The relevance of the study is explained by the need to consider the functioning of the mechanisms of discursive sensemaking and an impact on the recipients of information, taking into account the new linguosemiotic opportunities which modern political communication provides and which are manifested not only in the real life, but also in the virtual digital environment. The multimedia technologies are gaining an increasing role, allowing to encode discursive messages, using semiotically polycode format, combining the traditional verbal way of presenting information along with iconic, color, graphic, metatext and other formats. The aim of the study is to prove that semiotic parameters of visual metaphor as a polycode sign functioning in the discursive environment within the framework of the pragmatic situation “Elections”, determine its interpretative and semantic potential and interacting with other sign codes result in new meanings and interpretations. The materials for the study of visual metaphors are the official logos used during the 2024 election campaign in Mexico. The research methodology, which allows for a systematic study of visual metaphors semiotics, is based on the interdisciplinary and polyparadigmatic model of discourse studies and implies an integral approach that combines the general scientific methods of analysis and synthesis, comparison and analogy with the method of sociolinguistic interpretation, semantic, interpretive, linguocognitive and discourse analysis. It has been found out that the linguosemiotic paradigm of Mexican electoral discourse integrates verbal and non-verbal semiotic codes to express meanings, conceptualize and categorize reality. By using of various semiotic codes, the metaphor-logo condenses the semantic content and, as a cognitive model based on analogy, acts as a means of sensemaking, modelling the perception of the world in the interests of the addresser. The use of polysemantic iconic signs-images, understandable to a wide audience of voters, translates connotative, ideological and social meanings. Thus, a visual metaphor as a way of logical comprehension and verbalization of reality functions as a significant component of the cognitive space within electoral discourse.

Abstract Tongue cancer at a young age demonstrates an increase in incidence, aggressiveness, and poor response to therapy. Classic etiological factors for head and neck tumors such as tobacco, alcohol, and human papillomavirus are not related to early-onset tongue cancer. Mechanisms of development and progression of this cancer remain unclear. In this study, we performed spatial whole-transcriptome profiling of tongue cancer in young adults compared with older patients. Nine patients with tongue squamous cell carcinoma (T2-3N0-1M0) were included and divided into two groups: younger (n = 5) and older than 45 years (n = 4). Formalin-fixed paraffin-embedded (FFPE) and fresh frozen (FF) samples of tumor tissue from 4 young and 5 older patients, respectively, were used for spatial transcriptomic profiling using the 10 × Genomics Visium. The findings were validated using SeekGene single cell full-length RNA sequencing (1 young vs 1 older patient) and TCGA data (15 young vs 70 older patients). As a result, we performed the first successful integration of spatial transcriptomics data from FF and FFPE samples and revealed distinctive features of tongue cancer in young adults. Oxidative stress, vascular mimicry, and MAPK and JAK-STAT pathways were enriched in early-onset tongue cancer. Tumor microenvironment demonstrated increased gene signatures corresponding to myeloid-derived suppressor cells, tumor-associated macrophages, and plasma cells. The invasive front was accompanied by vascular mimicry with arrangement of tumor-associated macrophages and aggregations of plasma cells and lymphocytes organized into tertiary lymphoid structures. Taken together, these results indicate that early-onset tongue cancer has distinct transcriptomic features and molecular mechanisms compared to older patients.

Abstract Background Previous studies have reported that periderm (the outer ectodermal layer) in zebrafish partially expands into the mouth and pharyngeal pouches, but does not reach the medial endoderm, where the pharyngeal teeth develop. Instead, periderm-like cells, arising independently from the outer periderm, cover prospective tooth-forming epithelia and are crucial for tooth germ initiation. Here we test the hypothesis that restricted expansion of periderm is a teleost-specific character possibly related to the derived way of early embryonic development. To this end, we performed lineage tracing of the periderm in a non-teleost actinopterygian species possessing pharyngeal teeth, the sterlet sturgeon (Acipenser ruthenus), and a sarcopterygian species lacking pharyngeal teeth, the axolotl (Ambystoma mexicanum). Results In sturgeon, a stratified ectoderm is firmly established at the end of gastrulation, with minimally a basal ectodermal layer and a surface layer that can be homologized to a periderm. Periderm expands to a limited extent into the mouth and remains restricted to the distal parts of the pouches. It does not reach the medial pharyngeal endoderm, where pharyngeal teeth are located. Thus, periderm in sturgeon covers prospective odontogenic epithelium in the jaw region (oral teeth) but not in the pharyngeal region. In axolotl, like in sturgeon, periderm expansion in the oropharynx is restricted to the distal parts of the opening pouches. Oral teeth in axolotl develop long before mouth opening and possible expansion of the periderm into the mouth cavity. Conclusions Restricted periderm expansion into the oropharynx appears to be an ancestral feature for osteichthyans, as it is found in sturgeon, zebrafish and axolotl. Periderm behavior does not correlate with presence or absence of oral or pharyngeal teeth, whose induction may depend on ‘ectodermalized’ endoderm. It is proposed that periderm assists in lumenization of the pouches to create an open gill slit. Comparison of basal and advanced actinopterygians with sarcopterygians (axolotl) shows that different trajectories of embryonic development converge on similar dynamics of the periderm: a restricted expansion into the mouth and prospective gill slits.

Chlamydia is a group of zoonotic diseases caused by Chlamydiaceae spp., which carries high potential risks for animal welfare, public health and animal husbandry. The application of effective vaccines for livestock is still one of the major important tools of managing Chlamydia infection. In this study, the main characteristics of the currently available Russian commercial inactivated whole-cell mono-, polyvalent, and combination vaccines for the prevention of either a single Chlamydia infection (monoinfection) or multi-pathogen infection in livestock, cattle, small ruminants, pigs, farm birds, and fur animals are briefly reviewed. All of the vaccines are inactivated, absolutely safe for the immunization of target livestock and naive untreated contacting animals in the same herd and produce no documented serious vaccine-related complications. Since chlamydia has not yet been eradicated in the world, the infection persists in nature and can be transmitted from infected livestock, wild, domestic animals, and migratory birds to unprotected farm animals. Thus, the availability of effective and safe chlamydia vaccines remains an important aspect for the prevention and control over Chlamydia infection in Russian Federation and worldwide.

Introduction. Tumor-associated macrophages (TAMs) are essential innate immune cells in the tumor microenvironment. TAMs can stimulate cancer cell proliferation and primary tumor growth, angiogenesis, lymphangiogenesis, cancer cell invasiveness in vessels and metastatic niche formation as well as support chemotherapy resistance. TAMs are phenotypically diverse both in various cancer localizations and in intratumoral heterogeneous compartments. Tumor-specific modeling of TAMs is necessary to understand the fundamental mechanism of pro- and anti-tumor activity, to test their interaction with existing therapies, and to develop TAM- targeted immunotherapy. Aim of study: To investigate cancer-specific transcriptomic features of ex vivo human TAM models. Material and Methods. Here we compared transcriptomic profiles of TAMs for breast, colorectal, ovarian, lung, and prostate cancers ex vivo. Human monocytes were isolated from buffy coats, and then stimulated by the tumor cell conditioned medium ex vivo. Using real-time PCR, we quantified the expression of key TAM biomarkers including inflammatory cytokines, scavenger-receptors, angiogenesis-regulating genes, and matrix remodeling factors. Results. PCR analysis allowed revealing cancer-specific expression profiles of modeled TAMs. By comparing the existing knowledge about TAM phenotypes in human tumors in vivo with the collected data, we discuss the advantages and limitation of ex vivo TAM models derived from human blood monocytes. Conclusion. Monocytes-derived macrophages stimulated with cancer cell-conditioned medium can, to a certain extent, allow modeling of cancer-specific programming of TAMs. Our model system is valuable to examine agents reprogramming key TAM pro-tumoral activities, and for the reproducible analysis of mechanistic events that program tolerogenic status of TAMs towards cancer cells.

Cognitive studies describe explicit and implicit means of revealing cognitive and linguistic skills. Allusions play a particular role in distributed cognition in language and speech. This research featured the cognitive potential of linguistic allusion in line with the theory of distributed cognition. The authors used the methods of conceptual and contextual analyses to clarify the main research concepts, describe allusion as a cognitive phenomenon, and define its functions. The article covers such issues as the interaction between the speaker and the language within the linguistic eco-system, the concepts of linguistic ecology and ecological linguistics, and the biological approach in linguistics. Allusion appeared to have constitutive and verifying functions. The constitutive function arranged discourse by correlating the information given by the respondent with the additional meanings that occur in the process of communication. The verifying function provided the accuracy of perception. These functions made it possible to use allusion to describe distributed cognition in speech as consisting of three stages: evaluation, correlation, and interaction.

