β-N-methylamino-L-alanine (BMAA) is an environmental neurotoxin thought to be produced by cyanobacteria. However, the cyanobacterial origin of BMAA remains controversial. The detection method and culture conditions of cyanobacteria are often cited as factors behind the discrepancy of published results. We showed previously that BMAA was highly toxic to the cyanobacterium Nostoc PCC 7120, and it is taken up via an amino acid transport system. Using a mutant ΔnatAΔbgtA deficient in amino acid transport as a genetic control, we show here that BMAA taken up from the medium can be detected quantitatively at a threshold similar to, or below those reported, but was undetectable in the mutant. The BMAA isomer, 2,4-diaminobutanoic acids (DAB), but not BMAA, could be detected in cell free extracts of Nostoc PCC 7120. Long-term (20 ​days) diazotrophic growth or non-limiting supply of phosphate, conditions reported to enhance BMAA synthesis, did not lead to the detection of BMAA. An UPLC-MS/MS signal with a similar retention time to BMAA was found after prolonged diazotrophic incubation, but did not have fragment ions of BMAA after further analysis. When extended to 29 different cyanobacterial strains and 6 natural cyanobacterial bloom samples, none of them was found to produce BMAA. The cytotoxicity of BMAA to cyanobacteria, and the lack of a cellular protective mechanism against such toxicity, contradict the presence of a BMAA synthesis pathway in these organisms. More specific methods for BMAA detection in vivo need to be developed to clarify the cyanobacterial origin of BMAA.

The diatom Skeletonema costatum and the haptophyte Phaeocystis globosa often form blooms in the coastal waters of the South China Sea. Skeletonemacostatum commonly dominates in nutrient enrichment coastal waters, whereas P. globosa starts flourishing after the diatom blooms when phosphorus (P) is limited. Therefore, P limitation was proposed to be a critical factor affecting diatom–haptophyte transition. To elucidate the tolerance to P limitation in P. globosa compared with S. costatum, the effect of P limitation on their photosystem II (PSII) performance was investigated and their photosynthesis acclimation strategies in response to P limitation were evaluated. P limitation did not affect the growth of P. globosa over 7 days but decreased it for S. costatum. Correspondingly, the PSII activity of S. costatum was significantly inhibited by P limitation. The decline in PSII activity in S. costatum under P limitation was associated with the impairment of the oxygen-evolving complex (the donor side of PSII), the hindrance of electron transport from QA− to QB (the acceptor side of PSII), and the inhibition of electron transport to photosystem I (PSI). The 100% decrease in D1 protein level of S. costatum after P limitation for 6 days and PsbO protein level after 2 days of P limitation were attributed to its enhanced photoinhibition. In contrast, P. globosa maintained its photosynthetic activity with minor impairment of the function of PSII. With accelerated PSII repair and highly increased non-photochemical quenching (NPQ), P. globosa can avoid serious PSII damage under P limitation. On the contrary, S. costatum decreased its D1 restoration under P limitation, and the maximum NPQ value in S. costatum was only one-sixth of that in P. globosa. The present work provides extensive evidence that a close interaction exists between the tolerance to P limitation and photosynthetic responses of S. costatum and P. globosa.

Although programmed cell death (PCD) has been reported in phytoplankton, knowledge of the characterization of the PCD pathway and cascade process in different phytoplankton species is still limited. In this study, PCD progression in cyanobacterium Microcystis aeruginosa and green algae Chlorella luteoviridis by paraquat-induced oxidative stress was monitored. The results showed that paraquat-induced PCD in the two species belonged to the caspase-dependent pathway. Dose- and time-dependent PCD characteristics in the two strains under paraquat included the increase in caspase-like activity, DNA fragmentation, and chromatin condensation. However, the signaling pathway and cascade events of PCD in M. aeruginosa and C. luteoviridis differed. In M. aeruginosa, the free Ca2+ concentration was rapidly increased at 8 h, followed by a significant elevation of the reactive oxygen species (ROS) level at 24 h, and eventual cell death. In C. luteoviridis, the mitochondrial apoptosis pathway, revealed by the depolarization of the mitochondrial membrane potential at 1 h and increase in the ROS level and caspase-like activity at 8 h, might contribute to cell death. In addition, the dynamics of ROS levels and metacaspase activity were synchronized, suggesting that paraquat-triggered PCD was ROS-mediated in both M. aeruginosa and C. luteoviridis. These results provide insights into PCD patterns in prokaryotic cyanobacteria and eukaryotic green algae under similar stress.

Zinc antimonides have been widely studied owing to their outstanding thermoelectric properties. Unlike in the bulk state, where various structurally unknown phases have been identified through their specific physical properties, a number of intermediate phases in the thin-film state remain largely unexplored. Here, in situ X-ray diffraction and X-ray total scattering are combined with in situ measurement of electrical resistivity to monitor the crystallization process of as-deposited amorphous Zn-Sb films during post-deposition annealing. The as-deposited Zn-Sb films undergo a structural evolution from an amorphous phase to an intermediate crystalline phase and finally the ZnSb phase during heat treatment up to 573 K. An intermediate phase (phase B) is identified to be a modified β-Zn8Sb7 phase by refinement of the X-ray diffraction data. Within a certain range of Sb content (∼42–55 at%) in the films, phase B is accompanied by an emerging Sb impurity phase. Lower Sb content leads to smaller amounts of Sb impurity and the formation of phase B at lower temperatures, and phase B is stable at room temperature if the annealing temperature is controlled. Pair distribution function analysis of the amorphous phase shows local ordered units of distorted ZnSb4 tetrahedra, and annealing leads to long-range ordering of these units to form the intermediate phase. A higher formation energy is required when the intermediate phase evolves into the ZnSb phase with a significantly more regular arrangement of ZnSb4 tetrahedra.

