Abstract Background Heart rate is crucial for patients with septic shock, but there are few studies on the scope of heart rate. Therefore, we studied the relationship between different heart rates and mortality of critically ill patients with septic shock, and explored the optimal heart rate range, in order to provide new insights for clinical treatment of septic shock. Methods This retrospective study utilized time-series heart rate data from the Medical Information Mart for Intensive Care (MIMIC) IV database. Patients with septic shock were identified as the Sepsis 3.0 criteria and received vasopressor therapy in the first 24 h since ICU admission. We calculated the time-weighted average heart rate (TWA-HR) based on the time-series data. The restricted cubic spline (RCS) analysis was employed to investigate the nonlinear relationship between heart rate and 28-day mortality, aiming to explore the optimal heart rate control target for septic patients and using this target as the exposure factor. The primary outcome was 28-day mortality, and the secondary outcome were ICU and in-hospital mortality. For the original cohort, we applied the log-rank test to infer the relationship between heart rate and mortality. To control for bias introduced by confounders, we utilized propensity score matching (PSM) to reduce imbalances between normal TWA-HR and high TWA-HR groups, and we established a series of models [the multivariable Cox model, matching weight (MW)-adjusted Cox model, multivariable logistic regression, MW-adjusted logistic regression, and doubly robust model] as sensitivity analyses and subgroup analyses to demonstrate the robustness of our findings. Results A total of 13492 patients were included in our study. The RCS analysis based on Cox and logistic regression showed increased risk of mortality (P 85 beats per minute (bpm). The log-rank test revealed in terms of the 28-day mortality, the hazard ratio (HR) (95% confidence interval [CI]) was 1.92 (1.78—2.06, P 85 bpm. Conclusion Patients with septic shock whose heart rate was controlled no more than 85 bpm during ICU stay received survival benefit in terms of 28-day, ICU and in-hospital mortality.

Abstract Hepatocellular carcinoma (HCC) is the most common type of primary liver cancer, characterized by difficulties in early diagnosis, prone to distant metastasis, and high recurrence rates following surgery. Extracellular vesicles (EVs) are a class of cell-derived particles, including exosomes, characterized by a phospholipid bilayer. They serve as effective carriers for intercellular communication cargo, including proteins and nucleic acids, and are widely involved in tumor progression. They are being explored as potential tumor biomarkers and novel therapeutic avenues. We provide a brief overview of the biogenesis and characteristics of EVs to better understand their classification standards. The focus of this review is on the research progress of EV-associated proteins in the field of HCC. EV-associated proteins are involved in tumor growth and regulation in HCC, participate in intercellular communication within the tumor microenvironment (TME), and are implicated in events including angiogenesis and epithelial-mesenchymal transition (EMT) during tumor metastasis. In addition, EV-associated proteins show promising diagnostic efficacy for HCC. For the treatment of HCC, they also demonstrate significant potential including enhancing the efficacy of tumor vaccines, and as targeting cargo anchors. Facing current challenges, we propose the future directions of research in this field. Above all, research on EV-associated proteins offers the potential to enhance our comprehension of HCC and offer novel insights for developing new treatment strategies.