Immunotherapy represents a groundbreaking therapeutic approach, based on the immune system’s intrinsic capacity to interfere with tumor progression, that opens the horizons in the treatment of endometrial cancer. However, the clinical efficacy of immunotherapy is hampered by the development of resistance in patients. The resistance to immunotherapy is multifactorial mechanism, encompassed genetic and epigenetic alterations in tumor cells modulating immune checkpoint molecules, resulted in escaping immune surveillance. The tumor microenvironment can orchestrate an immunosuppressive milieu, attenuating the immune response and facilitating tumor progression. To overcome immunotherapeutic resistance in endometrial cancer we must bring to light the mechanisms of intricate interplay between neoplastic cells, the host immune system, and the tumor microenvironment. The identification of predictive biomarkers for immunotherapeutic response and the innovative agents capable of reversing resistance pathways must be developed. Our review summarizes accumulated data on the role of cells of the tumor microenvironment and their regulatory molecules in the mechanisms underlying therapeutic effects of immune checkpoint inhibitors, including resistance to therapy. Major question we raise here – which group of patients is the most favorable to achieve durable immunotherapy response in endometrial cancer?

Brain-derived neurotrophic factor (BDNF) is a secreted molecule that plays an important role in the survival and growth of nerve cells. BDNF undergoes complex post-translational processing with cellular proteases. Pro- and mature BDNF forms bind to different receptor types in the brain. BDNF is prominent in the neonatal cerebral cortex. The neonatal period is critical for the proper development of the brain and nervous system. Disruptions in these critical periods can have long-lasting effects on behavior and mental health. Individuals who experience adverse effects in the neonatal period have demonstrated a predisposition to depression and other neurobehavioral disorders. In this work we studied the influence of transient expression (P3–P8) elevation of pro-, mature and mutant forms of BDNF that could not be processed with cellular convertases in the neonatal medial prefrontal cortex (mPFC) on anxiety and depressive-like behavior in adolescents. Elevated expression of mature BDNF (LV-BDNF) increased anxiety and depressive-like behavior at P30. Only immobility in the tail suspension test was increased after expression of mutant BDNF (LV-pBDNF mut). Using our RNA-seq data and available online sn-RNAseq results, we investigated transcriptomic changes in the neonatal mPFC at P8 that underlie subsequent behavioral changes. Mature BDNF expression caused an increased transcriptional response in perivascular stromal cells (PSC) with such genes: Ptgds, Slc6a13, Slc22a6, Bnc2, Slc13a4, Aldh1a2. Based on GWAS data, Ptgds is a candidate gene associated with ADHD and bipolar disorder Pujol-Gualdo et al. (2021); Marín-Méndez et al. (2012); Munkholm et al. (2015). LV-pBDNF mut caused a complete opposite set of transcriptional changes in the PSC compared to LV-BDNF. The observed similar behavioral phenotype after expression of mature and mutant forms of BDNF together with the detected genes related to bipolar disorder underpinned that Bdnf could play a substantial role in the pathogenesis of this neurobehavioral disorder.

Abstract Background Assessing the historical dynamics of key food web components is crucial to understand how climate change impacts the structure of Arctic marine ecosystems. Most retrospective stable isotopic studies to date assessed potential ecosystem shifts in the Arctic using vertebrate top predators and filter-feeding invertebrates as proxies. However, due to long life histories and specific ecologies, ecosystem shifts are not always detectable when using these taxa. Moreover, there are currently no retrospective stable isotopic studies on various other ecological and taxonomic groups of Arctic biota. To test whether climate-driven shifts in marine ecosystems are reflected in the ecology of short-living mesopredators, ontogenetic changes in stable isotope signatures in chitinous hard body structures were analysed in two abundant squids (Gonatus fabricii and Todarodes sagittatus) from the low latitude Arctic and adjacent waters, collected between 1844 and 2023. Results We detected a temporal increase in diet and habitat-use generalism (= opportunistic choice rather than specialization), trophic position and niche width in G. fabricii from the low latitude Arctic waters. These shifts in trophic ecology matched with the Atlantification of the Arctic ecosystems, which includes increased generalization of food webs and higher primary production, and the influx of boreal species from the North Atlantic as a result of climate change. The Atlantification is especially marked since the late 1990s/early 2000s. The temporal patterns we found in G. fabricii’s trophic ecology were largely unreported in previous Arctic retrospective isotopic ecology studies. Accordingly, T. sagittatus that occur nowadays in the high latitude North Atlantic have a more generalist diet than in the XIXth century. Conclusions Our results suggest that abundant opportunistic mesopredators with short life cycles (such as squids) are good candidates for retrospective ecology studies in the marine ecosystems, and to identify ecosystem shifts driven by climate change. Enhanced generalization of Arctic food webs is reflected in increased diet generalism and niche width in squids, while increased abundance of boreal piscivorous fishes is reflected in squids’ increased trophic position. These findings support opportunism and adaptability in squids, which renders them as potential winners of short-term shifts in Arctic ecosystems.

The etiology of cerebral palsy in children with intrauterine hypotrophy at birth and developmental delay is often explained by chronic intrauterine hypoxia. However, children with muscle hypotonia and developmental delay require genetic examination. The aim of this study is to report a case of mitochondrial disease caused by FBXL4 gene mutations and to identify main diagnostic criteria for mitochondrial DNA (mtDNA) depletion syndromes (MDS) in early childhood. Mitochondrial DNA depletion syndrome-13 is associated with FBXL4 gene mutations located in the 6q16.1–q16.27 locus. This disorder was first described in 2013 by P.E. Bonnen and X. Gai independently. MDS are a clinically and genetically heterogeneous group of diseases inherited by an autosomal recessive type and caused by mutations in genes that support the biogenesis and integrity of mtDNA. Encephalomyopathic mtDNA depletion syndrome-13 (MTDPS13) (OMIM: 615471) is an exceedingly rare autosomal recessive disease caused by biallelic mutations in the FBXL4 gene (MIM: 605654) with an estimated prevalence of 1 case per 100,000– 400,000 newborns. The disease onset is usually observed in the neonatal period; 75 % of patients develop symptoms by the age of 3 months. In the majority of cases, mtDNA depletion syndrome-13 manifests itself in the early neonatal period; however, in some patients, the disease onset was registered by the age of 24 months. The disease is characterized by encephalopathy, hypotension, lactic acidosis, severe developmental delay, and changes in the area of basal ganglia revealed by magnetic resonance imaging of the brain. FBXL4-related encephalomyopathy is a multisystem disease primarily affecting the central nervous system, heart, and liver. It is characterized by different clinical manifestations such as lactic acidosis, developmental delay, generalized hypotension, nutritional disorders, and growth retardation. Some patients demonstrate specific facial features, including prominent forehead, sinus-shaped folds, thick eyebrows, long eyelashes, epicanthus, short eye slits, hypertelorism, wide and depressed nose bridge, long and smooth labial groove, thin upper lip, and low-set ears. The disease prognosis is extremely poor; most children die before the age of 4 years. Approximately half of the patients suffer from microcephaly and hyperammonemia. The outcome varies; death was reported in 30 % of cases. Mean time to death was 3 years (median – 2 years). The diagnosis is crucial for medical and genetic counseling and possible prenatal diagnosis.

Abstract Listeria monocytogenes (Lm) is a highly pathogenic bacterium that can cause listeriosis, a relatively rare food-borne infectious disease that affects farm, domestic, wild animals and humans as well. The infected livestock is the frequent sources of Lm. Vaccination is one of the methods of controlling listeriosis in target farm animals to prevent Lm-associated food contamination. Here we report the complete sequence of the Lm strain AUF attenuated from a fully-virulent Lm strain by ultraviolet irradiation, successfully used since the 1960s as a live whole-cell veterinary vaccine. The de novo assembled genome consists of a circular chromosome of 2,942,932 bp length, including more than 2,800 CDSs, 17 pseudogenes, 5 antibiotic resistance genes, and 56/92 virulence genes. Two wild Lm strains, the EGD and the 10403S that is also used in cancer Immunotherapy, were the closest homologs for the Lm strain AUF. Although all three strains belonged to different sequence types (ST), namely ST12, ST85, and ST1538, they were placed in the same genetic lineage II, CC7.

