Abstract Studies on the compounds of aromatic oils and their effects on psychophysiological changes in humans are often conducted separately. To obtain better validation, a suitable protocol is needed that can be extrapolated to large-scale olfactory stimulation experiments. Unfortunately, this type of study is still rarely performed. In this situation, we propose a randomized crossover pilot study on olfactory stimulation with aromatic oils in relation to changes in psychophysiological activity by focusing on white musk aromatic oil due to its popularity in the community. Chemical profiling by TDU-GC-MS (thermal desorption gas chromatography/mass spectrometry) was performed to understand the compounds of the aromatic oils presented. To understand the changes in the participants’ impressions and mood states, POMS 2 (Profile of Mood States 2nd Edition) and VAS (Visual analogue scale) were performed in addition to physiological evaluation by using EEG (electroencephalogram), ECG (electrocardiogram) and salivary amylase measurements. The proposed pilot study showed “gorgeous”, “sweet”, and “like” impression toward white musk aromatic oil under VAS evaluation. Mood evaluation under POMS 2 variables such as Fatigue-Inertia (FI), Tension-anxiety (TA) and TMD (total mood disturbance) were significantly decreased under white musk aromatic oil inhalation. Under current protocol, we can also see the changes in autonomic activity and brain activity during olfactory stimulation. This pilot study could be the first step towards a larger sample size experiment on olfactory stimulation. This experiment has been registered to UMIN Clinical Trials Registry with register ID : UMIN000051972 on 24/08/2023.

Abstract Lipidomics and metabolomics communities comprise various informatics tools; however, software programs handling multimodal mass spectrometry (MS) data with structural annotations guided by the Lipidomics Standards Initiative are limited. Here, we provide MS-DIAL 5 for in-depth lipidome structural elucidation through electron-activated dissociation (EAD)-based tandem MS and determining their molecular localization through MS imaging (MSI) data using a species/tissue-specific lipidome database containing the predicted collision-cross section values. With the optimized EAD settings using 14 eV kinetic energy, the program correctly delineated lipid structures for 96.4% of authentic standards, among which 78.0% had the sn-, OH-, and/or C = C positions correctly assigned at concentrations exceeding 1 μM. We showcased our workflow by annotating the sn- and double-bond positions of eye-specific phosphatidylcholines containing very-long-chain polyunsaturated fatty acids (VLC-PUFAs), characterized as PC n-3-VLC-PUFA/FA. Using MSI data from the eye and n-3-VLC-PUFA-supplemented HeLa cells, we identified glycerol 3-phosphate acyltransferase as an enzyme candidate responsible for incorporating n-3 VLC-PUFAs into the sn1 position of phospholipids in mammalian cells, which was confirmed using EAD-MS/MS and recombinant proteins in a cell-free system. Therefore, the MS-DIAL 5 environment, combined with optimized MS data acquisition methods, facilitates a better understanding of lipid structures and their localization, offering insights into lipid biology.

Abstract Sleep plays an important role in human health. Changes in the sleeping environment can affect sleep quality. The present study investigated the effects of natural wood on sleep quality. The interior material was a fully used Cryptomeria japonica wood board (Japanese cedar room), while the particleboard and medium-density fiberboard (MDF) were covered by a vinyl cloth with a woodgrain (resin room). In order to assess sleep quality, the sleep–depth index (SDI) is calculated based on the lengths of sleep stage 3 and the rapid eye movement (REM) stage using electroencephalography (EEG). Before the sleep experiments, the Pittsburgh Sleep Quality Inventory (PSQI) and Morningness–Eveningness Questionnaire (MEQ) were administered to confirm the participants’ usual sleep habits. Thirteen participants were invited to sleep in two different environments. The MEQ before sleep experiments confirmed that eight participants were of the intermediate type and five participants were of the moderate evening type. According to the PSQI, three participants were poor sleepers and ten were good sleepers. The sleep EEG demonstrated that 12 of 13 participants had higher SDI in the Japanese cedarwood room than in the resin room. A Wilcoxon signed-rank test revealed that SDI was significantly prolonged in the Japanese cedarwood room than in the resin room, regardless of the small sample size (n = 13, z = 2.48, p = 0.014, effect size = 0.78). The present study demonstrated that the indoor environment with C. japonica induced a significantly higher SDI compared to that with wood-grain vinyl cloth. This indicates that the interior materials of C. Japonica wood can positively influence the sleep environment.

In the face of increasingly serious environmental pollution, the health of human lung tissues is also facing serious threats. Mogroside ⅡE (M2E) is the main metabolite of sweetening agents mogrosides from the anti-tussive Chinese herbal Siraitia grosvenori. The study elucidated the anti-inflammatory action and molecular mechanism of M2E against acute lung injury (ALI). A lipopolysaccharide (LPS)-induced ALI model was established in mice and MH-S cells were employed to explore the protective mechanism of M2E through the western blotting, co-immunoprecipitation, and quantitative real time-PCR analysis. The results indicated that M2E alleviated LPS-induced lung injury through restraining the activation of secreted phospholipase A2 type IIA (Pla2g2a)-epidermal growth factor receptor (EGFR). The interaction of Pla2g2a and EGFR was identified by co-immunoprecipitation. In addition, M2E protected ALI induced with LPS against inflammatory and damage which were significantly dependent upon the downregulation of AKT and mTOR via the inhibition of Pla2g2a-EGFR. Pla2g2a may represent a potential target for M2E in the improvement of LPS-induced lung injury, which may represent a promising strategy to treat ALI.