Abstract Background Harmful cyanobacterial blooms have attracted wide attention all over the world as they cause water quality deterioration and ecosystem health issues. Microcystis aeruginosa associated with a large number of bacteria is one of the most common and widespread bloom-forming cyanobacteria that secret toxins. These associated bacteria are considered to benefit from organic substrates released by the cyanobacterium. In order to avoid the influence of associated heterotrophic bacteria on the target cyanobacteria for physiological and molecular studies, it is urgent to obtain an axenic M. aeruginosa culture and further investigate the specific interaction between the heterotroph and the cyanobacterium. Results A traditional and reliable method based on solid-liquid alternate cultivation was carried out to purify the xenic cyanobacterium M. aeruginosa FACHB-905. On the basis of 16S rDNA gene sequences, two associated bacteria named strain B905–1 and strain B905–2, were identified as Pannonibacter sp. and Chryseobacterium sp. with a 99 and 97% similarity value, respectively. The axenic M. aeruginosa FACHB-905A (Microcystis 905A) was not able to form colonies on BG11 agar medium without the addition of strain B905–1, while it grew well in BG11 liquid medium. Although the presence of B905–1 was not indispensable for the growth of Microcystis 905A, B905–1 had a positive effect on promoting the growth of Microcystis 905A. Conclusions The associated bacteria were eliminated by solid-liquid alternate cultivation method and the axenic Microcystis 905A was successfully purified. The associated bacterium B905–1 has the potentiality to promote the growth of Microcystis 905A. Moreover, the purification technique for cyanobacteria described in this study is potentially applicable to a wider range of unicellular cyanobacteria.

Abstract Background Plant NADPH oxidase (NOX), also known as respiratory burst oxidase homolog (rboh), encoded by the rboh gene, is a key enzyme in the reactive oxygen species (ROS) metabolic network. It catalyzes the formation of the superoxide anion (O2 •−), a type of ROS. In recent years, various studies had shown that members of the plant rboh gene family were involved in plant growth and developmental processes as well as in biotic and abiotic stress responses, but little is known about its functional role in upland cotton. Results In the present study, 26 putative Ghrboh genes were identified and characterized. They were phylogenetically classified into six subfamilies and distributed at different densities across 18 of the 26 chromosomes or scaffolds. Their exon-intron structures, conserved domains, synteny and collinearity, gene family evolution, regulation mediated by cis-acting elements and microRNAs (miRNAs) were predicted and analyzed. Additionally, expression profiles of Ghrboh gene family were analyzed in different tissues/organs and at different developmental stages and under different abiotic stresses, using RNA-Seq data and real-time PCR. These profiling studies indicated that the Ghrboh genes exhibited temporal and spatial specificity with respect to expression, and might play important roles in cotton development and in stress tolerance through modulating NOX-dependent ROS induction and other signaling pathways. Conclusions This comprehensive analysis of the characteristics of the Ghrboh gene family determined features such as sequence, synteny and collinearity, phylogenetic and evolutionary relationship, expression patterns, and cis-element- and miRNA-mediated regulation of gene expression. Our results will provide valuable information to help with further gene cloning, evolutionary analysis, and biological function analysis of cotton rbohs.

Toxic cyanobacterial blooms have become a severe global hazard to human and environmental health. Most studies have focused on the relationships between cyanobacterial composition and cyanotoxins production. Yet, little is known about the environmental conditions influencing the hazard of cyanotoxins. Here, we analysed a unique 22 sites dataset comprising monthly observations of water quality, cyanobacterial genera, zooplankton assemblages, and microcystins (MCs) quota and concentrations in a large-shallow lake. Missing values of MCs were imputed using a non-negative latent factor (NLF) analysis, and the results achieved a promising accuracy. Furthermore, we used the Bayesian additive regression tree (BART) to quantify how Microcystis bloom toxicity responds to relevant physicochemical characteristics and zooplankton assemblages. As expected, the BART model achieved better performance in Microcystis biomass and MCs concentration predictions than some comparative models, including random forest and multiple linear regression. The importance analysis via BART illustrated that the shade index was overall the best predictor of MCs concentrations, implying the predominant effects of light limitations on the MCs content of Microcystis. Variables of greatest significance to the toxicity of Microcystis also included pH and dissolved inorganic nitrogen. However, total phosphorus was found to be a strong predictor of the biomass of total Microcystis and toxic M. aeruginosa. Together with the partial dependence plot, results revealed the positive correlations between protozoa and Microcystis biomass. In contrast, copepods biomass may regulate the MC quota and concentrations. Overall, our observations arouse universal demands for machine-learning strategies to represent nonlinear relationships between harmful algal blooms and environmental covariates.