Abstract This study aimed to develop and validate prognostic nomograms that can estimate the probability of 1-, 3- and 5-year overall survival (OS) as well as cancer-specific survival (CSS) for Intrahepatic cholangiocarcinoma (ICCA) patients. Clinical data of 1446 patients diagnosed with ICCA between 2010 and 2017 from the Surveillance, Epidemiology, and End Results (SEER) database were analyzed. In both the OS and the CSS group, the training cohort and validation cohort were divided into a 7:3 ratio. Age, sex, AJCC T stage, AJCC N stage, AJCC M stage, surgical status, and tumor grade were selected as independent prognostic risk factors to build the nomograms. To compare the efficacy of predicting 1-, 3-, and 5-year OS and CSS rates of the nomogram with the 8th edition of the American Joint Committee on Cancer (AJCC) staging system, we evaluated the Harrell’s index of concordance (C-index), area under the receiver operating characteristic curve (AUC) and decision curve analysis (DCA) in both cohorts. The results showed the nomogram for 1-, 3-, and 5-year OS and CSS prediction performed better than the AJCC staging system. In the subgroup analysis for patients could not receive surgery as the primary treatment. We developed two nomograms for predicting the 1-, and 2-year OS and CSS rates following the same analysis procedure. Results indicate that the performance of both nomograms, which contained sex, AJCC T stage, AJCC M stage, chemotherapy, and tumor grade and prognostic factors, was also superior to the AJCC staging system. Meanwhile, four dynamic network-based nomograms were published. Abstract Background As the complexity and diversity of the tumor immune microenvironment (TIME) are becoming better understood, burgeoning research has progressed in this field. However, there is a scarcity of literature specifically focused on the bibliometric analysis of this topic. This study sought to investigate the development pattern of TIME-related research from 2006 to September 14, 2022, from a bibliometric perspective. Methods We acquired both articles and reviews related to TIME from the Web of Science Core Collection (WoSCC) (retrieved on September 14, 2022). R package “Bibliometrix” was used to calculate the basic bibliometric features, present the collaborative conditions of countries and authors, and generate a three-field plot to show the relationships among authors, affiliations, and keywords. VOSviewer was utilized for co-authorship analysis of country and institution and keyword co-occurrence analysis. CiteSpace was used for citation burst analysis of keywords and cited references. In addition, Microsoft Office Excel 2019 was used to develop an exponential model to fit the cumulative publication numbers. Results A total of 2545 publications on TIME were included, and the annual publication trend exhibited a significant increase over time. China and Fudan University were the most productive country and institution, with the highest number of publications of 1495 and 396, respectively. Frontiers in Oncology held the highest number of publications. A number of authors were recognized as the main contributors in this field. The clustering analysis revealed six clusters of keywords that highlighted the research hot spots in the fields of basic medical research, immunotherapy, and various cancer types separately. Conclusions This research analyzed 16 years of TIME-related research and sketched out a basic knowledge framework that includes publications, countries, journals, authors, institutions, and keywords. The finding revealed that the current research hot spots of the TIME domain lie in “TIME and cancer prognosis”, “cancer immunotherapy”, and “immune checkpoint”. Our researchers identified the following areas: “immune checkpoint-based immunotherapy”, “precise immunotherapy” and “immunocyte pattern”, which may emerge as frontiers and focal points in the upcoming years, offering valuable avenues for further exploration.

BackgroundPrimary gallbladder gastrinoma is an exceptionally uncommon tumor and is a rare form of neuroendocrine neoplasm. Until now, no cases of primary gallbladder gastrinoma and rare cases of primary gastrinoma from the biliary system have been reported.Case presentationWe report a case of a 50-year-old woman with watery diarrhea who intermittently received proton pump inhibitors (PPIs) as treatment. A serum gastrin level of 711 pg/ml was recorded after the withdrawal of PPI over 1 week. Enhanced computed tomography (CT) imaging and octreotide imaging uncovered a solitary tumor at the hepatic hilar region. During the laparoscopic surgery, it was determined that the tumor had its origin in the wall of the gallbladder neck, prompting the implementation of a laparoscopic cholecystectomy. Histological analysis revealed a primary neuroendocrine tumor from the neck of the gallbladder. The patient’s symptoms disappeared after the surgery with a follow-up of 6 months.ConclusionsThis case confirmed that primary gallbladder gastrinoma represents a distinct nosological entity. Immunohistochemical analysis plays a pivotal role in the diagnostic process. Given the limited understanding of primary gallbladder gastrinoma, our objective is to offer novel insights into this rare disease by delivering distinctive information and highlighting the therapeutic significance of surgical intervention.