Abstract Loquat (Eriobotrya japonica) leaves contain many bioactive components such as ursolic acid (UA) and amygdalin. We investigated the effects of loquat leaf powder and methanol extract in human neuroglioma H4 cells stably expressing the Swedish-type APP695 (APPNL-H4 cells) and C57BL/6 J mice. Surprisingly, the extract greatly enhanced cellular amyloid-beta peptide (Aβ) 42 productions in APPNL-H4 cells. Administration of leaf powder increased Aβ42 levels after 3 months and decreased levels after 12 months compared to control mice. Leaf powder had no effect on working memory after 3 months, but improved working memory after 12 months. Administration of UA decreased Aβ42 and P-tau levels and improved working memory after 12 months, similar to the administration of leave powder for 12 months. Amygdalin enhanced cellular Aβ42 production in APPNL-H4 cells, which was the same as the extract. Three-month administration of amygdalin increased Aβ42 levels slightly but did not significantly increase them, which is similar to the trend observed with the administration of leaf powder for 3 months. UA was likely the main compound contained in loquat leaves responsible for the decrease in intracerebral Aβ42 and P-tau levels. Also, amygdalin might be one of the compounds responsible for the transiently increased intracerebral Aβ42 levels.

Eriobotrya japonica (E. japonica) leaves have been used as an herbal traditional medicine in China and Japan owing to their anti-inflammatory and protective effects against skin conditions and allergy symptoms. These beneficial effects are likely mediated by the various triterpenoids present in E. japonica leaves. However, the efficacy of E. japonica leaves in the treatment of allergic rhinitis has not been evaluated in humans. Therefore, in the present study, a randomized, controlled, double-blind trial was performed on healthy adults of age >20 (n = 27) who were randomly assigned to receive either 2.5 g of placebo or E. japonica leaf supplements once daily for 4 weeks. The Japanese Allergic Rhinitis Quality of Life Standard Questionnaire (JRQLQ), dermatological allergy symptoms, Dermatology Life Quality Index, and skin condition parameters were assessed at baseline and after 4 weeks. Significant differences were observed in the variability of the itchy nose, itchy eyes, and eye symptoms between the E. japonica supplementation and placebo groups after 4 weeks. Arm skin transepidermal water loss was improved only in the E. japonica supplementation group. This study suggests that E. japonica leaves can be used as a functional food ingredient to relieve allergic symptoms.

Numerous health hazards have been connected to advanced glycation end products (AGEs). In this investigation, using reaction models including BSA-fructose, BSA- methylglyoxal (MGO), and BSA-glyoxal (GO), we examined the anti-glycation potential of eight different berry species on AGEs formation. Our results indicate that black chokeberry (Aronia melanocarpa) exhibited the highest inhibitory effects, with IC50 values of 0.35 ± 0.02, 0.45 ± 0.03, and 0.48 ± 0.11 mg/mL, respectively. Furthermore, our findings suggest that black chokeberry inhibits AGE formation by binding to BSA, which alleviates the conformation alteration, prevents protein cross-linking, and traps reactive α-dicarbonyls to form adducts. Notably, three major polyphenols, including cyanidin-3-O-galactoside, cyanidin-3-O-arabinoside, and procyanidin B2 from black chokeberry, showed remarkably inhibitory effect on MGO/GO capture, and new adducts formation was verified through LC-MS/MS analysis. In summary, our research provides a theoretical basis for the use of berries, particularly black chokeberry, as natural functional food components with potential anti-glycation effects.

Abstract Odor environments in living spaces can influence human physiological and psychological states. To elucidate the effect of volatile organic compounds (VOCs) of wood in living spaces on the modulations of cognitive processing, we built two experimental huts to simulate wooden construction living spaces. One hut was made of cedarwood (Cryptomeria japonica), and the other was made of resin. We used cedarwood and resin, because they are often used as construction materials in Japan. In both huts, we measured the participants’ first positivity in the early visual cortex (P1), the third positivity elicited by voluntary attention (P3b), and the automatic occipital negativity to infrequent visual stimuli in event-related potentials (ERPs), while they performed a visual discrimination task. The VOCs in both huts were measured by gas chromatography–mass spectrometry. The concentration of volatile sesquiterpenes was significantly increased in the cedarwood hut. Neither P1 nor P3b was affected by the cedarwood hut. Compared to the resin hut, we observed significantly larger occipital negativities to the infrequent stimuli in the cedarwood hut in the time windows of 280–300 ms at the mid-occipital region. These findings suggest that the increased concentration of volatile sesquiterpenes emitted from cedarwood facilitates the human brain’s response to changes in visual stimuli.

Abstract We examined the association of serum s-adenosylmethionine (SAM), s-adenosylhomocysteine (SAH) (methionine metabolites), and their ratio on the risk of dementia and death in a community-dwelling population of older Japanese individuals. 1371 residents of Hisayama, Japan, aged 65 years or older and without dementia, were followed for a median of 10.2 years (2007–2017). We divided serum SAM, SAH, and SAM/SAH ratio into quartiles. Cox proportional hazards models were used to estimate the hazard ratios (HRs) and their 95% confidence intervals (CIs) of serum SAM, SAH, and SAM/SAH ratio levels on the risk of a composite outcome of all-cause dementia or death, and each outcome. During the follow-up, 635 participants developed all-cause dementia and/or died, of which 379 participants developed dementia and 394 deaths occurred. The multivariable-adjusted HRs of the composite outcome decreased significantly with increasing serum SAM levels (P for trend = 0.01), while they increased significantly with higher serum SAH levels (P for trend = 0.03). Higher serum SAM/SAH ratio levels were significantly associated with a lower risk of the composite outcome (P for trend = 0.002), as well as with lower risk of each outcome. Our findings suggest that the balance of methionine metabolites may closely associate with the risk of dementia and death.

