Search

The geometric calibration of cameras becomes necessary when images should be undistorted, geometric image information is needed or data from more than one camera have to be fused. This process is often done using a target with a checkerboard or circular pattern and a given geometry. In this work, a coded checkerboard target for thermal imaging cameras and the corresponding image processing algorithm for iterative feature detection are presented. It is shown that, due in particular to the resulting better feature detectability at image borders, lower uncertainties in the estimation of the distortion parameters are achieved.

Abstract An event topology with two secondary vertices compatible with the decay of short-lived particles was found in the analysis of neutrino interactions in the OPERA target. The observed topology is compatible with tau neutrino charged current (CC) interactions with charm production and neutrino neutral current (NC) interactions with $$c\overline{c}$$ cc¯ pair production. However, other processes can mimic this topology. A dedicated analysis was implemented to identify the underlying process. A Monte Carlo simulation was developed and complementary procedures were introduced in the kinematic reconstruction. A multivariate analysis technique was used to achieve an optimal separation of signal from background. Most likely, this event is a $$\nu _{\tau }$$ ντ CC interaction with charm production, the tau and charm particle decaying into 1 prong and 2 prongs, respectively. The significance of this observation is evaluated.

Abstract Heavy Neutral Leptons (HNLs) are hypothetical particles predicted by many extensions of the Standard Model. These particles can, among other things, explain the origin of neutrino masses, generate the observed matter-antimatter asymmetry in the Universe and provide a dark matter candidate. The SHiP experiment will be able to search for HNLs produced in decays of heavy mesons and travelling distances ranging between O $$ \mathcal{O} $$ (50 m) and tens of kilometers before decaying. We present the sensitivity of the SHiP experiment to a number of HNL’s benchmark models and provide a way to calculate the SHiP’s sensitivity to HNLs for arbitrary patterns of flavour mixings. The corresponding tools and data files are also made publicly available.

Abstract The OPERA experiment has discovered the tau neutrino appearance in the CNGS muon neutrino beam, in agreement with the 3 neutrino flavour oscillation hypothesis. The OPERA neutrino interaction target, made of Emulsion Cloud Chambers, was particularly efficient in the reconstruction of electromagnetic showers. Moreover, thanks to the very high granularity of the emulsion films, showers induced by electrons can be distinguished from those induced by π 0s, thus allowing the detection of charged current interactions of electron neutrinos. In this paper the results of the search for electron neutrino events using the full dataset are reported. An improved method for the electron neutrino energy estimation is exploited. Data are compatible with the 3 neutrino flavour mixing model expectations and are used to set limits on the oscillation parameters of the 3+1 neutrino mixing model, in which an additional mass eigenstate m 4 is introduced. At high Δm 412 (≳0.1 eV2), an upper limit on sin2 2θ μe is set to 0.021 at 90% C.L. and Δm 412 ≳ 4 × 10− 3 eV2 is excluded for maximal mixing in appearance mode.

Abstract The OPERA experiment was designed to search for $$\nu _{\mu } \rightarrow \nu _{\tau }$$ νμ→ντ oscillations in appearance mode through the direct observation of tau neutrinos in the CNGS neutrino beam. In this paper, we report a study of the multiplicity of charged particles produced in charged-current neutrino interactions in lead. We present charged hadron average multiplicities, their dispersion and investigate the KNO scaling in different kinematical regions. The results are presented in detail in the form of tables that can be used in the validation of Monte Carlo generators of neutrino–lead interactions.

211Po ions in the ground and isomeric states were produced via 238U projectile fragmentation at 1000 MeV/u. The 211Po ions were spatially separated in flight from the primary beam and other reaction products by the fragment separator FRS. The ions were energy-bunched, slowed-down and thermalized in a gas-filled cryogenic stopping cell (CSC). They were then extracted from the CSC and injected into a high-resolution multiple-reflection time-of-flight mass spectrometer (MR-TOF-MS). The excitation energy of the isomer and, for the first time, the isomeric-to-ground state ratio were determined from the measured mass spectrum. In the subsequent experimental step, the isomers were spatially separated from the ions in the ground state by an ion deflector and finally collected with a silicon detector for decay spectroscopy. This pioneering experimental result opens up unique perspectives for isomer-resolved studies. With this versatile experimental method new isomers with half-lives longer than a few milliseconds can be discovered and their decay properties can be measured with highest sensitivity and selectivity. These experiments can be extended to studies with isomeric beams in nuclear reactions. The aim of the COBRA experiment is to prove the existence of neutrinoless double-beta-decay (0νββ-decay) and to measure its half-life. For this purpose a detector array made of cadmium-zinc-telluride (CdZnTe) semiconductor detectors is operated at the Gran Sasso Underground Laboratory (LNGS) in Italy. This setup is used to investigate the experimental issues of operating CdZnTe detectors in low-background mode and to identify potential background components, whilst additional studies are proceeding in surface laboratories. The experiment currently consists of monolithic, calorimetric detectors of coplanar grid design (CPG detectors). These detectors are 1 × 1 × 1 cm3 and are arranged in 4 × 4 detector layers. Ultimately four layers will be installed by the end of 2013, of which two are currently operating. To date 82.3 kg·days of data have been collected. In the region of interest for 116Cd around 2.8 MeV, the median energy resolution is 1.5% FWHM, and a background level near 1 counts/keV/kg/y has been reached. This paper gives an overview of the current status of the experiment and future perspectives.

