Abstract As a sport conducted in dynamically changing natural environments, orienteering places high demands on athletes' cognitive processing abilities and visual search efficiency. However, previous studies on orienteering have been primarily limited by the use of fixed stimulus materials on computer screens, which are unable to fully simulate authentic sports scenarios. To better understand the sports expertise of orienteering athletes in terms of their real scene image recognition performance and visual search characteristics, this study recruited 40 orienteering athletes, both experts and novices, as participants. By utilizing eye-tracking technology and setting observation points in real-world scenarios to conduct image recognition task tests, the ecological validity of the experiment was further enhanced. The results showed that the experts demonstrated a high level of accuracy and a short response time, with visual search characteristics including few saccade counts, low fixation frequency, concentrated fixation points, simple and clear fixation paths, and higher visual search efficiency. This study further reveals that long-term specialized training will lead to the formation of a unique cognitive structure related to the specific knowledge and long-term memory required by expert orienteering athletes, thereby promoting the development of expert advantage.

IntroductionMycoplasma synoviae (MS) is a globally important avian pathogen causing infectious synovitis and respiratory diseases in poultry, leading to significant economic losses. Despite advances in vaccine development, a commercially viable subunit vaccine against MS remains elusive.MethodsWe sequenced whole genomes of six clinical MS strains isolated from different Chinese provinces. Common genes were analyzed using Biopython software, identifying those with high copy numbers in virulent strains and shared among all strains. Vaxign2 and IEDB Antibody Epitope Prediction were used to analyze protein properties. We assessed immune protective effects of candidate proteins and developed a multivalent subunit vaccine.ResultsTen candidate vaccine proteins were initially selected. A multivalent subunit vaccine composed of MSPB, Ppht, Cfba, and EF-G displayed the best protective effect. The optimal immunization dosage was 20μg, with each protein accounting for 25%. The immune production period was determined to be 28 days post-first immunization, lasting 180 days. The immune protection rate against highly virulent strains reached 90%∼100%.DiscussionThis study provides a new approach for screening vaccine antigens and develops an effective candidate vaccine for MS prevention. The multivalent subunit vaccine shows promising results in protecting against MS infections, potentially offering a solution to reduce economic losses in the poultry industry.

African swine fever virus (ASFV) is one of the most complex viruses. ASFV is a serious threat to the global swine industry because no commercial vaccines against this virus are currently available except in Vietnam. Moreover, ASFV is highly stable in the environment and can survive in water, feed, and aerosols for a long time. ASFV is transmitted through the digestive and respiratory tract. Mucosal immunity is the first line of defense against ASFV. Saccharomyces cerevisiae (SC), which has been certified by the U.S. Food and Drug Administration and has a generally recognized as safe status in the food industry, was used for oral immunization in this study. ASFV antigens were effectively expressed in recombinant SC strains with high DNA copy numbers and stable growth though surface display technology and chromosome engineering (δ-integration). The recombinant SC strains containing eight ASFV antigens—KP177R, E183L, E199L, CP204L, E248R, EP402R, B602L, and B646L— induced strong humoral and mucosal immune responses in mice. There was no antigenic competition, and these antigens induced Th1 and Th2 cellular immune responses. Therefore, the oral immunization strategy using recombinant SC strains containing multiple ASFV antigens demonstrate potential for future testing in swine, including challenge studies to evaluate its efficacy as a vaccine against ASFV.

