A classical symphony orchestra consists of up to 29 musical instruments manufactured from up to 758 distinct natural materials. The interrelationships between the extraction of raw materials for instrument making, the international trade conditions, and the protection status of endangered species and their ecosystems are highly complex and have yet to be sufficiently scientifically examined. However, rapidly progressing climate and ecological change call for sustainable solutions. To address this challenging task, we present MusEcology, a new interactive decision support system based on visualizations. The interactive visualizations offer entry points for users of various backgrounds to explore the interrelationships between musical instruments, natural resources and ecosystems. The tool’s fundamental objectives are to guarantee that the (1) data processing correlates related data resources, that (2) visual interfaces and interaction schemes encourage new interdisciplinary research on complex systems interactions, and that (3) high-level decision-making is supported to identify alternative pathways towards sustainable instrument making.

Die Frage, inwieweit Datenmanagement jenseits von entweder generischen oder fachspezifischen Perspektiven denkbar ist, stand im Mittelpunkt eines Workshops im Rahmen der FORGE 2021. Im Workshop wurde das Konzept der „Data Communities“ (Cooper und Springer 2019) vorgestellt, seine Potenziale mit Blick auf die Sozial- und Geisteswissenschaften diskutiert und anschließend eruiert, welche strategischen wie operativen Kriterien sich daraus für forschungsadäquat unterstützende Datenmanagement-Services ableiten lassen. Der Beitrag fasst die wichtigsten Erkenntnisse aus dem Workshop zusammen und diskutiert, wie das bestehende Konzept um spezifisch sozial- und geisteswissenschaftliche Aspekte erweitert werden könnte.

Biological soil crusts (biocrusts) are a complex community of algae, cyanobacteria, lichens, bryophytes, and assorted bacteria, fungi, archaea, and bacteriophages that colonize the soil surface. Biocrusts are particularly common in drylands and are found in arid and semiarid ecosystems worldwide. While diminutive in size, biocrusts often cover large terrestrial areas, provide numerous ecosystem benefits, enhance biodiversity, and are found in multiple configurations and assemblages across different climate and disturbance regimes. Biocrusts have been a focus of many ecologists, especially those working in semiarid and arid lands, as biocrusts are foundational community members, play fundamental roles in ecosystem processes, and offer rare opportunities to study biological interactions at small and large spatial scales. Due to these same characteristics, biocrusts have the potential to serve as an excellent teaching tool. The purpose of this paper is to demonstrate the utility of biocrust communities as a model system in science education. Functioning as portable, dynamic mini ecosystems, biocrusts can be used to teach about organisms, biodiversity, biotic interactions, abiotic controls, ecosystem processes, and even global change, and can be easy to use in nearly every classroom setup. For example, education principles, such as evolution and adaptation to stress, or structure and function (patterns and processes) can be applied by bringing biocrusts into the classroom as a teaching tool. In addition, discussing the utility of biocrusts in the classroom – including theory, hypothesis testing, experimentation, and hands-on learning – this document also provides tips and resources for developing education tools and activities geared toward impactful learning.