Background and Objectives: The development of resistance of microorganisms to existing antibacterial agents requires constant updating of existing drugs and research in the search for alternative sources of active substances. In recent years, the problem of the emergence of microorganisms resistant to all existing antimicrobial drugs has become systematic and requires significant attention from researchers to search for alternative sources of active substances. The main problem in the development of drugs based on antimicrobial peptides is the search for optimal solutions in the preparation of these substances. Therefore, optimization and search for methods of isolation, analysis and control of protein fractions of water-soluble peptides used for the subsequent development of antibacterial drugs based on them is an urgent task. Materials and Methods: Optimal conditions and methods have been selected for the preparative production of water-soluble peptides isolated from the biomass of Hermetia illucens larvae. Optimization and search of methods for isolation, analysis and control of protein fractions of these water-soluble peptides will ensure the accuracy of the results and obtain optimal amounts of protein fractions. Results: It has been found that the use of molecular sieves makes it possible to obtain a mixture of three peptides with a difference in chromatographic retention time of less than 1 minute, which has been confirmed by three parallel experiments on the isolation and purification of peptides. During HPLC it has been noted that protein fractions 1 and 2 have similar values and the first and third protein fractions have a smaller difference in retention time, which is why there is no complete separation of these chromatographic peaks. Comparison of the percentage of the area of the peptides obtained allows us to talk about the possibility of obtaining peptides of the same size from H. illucens larvae by HPLC, and in combination with DLS to obtain protein fractions with very similar physicochemical and physical characteristics, since this type of chromatography separates substances according to their size. Conclusion: The use of high-performance liquid chromatography makes it possible to establish the reproducibility of the method of isolation of antimicrobial peptides by cold extraction with water and further stages of protein purification, salting and molecular sieve chromatography, which, in correlation with DLS analysis, makes it possible to reliably identify the peptides obtained, and the developed technology of isolation and purification makes it possible to produce these proteins on an industrial scale at low cost.

Background. Pityriasis amiantacea is a reaction of the skin to various inflammatory diseases (seborrheic dermatitis, psoriasis, atopic dermatitis, chronic lichen simplex and others) that manifests by thick gray-yellow scaly crusts surrounding the hair. Сrusts are often attached to hair quite firmly and an effort to remove them leads to hair pulling, that in combination with the inflammatory process and secondary infection can sometimes lead to the formation of secondary traumatic cicatricial alopecia or non-scarring alopecia with subsequent hair growth.Objective. The aim is to study and analyze modern scientific data regarding the clinical features and treatment of pityriasis amiantacea, and demonstrate our own clinical observation.Materials and methods. The article discusses current information about pityriasis amiantacea, a rare condition that can be a manifestation of various inflammatory diseases of the scalp. The article discusses in detail the clinical manifestations and variants of the process, and gives a description of the features of primary conditions that can lead to its development. The authors presented their own clinical case of a case of pityriasis amiantacea in a 21-year-old man, which developed as a complication of seborrheic dermatitis and accompanied by alopecia.Conclusion. There is no single approach to the treatment of pityriasis amiantacea; the choice of therapy depends on the underlying disease. Determining the primary disease that caused the development of pityriasis amiantacea is a difficult diagnostic task, since regardless of the etiology, the manifestations of pityriasis amiantacea on the scalp are almost the same. The persistence of the process and the lack of generally accepted approaches to therapy require further research. It seems especially significant due to the fact that long-term existence and the absence of treatment, it can lead to the formation of secondary traumatic cicatricial alopecia.

In this article, the author presents the methodology of organizing and conducting distance Olympiads in Korean as a second foreign language in the process of professional training of foreign language teachers.The purpose of the study is to examine specifics of second foreign language distance Olympiads organization in the context of professional training, main objectives of the Korean language Olympiad (increasing motivation, interest in learning Korean, exposure to Korean culture, etc.), and to analyze the main requirements for distance Olympiad tasks of the first round (tests), and tasks of the second round, which are aimed to identify the level of communication abilities. Interview, testing and observation are used as the main methods of research.Results. The methodology proposed by the author was tested in groups of 3–4 year students of Sarsen Amanzholov East Kazakhstan University (educational program 6B01703 foreign language: two foreign languages). The article presents analysis of the distance Olympiad’s results, examples of tasks for each round and the main criteria for evaluating the tasks. The requirements to the second foreign language Olympiad tasks implied: possibility of mass coverage of students with Olympiad tasks; compliance of the tasks with the curriculum; time limit; relying on the students’ experience, including their native language skills and abilities; increasing tasks complexity; variety of tasks forms and content, the tasks contents and novelty; problem nature tasks to stimulate students thinking, language guess; using authentic material for the Olympiad tasks; the use of local history and extralinguistic material. The general complexity of the tasks, and difficulties associated with the nature of the linguistic material were also taken into account.Conclusion. According to the results of the experimental work, the author comes to the following conclusion: the leading principle of organizing distance Olympiads assumes that the university faculty will be focused on the timely identification of the future teachers’ interest to the Korean language, their involvement in preparation for the Olympiad, thus forming their interest in Korean culture and language. Olympiad assignments should be creative so that students learn to find their own ways of solving problems and develop self-education skills. Participation in the Olympiad increases students’ learning motivation, broadens their horizons, and develops their ability to express their opinions. The variety of requirements to the work of participants makes the assessment of each member of the jury quite subjective, but the collective nature of the evaluation procedure ensures correctness and objectivity of the final decision, allows participants to familiarize with the results of their work and consider appeals after each round of the Olympiad participants.

The economic policy pursued has turned China into a major trading power. In terms of GDP, the Chinese economy has been the first economy in the world for a number of recent years; China’s rapidly growing trade flows have made it the largest trading partner for many countries. Over the past twenty years there has been rapid growth in exports and imports. The article puts forward a hypothesis about the presence of structural instability in the dynamics of Chinese exports for the period from 2000 to 2020. The hypothesis was confirmed and two periods were identified in the dynamics of exports and imports with different natures of the main development trend: from 2000 to 2008 and from 2009 to 2020. For each stage, trend equations were selected that describe the dynamics of exports and imports and an interval forecast of indexes for 2021 was made. Materials and methods. During the work, dynamic, structural analysis of analytical and statistical information was used; methods of analytical, logical, systemic, correlation and regression analysis were used, as well as analysis of structural changes. The analysis was carried out using the Statistica 10.0 program.Results. A change in the value of the import coverage ratio by exports was revealed before and after structural changes in the dynamics of the main foreign economic indexes and an assessment of their statistical significance was given. An analysis of the influence of export dynamics on GDP dynamics was carried out and it was found that before structural changes, the influence of export dynamics on GDP dynamics was statistically insignificant, and after a structural change, export dynamics began to have a direct impact on changes in China’s GDP. Conclusion. Analysis of official statistical data on the main indexes of China’s foreign economic activity for the period from 2000 to 2020 allowed us to note that the volume of Chinese exports increased over the period 2010-2019 from $1.58 trillion in 2010 (10.34% of global exports), to $2.50 trillion in 2019 (12.81% of global exports). In 2019, China continued to rank first in terms of export value among all countries in the world. Despite the challenging year of 2020, China’s exports reached almost 2.6 trillion US dollars and increased by 4% compared to 2019.China also occupies a leading position in the export of high-tech goods - their volume is 731.9 billion US dollars, the share of exports in the global volume is 25%. At the same time, the dynamics of Chinese exports for the period from 2000 to 2020 was heterogeneous and characterized by structural instability. Two stages can be distinguished with different characteristics of the main development trend: from 2000 to 2008 and from 2009 to 2020. Based on this, the authors came to the conclusion that to describe the main trend in the development of export and import dynamics for the period from 2000 to 2008, the exponential model is best suited. To describe the main trend in the development of export and import dynamics in the period from 2009 to 2020 - logarithmic model. A study of the consistency of changes in export and import volumes showed that in the period from 2009 to 2020 was characterized by greater consistency in China’s main foreign trade flows than the period from 2000 to 2008. In the period before the structural change in the dynamics of exports and imports, the coverage ratio varied from 105.7% to 127.6%; in the period after the structural change, the coverage ratio changed from 108.9% to 135.4%.Checking the statistical significance of the differences in the coefficient of coverage of imports by exports before and after the structural change showed that the differences are not statistically significant. In the period from 2000 to 2008 the dynamics of China’s exports did not have a statistically significant impact on the dynamics of the country’s GDP. From 2009 to 2020 after structural changes in the nature of the dynamics of the main indexes of China’s foreign trade, changes in exports began to have a statistically significant impact on the dynamics of GDP. A 1% increase in China’s exports now results in a 0.25% increase in GDP.The practical significance of the paper is determined by the developed methodology for analyzing structural changes in the dynamics of exports and imports, examined using the example of China.

