The objective of this study was to identify the dominant bacterial flora linked to the hygiene of dried and smoked fish in different geographical locations and to determine the presence of pesticide residues on the processed fish. Fish species were listed and collected. The bacterial ecology of dried and smoked fish from the northern zone of Cameroon was probed using a molecular technique employing variable regions of 16S rDNA profiles generated by PCR-DGGE (Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis). The bacterial species were obtained by sequencing the PCR-DGGE bands and the pesticide residues by Chromatography coupled with mass spectrometry (GC-MS / MS), tested on 11 fish samples. Fifty three percent of the fish were smoked. Three types of pesticides have been identified on the dried fish samples. Among them, there were Cypermethrin (15 to 3600 µg/kg), Chlorpyrifos (19 to 8800 µg/kg) and Profenophos (62 to 92 µg/kg). The analysis of 16S rDNA profile and DNA sequencing revealed the presence of 32 bacterial species grouped into 22 genera. Dried fish were the most contaminated. Species such as Vagococcus carniphilus, Kurthia gibsonii, Lysinibacillus endophyticus, Macrococcus caseolyticus and Bacillus atrophaeus were present in the samples from all the locations. On a general note, the fish samples are of poor microbiological and sanitary quality.

The antifungal and antiaflatoxinogenic activities of the essential oils (EOs) from the leaves of Cymbopogon schoenanthus, Cymbopogon citratus, Cymbopogon nardus, and their pair combinations were investigated. Antifungal susceptibility and the efficacy of paired combinations of EOs were assessed using agar microdilution and checkerboard methods, respectively. Identification and quantification of chemical components of the EOs were carried out by gas chromatography-mass spectrometry and gas chromatography-flame ionization detector (GC-MS and GC-FID), respectively. Aflatoxins were separated and identified by High-Performance Liquid Chromatography (HPLC) and then quantified by spectrofluorescence. The EO of C. nardus exhibited the highest inhibitory activity against Aspergillus flavus and Aspergillus parasiticus. The combination of C. citratus and C. nardus and that of C. nardus and C. schoenanthus exhibited a synergistic effect against Aspergillus flavus and Aspergillus, respectively. Both C. citratus and C. schoenanthus EOs totally inhibited the synthesis of aflatoxin B1 at 1 µL/mL. C. citratus blocked the production of aflatoxins B2 and G2 at 0.5 µL/mL. Both C. citratus and C. schoenanthus totally hampered the production of the aflatoxin G1 at 0.75 µL/mL. The combination of C. citratus and C. schoenanthus completely inhibited the production of the four aflatoxins. The study shows that the combinations can be used to improve their antifungal and antiaflatoxinogenic activities.

Bacteria generally ferment milk but, sometimes, yeasts are found in fermented milks. The presence of these yeasts in the microbial community of some fermented milks could be intentional or accidental. The Diversity of yeasts in the products was investigated using a molecular technique employing variable regions of 26S rDNA profiles generated by PCR-DGGE (Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis). Two types of samples: artisanal or handcraft yoghurt and traditionally fermented milks were collected in three towns of the three regions of the northern part of Cameroon. Firstly, a comparison was made between the 16 traditionally fermented milks collected in Maroua, Garoua and Ngaoundre each of the 3 regions. Secondly, it was between 26 artisanal fermented milks of each region and finally, between the two types of products. The different PCR-DGGE 26S rDNA profiles obtained were analyzed and DNA sequencing was used to compare yeasts from each method of production. Twelve (12) species of Yeasts were identified as: Malassezia globosa, Hanseniaspora uvarum, Galactomyces candidum, Candida tropicalis, Aureobasidium pullulans, Torulaspora globosa, Saccharomyces cerevisiae/paradoxus, Pichia kluyveri, Candida parapsilosis, Torulaspora delbrueckii, Kluyveromyces marxianus, Candida orthopsilosis and Pseudozyma sp. Yeast diversity was higher for artisanal fermented milks (yoghurt) with at least 10 species, while for traditionally non packed fermented milks only 5 species were identified with a predominance strain of Galactomyces candidum, Candida parapsilosis, Torulaspora delbrueckii, Saccharomyces cerevisiae/paradoxus and Kluyveromyces marxianus. The different species of yeast might be introduced accidentally in artisanal yoghurt; however, for traditionally fermented milks their presence might be associated to the starter. This study was conducted to assess the different risk factors related to practices of sale of peanuts for better control of aflatoxin contamination and also to improve the safety of peanuts during marketing. The sales practices are similar regardless of the area surveyed. Of all the respondents in this survey, 61.7% were men as against 38.3% women and 70% of the respondents do not have any formal education. The marketing of peanut seeds is generally an activity done by men (100%) while that of peanut paste is assigned to women (100%). The unsold peanut paste is kept from 1 day to 3 months at room temperature until sale, mainly in market stalls used to sell it (42.4%) or in stores (39.4%). The losses are mainly due to fungi (37.9%) and insects (13.6%) followed by humidity and high temperature (4.5%). During sale of peanut paste, the site, the surroundings of sales points and the storage containers are potential fungi growth factors and eventual risk points for mycotoxin contamination. Proper handling and hygiene might reduce the aflatoxin contamination of peanut to ensure better sanitary quality of peanuts and protect consumer’s heath.