Integrated constructed wetland-reservoirs (referred to as CW-Rs) are being built for ensuring drinking water supply in plain areas where the source water often cannot meet the quality criteria for drinking water. CW-Rs consist of a series of treatment units and have been reported to reduce the concentration of major nutrients. The efficiency of CW-Rs in mitigating odor compounds and their producer(s) remains largely unclear. In this study, Yanlonghu CW-R located in Jiangsu province, eastern China, was selected to monitor the occurrence and dynamics of 2-methylisoborneol (2-MIB). Two peaks of 2-MIB, attributed mainly to Pseudanabaena in April, and Pseudanabaena and Oscillatoria in July, were observed. This indicated that benthic Oscillatoria is also a threatening factor and should be considered. The concentrations of 2-MIB at the inlet and outlet were 9.75 and 50.08 ng/L in April and 73.11 and 25.21 ng/L in July, respectively. Yanlonghu CW-R was reported to be effective in reducing the content of major nutrients (total phosphorus in particular) throughout the year. In addition, it was effective in mitigating the levels of 2-MIB-producers/2-MIB during the summer season. It is proposed that qPCR for mic gene detection can be performed to screen and establish an early warning method. We revealed that the 2-MIB episode is related to the function of the CW-R, which is shown to be different from what is known in lakes or reservoirs. It is emphasized that each treatment unit of CW-R should be maintained at full capacity so that the frequency and hazardous effects caused by 2-MIB in the system can be controlled or reduced. This finding is implicated in the control strategy selection and contributes to the better management and improvement of future applications.

PurposeTo evaluate the potential of the texture features extracted from dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) intratumoral subregions to distinguish benign from malignant breast tumors.Materials and MethodsA total of 299 patients with pathologically verified breast tumors who underwent breast DCE-MRI examination were enrolled in this study, including 124 benign cases and 175 malignant cases. The whole tumor area was semi-automatically segmented on the basis of subtraction images of DCE-MRI in Matlab 2018b. According to the time to peak of the contrast agent, the whole tumor area was partitioned into three subregions: early, moderate, and late. A total of 467 texture features were extracted from the whole tumor area and the three subregions, respectively. Patients were divided into training (n = 209) and validation (n = 90) cohorts by different MRI scanners. The least absolute shrinkage and selection operator (LASSO) method was used to select the optimal feature subset in the training cohort. The Kolmogorov-Smirnov test was first performed on texture features selected by LASSO to test whether the samples followed a normal distribution. Two machine learning methods, decision tree (DT) and support vector machine (SVM), were used to establish classification models with a 10-fold cross-validation method. The performance of the classification models was evaluated with receiver operating characteristic (ROC) curves.ResultsIn the training cohort, the areas under the ROC curve (AUCs) for the DT_Whole model and SVM_Whole model were 0.744 and 0.806, respectively. In contrast, the AUCs of the DT_Early model (P = 0.004), DT_Late model (P = 0.015), SVM_Early model (P = 0.002), and SVM_Late model (P = 0.002) were significantly higher: 0.863 (95% CI, 0.808–0.906), 0.860 (95% CI, 0.806–0.904), 0.934 (95% CI, 0.891–0.963), and 0.921 (95% CI, 0.876–0.954), respectively. The SVM_Early model and SVM_Late model achieved better performance than the DT_Early model and DT_Late model (P = 0.003, 0.034, 0.008, and 0.026, respectively). In the validation cohort, the AUCs for the DT_Whole model and SVM_Whole model were 0.670 and 0.708, respectively. In comparison, the AUCs of the DT_Early model (P = 0.006), DT_Late model (P = 0.043), SVM_Early model (P = 0.001), and SVM_Late model (P = 0.007) were significantly higher: 0.839 (95% CI, 0.747–0.908), 0.784 (95% CI, 0.601–0.798), 0.890 (95% CI, 0.806–0.946), and 0.865 (95% CI, 0.777–0.928), respectively.ConclusionThe texture features from intratumoral subregions of breast DCE-MRI showed potential in identifying benign and malignant breast tumors.

ObjectiveTo evaluate whether texture features derived from semiquantitative kinetic parameter maps based on breast dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) can determine human epidermal growth factor receptor 2 (HER2) status of patients with breast cancer.Materials and MethodsThis study included 102 patients with histologically confirmed breast cancer, all of whom underwent preoperative breast DCE-MRI and were enrolled retrospectively. This cohort included 48 HER2-positive cases and 54 HER2-negative cases. Seven semiquantitative kinetic parameter maps were calculated on the lesion area. A total of 55 texture features were extracted from each kinetic parameter map. Patients were randomly divided into training (n = 72) and test (n = 30) sets. The least absolute shrinkage and selection operator (LASSO) was used to select features in the training set, and then, multivariate logistic regression analysis was conducted to establish the prediction models. The classification performance was evaluated by receiver operating characteristic (ROC) analysis.ResultsAmong the seven prediction models, the model with features extracted from the early signal enhancement ratio (ESER) map yielded an area under the ROC curve (AUC) of 0.83 in the training set (sensitivity of 70.59%, specificity of 92.11%, and accuracy of 81.94%), and the highest AUC of 0.83 in the test set (sensitivity of 57.14%, specificity of 100.00%, and accuracy of 80.00%). The model with features extracted from the slope of signal intensity (SIslope) map yielded the highest AUC of 0.92 in the training set (sensitivity of 82.35%, specificity of 97.37%, and accuracy of 90.28%), and an AUC of 0.79 in the test set (sensitivity of 92.86%, specificity of 68.75%, and accuracy of 80.00%).ConclusionsTexture features derived from kinetic parameter maps, calculated based on breast DCE-MRI, have the potential to be used as imaging biomarkers to distinguish HER2-positive and HER2-negative breast cancer.