Studying multiple properties of a material concurrently is essential for obtaining a comprehensive understanding of its behavior and performance. However, this approach presents certain challenges. For instance, simultaneous examination of various properties often necessitates extensive experimental resources, thereby increasing the overall cost and time required for research. Furthermore, the pursuit of desirable properties for one application may conflict with those needed for another, leading to trade-off scenarios. In this study, we focused on investigating adhesive joint strength and elastic modulus, both crucial properties directly impacting adhesive behavior. To determine elastic modulus, we employed a non-destructive indentation method for converting hardness measurements. Additionally, we introduced a specimen apparatus preparation method to ensure the fabrication of smooth surfaces and homogeneous polymeric specimens, free from voids and bubbles. Our experiments utilized a commercially available bisphenol A-based epoxy resin in combination with a Poly(propylene glycol) curing agent. We generated an initial dataset comprising experimental results from 32 conditions, which served as input for training a machine learning model. Subsequently, we used this model to predict outcomes for a total of 256 conditions. To address the high deviation in prediction results, we implemented active learning approaches, achieving a 50% reduction in deviation while maintaining model accuracy. Through our analysis, we observed a trade-off boundary (Pareto frontier line) between adhesive joint strength and elastic modulus. Leveraging Bayesian optimization, we successfully identified experimental conditions that surpassed this boundary, yielding an adhesive joint strength of 25.2 MPa and an elastic modulus of 182.5 MPa.

Pancreatic β-cell failure is a pathological feature in type 1 diabetes. One promising approach involves inducing transdifferentiation of related pancreatic cell types, specifically α cells that produce glucagon. The chemokine stromal cell-derived factor-1 alpha (SDF-1α) is implicated in pancreatic α-to-β like cell transition. Here, the serum level of SDF-1α was lower in T1D with C-peptide loss, the miR-23a was negatively correlated with SDF-1α. We discovered that exosomal miR-23a, secreted from β cells, functionally downregulates the expression of SDF-1α, leading to increased Pax4 expression and decreased Arx expression in vivo. Adenovirus-vectored miR-23a sponge and mimic were constructed to further explored the miR-23a on pancreatic α-to-β like cell transition in vitro, which yielded results consistent with our cell-based assays. Suppression of miR-23a upregulated insulin level and downregulated glucagon level in STZ-induced diabetes mice models, effectively promoting α-to-β like cell transition. Our findings highlight miR-23a as a new therapeutic target for regenerating pancreatic β cells from α cells.

BackgroundMicrobiome-directed therapies are increasingly utilized to optimize thyroid function in both healthy individuals and those with thyroid disorders. However, recent doubts have been raised regarding the efficacy of probiotics, prebiotics, and synbiotics in improving thyroid function. This systematic review aimed to investigate the potential relationship between probiotics/prebiotics and thyroid function by analyzing the impact on thyroid hormone levels.MethodsWe conducted a comprehensive systematic review and meta-analysis of randomized controlled trials that investigated the effects of probiotics, prebiotics, and synbiotics on free triiodothyronine (fT3), free thyroxine (fT4), thyroid stimulating hormone (TSH), and thyroid stimulating hormone receptor antibody (TRAb) levels. We searched for articles from PubMed, Scopus, Web of Science, and Embase up until April 1st, 2023, without any language restriction. Quantitative data analysis was performed using a random-effects model, with standardized mean difference (SMD) and 95% confidence interval as summary statistics. The methods and results were reported according to the PRISMA2020 statement.ResultsA total of eight articles were included in this review. The meta-analysis showed no significant alterations in TSH (SMD: -0.01, 95% CI: -0.21, 0.20, P = 0.93; I2: 0.00%), fT4 (SMD: 0.04, 95% CI: -0.29, 0.21, P = 0.73; I2: 0.00%) or fT3 (SMD: 0.45, 95% CI: -0.14, 1.03, P = 0.43; I2: 78.00%), while a significant reduction in TRAb levels was observed (SMD: -0.85, 95% CI: -1.54, -0.15, P = 0.02; I2: 18.00%) following probiotics/prebiotics supplementation. No indication of publication bias was found.ConclusionsProbiotics/prebiotics supplementation does not influence thyroid hormone levels, but may modestly reduce TRAb levels in patients with Graves' disease.