The wood of the pau-brasil tree (Paubrasilia echinata Lam., formerly Caesalpinia echinata Lam.) is used worldwide as raw material for the construction of high-quality bows for string instruments. Alternative tree species are rarely accepted by professional musicians, or by bow and violin makers. Historical overexploitation of this endemic species in the Brazilian Atlantic Forest biome (Mata Atlântica), a global biodiversity hotpot including UNESCO World Natural Heritage Sites, and illegal trade have caused drastic declines in its natural abundance. Pau-brasil is now classified as an endangered species and listed in Appendix II of the Convention on International Trade in Endangered Species. Traditional bow-making craftsmanship, an intangible cultural heritage, depends heavily on the high-quality pau-brasil wood. This complex situation presents unprecedented cross-continental transdisciplinary challenges. In order to target the protection of this coupled natural/cultural heritage, this work frames and examines the pau-brasil/bow-making cultural-ecological system as a complex telecoupled system linked by cultural ecosystem services provided by the pau-brasil, as well as the relationships and cultural exchanges among key actors. Using historical trajectory analysis, we identify past, present, and potential future trigger events, key drivers, and key system variables that explain the dynamics, feedback, and resilience of this complex multi-dimensional system. Furthermore, with a cross-scale social and power relations analysis, we examine the level of dependencies and influences of contemporary key actors on the ecosystem services provided by the pau-brasil and their interconnections, in order to ultimately identify their level of disadvantage regarding the pau-brasil. Finally, we discuss the potential of this novel cultural-ecological system approach to (i) interlink science, nature, and art, (ii) reconcile the currently competing protection aims of natural and cultural heritage elements, and (iii) provide future trajectories regarding the resilience and sustainable development of this pau-brasil/bow-making cultural-ecological system. We advocate for this novel path forward toward sustainable transformation of complex cultural-ecological systems urgently needed to navigate our increasingly telecoupled world.

In this paper we will analyze the dynamics of a social-ecological system (SES), which requires an integrated understanding of both the interrelatedness of biophysical and socioeconomic components and the adaptive capacity of these system’s components to external drivers. Social-ecological resilience, the adaptive cycle metaphor and livelihood development are presented as the guiding conceptual framework to analyze local strategies, aiming towards the sustainable use of natural resources and to encourage the participation of the community in the management of ecosystem services, thereby improving human well-being. Furthermore, in the light of recurring unpredictable changes, adaptive capacity building and a high responsiveness to these changes may serve as fundamental assets to increase both ecological resilience, including the protection of biodiversity, and social resilience, including social and human capital and institutional capacity. An integrated analysis of SESs considers i) the interplay of internal and external factors and their role in SES dynamics, ii) potential thresholds whose crossing may shift the system into an undesirable state, and iii) cross-scale spatial and temporal interactions. Ultimately, an SES approach favors ecosystem stewardship in that it enhances the sustainable use of natural resources and ecosystem services, and simultaneously resilient livelihood development.

Intestinal microbiota disruption is associated with acute gastrointestinal (GI) Graft-versus-Host Disease (GvHD) and poor outcome after allogeneic stem cell transplantation (ASCT). Here, in a retrospective analysis of 200 patients undergoing ASCT at the Regensburg University Medical Center, we assessed the relative expression of Paneth cell antimicrobial peptides (AMPs), Human Defensins (HD) 5 and 6 and regenerating islet-derived 3α (Reg3α), in 292 human intestinal biopsies as well as Reg3α serum levels in relation to acute GI GvHD. In the absence of GI GvHD, the relative expression of Paneth cell AMPs was significantly higher in the small intestine (duodenum to ileum) than in the stomach and large intestine (cecum to rectum) for Reg3α (p≤0.001), HD5 (p≤0.002) and HD6 (p≤0.02). Acute stage 2-4 GI GvHD was associated with reduced expression of AMPs in the small intestine (p≤0.01) in comparison to stage 0-1 disease, accompanied by a decrease in Paneth cell count in case of severe acute GI GvHD (p

Myelofibrosis is a myeloproliferative neoplasm that results in cytopenia, bone marrow fibrosis and extramedullary hematopoiesis. Allogeneic hematopoietic stem cell transplantation is the only curative treatment but is associated with a risk of delayed engraftment and graft failure. In this study, patients with myelofibrosis (n=31) and acute myeloid leukemia (n=31) were analyzed for time to engraftment, graft failure and engraftment-related factors. Early and late neutrophil engraftment and late thrombocyte engraftment were significantly delayed in patients with myelofibrosis as compared to acute myeloid leukemia, and graft failure only occurred in myelofibrosis (6%). Only spleen size had a significant influence on engraftment efficiency in myelofibrosis patients. To analyze the cause for the engraftment defect, clearance of hematopoietic stem cells from peripheral blood was measured and immunohistological staining of bone marrow sections was performed. Numbers of circulating CD34+ were significantly reduced at early time points in myelofibrosis patients, whereas CD34+CD38− and colony-forming cells showed no significant difference in clearance. Staining of bone marrow sections for homing proteins revealed a loss of VCAM-1 in myelofibrosis with a corresponding significant increase in the level of soluble VCAM-1 within the peripheral blood. In conclusion, our data suggest that reduced engraftment and graft failure in myelofibrosis patients is caused by an early pooling of CD34+ hematopoietic stem cells in the spleen and a bone marrow homing defect caused by the loss of VCAM-1. Improved engraftment in myelofibrosis might be achieved by approaches that reduce spleen size and cleavage of VCAM-1 in these patients prior to hematopoietic stem cell transplantation.