Ovarian cancer (OC) is the most lethal gynecologic malignancy worldwide. The major clinical challenge includes the asymptomatic state of the disease, making diagnosis possible only at advanced stages. Another OC complication is the high relapse rate and poor prognosis following the standard first-line treatment with platinum-based chemotherapy. At present, numerous clinical trials are being conducted focusing on immunotherapy in OC; nevertheless, there are still no FDA-approved indications. Personalized decision regarding the immunotherapy, including immune checkpoint blockade and immune cell–based immunotherapies, can depend on the effective antigen presentation required for the cytotoxic immune response. The major aim of our study was to uncover tumor-specific transcriptional and epigenetic changes in peripheral blood monocytes in patients with high-grade serous ovarian cancer (HGSOC). Another key point was to elucidate how chemotherapy can reprogram monocytes and how that relates to changes in other immune subpopulations in the blood. To this end, we performed single-cell RNA sequencing of peripheral blood mononuclear cells (PBMCs) from patients with HGSOC who underwent neoadjuvant chemotherapeutic treatment (NACT) and in treatment-naïve patients. Monocyte cluster was significantly affected by tumor-derived factors as well as by chemotherapeutic treatment. Bioinformatical analysis revealed three distinct monocyte subpopulations within PBMCs based on feature gene expression – CD14.Mn.S100A8.9hi, CD14.Mn.MHC2hi and CD16.Mn subsets. The intriguing result was that NACT induced antigen presentation in monocytes by the transcriptional upregulation of MHC class II molecules, but not by epigenetic changes. Increased MHC class II gene expression was a feature observed across all three monocyte subpopulations after chemotherapy. Our data also demonstrated that chemotherapy inhibited interferon-dependent signaling pathways, but activated some TGFb-related genes. Our results can enable personalized decision regarding the necessity to systemically re-educate immune cells to prime ovarian cancer to respond to anti-cancer therapy or to improve personalized prescription of existing immunotherapy in either combination with chemotherapy or a monotherapy regimen.

Introduction. Chemotherapy is a common treatment for breast cancer. Chemotherapeutic drugs effect blood monocytes, which are major contributors to cancer pathogenesis. However, to date, pro-tumor or anti-tumor programming by chemotherapy of monocytes is controversial.Aim. To characterize changes in phenotypic and transcriptomic profiles of monocytes of breast cancer patients before and after chemotherapeutic treatment.Materials and methods. In a cohort of 50 breast cancer patients, monocyte populations were identified based on their expression of CD14, CD16, CD163, and HLA-DR evaluated by flow cytometry before and after neoadjuvant chemotherapy. Bulk RNA sequencing was adopted to explore the transcriptomic profile of CD14+ monocytes before and after treatment. After treatment, we observed an increase in the activity of signaling pathways related to lipid metabolism and intracellular transport of vesicles from the endoplasmic reticulum, against the background of a decreased response to exposure to interferon γ and interferon α, and foreign molecules (exogenous nucleic acids, viruses and bacteria).Results. In breast cancer patients, neoadjuvant chemotherapy decreased in CD14+16+HLA-DR+ monocytes. Under cytostatic treatment, increased gene expression of MGLL, NR4A2, UCK1, YOD1, ABCA2, PAPSS2, ATP10 (log2FoldChange ≥0.8; false discovery rate (FDR) ≤0.01) and decreased gene expression of KPNA2, ERCC4, JAGN1, RUBCNL, SMYD4, B3GALT4 (log2FoldChange ≥0.8; FDR ≤0.01) were observed in monocytes of patients. Using discriminant analysis, the relative numbers of CD14+16–, CD14+16+, CD14-16+, CD14+16-HLA-DR+, CD14+16+HLA-DR+ and CD14–16+HLA-DR+ monocytes in the blood were found to be valuable in predicting response to neoadjuvant chemotherapy.Conclusion. Thus, association of blood monocytes with chemotherapeutic treatment in breast cancer was revealed.

Abstract Recognizing spelling errors is important for correct writing and reading, and develops over an extended period. The neural bases of the development of orthographic sensitivity remain poorly understood. We investigated event-related potentials (ERPs) associated with spelling error recognition when performing the orthographic decision task with correctly spelled and misspelled words in children aged 8–10 years old, early adolescents aged 11–14 years old, and adults. Spelling processing in adults included an early stage associated with the initial recognition of conflict between orthography and phonology (reflected in the N400 time window) and a later stage (reflected in the P600 time window) related to re-checking the spelling. In children 8–10 years old, there were no differences in ERPs to correct and misspelled words; in addition, their behavioral scores were worse than those of early adolescents, implying that the ability to quickly recognize the correct spelling is just beginning to develop at this age. In early adolescents, spelling recognition was reflected only at the later stage, corresponding to the P600 component. At the behavioral level, they were worse than adults at recognizing misspelled words. Our data suggest that orthographic sensitivity can develop beyond 14 years.

Introduction. The prevalence of patients with extended ureteral strictures has been steadily increasing over the past decades. When it is not possible to perform reconstruction with the tissues of the urinary system, the best solution is intestinal replacement plasty. However, despite the great world experience, the search for the optimal method of intestinal reconstruction does not stop, which determines the relevance of conducting an experimental prospective study.Objective. To evaluate morphological changes in renal parenchyma after ureteral replacement reconstruction by segments of small and large intestine.Materials & methods. Fifteen white giant rabbits (6-months-old, weighing 5 kg) were divided into three groups. Group A (n = 5) underwent small intestine [ilealplasty] replacement plasty. Group B (n = 5) double-flap ileoplasty using the Yang-Monti technique. Group C (n = 5) – large intestine [coloplasy] ureteral reconstruction. Before the operation and before withdrawal from the experiment, the level of creatinine and electrolytes was assessed, kidney ultrasound was performed on days 2, 5, 10. Excretory urography was performed to assess the patency of the anastomoses. Animals were withdrawn from the experiment from day 10 to 30. The material used for morphological study included kidneys from the operated and intact sides, proximal and distal anastomoses.Results. No increase in creatinine level, metabolic disorders were detected in animals. According to ultrasound data, hydronephrosis developed in all animals on day 2 and remained unchanged during the entire follow-up. Group A: no complications were detected. Histologically, the renal parenchyma showed moderate signs of chronic inflammation, single foci of lymphoid infiltration, but there were no irreversible processes in the form of necrosis and sclerosis. Group B: complications — stricture in the anastomosis area of the detubularised fragments and necrosis of the small intestine due to compression of the mesentery by the graft vascular stem. Histologically there were marked dilatation of the tubules at all levels, enlargement of the Bowman-Shumlansky capsule, as well as signs of moderate inflammatory process, there were hydropic and hyaline-droplet dystrophy, tubular necrosis foci. Group C: complications — accumulation of a significant amount of grit, mucus, and fibrin in the colocystoanastomosis area. Histologically, the renal parenchyma showed a marked inflammatory process, in particular, purulent inflammation with demarcation zones, infiltration with polymorphonuclear leukocytes and bacterial cells, foci of necrosis and sclerosis of both stroma and tubules, dilatation of tubules and Bowman-Schumlansky capsules.Conclusion. The use of an unchanged ileal-graft reconstruction of an extended ureteral defect showed acceptable histological results in an animal model, which confirms the feasibility of its use in clinical practice. The absence of foci of necrosis and fibrosis in the renal parenchyma indicates the preserved functional potential, which suggests the stabilization of renal function in the long term.