The prevalence of diarrheal diseases in children aged from 6 to 24 months in Burkina Faso is 38%. These diarrheas may be due to the consumption of contaminated weaning food. Therefore, the microbiological quality of follow up infant flours used as supplement foods is a key-point to reduce children diseases. In this study, the microbiological safety of locally-produced infant flours was investigated. One hundred and ninety-nine (199) samples were collected mainly in retails outlets and in Recovery and Nutrition Education Centers. According to the Burkina Faso regulations, microbiological analyses were carried out for Total Aerobic Mesophilic Flora (TAMF), thermotolerant coliforms, Salmonella spp. and yeasts/molds. The bacterial and fungal isolates were identified using phenotypic and genotypic methods and the study of the production of mycotoxins was carried out from the fungal isolates. In collected samples, the TAMF count ranged from 0 to 1.8 × 106 CFU/g with a total average of 6.3 × 104 CFU/g. About 2% of the samples had a microbial load exceeding the standards (105 CFU/g). No Salmonella spp. was isolated in the final infant flours. However, the presence of Enterobacteriaceae (Klebsiella spp. Enterobacter spp. and Cronobacter spp.) was detected and molecular characterization revealed also the presence of fungal species of the genus Aspergillus spp., Penicillium spp. and Fusarium spp. Some of these species were found to produce aflatoxins, ochratoxin A and fumonisins, which are potential carcinogenic toxins. These results demonstrated the need for a preventive approach based on the application of Hazard Analysis Critical Control Point in the food industry to ensure food safety of infant flours in Burkina Faso.

Acetic acid bacteria are involved in many food and beverage fermentation processes. They play an important role in cocoa bean fermentation through their acetic acid production. They initiate the development of some of the flavor precursors that are necessary for the organoleptic quality of cocoa, and for the beans’ color. The development of starter cultures with local strains would enable the preservation of the microbial biodiversity of each country in cocoa-producing areas, and would also control the fermentation. This approach could avoid the standardization of cocoa bean fermentation in the producing countries. One hundred and thirty acetic acid bacteria were isolated from three different cocoa-producing countries, and were identified based on their 16S rRNA gene sequence. The predominate strains were grown in a cocoa pulp simulation medium (CPSM-AAB) in order to compare their physiological traits regarding their specific growth rate, ethanol and lactic acid consumption, acetic acid production, and relative preferences of carbon sources. Finally, the intraspecific diversity of the strains was then assessed through the analysis of their genomic polymorphism by (GTG)5-PCR fingerprinting. Our results showed that Acetobacter pasteurianus was the most recovered species in all of the origins, with 86 isolates out of 130 cultures. A great similarity was observed between the strains according to their physiological characterization and genomic polymorphisms. However, the multi-parametric clustering results in the different groups highlighted some differences in their basic metabolism, such as their efficiency in converting carbon substrates to acetate, and their relative affinity to lactic acid and ethanol. The A. pasteurianus strains showed different behaviors regarding their ability to oxidize ethanol and lactic acid into acetic acid, and in their relative preference for each substrate. The impact of these behaviors on the cocoa quality should be investigated, and should be considered as a criterion for the selection of acetic acid bacteria starters.