ABSTRACT The detection of soil-borne pathogens by quantitative PCR (qPCR) has been challenging due to the pronounced influence of soil type on DNA extraction and PCR reactions. In the present study, we developed a novel qPCR system and an internal sample process control (ISPC) strain, RsPC, for the detection of Ralstonia solanacearum species complex (RSSC), the pathogens causing bacterial wilt. Specific primers and TaqMan probe were designed based on analyses of 16S rRNA gene sequences from 581 Ralstonia genomes, and the RsPC was constructed by insertion of an artificial fragment, which consisted of two fragments from the kanamycin-resistant gene and the gfp gene, into the chromosome of a phylogenetically closely related strain, Ralstonia pickettii JCM 5969. The qPCR target sequences of RSSC and RsPC shared primers; however, different TaqMan probes were used to distinguish them from each other. Abstract Background Intrahepatic cholangiocarcinoma (ICC) is a highly lethal hepatobiliary cancer, and very few patients can undergo surgery. The prognosis of advanced ICC is poor, especially in patients who progress after first-line chemotherapy, with a median overall survival of less than 10 months. Case presentation A 64-year-old male was diagnosed with advanced intrahepatic cholangiocarcinoma with ERBB2 (HER2) 3 + amplification determined by tissue-based testing and confirmed by next-generation sequencing. The patient was treated with pyrotinib added to pembrolizumab and lenvatinib after progressing with pyrotinib and tegafur and responded very well with regression of the tumor on imaging as well as normalization of tumor marker levels without serious adverse events. PET-CT after 6 months of treatment showed a partial response. The progression-free survival with second-line treatment was 17 months. For the third line of therapy, lenvatinib and pembrolizumab were used in combination with bevacizumab. Currently, he has had stable disease for approximately 6 months during third-line treatment. Conclusion Adding pyrotinib to pembrolizumab and lenvatinib may represent a promising strategy for advanced ICC patients who have high levels of HER2.

Purpose: The long-term situation of children with spinal cord injury (SCI) was investigated, and suggestions for helping them better return to the society were provided. Methods: SCI patients less than 18 years old hospitalized in Beijing Boai Hospital from January 2011 to December 2020 were retrospectively analyzed. Information including motor function, complications, characteristic changes, self-care abilities, school attendance and social participation were collected by telephone interview and electronic questionnaire. All the answers were statistically analyzed. Results: A total of 86 cases were enrolled, 77 girls and 9 boys, with a median injury age of 6 years and 2 months. The follow-up time was 3–130 months. The main cause of trauma in these children was sport injury (66.3%), the thoracic spinal cord was involved the most (91.9%), and complete SCIs accounted for the majority (76.7%). In order to further strengthen the operation protection measures of the optical cable and realize real-time monitoring of the hidden danger of external force damage in the optical cable line area, this paper uses the principle of Rayleigh backscattering and coherent detection to design and construct an optical cable external damage event monitoring based on distributed optical fiber vibration sensing early warning system. Using Phase Sensitive Optical Time Domain Reflectometer (Φ-OTDR) technology, the optical fiber vibration signal is collected in real time. The key characteristics of vibration sensing are extracted, and the type of hidden danger is sent to the operation and maintenance staff to locate and warn the hidden danger of external force damage in real time. Field verification results show that the system can simultaneously detect and locate multiple vibration sources along the sensing fiber, and can identify various types of disturbance signals. The monitoring and early warning system has a complete system and a simple structure. The recognition rate of various construction signals is as high as 95%. It realizes double early warning of the type of external breakage and precise location, which can effectively prevent and stop external damage events and improve the work efficiency of anti-external breakage of optical cables.

