Abstract Microglia and the border-associated macrophages contribute to the modulation of cerebral blood flow, but the mechanisms have remained uncertain. Here, we show that microglia regulate the cerebral blood flow baseline and the responses to whisker stimulation or intra-cisternal magna injection of adenosine triphosphate, but not intra-cisternal magna injection of adenosine in mice model. Notably, microglia repopulation corrects these cerebral blood flow anomalies. The microglial-dependent regulation of cerebral blood flow requires the adenosine triphosphate-sensing P2RY12 receptor and ectonucleotidase CD39 that initiates the dephosphorylation of extracellular adenosine triphosphate into adenosine in both male and female mice. Pharmacological inhibition or CX3CR1-CreER-mediated deletion of CD39 mimics the cerebral blood flow anomalies in microglia-deficient mice and reduces the upsurges of extracellular adenosine following whisker stimulation. Together, these results suggest that the microglial CD39-initiated breakdown of extracellular adenosine triphosphate co-transmitter is an important step in neurovascular coupling and the regulation of cerebrovascular reactivity.

Background and aims – Ongoing studies on Colombian mistletoes revealed an undescribed species of Maracanthus (Loranthaceae) endemic to the Colombian Eastern Cordillera (Cordillera Oriental). This species is described and illustrated here, and a checklist of the species accepted in Maracanthus is also provided, including a taxonomic key, references to the known iconography of the genus, and taxonomic and geographic comments. Material and methods – This study is based on a literature review according to the International Code of Nomenclature for Algae, Fungi, and Plants, examination of herbarium specimens using physical specimens and digital images, and morphological studies based on samples from dried and rehydrated specimens. Key results – A detailed morphological analysis revealed that the new species is part of the little-known genus Maracanthus. The dichotomous key, as well as the taxonomic and geographic notes, allow users to recognise the five known species in this genus, showing that they occur in restricted distribution areas.

Abstract The present study was aimed at revealing the metabolic changes that occurred in the cellular lipid pattern of acute and chronic myeloid leukaemia cells following treatment with cannabidiol (CBD). CBD is a non-psychoactive compound present in Cannabis sativa L., which has shown an antiproliferative action in these type of cancer cells. CBD treatment reduced cell viability and initiated apoptotic and necrotic processes in both cancer cell lines in a time and dose-dependent manner, showing acute myeloid leukaemia (HL-60) cells greater sensitivity than chronic myeloid leukaemia ones (K-562), without differences in the activation of caspases 3/7. Then, control and treated cells of HL-60 and K-562 cell lines were studied through an untargeted lipidomic approach. The treatment was carried out with CBD at a concentration of 10 μM for HL-60 cells and 23 µM CBD for K-562 cells for 48 h. After the extraction of the lipid content from cell lysates, the samples were analysed by UHPLC-QTOF-MS/MS both in the positive and the negative ionization modes. The comprehensive characterization of cellular lipids unveiled several classes significantly affected by CBD treatment. Most of the differences correspond to phospholipids, including cardiolipins (CL), phosphatidylcholines (PC) and phosphosphingolipids (SM), and also triacylglycerols (TG), being many TG species increased after CBD treatment in the acute and chronic models, whereas phospholipids were found to be decreased. The results highlight some important lipid alterations related to CBD treatment, plausibly connected with different metabolic mechanisms involved in the process of cell death by apoptosis in cancer cell lines.

Abstract Symbolic anchoring is an important topic in robotics, as it enables robots to obtain symbolic knowledge from the perceptual information acquired through their sensors and maintain the link between that knowledge and the sensory data. In cognitive-based robots, this process of transforming sub-symbolic data generated by sensors to obtain and maintain symbolic knowledge is still an open problem. To address this issue, this paper presents SAILOR, a framework for symbolic anchoring integrated into ROS 2. SAILOR aims to maintain the link between symbolic data and perceptual data in real robots over time. It provides a semantic world modeling approach using two deep learning-based sub-symbolic robotic skills: object recognition and matching function. The object recognition skill allows the robot to recognize and identify objects in its environment, while the matching function enables the robot to decide if new perceptual data corresponds to existing symbolic data. This paper describes the proposed method and the development of the framework, as well as its integration in MERLIN2 (a hybrid cognitive architecture fully functional in robots running ROS 2) and the validation of SAILOR using public datasets and a real-world scenario.

Abstract Climate classification systems (CCSs) are emerging as essential tools in climate change science for mitigation and adaptation. However, their limitations are often misunderstood by non-specialists. This situation is especially acute when the CCSs are derived from Global Climate Model outputs (GCMs). We present a set of uncertainty maps of four widely used schemes -Whittaker-Ricklefs, Holdridge, Thornthwaite-Feddema and Köppen- for present (1980–2014) and future (2015–2100) climate based on 52 models from the Coupled Intercomparison Model Project Phase six (CMIP6). Together with the classification maps, the uncertainty maps provide essential guidance on where the models perform within limits, and where sources of error lie. We share a digital resource that can be readily and freely integrated into mitigation and adaptation studies and which is helpful for scientists and practitioners using climate classifications, minimizing the risk of pitfalls or unsubstantiated conclusions.

Abstract Sponges are key ecosystem engineers that shape, structure and enhance the biodiversity of marine benthic communities globally. Sponge aggregations and reefs are recognized as vulnerable marine ecosystems (or VMEs) due to their susceptibility to damage from bottom-contact fishing gears. Ensuring their long-term sustainability, preservation, and ecosystem functions requires the implementation of sound scientific conservation tools. Here, the genetic diversity, structure, and connectivity of the deep-sea glass sponge, Vazella pourtalesii (Schmidt, 1870), was investigated using 1,102 neutral SNPs obtained in RADseq. This species is distributed across the northwest Atlantic from Florida, USA to Nova Scotia, Canada and we sequenced samples covering this full distribution and provided evidence of strong genetic structure with two distinct clusters: Florida together with the Carolina Shelves and the Scotian Shelf. We estimated moderate levels of diversity with low migration across large distances (> 1000 kms) and high connectivity at smaller scales (

Abstract The screening of Parkinson’s Disease (PD) through speech is hindered by a notable lack of publicly available datasets in different languages. This fact limits the reproducibility and further exploration of existing research. To address this gap, this manuscript presents the NeuroVoz corpus consisting of 112 native Castilian-Spanish speakers, including 58 healthy controls and 54 individuals with PD, all recorded in ON state. The corpus showcases a diverse array of speech tasks: sustained vowels; diadochokinetic tests; 16 Listen-and-Repeat utterances; and spontaneous monologues. The dataset is also complemented with subjective assessments of voice quality performed by an expert according to the GRBAS scale (Grade/Roughness/Breathiness/Asthenia/Strain), as well as annotations with a thorough examination of phonation quality, intensity, speed, resonance, intelligibility, and prosody. The corpus offers a substantial resource for the exploration of the impact of PD on speech. This data set has already supported several studies, achieving a benchmark accuracy of 89% for the screening of PD. Despite these advances, the broader challenge of conducting a language-agnostic, cross-corpora analysis of Parkinsonian speech patterns remains open.