Hericium erinaceus, a mushroom species commonly known as Yamabushitake in Japan, is known to have a stimulatory effect on neurotrophic factors, such as brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF). Hericenone C, a meroterpenoid with palmitic acid as the fatty acid side chain, is reported to be one such stimulant. However, according to the structure of the compound, the fatty acid side chain seems highly susceptible to lipase decomposition, under in vivo metabolic conditions. To study this phenomenon, hericenone C from the ethanol extract of the fruiting body was subjected to lipase enzyme treatment and observed for changes in the chemical structure. The compound formed after the lipase enzyme digestion was isolated and identified using LC-QTOF-MS combined with 1H-NMR analysis. It was found to be a derivative of hericenone C without its fatty acid side chain and was named deacylhericenone. Interestingly, a comparative investigation of the neuroprotective properties of hericenone C and deacylhericenone showed that the BDNF mRNA expression in human astrocytoma cells (1321N1) and the protection against H2O2-induced oxidative stress was considerably higher in the case of deacylhericenone. These findings suggest that the stronger bioactive form of the hericenone C compound is in fact deacylhericenone.

Multiple sclerosis (MS) is a demyelinating disease of the central nervous system, characterized by apoptotic death of mature oligodendrocytes, neuroinflammation, and motor dysfunction. A pentacyclic triterpenoid compound, ursolic acid (UA), has various pharmacological activities, such as anti-inflammatory, anti-oxidative, and anti-apoptotic effects. In the present study, we investigated the effects of UA on cuprizone-induced demyelination, which is a model of MS. Oral administration of UA effectively suppressed cuprizone-induced demyelination and motor dysfunction via the enhancement of IGF-1 levels in the demyelinating lesions. Our results suggest that UA might be therapeutically useful for demyelination in MS.

Abstract Japanese cedar (Cryptomeria japonica) wood is widely used as a traditional construction material in Japan. The relationship between an individual’s perceived comfort level and a preference for Japanese cedar wood interiors is of interest. We compared volunteers’ physiological responses and subjective evaluations of wooden dwelling spaces with different wood materials: planed Japanese cedar lumber, or printed grain resin sheet overlay boards. Eighty-three subjects were asked to stay in each room for 30 min. We evaluated salivary stress markers, blood pressure, the profile of mood states-brief form (POMS), and a questionnaire that used the semantic differential method to evaluate the subjects’ feeling state for both rooms. The concentrations of the volatile organic compounds in both rooms were also quantified after the experiment. The results demonstrated that the subjects’ evaluation of each room was highly dependent on their preference; each room was evaluated more positively by subjects who preferred it. Although the subjects’ feelings were also influenced by their preference, the room with Japanese cedar did not elicit negative feelings, even from the subjects who disliked it. The subjects’ physiological responses were totally independent of their preferences. Their blood pressure decreased in the Japanese cedar room, and their salivary alpha-amylase activity was repressed in both rooms. These results indicated that the subjective evaluations were influenced in part by the subjects’ preferences, while their physiological responses were not affected. Regardless of which room the subjects preferred, the Japanese cedar room reduced the subjects’ blood pressure compared to the room with artificial materials.

Abstract Introduction Placenta extract is used as an ingredient in ointments for treating dermatological diseases, skin dryness, and for skin beautification. However, the clinical effects of the equine placenta on humans and the underlying mechanism of action are unclear. This randomized, controlled, double‐blind study aimed to clinically evaluate the effect of oral intake of equine placental extract on human skin quality. Methods Healthy women volunteers between the ages of 30 and 59 years (n = 29) were randomly assigned to receive 220 mg of equine placental extract–placebo orally, once daily for 4 weeks. Skin quality parameters such as skin hydration, skin barrier function (transepidermal water loss [TEWL]), and melanin index were assessed at baseline and after 4 weeks of administration. Results The melanin index was significantly increased in the placebo group, whereas it remained unchanged in the equine placenta group. The pattern of melanin index change was significantly different due to intake or no intake of equine placenta supplements over 4 weeks. No significant difference was found in skin hydration and TEWL between the two groups at 4 weeks of postadministration. It was shown that the intake of the equine placenta was more effective in protecting the skin condition against the change of ultraviolet (UV) sensitively than the change in temperature and humidity. Conclusions Effect of equine placental extract intake was evident on the cheek skin of the equine placenta group where participants were protected from UV‐induced pigmentation. Equine placental extract is useful for decreasing melanin synthesis and melanin content in the human skin and can be used as an effective food supplement to maintain human skin quality.

Nutritional foods are concentrated sources of molecules with a nutritional or physiological effect which contain nutrients. There is a category, “nutricosmetics”, defined as ingestible natural health products that enhance the function and appearance of human skin, nails, and hair. A new variety of Flammulina velutipes (F. velutipes), Oki-Shirayuki 919, was explored to ascertain whether its components have functions of nutricosmetics. We focused on physiological effects for enhancing the human skin condition, such as moisturizing or barrier functions in F. velutipes. A randomized, double-blind, placebo-controlled clinical study was performed between January and March 2022. Among healthy men and women (n = 30) aged 20 to 59 years, the test group (n = 15) took a test product which included F. velutipes dry powder, and the placebo group (n = 15) took a placebo (a similar product in which the F. velutipes dry powder was replaced with plum fruit paste). Since the amount of increase in skin hydration over four weeks in the test group was significantly larger than that in the placebo group, a significant difference between the two groups was observed (p = 0.033). F. velutipe was suggested to have some physiological functions such as improving skin moisture.