Heterostructure engineering of electrocatalysts provides a fascinating platform to reasonably manipulate the physicochemical properties of nanomaterials and further improve their catalytic efficiency for water electrolysis. However, it still remains a huge challenge to construct well-designed core-shell heterostructured catalysts and identify the key role of components for synergistic catalysis. Herein, a melted polymeric salt tactics was innovatively developed to synthesize heterostructured Ni@Ni(OH)2 core-shell nanomaterials supported on porous carbon (named Ni@Ni(OH)2/PC), wherein well-defined Ni(OH)2-Ni heterostructure plays a pivotal role in improving electrocatalytic activity for water reduction and oxidation. Besides, the stable porous carbon support functions as a highway for continuous electron transfer between Ni and Ni(OH)2, and simultaneously enables the full exposure of accessible active sites. The fabricated Ni@Ni(OH)2/PC exhibits outstanding bifunctional electrocatalytic performance for overall water splitting (ƞ10 = 1.55 V) with a good long-time stability. This work sheds new light on the design of engineering heterostructure of active bifunctional electrocatalysts for efficient energy conversion system.

The disease Fusarium crown and root rot (FCRR), caused mainly by Fusarium oxysporum f. sp. radicis-lycopersici (FORL), seriously affects commercial tomato [Solanum lycopersicum (Sl)] yields. However, the genes that offer resistance to FORL are limited and the mechanism of resistance to FCRR is poorly understood. Lectin receptor-like kinases (LecRKs) play critical roles in defensive responses and immunity in many plant species; however, whether specific LecRKs are involved in the response of tomato plants to FORL is unclear. Here, we report that the expression of SlLecRK1/Solyc09g011070.1 was obviously induced by the infection of FORL. Biochemical and cell biological data revealed that SlLecRK1 is an active kinase that is located at the cell membrane, while real-time quantitative PCR data suggested that SlLecRK1 is mainly expressed in stems and roots. Genetic studies showed that overexpression of SlLecRK1 significantly improved the resistance of tomato plants to FORL but did not cause visible changes in plant growth and development compared with wild-type control plants. RNA-Seq data suggested that the positive effects of SlLecRK1 on the resistance of tomato plants to FORL occur mainly by triggering the expression of ethylene-responsive transcription factor (ERF) genes. Together, our findings not only identify a new target for the development of FCRR-resistant tomato varieties, they also demonstrate a molecular mechanism linking SlLecRK1 and ERFs in regulating the immune responses of tomato plants to FORL.

Wildlife is increasingly forced to live in close proximity to humans, resulting in human-wildlife conflict and anthropogenic mortality. Carnivores persisting in human-dominated landscapes respond to anthropogenic threats through fine-scaled spatial and temporal behavioral adjustments. Although crucial for conservation, quantitative information on these adjustments is scarce. Taiwan’s endangered leopard cat occurs in rural human-dominated landscapes with a high anthropogenic mortality risk. To survive, the nocturnal leopard cat needs suitable habitats for foraging and safe refuge for resting during daytime hours when human activity peaks. In this study, we tracked seven VHF-collared leopard cats. To determine habitat selection patterns, we compared land use at nighttime locations and daytime resting sites with random points and fine-scaled vegetation characteristics at daytime resting sites with random points. Leopard cats selected natural habitats for nighttime hunting and avoided manmade and, to a lesser extent, agricultural habitats or used them according to availability. For daytime resting, leopard cats selected natural habitats and, to a lesser extent semi-natural habitats, such as unused land and abandoned orchards. Xinyang Maojian (XYMJ) is one of the premium green teas and originates from Xinyang, which is the northernmost green tea production area in China. The special geographic location, environmental conditions, and manufacturing process contribute to the unique flavor and rich nutrition of XYMJ green tea. Aroma is an important quality indicator in XYMJ green tea. In order to illustrate the aroma of XYMJ green tea, the key odorants in XYMJ green tea and their dynamic changes during the manufacturing processes were analyzed by headspace solid-phase microextraction (HS-SPME) combined with gas chromatography-mass spectrometry (GC-MS). A total of 73 volatile compounds of six different chemical classes were identified in the processed XYMJ green tea samples, and the manufacturing processes resulted in the losses of total volatile compounds. Among the identified volatile compounds, twenty-four aroma-active compounds, such as trans-nerolidol, geranylacetone, nonanal, (+)-δ-cadinene, linalool, (Z)-jasmone, cis-3-hexenyl butyrate, cis-3-hexenyl hexanoate, methyl jasmonate, and β-ocimene, were identified as the key odorants of XYMJ green tea based on odor activity value (OAV). The key odorants are mainly volatile terpenes (VTs) and fatty acid-derived volatiles (FADVs). Except for (+)-δ-cadinene, copaene, cis-β-farnesene, (Z,E)-α-farnesene and phytol acetate, the key odorants significantly decreased after fixing. The principal coordinate analysis (PCoA) and the hierarchical cluster analysis (HCA) analyses suggested that fixing was the most important manufacturing process for the aroma formation of XYMJ green tea. These findings of this study provide meaningful information for the manufacturing and quality control of XYMJ green tea.