Abstract In this work, we present a novel modeling framework for understanding the dynamics of homeostatic regulation. Inspired by engineering control theory, this framework incorporates unique features of biological systems. First, biological variables often play physiological roles, and taking this functional context into consideration is essential to fully understand the goals and constraints of homeostatic regulation. Second, biological signals are not abstract variables, but rather material molecules that may undergo complex turnover processes of synthesis and degradation. We suggest that the particular nature of biological signals may condition the type of information they can convey, and their potential role in shaping the dynamics and the ultimate purpose of homeostatic systems. We show that the dynamic interplay between regulated variables and control signals is a key determinant of biological homeostasis, challenging the necessity and the convenience of strictly extrapolating concepts from engineering control theory in modeling the dynamics of homeostatic systems. This work provides a simple, unified framework for studying biological regulation and identifies general principles that transcend molecular details of particular homeostatic mechanisms. We show how this approach can be naturally applied to apparently different regulatory systems, contributing to a deeper understanding of homeostasis as a fundamental process in living systems.

The shipping industry faces the challenge of expanding its capabilities and mitigating its environmental impact. The utilization of adhesive bonding techniques has the potential to facilitate the construction of lighter ships with a reduced carbon footprint. In this work, a complete methodology has been established to study the properties of an adhesive bond between naval steel and the behavior of the bulk adhesive when introduced into seawater. Thermogravimetric analysis (TGA) has been used to determine the thermal properties. Single-lap-joint and tensile tests have been performed showing the negative influence of temperature on the mechanical properties. Seawater absorption has been studied by gravimetric tests. Differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), and mechanical bulk tests have been carried out before and after aging. The water absorption is Fickian, and when a constant load is applied, the absorption occurs faster and in greater quantity, decreasing the stiffness and strength. An increase in the glass transition temperature was observed in the absence of a load, with no discernible change upon the application of load condition. Also, aged specimens with constant load exhibited a reduction in water content. These phenomena may be attributed to several diffusion processes.

La investigación analiza la percepción de los consumidores sobre los canales cortos de comercialización de alimentos (CCCA) y los desafíos para productores. Utilizando un enfoque cualitativo, se realizaron 12 grupos de discusión en diversas poblaciones españolas, lo que permitió identificar ventajas de los CCCA, como la calidad del producto y el trato personalizado. No obstante, se detecta un desconocimiento significativo entre los consumidores, especialmente en áreas urbanas. Los productores resaltan la necesidad de apoyo público y estrategias efectivas para mejorar la visibilidad y superar los desafíos de la comercialización directa, destacando la capacidad de los CCCA para revitalizar el comercio.

EU's rural areas are an essential part of the European way of life, covering more than 75 % of the Union's territory and housing over 25 % of its population. However, they experience major imbalances compared to urban areas. Thus, it is important to analyse whether EU's rural development (RD) policy is effective in promoting the development of rural areas. This study uses a RD index for 9 Portuguese NUTS 3 regions, with data from 2011 and 2021. The results are then compared with the financial support delivered by EU funds and show their impact on RD in Portugal.

Abstract The three members of the genus capripoxvirus (CaPV), lumpy skin disease virus (LSDV), sheeppox virus (SPPV), and goatpox virus (GTPV) have common hosts and areas of overlapping geographical distribution with Rift Valley fever virus (RVFV). Hence, to ensure more cost-effective disease surveillance we developed and evaluated a Luminex assay for the simultaneous detection of antibodies against CaPV and RVFV in domestic ruminants. In cattle, the assay had a sensitivity (Se) of 98.7% and a specificity (Sp) of 98.3% in detecting anti-LSDV antibodies; both diagnostic parameters were 100% for the detection of anti-RVFV antibodies in this species. In sheep and goats, Se and Sp were 100% for the detection of anti-SPPV and anti-GTPV antibodies while they were 100% and 98.9%, respectively for the detection of anti-RVFV antibody. The assay did not cross react with anti-parapoxvirus antibodies of cattle, sheep, and goats. This multiplex serological assay offers a practical tool for accurate detection and monitoring of the immunological status of domestic ruminant populations against veterinary and socio-economically important capripox- and phleboviral infections, thus has the potential to aid in the strategic application of vaccination programmes.

Abstract We developed a 4-parameter clinical assay using Electric Field Induced Release and Measurement (EFIRM) technology to simultaneously assess SARS-CoV-2 RNA (vRNA), nucleocapsid antigen, host binding (BAb) and neutralizing antibody (NAb) levels from a drop of saliva with performance that equals or surpasses current EUA-approved tests. The vRNA and antigen assays achieved lower limit of detection (LOD) of 100 copies/reaction and 3.5 TCID₅₀/mL, respectively. The vRNA assay differentiated between acutely infected (n = 10) and infection-naïve patients (n = 33) with an AUC of 0.9818, sensitivity of 90%, and specificity of 100%. The antigen assay similarly differentiated these patient populations with an AUC of 1.000. The BAb assay detected BAbs with an LOD of 39 pg/mL and distinguished acutely infected (n = 35), vaccinated with prior infection (n = 13), and vaccinated infection-naïve patients (n = 13) from pre-pandemic (n = 81) with AUC of 0.9481, 1.000, and 0.9962, respectively. The NAb assay detected NAbs with a LOD of 31.6 Unit/mL and differentiated between COVID-19 recovered or vaccinated patients (n = 31) and pre-pandemic controls (n = 60) with an AUC 0.923, sensitivity of 87.10%, and specificity of 86.67%. Our combo assay represents a significant technological advancement to simultaneously address SARS-CoV-2 infection and immunity, and it lays the foundation for tackling potential future pandemics.

The electrorheological (ER) characterization of low-viscosity fluids is paramount for producing micro- and nanoscale products through electrohydrodynamic (EHD) techniques, such as EHD-jet printing, electrospray, and electrospinning. Key properties such as viscosity, surface tension, dielectric properties, electrical conductivity, and relaxation time significantly influence both the quality and properties of the final products and the efficiency of the industrial process. ER characterization is essential for studying the macroscopic effects of the interaction between these physicochemical properties under controlled flow kinematics. Researchers may face several technical challenges in performing rigorous ER characterization of moderate conductive fluids typically used in EHD processes. This characterization is crucial for formulating inks compatible with these processes and for understanding fluid dynamics in EHD processes to ensure stable printing conditions and achieve high-resolution, accurate prints. This work highlights the inherent limitations of current ER cells and proposes methodologies to mitigate their impact on measurement accuracy. Furthermore, we propose the use of microfluidic devices as a solution for the ER characterization of moderate conductive fluids.

