Two new species of the family Aoridae, one from the genus Aoroides Walker, 1898, and other from the genus Grandidierella Coutière, 1904, are reported from Korean waters. Aoroides gracilicrus sp. nov. is similar to A. longimerus in having numerous plumose setae on the basis and carpus of gnathopod 1. However, the new species can be distinguished from A. longimerus by the numerous plumose setae on the bases of pereopods 3 and 4 and the slender basis of pereopod 7. Grandidierella naroensis sp. nov. is morphologically most similar to G. pseudosakaensis. However, the new species can be distinguished by the presence of small distal and proximal processes and a large middle process on the carpus of gnathopod 1, and the subovate propodus of gnathopod 1. Both new species are illustrated and compared to related species. A key to species in the family Aoridae from Korean waters is also provided.

Abstract Dietary inclusion effects of various levels of jack mackerel meal (JMM) on growth, feed utilization, biochemical composition, and innate immune response of rockfish were determined. The control (Con) diet included 550 g/kg fish meal (FM). Ten, 30, 50, 100, 200, 400, 600, and 1000 g/kg of JMM were included in the Con diet, referred to as the JMM10, JMM30, JMM50, JMM100, JMM200, JMM400, JMM600, and JMM1000 diets, respectively. Eight hundred and ten fish (initial mean weight of 2.3 g) were distributed into 27 flow‐through tanks. The experimental diets were assigned to triplicate groups of fish. Fish were hand‐fed for 8 weeks. Weight gain of fish fed the JMM400 diet was statistically better than that of fish fed the Con, JMM10, JMM30, JMM50, JMM100, and JMM200 diets. Feed consumption of fish fed the JMM400 diet was statistically higher than that of fish fed the Con, JMM10, JMM30, and JMM50 diets. Dietary inclusion of JMM did not influence feed utilization, biochemical composition, and innate immune responses of fish. In conclusion, inclusion of JMM up to 400 g/kg of FM in diet improved growth and feed consumption of rockfish, and dietary optimum inclusion levels of JMM were estimated to be 396.7 and 403.1 g/kg based on regression analysis of weight gain and feed consumption, respectively.

Incorporating feed stimulants and attractants in low fish meal (FM) diets is a very effective way to improve palatability and increase feed intake and fish growth. This research aims to elucidate the substitution impact of different levels of FM with various plant proteins in diets with jack mackerel meal (JMM) inclusion as feed stimulants on the growth and feed utilization of rockfish (Sebastes schlegeli). A two-way (2 substitution levels [25% and 50%] × 3 substitution sources [corn gluten meal (CGM), soybean protein concentrate (SPC), and corn protein concentrate (CPC)]) ANOVA experimental design was adopted. In total, 525 fish averaging 8.3 g were assigned to 21 tanks (25 juvenile/tank). Seven isoprotetic and isolipidic feeds were formulated. The control (Con) diet contained 55% FM. CGM, SPC, and CPC were replaced for 25% and 50% FM in the Con diet, and then 22% JMM as feed stimulants was included at the cost of FM, named as the CGM25, CGM50, SPC25, SPC50, CPC25, and CPC50 diets, respectively. All diets were assigned to triplicate groups of rockfish. Rockfish were hand-fed to apparent satiation twice a day for 8 weeks. Rockfish fed the 25% FM substitution diets achieved superior (p < 0.0001 for both) weight gain and specific growth rate (SGR) compared to rockfish fed the 50% FM substitution diets, while rockfish fed the CGM-replaced diets achieved superior (p < 0.0001 for both) weight gain and SGR compared to rockfish fed the SPC- and CPC-replaced diets. The weight gain and SGR of rockfish fed the Con and CGM25 diets were superior to rockfish fed all other diets. Rockfish fed the 25% FM substitution diets achieved superior (p < 0.0001 for both) feed efficiency (FE) and protein efficiency ratio (PER) compared to rockfish fed the 50% FM substitution diets, while rockfish fed the CGM-replaced diets achieved superior FE and PER compared to rockfish fed the CPC-replaced diets. Rockfish fed the 25% FM substitution diets led to superior (p < 0.0001) protein retention (PR) compared to rockfish fed the 50% FM substitution diets. The biological indices, proximate composition, and amino acid profiles of the whole body, as well as the plasma and serum parameters of rockfish, were not changed by either substitution level or substitution source. In conclusion, the replacement of CGM for 25% FM in the rockfish diet with JMM inclusion can be made without deteriorating growth performance, feed consumption and utilization, biochemical composition, and plasma and serum parameters.

Abstract Reference materials are critical in assay development for calibrating and assessing their suitability. The devasting nature of the COVID-19 pandemic and subsequent proliferation of vaccine platforms and technologies has meant that there is even a greater need for standards for immunoassay development, which are critical to assess and compare vaccines’ responses. Equally important are the standards needed to control the vaccine manufacturing processes. Standardized vaccine characterization assays throughout process development are essential for a successful Chemistry, Manufacturing and Controls (CMC) strategy. In this perspective paper, we advocate for reference material incorporation into assays and their calibration to International Standards from preclinical vaccine development through control testing and provide insight into why this is necessary. We also provide information on the availability of WHO international antibody standards for CEPI-priority pathogens.

The production of phytylated chlorophyll a (Chl aP) in Rhodobacter sphaeroides, which uses phytylated bacteriochlorophyll a (BChl aP), is the first step in expanding the light absorption spectra. Unlike the chlorophyll synthase (ChlG) of the Synechocystis sp. PCC6803, ChlGs of angiosperms, including Arabidopsis thaliana, Nicotiana tabacum, Avena sativa, and Oryza sativa, showed bacteriochlorophyll synthase activity and resistance to inhibition by bacteriochlorophyllide a (BChlide a), geranylgeranylated BChl a (BChl aGG), and BChl aP, collectively called bacteriochlorins. Among the angiosperm ChlGs, N. tabacum ChlG had the highest bacteriochlorophyll synthase activity and resistance to inhibition by bacteriochlorins. Expression of N. tabacum chlG in R. sphaeroides resulted in the formation of free Chl aP in the presence of BChl aP during photoheterotrophic growth, even though reactive oxygen species were generated.

Fish meal (FM) usage in aquafeed is fraught with cost and environmental challenges. Alternatives to FM must be readily available and with adequate protein. This 8-weeks study explored the utilization of tuna by-product meal (TBM) without fermentation as a substitute for FM in diet on growth, feed utilization, chemical composition, plasma chemistry, and non-immune responses of olive flounder (Paralichthys olivaceus). Four hundred juveniles (initial weight of 14.7 g) were randomly distributed to 20 tanks (20 fish per tank). The control (Con) diet contained 65% FM without replacement, and in the TBM10, TBM20, TBM30, TBM40, and TBM50 diets, FM was substituted with 10%, 20%, 30%, 40%, and 50% TBM, respectively. Each diet was fed to triplicate groups of fish, but quadruplicate groups for the Con and TBM30 diets. Survival was not different among the experimental diets. Superior weight gain, specific growth rate (SGR), feed efficiency ratio (FER) and protein efficiency ratio were observed in fish fed the TBM40 and TBM50 diets to fish fed all other diets. Condition factor of fish fed the TBM40 and TBM50 diets was higher than that of fish fed the Con diet. Feed consumption, chemical composition, and amino acid profiles of the whole body fish, except for the tryptophan content, viscerosomatic and hepatosomatic indices, plasma biochemical parameters, non-specific immune responses were not affected by the experimental diets. In conclusion, FM up to 50% can be replaced with TBM without impairing growth, feed utilization, nutritional composition, plasma chemistry, and non-specific immune responses of olive flounder when 65% FM was included. The greatest weight gain, SGR, and FER of fish were estimated to be 47.1%, 48.6%, and 46.3% of dietary substitution of FM with TBM, respectively.