Vast amounts of information can be obtained by systematic explorations of synergy between phytochemicals and probiotics, which is required for the development of non-dairy probiotic products, globally. Evidence confirms that the same probiotic strain can have different efficiencies depending on the food matrix. One such functional property, viz., antipathogenicity of the probiotic strain against Shigella was investigated in this study. The potential of two fruit based (apple and sea buckthorn) beverage matrices fortified with Lactobacillus rhamnosus GG (ATCC 53103), against outbreak-causing serotypes of Shigella dysenteriae (ATCC 29026) and Shigella flexneri (ATCC 12022) was evaluated. The originality of this study lies in the fact that the functionality assessment was performed with a more realistic approach under storage conditions from 0–14 days at 4 °C. The finding confirms that Lactobacillus rhamnosus GG (LGG) differs in its potential depending on beverage matrices. Principal Component Analysis (PCA) clustered the matrices based on their pathogen clearance. LGG fortified sea buckthorn beverage matrix showed 99% clearance of S. dysenteriae within the first hour compared to 11% in apple beverage matrix. Interestingly, S. flexneri showed more resistance and was cleared (99%) in the LGG fortified sea buckthorn beverage matrix within three hours compared to 5.6% in apple matrix.

Several strains of a new aflatoxigenic species of Aspergillus, A. korhogoensis, were isolated in the course of a screening study involving species from section Flavi found contaminating peanuts (Arachis hypogaea) and peanut paste in the Côte d’Ivoire. Based on examination of four isolates, this new species is described using a polyphasic approach. A concatenated alignment comprised of nine genes (ITS, benA, cmdA, mcm7, amdS, rpb1, preB, ppgA, and preA) was subjected to phylogenetic analysis, and resulted in all four strains being inferred as a distinct clade. Characterization of mating type for each strain revealed A. korhogoensis as a heterothallic species, since three isolates exhibited a singular MAT1-1 locus and one isolate exhibited a singular MAT1-2 locus. Morphological and physiological characterizations were also performed based on their growth on various types of media. Their respective extrolite profiles were characterized using LC/HRMS, and showed that this new species is capable of producing B- and G-aflatoxins, aspergillic acid, cyclopiazonic acid, aflavarins, and asparasones, as well as other metabolites. Altogether, our results confirm the monophyly of A. korhogoensis, and strengthen its position in the A. flavus clade, as the sister taxon of A. parvisclerotigenus.

In vitro adhesion assay using Lactobacillus reuteri KUB-AC5 as a test strain has been studied by applying simple PCR reaction together with image analysis and plate count techniques. Critical factor affecting the PCR method was quality and quantity of DNA. The cell lysis technique was modified to optimize this method. Thus, lysozyme and proteinase K were added to lyse the cells, followed by SDS solution to obtain a complete cell lysis. Only PCR products from total cells (TC) were obtained, with low consistency, but none from cells bound to mucus (BC) at either 0.1 or 0.5 mg/mL concentration. It was hypothesized that the attached cells might not be extracted into the cell suspension. Therefore, 1% SDS solution and 0.1M NaOH were used directly in the extraction. As expected, PCR products were observed when both TC and BC were used as a DNA template. Adhesion appeared at a wide range of 0-45%, with low consistency. Therefore, a simple microbiological method (plate count) was used. The extraction of bound cells into cell suspension was critical in this method. Extraction times of 20, 60, 120 and 150 min were tried. Results showed that maximum cell number was obtained with 120 min extraction. L. reuteri KUB-AC5, L. reuteri KUB-AC16, L. reuteri KUB-AC20, L. salivarius KUB-AC21, L. acidophilus KV-1, Escherichia coli E010, Salmonella sp. S003, E. coli ATCC8739, and S. typhimurium ATCC 13311 exhibited adhesion activity of 21.6%, 0.8%, 5.7%, 1.1%, 23.1%, 10.7%, 10.3%, 4.4% and 3.2%, respectively. Among the 9 types of microorganisms tested L. acidophilus KV-1 and L. reuteri KUB-AC5 showed higher adhesion activity than the others.

Although Yersinia enterocolitica is usually transmitted through contaminated food and untreated water, occasional transmission such as human-to-human, animal-to-human and blood transfusion associated transmission have also identified in human disease. Of the six Y. enterocolitica biotypes, the virulence of the pathogenic biotypes, namely, 1B and 2–5 is attributed to the presence of a highly conserved 70-kb virulence plasmid, termed pYV/pCD and certain chromosomal genes. Some biotype 1A strains, despite lacking virulence plasmid (pYV) and traditional chromosomal virulence genes, are isolated frequently from humans with gastrointestinal diseases similar to that produced by isolates belonging known pathogenic biotypes. Y. enterocolitica pathogenic biotypes have evolved two major properties: the ability to penetrate the intestinal wall, which is thought to be controlled by plasmid genes, and the production of heat-stable enterotoxin, which is controlled by chromosomal genes.