Abstract Introduction In contrast with patients receiving therapy for retinal disease during clinical trials, those treated in routine clinical practice experience various challenges (including administrative, clinic, social, and patient-related factors) that can often result in high patient and clinic burden, and contribute to suboptimal visual outcomes. The objective of this study was to understand the challenges associated with clinical management of diabetic macular edema from the perspectives of patients, healthcare providers, and clinic staff, and identify opportunities to improve eye care for people with diabetes. Methods We conducted a survey of patients with diabetic macular edema, providers, and clinic staff in 78 clinics across 24 countries on six continents, representing a diverse range of individuals, healthcare systems, settings, and reimbursement models. Surveys comprised a series of single- and multiple-response questions completed anonymously. Data gathered included patient personal characteristics, challenges with appointment attendance, treatment experiences, and opportunities to improve support. Provider and clinic staff surveys asked similar questions about their perspectives; and clinic characteristics were also captured. Results Overall, 5681 surveys were gathered: 3752 from patients with diabetic macular edema, 680 from providers, and 1249 from clinic staff. Too many appointments, too short treatment intervals, difficulties in traveling to the clinic or arranging adequate support to travel, out-of-pocket costs, office/parking fees, and long waiting times were noted by all as contributing to increase the burden on the patient and caregiver. Patients generally desired more in-depth discussions with their provider, which would help with information exchange and better expectation-setting. Conclusions The wealth of systematic data generated by this global survey highlights the breadth and scale of challenges associated with the clinical management of patients with diabetic macular edema. Addressing the opportunities for improvement raised by patients, providers, and clinic staff could increase patient adherence to treatment, reduce appointment burden, and improve clinic capacity.

Abstract Introduction Neovascular age-related macular degeneration is a global public-health concern, associated with a considerable burden to individuals, healthcare systems, and society. The objective of this study was to understand different perspectives on the challenges associated with the clinical management of neovascular age-related macular degeneration, which could elucidate measures to comprehensively improve clinical care and outcomes. Methods A survey was carried out of patients with neovascular age-related macular degeneration, their providers, and clinic staff in 77 clinics across 24 countries on six continents, from a diverse range of healthcare systems, settings, and reimbursement models. Surveys comprised a series of single/multiple-response questions completed anonymously. Data gathered included patient personal characteristics, appointment attendance challenges, treatment experiences, and opportunities to improve support. Provider and clinic staff surveys asked similar questions about their perspectives; clinic characteristics were also captured. Results There were 6425 responses; 4558 patients with neovascular age-related macular degeneration, 659 providers, and 1208 clinic staff. Challenges identified included concern about patient burden to family/friends, high frequency of treatment, difficulties in traveling to appointments, long waiting times, and insufficient comprehension of neovascular age-related macular degeneration. Participants identified logistical (improved financial assistance with treatment and out-of-pocket costs, and appointment reminders), operational (addressing clinic set up to reduce waiting times and improving the amount of time providers spend with patients), and educational (improving quality and provision of patient information and expectation-setting) opportunities to improve care. Conclusions The wealth of data generated by this global survey highlights the breadth of challenges associated with clinical management of patients with neovascular age-related macular degeneration. Addressing the opportunities raised could improve patient adherence to treatment and potentially outcomes, reduce appointment burden, and increase clinic capacity.

Abstract Objective This study investigated the effectiveness and tolerability of brivaracetam (BRV) monotherapy in a large series of patients with epilepsy. Method This was a multicenter, retrospective, observational, non‐interventional study in 24 hospitals across Spain. Patients aged ≥18 years who started on BRV monotherapy, either as first‐line or following conversion, at least 1 year before database closure were included. Patients were evaluated at baseline and at 3, 6 and 12 months after initiation of BRV monotherapy, in accordance with usual clinical practice at these centers. Data were collected retrospectively from patients' individual charts by participating physicians. The primary effectiveness and safety endpoints were the percentage of seizure‐free patients 1 year after initiation of BRV monotherapy and the proportion of patients reporting adverse events (AEs) over the complete follow‐up period. Retention rates and subpopulation analysis (levetiracetam switchers, elderly and different etiologies) were also investigated. Results A total of 276 patients were included (48 with BRV as first‐line monotherapy and 228 who converted to BRV monotherapy). The overall retention rate in monotherapy at 12 months was 89.9% (87.5% for first‐line monotherapy group; 90.4% for conversion‐to‐monotherapy group). Seizure‐freedom rates at 12 months were 77.8% (75% for first‐line monotherapy group; 78.4% for conversion‐to‐monotherapy group). AEs occurred in 39.5% of patients at 12 months (35.4% for first‐line monotherapy group; 40.4% for conversion‐to‐monotherapy group). Most AEs were mild‐to‐moderate. The most frequent AEs were irritability (12.3%) and dizziness (10.1%). The most frequent AEs leading to BRV withdrawal were dizziness (1.8%) and memory problems (1.4%). Similar outcomes in terms of effectiveness and tolerability of BRV monotherapy were observed in patients switching from levetiracetam, those with different epilepsy etiologies, and elderly patients. Significance BRV was effective and well tolerated both as first‐line monotherapy and following conversion to monotherapy in a real‐world setting of patients with epilepsy. Plain Language Summary The goal of the medical treatment of epilepsy is to ensure best possible patient quality of life, by maximizing seizure control and minimizing medication toxicity. Brivaracetam (BRV) is a new‐generation epilepsy treatment that is well tolerated by patients. In our study, monotherapy with BRV reduced seizures in patients who had not received other treatments and in patients who switched from a previous treatment to BRV monotherapy. BRV was well tolerated and also effective in sensitive patients (i.e., the elderly and those who had epilepsy caused by a brain tumor or a brain injury).

Abstract The Canary Islands are an archipelago of significant natural value, with a large variety of endemic species living in them, making essential the study of the effects of climate change. This research is based on the use of dynamic techniques of climatic regionalisation to obtain climate projections that contribute to the conservation of ecosystems in the Canary Islands. In this work, climate projections derived from WRF simulations with a spatial resolution of 3 km were used to obtain a novel dataset of 53 bioclimatic indicators, called BICI–ULL (Bioclimatic Indicators in the Canary Islands, University of La Laguna). The regional climate simulations were driven by three CMIP5 models (GFDL–ESM2M, IPSL–CM5A–MR, and MIROC–ESM) over three periods (1980–2009, 2030–2059, 2070–2099) and two emission scenarios (representative concentration pathway 4.5 and 8.5), to obtain, among others, the standard climatic variables used to generate the bioclimatic indicators: temperature, precipitation, solar radiation, humidity and wind speed.