This study targets the evaluation of melanin synthesis inhibition activity of the bamboo shoot skin as agro-waste. The total methanolic extract of bamboo peel extract was evaluated for its skin protective effects via measuring its melanin inhibitory activity and its suppression activity on the expression of tyrosinase mRNA levels. Results showed that bamboo peel extract has a good ability for the inhibition of melanin synthesis so further studies were performed for the isolation of its constituents. Twelve compounds have been isolated from the shoot skin of Phyllostachys pubescens. Their structures were elucidated based on extensive spectroscopic methods. The melanin inhibition potential of the isolates was tested with their collagen-production-promoting activity for the determination of active principles. Results showed that Betulinic acid, tachioside, and 1,2-dilinolenin significantly suppressed melanin production per cell compared to control. Triacontanol, tricin, and (+)-lyoniresinol 9′-O-glucoside also tended to decrease melanin production per cell. These findings indicated that the skin of bamboo shoots, a significant agricultural waste, is a useful natural source for further research on its potential for aging problems such hyperpigmentation and cognitive function impairment.

We report the isolation and anti-allergic activity (degranulation reduction) of six polyphenolic compounds, one of them being novel, from olive mill wastes (OMW). Additionally, we evaluated their ability to reduce; intracellular Ca2+ levels and expression of calcium channel proteins in RBL-2H3 cells, to assist the possible characterization of the mechanisms involved. The novel compound, new hemialdehydic decarboxymethylated oleuropein aglycone, HDOA 1 (IC50 = 108.3 µM), hydroxytyrosol acetate 3 (IC50 = 40.2 µM) and 1-acetoxypinoresinol 4 (IC50 = 43.8 µM) reduced intracellular Ca2+ levels after challenging by both A23187 and DNP-BSA, suggesting a mechanism beyond just blockage of extracellular Ca2+ influx. While luteolin 2, despite its strongest activity (IC50 = 3.9 µM), reduced Ca2+ levels only after challenging by A23187, suggesting its main mechanism is by blocking extracellular Ca2+ influx. We confirmed further, that, each compound decreased the expression of, two or more, calcium channel proteins suggesting that they act as ‘mast cell stabilizers’ by reducing the expression of calcium channel proteins. This work attracts more attention to OMW as a potential source of lead compounds for structural modification to improve and develop new anti-allergic drugs.

Chemical investigation of the total extract of the Egyptian soft coral Heteroxenia fuscescens, led to the isolation of eight compounds, including two new metabolites, sesquiterpene fusceterpene A (1) and a sterol fuscesterol A (4), along with six known compounds. The structures of 1–8 were elucidated via intensive studies of their 1D, 2D-NMR, and HR-MS analyses, as well as a comparison of their spectral data with those mentioned in the literature. Subsequent comprehensive in-silico-based investigations against almost all viral proteins, including those of the new variants, e.g., Omicron, revealed the most probable target for these isolated compounds, which was found to be Mpro. Additionally, the dynamic modes of interaction of the putatively active compounds were highlighted, depending on 50-ns-long MDS. In conclusion, the structural information provided in the current investigation highlights the antiviral potential of H. fuscescens metabolites with 3β,5α,6β-trihydroxy steroids with different nuclei against SARS-CoV-2, including newly widespread variants.

Hibiscus sabdariffa L. (HS) has a long history of edible and medicinal uses. In this study, the biological activities of the extracts, chromatographic fractions, and hibiscus acid obtained from HS were evaluated for their potential bioactivities. Their ability to promote extracellular matrix synthesis in skin fibroblasts was evaluated by enzyme-linked immunosorbent assays. Their anti-inflammatory activity was evaluated in a nitric oxide (NO)–Griess inflammatory experiment. Furthermore, hibiscus acid was found to have a strong anti-oxidative stress effect through the establishment of an oxidative stress model induced by hydrogen peroxide. Several assays indicated that hibiscus acid treatment can effectively reduce extracellular adenosine triphosphate (ATP) secretion and carbonyl protein production, as well as maintain a high level of reduced/oxidized glutathione (GSH/GSSG) in skin cells, thus providing a possible mechanism by which hibiscus acid can counter antioxidative stress. The present study is the first to explore the reversing skin aging potential and the contributory component of HS.