Hedgerow is a vital soil and water conservation technology for sloping farmland that can significantly reduce erosion and nutrient loss via their above- and below-ground parts. The effect of hedgerow on erosion and nutrient loss has been widely investigated, while the respective effects of their above- and below-ground parts are still unclear. Therefore, the purple soil from the Three Gorges Reservoir Area was used, and 3 slope conditions of control check, whole hedgerow and hedgerow roots only combined with 2 slope gradients (15° and 25°) were constructed and the simulated rainfall tests were researched at 3 rainfall intensities (60, 90 and 120 mm h−1). The runoff initiation time, runoff and erosion rates, and loss of nitrogen via runoff and sediment were analysed. In comparison to the control check slope condition, it was indicated that hedgerow increased the runoff initiation time by 43.38 %, decreased runoff and erosion by 15.59 % and 78.37 %, and decreased nitrogen loss via runoff by approximately 40 % and via sediment by approximately 70 % on average, respectively. The average contribution rates of the below-ground part of the hedgerow was 49.89 % for the increase in runoff initiation time, 33.99 % for runoff reduction, and 39.91 % for erosion reduction. In addition, more than 2/3 and 58.49 % of nitrogen loss reduction via runoff and sediment, respectively, was contributed by the above-ground part of the hedgerow. These results are beneficial for comprehending the control mechanism of hedgerow on soil erosion and nitrogen loss, thereby provide a scientific foundation for the sustainable and efficient utilization of soil and water resources on sloping farmland.

The use of affine maneuver control to maintain the desired configuration of unmanned aerial vehicle (UAV) swarms has been widely practiced. Nevertheless, the lack of capability to interact with obstacles and navigate autonomously could potentially limit its extension. To address this problem, we present an innovative formation flight system featuring a virtual leader that seamlessly integrates global control and local control, effectively addressing the limitations of existing methods that rely on fixed configuration changes to accommodate real-world constraints. To enhance the elasticity of an algorithm for configuration change in an obstacle-laden environment, this paper introduces a second-order differentiable virtual force-based metric for planning local trajectories. The virtual field comprises several artificial potential field (APF) forces that adaptively adjust the formation compared to the existing following control. Then, a distributed and decoupled trajectory optimization framework that considers obstacle avoidance and dynamic feasibility is designed. This novel multi-agent agreement strategy can efficiently coordinate the global planning and local trajectory optimizations of the formation compared to a single method. Finally, an affine-based maneuver approach is employed to validate an optimal formation control law for ensuring closed-loop system stability. The simulation results demonstrate that the proposed scheme improves track accuracy by 32.92% compared to the traditional method, while also preserving formation and avoiding obstacles simultaneously.

Enterotoxigenic Escherichia coli (ETEC) causes severe diarrhea in piglets. The current primary approach for ETEC prevention and control relies on antibiotics, as few effective vaccines are available. Consequently, an urgent clinical demand exists for developing an effective vaccine to combat this disease. Here, we utilized food-grade Lactococcus lactis NZ3900 and expression plasmid pNZ8149 as live vectors, together with the secreted expression peptide Usp45 and the cell wall non-covalent linking motif LysM, to effectively present the mutant LTA subunit, the LTB subunit of heat-labile enterotoxin, and the FaeG of F4 pilus on the surface of recombinant lactic acid bacteria (LAB). Combining three recombinant LAB as a live vector oral vaccine, we assessed its efficacy in preventing F4+ ETEC infection. The results demonstrate that oral immunization conferred effective protection against F4+ ETEC infection in mice and piglets lacking maternal antibodies during weaning. Sow immunization during late pregnancy generated significantly elevated antibodies in colostrum, which protected piglets against F4+ ETEC infection during lactation. Moreover, booster immunization on piglets during lactation significantly enhanced their resistance to F4+ ETEC infection during the weaning stage. This study highlights the efficacy of an oral LAB vaccine in preventing F4+ ETEC infection in piglets by combining the sow immunization and booster immunization of piglets, providing a promising vaccination strategy for future prevention and control of ETEC-induced diarrhea in piglets.