Abstract Dung serves as a critical resource for diverse organisms, including dung-inhabiting fungi, which play a key role in nutrient cycling. In this study, we examined the decomposition rates and half-lives of dung from ruminant and monogastric herbivores in a microcosm experiment, assessing the impact of autoclaving (fungal exclusion) on decomposition dynamics. Over six months, autoclaved dung decomposed more slowly, retaining greater biomass and highlighting the fungi’s role in matter cycling. Decomposition followed a Gaussian linear model, with constants k ranging from 0.02 to 0.03 and half-lives of 19–23 days. Nutrient mineralization varied significantly between the start and end of the experiment, underscoring the contribution of the fungi to nutrient release. Our findings emphasize the ecological importance of dung-inhabiting fungi and suggest areas for future research on factors influencing dung decomposition in terrestrial ecosystems.

Abstract Background The gut microbiome has been reported to induce epigenetic modifications in the host, which may be involved in the pathophysiology of metabolic diseases. Objective To evaluate the potential interactions between the gut microbiome and DNA methylome in subjects with different metabolic characteristics. Methods Sixty-four participants with different metabolic characteristics (i.e., participants without obesity -healthy controls-, and participants with obesity and normal insulin sensitivity/insulin resistance/ type 2 diabetes-T2DM-) were included in this study. A machine learning approach was performed in order to identify distinctive patterns in three omics (gut microbiome, blood DNA methylome, and visceral adipose tissue-VAT- DNA methylome) according to the different study groups. Results Robust distinctive distribution patterns of the three different omics were found between healthy controls and patients with obesity; participants with and without T2DM, and patients with obesity with and without insulin resistance. Importantly, strong correlations between the gut microbiome (including Odoribacteriaceae and Christensenllaceae families) and both blood and VAT DNA methylome were found. Moreover, in the entire study population, three main bacterial genera (Sutterella, Collinsella and Eubacterium) were related to the epigenetic regulation of different genes involved in distinct processes related to cellular metabolism and metabolic diseases, including small ubiquitin-related modifier (SUMO) transferase activity or lipid binding. Conclusion We show that distinctive interactions between the gut microbiome and DNA methylome may occur in subjects with different metabolic characteristics. Further research is needed to elucidate the potential role of these interactions in the pathophysiology of obesity and related comorbidities.

Abstract Interleukins are central in the modulation of immune responses. This narrative review aims to summarize the growing evidence on their significance as key drivers of numerous cutaneous diseases with a special focus in some of the more prevalent chronic inflammatory dermatologic diseases such as psoriasis, atopic dermatitis, allergic contact dermatitis, urticaria, and hidradenitis suppurativa. Additionally, we discuss their relevance in the recent developments in targeted therapies that have significantly transformed the management of these skin conditions. To this end, we have conducted a comprehensive search through the Cochrane Library and Database of Systematic Reviews and the MEDLINE search engine, and we have summarized the available clinical evidence considering up to 466 records including meta‐analyses, systematic reviews, reviews and clinical trials. Ultimately, this review intents to foster both dermatologist and non‐dermatologist physicians' understanding of the immunology behind the clinical manifestations of some of the most common inflammatory skin diseases and engage with the novel therapeutic approaches by providing accessible insights into the implications of interleukin pathways dysregulation.

Neurological disorders such as Autism Spectrum Disorder (ASD), Schizophrenia (SCH), Bipolar Disorder (BD), and Major Depressive Disorder (MDD) affect millions of people worldwide, yet their molecular mechanisms remain poorly understood. This study describes the application of the Comparative Analysis of Shapley values (CASh) to transcriptomic data from nine datasets associated with these complex disorders, demonstrating its effectiveness in identifying differentially expressed genes (DEGs). CASh, which combines Game Theory with Bootstrap resampling, offers a robust alternative to traditional statistical methods by assessing the contribution of each gene in the broader context of the complete dataset. Unlike conventional approaches, CASh is highly effective at detecting subtle but meaningful molecular patterns that are often missed. These findings highlight the potential of CASh to enhance the precision of transcriptomic analysis, providing a deeper understanding of the molecular mechanisms underlying these disorders and establishing a solid basis to improve diagnostic techniques and developing more targeted therapeutic interventions.

In this work, we present the exact equations for the back and effective focal lengths of a plano-concave thick lens. These equations are derived through a detailed ray-tracing approach, calculating the meridional ray’s path parallel to the optical axis while taking into account lens thickness, refractive index, and surface curvature. Our analysis bridges the gap between paraxial approximations and exact ray behavior, highlighting how traditional models (e.g., thin-lens equations) can be extended to thick lenses. Additionally, we review recent advancements in negative refraction and surface phase manipulation techniques to demonstrate how our equations compare to modern numerical methods and metacoating-enhanced designs. This work provides a comprehensive and exact framework for understanding the optical properties of plano-concave thick lenses, offering new insights into their application in precision optics.

Abstract Atmospheric new particle formation (NPF) is a naturally occurring phenomenon, during which high concentrations of sub-10 nm particles are created through gas to particle conversion. The NPF is observed in multiple environments around the world. Although it has observable influence onto annual total and ultrafine particle number concentrations (PNC and UFP, respectively), only limited epidemiological studies have investigated whether these particles are associated with adverse health effects. One plausible reason for this limitation may be related to the absence of NPF identifiers available in UFP and PNC data sets. Until recently, the regional NPF events were usually identified manually from particle number size distribution contour plots. Identification of NPF across multi-annual and multiple station data sets remained a tedious task. In this work, we introduce a regional NPF identifier, created using an automated, machine learning based algorithm. The regional NPF event tag was created for 65 measurement sites globally, covering the period from 1996 to 2023. The discussed data set can be used in future studies related to regional NPF.

Abstract The wind energy requirements for high temporal and spatial observations, unavailable in many countries, are met by reanalysed wind data, which are increasingly used in wind power assessments. However, these data also include unrealistic features, making their validation compulsory. The validation analysis of ERA5 and MERRA-2 wind data, two of the most successful wind reanalyses, with observations near Morocco’s coast, highlighted the existence of bias in both datasets, and other shortcomings. The study was conducted at eight selected sites in the period 2011-2020 using linear and probability density function based statistical tests. The local ERA5-BA and MERRA-2 BA velocity fields were created using Bias Adjustment (BA) Techniques. Only part of the observed and modelled datasets (training period) were used to identify the transfer function. The BA fields were verified against observations during the remaining period (verification period). They outperformed their respective non-adjusted fields and also the WFDE5 (the global bias-corrected ERA5 wind field) in the wind potential assessments, represented by capacity factor and low-wind day. At every location, the relative errors in the latter’s estimates using ERA5-BA were lowered to less than 10 %, and the same was true for MERRA-2-BA. Thus, an observational dataset of reduced length can help overcome some reanalysed wind data limitations for wind power estimations. Additionally, this methodology can be applied to the correction of each reanalysis update.

