Isoetes baodongii is a diploid species of Isoetaceae distributed in low altitude area, its megaspore ornamentation is similar to tetraploid species I. sinensis. We collected leaf material of I. baodongii and sequenced it for low depth whole genome sequence, then, a complete chloroplast genome of I. baodongii was assembled and annotated. This chloroplast genome has a circular structure of 145,494 bp in length with a GC content of 38.0%, comprising a large single copy (LSC) region of 91,860 bp, a pair of inverted repeat (IR) regions of 13,207 bp each, and a small single copy (SSC) region of 27,220 bp. 136 genes were annotated, including 84 protein-coding genes, 38 tRNA genes, and 8 rRNA genes. A maximum likelihood phylogeny tree was reconstructed after the sequences alignment, the result showed that I. baodongii formed a sister clade to the one clustered by I. sinensis, I. taiwanensis and I. orientalis. Although the chloroplast genome structure of I. baodongii is extremely similar to other species distributed in China, a well-supported phylogenetic relationship was reconstructed here, these results may provide new messages for further studies on phylogeny and evolution of vascular plant on the earth.

Cryptic species are commonly misidentified because of high morphological similarities to other species. One group of plants that may harbor large numbers of cryptic species is the quillworts (Isoëtes spp.), an ancient aquatic plant lineage. Although over 350 species of Isoëtes have been reported globally, only ten species have been recorded in China. The aim of this study is to better understand Isoëtes species diversity in China. For this purpose, we systematically explored the phylogeny and evolution of Isoëtes using complete chloroplast genome (plastome) data, spore morphology, chromosome number, genetic structure, and haplotypes of almost all Chinese Isoëtes populations. We identified three ploidy levels of Isoëtes in China—diploid (2n = 22), tetraploid (2n = 44), and hexaploid (2n = 66). We also found four megaspore and microspore ornamentation types in diploids, six in tetraploids, and three in hexaploids. Phylogenetic analyses confirmed that I. hypsophila as the ancestral group of the genus and revealed that Isoëtes diploids, tetraploids, and hexaploids do not form monophyletic clades. Most individual species possess a single genetic structure; however, several samples have conflicting positions on the phylogenetic tree based on SNPs and the tree based on plastome data. All 36 samples shared 22 haplotypes. Divergence time analysis showed that I. hypsophila diverged in the early Eocene (∼48.05 Ma), and most other Isoëtes species diverged 3–20 Ma. Additionally, different species of Isoëtes were found to inhabit different water systems and environments along the Yangtze River. These findings provide new insights into the relationships among Isoëtes species in China, where highly similar morphologic populations may harbor many cryptic species.

Isoetes orientalis is an endangered hexaploidy species of Isoetaceae in China and the complete chloroplast genome of this species has not been reported. In the present study, a complete chloroplast genome of Isoetes orientalis (Isoetaceae) was assembled and annotated. This chloroplast genome has a circular structure of 145,504 bp in length, comprising a pair of inverted repeat (IR) regions of 13,207 bp each, a large single-copy (LSC) region of 91,864 bp, and a small single-copy (SSC) region of 27,226 bp. The chloroplast genome contains 136 genes, including 84 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Phylogenetic analysis showed that I. orientalis was closely related to I. sinensis. These results provide additional resources for future studies on Isoetes from China and across the globe.

Abstract The quillwort Isoëtes cangae is a critically endangered species occurring in a single lake in Serra dos Carajás, Eastern Amazon. Low genetic diversity and small effective population sizes (Ne) are expected for narrow endemic species (NES). Conservation biology studies centered in a single species show some limitations, but they are still useful considering the limited time and resources available for protection of species at risk of extinction. Here, we evaluated the genetic diversity, population structure, Ne, and minimum viable population (MVP) of I. cangae to provide information for effective conservation programs. Our analyses were based on 55 individuals collected from the Amendoim Lake and 35,638 neutral SNPs. Our results indicated a single panmictic population, moderate levels of genetic diversity, and Ne in the order of thousands, contrasting the expected for NES. Negative FIS values were also found, suggesting that I. cangae is not under risk of inbreeding depression. Our findings imply that I. cangae contains enough genetic diversity to ensure evolutionary potential and that all individuals should be treated as one demographic unit. These results provide essential information to optimize ex situ conservation efforts and genetic diversity monitoring, which are currently applied to guide I. cangae conservation plans.

Umkomasiaceans are the most commonly recorded components of Gondwanan Triassic floras but they are not represented subsequently in that region. Their diversity appears to have substantially declined prior to the Rhaetian. Lower and Middle Jurassic floras in Australia and elsewhere in Gondwana are strikingly different from those of the Triassic, comprising ferns, lycopods, conifers, cycads, and bennettitaleans. The fern Cladophlebis Brongniart 1849 emend. Seward 1894 dominated coal-forming paludal environments during the Middle Jurassic in Queensland and it is one of the most commonly preserved plants in the Eastern Gondwanan Jurassic. Ginkgoaleans were present in Eastern Gondwana until near the end-Triassic but were absent below ca 60°S palaeolatitude during the Early and Middle Jurassic.

Las licofitas son las plantas vasculares más antiguas, comprenden a las Lycopodiaceae, Isoëtaceae y Selaginellaceae, constituyendo un grupo monofilético separado del resto de los helechos y plantas con semilla. Debido a su falta de relaciones coevolutivas con vectores bióticos, su monofilia y su patrón morfológico altamente estable son especialmente aptas para establecer patrones biogeográficos. Con este fin se construyeron los trazos individuales de los siete taxones encontrados. El trazo generalizado resultante indica que las sierras centrales de Argentina constituyen el extremo austral de un componente biótico neotropical, presentando una relación más estrecha con las Sierras Subandinas y los Andes bolivianos y peruanos que con el Chaco (donde están incluidas), bordeando la Zona de Transición Sudamericana, la que, caracterizada por ambientes áridos, constituye una barrera exitosa para las licofitas, separando la región austral de Argentina y Chile del resto de América del Sur.Lycophytes are the most ancient lineage of vascular plants and include Lycopodiaceae, Isoëtaceae and Selaginellaceae. They are a monophyletic group separated from ferns and seed plants and are especially useful for establishing biogeographic patterns, due to their lack of coevolutionary relationships with biotic vectors, their monophyly and their remarkable morphological conservatism. The individual tracks of the seven taxa of lycophytes from central Argentina were drawn and the generalized track obtained shows that the central hills of Argentina are the austral extreme of the distribution of a Neotropical biotic component, with closer relationships with the Subandean hills and the Bolivian and Peruvian Andes than to the Chaco (where they are included), bordering the South American Transition Zone. The latter is characterized by arid environments and is a successful barrier for Lycophytes, separating the austral region of Argentina and Chile from the other parts of South America.

Entre las familias de las Lycophyta, las Isoetaceae son de fácil reconocimiento por su hábito herbáceo que semeja a una gramínea y por su hábitat palustre o acuático. Esta familia es reconocida en el Perú con un solo género, Isoetes y 10 especies (Hickey, 1994; Smith et al., 2005). Todas las especies, con la excepción de dos, están representadas en la región Altoandina, asociadas a los lagos y bofedales. Cuatro especies son consideradas endemismos para la flora, dos de las cuales son propiamente acuáticas y una es facultativa. Solamente una de estas especies cuenta con poblaciones en un área protegida, si bien amenazas a sus poblaciones son alarmantes (León & Young, 1996).