In this study, an accurate, rapid, green, and environment friendly method for the extraction and quantitative analysis of flavonoids in honey was established by using the aqueous two-phase extraction combined with the chemometrics-assisted HPLC-DAD. The first purpose of this study was to extract seven flavonoids in five different types of honey using alcohol/salt aqueous two-phase system (ATPS). The system with 2.82 mL sodium citrate (30%), 1.58 mL water, and 3.10 mL isopropanol, showed the highest flavonoids extraction yields in the top phase (87.66–101.50%). Additionally, the three-way array of honey samples based on HPLC-DAD was decomposed mathematically by the alternating trilinear decomposition (ATLD) algorithm to obtain reasonable chromatograms, spectra, and concentration profiles for each analyte. Compared with the traditional solid-phase extraction method, the ATPS-ATLD-based method showed satisfactory spiked recoveries, lower limit of detection, and higher sensitivity, further verifying its accuracy and stability.

As harvesting at the right time is crucial to ensuring the best quality and maximizing the yield of tobacco leaves, great attention has been paid to the research of “harvest maturity”. Fresh tobacco leaves can be manually categorized into four maturity stages, including immature, pseudomature, mature, and hypermature, according to the leaf tissue structure and color. To improve the discriminative accuracy, convenience, and automation of maturity levels, a tobacco leaf maturity discriminative method based on machine vision and deep learning was developed and integrated into a system based on the mobile terminal. In this method, the chromatic features and texture features can be automatically extracted from the acquired digital image of tobacco leaf by convolutional neural networks (CNNs). Experimental results of an independent test set consisting of 480 tobacco leaf samples demonstrated that the proposed system was able to classify the maturity stage of tobacco leaf efficiently and accurately. The system's ability to run on mobile devices, like smartphones, makes it easier to accurately collect mature tobacco leaves and reduces subjectivity compared to the current visual identification approach. Moreover, this system can provide a picking date recommendation to tobacco growers.

Study region: The Three Gorges Reservoir area (TGR), China Study focus: Soil erosion on purple soil hillslope and its negative effects are deleterious to the ecological environment of the TGR. Sheet erosion is the initial form of the slope erosion, which is notably impacted by land degradation, polyacrylamide (PAM) is commonly applied for soil and water conservation purpose, while the sheet erosion on purple soil hillslope treated with PAM remains unclear. New hydrological insights for the region: The findings suggested that the sheet erosion and runoff rate increased as the rainfall intensity was increased and the slope gradient was decreased with increasing PAM application rate until reaching minimum values at 0.80 g m−2, and these quantities increased afterward. The single and combined impacts of the PAM application rate, rainfall intensity and slope gradient on erosion and runoff were significant. The slope gradient, rainfall intensity and PAM application rate also influenced the hydrodynamic parameters, and the stream power was the optimal parameter for describing the sheet erosion. An equation encompassing the rainfall intensity, slope gradient and PAM application rate was used for sheet erosion rate prediction with high accuracy relative to the stream power and PAM application rate. These results could be helpful to better understand sheet erosion on purple soil slopes treated with PAM and offer fundamental guidelines for soil and water conservation in the TGR.

In order to screen out novel energetic compounds with excellent comprehensive properties and provide theoretical guidance for design researcher, a series of difluoroamino energetic compounds were designed by combining energetic molecular scaffolds and NF2 groups. The effect of energetic molecular scaffold, group number and connection position on the performance of difluoroamino energetic compounds was systematically studied, and its design basis was given. From results, we could draw a conclusion that none of the designed energetic compounds could simultaneously satisfy high density, high detonation, good stability, low sensitivity, high explosive power and high specific impulse. When designing a compound structure, the selection of the scaffold type and number of difluoroamino group should be considered comprehensively weighing each performance to meet the actual needs.