Abstract Impulsivity is an important determinant of human behaviour, affecting self-control, reasonable thinking and food choices. Recent evidence suggests a role for gut microbiota in human behaviour, but the relationship between gut microbiota and impulsive behaviours remains largely unexplored. To address this knowledge gap, the present study aims to explore the associations between faecal microbiota composition with trait and behavioural impulsivity, in a subcohort of the PREDIMED-Plus trial, including older adults presenting overweight/obesity. Fecal samples (n = 231) were profiled for their microbiota composition using 16 S rRNA amplicon sequencing and impulsivity was determined through four different assessments. Adherence to different dietary patterns was estimated through questionnaires. Beta diversity analyses showed a significant association with the Conner’s Performance Test (CPT) in multivariate-adjusted models, and, in total, 13 bacterial genera associated with CPT. Erysipelotrichaceae UCG 003 showed the highest association with CPT and known butyrate producers such as Butyricicoccus spp., Roseburia spp., and Eubacterium hallii were among the identified bacteria. The bacteria Lachnospiraceae UCG 001, Anaerostipes and Blautia were associated with CPT and also the adherence to healthy and unhealthy plant-based diets. In addition, functional analysis showed a significant negative association between the CPT and the glucuronate and galacturonate metabolic pathways. From the other impulsivity assessments, two more associations were identified, for the genus Phascolarctobacterium with the Stroop test, and the genus Lachnospiraceae GAG 54 with the positive urgency subscore of UPPS-P Impulsive Behaviour Scale. Overall, our findings suggest potential links between the faecal microbiota composition and function with behavioural impulsive inattention as determined by the CPT.

Abstract To investigate the effect of physical activity (PA) (both general and its type) on self-esteem during and after cancer treatment. A systematic search was conducted across PubMed, Web of Science, Scopus, SPORTDiscuss, and PsycINFO from their inception to February 2024. The systematic review included 32 studies, with 15 studies (13 RCT and 2 quasi-experimental) and 3604 participants (66.7% female) included in the meta-analysis involving controlled trials using a control group and at least one PA intervention group. The study was registered in PROSPERO (CRD42022309771). Abstract Background Vasculogenic therapies explored for the treatment of peripheral artery disease (PAD) have encountered minimal success in clinical trials. Addressing this, B55α, an isoform of protein phosphatase 2A (PP2A), emerges as pivotal in vessel remodeling through activation of hypoxia-inducible factor 1α (HIF-1α). This study delves into the pharmacological profile of VCE-004.8 (Etrinabdione) and evaluates its efficacy in a preclinical model of critical limb ischemia, with a focus on its potential as a PP2A/B55α activator to induce angiogenesis and arteriogenesis. Methods Vascular endothelial cells were used for in vitro experiments. Aorta ring assay was performed to explore sprouting activity. Matrigel plug-in assay was used to assess the angiogenic potential. Critical limb ischemia (CLI) in mice was induced by double ligation in the femoral arteria. Endothelial vascular and fibrotic biomarkers were studied by immunohistochemistry and qPCR. Arteriogenesis was investigated by microvascular casting and micro-CT. Proteomic analysis in vascular tissues was analyzed by LC–MS/MS. Ex-vivo expression of B55α and biomarkers were investigated in artery samples from PAD patients. Results VCE-004.8 exhibited the ability to induce B55α expression and activate the intersecting pathways B55α/AMPK/Sirtuin 1/eNOS and B55α/PHD2/HIF-1α. VCE-004.8 prevented OxLDL and H2O2-induced cytotoxicity, senescence, and inflammation in endothelial cells. Oral VCE-004.8 increased aorta sprouting in vitro and angiogenesis in vivo. In CLI mice VCE-004.8 improved collateral vessel formation and induced endothelial cells proliferation, angiogenic gene expression and prevented fibrosis. The expression of B55α, Caveolin 1 and Sirtuin-1 is reduced in arteries from CLI mice and PAD patient, and the expression of these markers was restored in mice treated with VCE-004.8. Conclusions The findings presented in this study indicate that Etrinabdione holds promise in mitigating endothelial cell damage and senescence, while concurrently fostering arteriogenesis and angiogenesis. These observations position Etrinabdione as a compelling candidate for the treatment of PAD, and potentially other cardiovascular disorders.

In this research work, the feasibility of using fusel oil, a by-product of the sugar–alcohol industry, as an LVLC solvent in blends with straight vegetable oils (SVOs) and diesel was investigated. Concretely, diesel/fusel oil/sunflower oil (D/FO/SO) and diesel/fusel oil/castor oil (D/FO/CO) triple blends were prepared and characterized by measuring the most important physicochemical properties, i.e., viscosity, density, cold flow properties, flash point and cetane number. An appreciable improvement in cold flow values has been achieved with triple blends, without compromising properties such as calorific value and cetane number. Likewise, the triple blends meet the viscosity and density requirements specified by the European quality standard EN 14214 and the American standard ASTM D6751. After characterization, the triple blends were used on a diesel engine, evaluating different parameters such as power output, opacity, exhaust emissions (CO and NOx) and consumption at different engine loads. The results indicate that as the biofuel content in the blend increases, engine power decreases while fuel consumption rises. Nevertheless, the values obtained with D/FO/CO are better than those for D/FO/SO and are also very similar to those of fossil diesel. Regarding opacity values and NOx emissions obtained with the utilization of the triple blends, they are lower than those produced by diesel. However, in the case of CO emissions, it depends on the type of oil used, with the samples prepared with castor oil exhibiting the best results.

The World Health Organization (WHO) cancer agency predicts that more than 35 million cases of cancer will be experienced in 2050, a 77% increase over the 2022 estimate. Currently, the main cancers diagnosed are breast, lung, and colorectal. There is no standardized tool for cancer diagnoses; initially, clinical procedures are guided by the patient symptoms and usually involve biochemical blood tests, imaging, and biopsy. Label-free non-linear optical approaches are promising tools for tumor imaging, due to their inherent non-invasive biosafe contrast mechanisms and the ability to monitor collagen-related disorders, and biochemical and metabolic changes during cancer progression. In this review, the main non-linear microscopy techniques are discussed, according to three main contrast mechanisms: biochemical, metabolic, and structural imaging.