Abstract The jaguar (Panthera onca) is the dominant predator in Central and South America, but is now considered near‐threatened. Estimating jaguar population size is difficult, due to uncertainty in the underlying dynamical processes as well as highly variable and sparse data. We develop a stochastic temporal model of jaguar abundance in the Peruvian Amazon, taking into account prey availability, under various climate change scenarios. The model is calibrated against existing data sets and an elicitation study in Pacaya Samiria. In order to account for uncertainty and variability, we construct a population of models over four key parameters, namely three scaling parameters for aquatic, small land, and large land animals and a hunting index. We then use this population of models to construct probabilistic evaluations of jaguar populations under various climate change scenarios characterized by increasingly severe flood and drought events and discuss the implications on jaguar numbers. Results imply that jaguar populations exhibit some robustness to extreme drought and flood, but that repeated exposure to these events over short periods can result in rapid decline. However, jaguar numbers could return to stability—albeit at lower numbers—if there are periods of benign climate patterns and other relevant factors are conducive.

2-Oxoglutarate:ferredoxin oxidoreductase from Chlorobaculum tepidum (CtOGOR) is a carbon-fixing enzyme in the reductive TCA cycle that reversibly carboxylates succinyl-CoA to yield 2-oxoglutarate. CtOGOR is a heterotetramer of two large (α = 68 kDa) and two small (β = 38 kDa) subunits. The αβ protomer harbors one thiamine pyrophosphate and two 4Fe-4S clusters. Nonetheless, the enzyme has a considerable oxygen tolerance with a half-life of 143 min at 215 μM dissolved oxygen. Kinetic analyses of the purified recombinant CtOGOR revealed a lower Km for succinyl-CoA than for 2-oxoglutarate. Cellular levels of 2-oxoglutarate and glutamate—a product of glutamine oxoglutarate aminotransferase and glutamate dehydrogenase—increased more than twofold in the exponential phase compared with the control strain, leading to an approximately >30% increase in the photoautotrophic growth rate. Thus, CtOGOR was successfully produced in Synechocystis, thereby boosting carboxylation, resulting in enhanced photoautotrophic growth.

OBJECTIVES As HIV/AIDS is becoming a chronic disease, the risk of developing cardiovascular disease (CVD) among people living with HIV/AIDS is rising. Anxiety and depression, which are common among people living with HIV/AIDS, have been linked with CVD. This study investigated the risk of CVD in people living with HIV/AIDS and explored the effects of depression and anxiety on CVD risk. METHODS Data were collected for 457 people enrolled in the Korea Cohort HIV/AIDS study after 2010. Framingham risk scores were calculated to quantify the 10-year risk of developing CVD. Depression and anxiety variables were re-coded as a single combined variable. Multivariable logistic regression analysis was performed, adjusting for age, body mass index, low-density lipoprotein (LDL) cholesterol, triglycerides (TG), duration of human immunodeficiency virus (HIV) positivity after entry into the cohort, and depression/anxiety. RESULTS Participants with both depression and anxiety were 2.28 times more likely than those with neither depression nor anxiety to have moderate/high-risk CVD risk. The 10-year risk of developing CVD was affected by LDL cholesterol, TG, age, and duration of HIV infection. LDL cholesterol and TG levels change according to the duration of HIV infection, and metabolic disorders affect the risk of CVD. Thus, a longer duration of HIV infection is associated with a higher risk of developing CVD. CONCLUSIONS Screenings for depression and anxiety need to be provided regularly to assess the severity of those symptoms. To help decrease their risk of developing CVD, people living with HIV/AIDS should be offered behavioral modification interventions aimed at developing healthy lifestyle habits.

Background COPD patients account for a large proportion of lung transplants; lung transplantation survival benefit for COPD patients is not well established. Methods We identified 4521 COPD patients in the United Network for Organ Sharing (UNOS) dataset transplanted from May 2005 to August 2016, and 604 patients assigned to receive pulmonary rehabilitation and medical management in the National Emphysema Treatment Trial (NETT). After trimming the populations for NETT eligibility criteria and data completeness, 1337 UNOS and 596 NETT patients remained. Kaplan–Meier estimates of transplant-free survival from transplantation for UNOS, and NETT randomisation, were compared between propensity score-matched UNOS (n=401) and NETT (n=262) patients. Results In propensity-matched analyses, transplanted patients had better survival compared to medically managed patients in NETT (p=0.003). Abstract Inclusion effect of soybean meal (SBM) and fermented SBM (FSM) in extruded pellet for juvenile abalone (Haliotis discus) was compared in abalone farm. Dietary inclusion effect of the combined macroalgae (MA) (Undaria pinnatifida and Hizikia fusiforme) and a single Saccharina japonica on abalone was also compared. Three thousand six hundred juvenile abalone were purchased from a private hatchery and acclimated to the experimental conditions for 2 weeks. Six 5-ton flow-through raceway tanks were used, and abalone were randomly distributed into tanks (n = 600 per tank). Three experimental diets were prepared in duplicate. Fish meal, FSM, corn gluten meal, and shrimp meal and wheat flour and dextrin were used as the protein and carbohydrate sources, respectively, in the FSM diet. MA was also included in the FSM diet. FSM and MA in the FSM diet were substituted with SBM at the expense of wheat flour and S. japonica, referred to as the SBM and SJ diets. The experimental diets were pelletized by an extruded pelleter. Water stability of nutrients in the experimental diets was monitored at 12, 24, and 48 h after seawater immersion. The experimental diets were fed to abalone once a day to satiation with a little leftover for 120 days. The retained crude protein and lipid and ash content of the extruded pellets were changed over all period of time. Weight gain and specific growth rate (SGR) of abalone fed the SBM diet were greater than those of abalone fed the FSM and SJ diets. Weight gain and SGR of abalone fed the SJ diet were also greater than those of abalone fed the FSM diet. The longest shell length, widest shell width, highest shell height, and greatest soft body weight were obtained in abalone fed the SBM diet, followed by the SJ and FSM diets. Proximates of the soft body of abalone were not different among the experimental diets. In conclusion, SBM was a superior protein source to FSM in extruded pellet for growth performance of abalone. Dietary inclusion of a single S. japonica was superior to the combined inclusion of U. pinnatifida and H. fusiforme in the production of abalone.