Averrhoa carambola L. is reported for its anti-obese and anti-diabetic activities. The present study aimed to investigate its aqueous methanol leaf extract (CLL) in vivo anti-obese activity along with the isolation and identification of bioactive compounds and their in vitro α-glucosidase inhibition assessment. CLL improved all obesity complications and exhibited significant activity in an obese rat model. Fourteen compounds, including four flavone glycosides (1–4) and ten dihydrochalcone glycosides (5–12), were isolated and identified using spectroscopic techniques. New compounds identified in planta included (1) apigenin 6-C-(2-deoxy-β-D-galactopyranoside)-7-O-β-D-quinovopyranoside, (8) phloretin 3′-C-(2-O-(E)-cinnamoyl-3-O-β-D-fucopyranosyl-4-O-acetyl)-β-D-fucopyranosyl-6′-O-β-D fucopyranosyl-(1/2)-α-L arabinofuranoside, (11a) phloretin3′-C-(2-O-(E)-p-coumaroyl-3-O-β-D-fucosyl-4-O-acetyl)-β-D-fucosyl-6′-O-(2-O-β-D-fucosyl)-α-L-arabinofuranoside, (11b) phloretin3′-C-(2-O-(Z)-p-coumaroyl-3-O-β-D-fucosyl-4-O-acetyl)-β-D-fucosyl-6′-O-(2-O-β-D-fucosyl)-α-L-arabinofuranoside. Carambolaside M (5), carambolaside Ia (6), carambolaside J (7), carambolaside I (9), carambolaside P (10a), carambolaside O (10b), and carambolaside Q (12), which are reported for the first time from A. carambola L. leaves, whereas luteolin 6-C-α-L-rhamnopyranosyl-(1-2)-β-D-fucopyranoside (2), apigenin 6-C-β-D-galactopyranoside (3), and apigenin 6-C-α-L-rhamnopyranosyl-(1-2)-β-L-fucopyranoside (4) are isolated for the first time from Family. Oxalidaceae. In vitro α-glucosidase inhibitory activity revealed the potential efficacy of flavone glycosides, viz., 1, 2, 3, and 4 as antidiabetic agents. In contrast, dihydrochalcone glycosides (5–11) showed weak activity, except for compound 12, which showed relatively strong activity.

Pulmonary fibrosis is a chronic and aggresitive interstitial lung disease. We evaluated the anti-fibrotic effect of mogrol isolated from Siratia grosvenorii against pulmonary fibrosis and the underlying mechanism. In vitro, mogrol (10 µM) show anti-fibrotic effects by suppressing TGF-β1-induced the epithelial mesenchymal transition of alveolar epithelial cells and myofibroblasts trans-differentiation. Then, mogrol was given after intratracheal instillation of bleomycin in mice. Results showed that mogrol treatments (10 mg/kg/d) improved characteristic changes in histopathology, inflammation infiltration and collagen deposition in lung tissues, and that mogrol significantly inhibited TGF-β1-Smad2/3 pathway, restored NOX4 abnormal expression but promoted AMPK phosphorylation. Further research identified that co-treatment with AMPK inhibitor, significantly blocked the anti-fibrotic properties of mogrol, suggesting that the protective effect of mogrol may be attributed to activated AMPK with consequent reduction in inflammatory, fibrosis and oxidative stress. Overall, Siratia grosvenorii effectively protects against lung fibrosis via AMPK activation and amelioration of TGF-β1 signalling pathway.

Lotus root (Nelumbo nucifera) is a popular food in Japan and southeast Asia. It is reported that lotus root includes dietary fiber, vitamin C, and abundant polyphenolic compounds. In Chinese medicine, lotus powder is also utilized in anti-tussive or anti-inflammatory drugs. In this study, we focused on the effects of lotus root powder on allergy symptoms in mice. First, we assessed nasal allergy symptoms in 2,4-diisocyanic acid toluene (TDI)-sensitized nasal allergy model mice. We found that lotus root powder alleviated nasal allergy symptoms in the mice. In addition, we estimated serum inflammatory parameters such as serum total IgE, histamine, and leukotriene B4. Lotus root powder intake significantly decreased serum total IgE and leukotriene B4 compared with standard food intake. We suggest that lotus root powder intake may alleviate allergic symptoms.

Essentially, flavonoids have been found to be among the most promising molecules identified as coronavirus inhibitors. Recently, a new SARS-CoV-2 B.1.1.529 variant has spread in many countries, which has raised awareness of the role of natural constituents in attempts to contribute to therapeutic protocols. (2) Methods: Using various chromatographic techniques, triterpenes (1–7), phenolics (8–11), and flavonoids (12–17) were isolated from Euphorbia dendroides and computationally screened against the receptor-binding domain (RBD) of the SARS-CoV-2 Omicron variant. As a first step, molecular docking calculations were performed for all investigated compounds. Promising compounds were subjected to molecular dynamics simulations (MD) for 200 ns, in addition to molecular mechanics Poisson–Boltzmann surface area calculations (MM/PBSA) to determine binding energy. (3) Results: MM/PBSA binding energy calculations showed that compound 14 (quercetin-3-O-β-D-glucuronopyranoside) and compound 15 (quercetin-3-O-glucuronide 6″-O-methyl ester) exhibited strong inhibition of Omicron, with ΔGbinding of −41.0 and −32.4 kcal/mol, respectively. Finally, drug likeness evaluations based on Lipinski’s rule of five also showed that the discovered compounds exhibited good oral bioavailability. (4) Conclusions: It is foreseeable that these results provide a novel intellectual contribution in light of the decreasing prevalence of SARS-CoV-2 B.1.1.529 and could be a good addition to the therapeutic protocol.

Syzygium polycephalum (Kupa) is a plant of the Myrtaceae family which is one of the endemic plants in Indonesia, commonly called as Gowok. The chemical components of the plant have not been reported so far. This study is intended to know the molecular structures of isolated compounds of chloroform fraction from S. polycephalum stem bark.The steam bark of the plant is dried, powdered and macerated with methanol to yield methanolic extract. The methanolic extract was then conducted to fractionation using hexane and chloroform to obtain hexane and chloroform fractions. The chloroform fraction was further subjected to separation using column chromatography to obtain pure isolates and followed by measuring of their spectroscopic evidences. The isolation of chloroform fraction had led to the findings of two pure isolates. Their structures of isolates were elucidated by extensive spectroscopic methods and by comparison with the literature data to gain two phenolic compounds that are gallic acid and 3,4,3’-tri-O-methylellagic acid.