In our case, a 45-year-old male patient had multiple fractures accompanied by hypophosphatemia. FGF-23 levels were significantly increased, and total body magnetic resonance imaging (MRI) revealed a tumor mass located at the distal tibia leading to the diagnosis of tumor-induced osteomalacia (TIO). After resection of the tumor, hypophosphatemia and the increased levels of FGF-23 normalized within a few days. Subsequent microscopic examination and immunohistochemical analysis revealed a phosphaturic mesenchymal tumor mixed connective tissue variant (PMTMCT) showing a positive expression of somatostatin receptor 2A (SSTR2A), CD68, and Periostin. Electron microscopy demonstrated a poorly differentiated mesenchymal tumor with a multifocal giant cell component and evidence of neurosecretory-granules. However, the resected margins showed no tumor-free tissue, and therefore a subsequent postoperative radiotherapy was performed. The patient is still in complete remission after 34 months. Tumor resection of PMTMCTs is the therapy of choice. Subsequent radiotherapy in case of incompletely resected tumors can be an important option to avoid recurrence or metastasis even though this occurs rarely. The prognostic value of expression of Periostin has to be evaluated more precisely in a larger series of patients with TIO.

The success of allogeneic (allo) hematopoietic cell transplantation (HCT) is limited by its treatment related complications, mostly graft versus host disease (GVHD) and fungal and viral infections. CMV reactivation after HCT has been associated with increased morbidity and mortality, and a causal relation between GVHD, immunosuppressive therapy and vice versa has been postulated. Using a low GVHD severity murine HCT model, we assessed the role of MCMV reactivation and GVHD development. BALB/c mice were infected with either murine CMV (MCMV) or mock and monitored for 25 weeks to establish latency, followed by sublethal irradiation conditioning and infusion of bone marrow plus splenocytes from either syngeneic (syn) BALB/c or allo B10.D2 donors. Engraftment of allo donor cells was confirmed by PCR for D2Mit265 gene product size. Day+100 mortality and overall GVHD severity in allo MCMV pre-infected recipients was higher than in allo mock controls. Pathologic changes of lung and liver GVHD in immediate-early gene 1 (IE1) positive recipients were significantly increased compared to mock controls, and were only slightly increased in IE1 negative. No significant gut injury was seen in any group. Aggravated lung injury in IE1 positive recipients correlated with higher BAL cell counts both for total cells and for CD4+ T cells when compared with mock controls, and also with protein expression of lung IFN-gamma and liver TNF. No evidence for CMV specific morphologic changes was seen on histopathology in any organ of IE1 positive recipients, suggesting that CMV reactivation is related to increased GVHD severity but does not require active CMV disease, strengthening the concept of a reciprocal relationship between CMV and GVHD.