Abstract Substitution effect of white radish’ by-product (WRB) and tunic of sea squirt (TSS) for Undaria in feed on growth and carcass composition of abalone was determined. Eight-hundred forty juveniles were distributed into 12 containers. Three formulated diets were prepared in triplicate. A 15% Undaria was included in the control diet. The 15% Undaria was substituted with a same amount of WRB and TSS, referred to as the DRG and TSS diets, respectively. Finally, dry Undaria was also prepared to compare effect of the formulated diets on performance of abalone. The experimental diets were fed to abalone once a day at a satiation level with a little leftover. The feeding trial lasted for 16 weeks. Crude protein, crude lipid, and ash content of the experimental diets changed over all period of time. Higher crude protein and lipid and ash contents retained in all formulated diets compared to the Undaria were observed at 24 and 48 h after seawater immersion. Survival of abalone was not affected by the experimental diets. Weight gain and specific growth rate (SGR) of abalone fed the TSS diet was greater than all other diets. Weight gain and SGR of abalone fed the control diet was not different from those of abalone fed the WRB diet. Proximates of the soft body of abalone was affected by the diets. In conclusion, WRB and TSS are the promising feed ingredients to replace Undaria in abalone feed; especially, TSS is superior to either Undaria or WRB. The formulated diets produced improved growth performance of abalone over the single Undaria.

PurposeChloramphenicol and tetracycline are not recommended for treating scrub typhus in pediatric patients because of potential side effects, such as aplastic anemia or tooth discoloration. While clarithromycin has recently been used in adults, few reports have been published on its effects in pediatric patients. We report the clinical profiles of pediatric scrub typhus and the effects of clarithromycin on scrub typhus in children.MethodsWe retrospectively analyzed medical records of 56 children with scrub typhus who were admitted between 2004 and 2013 to Chonbuk National University Hospital, Jeonju, Korea. Cases were divided into 3 groups based on thetreatment drug (chloramphenicol, azithromycin, and clarithromycin). We compared their clinical manifestations and laboratory findings.ResultsAll patients exhibited fever and rash. Other common clinical manifestations were eschars (66%), lymphadenopathy (48%), upper respiratory symptoms (42%), abdominal pain (32%), and hepatosplenomegaly (14%). Elevated levels of C-reactive protein, erythrocyte sedimentation rates, aspartate transaminase, and alanine transaminase were detected in 95%, 96%, 84%, and 77% of patients, respectively. Additionally, decreased platelet and white blood cell levels were observed in 43% and 36% of patients, respectively. There were no statistical differences between the treatment groups in mean age (P=0.114) or sex (P=0.507). However, time to defervescence after the treatments differed significantly, being the shortest in the clarithromycin group (P=0.019). All patients recovered without complications related to the disease or drugs.ConclusionClarithromycin was as effective as chloramphenicol and azithromycin in pediatric scrub typhus patients and may be used as a first-line treatment drug.

Dietary inclusion effect of yacon (YC), ginger (GG) and blueberry (BB) on the growth, feed utilization, body composition and plasma chemistry of olive flounder and immunity test against Streptococcus iniae compared to a commercial probiotic (Lactobacillus fermentum) was investigated. Six hundred fish were randomly distributed into 15, 50 L flow-through tanks (40 fish per tank). Five experimental diets were prepared in triplicates. No additives were added in the control (CON) diet. The commercially available 0.5% L. fermentum and 1% each of YC, GG and BB powder were included instead of an equal amount of water and wheat flour, referred to as the LF, YCP, GGP and BBP diets, respectively. Fish were daily fed with one of the experimental diets for 8 weeks. At the end of the 4 and 8-week feeding trials, fish were artificially infected with S. iniae and the cumulative mortality was monitored for the next 8 days. No differences in growth performance, feed utilization, body composition and plasma chemistry were observed at the end of the 8-week feeding trial. However, the highest cumulative mortality was observed in fish fed the CON diet starting at 42 h until the end of 8-day post observation after the both 4 and 8-week feeding trials. The cumulative mortality of fish fed the YCP, GGP and BBP diets was comparable to the LF diet. Therefore, YC, GG and BB were effective as immunostimulant against S. iniae rather than growth promoter of olive flounder. OBJECTIVES To manage evidence-based diseases, it is important to identify the characteristics of patients in each country. METHODS The Korea HIV/AIDS Cohort Study seeks to identify the epidemiological characteristics of 1,442 Korean individuals with human immunodeficiency virus (HIV) infection (12% of Korean individuals with HIV infection in 2017) who visited 21 university hospitals nationwide. The descriptive statistics were presented using the Korea HIV/AIDS cohort data (2006-2016). RESULTS Men accounted for 93.3% of the total number of respondents, and approximately 55.8% of respondents reported having an acute infection symptom. According to the transmission route, infection caused by sexual contact accounted for 94.4%, of which 60.4% were caused by sexual contact with the same sex or both males and females. Participants repeatedly answered the survey to decrease depression and anxiety scores. Of the total participants, 89.1% received antiretroviral therapy (ART). In the initial ART, 95.3% of patients were treated based on the recommendation. The median CD4 T-cell count at the time of diagnosis was 229.5 and improved to 331 after the initial ART. Of the patients, 16.6% and 9.4% had tuberculosis and syphilis, respectively, and 26.7% had pneumocystis pneumonia. In the medical history, sexually transmitted infectious diseases showed the highest prevalence, followed by endocrine diseases. The main reasons for termination were loss to follow-up (29.9%) and withdrawal of consent (18.7%). CONCLUSIONS Early diagnosis and ART should be performed at an appropriate time to prevent the development of new infection.

The number of persons infected by HIV/AIDS has consistently increased in Korea since the first case of HIV/AIDS infection in 1985 and reached 15,208 by 2016. About 1,100 new patients with HIV/ AIDS infections have emerged every year since 2013. In Korea, the Korea HIV/AIDS Cohort Study was established for the evidenced-based prevention, treatment, and effective management of patients infected with human immunodeficiency virus (HIV) in December 2006. This study monitored 1,438 patients, who accounted for about 10% of all patients with HIV/AIDS in Korea, for 10 years with the following aims: (1) to develop an administrative system for the establishment of a HIV/AIDS cohort-based study; (2) to standardize methodologies and the case report forms; and (3) to standardize multi-cohort data and develop a data cleaning method. This study aims to monitor at least 1,000 patients (excluding those for whom investigation had been completed) per year (estimated number of patients who can be monitored by January 2018: 939). By December 2016, the sex distribution was 93.3% for men, and 6.7% for women (gender ratio, 13.9:1.0), and 98.9% of all participants were Korean. More than 50.0% of the participants were confirmed as HIV positive after 2006. This study reports competitive, long-term research that aimed to develop policies for the prevention of chronic infectious diseases for patients with HIV. The data collected over the last decade will be used to develop indices for HIV treatment and health promotion.