Abstract Background Long non-coding (lnc) RNAs are a class of functional RNA molecules greater than 200 nucleotides in length, and lncRNAs play important roles in various biological regulatory processes and response to the biotic and abiotic stresses. LncRNAs associated with salt stress in cotton have been identified through RNA sequencing, but the function of lncRNAs has not been reported. We previously identified salt stress-related lncRNAs in cotton (Gossypium spp.), and discovered the salt-related lncRNA-lncRNA973. Results In this study, we identified the expression level, localization, function, and preliminary mechanism of action of lncRNA973. LncRNA973, which was localized in the nucleus, was expressed at a low level under nonstress conditions but can be significantly increased by salt treatments. Here lncRNA973 was transformed into Arabidopsis and overexpressed. Along with the increased expression compared with wild type under salt stress conditions in transgenic plants, the seed germination rate, fresh weights and root lengths of the transgenic plants increased. We also knocked down the expression of lncRNA973 using virus-induced gene silencing technology. The lncRNA973 knockdown plants wilted, and the leaves became yellowed and dropped under salt-stress conditions, indicating that the tolerance to salt stress had decreased compared with wild type. LncRNA973 may be involved in the regulation of reactive oxygen species-scavenging genes, transcription factors and genes involved in salt stress-related processes in response to cotton salt stress. Conclusions LncRNA973 was localized in the nucleus and its expression was increased by salt treatment. The lncRNA973-overexpression lines had increased salt tolerance compared with the wild type, while the lncRNA973 knockdown plants had reduced salt tolerance. LncRNA973 regulated cotton responses to salt stress by modulating the expression of a series of salt stress-related genes. The data provides a basis for further studies on the mechanisms of lncRNA973-associated responses to salt stress in cotton.

Objective: To investigate the value of texture features derived from T2-weighted magnetic resonance imaging (T2WI) for predicting preoperative lymph node invasion (N stage) in rectal cancer.Materials and Methods: One hundred and eighty-two patients with histopathologically confirmed rectal cancer and preoperative magnetic resonance imaging were retrospectively analyzed, who were divided into high (N1-2) and low N stage (N0). Texture features were calculated from histogram, gray-level co-occurrence matrix, and gray-level run-length matrix from sagittal fat-suppression and oblique axial T2WI. Independent sample t-test or Mann-Whitney U-test were used for statistical analysis. Multivariate logistic regression analysis was conducted to build the predictive models. Predictive performance was evaluated by receiver operating characteristic (ROC) analysis.Results: Energy (ENE), entropy (ENT), information correlation (INC), long-run emphasis (LRE), and short-run low gray-level emphasis (SRLGLE) extracted from sagittal fat-suppression T2WI, and ENE, ENT, INC, low gray-level run emphasis (LGLRE), and SRLGLE from oblique axial T2WI were significantly different between stage N0 and stage N1-2 tumors. The multivariate analysis for features from sagittal fat-suppression T2WI showed that higher SRLGLE and lower ENE were independent predictors of lymph node invasion. The model reached an area under ROC curve (AUC) of 0.759. The analysis for features from oblique axial T2WI showed that higher INC and SRLGLE were independent predictors. The model achieved an AUC of 0.747. The analysis for all extracted features showed that lower ENE from sagittal fat-suppression T2WI and higher INC and SRLGLE from oblique axial T2WI were independent predictors. The model showed an AUC of 0.772.Conclusions: Texture features derived from T2WI could provide valuable information for identifying the status of lymph node invasion in rectal cancer.

ObjectiveTo investigate whether texture features extracted from dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) are associated with human epidermal growth factor receptor type 2 (HER2) 2+ status of breast cancer.Materials and methods92 MRI cases including 52 HER2 2+ positive and 40 negative patients confirmed by fluorescence in situ hybridization were retrospectively selected. The lesion area was semi-automatically delineated, and a total of 488 texture features were respectively extracted from precontrast, postcontrast, and subtraction images. The Student's t-test or Mann-Whitney U test was performed to identify statistically significant features between different HER2 2+ amplification groups. Least absolute shrinkage and selection operator (LASSO) was used to search for the optimal feature subsets. Three machine learning classifiers, logistic regression analysis (LRA), quadratic discriminant analysis (QDA), and support vector machine (SVM), were used with a leave-one-out cross validation method to establish the classification models of HER2 2+ status. Classification performance was evaluated by receiver operating characteristic (ROC) analysis.ResultsBased on the texture analysis with SVM model, the areas under the ROC curve (AUCs) were 0.890 for subtraction images, 0.736 for postcontrast images, and 0.672 for precontrast images, respectively. For LRA model, the AUCs were 0.884, 0.733, and 0.623, respectively. For QDA model, the AUCs were 0.831, 0.726, and 0.568, respectively. LRA and the SVM model with subtraction images reached significantly better performance than the QDA model (P = 0.0227 and P = 0.0088, respectively).ConclusionTexture features of breast cancer extracted from DCE-MRI are associated with HER2 2+ status. Additional studies are necessary to confirm the present preliminary findings.