Feline calicivirus (FCV) is one of the most important pathogens causing upper respiratory tract diseases in cats, posing a serious health threat to these animals. At present, FCV is mainly prevented through vaccination, but the protective efficacy of vaccines in China is limited. In this study, based on the differences in capsid proteins of isolates from different regions in China, as reported in our previous studies, seven representative FCV epidemic strains were selected and tested for their viral titers, virulence, immunogenicity, and extensive cross-protection. Subsequently, vaccine strains were selected to prepare inactivated vaccines. The whole-genome sequencing and analysis results showed that these seven representative FCV strains and 144 reference strains fell into five groups (A, B, C, D, and E). The strains isolated in China mainly fall into groups C and D, exhibiting regional characteristics. These Chinese isolates had a distant evolutionary relationship and low homology with the current FCV-255 vaccine strain. The screened FCV-HB7 and FCV-HB10 strains displayed desirable in vitro culture characteristics, with the highest virus proliferation titers (109.5 TCID50/mL) at 36 h post inoculation at a dose of 0.01 MOI. All five cats infected intranasally with FCV-HB7 or FCV-HB10 strains showed obvious clinical symptoms of FCV. The symptoms of cats infected with the FCV-HB7 strain were more severe than those infected with the FCV-HB10 strain. Both the single-strain inactivated immunization and combined bivalent inactivated vaccine immunization of FCV-HB7 and FCV-HB10 induced high neutralizing antibody titers in five cats immunized. Moreover, bivalent inactivated vaccine immunization protected cats from FCV-HB7 and FCV-HB10 strains. The cross-neutralizing antibody titer against seven representative FCV epidemic strains achieved by combined bivalent inactivated vaccine immunization was higher than that achieved by single-strain immunization, which was much higher than that achieved by commercial vaccine FCV-255 strain immunization. The above results suggest that the FCV-HB7 and FCV-HB10 strains screened in this study have great potential to become vaccine strains with broad-spectrum protective efficacy. However, their immune protective efficacy needs to be further verified by multiple methods before clinical application.

In grounding fault of DC system, overvoltage will appear on equipment nodes in converter station. Due to different operation mode and equipment type, it is very different from overvoltage in AC system. Based on Shanghaimiao–Linyi​ ±800kV Ultra High Voltage Direct Current (UHVDC) transmission project, this paper studies the converter station overvoltage characteristics under the conditions of DC pole line grounding, DC bus grounding and converter transformer valve side grounding fault, then analyzes the influence of smoothing reactor, filter capacitor and grounding pole line on overvoltage. In grounding fault at DC pole line near the converter station, the maximum overvoltage occurs on the inductance of DC filter, which is 592.75kV; In grounding fault at the top of the high voltage converter valve, the maximum overvoltage occurs on the neutral bus, which is 272.17kV. Increasing the inductance or capacitance of the smoothing reactor and filter capacitor, shortening the length of grounding electrode wire can significantly reduce the amplitude and steepness of overvoltage. The calculation results provide a reference for the internal overvoltage suppression of DC converter station with layered access at the inverter side.

It is of great importance to simultaneously stabilize output power and suppress platform motion and fatigue loads in floating offshore wind turbine control systems. In this paper, a novel composite blade pitch control scheme considering actuator fault is proposed based on an augmented linear quadratic regulator (LQR), a fuzzy proportional integral (PI) and an adaptive second-order sliding-mode observer. Collective pitch control was achieved via the fuzzy PI, while individual pitch control was based on the augmented LQR. In the case of actuator fault, an adaptive second-order sliding-mode observer was constructed to effectively eliminate the need for the upper bound of unknown fault derivatives and suppress the chattering effect. This paper conducted co-simulations based on FAST (Fatigue, Aerodynamics, Structures, and Turbulence) and MATLAB/Simulink to verify the effectiveness and superiority of the proposed scheme under different environmental conditions. It is shown that platform roll was reduced by approximately 54% compared to that under PI control. For the tower fore–aft moment, load reductions of 45% or more were achievable. The proposed scheme can greatly reduce the pitch and roll of the floating platform and loads in the windward direction of the wind turbine.