Helicobacter pylori genetic variation is a crucial component of colonization and persistence within the inhospitable niche of the gastric mucosa. As such, numerous H. pylori genes have been shown to vary in terms of presence and genomic location within this pathogen. Among the variable factors, the Bab family of outer membrane proteins (OMPs) has been shown to differ within subsets of strains. To better understand genetic variation among the bab genes and to determine whether this variation differed among isolates obtained from different geographic locations, we characterized the distribution of the Bab family members in 80 American H. pylori clinical isolates (AH) and 80 South Korean H. pylori clinical isolates (KH). Overall, we identified 23 different bab genotypes (19 in AH and 11 in KH), but only 5 occurred in greater than 5 isolates. Regardless of strain origin, a strain in which locus A and locus B were both occupied by a bab gene was the most common (85%); locus C was only occupied in those isolates that carried bab paralog at locus A and B. While the babA/babB/- genotype predominated in the KH (78.8%), no single genotype could account for greater than 40% in the AH collection. Comprehensive evaluation of cancer screening activities based on individual experiences is urgently needed to address the burden of cancer among Métis people. In this co-designed and co-led study, a cancer screening questionnaire developed for Métis people to evaluate their cancer screening histories and to explore barriers and facilitators to cancer screening was used. Adult Métis Albertans were invited to participate in the anonymous survey through a multi-modal strategy used for community consultations. Descriptive analyses compared responses between regions, age groups and geographic locations. In total, 370 participants who identified as Métis consented and contributed responses between 12 September and 2 December 2022. Female respondents reported higher rates of cervical and breast cancer screening (>94%) and lower rates of colorectal cancer screening (67–78%). Most of the barriers and facilitators were rated as very important, especially access to reliable and accurate information on screening, risks and benefits of cancer screening, explanation of the test results or procedures, trust in their health care provider(s) and health care system and access to a primary health care provider. This study fills a crucial gap that can inform targeted interventions to increase cancer screening awareness and rates among Métis Albertans and reduce their cancer burden.

Abstract This letter presents a wide dynamic range (WDR) feature extraction (FE) readout scheme for machine vision applications using CMOS image sensors (CISs). The proposed scheme with the proposed pixel structure has two operating modes, the normal and WDR modes. In the normal operating mode, the proposed CIS captures a normal image with high sensitivity. In addition, as a unique function, a bi‐level image is obtained for real‐time FE even if a pixel is saturated in strong illumination conditions. Thus, compared to typical CISs for machine vison, the proposed CIS can reveal object features that are blocked by light in real time. In the WDR operating mode, the proposed CIS produces a WDR image with its corresponding bi‐level image. A prototype CIS was fabricated using a standard 0.35‐μm 2P4M CMOS process with a 320 × 240 format (QVGA) with 10‐μm pitch pixels. At 60 fps, the measured power consumption was 5.98 mW at 3.3 V for pixel readout and 2.8 V for readout circuitry. The dynamic range of 73.1 dB was achieved in the WDR operating mode.

Background: We evaluated the accuracy of a sagittal alignment of tibial component in total knee arthroplasty (TKA) with the sagittal reference line using the preoperative radiograph. Methods: We evaluated 151 patients who underwent primary TKA with posterior cruciate substituting type implant. For 75 patients, the sagittal reference line using preoperative radiograph (group A) was used, while for 76 patients the sagittal reference using intraoperative fibular shaft line (group B) was used. The parallel line (line S) to the anatomical axis of the tibia in the lateral plain radiograph was used as the sagittal reference. The distance from line S to proximal tibia cutting area and to skin surface 20 cm distal to the cutting area was measured in preoperative radiographs. Next, the distance to the extramedullary guide rod was applied intraoperatively, reflecting the results. The intraoperative fibular shaft line was determined using the connecting line between the tip of fibular head and the lateral malleolus. The postoperative tibial component slope angle and the difference to the target slope angle were compared. Results: The difference to the target angle was 1.8 ± 1.3° in group A, whereas the difference was 2.5 ± 1.4° in group B ( p = 0.04). The patients with difference within 3° to the target angle were 78.7% in group A and 61.8% in group B ( p = 0.024). Conclusion: The sagittal reference line using the preoperative lateral radiograph was helpful as a reference guide for the tibial slope in TKA.

Based on an analysis of the signal characteristics of gas sensors, this work presents a chemoresistive sensor readout circuit design for detecting gases with slow response time characteristics. The proposed readout circuit directly generates a reference voltage corresponding to the initial value of the gas sensor and extracts only the amount of gas concentration change in the sensor. Because the proposed readout circuit can adaptively regenerate the suitable reference voltage under various changing ambient conditions, it can alleviate the variation in output values at the same gas concentration caused by non-uniformities among gas sensors. Furthermore, this readout circuit effectively eliminates the initial value shifts due to the poor reproducibility of the gas sensor itself without requiring complex digital signal calibrations. This work focuses on a commercially viable readout circuit structure that can effectively obtain slow response gas information without requiring a large capacitor. The proposed readout circuit operation was verified by simulations using spectre in cadence simulation software. It was then implemented on a printed circuit board with discrete components to confirm the effectiveness with existing gas sensor systems and its commercial viability.

Spinal cord stimulation is a therapy to treat the severe neuropathic pain by suppressing the pain signal via electrical stimulation of the spinal cord. The conventional metal packaged and battery-operated implantable pulse generator (IPG) produces electrical pulses to stimulate the spinal cord. Despite its stable operation after implantation, the implantation site is limited due to its bulky size and heavy weight. Wireless communications including wireless power charging is also restricted, which is mainly attributed to the electromagnetic shielding of the metal package. To overcome these limitations, here, we developed a fully implantable miniaturized spinal cord stimulator based on a biocompatible liquid crystal polymer (LCP). The fabrication of electrode arrays in the LCP substrate and monolithically encapsulating the circuitries using LCP packaging reduces the weight (0.4 g) and the size (the width, length, and thickness are 25.3, 9.3, and 1.9 mm, respectively). An inductive link was utilized to wirelessly transfer the power and the data to implanted circuitries to generate the stimulus pulse. Prior to implantation of the device, operation of the pulse generator was evaluated, and characteristics of stimulation electrode such as an electrochemical impedance spectroscopy (EIS) were measured. The LCP-based spinal cord stimulator was implanted into the spared nerve injury rat model. The degree of pain suppression upon spinal cord stimulation was assessed via the Von Frey test where the mechanical stimulation threshold was evaluated by monitoring the paw withdrawal responses. With no spinal cord stimulation, the mechanical stimulation threshold was observed as 1.47 ± 0.623 g, whereas the stimulation threshold was increased to 12.7 ± 4.00 g after spinal cord stimulation, confirming the efficacy of pain suppression via electrical stimulation of the spinal cord. This LCP-based spinal cord stimulator opens new avenues for the development of a miniaturized but still effective spinal cord stimulator.