Microbial degradation is an important route for removing environmental microcystins (MCs). Here, we investigated the ecological distribution of microcystin degraders (mlr-genotype), and the relationship between the substrate specificity of the microcystin degrader and the profile of microcystin congener production in the habitat. We showed that microcystin degraders were widely distributed and closely associated with Microcystis abundance in Lake Taihu, China. We characterized an indigenous degrader, Sphingopyxis N5 in the northern Lake Taihu, and it metabolized six microcystin congeners in increasing order (RR > LR > YR > LA > LF and LW). Such a substrate-specificity pattern was congruent to the order of the dominance levels of these congeners in northern Lake Taihu. Furthermore, a meta-analysis on global microcystin degraders revealed that the substrate-specificity patterns varied geographically, but generally matched the profiles of microcystin congener production in the degrader habitats, and the indigenous degrader typically metabolized well the dominant MC congeners, but not the rare congeners in the habitat. This highlighted the phenotypic congruence between microcystin production and degradation in natural environments. We theorize that such congruence resulted from the metabolic adaptation of the indigenous degrader to the local microcystin congeners. Under the nutrient microcystin selection, the degraders might have evolved to better exploit the locally dominant congeners. This study provided the novel insight into the ecological distribution and adaptive degradation of microcystin degraders.

Mitigation of harmful cyanobacterial blooms that constitute a serious threat to water quality, particularly in eutrophic water, such as in aquaculture, is essential. Thus, in this study, we tested the efficacy of selected cyanocides towards bloom control in laboratory and outdoor mesocosm experiments. Specifically, we focused on the applicability of a group of cationic disinfectants, alkyltrimethyl ammonium (ATMA) compounds and H2O2. The biocidal effect of four ATMA cations with different alkyl chain lengths was evaluated ex situ using Microcystis colonies collected from a fish pond. The most effective compound, octadecyl trimethyl ammonium (ODTMA), was further evaluated for its selectivity towards 24 cyanobacteria and eukaryotic algae species, including Cyanobacteria, Chlorophyta, Bacillariophyta, Euglenozoa and Cryptophyta. The results indicated selective inhibition of cyanobacteria by ODTMA-Br (C18) on both Chroccocales and Nostocales, but a minor effect on Chlorophytes and Bacillariophytes. The efficacy of ODTMA-Br (C18) (6.4 μM) in mitigating the Microcystis population was compared with that of a single low dose of H2O2 treatments (117.6 μM). ODTMA-Br (C18) suppressed the regrowth of Microcystis for a longer duration than did H2O2. The results suggested that ODTMA-Br (C18) may be used as an effective cyanocide and that it is worth further evaluating this group of cationic compounds as a treatment to mitigate cyanobacterial blooms in aquaculture.

The utility of cyanobacterial bloom is often hindered by concerns about the toxin content. Over three years of investigation, we found that the toxin content of cyanobacterial bloom in Lake Taihu was always low in June and higher in late summer and autumn. The findings enabled us to compare the effects of diets containing low and high toxic cyanobacterial blooms on the growth and consumption safety of tilapia. There were no negative effects on the growth of tilapia, and the muscle seemed to be safe for human consumption in the treatment of 18.5% low toxic cyanobacterial bloom. Therefore, limitations of the utilization of cyanobacterial biomass can be overcome by selecting low toxic cyanobacterial bloom that can be found and collected in large lakes.

Freshwater cyanobacterial blooms (e.g., Microcystis blooms) constitute a major global environmental problem because of their risks to public health and aquatic ecological systems. Current physicochemical treatments of toxic cyanobacteria cause the significant release of cyanotoxin microcystins from damaged cells. Biological control is a promising eco-friendly technology to manage harmful cyanobacteria and cyanotoxins. Here, we demonstrated an efficient biological control strategy at the laboratory scale to simultaneously remove Microcystis and microcystins via the combined use of the algicidal bacterial filtrate and the microcystin-degrading enzymatic agent. The algicidal indigenous bacterium Paenibacillus sp. SJ-73 was isolated from the sediment of northern Lake Taihu, China, and the microcystin-degrading enzymatic agent (MlrA) was prepared via the heterologous expression of the mlrA gene in the indigenous microcystin-degrading bacterium Sphingopyxis sp. HW isolated from Lake Taihu. The single use of a fermentation filtrate (5%, v/v) of Paenibacillus sp. SJ-73 for seven days removed the unicellular Microcystis aeruginosa PCC 7806 and the native colonial Microcystis strain TH1701 in Lake Taihu by 84% and 92%, respectively, whereas the single use of MlrA removed 85% of microcystins. Used in combination, the fermentation filtrate and MlrA removed Microcystis TH1701 and microcystins by 92% and 79%, respectively. The present biological control thus provides an important technical basis for the further development of safe, efficient, and effective measures to manage Microcystis blooms and microcystins in natural waterbodies.