Frost damage in permafrost tunnels is very common, and this can have a negative influence on traffic. The most serious frost damage typically occurs at a certain length from the tunnel opening. Thus the antifreeze measures of the lining structure in this area need to be strengthened. In this study, the antifreeze disease fortification length for permafrost tunnels is determined from heat transfer and mathematical physics equations by the theoretical analysis method. The temperature distribution characteristics of the lining along the tunnel axis under the influence of the tunnel depth, the tunnel radius, the wind velocity at the tunnel opening, and the thermal conductivity of the insulation layer are analysed. The results show that the longitudinal temperature characteristics in the tunnel axis are influenced by many factors. The proposed antifreeze disease length of the permafrost tunnel was found to be approximately 31 times of the tunnel diameter, which agrees with the results of the numerical simulation. It verifies the rationality of the theoretical calculation. This value, 31 times of the tunnel diameter, can be used as a reference for the design of the tunnel antifreeze disease fortification length.

Background/Aims: Erinacine, which is extracted from the medicinal mushroom Hericium erinaceus, is known to play anticancer roles in human cancers. The following study aims to investigate the role of erinacine in the opening of the mitochondrial permeability transition pore (MPTP) in hepatocellular carcinoma (HCC) through the PI3K/Akt/GSK-3β signaling pathway and highlights the applicability of erinacine in HCC treatments. Methods: HCC and paracancerous tissues were obtained from 85 HCC patients who’ve undergone surgical resection. Immunohistochemistry was adopted to detect positive expression of PI3K, Akt, and GSK-3β. Treatment of HepG-2 with LY294002 (an inhibitor of the PI3K/Akt/GSK-3β signaling pathway) and different concentration of erinacine was performed to determine the involvement of LY294002 in erinacine action. The expressions of PI3K, Akt, GSK-3β, CyclinD1, Vimentin, β-catenin, Bcl-2, E-cadherin, Bax, and caspase-9 were determined by RT-qPCR and Western blot analysis. Cell viability, colony formation rate, migration, invasion, cycle, and apoptosis were detected by MTT, colony formation, wound healing assay, Transwell assay, and flow cytometry, respectively. The size and weight of xenograft tumors were observed in nude mice. Mitochondrial membrane potential in HepG-2 was determined using laser scanning confocal microscopy following JC-1 staining. Mitochondrial Ca2+ indicator Rhod-2, AM was used to detect the changes of mitochondrial Ca2+, while western blot analysis was employed to detect the presence levels of cytochrome C (cyt-C). Results: The results revealed that PI3K, Akt, and GSK-3β were up-regulated in HCC tissues. Erinacine or LY294002 led to a decrease in mitochondrial membrane potential, increase in intracellular mitochondrial Ca2+, and the release of cyt-C in mitochondria. In addition, Erinacine was found to decrease the mitochondrial membrane potential, expression of PI3K, Akt, GSK-3β, CyclinD1, Vimentin, β-catenin, and Bcl-2, cell proliferation, colony formation ability, migration, invasion, and xenograft tumor size, while E-cadherin, Bax, and caspase-9 expression, and cell apoptosis were elevated in a dose-dependent manner. Erinacine also stimulated the effects of LY294002 on the HCC. Following the addition of 500 μM Erinacine and MPTP opening inhibitor CsA, we found that the mitochondrial membrane potential level increased, while mitochondrial Ca2+ and Cyt-C decreased from the mitochondria. Conclusion: The results from the study demonstrated that erinacine induced MPTP opening, facilitates the release of cyt-C, and inhibited cell proliferation, migration, and invasion, while it promotes apoptosis by inactivating the PI3K/Akt/GSK-3β signaling pathway, preventing the progression of HCC.