Background. Colorectal cancer is one of the most common cancers and the second leading cause of cancer-related deaths worldwide. Population-based studies have shown that 25–30 % of patients with colorectal cancer have synchronous liver metastases at the time of diagnosis. despite modern advances in oncology and surgery, only 25 % of patients with metastatic colorectal cancer (mCRC) are suitable for liver resection, which is the only curative treatment option for these patients. In recent years, the indications for curative treatment of mCRC have expanded. due to the introduction of new targeted drugs into clinical practice, the tumor response rate to preoperative therapy has increased, thus increasing surgical resection rate. Some patients experience a complete clinical response, which is defined as the complete disappearance of liver metastases. However, 30–70 % of patients develop recurrent metastases in the liver within the first year of follow-up, and currently, even in the presence of complete regression of metastases, it is recommended to perform resection of the initially affected hepatic segments. Case presentation. We describe a case of complete clinical and radiological response of liver metastases after chemotherapy in a patient with sigmoid colon cancer. Molecular genetic analysis revealed the wild type of the Kras, Nras and Braf genes. The patient received 3 courses of preoperative chemotherapy according to the FOLFOxIRI + Cetuximab regimen. Laparoscopic sigmoid colon resection with d3 lymph node dissection was followed by adjuvant chemotherapy with oxaliplatin and 5-fluorouracil (12 cycles). After 16 months of follow-up, no evidence of colon cancer recurrence and liver metastasis was found. Conclusion. Current targeted therapy has demonstrated efficacy in treating mCRC with synchronous liver metastases and makes it possible, in selected cases, to avoid the liver resection provided that a complete clinical and radiological response of the metastases is achieved.

In this work, the effect of high-entropy alloy powder preparation on the coatings deposited via high-velocity oxygen fuel sprayings was studied. The powders of FeNiCoCrMo0.5Al1.3 composition were prepared by milling and gas atomization. The structures, porosity, phase composition, and microhardness of the coatings produced from mechanically alloyed and gas-atomized powders were compared. The influence of milling parameters on the powder phase composition and morphology was studied. Milling at 600 rpm for 1.5 h allowed the production of mechanically alloyed powder with a homogeneous distribution of Fe, Ni, and Al and thin lamellas enriched with Co, Cr, and Mo. Despite the difference in the feedstock powders’ phase compositions, the phase compositions of the coatings deposited from mechanically alloyed and gas-atomized powders are the same consisting of BCC, FCC solutions, and oxide. The amount of FCC solutions and oxide in the coating depends on the size distribution of the sprayed powder. It was found that the phase composition and the properties of the coatings deposited from the mechanically alloyed and gas-atomized powders of similar sizes are similar.

Prussian blue nanoparticles (PBNPs) have been identified as a promising candidate for biomimetic peroxidase (POD)-like activity, specifically due to the metal centres (Fe3+/Fe2+) of Prussian blue (PB), which have the potential to function as catalytically active centres. The decoration of PBNPs with desired functional polymers (such as amino- or carboxylate-based) primarily facilitates the subsequent linkage of biomolecules to the nanoparticles for their use in biosensor applications. Thus, the elucidation of the catalytic POD mimicry of these systems is of significant scientific interest but has not been investigated in depth yet. In this report, we studied a series of poly(ethyleneimine) (PEI)-mediated PBNPs (PB/PEI NPs) prepared using various synthesis protocols. The resulting range of particles with varying size (~19–92 nm) and shape combinations were characterised in order to gain insights into their physicochemical properties. The POD-like nanozyme activity of these nanoparticles was then investigated by utilising a 3,3′,5,5′-tetramethylbenzidine (TMB)/H2O2 system, with the catalytic performance of the natural enzyme horseradish peroxidase (HRP) serving as a point of comparison. It was shown that most PB/PEI NPs displayed higher catalytic activity than the PBNPs, with higher activity observed in particles of smaller size, higher Fe content, and higher Fe2+/Fe3+ ratio. Furthermore, the nanoparticles demonstrated enhanced chemical stability in the presence of acid, sodium azide, or high concentrations of H2O2 when compared to HRP, confirming the viability of PB/PEI NPs as a promising nanozymatic material. This study disseminates fundamental knowledge on PB/PEI NPs and their POD-like activities, which will facilitate the selection of an appropriate particle type for future biosensor applications.

The coordination environment of magneto-luminescent Dy3+-based Single-Molecule Magnets (SMM) is a crucial factor influencing both magnetic and luminescent properties. In this work, we explore how triphenylmethanolate (Ph3CO−), in combination with other ligands, can modulate the structure and, therefore, the magnetic properties of Dy3+-based SMM. Using triphenylmethanolate in combination with THF and pyridine (Py) as co-ligands, we synthesized a series of mononuclear cis-[Dy(OCPh3)2(THF)4][BPh4]·(2,6-Me2C5H3N) (1), trans-Dy(OCPh3)3(THF)2 (2), fac-Dy(OCPh3)3(py)3 (3) and dinuclear [(Ph3CO)Dy(THF){(μ2–Cl)2Li(THF)2}μ2–Cl]2 (4) complexes where the Dy3+ ion presents five- or six-coordinate geometries. Dinuclear compound 4 exhibits a genuine SMM behavior with a relatively high energy barrier of 421 cm−1, while mononuclear complexes 1–3 are field-induced SMM. These complexes also present Dy3+-characteristic luminescence, highlighting their multifunctional character.

Background. The problem of eliminating the surgical soft tissue defect after radical surgical treatment of the epithelial coccygeal tract is relevant. The purpose was to develop and clinically test an original method of radical surgical treatment of epithelial coccygeal cysts, in which vertical polyspast sutures are used to eliminate a postoperative wound. Materials and methods. According to the proposed method, 15 patients were operated on. In the postoperative period, an assessment was made of the average time of intervention, an analysis of the presence of postoperative complications, an assessment of the severity of pain on a visual analogue scale, an average length of hospital stay, and an examination of patients 3-7 months after surgery was carried out. Results. The average intervention time was 21±2 minutes. Complications were not recorded. The severity of pain syndrome ranged from 3.6±0.3 to 0.005±0.005 points, depending on the day of the postoperative period. The average duration of hospitalization was 7±1 days. There were no recurrences of the disease within 3-7 months after the operation. Conclusions. The first experience of applying the proposed operation showed the effectiveness of the technique. Further comparison of the results of the proposed technique with those after the use of traditional surgical interventions seems promising.

Background. Desmoid fibroma is a fibroblastic neoplasm of intermediate biological significance without metastatic potential, but with a tendency to local recurrence and infiltration of surrounding tissues. These tumors are extremely rare (2-4 cases per 1 million people). Information about desmoid fibromatosis is presented in isolated publications. The purpose of the study: to describe a clinical case of desmoid abdominal fibroma in a man of working age, including: identification of the features of the histological picture of the tumor, interpretation of the results of immunohistochemical examination, differential diagnosis with other diseases. Materials and methods. The data of the results of multispiral computed tomography (MSCT) of abdominal organs, macroscopic and microscopic examination of surgical material in the form of tumors, fragments of the abdominal cavity, omentum and aponeurosis, immunohistochemical examination with the following markers: CD34, CD31, LMO2, Podoplanin, CD68, anti-S100, HMB45, B-Catenin, SMA, Desmin. Results and conclusions. In this study, an analysis of a clinical case of rare abdominal fibroma was presented, and important points for differential diagnosis with other soft tissue neoplasms were identified. Raising awareness on this topic increases the likelihood of timely detection and verification of desmoid fibroma.

The purpose of the study is to establish significance of graphic symbols - semiotic, cognitive and functional characteristics of emoticons and emoji as paralinguistic means of written communication in Spanish political Internet communication. The materials of the study are the publications of Spanish government bodies, political parties and politicians in the official Twitter accounts. The relevance of the study stems from the need to analyze the functional and impact potential of emoticons and emoji as an integral part of modern Spanish political discourse in the Internet and determine their characteristics from the standpoint of theoretical linguistics, discourse theory, semiotics and pragmalinguistics. The paper concludes that the semiotic characteristics of emoji and emoticons as iconic signs and symbols allow them to perform a number of important functions in the discursive Internet communication, for instance, nominative - identifying referents, due to the fact that they are associated with them by physical similarity; syntactic - structuring the punctuation and intonation contour of the message; cognitive - conveying additional conventional meanings implied by the interpretation of the represented objects; communicative - creating an intonation contour and emotional background of the message; influencing - setting the vector of pragmatic interpretation of the message and its empathic component. At the same time, the verbal and non-verbal components of polycode Internet messages are not opposed to each other, but mutually determine the integral perception, representation and interpretation of reality.