ABSTRACT Animal experiments on African swine fever virus (ASFV) are vital to the study of ASFV; however, ASFV can only infect pigs, and animal experiments need to be performed in animal biosafety level 3 (ABSL-3) laboratories, meaning that many small ABSL-3 laboratories are unable to carry out in vivo ASFV experiments. Therefore, miniaturized experimental animals for ASFV infection are urgently needed. Here, we successfully isolated genotype II of ASFV SY-1 from wild boars and evaluated ASFV-infected Bama minipigs in a negative-pressure isolator of a small ABSL-3 laboratory. The pathological changes of ASFV-infected Bama minipigs were consistent with characteristic lesions of ASFV-infected domestic pigs and wild boars. All pigs died 5 to 14 days postinfection (dpi) through intramuscular injection. Viral genomic DNA from nasal, oral, and rectal swab samples was first detectable at 2 to 4 dpi. The common differentially expressed genes were clustered in the immune-related, metabolic, and inflammatory response pathways from the spleen and inguinal lymph node samples comparing infected to mock. In summary, these results demonstrated that the Bama minipig was an appropriate model for ASFV infection in small ABSL-3 laboratories that can accelerate the research of vaccines and antiviral drugs and uncover pathogenic mechanisms of ASFV infection. IMPORTANCE African swine fever virus (ASFV) can only infect pigs rather than other animals. However, the domestic pigs cannot be kept in small ABSL-3 laboratories for a long time due to the characteristics of rapid growth and large size, which hinder ASFV research, including research of vaccines, antiviral drugs, and mechanisms. In contrast, Bama minipigs have unique advantages consisting of low growth and small size. In the research, Bama minipigs were used to evaluate the characteristics of ASFV infection in small ABSL-3 laboratories. The pathological changes, viral shedding, and gene regulation were consistent with those of domestic pigs infected with ASFV. Therefore, Bama minipigs can be a suitable model for ASFV infection in small ABSL-3 laboratories.

Abstract In this study, we investigated the effect of exenatide (EXE), a glucagon-like peptide (GLP)-1 receptor agonist, on kidney function, obesity indices, and glucose control in overweight/obese patients with type 2 diabetes mellitus (T2DM). A total of 159 overweight/obese patients with T2DM were randomized to the EXE group or insulin glargine (GLAR) control group for a total treatment period of 24 weeks. Chinese water chestnut (CWC) is one of the most popular hydrophytic vegetables in China due to its unique aroma and taste. The volatile compounds (VOCs) of CWC with different processing methods were evaluated. The results of headspace-gas chromatography–ion mobility spectrometry (HS-GC-IMS) showed that alcohols, aldehydes, aromatics, and esters were the main VOCs, and the headspace solid-phase microextraction-gas chromatography–mass spectrometry (HS-SPME-GC–MS) results indicated that esters, amines and aromatics were the main flavor components. Compared with other processes, the VOC alcohol levels in fresh cutting (FF) decreased, while esters and sulfide increased. The flavor of CWC in the cooking with peeling (PC) and steaming with peeling (PS) were similar, the FF and steaming with skin (WPC) are very different from the other samples. This study is the first to provide results regarding differences in the aroma compounds of CWC processed with different methods, and 146 aroma compounds were characterized.

Abstract In recent years, considerable attention has been paid in time–frequency analysis (TFA) methods, which is an effective technology in processing the vibration signal of rotating machinery. However, TFA techniques are not sufficient to handle signals having a strong non-stationary characteristic. To overcome this drawback, taking short-time Fourier transform as a link, a TFA methods that using the generalized Warblet transform (GWT) in combination with the second order synchroextracting transform (SSET) is proposed in this study. Firstly, based on the GWT and SSET theories, this paper proposes a method combining the two TFA methods to improve the TFA concentration, named GWT–SSET. Secondly, the method is verified numerically with single-component and multi-component signals, respectively. Quantized indicators, Rényi entropy and mean relative error (MRE) are used to analyze the concentration of TFA and accuracy of instantly frequency (IF) estimation, respectively. Finally, the proposed method is applied to analyze nonstationary signals in variable speed. The numerical and experimental results illustrate the effectiveness of the GWT–SSET method.