The enhancement of cultural heritage for tourism can greatly benefit from the opportunities offered by Information and Communication Technologies (ICTs), promoting greater interaction and integration between all stakeholders and service providers. In recent years, ICT applications and platforms have tended to provide a platform to translate the strategic goals and performance metrics of cultural heritage tourism into comprehensive services tailored to stakeholders’ needs by participating stakeholders. A successful co-designed platform integrates explicit services—requiring specialized software tools—and implicit services, facilitating evaluation processes. This adaptability ensures continuous improvement and alignment with evolving strategies and business objectives. As part of the European project, the development of the TExTOUR open ICT platform marks a significant advancement in leveraging technology within cultural tourism. It exemplifies the creation of a sustainable and adaptable framework, co-designed specifically to support and amplify the strategic objectives of cultural heritage tourism. This approach not only fosters better engagement and management within the sector but also sets a new standard for innovation in cultural tourism practices.

Abstract River networks contribute disproportionately to the global carbon cycle. However, global estimates of carbon emissions from inland waters are based on perennial rivers, even though more than half of the world's river length is prone to drying. We quantified CO2 and CH4 emissions from flowing water and dry riverbeds across six European drying river networks (DRNs, 120 reaches) and three seasons and identified drivers of emissions using local and regional variables. Drivers of emissions from flowing water differed between perennial and non‐perennial reaches, both CO2 and CH4 emissions were controlled partly by the annual drying severity, reflecting a drying legacy effect. Upscaled CO2 emissions for the six DRNs at the annual scale revealed that dry riverbeds contributed up to 77% of the annual emissions, calling for an urgent need to include non‐perennial rivers in global estimates of greenhouse gas emissions.

Abstract Background Sarcopenia and low areal bone mineral density (aBMD) are prevalent musculoskeletal complications after paediatric cancer treatment. However, their relationship has not been examined in young paediatric cancers survivors. This study aimed to evaluate aBMD differences according to sarcopenia status and the risk of low aBMD Z‐score in young paediatric cancer survivors with sarcopenia confirmed/probable. Methods This cross‐sectional study included 116 paediatric cancer survivors (12.1 ± 3.3 years old; 42.2% female). Handgrip strength was used to assessed muscle strength. Dual‐energy X‐ray absorptiometry estimated aBMD (g/cm2) and appendicular lean mass index (ALMI, kg/m2). ‘No sarcopenia’ was defined when muscle strength was >decile 2. ‘Sarcopenia probable’ was defined when muscle strength was ≤ decile 2 and ALMI Z‐score was > −1.5 standard deviation (SD). ‘Sarcopenia confirmed’ was defined when muscle strength was ≤ decile 2 and ALMI Z‐score ≤ −1.5 SD. Analysis of covariance and logistic regression, adjusted for time from treatment completion, radiotherapy exposure, calcium intake, and physical activity, was used to evaluate aBMD and estimate the odds ratios (ORs) of low aBMD (aBMD Z‐score < −1.0). ABSTRACT Introduction: Although nerves and vessels of the penis play important role in erection, there are few studies on their development in human fetus. Therefore, the objective of the present study is to analyze, quantitatively, in the corpora cavernosa and corpus spongiosum, the development of the nerves and vessels in the fetal penis at different gestational ages. Material and Methods: Fifty-six fresh, macroscopically normal human fetuses aged from 13 to 36 weeks post-conception (WPC) were used. Gestational age was determined by the foot length criterion. Penises were immediately fixed in 10% formalin, and routinely processed for paraffin embedding, after which tissue sections from the mid-shaft were obtained. We used immunohistochemical staining to analyze the nerves and vessels in the corpus cavernous and in the corpus spongiosum. These elements were identified and quantified as percentage by using the Image-J software. Results: The quantitative analysis showed that the percentage of nerves varied from 3.03% to 20.35% in the corpora cavernosa and from 1.89% to 23.88% in the corpus spongiosum. Abstract Background Benzodiazepines are frequently prescribed drugs; however, their prolonged use can lead to tolerance, dependence, and other adverse effects. Despite these risks, long-term use remains common, presenting a public health concern. This study aims to explore the beliefs and opinions held by the public regarding benzodiazepines, as understanding these perspectives may provide insights into their usage patterns. Methods We collected public tweets published in English between January 1, 2019, and October 31, 2020, that mentioned benzodiazepines. The content of each tweet and the characteristics of the users were analyzed using a mixed-method approach, including manual analysis and semi-supervised machine learning. Results Over half of the Twitter users highlighted the efficacy of benzodiazepines, with minimal discussion of their side effects. The most active participants in these conversations were patients and their families, with health professionals and institutions being notably absent. Additionally, the drugs most frequently mentioned corresponded with those most commonly prescribed by healthcare professionals. Conclusions Social media platforms offer valuable insights into users’ experiences and opinions regarding medications. Notably, the sentiment towards benzodiazepines is predominantly positive, with users viewing them as effective while rarely mentioning side effects. This analysis underscores the need to educate physicians, patients, and their families about the potential risks associated with benzodiazepine use and to promote clinical guidelines that support the proper management of these medications. [Purpose] This paper seeks to develop, from the current doctrine and regulations, some guidelines for the debate that the Law will have to give shortly, about a new type of personality and its attributes those humanoid robots that are capable of questioning their initial programming and take actions on your own. [Methodology] It is a work developed through a legal-hermeneutical and documentary analysis methodology. The authors used legal doctrine as sources, especially books and book chapters, research results, as well as articles in scientific journals evaluated by peers and indexed in legal databases. To develop the objective of the paper, the authors outlined several stages, first, the search and recovery of information, second a critical analysis, and finally the interpretation of all of the above. [Findings] The authors present their results, on the one hand, with an analysis of the personality of humanoid robots, and on the other, with some reflections on their attributes. The paper presents a critical study on the legal situation of the regulation of the autonomy of the will; which must be understood as the materialized faculty of self-determination that these robots could have in making their own decisions and concerning the rights, obligations, and responsibilities that the Law decides to recognize. All of the above, from artificial intelligence systems that are not subordinated to their initial programming, or the immediate and permanent control of a natural person.

Abstract Current influenza vaccines are strain-specific and require frequent updates to combat new strains, making a broadly protective influenza vaccine (BPIV) highly desirable. A promising strategy is to induce T-cell responses against internal proteins conserved across influenza strains. In this study, pH1N1 pre-exposed pigs were immunized by aerosol using viral vectored vaccines (ChAdOx2 and MVA) expressing matrix (M1) and nucleoprotein (NP). Following H3N2 challenge, all immunizations (M1, NP or NPM1) reduced lung pathology, but M1 alone offered the greatest protection. NP or NPM1 immunization induced both T-cell and antibody responses. M1 immunization generated no detectable antibodies but elicited M1-specific T-cell responses, suggesting T cell-mediated protection. Additionally, a single aerosol immunization with the ChAdOx vaccine encoding M1, NP and neuraminidase reduced lung pathology. These findings provide insights into BPIV development using a relevant large natural host, the pig.