Classical reassortment in developing chicken eggs is a well-established technique for obtaining LAIV strains. Naturally generated reassortant vaccine strains are characterized by high reproductive capacity, genetically stable characteristics of temperature sensitivity and cold resistance, which correspond to the characteristics of the MDV involved in crossing with the epidemic virus. Along with antigenic relevance, natural reassortment ensures attenuation of vaccine strains, good reproduction capacity in upper respiratory tract cells and inability to reproduction in the lower respiratory tract. With classical reassortment, the speed and efficiency of obtaining vaccine reassortants largely depend on the properties of epidemic virus, and therefore cannot be stable. The potential of reverse genetics is attractive because it allows to obtain vaccine reassortants quickly and efficiently, reduce the likelihood of spontaneous mutations; however, the vaccine strain is deprived of the advantages of natural selection, in which the most viable clones are selected. This study presents the results of comparatively assessed A(H3N2) LAIVs obtained in parallel by classical reassortment and reverse genetics according to criteria confirming that vaccine strains inherit the necessary properties that guarantee their harmlessness and high reproduction in chicken embryos. Strains for LAIV obtained by both methods retained all attenuating mutations inherited from the MDV, were highly reproductive at the optimal temperature, with temperature sensitivity corresponded to the MDV. However, strains obtained by reverse genetics, was observed to have partial loss of cold resistance in comparison with that of the MDV and classical reassortants. Reduced cold adaptation may negatively affect vaccine effectiveness. It is important that after several additional passages in chicken embryos at low temperature, the cold resistance of the vaccine strain, assembled by reverse genetics, was increased. Credibly that cold resistance is a phenotypic trait, the degree of manifestation of which depends on the temperature conditions of virus multiplication. The selective factor of reduced incubation temperature is missing in reverse genetics. In order for the cold-adapted phenotype to be fully realized, additional passages at low temperature of RG-reassortants are necessary. Thus, the reverse genetics method using plasmid technology allows to effectively prepare reassortant strains for LAIV. An important stage in obtaining vaccine strains using genetic engineering techniques should be the control of their cold-adapted phenotype and its optimization by additional passages at low temperature.

This article presents the results of a study of the influence of radionuclide and chemical pollution on the morphological and anatomical parameters of Calamagróstis epigéjos plants growing in the territory of “Degelen” at the Semipalatinsk Test Site (STS). Quantitative data of morphological and anatomical parameters are given, and the content of radionuclides and toxic elements in samples of plants obtained. Statistical processing of the obtained data was conducted. The results revealed that elevated concentrations of radionuclides 137Cs and 90Sr, and the calculated absorbed dose, do not have effects on plants. Changes in the anatomical parameters of leaves and stems were observed at elevated concentrations of the elements: for leaves—Al, Pb, Sr, U, Ni, Rb, Sm; for stems—Al, Cr, Cd, U, Cu, Be, Ni, Sm, Fe. The mesophyll of the leaves and the epidermis of the stems were the most exposed to toxic elements. The data of the anatomical parameters are recommended to be used as indicative parameters of plants grown in chemically contaminated areas.

Triboelectric nanogenerators (TENGs), as novel electronic devices for converting mechanical energy into electrical energy, are better suited as signal-testing sensors or as components within larger wearable Internet of Things (IoT) or Artificial Intelligence (AI) systems, where they handle small-device power supply and signal acquisition. Consequently, TENGs hold promising applications in self-powered sensor technology. As global energy supplies become increasingly tight, research into self-powered sensors has become critical. This study presents a self-powered sport sensor system utilizing a triboelectric nanogenerator (TENG), which incorporates a thermoplastic polyurethane (TPU) film doped with graphene and polytetrafluoroethylene (PTFE) as friction materials. The graphene-doped TPU nanocomposite film-based TENG (GT-TENG) demonstrates excellent working durability. Furthermore, the GT-TENG not only consistently powers an LED but also supplies energy to a sports timer and an electronic watch. It serves additionally as a self-powered sensor for monitoring human movement. The design of this self-powered motion sensor system effectively harnesses human kinetic energy, integrating it seamlessly with sport sensing capabilities.

Background. People who previously had COVID-19 infection have an increased risk of developing cardiovascular diseases. Left ventricular diastolic dysfunction is an early marker of the development of cardiac pathology. Its early detection is important for the adequate therapy order and dynamic monitoring of patients. In this regard, it seems relevant to study the effect of a recent COVID-19 infection on the left ventricular diastolic function and transmitral blood flow parameters in apparently healthy individuals without clinical and instrumental signs of cardiovascular pathology. The aim of the study. To assess the changes in the diastolic and systolic function of the left ventricle, its anatomical parameters and transmitral blood flow parameters in two groups of apparently healthy individuals: those who had and those who had not COVID-19 infection. Materials and methods. Transthoracic echocardiography was performed according to the standard technique and its results were analyzed in 66 examined patients who were recognized as apparently healthy according to the results of regular comprehensive clinical and instrumental studies. The first group included 30 individuals who underwent an echocardiographic study before or during the COVID-19 pandemic, but did not have a coronavirus infection; the second group consisted of 36 people who recovered from COVID-19. The indicators of the presence of left ventricular diastolic dysfunction and the transmitral blood flow parameters were assessed. The study was approved at a meeting of the Medical Ethics Committee under the Ministry of Health of the Republic of Karelia and of Petrozavodsk State University (Minutes No. 47 of 01.11.2023). Results. The frequency of the left ventricular diastolic dysfunction did not differ in the first and second groups of patients. Statistically significant differences were recorded in the average flow deceleration time for both early and late filling of the left ventricle in people who had and did not have COVID-19 infection. A change in the phase structure of the transmitral blood flow may be an early manifestation of intracardiac hemodynamic disorders in people who have recovered from COVID-19.

Objective: To study the current epidemiological trends related to PD in children Methods: In 2015-2016, a cross-sectional one-stage study was conducted to examine the PD of children. The body height and weight were determined during the examination program using standardized equipment and accepted methodology. The Body Mass Index (BMI, Quetelet's index) was expressed in kg/m2 , along with SD values (standard deviation) for both height and BMI. We utilized BMI-for-age percentile growth charts. Results: : 9,662 children between the ages of 1 and 17 were assessed, with 49.7% boys and 50.3% girls. The sample was split equally between urban and rural areas, with 50.0% residing in each. The age distribution of the participants was divided into five age groups: 1-2 y/o (14.5%), 3-6 y/o (19.6%), 7-11 y/o (30.7%), 12-14 y/o (19.1%), and 15-17 y/o (16.1%). Of those surveyed, 65.8% had an average height, 19.0% were below average or short, and 15.2% were above average and tall. The height 2SD below and above the mean height for age and gender was found in 3.7% and 2.7% of children, respectively. Most below-average and low height values were found in early and preschool ages. In boys, gender characteristics were associated with more significant variations in heights towards low and high values. Research shows that children residing in urban areas tend to have better physical development than those in rural areas. Conclusion: Data have been gathered on the current PD of children, focusing on age, gender, and geographical factors.

Abstract Juvenile myelomonocytic leukemia (JMML) is a rare myeloproliferative disease of early childhood that develops due to mutations in the genes of the RAS-signaling pathway. Next-generation high throughput sequencing (NGS) enables identification of various secondary molecular genetic events that can facilitate JMML progression and transformation into secondary acute myeloid leukemia (sAML). The methods of single-cell DNA sequencing (scDNA-seq) enable overcoming limitations of bulk NGS and exploring genetic heterogeneity at the level of individual cells, which can help in a better understanding of the mechanisms leading to JMML progression and provide an opportunity to evaluate the response of leukemia to therapy. In the present work, we applied a two-step droplet microfluidics approach to detect DNA alterations among thousands of single cells and to analyze clonal dynamics in two JMML patients with sAML transformation before and after hematopoietic stem cell transplantation (HSCT). At the time of diagnosis both of our patients harbored only “canonical” mutations in the RAS signaling pathway genes detected by targeted DNA sequencing. Analysis of samples from the time of transformation JMML to sAML revealed additional genetic events that are potential drivers for disease progression in both patients. ScDNA-seq was able to measure of chimerism level and detect a residual tumor clone in the second patient after HSCT (sensitivity of less than 0.1% tumor cells). The data obtained demonstrate the value of scDNA-seq to assess the clonal evolution of JMML to sAML, response to therapy and engraftment monitoring.