Abstract The performance models are the critical step for condition monitoring and fault diagnosis of diesel engines, and are an important bridge to describe the link between input parameters and targets. Large-scale experimental methods with higher economic costs are often adopted to construct accurate performance models. To ensure the accuracy of the model and reduce the cost of the test, a novel method for modeling the performances of marine diesel engine is proposed based on deep neural network method coupled with virtual sample generation technology. Firstly, according to the practical experience, the four parameters including speed, power, lubricating oil temperature and pressure are selected as the input factors for establishing the performance models. Besides, brake specific fuel consumption, vibration and noise are adopted to assess the status of marine diesel engine. Secondly, small sample experiments for diesel engine are performed under multiple working conditions. Moreover, the experimental sample data are diffused for obtaining valid extended data based on virtual sample generation technology. Then, the performance models are established using the deep neural network method, in which the diffusion data set is adopted to reduce the cost of testing. Finally, the accuracy of the developed model is verified through experiment, and the parametric effects on performances are discussed. The results indicate that the overall prediction accuracy is more than 93%. Moreover, power is the key factor affecting brake specific fuel consumption with a weighting of 30% of the four input factors. While speed is the key factor affecting vibration and noise with a weighting of 30% and 30.5%, respectively.

The market for water infrastructure rehabilitation is growing rapidly due to the increasing age of underground utilities. Currently, two common water main rehabilitation methods exist: cured-in-place pipe (CIPP) and polymer spray-on coatings. CIPP can provide structural support for both internal and external loads, while spray-on techniques provide chemical resistance as well as adding minor strength to the existing pipe. This paper summarizes water main rehabilitation practice using CIPP and spray-on methods. The history of trenchless rehabilitation technology is discussed, as well as current methodologies and products for water mains. The design, installation, and monitoring of water main rehabilitation products are also summarized, along with the associated risks. In order to screen novel high energetic density compounds with high power and specific impulse, eight compounds containing an aza-cage or fused ring structure were designed. The B3PW91/6–31G(d,p) theoretical method was used to optimize their structures. Their power, specific impulse, impact sensitivity and the other detonation performance were predicted, and compared with that of HMX and CL-20. The results showed that the explosive powers and the specific impulses of all eight compounds were over 150 and 2.60 N s g−1, respectively, and higher than that of HMX and CL-20. Especially, the compd-1 and compd-4 compounds have excellent comprehensive performance.

The 9-cis-epoxycarotenoid dioxygenase (NCED) is a key enzyme for the process of ABA synthesis that plays key roles in a variety of biological processes. In the current investigation, genome-wide identification and comprehensive analysis of the NCED gene family in ‘Kuerle Xiangli’ (Pyrus sinkiangensis Yu) were conducted using the pear genomic sequence. In total, nineteen members of PbNCED genes were identified from the whole genome of pear, which are not evenly distributed over the scaffolds, and most of which were focussed in the chloroplasts. Sequence analysis of promoters showed many cis-regulatory elements, which presumably responded to phytohormones such as abscisic acid, auxin, etc. Synteny block indicated that the PbNCED genes have experienced strong purifying selection. Multiple sequence alignment demonstrated that these members are highly similar and conserved. In addition, we found that PbNCED genes were differentially expressed in various tissues, and three PbNCED genes (PbNCED1, PbNCED2, and PbNCED13) were differentially expressed in response to exogenous Gibberellin (GA3) and Paclobutrazol (PP333). PbNCED1 and PbNCED13 positively promote ABA synthesis in sepals after GA3 and PP333 treatment, whereas PbNCED2 positively regulated ABA synthesis in ovaries after GA3 treatment, and PbNCED13 positively regulated ABA synthesis in the ovaries after PP333 treatment. This study was the first genome-wide report of the pear NCED gene family, which could improve our understanding of pear NCED proteins and provide a solid foundation for future cloning and functional analyses of this gene family. Meanwhile, our results also give a better understanding of the important genes and regulation pathways related to calyx abscission in ‘Kuerle Xiangli’.