Abstract Background A hypoxic-preconditioned secretome from stem cells reportedly promotes the functional and regenerative capacity of the liver more effectively than a control secretome. However, the optimum oxygen partial pressure (pO2) in the cell culture system that maximizes the therapeutic potential of the secretome has not yet been determined. Methods We first determined the cellular alterations in adipose tissue-derived stem cells (ASCs) cultured under different pO2 (21%, 10%, 5%, and 1%). Subsequently, partially hepatectomized mice were injected with the secretome of ASCs cultured under different pO2, and then sera and liver specimens were obtained for analyses. Results Of all AML12 cells cultured under different pO2, the AML12 cells cultured under 1% pO2 showed the highest mRNA expression of proliferation-associated markers (IL-6, HGF, and VEGF). In the cell proliferation assay, the AML12 cells cultured with the secretome of 1% pO2 showed the highest cell proliferation, followed by the cells cultured with the secretome of 21%, 10%, and 5% pO2, in that order. When injected into the partially hepatectomized mice, the 1% pO2 secretome most significantly increased the number of Ki67-positive cells, reduced serum levels of proinflammatory mediators (IL-6 and TNF-α), and reduced serum levels of liver transaminases. In addition, analysis of the liver specimens indicated that injection with the 1% pO2 secretome maximized the expression of the intermediate molecules of the PIP3/Akt and IL-6/STAT3 signaling pathways, all of which are known to promote liver regeneration. Conclusions The data of this study suggest that the secretome of ASCs cultured under 1% pO2 has the highest liver reparative and regenerative potential of all the secretomes tested here.

Purpose: This study compared the results of a culture method using sonication with those yielded by the conventional culture method, for patients with infected total knee arthroplasty (TKA). We also evaluated the usefulness of sonication for the identification of pathogens in infected TKA cases. Methods: Isolates were cultured from 13 implants that had been removed from 13 patients with infected TKA. Preoperative culture was performed on aspirated joint fluid, and during the operation, infected tissue was collected for culture. The removed prosthetic implants were cultured before and after sonication. Next, we identified the cultured bacteria using API biochemical kits and 16 S rRNA sequencing. Results: The cultures from preoperative joint fluid and intraoperative tissue were positive in 9 of 13 cases (69.2%). For the removed implants, 10 cases were positive before sonication. After sonication, 12 cases (92.3%) had positive cultures. The pathogen most commonly isolated from the cultures was Staphylococcus aureus . Conclusions: This study found that a culturing workflow incorporating sonication diagnosed pathogens in patients with infected TKA with higher sensitivity than did the conventional culturing method.

Understanding the barriers to and facilitators of cancer screening programs among Indigenous populations remains limited. In the spirit of mutual respect, this co-led, collaborative project was carried out between the Métis Nation of Alberta and Screening Programs from Alberta Health Services (AHS). This scoping review assessed the cancer screening literature for available questionnaires and then identified themes and suitable questions for a Métis-specific cancer screening questionnaire. Literature searches on cervical, breast, and colorectal cancer screening programs and related concepts were conducted in electronic databases, including the Native Health Database, MEDLINE (Ovid), PsycINFO, PubMed, PubMed Central, CINAHL, MEDLINE (Ebsco), Psychology & Behavioral Sciences Collection, and Web of Science. Grey literature was collected from AHS Insite, Open Archives Initiative repository, American Society of Clinical Oncology, European Society of Medical Oncology, Google, and Google Scholar. 135 articles were screened based on the eligibility criteria with 114 articles selected, including 14 Indigenous-specific ones. Knowledge, attitude, belief, behaviour, barrier, and facilitator themes emerged from the review, but no Métis-specific cancer screening instruments were found. Thus, one was developed using existing cancer screening instruments, with additional questions created by the project team. A survey of the Métis population in Alberta will use this questionnaire and provide data to address the burden of cancer among Métis people.

In this study, a genetic algorithm was used to calculate the scheduled waiting time according to the train operation frequency of heterogeneous trains operating on one track. The acquired data were then used to determine the appropriate subsidiary track at which high-speed trains can load or release cargo away from low-speed trains. A metaheuristic genetic algorithm was applied and implemented using Javascript/jQuery. Six cases were investigated, which provided values of subsidiary track that vary according to the operation frequencies of different types of trains, and solutions were derived through 100 simulations using a stochastic method. The analysis results showed that the train overtaking frequency was the highest at the third intermediate station within the simulation, suggesting that this particular station requires a subsidiary track, even if the operating frequency of each train differs across the entire track considered in this study. The results of this study are expected to facilitate objective and practical planning during railway construction.

Retinal prostheses are implantable devices that aim to restore the vision of blind patients suffering from retinal degeneration, mainly by artificially stimulating the remaining retinal neurons. Some retinal prostheses have successfully reached the stage of clinical trials; however, these devices can only restore vision partially and remain insufficient to enable patients to conduct everyday life independently. The visual acuity of the artificial vision is limited by various factors from both engineering and physiological perspectives. To overcome those issues and further enhance the visual resolution of retinal prostheses, a variety of retinal prosthetic approaches have been proposed, based on optimization of the geometries of electrode arrays and stimulation pulse parameters. Other retinal stimulation modalities such as optics, ultrasound, and magnetics have also been utilized to address the limitations in conventional electrical stimulation. Although none of these approaches have been clinically proven to fully restore the function of a degenerated retina, the extensive efforts made in this field have demonstrated a series of encouraging findings for the next generation of retinal prostheses, and these could potentially enhance the visual acuity of retinal prostheses. In this article, a comprehensive and up-to-date overview of retinal prosthetic strategies is provided, with a specific focus on a quantitative assessment of visual acuity results from various retinal stimulation technologies. The aim is to highlight future directions toward high-resolution retinal prostheses.

In the multiple asynchronous bearing-only (BO) sensors tracking system, there usually exist two main challenges: (1) the presence of clutter measurements and the target misdetection due to imperfect sensing; (2) the out-of-sequence (OOS) arrival of locally transmitted information due to diverse sensor sampling interval or internal processing time or uncertain communication delay. This paper simultaneously addresses the two problems by proposing a novel distributed tracking architecture consisting of the local tracking and central fusion. To get rid of the kinematic state unobservability problem in local tracking for a single BO sensor scenario, we propose a novel local integrated probabilistic data association (LIPDA) method for target measurement state tracking. The proposed approach enables eliminating most of the clutter measurement disturbance with increased target measurement accuracy. In the central tracking, the fusion center uses the proposed distributed IPDA-forward prediction fusion and decorrelation (DIPDA-FPFD) approach to sequentially fuse the OOS information transmitted by each BO sensor. The track management is carried out at local sensor level and also at the fusion center by using the recursively calculated probability of target existence as a track quality measure. The efficiency of the proposed methodology was validated by intensive numerical experiments.

The point detections obtained from radars or sonars in surveillance environments include clutter measurements, as well as target measurements. Target tracking with these data requires data association, which distinguishes the detections from targets and clutter. Various algorithms have been proposed for clutter measurement density estimation to achieve accurate and robust target tracking with the point detections. Among them, the spatial clutter measurement density estimator (SCMDE) computes the sparsity of clutter measurement, which is the reciprocal of the clutter measurement density. The SCMDE considers all adjacent measurements only as clutter, so the estimated clutter measurement density is biased for multi-target tracking applications, which may result in degraded target tracking performance. Through the study in this paper, a major source of tracking performance degradation with the existing SCMDE for multi-target tracking is analyzed, and the use of the clutter measurement probability is proposed as a remedy. It is also found that the expansion of the volume of the hyper-sphere for each sparsity order reduces the bias of clutter measurement density estimates. Based on the analysis, we propose a new adaptive clutter measurement density estimation method called SCMDE for multi-target tracking (MTT-SCMDE). The proposed method is applied to multi-target tracking, and the improvement of multi-target tracking performance is shown by a series of Monte Carlo simulation runs and a real radar data test. The clutter measurement density estimation performance and target tracking performance are also analyzed for various sparsity orders.