Microcystis blooms are the most widely distributed and frequently occurring cyanobacterial blooms in freshwater. Reducing phosphorus is suggested to be effective in mitigating cyanobacterial blooms, while the underlying molecular mechanisms are yet to be elucidated. In the present study, isobaric tags for relative and absolute quantitation (iTRAQ)-based quantitative proteomics was employed to study the effects of phosphorus depletion on Microcystis aeruginosa FACHB-905. The production of microcystins (MCs), a severe hazard of Microcystis blooms, was also analyzed. In total, 230 proteins were found to be differentially abundant, with 136 downregulated proteins. The results revealed that, upon phosphorus limitation stress, Microcystis aeruginosa FACHB-905 raised the availability of phosphorus primarily by upregulating the expression of orthophosphate transport system proteins, with no alkaline phosphatase producing ability. Phosphorus depletion remarkably inhibited cell growth and the primary metabolic processes of Microcystis, including transcription, translation and photosynthesis, with structures of photosystems remaining intact. Moreover, expression of nitrogen assimilation proteins was downregulated, while proteins involved in carbon catabolism were significantly upregulated, which was considered beneficial for the intracellular balance among carbon, nitrogen and phosphorus. The expression of MC synthetase was not significantly different upon phosphorus depletion, while MC content was significantly suppressed. It is assumed that phosphorus depletion indirectly regulates the production of MC by the inhibition of metabolic processes and energy production. These results contribute to further understanding of the influence mechanisms of phosphorus depletion on both biological processes and MC production in Microcystis cells.

Zintl-phase thermoelectrics are predominantly p-type. Here, Zhanget al. use tellurium to n-dope Mg3Sb1.5Bi0.5and obtain thermoelectric figures of merit up to 1.6 at 700 K. Calculations show that these performances result from a conduction band with sixfold valley degeneracy.

Purpose: To evaluate the ability of texture analysis of breast dynamic contrast enhancement-magnetic resonance (DCE-MR) images in differentiating human epidermal growth factor receptor 2 (HER2) 2+ status of breast tumors.Methods: A total of 73 cases were retrospectively selected. HER2 2+ status was confirmed by fluorescence in situ hybridization. For each case, 279 textural features were derived. A student's t-test or Mann-Whitney U test was used to select features with statistically significant differences between HER2 2+ positive and negative groups. A principal component analysis was applied to eliminate feature correlation. Three machine learning classifiers, logistic regression (LR), quadratic discriminant analysis (QDA), and a support vector machine (SVM), were trained and tested using a leave-one-out cross-validation method. The area under a receiver operating characteristic curve (AUC) was measured to assess the classifier's performance.Results: The AUCs for the different classifiers were satisfactory, ranging from 0.808 to 0.865. The classification methods derived with LR and SVM demonstrated similarly high performances, and the accuracy levels were 81.06 and 81.18%, respectively. The AUC for the classifier derived with SVM was the highest (0.865), and a marked specificity (88.90%) was presented. For the classifier with LR, the AUC was 0.851, and the corresponding sensitivity (94.44%) was the highest.Conclusion: The texture analysis for breast DCE-MRI proposed in this study demonstrated potential utility in HER2 2+ status discrimination.

Cyanotoxins are the underlying cause of the threat that globally pervasive Cyanobacteria Harmful algal blooms (CyanoHABs) pose to humans. Major attention has been focused on the cyanobacterial hepatotoxin microcystins (MCs); however, there is a dearth of studies on cyanobacterial neurotoxin anatoxins. In this study, we explored how an anatoxin-producing Cuspidothrix issatschenkoi strain responded to culture with inorganic and organic nitrogen sources in terms of growth and anatoxins production. The results of our study revealed that ʟ- alanine could greatly boost cell growth, and was associated with the highest cell productivity, while urea significantly stimulated anatoxin production with the maximum anatoxin yield reaching 25.86 μg/mg dry weight, which was 1.56-fold higher than that in the control group (BG11). To further understand whether the carbon/nitrogen balance in C. issatschenkoi would affect anatoxin production, we explored growth and toxin production in response to different carbon/nitrogen ratios (C/N). Anatoxin production was mildly promoted when the C/N ratio was within low range, and significantly inhibited when the C/N ratio was within high range, showing approximately a three-fold difference. Furthermore, the transcriptional profile revealed that anaC gene expression was significantly up-regulated over 2–24 h when the C/N ratio was increased, and was significantly down-regulated after 96 h. Overall, our results further enriched the evidence that urea can stimulate cyanotoxin production, and ʟ-alanine could boost C. issatschenkoi proliferation, thus providing information for better management of aquatic systems. Moreover, by focusing on the intracellular C/N metabolic balance, this study explained the anatoxin production dynamics in C. issatschenkoi in response to different N sources.

Although the nutrient as a driving force for the red tide was intensively studied, the spatial patterns of the phytoplankton community and its response to benthic nutrient cycling remain unclear. We determined the pelagic phytoplankton community and its extracellular alkaline phosphatase qualitatively using enzyme-labeled fluorescence (ELF) technique, concomitantly with the concentrations of phosphorus (P) and nitrogen (N) in the water and sediments in the Taiwan Strait in spring 2018. A total of 30 phytoplankton genera were identified with a higher abundance of the abundance of Prorocentrum and Trichodesmium being observed at the north coast and the center of the southern strait, respectively. Both phytoplankton abundances and Trichodesmium were negatively correlated with the ratios of dissolved inorganic N and ammonium to soluble reactive P (DIN/SRP, NH4+/SRP) in the bottom. Furthermore, the ELF-labeling percentage in Trichodesmium was negatively correlated with total P and SRP but positively correlated with TN/TP, DIN/SRP, and NH4+/SRP in the bottom. In contrast to high DIN/SRP of the surface, lower DIN/SRP in the bottom was owing to a high P release potential and weak sequestration of P as evidenced by the distribution of P solubilizing bacteria and P content. Our findings indicated that the benthic nutrient regime might shape the structure of the pelagic phytoplankton community.