The extract from Entada phaseoloides was employed as active ingredients of natural origin into cosmetic products, while the components analysis was barely reported. Using LC-DAD-MS/qTOF analysis, eleven compounds (1–11) were proposed or identified from acetone extract of E. phaseoloides leaves (AE). Among them, six phenolic compounds, protocatechuic acid (2), 4-hydroxybenzoic acid (3), luteolin-7-O-β-d-glucoside (5), cirsimaritin (6), dihydrokaempferol (9), and apigenin (10), were isolated by various chromatographic techniques. Protocatechuic acid (2), epicatechin (4), and kaempferol (11) at a concentration 100 μM increased the HaCaT cells viability of the UVB-irradiated cell without any cytotoxicity effect and reduced the expression of COX-2 and iNOS inflammation gene. Moreover, compounds 2 and 4 could have potent effects on cell migration during wound closure. These results suggest that compounds 2, 4, and 11 from AE have anti-photoaging properties and could be employed in pharmaceutical and cosmeceutical products.

Deep eutectic solvents (DESs) have recently attracted attention as a promising green alternative to conventional hazardous solvents by virtue of their simple preparation, low cost, and biodegradability. Even though the application of DESs in analytical chemistry is still in its early stages, the number of publications on this topic is growing. Analytical procedures applying dispersive liquid–liquid microextraction based on the solidification of floating organic droplets (DLLME-SFOD) are among the more appealing approaches where DESs have been found to be applicable. Herein, we provide a summary of the articles that are concerned with the application of DESs in the DLLME-SFOD of target analytes from diverse samples to provide up-to-date knowledge in this area. In addition, the major variables influencing enrichment efficiency and the microextraction mechanism are fully investigated and explained. Finally, the challenges and future perspectives of applying DESs in DLLME-SFOD are thoroughly discussed and are critically analyzed.

Chromone glycosides comprise an important group of secondary metabolites. They are widely distributed in plants and, to a lesser extent, in fungi and bacteria. Significant biological activities, including antiviral, anti-inflammatory, antitumor, antimicrobial, etc., have been discovered for chromone glycosides, suggesting their potential as drug leads. This review compiles 192 naturally occurring chromone glycosides along with their sources, classification, biological activities, and spectroscopic features. Detailed biosynthetic pathways and chemotaxonomic studies are also described. Extensive spectroscopic features for this class of compounds have been thoroughly discussed, and detailed 13C-NMR data of compounds 1–192, have been added, except for those that have no reported 13C-NMR data.

The investigation of the Syzygium polycephalum Miq. (Myrtaceae) aimed to assess the phytochemical contents and antioxidant activity of the chloroform fraction. In this study, the fraction was obtained from methanol extract of S. polycephalum stem bark partitionated by chloroform. An ellagic acid derivative was successively isolated from the chloroform fraction. The molecular structure of isolated compound was elucidated and established as 3,4,3’-tri-O-methylellagic acid through extensive spectroscopic studies including UV-Vis, FTIR, NMR and LC-MS analyses and by comparison with literature data. The finding of the isolated compound is the first time from the plant, although the isolated compound previously have been found in the other Syzygium species such as S. cumini together with ellagic acid and 3,3’-di-O-methylellagic acid. The chloroform fraction, isolated compound, and vitamin C showed antioxidant activity against 2,2’-diphenyl-1-picrylhydrazyl (DPPH) with IC50 value of 163.6, 72.1, and 11.5 μg/mL, respectively.

The consumption of mushroom Ganoderma lingzhi (G. lingzhi) is believed to help people fight influenza. However, few scientific studies, particularly in animals, has addressed that. Here, a hot-water extract of G. lingzhi was orally or intranasally administered to mice infected with influenza A virus, following by a 21-day post-infection observation. An in vitro neuraminidase (NA) assay, with four subtypes of NA, was used to assess inhibition. The total content of triterpenoids and carbohydrates in the extract were determined with colorimetric assays. Results showed that intranasal administration of the extract reduced the severe weight loss in infected mice by 55.1%. Oral administration of the extract did not significantly interfere the virus infection. In addition, the extract strongly inhibited NA from influenza virus H1N1 and H5N1. These findings suggest that short-term oral consumption of hot-water extract of G. lingzhi has limited anti-influenza function, which also inform further research for its activity observed.

Antioxidant activities of Syzygium polyanthum Wight leaves extracts (methanol, ethyl acetate, dichloromethane, and hexane) were evaluated by using DPPH (2,2-diphenyl-2-picrylhydrazyl hydrate) and ABTS (2,2’-azinobis (3-ethylbenzothiazole-6-sulfonic acid) methods. The methanol extract showed the highest antioxidant activity of DPPH assay among extracts with IC50 value of 55.2 mg/ml. In addition, methanol extract also showed the highest antioxidant activity of ABTS assay among extracts with IC50 value of 17.41 mg/ml. It indicated that methanol extract of S. polyanthum Wight leaves potential for antioxidant.

Plant derived compounds are a source of long term research focus due to their applications in a variety of fields, particularly food preservation. One key way in which phytochemicals are crucial in this area is by disrupting enzyme functionality. In this work, essential oil was extracted by steam distillation from the fresh leaves of Polygonum odoratum (Polygonaceae), commonly known as Vietnamese coriander, and shown to effectively inhibit the oxidation of L-3,4-dihydroxyphenylalanine (L-DOPA) catalyzed by mushroom tyrosinase (EC1.14.18.1). Using GC-MS analysis, twenty five compounds were identified in the essential oil. The most abundant compounds in the essential oil were Alkanals - dodecanal (55.49%), and decanal (11.57%) - followed by anisaldehyde (6.35%); these compounds were individually investigated for inhibitory activity by performing single-compound screening. Each of the top three most abundant compounds inhibited the tyrosinase-catalyzed oxidation of L-DOPA, as identified by UV-VIS spectroscopy and oxygen consumption assays. The inhibitory activity of the major compounds increased when pre-incubated with tyrosinase and without significant additional oxygen consumption, suggesting kcat-type inactivation is not involved. Interactions of the head and tail components of the major alkanals may disrupt the tertiary structure of the enzyme, presenting a potential inhibitory mechanism.