Level lifetimes for the candidate chiral doublet bands of 80Br were extracted by means of the Doppler-shift attenuation method. The absolute transition probabilities derived from the lifetimes agree well with the M1 and E2 chiral electromagnetic selection rules, and are well reproduced by the triaxial particle rotor model calculations. Such good agreements among the experimental data, selection rules of chiral doublet bands and theoretical calculations are rare and outstanding in researches of nuclear chirality. Besides odd-odd Cs isotopes, odd-odd Br isotopes in the A≈ 80 mass region represent another territory that exhibits the ideal selection rules expected for chiral doublet bands.

The β-decay of the even-even nucleus 70Kr with Z=N+2, has been investigated at the Radioactive Ion Beam Factory (RIBF) of the RIKEN Nishina Center using the BigRIPS fragment separator, the ZeroDegree Spectrometer, the WAS3ABI implantation station and the EURICA HPGe cluster array. Fifteen γ-rays associated with the β-decay of 70Kr into 70Br have been identified for the first time, defining ten populated states below Eexc=3300 keV. The half-life of 70Kr was derived with increased precision and found to be t1/2=45.19±0.14 ms. The β-delayed proton emission probability has also been determined as εp=0.545(23)%. An increase in the β-strength to the yrast 1+ state in comparison with the heaviest Z=N+2 system studied so far (62Ge decay) is observed that may indicate increased np correlations in the T=0 channel. The β-decay strength deduced from the results is interpreted in terms of the proton-neutron quasiparticle random-phase approximation (pnQRPA) and also with a schematic model that includes isoscalar and isovector pairing in addition to quadrupole deformation. The application of this last model indicates an approximate realization of pseudo-SU(4) symmetry in this system.

The γ-ray emission from the nuclei 62,64Fe following Coulomb excitation at bombarding energy of 400-440 AMeV was measured with special focus on E1 transitions in the energy region 4-8 MeV. The unstable neutron-rich nuclei 62,64Fe were produced at the FAIR-GSI laboratories and selected with the FRS spectrometer. The γ decay was detected with AGATA. From the measured γ-ray spectra the summed E1 strength is extracted and compared to microscopic quasi-particle phonon model calculations. The trend of the E1 strength with increasing neutron number is found to be fairly well reproduced with calculations that assume a rather complex structure of the 1− states (three-phonon states) inducing a strong fragmentation of the E1 nuclear response below the neutron binding energy.

The lifetimes of the low-lying excited states 2+ and 4+ have been directly measured in the neutron-deficient 106,108Sn isotopes. The nuclei were populated via a deep-inelastic reaction and the lifetime measurement was performed employing a differential plunger device. The emitted γ rays were detected by the AGATA array, while the reaction products were uniquely identified by the VAMOS++ magnetic spectrometer. Large-Scale Shell-Model calculations with realistic forces indicate that, independently of the pairing content of the interaction, the quadrupole force is dominant in the B(E2;21+→0g.s.+) values and it describes well the experimental pattern for 104−114Sn; the B(E2;41+→21+) values, measured here for the first time, depend critically on a delicate pairing-quadrupole balance, disclosed by the very precise results in 108Sn.

Electric quadrupole matrix elements, Mp, for the Jπ=2+→0+, ΔT=0, T=1 transitions across the A=46 isobaric multiplet 46Cr-46V-46Ti have been measured at GSI with the FRS-LYCCA-AGATA setup. This allows direct insight into the isospin purity of the states of interest by testing the linearity of Mp with respect to Tz. Pairs of nuclei in the T=1 triplet were studied using identical reaction mechanisms in order to control systematic errors. The Mp values were obtained with two different methodologies: (i) a relativistic Coulomb excitation experiment was performed for 46Cr and 46Ti; (ii) a “stretched target” technique was adopted here, for the first time, for lifetime measurements in 46V and 46Ti. A constant value of Mp across the triplet has been observed. Shell-model calculations performed within the fp shell fail to reproduce this unexpected trend, pointing towards the need of a wider valence space. This result is confirmed by the good agreement with experimental data achieved with an interaction which allows excitations from the underlying sd shell. A test of the linearity rule for all published data on complete T=1 isospin triplets is presented.