This paper introduces a digital-assisted multiple echo detection scheme, which utilizes the waste time of the full serial data readout period in a focal plane array (FPA)-based laser detection and ranging (LADAR) receiver. With the support of an external digital signal processor (DSP) and additional analog memory inserted into the receiver, the proposed readout scheme can effectively enhance multi-target resolution (MTR) three times higher than the conventional FPA-based LADAR, while maintaining low power consumption and a small area. A prototype chip was fabricated in a 0.18-μm CMOS process with an 8 × 8 FPA configuration, where each single receiver pixel occupied an area of 100 μm × 100 μm. The single receiver achieved an MTR of 20 ns with 7.47 mW power dissipation, an input referred noise current of 4.48 pA/√Hz with a bandwidth 530 MHz, a minimum detectable signal (MDS) of 340 nA, a maximum walk error of 2.2 ns, and a maximum non-linearity of 0.05% among the captured multiple echo images.

Microfabrication technology for cortical interfaces has advanced rapidly over the past few decades for electrophysiological studies and neuroprosthetic devices offering the precise recording and stimulation of neural activity in the cortex. While various cortical microelectrode arrays have been extensively and successfully demonstrated in animal and clinical studies, there remains room for further improvement of the probe structure, materials, and fabrication technology, particularly for high-fidelity recording in chronic implantation. A variety of non-conventional probes featuring unique characteristics in their designs, materials and fabrication methods have been proposed to address the limitations of the conventional standard shank-type (“Utah-” or “Michigan-” type) devices. Such non-conventional probes include multi-sided arrays to avoid shielding and increase recording volumes, mesh- or thread-like arrays for minimized glial scarring and immune response, tube-type or cylindrical probes for three-dimensional (3D) recording and multi-modality, folded arrays for high conformability and 3D recording, self-softening or self-deployable probes for minimized tissue damage and extensions of the recording sites beyond gliosis, nanostructured probes to reduce the immune response, and cone-shaped electrodes for promoting tissue ingrowth and long-term recording stability. Herein, the recent progress with reference to the many different types of non-conventional arrays is reviewed while highlighting the challenges to be addressed and the microfabrication techniques necessary to implement such features.

Urothelial carcinomas of the urinary bladder have diverse biological and functional characteristics, and numerous factors are likely to be involved in recurrence, progression, and patient survival. While several molecular markers used to evaluate the development and prognosis of bladder cancer have been studied, they are of limited value; therefore, new molecular parameters useful for predicting the prognosis of bladder cancer patients (particularly patients at high risk of progression and recurrence) are required. Recent progress in the understanding of epigenetic modification and gene silencing has provided new opportunities for the detection, treatment, and prevention of cancer. Methylation is an important molecular mechanism in bladder cancer and may have utility as a prognostic and/or diagnostic marker. This review discusses the epigenetic issues involved in the detection and prediction of bladder cancer.

Purpose To investigate the success rate of trial without catheter (TWOC) for postoperative urinary retention (POUR) after non-urological surgery and to determine predictors of successful TWOC. Methods A total of 104 patients who underwent non-urological surgery and were referred to the department of urology for POUR were included in this retrospective study. All eligible patients underwent indwelling catheterization as an initial treatment and then TWOC was performed 3 to 7 days later. POUR was defined as micturition difficulty with greater than 400 mL of postvoid residual (PVR) urine volume measured by catheterization after non-urological surgery. Successful TWOC was defined as voiding with less than 100 mL of PVR urine volume. Predictive factors were identified by multivariate regression analysis. All definitions corresponded to recommendations of the International Continence Society. Results The mean age of the patients was 65.2 (range, 23 to 92) years. There were 45 male and 59 female patients. Intraoperative indwelling catheterization was performed in 69 (66.3%) patients. Mean duration of indwelling catheterization for POUR was 5.0 (range, 3.0 to 7.0) days and 83 (79.8%) patients received medication with an alpha-blocker. A successful TWOC was observed in 70 (67.4%) patients. The mean age of the patients with failure of TWOC was significantly higher than that of the patients with successful TWOC. The percentages of female patients, spinal surgery, and prone position during surgery in patients with unsuccessful TWOC were higher than in those with successful TWOC. In the multivariate logistic regression analysis, age and location of surgery (spine vs. non-spine) were the independent predictors of successful TWOC for POUR. Conclusions Our data suggest that older age and spinal surgery may be important risk factors for failure of TWOC for POUR after non-urological surgery. Thus, adequate prevention measures may be necessary for POUR after non-urological surgery, especially in patients with these risk factors.

Polyimide has been widely applied to neural prosthetic devices, such as the retinal implants, due to its well-known biocompatibility and ability to be micropatterned. However, planar films of polyimide that are typically employed show a limited ability in reducing the distance between electrodes and targeting cell layers, which limits site resolution for effective multi-channel stimulation. In this paper, we report a newly designed device with a pillar structure that more effectively interfaces with the target. Electrode arrays were successfully fabricated and safely implanted inside the rabbit eye in suprachoroidal space. Optical Coherence Tomography (OCT) showed well-preserved pillar structures of the electrode without damage. Bipolar stimulation was applied through paired sites (6:1) and the neural responses were successfully recorded from several regions in the visual cortex. Electrically evoked cortical potential by the pillar electrode array stimulation were compared to visual evoked potential under full-field light stimulation.

Abstract Background The accurate segmentation of spine muscles plays a crucial role in analyzing musculoskeletal disorders and designing effective rehabilitation strategies. Various imaging techniques such as MRI have been utilized to acquire muscle images, but the segmentation process remains complex and challenging due to the inherent complexity and variability of muscle structures. In this systematic review, we investigate and evaluate methods for automatic segmentation of spinal muscles. Methods Data for this study were obtained from PubMed/MEDLINE databases, employing a search methodology that includes the terms 'Segmentation spine muscle’ within the title, abstract, and keywords to ensure a comprehensive and systematic compilation of relevant studies. Systematic reviews were not included in the study. Results Out of 369 related studies, we focused on 12 specific studies. All studies focused on segmentation of spine muscle use MRI, in this systematic review subjects such as healthy volunteers, back pain patients, ASD patient were included. MRI imaging was performed on devices from several manufacturers, including Siemens, GE. The study included automatic segmentation using AI, segmentation using PDFF, and segmentation using ROI. Conclusion Despite advancements in spine muscle segmentation techniques, challenges still exist. The accuracy and precision of segmentation algorithms need to be improved to accurately delineate the different muscle structures in the spine. Robustness to variations in image quality, artifacts, and patient-specific characteristics is crucial for reliable segmentation results. Additionally, the availability of annotated datasets for training and validation purposes is essential for the development and evaluation of new segmentation algorithms. Future research should focus on addressing these challenges and developing more robust and accurate spine muscle segmentation techniques to enhance clinical assessment and treatment planning for musculoskeletal disorders.