Summary: Heterozygous mutations in COL10A1 lead to metaphyseal chondrodysplasia type Schmid (MCDS), a skeletal disorder characterized by epiphyseal abnormalities. Prior analysis revealed impaired trimerization and intracellular retention of mutant collagen type X alpha 1 chains as cause for elevated endoplasmic reticulum (ER) stress. However, how ER stress translates into structural defects remained unclear. We generated a medaka (Oryzias latipes) MCDS model harboring a 5 base pair deletion in col10a1, which led to a frameshift and disruption of 11 amino acids in the conserved trimerization domain. col10a1Δ633a heterozygotes recapitulated key features of MCDS and revealed early cell polarity defects as cause for dysregulated matrix secretion and deformed skeletal structures. Carbamazepine, an ER stress-reducing drug, rescued this polarity impairment and alleviated skeletal defects in col10a1Δ633a heterozygotes. Our data imply cell polarity dysregulation as a potential contributor to MCDS and suggest the col10a1Δ633a medaka mutant as an attractive MCDS animal model for drug screening.

M1/M2 paradigm of macrophage plasticity has existed for decades. Now it becomes clear that this dichotomy doesn't adequately reflect the diversity of macrophage phenotypes in tumor microenvironment (TME). Tumor-associated macrophages (TAMs) are a major population of innate immune cells in the TME that promotes tumor cell proliferation, angiogenesis and lymphangiogenesis, invasion and metastatic niche formation, as well as response to anti-tumor therapy. However, the fundamental restriction in therapeutic TAM targeting is the limited knowledge about the specific TAM states in distinct human cancer types. Here we summarized the results of the most recent studies that use advanced technologies (e.g. single-cell RNA sequencing and spatial transcriptomics) allowing to decipher novel functional subsets of TAMs in numerous human cancers. The transcriptomic profiles of these TAM subsets and their clinical significance were described. We emphasized the characteristics of specific TAM subpopulations – TREM2+, SPP1+, MARCO+, FOLR2+, SIGLEC1+, APOC1+, C1QC+, and others, which have been most extensively characterized in several cancers, and are associated with cancer prognosis. Spatial transcriptomics technologies defined specific spatial interactions between TAMs and other cell types, especially fibroblasts, in tumors. Spatial transcriptomics methods were also applied to identify markers of immunotherapy response, which are expressed by macrophages or in the macrophage-abundant regions. We highlighted the perspectives for novel techniques that utilize spatial and single cell resolution in investigating new ligand-receptor interactions for effective immunotherapy based on TAM-targeting.

We report the synthesis and single-crystal X-ray diffraction, magnetic, and luminescence measurements of a novel family of luminescent cage-like tetranuclear silsesquioxanes (PhSiO1.5)8(LnO1.5)4(O)(C5H8O2)6(EtOH)2(CH3CN)2⋅2CH3CN (where Ln = Tb, 1; Tb/Eu, 2; and Gd, 3), featuring seven-coordinated lanthanide ions arranged in a one-capped trigonal prism geometry. Compounds 1 and 2 exhibit characteristic Tb3+ and Tb3+/Eu3+-related emissions, respectively, sensitized by the chelating antenna acetylacetonate (acac) ligands upon excitation in the UV and visible spectral regions. Compound 3 is used to assess the energies of the triplet states of the acac ligand. For compound 1, theoretical calculations on the intramolecular energy transfer and multiphonon rates indicate a thermal balance between the 5D4 Stark components, while the mixed Tb3+/Eu3+ analog 2, with a Tb:Eu ratio of 3:1, showcases intra-cluster Tb3+-to-Eu3+ energy transfer, calculated theoretically as a function of temperature. By utilizing the intensity ratio between the 5D4→7F5 (Tb3+) and 5D0→7F2 (Eu3+) transitions in the range 11–373 K, we demonstrate the realization of a ratiometric luminescent thermometer with compound 2, operating in the range 11–373 K with a maximum relative sensitivity of 2.0% K−1 at 373 K. These findings highlight the potential of cage-like silsesquioxanes as versatile materials for optical sensing-enabled applications.

Phase evolution and strengthening of the FeNiCoCrMo0.5Al1.3 powder alloy produced via inert gas atomization and annealed in the temperature interval of 300–800 °C have been studied by X-ray diffraction, scanning electron microscopy, energy dispersive X-ray spectroscopy, and microhardness testing. It was found that annealing at 300–600 °C leads to an increase of the element segregations between the several solid solutions with a rise of the lattice misfit (ε) to 1.5 % and microhardness growth to 1070 HV. It was assumed that elastic stress caused by the element partitioning is the main strengthening mechanism: microhardness rises linearly with misfit rise with dHV/dε = 43400 MPa. Sigma arises after the maximum elastic deformation (in 1.5 %) was reached. Formation of the dispersed coherent sigma phase in the annealing interval 600–800 °C results in the microhardness rise. Oxidation that began at 800 °C in 27 h is accompanied with FCC formation due to a depletion of the B2 in Al caused by Al2O3 formation. Estimation of the activation energy of the initial stage of the solid solution decomposition gives a very low value in 0.65eV, apparently caused by the high concentration of quenched vacancies. The activation energy of sigma formation approximately coincides with the activation energy of self-diffusion in BCC metals (about 2.60 eV).

Abstract The potential of photoactive Prussian blue nanoparticles dispersed in thermo‐responsive PNIPAM hydrogels in 4D printing using a stereolithography is investigated with digital light processing. The proportion of Prussian blue nanoparticles used in the resin is chosen to deliver a significant photothermal effect providing a heating above the volume phase transition temperature of the final nanocomposite hydrogels; while, giving only a limited effect on light scattering during the 3D printing process. Four formulations with various amounts of Prussian blue nanoparticles are used to print 3D structures with different shapes, such as Aztec pyramids, cylinders, gyroid porous cubes, and porous films. The shrinkage effect triggered by light irradiation at 808 nm on the as‐obtained nanocomposite hydrogels is demonstrated through the delivery of a representative molecule fluorescein and a triggered 4D shape‐morphing effect.

The modern digital economy requires “new finance”. The subject of the study is to clarify their content, and the goal is to develop the author’s view of the term of “finance”. The relevance of the topic is dictated by the need to analyze complex theoretical questions of technological transformation of financial markets and activities of financial intermediaries. The scientific novelty consists of the theoretical contribution of the theory of finance evolution in the digital era. The main research methods are systemic and logical approaches, the method of theoretical cognition, the historical method and the method of scientific abstraction. The results of the study are clarification the essence of finance and applied issues of their use in Russia and abroad. It will contribute to the development of a scientific doctrine of digital assets circulation; will allow to refine the model of the activities of financial intermediaries’ regulation in our country and lay the foundation for the formation of global norms and rules for the development of national and international financial markets of the CIS, the EAEU, the SCO, and BRICS countries. The conclusions of the study are: a) confirmation of the author’s position on the essence of finance. The paper indicates that, in the context of the economy’s digital revolution, the substance of economic categories stays constant, although their forms vary according to innovation, behavioral characteristics, and other factors; b) without rejecting the rational knowledge formed in the Western scientific school, the authors emphasize that the social sciences cannot be global, they have a national character; c) based on the investigation, the author defines “finance” as a distribution category in the development and utilization of financial resources.

In this article, we report a case of CTNNB1 syndrome (CTNNB1-NDD) in a child with cerebral palsy and also provide a literature review on the problem. CTNNB1 syndrome is an exceedingly rare and poorly studied disorder, which makes it particularly interesting due the difficulties associated with its diagnosis and description of the disease phenotype, as well as highly polymorphic clinical manifestations. Verification of the diagnosis is important to determine the prognosis of a child with cerebral palsy and visual impairment, as well as for reproductive planning in the family.

In this work, we report on the synthesis and investigation of new hybrid multifunctional iron oxide nanoparticles (IONPs) coated by coumarin-bound copolymer, which combine magneto- or photothermal heating with luminescent thermometry. A series of amphiphilic block copolymers, including Coum-C11-PPhOx27-PMOx59 and Coum-C11-PButOx8-PMOx42 bearing luminescent and photodimerizable coumarin moiety, as well as coumarin-free PPhOx27-PMOx57, were evaluated for their utility as luminescent thermometers and for encapsulating spherical 26 nm IONPs. The obtained IONP@Coum-C11-PPhOx27-PMOx59 nano-objects are perfectly dispersible in water and able to provide macroscopic heating remotely triggered by an alternating current magnetic field (AMF) with a specific absorption rate (SAR) value of 240 W.g−1 or laser irradiation with a photothermal conversion efficiency of η = 68%. On the other hand, they exhibit temperature-dependent emission of coumarin offering the function of luminescent thermometer, which operates in the visible region between 20 °C and 60 °C in water displaying a maximal relative thermal sensitivity (Sr) of 1.53%·°C−1 at 60 °C.