A sharing multi-owner setting where data was owned by a fixed number of data owners,the existing searchable encryption schemes could not support ciphertext retrieval and fine-grained access control at the same time.For this end,an efficient cryptographic primitive called as multi-owner accredited keyword search over encrypted data scheme was designed,through combining linear secret-sharing technique with searchable encryption schemes,only the data users authorized bymulti-owner by could decrypt the returned results.The formal security analysis shows that the scheme can protect security and privacy under the bilinear Diffie-Hellman assumption.As a further contribution,an empirical study over real-world dataset was conelucted to show the effectiveness and practicability of the scheme.

A mu1ti-keyword search over encrypted data was proposec1 with fi1e-1eve1 access authorization and data user revocation scheme through emp1oying ciphertext-po1icy attribute-based encryption（CP-ABE）.The scheme supports fi1e-1eve1 access authorization in the mu1ti-owner mu1ti-user settings，which means that data users cou1d on1y gain the authorized fi1es encrypted by different data owners with different keys.Moreover，the scheme cou1d achieve data user revocation.Forma1 security ana1ysis shows that the proposed scheme cou1d resist the chosen keyword attack in random orac1e.Moreover，the experimenta1 study over rea1-wor1d dataset demonstrates its efficiency and feasibi1ity in practice.

Multi-authority attribute-based encryption was very suitable for data access control in a cloud storage environment.However,efficient user revocation in multi-authority attribute-based encryption remains a challenging problem that prevents it from practical applications.A multi-authority ciphertext-policy attribute-based encryption scheme with efficient revocation was proposed in prime order bilinear groups,and was further proved statically secure and revocable in the random oracle model.Extensive efficiency analysis results indicate that the proposed scheme significantly reduce the computation cost for the users.In addition,the proposed scheme supports large universe and any monotone access structures,which makes it more flexible for practical applications.

Post traumatic stress disorder (PTSD) is a mental illness that affected numerous people. The anti-PTSD-like effects of puerarin is unknown, although the antidepressant- and anxiolytic- like effects of puerarin have been reported. The PTSD behavioral deficits in rats were induced by single prolonged stress (SPS), mainly including the reduced time/entries in the open arms and the elevated time/entries in the closed arms in elevated plus maze test, increased freezing duration in contextual fear paradigm and lowered time/entries in the central zone in open field test. However, the behavioral deficits were attenuated by puerarin (50 and 100 mg/kg) without affecting the locomotor activity. For the evaluation of mechanism, the decreased levels of progesterone, allopregnanolone, and the increased levels of corticosterone, corticotropin releasing hormone, and adrenocorticotropic hormone in the brain or serum were induced by SPS, which is blocked by puerarin. In summary, the anti-PTSD-like effects of puerarin were associated with biosynthesis of neurosteroids and normalized levels of stress hormones in HPA axis.