Moringa oleifera have been evaluated for its antioxidant activity. M. oleifera leaves were extracted with methanol, ethyl acetate, dichloromethane and n-hexane. The antioxidant activity of extracts were evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity assay and an improved 2,2’-azino-bis-[3-ethylbenzothiazoline sulphonate] (ABTS) radical cation decolorization assay in vitro. Trolox was used as standard with IC50 5.89 μg/mL in DPPH assay and 3.06 μg/mL in ABTS assay. The methanol extract showed the highest free radical scavenging activity with IC50 value of 49.30 μg/mL in DPPH assay and 11.73 μg/mL in ABTS assay. This study provided that M. oleifera leaves possess antioxidant.

A phenolic acid had been isolated from chloroform soluble fractions of a methanol extract of stem bark of Syzygium litorale, Fam. Myrtaceae. The structure of the isolated compound was elucidated and established as gallic acid through extensive spectroscopic studies (UV-Vis, FTIR, and NMR) and by comparison with literature data and authentic sample. This is the first report of the isolation of compound from this plant, although it has previously been found in Myrtaceae family such as S. aromaticum, S. cumini, S. polyanthum, S. cordatum, etc. The chloroform fraction, isolated compound, and vitamin C showed very strong antioxidant activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) with IC50 value of 23.2, 7.5, and 12.5 mg/mL, respectively.

Angelica shikokiana is widely marketed in Japan as a dietary food supplement. With a focus on neurodegenerative conditions such as Alzheimer’s disease, the aerial part was extracted and through bio-guided fractionation, fifteen compounds [α-glutinol, β-amyrin, kaempferol, luteolin, quercetin, kaempferol-3-O-glucoside, kaempferol-3-O-rutinoside, methyl chlorogenate, chlorogenic acid, hyuganin E, 5-(hydroxymethyl)-2-furaldehyde, β-sitosterol-3-O-glucoside, adenosine (isolated for the first time from A. shikokiana), isoepoxypteryxin and isopteryxin] were isolated. Isolated compounds were evaluated for in vitro neuroprotection using acetylcholine esterase inhibitory, protection against hydrogen peroxide and amyloid β peptide (Aβ25-35)-induced neurotoxicity in neuro-2A cells, scavenging of hydroxyl radicals and intracellular reactive oxygen species and thioflavin T assays. Quercetin showed the strongest AChE inhibition (IC50 value = 35.5 µM) through binding to His-440 and Tyr-70 residues at the catalytic and anionic sites of acetylcholine esterase, respectively. Chlorogenic acid, its methyl ester, quercetin and luteolin could significantly protect neuro-2A cells against H2O2-induced neurotoxicity and scavenge hydroxyl radical and intracellular reactive oxygen species. Kaempferol-3-O-rutinoiside, hyuganin E and isoepoxypteryxin significantly decreased Aβ25-35-induced neurotoxicity and Th-T fluorescence. To the best of our knowledge, this is the first report about neuroprotection of hyuganin E and isoepoxypteryxin against Aβ25-35-induced neurotoxicity.

The in vitro antioxidant, anti-obesity, anti-allergy and skin-lightening effects of extracts from strawberry fruits (ripe and unripe) and nine other plant parts of Amaou strawberry (Fragaria × ananassa var. Amaou) were comparatively investigated by using the antioxidant assay and inhibition assays of lipase, adipocyte differentiation, β-hexosaminidase release, melanogenesis and tyrosinase. Their total phenolic content and the main phenolic compounds were also determined. The ripe strawberry fruits were found to possess potential anti-obesity and skin-lightening effects. The ethanol extracts from unripe strawberry fruits and several other parts like the crown, stolon leaf and flower showed more strong activities in the bioactivity assays conducted. The phytochemical profile varied among extracts from various parts of the plant. Ellagic acid was found in every part of the strawberry plant except the root. Our study provides valuable evidence that strawberry fruit is a food with potential anti-obesity and skin-lightening functions. The findings also support that several inedible parts of the strawberry plant have great potential to be used as the ingredients of functional food products.

Reishi (Ganoderma lingzhi) has been used as a traditional medicine for millennia. However, relatively little is known about this mushroom’s proteins and their bioactivities. In this study, we used reishi’s own proteases to hydrolyze its protein and obtained auto-digested reishi (ADR) extract. The extract was subjected to in vitro assays and administered to spontaneous hypertensive rats (SHRs) to determine its potential for use as a hypotensive medication. Bioassay-guided fractionation and de novo sequencing were used for identifying the active compounds. After 4 h administration of ADR, the systolic pressure of SHRs significantly decreased to 34.3 mmHg (19.5% change) and the effect was maintained up to 8 h of administration, with the decrease reaching as low as 26.8 mmHg (15% reduction–compare to base line a decrease of 26.8 mmHg is less than a decrease of 34.3 mmHg so it should give a smaller % reduction). Eleven peptides were identified and four of them showed potent inhibition against ACE with IC50 values ranging from 73.1 μM to 162.7 μM. The results showed that ADR could be a good source of hypotensive peptides that could be used for antihypertensive medication or incorporation into functional foods.