The γ decay from the high-lying states of 124Sn was measured using the inelastic scattering of 17O at 340 MeV. The emitted γ rays were detected with high resolution with the AGATA demonstrator array and the scattered ions were detected in two segmented ΔE–E silicon telescopes. The angular distribution was measured both for the γ rays and the scattered 17O ions. An accumulation of E1 strength below the particle threshold was found and compared with previous data obtained with (γ,γ′) and (α,α′γ) reactions. The present results of elastic scattering, and excitation of E2 and E1 states were analysed using the DWBA approach. From this comprehensive description the isoscalar component of the 1− excited states was extracted. The obtained values are based on the comparison of the data with DWBA calculations including a form factor deduced using a microscopic transition density. The νg9/2,d5/2,s1/2 orbitals are assumed to be responsible for the swift onset of collectivity observed in the region below 68Ni. Especially the single-particle energies and strengths of these orbitals are of importance. We studied such properties in the nearby 67Ni nucleus, by performing a (d,p)-experiment in inverse kinematics employing a post-accelerated radioactive ion beam (RIB) at the REX-ISOLDE facility. The experiment was performed at an energy of 2.95 MeV/u using a combination of the T-REX particle detectors, the Miniball γ-detection array and a newly-developed delayed-correlation technique as to investigate μs-isomers. Angular distributions of the ground state and multiple excited states in 67Ni were obtained and compared with DWBA cross-section calculations, leading to the identification of positive-parity states with substantial νg9/2 (1007 keV) and νd5/2 (2207 keV and 3277 keV) single-particle strengths up to an excitation energy of 5.8 MeV. 50% of the νd5/2 single-particle strength relative to the νg9/2-orbital is concentrated in and shared between the first two observed 5/2+ levels. A comparison with extended Shell Model calculations and equivalent (3He, d) studies in the region around 9040Zr50 highlights similarities for the strength of the negative-parity pf and positive-parity g9/2 state, but differences are observed for the d5/2 single-particle strength.

Throughout the history of human spaceflight, spacefarers have experienced and reported the occurrence of medical conditions, including various illnesses and injuries. Therefore, future spaceflight missions to the Moon and Mars will require the capabilities necessary for maintaining the health of these new space travelers. Mass, power, and volume available in the space vehicles used for these missions will be severely constrained. The ability to resupply or evacuate to Earth will be limited or non-existent, and ground-based support will no longer be immediate due to communication latencies and blackouts. These vehicle and mission constraints will necessitate healthcare be provided from an efficiently planned medical system. To provide the necessary care, these medical systems will need to include at a minimum, several different types of medical devices, consumable resources, centralized data management, procedural guidance, and decision support technologies. Medical devices needed for diagnosing and treating medical conditions that are expected to occur during future spaceflight missions may include real-time health monitoring, medical imaging capabilities, as well as blood and urine analysis. Novel methods for interacting with onboard patient medical records will be necessary, as will resource tracking. Terrestrial medicine shares many of these same needs, therefore a multitude of these required medical capabilities can likely be satisfied by currently available, Commercial-Off-The-Shelf (COTS) devices and methodologies; however, in some cases the unique space environment and increased mission durations will drive the need for modifications or customization of standard technologies and treatment procedures. This article will provide a review of medical devices and technologies that have been considered for inclusion within future spaceflight medical systems. It will also include a discussion about the modifications and customized development that have been performed, as well as descriptions of the technology demonstrations that have been conducted in analog and spaceflight environments.

This study characterizes expression of pharmacogenes across the histological NAFLD severity spectrum. Here, the authors show the downregulation of CYP2C19 in NAFLD which supports developing personalized medicine approaches for drugs sensitive to metabolism by the CYP2C19 enzyme.