Purpose We compared the surgical outcomes of radical nephrectomy (RN) and partial nephrectomy (PN) in adult patients with clinical T2 stage (cT2) renal cell carcinoma (RCC) by utilizing data from the Korean Renal Cell Carcinoma (KORCC) database. Materials and Methods We retrospectively analyzed adult patients with cT2 RCC from 8 tertiary hospitals who were registered in the KORCC between 2003 and 2023. Patients with a solitary kidney or bilateral tumors were excluded. The patient cohort was divided into RN and PN arms, and propensity score matching (PSM) was conducted with a 1:3 ratio. Perioperative and survival outcomes were compared between arms. Results After PSM, the PN and RN arms included 44 and 132 patients, respectively. No significant differences were observed in baseline characteristics, apart from laterality, following PSM. Validated at the PCB level, the design shows strong commercial potential by addressing power efficiency and signal accuracy challenges. The technology is adaptable for applications in environmental monitoring, industrial safety, and medical diagnostics, where efficient and reliable gas sensing is essential.

Background: Symmetrical peripheral gangrene (SPG) is a destructive clinical condition where amputation is often the final treatment option. We aimed to identify the predictors of SPG using the common data model (CDM) and propose a new scoring system for predicting hospitalized patients at risk of developing SPG. Methods: A cohort of patients treated with intravenous noradrenaline, epinephrine, and vasopressin between 2011 and 2020 was retrospectively analyzed using the CDM database. The main outcome was amputation performed as a resuscitation measure. We investigated risk factors including demographic characteristics, comorbidities, and preoperative laboratory values. Based on demographic variables such as age and sex, a 1:10 propensity score matching (PSM) was performed. The odds ratio (OR) was calculated using logistic regression analysis. Results: Amputation was performed in 308 (0.4%) patients out of a cohort of 73,902 patients. Age, sex, hypertension, diabetes mellitus (DM), renal disease (RD), heart failure, anemia, hypercholesterolemia, peripheral vascular disease (PVD), and laboratory markers such as albumin, eosinophils, hematocrit, lymphocytes, monocytes, neutrophils, ESR, aPTT, creatinine, and BUN were statistically significant. Logistic regression analysis revealed statistically significant differences in DM (OR 5.51), RD (OR 2.90), PVD (OR 9.67), and cerebrovascular disease (CVD) (OR 0.49). Compared to the group without amputation, logistic regression analysis after matching the age and sex group with 1:10 PSM showed statistically significant results in DM (OR 3.59), RD (OR 2.59), PVD (OR 7.76), and CVD (OR 0.40). Conclusion: Early recognition of high-risk patients may help medical providers prevent severe outcomes, including amputation surgery.

This study investigates the complex relationship between upper limb movement direction and macroscopic neural signals in the brain, which is critical for understanding brain-computer interfaces (BCI). Conventional BCI research has primarily focused on a local area, such as the contralateral primary motor cortex (M1), relying on the population-based decoding method with microelectrode arrays. In contrast, macroscopic approaches such as electroencephalography (EEG) and magnetoencephalography (MEG) utilize numerous electrodes to cover broader brain regions. This study probes the potential differences in the mechanisms of microscopic and macroscopic methods. It is important to determine which neural activities effectively predict movements. To investigate this, we analyzed MEG data from nine right-handed participants while performing arm-reaching tasks. We employed dynamic statistical parametric mapping (dSPM) to estimate source activity and built a decoding model composed of long short-term memory (LSTM) and a multilayer perceptron to predict movement trajectories. This model achieved a high correlation coefficient of 0.79 between actual and predicted trajectories. Subsequently, we identified brain regions sensitive to predicting movement direction using the integrated gradients (IG) method, which assesses the predictive contribution of each source activity. The resulting salience map demonstrated a distribution without significant differences across motor-related regions, including M1. Predictions based solely on M1 activity yielded a correlation coefficient of 0.42, nearly half as effective as predictions incorporating all source activities. This suggests that upper limb movements are influenced by various factors such as movement coordination, planning, body and target position recognition, and control, beyond simple muscle activity. All of the activities are needed in the decoding model using macroscopic signals. Our findings also revealed that contralateral and ipsilateral hemispheres contribute equally to movement prediction, implying that BCIs could potentially benefit patients with brain damage in the contralateral hemisphere by utilizing brain signals from the ipsilateral hemisphere. In conclusion, this study demonstrates that macroscopic activity from large brain regions significantly contributes to predicting upper limb movement. Non-invasive BCI systems would require a comprehensive collection of neural signals from multiple brain regions.

Abstract Background Patients with renal cell carcinoma (RCC) have an elevated risk of chronic kidney disease (CKD) following nephrectomy. Therefore, continuous monitoring and subsequent interventions are necessary. It is recommended to evaluate renal function postoperatively. Therefore, a tool to predict CKD onset is essential for postoperative follow-up and management. Methods We constructed a cohort using data from eight tertiary hospitals from the Korean Renal Cell Carcinoma (KORCC) database. A dataset of 4389 patients with RCC was constructed for analysis from the collected data. Nine machine learning (ML) models were used to classify the occurrence and nonoccurrence of CKD after surgery. The final model was selected based on the area under the receiver operating characteristic (AUROC), and the importance of the variables constituting the model was confirmed using the shapley additive explanation (SHAP) value and Kaplan-Meier survival analyses. Results The gradient boost algorithm was the most effective among the various ML models tested. The gradient boost model demonstrated superior performance with an AUROC of 0.826. The SHAP value confirmed that preoperative eGFR, albumin level, and tumor size had a significant impact on the occurrence of CKD after surgery. Conclusions We developed a model to predict CKD onset after surgery in patients with RCC. This predictive model is a quantitative approach to evaluate post-surgical CKD risk in patients with RCC, facilitating improved prognosis through personalized postoperative care.

Abstract Purpose The aim of this study is to evaluate the results of high tibial osteotomy (HTO) in patients with bone marrow edema (BME) and assess the effect of factors on the subsequent results. Methods A total of 138 patients who underwent medial open-wedge HTO using locking plate were included in this study. BME was observed in 108 patients using preoperative magnetic resonance imaging. Clinical results were evaluated before HTO and postoperatively at 12 months. Moreover, we evaluated the factors affecting the clinical results. Results The clinical scores were all significantly improved regardless of the presence of BME. There were no differences in improvement of clinical scores between patients with BME and without BME. Patients with BME showed higher postoperative Western Ontario and McMaster Universities Arthritis Index (WOMAC) stiffness scores than patients without BME. Furthermore, patients with BME on both the femur and tibia showed lower Knee Society function scores than patients with BME on either the femur or the tibia. Patients with large BME lesions exhibited a lower Hospital for Special Surgery score and WOMAC pain scores, postoperatively. In patients with BME, patients with undercorrection showed significantly lower improvement in WOMAC pain scores compared with patients with acceptable correction. Conclusions The clinical improvement after HTO in patients with varus and medial osteoarthritis was not different regardless of the presence or absence of BME. However, accurate alignment should be considered essential for achieving better clinical outcomes in patients with preoperative BME.