En los pastizales del norte de México se han introducido gramíneas de origen africano, con el propósito fundamental de controlar la erosión del suelo e incrementar la productividad. Estas especies en lo general exhiben mayor talla y mayor producción de semilla, por lo que existe el riesgo, en caso de su naturalización, de que desplacen a las especies nativas. La invasión de ecosistemas semiáridos por estas especies exóticas ocasiona múltiples problemas, por lo que es fundamental identificar con anticipación los mecanismos que contribuyen a su propagación. En ensayos con condiciones controladas se trabajó con las gramíneas nativas Bouteloua gracilis y B. curtipendula así como las exóticas africanas Eragrostis curvula, E. lehmanniana, E. superba, Melinum repens y Panicum coloratum. Los mecanismos de adaptación de las plantas a los factores ambientales se manifiestan en sus rasgos morfológicos, fisiológicos y de crecimiento. La identificación de estos rasgos en especies de gramíneas nativas permite identificar germoplasma con características particulares de tolerancia a estrés o de alto rendimiento. En este estudio se examinaron divergencias morfológicas, de asignación de biomasa y de crecimiento de dos poblaciones del zacate navajita (Bouteloua gracilis) relacionadas con historiales de pastoreo contrastantes (sobrepastoreo vs pastoreo moderado) como un primer paso para identificar material genético sobresaliente. De cada población se examinaron 20 individuos que crecieron en condiciones controladas, y se evaluaron las siguientes variables, asignación de biomasa, altura, área foliar, anchura de hojas, área basal, área foliar específica, contenido de materia seca y mortalidad de tallos. A 61-year-old man presented with hematuria and intermittent right pelvic pain. Intravenous urography showed a tubular filling defect and ureteroscopy a tumor in the right mid ureter. Urine cytology and tumor biopsy showed nonmalignant results. Open surgery was performed, and an intraoperative frozen section revealed a fibroepithelial polyp of the right mid ureter. A fibroepithelial polyp is a rare benign lesion that can occur in childhood but is an important differential diagnosis of an upper urinary tract urothelial cell carcinoma in adults.

Valid and multidisciplinary assessment of a stroke patient's ability to perform activities of daily living is very important to define individual goals and to plan targeted rehabilitation. Until today, there is no observation scale that relies on International Classification of Functioning, Disability and Health (ICF). The aim of the present study was to develop and evaluate the reliability and validity of a new multidisciplinary observation scale for stroke patients, shortly called LIMOS, which is based on ICF.In a first phase, LIMOS was defined, using a Delphi approach, by an expert panel and a pilot testing was conducted in a small group of stroke patients (n =10) to investigate feasibility and practicability. In a second phase, LIMOS was assessed for its reliability (internal consistency and test-retest reliability) and validity in a large cohort of stroke patients (n = 102). For convergent validity, the correlation between total scores of the LIMOS and the Functional Independence Measure (FIM) was assessed.LIMOS consisted of seven ICF chapters incorporating 45 domains. A high internal consistency (=0.98) of LIMOS was found. Furthermore, good test-retest reliability at item and subscale level was found. Principal component analysis revealed that among the seven ICF chapters, four components could be found: (1) interpersonal activities, mobility and self-care, (2) communication, (3) knowledge and general tasks, and (4) domestic life. Significant associations were found between LIMOS and the FIM indicating good convergent validity.The new LIMOS is a reliable and valid observation scale for stroke patients based on ICF, which can be used by a multidisciplinary team working in a neurorehabilitation setting.

While early and higher visual areas along the ventral visual pathway in the inferotemporal cortex are critical for the recognition of individual objects, the neural representation of human perception of complex global visual scenes remains under debate. Stroke patients with a selective deficit in the perception of a complex global Gestalt with intact recognition of individual objects - a deficit termed simultanagnosia - greatly helped to study this question. Interestingly, simultanagnosia typically results from bilateral lesions of the temporo-parietal junction (TPJ). The present study aimed to verify the relevance of this area for human global Gestalt perception. We applied continuous theta-burst TMS either unilaterally (left or right) or bilateral simultaneously over TPJ. Healthy subjects were presented with hierarchically organized visual stimuli that allowed parametrical degrading of the object at the global level. Identification of the global Gestalt was significantly modulated only for the bilateral TPJ stimulation condition. Our results strengthen the view that global Gestalt perception in the human brain involves TPJ and is co-dependent on both hemispheres.