Purpose: To describe an atypical case of a choroidal melanoma presenting with eyelid edema, chemosis, pain and diplopia and demonstrating significant extraocular extension on ultrasonography and neuroimaging. Observations: A 69-year-old woman presented with a headache, eyelid edema, chemosis and pain in the right eye. Upon subsequent onset of diplopia, MRI of the orbits was performed and demonstrated a predominantly extraocular, intraconal mass with a small intraocular component. She was started on corticosteroids and referred to the ocular oncology service for evaluation. On fundus examination, she was noted to have a pigmented choroidal lesion consistent with melanoma, and ultrasound showed a large area of extraocular extension. Enucleation, enucleation with subsequent radiation and exenteration were discussed, and the patient requested an opinion from radiation oncology. A repeat MRI obtained by radiation oncology demonstrated a decrease in the extraocular component after corticosteroid treatment. The improvement was interpreted as suggestive of lymphoma by the radiation oncologist who recommended external beam radiation (EBRT). Fine needle aspiration biopsy was insufficient for cytopathologic diagnosis, and the patient elected to proceed with EBRT in the absence of a definitive diagnosis. Next generation sequencing revealed GNA11 and SF3B1 mutations, which supported the diagnosis of uveal melanoma and led to enucleation. Conclusion and Importance: Choroidal melanoma may present with pain and orbital inflammation secondary to tumor necrosis, which may delay diagnosis and decrease the diagnostic yield of fine-needle aspiration biopsy. Next generation sequencing may aid the diagnosis of choroidal melanoma when there is clinical uncertainty and cytopathology is unavailable.

Abstract Anti-EGFR antibodies are effective in therapies for late-stage colorectal cancer (CRC); however, many tumours are unresponsive or develop resistance. We performed genomic analysis of intrinsic and acquired resistance to anti-EGFR therapy in prospectively collected tumour samples from 25 CRC patients receiving cetuximab (an EGFR inhibitor). Of 25 CRC patients, 13 displayed intrinsic resistance to cetuximab; 12 were intrinsically sensitive. We obtained six re-biopsy samples at acquired resistance from the intrinsically sensitive patients. NCOA4–RET and LMNA–NTRK1 fusions and NRG1 and GNAS amplifications were found in intrinsic-resistant patients. In cetuximab-sensitive patients, we found KRAS K117N and A146T mutations in addition to BRAF V600E, AKT1 E17K, PIK3CA E542K, and FGFR1 or ERBB2 amplifications. The comparison between baseline and acquired-resistant tumours revealed an extreme shift in variant allele frequency of somatic variants, suggesting that cetuximab exposure dramatically selected for rare resistant subclones that were initially undetectable. There was also an increase in epithelial-to-mesenchymal transition at acquired resistance, with a reduction in the immune infiltrate. Furthermore, characterization of an acquired-resistant, patient-derived cell line showed that PI3K/mTOR inhibition could rescue cetuximab resistance. Thus, we uncovered novel genomic alterations that elucidate the mechanisms of sensitivity and resistance to anti-EGFR therapy in metastatic CRC patients.

Risk factors include medial patellofemoral ligament (MPFL) tears, patella alta, trochlear dysplasia, and an increased tibial tubercle–trochlear groove distance. Surgical management must be based on a patient’s unique joint pathoanatomy and may require MPFL reconstruction with tibial tubercle osteotomy or trochleoplasty either alone or in combination. This article discusses our preferred technique for surgical treatment of recurrent patellar instability with MPFL reconstruction using a quadriceps tendon autograft, an open trochleoplasty, and a tibial tubercle osteotomy for patients with patella alta, trochlear dysplasia, and an increased tibial tubercle–trochlear groove distance.

Chronic patellar instability is a disabling issue for some patients and may require surgical intervention for proper treatment. Risk factors for recurrent dislocations include medial patellofemoral ligament (MPFL) tears, patella alta, trochlear dysplasia, and increased tibial tubercle to trochlear groove distance. Surgical management must be based on a patient’s unique joint pathoanatomy and typically requires medial patellofemoral ligament reconstruction, with or without accompanying procedures such as tibial tubercle osteotomy or sulcus-deepening trochleoplasty. Chronic patellar instability in minors with open growth plates, requires alternative MPFL reconstruction techniques to prevent physeal injury, because of the close proximity of the femoral physis to the MPFL insertion. This article discusses the authors’ preferred technique for surgical treatment of recurrent patellar instability with a medial patellofemoral ligament reconstruction using a quadriceps tendon autograft.