Produced by cyanobacteria and some plants, BMAA is considered as an important environmental factor in the occurrence of some neurodegenerative diseases. Neither the underlying mechanism of its toxicity, nor its biosynthetic or metabolic pathway in cyanobacteria is understood. Interestingly, BMAA is found to be toxic to some cyanobacteria, making it possible to dissect the mechanism of BMAA metabolism by genetic approaches using these organisms. In this study, we used the cyanobacterium Anabaena PCC 7120 to isolate BMAA-resistant mutants. Following genomic sequencing, several mutations were mapped to two genes involved in amino acids transport, suggesting that BMAA was taken up through amino acid transporters. This conclusion was supported by the protective effect of several amino acids against BMAA toxicity. Furthermore, targeted inactivation of genes encoding different amino acid transport pathways conferred various levels of resistance to BMAA. One mutant inactivating all three major amino acid transport systems could no longer take up BMAA and gained full resistance to BMAA toxicity. Therefore, BMAA is a substrate of amino acid transporters, and cyanobacteria are interesting models for genetic analysis of BMAA transport and metabolism.

Eutrophication has become one of the major environmental issues of global concern due to the adverse effects on water quality, public health and ecosystem sustainability. Fundamental research on the restoration of eutrophic freshwaters, i.e., lakes and rivers, is crucial to support further evidence-based practical implementations. This Special Issue successfully brings together recent research findings from scientists in this field and assembles contributions on lake and river restoration. The 12 published papers can be classified into, and contribute to, three major aspects of this topic. Firstly, a background investigation into the migration of nutrients, and the characteristics of submerged biota, will guide and assist the understanding of the mechanisms of future restoration. Secondly, various restoration strategies, including control of both external and internal nutrients loading, are studied and evaluated. Thirdly, an evaluation of the field sites after restoration treatment is reported in order to support the selection of appropriate restoration approaches. This paper focuses on the current environmental issues related to lake and river restoration and has conducted a comprehensive bibliometric analysis in order to emphasise the fast-growing attention being paid to the research topic. The research questions and main conclusions from all papers are summarised to focus the attention toward how the presented studies aid gains in scientific knowledge, engineering experience and support for policymakers.

Harmful cyanobacterial blooms frequently occur in shallow eutrophic lakes and usually cause the decline of submerged vegetation. Therefore, artificial aquatic plants (AAPs) were introduced into enclosures in the eutrophic Dianchi Lake to investigate whether or not they could reduce cyanobacterial blooms and promote the growth of submerged macrophytes. On the 60th day after the AAPs were installed, the turbidity, total nitrogen (TN), total phosphorous (TP), and the cell density of phytoplankton (especially cyanobacteria) of the treated enclosures were significantly reduced as compared with the control enclosures. The adsorption and absorption of the subsequently formed periphyton biofilms attached to the AAPs effectively decreased nutrient levels in the water. Moreover, the microbial diversity and structure in the water changed with the development of periphyton biofilms, showing that the dominant planktonic algae shifted from Cyanophyta to Chlorophyta. The biodiversity of both planktonic and attached bacterial communities in the periphyton biofilm also gradually increased with time, and were higher than those of the control enclosures. The transplanted submerged macrophyte (Elodea nuttallii) in treated enclosures recovered effectively and reached 50% coverage in one month while those in the control enclosures failed to grow. The application of AAPs with incubated periphyton presents an environmentally-friendly and effective solution for reducing nutrients and controlling the biomass of phytoplankton, thereby promoting the restoration of submerged macrophytes in shallow eutrophic waters.

Catalases (CATs), which were coded by the catalase gene family, were a type notably distinguished ROS-metabolizing proteins implicated to perform various physiological functions in plant growth, development and stress responses. However, no systematical study has been performed in cotton. In the present study, we identified 7 and 7 CAT genes in the genome of Gossypium hirsutum L. Additionally, G. barbadense L., respectively. The results of the phylogenetic and synteny analysis showed that the CAT genes were divided into two groups, and whole-genome duplication (WGD) or polyploidy events contributed to the expansion of the Gossypium CAT gene family. Expression patterns analysis showed that the CAT gene family possessed temporal and spatial specificity and was induced by the Verticillium dahliae infection. In addition, we predicted the putative molecular regulatory mechanisms of the CAT gene family. Based on the analysis and preliminary verification results, we hypothesized that the CAT gene family, which might be regulated by transcription factors (TFs), alternative splicing (AS) events and miRNAs at different levels, played roles in cotton development and stress tolerance through modulating the reactive oxygen species (ROS) metabolism. This is the first report on the genome-scale analysis of the cotton CAT gene family, and these data will help further study the roles of CAT genes during stress responses, leading to crop improvement.