Abstract In the era of renewed space exploration, comprehending the effects of the space environment on human health, particularly for deep space missions, is crucial. While extensive research exists on the impacts of spaceflight, there is a gap regarding female reproductive risks. We hypothesize that space stressors could have enduring effects on female health, potentially increasing risks for future pregnancies upon return to Earth, particularly related to small-for-gestational-age (SGA) fetuses. To address this, we identify a shared microRNA (miRNA) signature between SGA and the space environment, conserved across humans and mice. These miRNAs target genes and pathways relevant to diseases and development. Employing a machine learning approach, we identify potential FDA-approved drugs to mitigate these risks, including estrogen and progesterone receptor antagonists, vitamin D receptor antagonists, and DNA polymerase inhibitors. This study underscores potential pregnancy-related health risks for female astronauts and proposes pharmaceutical interventions to counteract the impact of space travel on female health.

Abstract MicroRNAs (miRNAs) have been implicated in human disorders, from cancers to infectious diseases. Targeting miRNAs or their target genes with small molecules offers opportunities to modulate dysregulated cellular processes linked to diseases. Yet, predicting small molecules associated with miRNAs remains challenging due to the small size of small molecule-miRNA datasets. Herein, we develop a generalized deep learning framework, sChemNET, for predicting small molecules affecting miRNA bioactivity based on chemical structure and sequence information. sChemNET overcomes the limitation of sparse chemical information by an objective function that allows the neural network to learn chemical space from a large body of chemical structures yet unknown to affect miRNAs. We experimentally validated small molecules predicted to act on miR-451 or its targets and tested their role in erythrocyte maturation during zebrafish embryogenesis. We also tested small molecules targeting the miR-181 network and other miRNAs using in-vitro and in-vivo experiments. We demonstrate that our machine-learning framework can predict bioactive small molecules targeting miRNAs or their targets in humans and other mammalian organisms.

Abstract The development of computational tools for the systematic prediction of metabolic vulnerabilities of cancer cells constitutes a central question in systems biology. Here, we present gmctool, a freely accessible online tool that allows us to accomplish this task in a simple, efficient and intuitive environment. gmctool exploits the concept of genetic Minimal Cut Sets (gMCSs), a theoretical approach to synthetic lethality based on genome-scale metabolic networks, including a unique database of synthetic lethals computed from Human1, the most recent metabolic reconstruction of human cells. gmctool introduces qualitative and quantitative improvements over our previously developed algorithms to predict, visualize and analyze metabolic vulnerabilities in cancer, demonstrating a superior performance than competing algorithms. A detailed illustration of gmctool is presented for multiple myeloma (MM), an incurable hematological malignancy. We provide in vitro experimental evidence for the essentiality of CTPS1 (CTPS synthase) and UAP1 (UDP-N-Acetylglucosamine Pyrophosphorylase 1) in specific MM patient subgroups.

ABSTRACT Purpose: The gubernaculum seems to be the most important anatomical structure in the testicular migration process. The objective of this paper is to review current literature regarding the role of gubernaculum testis nerves in testicular migration. We conducted a comprehensive literature review about the gubernaculum testis innervation. A PubMed database search was performed in April 2024, focusing on gubernaculum testis and cryptorchidism and genitofemoral nerve (GFN) and calcitonin gene-related peptide (CGRP) gene. The gubernaculum has its own nerve supply, the GFN, descending on the anteromedial surface of the psoas muscle from L1-L2 segments. The second phase of testicular descent is regulated by androgens and CGRP, released from the sensory nucleus of the GFN. The GFN doesn't directly play a role in testicular migration but there is a theory that shows a regulatory function of this nerve in hormonal action during this process. The gubernaculum testis has important structural alterations during the testicular migration and the genitofemoral nerve and CGRP gene are of great importance in this process. The genitofemoral nerve provides motor innervation to the cremaster muscle and gubernaculum, which helps regulate the position of the testes within the scrotum.

Background/Objective: Stiff person syndrome (SPS) and type 1 diabetes (T1D) are heterogeneous disorders characterized by antibodies (Abs) against glutamic acid decarboxylase (GAD). Case Report: We describe 2 patients with T1D and autoimmune thyroid disease who presented with muscle rigidity and intermittent spasms that affected gait and with elevated circulating anti-GAD titers. Classic SPS and stiff limb syndrome were diagnosed, respectively. Muscle spasms resolved with immunotherapy and muscle relaxants in both patients, and the ability to ambulate without an assistive device was restored in 1 patient. Patients also had brittle diabetes with high glycemic variability, requiring the use of flash glucose monitoring with an insulin pump and a second-generation basal insulin analog, respectively. Discussion: GAD Ab–associated syndromes include SPS, T1D, and other endocrinopathies. The clinical heterogeneity implies variable susceptibility of γ-aminobutyric acid-ergic neurons and pancreatic beta cells to anti-GAD or other autoantibodies. Conclusion: Our case series represent the heterogeneity in natural history, clinical course, and response to therapy in patients with Abs against GAD-spectrum disorders.

Abstract Neuroligin-2 (Nlgn2) is a key synaptic adhesion protein at virtually all GABAergic synapses, which recruits GABAARs by promoting assembly of the postsynaptic gephyrin scaffold. Intriguingly, loss of Nlgn2 differentially affects subsets of GABAergic synapses, indicating that synapse-specific interactors and redundancies define its function, but the nature of these interactions remain poorly understood. Here we investigated how Nlgn2 function in hippocampal area CA1 is modulated by two proposed interaction partners, MDGA1 and MDGA2. We show that loss of MDGA1 expression, but not heterozygous deletion of MDGA2, ameliorates the abnormal cytosolic gephyrin aggregation, the reduction in inhibitory synaptic transmission and the exacerbated anxiety-related behaviour characterizing Nlgn2 knockout (KO) mice. Additionally, combined Nlgn2 and MDGA1 deletion causes an exacerbated layer-specific loss of gephyrin puncta. Given that both Nlgn2 and the MDGA1 have been correlated with many psychiatric disorders, our data support the notion that cytosolic gephyrin aggregation may represent an interesting target for novel therapeutic strategies.

This research explores the efficacy of the YOLOR (You Only Learn One Representation) algorithm integrated with the Deep Sort algorithm for real-time vehicle detection, classification, and counting in Morelia, Mexico. The study aims to enhance traffic monitoring and management by leveraging advanced deep learning techniques. The methodology involves deploying the YOLOR model at six key monitoring stations, with varying confidence levels and pre-trained weights, to evaluate its performance across diverse traffic conditions. The results demonstrate that the model is effective compared to other approaches in classifying multiple vehicle types. The combination of YOLOR and Deep Sort proves effective in tracking vehicles and distinguishing between different types, providing valuable data for optimizing traffic flow and infrastructure planning. This innovative approach offers a scalable and precise solution for intelligent traffic management, setting new methodologies for urban traffic monitoring systems.

