysaccharide enzymes and provided insights into the ecological differentiation of marine bacteria.

Liver fibrosis is characterized by deposition of excessive extracellular matrix (ECM). The major source of ECM is activated hepatic stellate cells (HSCs). Previously, we reported that hypoxia‐inducible factor‐1 (Hif‐1) regulates activation of HSCs through autophagy. In current work, human HSC cell line LX‐2 was used as cell model. It was determined that trimethylation of H3 histone on lysine 4 (H3K4me3) occurred in the Hif‐1 transcriptional complex. Inhibition of modifications of histone methylation suppressed Hif‐1 nuclear transport, autophagosome formation, and activation of LX‐2 cells. These data suggest that histone methylation modification plays an important role in the Hif‐1 signaling cascade regulating HSC activation.

Abstract Background Missed abortion is a common occurrence for otherwise healthy women. Immunological factor is one of the most important reasons. Tumor necrosis factor-α-induced protein-8 like-2 (TIPE2) is a novel negative immune regulator related to several human diseases. However, the expression level and clinical significance of TIPE2 in missed abortion remain unclear. Methods The expression of TIPE2 mRNA and protein in decidua and chorion from 36 missed abortion patients and 36 healthy controls was detected using quantitative real-time PCR, western blot and immunohistochemistry. In addition, serum TNF-ɑ and IL-10 levels were measured using flow cytometry. Serum estradiol and progesterone levels were measured by radioimmunoassay test. The correlations of TIPE2 protein levels with TNF-ɑ, IL-10, estradiol and progesterone were further analyzed. Results TIPE2 protein levels were significantly lower in decidual tissues of missed abortion patients than those in healthy controls. The patients with missed abortion had significantly higher levels of serum TNF-ɑ, and lower levels of serum IL-10, estradiol and progesterone compared with healthy controls. The TIPE2 protein levels were positively related to serum IL-10 levels. Conclusion Our data indicate TIPE2 could play important roles in maintaining the maternal-fetal tolerance and decreased TIPE2 expression in the decidua may be related to the development of missed abortion.

AIM:To investigate the effect of emodin on pseudomonas aeruginosa lipopolysaccharides (LPS)-induced corneal inflammation in rats.METHODS:Corneal infection was induced by pseudomonas aeruginosa LPS in Wistar rats. The inflammation induced by LPS were examined by slit lamp microscope and cytological checkup of aqueous humor. Corneal tissue structure was observed by hematoxylin and eosin (HE) staining. The activation of nuclear factor kappaB (NF-κB) was determined by Western blot. Messenger ribonucleic acid (mRNA) of tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule-1 (ICAM-1) in LPS-challenged rat corneas were measured with reverse transcription-polymerase chain reaction (RT-PCR).RESULTS:Typical manifestations of acute corneal inflammation were observed in LPS-induce rat model, and the corneal inflammatory response and structure were improved in rats pretreated with emodin. Treatment with emodin could improve corneal structure, reduce corneal injure by reducing corneal inflammatory response. Emodin could inhibit the decreasing lever of inhibitor of kappaB alpha (IкBα) express, and the mRNA expression of TNF-α and ICAM-1 in corneal tissues was also inhibited by emodin. The differences were statistically significant between groups treated with emodin and those without treatment (PCONCLUSION:Emodin could ameliorate LPS-induced corneal inflammation, which might via inhibiting the activation of NF-κB.

The scorpion has been extensively used in various pharmacological profiles or as food supplies. The exploration of scorpion venom has been reported due to the presence of recombinant peptides. rBmαTX14 is an α-neurotoxin extracted from the venom gland of the East Asian scorpion Buthus martensii Karsch and can affect ion channel conductance. Here, we investigated the functions of rBmαTX14 using the Caenorhabditis elegans model. Using western blot analysis, rBmαTX14 was shown to be expressed both in the cytoplasm and inclusion bodies in the E.coli Rosetta (DE3) strain. Circular dichroism spectroscopy analysis demonstrated that purified rBmαTX14 retained its biological structures. Next, feeding nematodes with E.coli Rosetta (DE3) expressing rBmαTX14 caused extension of the life span and promoted the locomotion of the nematodes. In addition, we identified several genes that play various roles in the life span and locomotion of C. elegans through microarray analysis and quantitative real-time PCR. Furthermore, if the amino acid site H15 of rBmαTX14 was mutated, rBmαTX14 no longer promoted the C. elegans life span. In conclusion, the results not only demonstrated the functions and mechanism of rBmαTX14 in C. elegans, but also provided the new sight in the utility of recombinant peptides from scorpion venom.