To overcome the limitations associated with studying the interactions between bacterial communities (BCs) and cyanobacteria in natural environments, we compared the structural and functional diversities of the BCs associated with fifteen non-axenic cyanobacterial strains in culture and two natural BCs sampled during cyanobacterial blooms. No significant differences in richness and diversity were found between the natural and cultivated BCs, although some of the cyanobacterial strains had been isolated 11 years earlier. Moreover, these BCs shared some similar characteristics, such as a very low abundance of Actinobacteria, but they display significant differences at the OTU level. Overall, our findings suggest that BCs associated with cyanobacteria in culture are good models to better understand the interactions between heterotrophic bacteria and cyanobacteria. Additionally, BCs associated with heterocystous cyanobacterial strains cultivated in Z8X culture medium without nitrate (Aphanizomenon-Dolichospermum) demonstrated significant differences compared to BCs associated with non-heterocystous strains cultivated in Z8 culture medium (Planktothrix-Microcystis) in terms of their composition and their ability to utilize different carbon sources, suggesting the potential influence of cyanobacterial metabolism and/or culture media on associated BCs. Finally, half of the dominant operational taxonomic units (OTUs) in these BCs were specifically associated with cyanobacteria or other phytoplankton, whereas the remaining OTUs were generally associated with ecosystems containing high Organic Matter (OM) content, such as sludge or intestines.

Due to the stratospheric ozone depletion, several organisms will become exposed to increased biologically active UVB (280-320 nm) radiation, not only at polar but also at temperate and tropical latitudes. Bloom forming cyanobacteria are exposed to UVB radiation on a mass scale, particularly during the surface bloom and scum formation that can persist for long periods of time. All buoyant species of cyanobacteria are at least periodically exposed to higher irradiation during their vertical migration to the surface that usually occurs several times a day. The aim of this study is to assess the influence on cyanobacteria of UVB radiation at realistic environmental intensities. The effects of two UVB intensities of 0.5 and 0.99 W/m(2) in up to 0.5 cm water depth were studied in vitro on Microcystis aeruginosa strains, two microcystin producing and one non-producing. After UVB exposure their ability to proliferate was estimated by cell counting, while cell fitness and integrity were evaluated using light microscopy, autofluorescence and immunofluorescence. Gene damage was assessed by TUNEL assay and SYBR Green staining of the nucleoide area. We conclude that UVB exposure causes damage to the genetic material, cytoskeletal elements, higher sedimentation rates and consequent cell death. In contrast to microcystin producers (PCC7806 and FACHB905), the microcystin non-producing strain PCC7005 is more susceptible to the deleterious effects of radiation, with weak recovery ability. The ecological relevance of the results is discussed using data from eleven years' continuous UVB radiation measurements within the area of Ljubljana city (Slovenia, Central Europe). Our results suggest that increased solar radiation in temperate latitudes can have its strongest effect during cyanobacterial bloom formation in spring and early summer. UVB radiation in this period may significantly influence strain composition of cyanobacterial blooms in favor of microcystin producers.

Micocystin (MC) exists in Microcystis cells in two different forms, free and protein-bound. We examined the dynamic change in extracellular free MCs, intracellular free MCs and protein-bound MCs in both batch cultures and semi-continuous cultures, using high performance liquid chromatography and Western blot. The results showed that the free MC per cell remained constant, while the quantity of protein-bound MCs increased with the growth of Microcystis cells in both kinds of culture. Significant changes in the dominant MC-bound proteins occurred in the late exponential growth phase of batch cultures, while the dominant MC-bound proteins in semi-continuous cultures remained the same. In field samples collected at different months in Lake Taihu, the dominant MC-bound proteins were shown to be similar, but the amount of protein-bound MC varied and correlated with the intracellular MC content. We identified MC-bound proteins by two-dimensional electrophoresis immunoblots and mass spectrometry. The 60 kDa chaperonin GroEL was a prominent MC-bound protein. Three essential glycolytic enzymes and ATP synthase alpha subunit were also major targets of MC-binding, which might contribute to sustained growth in semi-continuous culture. Our results indicate that protein-bound MC may be important for sustaining growth and adaptation of Microcystis sp.

Lake Taihu is the third-largest freshwater lake in China and has been suffering from cyanobacterial blooms for over two decades. The northern part of the lake, Meiliang Bay, is known to be at high risk of dense and sustained Microcystis blooms and toxins. This study aimed to investigate and record the annual and seasonal dynamics of toxic genotype, Microcystis morphospecies succession and microcystin variation. It also aimed to find out the underlying driving factors influencing the dynamic changes. Microcystin (MC) and the Microcystis genotype were quantified using HPLC and quantitative real-time PCR, respectively. Our study, over three consecutive years, showed that the pattern of morphospecies succession was seasonally distinct and annually consistent. During the same period in 2012, 2013 and 2014, the average MC were, on dry weight basis, 733 μg·g−1, 844 μg·g−1, 870 μg·g−1, respectively. The proportion of toxic Microcystis accounted for 41%, 44% and 52%, respectively. Cell bound microcystin was found to correlate with the percentage of toxic Microcystis. Based on historical and current data, we conclude that annual bloom toxicity was relatively stable or possibly increased over the last decade.