This study is aimed at exploring the effects of multi-stage homogenization and trace amounts of Zr and Sc on the microstructures, mechanical properties, and recrystallization of Al–4.5Zn–1.5Mg alloys. The mechanical properties of the AA7005 aluminum alloys after the T6 heat treatment were evaluated through a hardness test and tensile test. The microstructures were analyzed by an optical microscope (OM), a differential scanning calorimeter (DSC), a transmission electron microscope (TEM), a scanning electron microscope (SEM), and electron backscattered diffraction (EBSD). The results show that the grain refinement effect of the as-cast, homogenized, and recrystallized Al–4.5Zn–1.5Mg alloy containing 0.05Sc (wt%) after the T6 heat treatment was more significant than that of the alloy containing 0.1Zr (wt%). In addition, compared with the aforementioned one-stage homogenization heat treatment, the two-stage homogenization made the dispersed grain phase (Al3Zr/Al3Sc) smaller. As a result, the T6 mechanical strength of the alloy after the two-stage homogenization heat treatment was better than that of the contrastive alloy after the one-stage homogenization heat treatment. However, the two different homogenization heat treatments caused a greater divergence between the sizes of the dispersed grain phases of the Al–4.5Zn–1.5Mg alloys containing Zr than between the sizes of the dispersed grain phases of the alloys containing Sc. Therefore, after the two-stage homogenization heat treatment, the alloy with 0.1Zr (wt%) promoted the mechanical properties better than the alloy with 0.05Sc (wt%).

The security flaws of a direct anonymous attestation scheme proposed by Ge,et al.Were analyzed,and the re-sult shows that the authentication protocol of the scheme is vulnerable to reply attacks and platform masquerade attacks when being used for remote attestation.An improved direct anonymous attestation authentication scheme with the in-volvement of key agreement was proposed to provide the property of mutual authentication.The analysis shows that the proposal can realize direct anonymous attestation with the properties of forgery-resistance and anonymity,and resist reply attacks and platform masquerade attacks; the scheme is effective and suitable for the mobile trusted computing platforms.

An interference sensing based(ISB)routing metric which mainly depend on the estimation of signal power was introduced for the multi-radio multi-channel wireless mesh networks.The existing metric such like WCETT and iAWARE all have their drawbacks were shown.As shown in the equations of iAWARE,it could be proved that worse background noise corresponds to a better metric value which was imprecise.The iAWARE to make it suited for the environment with background noise was improved,and the isotonic property was regained that the Bellman-Ford and Dijkstra algorithm could be deployed in ISB.The simulation results prove the novel algorithm takes the advantages of higher efficiency both in throughput and end to end delays,compared with the traditional ones HopCount,ETT,WCETT and iAWARE.

IntroductionDiabetic nephropathy (DN), distinguished by detrimental changes in the renal glomeruli, is regarded as the leading cause of death from end-stage renal disease among diabetics. Cellular senescence plays a paramount role, profoundly affecting the onset and progression of chronic kidney disease (CKD) and acute kidney injuries. This study was designed to delve deeply into the pathological mechanisms between glomerulus-associated DN and cellular senescence.MethodsGlomerulus-associated DN datasets and cellular senescence-related genes were acquired from the Gene Expression Omnibus (GEO) and CellAge database respectively. By integrating bioinformatics and machine learning methodologies including the LASSO regression analysis and Random Forest, we screened out four signature genes. The receiver operating characteristic (ROC) curve was performed to evaluate the diagnostic performance of the selected genes. Rigorous experimental validations were subsequently conducted in the mouse model to corroborate the identification of three signature genes, namely LOX, FOXD1 and GJA1. Molecular docking with chlorogenic acids (CGA) was further established not only to validate LOX, FOXD1 and GJA1 as diagnostic markers but also reveal their potential therapeutic effects.Results and discussionIn conclusion, our findings pinpointed three diagnostic markers of glomerulus-associated DN on the basis of cellular senescence. These markers could not only predict an increased risk of DN progression but also present promising therapeutic targets, potentially ushering in innovative treatments for DN in the elderly population.