IntroductionImmunometabolism is essential factor of tumor progression, and tumor-associated macrophages are characterized by substantial changes in their metabolic status. In this study for the first time, we applied targeted amino acid LC-MS/MS analysis to compare amino acid metabolism of circulating monocytes isolated from patients with breast, ovarian, lung, and colorectal cancer.MethodsMonocyte metabolomics was analyzed by liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/ MS) analysis of amino acid extracts. The targeted analysis of 26 amino acids was conducted by LCMS/MS on an Agilent 6460 triple quadrupole mass spectrometer equipped with an electrospray ionization source and an Agilent 1260 II liquid chromatograph.ResultsComparison of monocytes of cancer patients with monocytes of healthy control individuals demonstrated that in breast cancer most pronounced changes were identified for tryptophan (AUC = 0.76); for ovarian cancer, aminobutyric acid was significantly elevated (AUC= 1.00); for lung cancer significant changes we indented for citrulline (AUC = 0.70). The article presents the results of a study of groundwater contaminated with tritium in the vicinity of the 'Atomic Lake' - a crater filled with water as a result of a thermonuclear explosion on the territory of the former Semipalatinsk test site. This crater was created as part of an experimental thermonuclear explosion in 1965 with the aim of creating an artificial reservoir in arid areas. The study was carried out to identify the source of groundwater contamination near the crater formed from a thermonuclear test. There were two possible factors of pollution: the influence of contaminated water from the crater on the groundwater of the adjacent area, or groundwater polluting the water in the crater. It was necessary to find out the source of groundwater contamination and its connection with the water in the funnel. For this purpose, a study of the geological and lithological conditions of the territory adjacent to the funnel was carried out, which was carried out using drilling operations and hydrological measurements. Drilling work made it possible to study the depth of distribution of groundwater, hydrological work made it possible to determine the conditions of distribution of groundwater, as well as to take samples of groundwater. The assessment of the degree of groundwater contamination was carried out through water sampling and laboratory analysis. As a result, it was established that the geological and lithological conditions of the area limit the flow of contaminated groundwater to the water in the crater - the 'Atomic Lake'. Despite the fact that the waters in the crater from a thermonuclear explosion and the groundwater of the adjacent territory are contaminated with the radionuclide tritium, they have different sources of contamination and are not interconnected. Radionuclide analysis of groundwater showed that increased concentrations of tritium with a specific activity of up to 95 000 Bq/l are found in groundwater near the river bed. Shagan and this is due to the influence of the flow of groundwater coming from other parts of the landfill.

This article presents research findings on 3H in abiotic environmental compartments, specifically, the 'water-soil-air' system. All of the research areas are located within the Semipalatinsk Test Site (STS): the perimeter of the 'Degelen' site, the riverside zone of the Shagan river and the 'background' area-the southeastern part of the STS (SEP). As research progressed, numerical values of 3H and its species were revealed in various environmental compartments. The presence of 3H was registered not only in underground nuclear test locations but also in the 'background' area-SEP. Maximum 3H tritium concentrations in the water were detected at the 'Degelen' site (up to 57000±5000 Bq/kg) and the Shagan riv (up to 61500±6000 Bq/kg), in the air of the 'Degelen' site (up to 56±11 Bq/m3), in the soil of the 'Degelen' site (up to 5170±500 Bq/kg) and the Shagan riv (4100±400 Bq/kg) in the free water, at SEP (up to 1710±170 Bq/kg) in the organic constituent. Based upon all of the findings, 3H was found to be readily distributed in abiotic environmental compartments depending on certain conditions. Research suggests that water plays a key role in 3H migration processes in the natural system of interest. The second most but equally important constituent is soil and microorganisms of plant and animal origin living there. These assumptions are indirectly proven by research findings that show the HTO and HT air concentration dynamics depending on the sampling location.

This paper presents the determination method of the exact geographical coordinates of aboveground nuclear tests (NT) epicenters based on the radioecological study results the example of the Semipalatinsk Nuclear Test Site. By testing the NT Epicenter software for determining the exact geographic coordinates of the NT centers, it was established that it is indeed possible to determine the exact coordinates of most of the aboveground NTs. Their locations are currently determined by the presence are currently determined by the presence of technogenic disturbance of the soil surface in the area of the alleged epicenter (the presence of a crater), as well as by comparing maps of radioactive contamination and a space image. The accuracy of the precise coordinates of the NT is highly dependent on the density of the auxiliary grid: the smaller the pitch of the auxiliary grid, the higher the accuracy of the NT epicenter.

Background and Objectives: Currently, antimicrobial peptides are one of the main sources of alternative antibiotics because they can easily interact with bacterial peptidoglycan by penetrating or dissolving biofilms with minimal side effects. However, from a practical point of view, questions remain open about choosing the optimal method for obtaining and scaling the process of isolating peptides from insect biomass, as well as analyzing drug prototypes. In this regard, the search for methods of analysis and control of protein fractions of water-soluble peptides used for the subsequent development of antibacterial drugs based on them is an urgent task. The aim of this work was to study the protein fractions of water-soluble peptides isolated from Musca domestica larvae by dynamic light scattering. Materials and Methods: The results of the study of fractions of water-soluble peptides from Musca domestica larvae by dynamic light scattering are presented. The selection of optimal methods for the analysis and control of prototypes of antibacterial drugs based on antimicrobial peptides will reduce the time of research and ensure the accuracy of the results obtained. Results: It has been found that all the analyzed peptides have a sufficiently high stability in the aqueous medium which is confirmed by values of the zeta potential from −11.2 mV to −12 mV. The peptides at a concentration of 666 µg/ml with a molecular weight of less than 3.5 kDa, it has been found that their size was in the range of 68–142 nm; with a molecular weight of 3.5–7 kDa – 43–68 nm; with a molecular weight of 7–14 kDa – 43–105 nm; with a molecular weight of more than 14 kDa – 79–190 nm. The use of the dynamic light scattering method for the control and analysis of protein fractions of water-soluble peptides is established. The use of this method will reduce the time of analysis, identify micro-impurities, ensure simplicity of execution and almost complete absence of consumables. Conclusion: Use of dynamic light scattering is justified as a fast method of analyzing the obtained fractions of peptides, establishing the absence of trace impurities. Taking into account the almost complete absence of consumables, reduced analysis time and ease of execution in comparison with high-performance liquid chromatography, this detection method can be successfully used in everyday practice.

Abstract Background The Illumina sequencing systems demonstrate high efficiency and power and remain the most popular platforms. Platforms with similar throughput and quality profiles but lower costs are under intensive development. In this study, we compared two platforms Illumina NextSeq 2000 and GeneMind Genolab M for 10x Genomics Visium spatial transcriptomics. Results The performed comparison demonstrates that GeneMind Genolab M sequencing platform produces highly consistent with Illumina NextSeq 2000 sequencing results. Both platforms have similar performance in terms of sequencing quality and detection of UMI, spatial barcode, and probe sequence. Raw read mapping and following read counting produced highly comparable results that is confirmed by quality control metrics and strong correlation between expression profiles in the same tissue spots. Downstream analysis including dimension reduction and clustering demonstrated similar results, and differential gene expression analysis predominantly detected the same genes for both platforms. Conclusions GeneMind Genolab M instrument is similar to Illumina sequencing efficacy and is suitable for 10x Genomics Visium spatial transcriptomics.

Unregulated intersections, according to E.A. Yablokov, are artistic rapprochement of writers not connected to each other biographically. At first glance, the oeuvres of Chekhov and Sholokhov form such an unregulated intersection. However, it is seemingly unregulated, seemingly uncrossed. The article highlights the commonality of writers, who belonged to one ethno-cultural space. The author indicates the consilience of functions of the image of steppe in Chekhov’s short-stories and Sholokhov’s epic novel The Silent Don (Tikhy Don), the similarities and differences when it comes to the writers focus on the Cossacks and related issues. Thus, the comparison of the writers may be both within their typological commonality and genetic succession.