Besides being a useful building material, bamboo also is a potential source of bioactive substances. Although some studies have been performed to examine its use in terms of the biological activity, only certain parts of bamboo, especially the leaves or shoots, have been studied. Comprehensive and comparative studies among different parts of bamboo would contribute to a better understanding and application of this knowledge. In this study, the biological activities of ethanol and water extracts from the leaves, branches, outer culm, inner culm, knots, rhizomes and roots of Phyllostachys pubescens, the major species of bamboo in Japan, were comparatively evaluated. The phytochemical profiles of these extracts were tentatively determined by liquid chromatography-mass spectrometry (LC-MS) analysis. The results showed that extracts from different parts of bamboo had different chemical compositions and different antioxidative, antibacterial and antiallergic activities, as well as on on melanin biosynthesis. Outer culm and inner culm were found to be the most important sources of active compounds. 8-C-Glucosylapigenin, luteolin derivatives and chlorogenic acid were the most probable compounds responsible for the anti-allergy activity of these bamboo extracts. Our study suggests the potential use of bamboo as a functional ingredient in cosmetics or other health-related products.

Lingzhi is a Ganoderma mushroom species which has a wide range of bioactivities. Analysis of the changes in metabolites during the developmental stages of lingzhi is important to understand the underlying mechanism of its biosynthesis, as well as its bioactivity. It may also provide valuable information for the cultivation efficiency of lingzhi. In this study, mass spectrometry based untargeted metabolomics was carried out to analyze the alteration of metabolites during developmental stages of lingzhi. Eight developmental stages were categorized on the basis of morphological changes; starting from mycelium stage to post-mature stage. GC/MS and LC/MS analyses along with multivariate analysis of lingzhi developmental stages were performed. Amino acids, organic acids, sugars, polyols, fatty acids, fatty alcohols, and some small polar metabolites were extracted as marker metabolites from GC/MS analysis, while, lanostane-type triterpenoids were observed in LC/MS analysis of lingzhi. The marker metabolites from untargeted analysis of lingzhi developmental stages were correlated with the α-glucosidase inhibitory activity. Two metabolites, compounds 34 and 35, were identified as potential contributors of the α-glucosidase inhibitory activity. The current result shows that some metabolites are involved in the developmental process and α-glucosidase inhibitory activity of lingzhi.

Phytochemical examination of corncob extracts led to the isolation of a new lignan identified as 7,7'-dihydroxy-3'-O-demethyl-4-methoxymatairesinol, together with seven known compounds, identified as β-sitosterol, β-sitosteryl-β-D-glucoside, 6β-hydroxy-campest-4-en-3-one, 5α,8α-epidioxyergosta-6,22-dien-3β-ol, tricin, kaempferol and p-coumaric acid. The isolated compounds were identified by one and two-dimensional NMR spectroscopies and mass spectrometry.

An ester derivative of 4-methoxycynnamic acid, i.e. 2-ethylhexyl 4-methoxy cynnamate was isolated for the first time from the chloroform extract of stem bark of Xylocarpus moluccencis (Lamk) M. Roem) (Meliaceae) along with β-sitosterol and stigmasterol. The first structure was elucidated with the help of various spectroscopic techniques, including IR, GC-MS, and NMR spectra. Two last structures were determined by comparison with the reported compounds in literature. These compounds were also found in the hexane extract of the plant.

Two known alkylated flavonoid namely matteucinol dan matteucinol-7-O-b-D-glucoside was isolated for the first time from the fern Chingia sakayensis (Zeiller) Holtt's stem. Their structures were elucidated on the basis of spectroscopic evidence and by comparation with those reported data in literature. A long chain alcohol, 1-eicosanol together with two sterols, β-sitosterol and stigmasterol had been isolated from hexane extract of stem bark of pacar cina (Aglaia odorata Lour) (Meliaceae). Regeneration of planted forests of Cryptomeria japonica (sugi) and Chamaecyparis obtuse (hinoki) is the pressing importance to the forest administration in Japan. Low seed germination rate of these species, however, has hampered low-cost production of their seedlings for reforestation. The primary cause of the low germinability has been attributed to highly frequent formation of anatomically unsound seeds, which are indistinguishable from sound germinable seeds by visible observation and other common criteria such as size and weight. To establish a method for sound seed selection in these species, hyperspectral imaging technique was used to identify a wavelength range where reflectance spectra differ clearly between sound and unsound seeds. In sound seeds of both species, reflectance in a narrow waveband centered at 1,730 nm, corresponding to a lipid absorption band in the short-wavelength infrared (SWIR) range, was greatly depressed relative to that in adjacent wavebands on either side. Such depression was absent or less prominent in unsound seeds. Based on these observations, a reflectance index SQI, abbreviated for seed quality index, was formulated using reflectance at three narrow SWIR wavebands so that it represents the extent of the depression. SQI calculated from seed area-averaged reflectance spectra and spatial distribution patterns of pixelwise SQI within each seed area were both proven as reliable criteria for sound seed selection. Enrichment of sound seeds was accompanied by an increase in germination rate of the seed lot. Thus, the methods described are readily applicable toward low-cost seedling production in combination with single seed sowing technology.

An oleane-type triterpenoid, briononic acid was isolated from hexane extract of the stem bark of Sandoricum koetjape Merr. (Meliaceae). This structure had been established based on spectroscopic data (UV, IR, and NMR) and by comparison with spectroscopic data of related compound that had been reported.