The 5-year survival rate of kidney cancer drops dramatically from 93% to 15% when it is metastatic. Metastasis constitutes for 30% of kidney cancer cases, in which clear cell renal cell carcinoma (ccRCC) is the most prominent subtype. By sequencing mRNA of ccRCC patient samples, we found that apolipoprotein L1 (APOL1) was highly expressed in tumors compared to their adjacent normal tissues. This gene has been previously identified in a large body of kidney disease research and was reported as a potential prognosis marker in many types of cancers. However, the molecular function of APOL1 in ccRCC, especially in metastasis, remained unknown. In this study, we modulated the expression of APOL1 in various renal cancer cell lines and analyzed their proliferative, migratory, and invasive properties. Strikingly, APOL1 overexpression suppressed ccRCC metastasis both in vitro and in vivo. We then explored the mechanism by which APOL1 alleviated ccRCC malignant progression by investigating its downstream pathways. APOL1 overexpression diminished the activity of focal adhesive molecules, Akt signaling pathways, and EMT processes. Furthermore, in the upstream, we discovered that miR-30a-3p could inhibit APOL1 expression. In conclusion, our study revealed that APOL1 play a role as a tumor suppressor in ccRCC and inhibit metastasis, which may provide novel potential therapeutic approaches for ccRCC patients.

Abstract Solar ultraviolet B (UVB) radiation triggers excessive inflammation, disrupting the epidermal barrier, and can eventually cause skin cancer. A previous study reported that under UVB irradiation, epidermal keratinocytes synthesize the proopiomelanocortin-derived peptide β-endorphin, which is known for its analgesic effect. However, little is known about the role of β-endorphin in UVB-exposed skin. Therefore, in this study, we aimed to explore the protective role of β-endorphin against UVB irradiation-induced damage to the skin barrier in normal human keratinocytes (NHKs) and on a human skin equivalent model. Treatment with β-endorphin reduced inflammatory responses in UVB-irradiated NHKs by inactivating the NF-κB signaling pathway. Additionally, we found that β-endorphin treatment reversed UVB-induced abnormal epidermal proliferation and differentiation in NHKs and, thus, repaired the skin barrier in UVB-treated skin equivalents. The observed effects of β-endorphin on UVB-irradiated NHKs were mediated via blockade of the Akt/mTOR signaling pathway. These results reveal that β-endorphin might be useful against UVB-induced skin injury, including the disruption of the skin barrier function.

Background: Recently, open pose estimation using artificial intelligence (AI) has enabled the analysis of time series of human movements through digital video inputs. Analyzing a person's actual movement as a digitized image would give objectivity in evaluating a person's physical function. In the present study, we investigated the relationship of AI camera-based open pose estimation with Harris Hip Score (HHS) developed for patient-reported outcome (PRO) of hip joint function. Method: HHS evaluation and pose estimation using AI camera were performed for a total of 56 patients after total hip arthroplasty in Gyeongsang National University Hospital. Joint angles and gait parameters were analyzed by extracting joint points from time-series data of the patient's movements. A total of 65 parameters were from raw data of the lower extremity. Principal component analysis (PCA) was used to find main parameters. K-means cluster, X-squared test, Random forest, and mean decrease Gini (MDG) graph were also applied. Results: The train model showed 75% prediction accuracy and the test model showed 81.8% reality prediction accuracy in Random forest. “Anklerang_max”, “kneeankle_diff”, and “anklerang_rl” showed the top 3 Gini importance score in the Mean Decrease Gini (MDG) graph. Conclusion: The present study shows that pose estimation data using AI camera is related to HHS by presenting associated gait parameters. In addition, our results suggest that ankle angle associated parameters could be key factors of gait analysis in patients who undergo total hip arthroplasty.

BackgroundUltraviolet B (UVB) from sunlight represents a major environmental factor that causes toxic effects resulting in structural and functional cutaneous abnormalities in most living organisms. Although numerous studies have indicated the biological mechanisms linking UVB exposure and cutaneous manifestations, they have typically originated from a single study performed under limited conditions.MethodsWe accessed all publicly accessible expression data of various skin cell types exposed to UVB, including skin biopsies, keratinocytes, and fibroblasts. We performed biological network analysis to identify the molecular mechanisms and identify genetic biomarkers.ResultsWe interpreted the inflammatory response and carcinogenesis as major UVB-induced signaling alternations and identified three candidate biomarkers (IL1B, CCL2, and LIF). Moreover, we confirmed that these three biomarkers contribute to the survival probability of patients with cutaneous melanoma, the most aggressive and lethal form of skin cancer.ConclusionOur findings will aid the understanding of UVB-induced cutaneous toxicity and the accompanying molecular mechanisms. In addition, the three candidate biomarkers that change molecular signals due to UVB exposure of skin might be related to the survival rate of patients with cutaneous melanoma.

Background: The human hair follicle undergoes cyclic phases—anagen, catagen, and telogen—throughout its lifetime. This cyclic transition has been studied as a target for treating hair loss. Recently, correlation between the inhibition of autophagy and acceleration of the catagen phase in human hair follicles was investigated. However, the role of autophagy in human dermal papilla cells (hDPCs), which is involved in the development and growth of hair follicles, is not known. We hypothesized that acceleration of hair catagen phase upon inhibition of autophagy is due to the downregulation of Wnt/β-catenin signaling in hDPCs, and that components of Panax ginseng extract can increase the autophagic flux in hDPCs. Methods: We generated an autophagy-inhibited condition using 3-methyladenine (3-MA), a specific autophagy inhibitor, and investigated the regulation of Wnt/β-catenin signaling using the luciferase reporter assay, qRT-PCR, and western blot analysis. In addition, cells were cotreated with ginsenoside Re and 3-MA and their roles in inhibiting autophagosome formation were investigated. Results: We found that the unstimulated anagen phase dermal papilla region expressed the autophagy marker, LC3. Transcription of Wnt-related genes and nuclear translocation of β-catenin were reduced after treatment of hDPCs with 3-MA. In addition, treatment with the combination of ginsenoside Re and 3-MA changed the Wnt activity and hair cycle by restoring autophagy. Conclusions: Our results suggest that autophagy inhibition in hDPCs accelerates the catagen phase by downregulating Wnt/β-catenin signaling. Furthermore, ginsenoside Re, which increased autophagy in hDPCs, could be useful for reducing hair loss caused by abnormal inhibition of autophagy.

Abstract We developed a novel prediction model for recurrence and survival in patients with localized renal cell carcinoma (RCC) after surgery and a novel statistical method of machine learning (ML) to improve accuracy in predicting outcomes using a large Asian nationwide dataset, updated KOrean Renal Cell Carcinoma (KORCC) database that covered data for a total of 10,068 patients who had received surgery for RCC. After data pre-processing, feature selection was performed with an elastic net. Nine variables for recurrence and 13 variables for survival were extracted from 206 variables. Synthetic minority oversampling technique (SMOTE) was used for the training data set to solve the imbalance problem. We applied the most of existing ML algorithms introduced so far to evaluate the performance. We also performed subgroup analysis according to the histologic type. Diagnostic performances of all prediction models achieved high accuracy (range, 0.77–0.94) and F1-score (range, 0.77–0.97) in all tested metrics. In an external validation set, high accuracy and F1-score were well maintained in both recurrence and survival. In subgroup analysis of both clear and non-clear cell type RCC group, we also found a good prediction performance.