The myozenin family of proteins binds calcineurin, which is involved in myocyte differentiation of skeletal muscle. Moreover, gene expression of myozenin is closely related to meat quality. To further understand the functions and effects of myozenin2 (MYOZ2) and myozenin3 (MYOZ3) genes in goat, we cloned them from Tianfu goat longissimus dorsi muscle. Sequence analyses revealed that full-length coding sequence of MYOZ2 consisted of 795 bp and encoded 264 amino acids, and full-length coding sequence of MYOZ3 consisted of 735 bp and encoded 244 amino acids. RT-qPCR analyses revealed that mRNA expressions of MYOZ2 and MYOZ3 were detected in heart, liver, spleen, lung, kidney, leg muscle, abdominal muscle, and longissimus dorsi muscle. Particularly high expression levels of MYOZ2 were seen in abdominal muscle and heart (P0.05) and very little expression were detected in liver, spleen, lung and kidney (P>0.05). In addition, high expression levels of MYOZ3 were seen in abdominal muscle, leg muscle, lungs and kidney (P0.05). Temporal mRNA expression results showed that MYOZ2 and MYOZ3 gene expression varied across four muscle tissues with different ages of the goats. Western blotting further revealed that MYOZ2 and MYOZ3 proteins were only expressed in goat muscle, with notable temporal expression differences in specialized muscle tissues from five development age stages. This work provides the first evidence that MYOZ2 and MYOZ3 genes are expressed abundantly in Tianfu goat muscle tissues from different development age stages, and lay a foundation for understanding the functions of MYOZ2 and MYOZ3 genes in muscle fiber differentiation.

Positron emission tomography (PET) is an important imaging modality for clincial use. Conventionally, the PET scanner is generally built to provide a roomy enough transverse field-of-view (FOV) for imaging most adults' torsos. However, in many cases, the region-of-interest (ROI) for imaging is usually a small area inside the human body. Therefore, to fulfill a PET system which provides an FOV comparable in size to the target ROI seems appealing and more cost effective. Meanwhile, such a PET system has the potential for portable or bedside application with the reduced system size. In this work, we have investigated the feasibility of using dual-headed panel-detectors to build an ROI-focused PET scanner. A novel windowed list-mode ordered subset expectation maximization method was developed to perform the ROI image reconstruction. With this method, the ROI of the object can be reconstructed from the coincidences whose position determined by time-of-flight (TOF) measurements was inside the ROI. Monte Carlo simulation demonstrates the feasibility of detecting lesions not less than 1 cm in diameter, with a 300 ps full width at half maximum timing resolution. As a critical system performance, the impact of TOF information on image quality has been studied and the required TOF capability was assessed. With enhanced timing resolution, the distortions and artifacts were reduced effectively. The further improved TOF capability also shows a noticeable improvement of detection performance for low uptake lesions, as well as the recovery speed of lesion contrast, which is of practical significance in the lesion detection task.

Abstract Background Platelet activation is relevant to a variety of coronary heart diseases. Our previous studies revealed that sesamol possesses potent antiplatelet activity through increasing cyclic AMP formation. Although platelets are anucleated cells, they also express the transcription factor, NF-κB, that may exert non-genomic functions in platelet activation. Therefore, we further investigated the inhibitory roles of sesamol in NF-κB-mediated platelet function. Methods Platelet aggregation, Fura 2-AM fluorescence, and immunoblotting analysis were used in this study. Results NF-κB signaling events, including IKKβ phosphorylation, IκBα degradation, and p65 phosphorylation, were markedly activated by collagen (1 μg/ml) in washed human platelets, and these signaling events were attenuated by sesamol (2.5~25 μM). Furthermore, SQ22536 and ODQ, inhibitors of adenylate cyclase and guanylate cyclase, respectively, strongly reversed the sesamol (25 μM)-mediated inhibitory effects of IKKβ phosphorylation, IκBα degradation, and p65 phosphorylation stimulated by collagen. The protein kinase A (PKA) inhibitor, H89, also reversed sesamol-mediated inhibition of IκBα degradation. Moreover, BAY11-7082, an NF-κB inhibitor, abolished IκBα degradation, phospholipase C (PLC)γ2 phosphorylation, protein kinase C (PKC) activation, [Ca2+]i mobilization, and platelet aggregation stimulated by collagen. Preincubation of platelets with the inhibitors, SQ22536 and H89, both strongly reversed sesamol-mediated inhibition of platelet aggregation and [Ca2+]i mobilization. Conclusions Sesamol activates cAMP-PKA signaling, followed by inhibition of the NF-κB-PLC-PKC cascade, thereby leading to inhibition of [Ca2+]i mobilization and platelet aggregation. Because platelet activation is not only linked to hemostasis, but also has a relevant role in inflammation and metastasis, our data demonstrating that inhibition of NF-κB interferes with platelet function may have a great impact when these types of drugs are considered for the treatment of cancer and various inflammatory diseases.