A novel theoretical framework and method, which is based on a novel space-time-coding metasurface (STCM) with nonlinearly periodic phases, is proposed to generate frequency-modulated continuous waves (FMCWs) and dynamically control their spatial propagation behaviors simultaneously.

In this paper, Ni-based alloy coating was prepared on pure iron substrate by steady magnetic field assisted laser cladding technology. The influence mechanism of steady magnetic field on the Ni-based coating was investigated by analyzing the macro-morphology, phase composition, microstructure, element composition and properties of the coatings, respectively. The results reveal that external magnetic field can restrain the surface fluctuation of the coatings. It has no effect on the phase composition of the coatings. Compared with the coating prepared without magnetic field assistance, the grains of the coatings prepared under the magnetic induction intensities of 0.1 T, 0.15 T and 0.2 T are all refined. The solid solubility of the γ-Ni matrix is also increased. Hard phases CrB in the microstructures appear clustered aggregation and preferential orientation with the increase of the magnetic induction intensity. The application of steady magnetic field is beneficial to reduce the number of cracks on the surface and improve the microhardness of the coatings. The coating exhibits the best comprehensive property under the magnetic induction intensity of 0.2 T. Under the action of steady magnetic field, the electromagnetic braking effect and the thermoelectric magnetic fluid effect can both influence the laser cladding process. The surface morphology of the coating is mainly affected by the electromagnetic braking effect. The solidification microstructure is mainly controlled by the thermoelectric magnetic fluid effect.

Fe-based alloy coatings with different Mo contents were fabricated on 45 steel substrates by laser cladding. X-ray diffractometer (XRD), scanning electron microscope (SEM) and energy dispersive spectrometer (EDS) were used to analyze the phase composition and microstructural evolution of the coatings. The effects of Mo addition on the wear and electrochemical behaviors of the coatings were analyzed by ball-on-disc wear tester and electrochemical workstation. The results showed that the addition of Mo could help refine grains and promote the transition of dendrites to equiaxed grains. With the increase of Mo content, the hard phases changed from Fe _2 B to Mo _2 FeB _2 . Mo and Cr tended to aggregate in the intercrystalline regions in the form of hard phases. When the Mo content was 4.8 wt%, the hardness increased by 3.6 times compared with that of the substrate. The wear resistance of the coatings increased with the Mo addition. Mo played the role of fine grain strengthening and solid solution strengthening, which was beneficial for the improvement of the hardness and wear resistance. Polarization curves and electrochemical impedance spectroscopy test results revealed that the addition of Mo could improve the corrosion resistance of Fe-based coating.

The PI3K/Akt/mTOR and JAK/STAT3 signaling pathways are important for regulating apoptosis, and are frequently activated in cancers. In this study, we targeted STAT3 and mTOR in human hepatocellular carcinoma Bel-7402 cells and examined the subsequent alterations in cellular apoptosis. The expression of STAT3 was silenced with small interfering RNA (siRNA)-expressing plasmid. The activity of mTOR was inhibited using rapamycin. Following treatment, Annexin V/propidium iodide staining followed by flow cytometry and Hoechst33258 immunofluorescence staining was used to examine cellular apoptosis. JC-1 staining was used to monitor depolarization of mitochondrial membrane (ΔΨm). Furthermore, the expression of activated caspase 3 protein was analyzed by Western blotting. Compared to non-treated or control siRNA-transfected cells, significantly higher levels of apoptosis were detected in siSTAT3-transfected or rapamycin-treated cells (P < 0.05), which was further enhanced in cells targeted for both molecules (P < 0.05). The pro-apoptotic effects were accompanied with concomitant depolarization of mitochondrial membrane and up-regulation of activated caspase 3. Combined treatments using rapamycin and STAT3 gene silencing significantly increases apoptosis in Bel-7402 cells, displaying more dramatic effect than any single treatment. This study provides evidence for targeting multiple molecules in cancer therapy.