Abstract Background The handstand is an essential skill in acrobatic sports. This skill requires the athlete to maintain an inverted upright stance with only the hands supported, which requires a great effort of muscular coordination and motor control. Several factors influence the ability to control the posture, including fatigue, which is a bit studied constraint of handstand performance. Research question With the aim to find out whether variability in movement control can be an indicator of fatigue, the present study was carried out. Method Fourteen male acrobatic gymnasts were required to perform handstands. The time series for analyzing variability were capturing using Force Platforms, which is a traditional laboratory instrument, and Inertial Measurement Units (IMU), which is a more recent and less widely used, but more accessible tool. For this purpose, an analysis of the amount of variability was carried out, using the standard deviation. And analysis of the structure of variability (or complexity), using Detrended Fluctuation Analysis (DFA) and Fuzzy Entropy (FuEn). Results Our results reveal that fatigue causes significant increases in the amount of variability in the medio-lateral axis on the force platform, and in the IMU located in the area of the L5 vertebra. These changes are accompanied by increased auto-correlation in the medio-lateral axis of the force platform, and more unpredictable behavior in the L5 IMU.

Abstract Background Polymorphisms are common in nature, but they are rarely shared among closely related species. Polymorphisms could originate through convergence, ancestral polymorphism, or introgression. Although shared neutral genomic variation across species is commonplace, few examples of shared functional traits exist. The blue-orange petal color polymorphisms in two closely related species, Lysimachia monelli and L. arvensis were investigated with UV-vis reflectance spectra, flavonoid biochemistry, and transcriptome comparisons followed by climate niche analysis. Results Similar color morphs between species have nearly identical reflectance spectra, flavonoid biochemistry, and ABP gene expression patterns. Transcriptome comparisons reveal two orange-specific genes directly involved in both blue-orange color polymorphisms: DFR-2 specificity redirects flux from the malvidin to the pelargonidin while BZ1-2 stabilizes the pelargonidin with glucose, producing the orange pelargonidin 3-glucoside. Moreover, a reduction of F3’5’H expression in orange petals also favors pelargonidin production. The climate niches for each color morph are the same between the two species for three temperature characteristics but differ for four precipitation variables. Conclusions The similarities in reflectance spectra, biochemistry, and ABP genes suggest that a single shift from blue-to-orange shared by both lineages is the most plausible explanation. Our evidence suggests that this persistent flower color polymorphism may represent an ancestrally polymorphic trait that has transcended speciation, yet future analyses are necessary to confidently reject the alternative hypotheses.

Adolescence is a period of special significance for the development of personal and social values. Positive adolescent development (PYD) can be an ideal perspective from which to promote values in young people through their sports practice. The aim of the study was to adapt and analyse the measurement properties of the Values Scale for Positive Youth Development for use in the context of sport in young athletes. A total of 599 adolescents, competitors of different sports modalities, participated in the study. Confirmatory factor analysis and reliability and invariance analyses were performed. The results confirmed a good model fit, with adjustment indexes (CFI, GFI and AGFI) above 0.90 and error (RMSEA and SRMR) below 0.08. The factor loadings above 0.50 were obtained for all items. Adequate reliability of the scales was also confirmed, between 0.72 and 0.89, and gender, age and sport invariance were confirmed. In conclusion, the analysed scale is a valid and reliable instrument with adequate psychometric properties, which makes it an appropriate assessment tool to be used in sports contexts, based on the positive adolescent development approach.

Background:: Deep brain stimulation of the central thalamus (CT-DBS) has potential for modulating states of consciousness, but it can also trigger electrographic seizures, including poly-spike-wave trains (PSWT). Objectives:: To report the probability of inducing PSWTs during CT-DBS in awake, freely-moving mice. Methods:: Mice were implanted with electrodes to deliver unilateral and bilateral CT-DBS at different frequencies while recording electroencephalogram (EEG). We titrated stimulation current by gradually increasing it at each frequency until a PSWT appeared. Subsequent stimulations to test arousal modulation were performed at the current one step below the current that caused a PSWT during titration. Results:: In 2.21% of the test stimulations (10 out of 12 mice), CT-DBS caused PSWTs at currents lower than the titrated current, including currents as low as 20 μA. Conclusion:: Our study found a small but significant probability of inducing PSWTs even after titration and at relatively low currents. EEG should be closely monitored for electrographic seizures when performing CT-DBS in both research and clinical settings.

Chiral optical forces exhibit opposite signs for the two enantiomeric versions of a chiral molecule or particle. If large enough, these forces might be able to separate enantiomers all optically, which would find numerous applications in different fields, from pharmacology to chemistry. Longitudinal chiral forces are especially promising for tackling the challenging scenario of separating particles of realistically small chiralities. In this work, we study the longitudinal chiral forces arising in dielectric integrated waveguides when the quasi-TE and quasi-TM modes are combined as well as their application to separate absorbing and non-absorbing chiral particles. We show that chiral gradient forces dominate in the scenario of beating of non-denegerate TE and TM modes when considering non-absorbing particles. For absorbing particles, the superposition of degenerate TE and TM modes can lead to chiral forces that are kept along the whole waveguide length. We accompany the calculations of the forces with particle tracking simulations for specific radii and chirality parameters. We show that longitudinal forces can separate non-absorbing chiral nanoparticles in water even for relatively low values of the particle chirality and absorbing particles with arbitrarily low values of chirality can be effectively separated after enough interaction time.

ABSTRACT Computer graphics aim to create visual representations for screens, where depth is simulated. In contrast, Computed Generated Holograms (CGH) focus on encoding and recreating light patterns to generate a true 3D holographic image that appears as a physical object in space. Therefore, although both use digital models, the computation of CGHs necessitates additional phase‐related calculations, which in turn escalate computational demands. These calculations often result in excessively long development times or, at worst, render the process unfeasible. In order to reduce computational time, Partial Monte–Carlo Sampling (PMCS) techniques for CGH generation are presented, integrating them into the whole process of generating a CGH for a synthetic 3D scene, from design to rendering. PMCS is based on the random choice of a subset of rays used to compute the CGH and relates the computation time spent to the quality of the reconstructed scene. Quantitative analysis shows that PMCS does not significantly compromise image quality. Both simulated and in‐laboratory image reconstruction from holograms demonstrates consistent trends, showcasing improved quality with higher numbers of rays and increased resolution. Furthermore, we establish a direct relationship between image quality and computational time, which effectively addresses specific requirements.