Abstract Background Activation of microglia, the resident immune cells of the central nervous system, is a prominent pathological hallmark of Alzheimer’s disease (AD). However, the gene expression changes underlying microglia activation in response to tau pathology remain elusive. Furthermore, it is not clear how murine gene expression changes relate to human gene expression networks. Methods Microglia cells were isolated from rTg4510 tau transgenic mice and gene expression was profiled using RNA sequencing. Four age groups of mice (2-, 4-, 6-, and 8-months) were analyzed to capture longitudinal gene expression changes that correspond to varying levels of pathology, from minimal tau accumulation to massive neuronal loss. Statistical and system biology approaches were used to analyze the genes and pathways that underlie microglia activation. Differentially expressed genes were compared to human brain co-expression networks. Results Statistical analysis of RNAseq data indicated that more than 4000 genes were differentially expressed in rTg4510 microglia compared to wild type microglia, with the majority of gene expression changes occurring between 2- and 4-months of age. These genes belong to four major clusters based on their temporal expression pattern. Genes involved in innate immunity were continuously up-regulated, whereas genes involved in the glutamatergic synapse were down-regulated. Up-regulated innate inflammatory pathways included NF-κB signaling, cytokine-cytokine receptor interaction, lysosome, oxidative phosphorylation, and phagosome. NF-κB and cytokine signaling were among the earliest pathways activated, likely driven by the RELA, STAT1 and STAT6 transcription factors. The expression of many AD associated genes such as APOE and TREM2 was also altered in rTg4510 microglia cells. Differentially expressed genes in rTg4510 microglia were enriched in human neurodegenerative disease associated pathways, including Alzheimer’s, Parkinson’s, and Huntington’s diseases, and highly overlapped with the microglia and endothelial modules of human brain transcriptional co-expression networks. Conclusion This study revealed temporal transcriptome alterations in microglia cells in response to pathological tau perturbation and provides insight into the molecular changes underlying microglia activation during tau mediated neurodegeneration.

Summary: We present a consensus atlas of the human brain transcriptome in Alzheimer’s disease (AD), based on meta-analysis of differential gene expression in 2,114 postmortem samples. We discover 30 brain coexpression modules from seven regions as the major source of AD transcriptional perturbations. We next examine overlap with 251 brain differentially expressed gene sets from mouse models of AD and other neurodegenerative disorders. Human-mouse overlaps highlight responses to amyloid versus tau pathology and reveal age- and sex-dependent expression signatures for disease progression. Human coexpression modules enriched for neuronal and/or microglial genes broadly overlap with mouse models of AD, Huntington’s disease, amyotrophic lateral sclerosis, and aging. Other human coexpression modules, including those implicated in proteostasis, are not activated in AD models but rather following other, unexpected genetic manipulations. Our results comprise a cross-species resource, highlighting transcriptional networks altered by human brain pathophysiology and identifying correspondences with mouse models for AD preclinical studies.

Purpose: To present a rare presentation of abusive head trauma (AHT) in an infant with a hereditary vitreoretinopathy. Observations: A two-month-old infant female victim of AHT presented with bilateral rhegmatogenous retinal detachments from giant retinal tears. She had rib fractures, a subdural hematoma, and hyphemas bilaterally. Retinal hemorrhages were not observed. The left eye was repaired by vitrectomy with intermediate-term perfluorocarbon liquid tamponade. Genetic testing demonstrated a pathogenic COL2A1 mutation confirming Stickler syndrome. Conclusions and Importance: Ophthalmic complications of AHT classically manifest as retinal hemorrhages in multiple layers. Instead, bilateral RRDs from GRTs were observed in this infant with Stickler syndrome. OPERA is a long-baseline experiment designed to search for $\nu_{\mu}\to\nu_{\tau}$ oscillations in appearance mode. It was based at the INFN Gran Sasso laboratory (LNGS) and took data from 2008 to 2012 with the CNGS neutrino beam from CERN. After the discovery of $\nu_\tau$ appearance in 2015, with $5.1\sigma$ significance, the criteria to select $\nu_\tau$ candidates have been extended and a multivariate approach has been used for events identification. In this way the statistical uncertainty in the measurement of the oscillation parameters and of $\nu_\tau$ properties has been improved. Results are reported.

Autism is a severe disorder that involves both genetic and environmental factors. Expression profiling of the superior temporal gyrus of six autistic subjects and matched controls revealed increased transcript levels of many immune system-related genes. We also noticed changes in transcripts related to cell communication, differentiation, cell cycle regulation and chaperone systems. Critical expression changes were confirmed by qPCR (BCL6, CHI3L1, CYR61, IFI16, IFITM3, MAP2K3, PTDSR, RFX4, SPP1, RELN, NOTCH2, RIT1, SFN, GADD45B, HSPA6, HSPB8 and SERPINH1). Overall, these expression patterns appear to be more associated with the late recovery phase of autoimmune brain disorders, than with the innate immune response characteristic of neurodegenerative diseases. Moreover, a variance-based analysis revealed much greater transcript variability in brains from autistic subjects compared to the control group, suggesting that these genes may represent autism susceptibility genes and should be assessed in follow-up genetic studies.