Debonding of solidified splats is a crucial issue for thermal sprayed coatings, which greatly influences the performance and lifetime of industrial components. The purpose of this study is to provide an in-depth understanding of the effects of impact parameters as well as residual stress on the debonding behaviors, and the related adhesion strength and driving forces of single splats. In this study, debonding behaviors of molten paraffin droplets which were impacted and solidified on stainless steel substrates, were observed considering the effects of substrate pre-set temperature, drop height (impact velocity), and droplet temperature. It was found that the debonding is prone to take place at lower substrate pre-set temperatures, lower drop heights, and lower droplet temperatures. A scraping method was then employed to measure the adhesion strength of splats formed under various conditions. The results showed good accordance with debonding behaviors in the drop impact test. The scraping tests also indicated that the residual tensile stress in splats reduces the scraping forces and prevents the complete removal of splats during the scraping processes. Peeling stress and shear stress along the splat-substrate interface, which are driving forces for debonding, were calculated using coupled thermomechanical finite element analyses. The calculated driving forces were larger for lower drop heights and lower droplet temperatures under which the debonding was more easily to occur. The results of numerical simulations coupled with scraping tests provide reasonable explanations for observed debonding behaviors of single splats. This study provides a comprehensive understanding of the effects of several droplet impact variables and residual stress on the debonding behaviors, and the related adhesion strength as well as driving forces of single splats.

With the rapid development of aero-engine manufacturing technology, the dual-rotor system has been employed in part of turbofan engine in order to improve the working performance of aircraft more efficiently. In this study, taking the counter-rotation dual-rotor as the research object, the dynamic model of dual rotor-casing coupling system is established by the aid of MATLAB. The dynamic frequency curves are in good agreement with the results in references and calculated by FEM method, that shows the validity and feasibility of the model. The local rub-impact dynamic model of dual rotor-casing coupling system is established, and rubbing analysis is carried out using Newmark- β method. The effects of rotating speed and speed ratio on local rub-impact response are deeply discussed. The results show that with the increase of rotating speed, combined frequencies and frequency multiplication components are more significant. In addition, speed ratio has a great influence on the periodic motion of the system. With the increase of the absolute value of the speed ratio, the whirl radius of the outer rotor and the normal rubbing force increase dramatically.

Circular histogram thresholding is a new threshold selection method in color image segmentation. However, the method of the existing circular histogram thresholding based on the Otsu Criteria lacks the generality of using the circular histogram. In order to improve the effectiveness and reduce the complexity of thresholding on circular histogram, this paper firstly introduces the Lorenz curve into circular histogram. Then the circular histogram is expanded into the linearized histogram in clockwise or anti-clockwise direction by the optimal index of the Lorenz curve. In the end, the entropy thresholding of the linearized circular histogram is adopted to choose the optimal threshold to obtain the object of color images. Many experimental results show that the proposed method has better effectiveness and adaptability than the existing circular thresholding utilizing Otsu Criteria.

The GRF gene family plays an important role in plant growth and development as regulators involved in plant hormone signaling and metabolism. However, the Juglandaceae GRF gene family remains to be studied. Here, we identified 15, 15, 19, and 20 GRF genes in J. regia, C. illinoinensis, J. sigillata, and J. mandshurica, respectively. The phylogeny shows that the Juglandaceae family GRF is divided into two subfamilies, the ε-group and the non-ε-group, and that selection pressure analysis did not detect amino acid loci subject to positive selection pressure. In addition, we found that the duplications of the Juglandaceae family GRF genes were all segmental duplication events, and a total of 79 orthologous gene pairs and one paralogous homologous gene pair were identified in four Juglandaceae families. The Ka/KS ratios between these homologous gene pairs were further analyzed, and the Ka/KS values were all less than 1, indicating that purifying selection plays an important role in the evolution of the Juglandaceae family GRF genes. The codon bias of genes in the GRF family of Juglandaceae species is weak, and is affected by both natural selection pressure and base mutation, and translation selection plays a dominant role in the mutation pressure in codon usage. Finally, expression analysis showed that GRF genes play important roles in pecan embryo development and walnut male and female flower bud development, but with different expression patterns. In conclusion, this study will serve as a rich genetic resource for exploring the molecular mechanisms of flower bud differentiation and embryo development in Juglandaceae. In addition, this is the first study to report the GRF gene family in the Juglandaceae family; therefore, our study will provide guidance for future comparative and functional genomic studies of the GRF gene family in the Juglandaceae specie.

Plasmonic nanostructures with sharp tips are widely used for optical signal enhancement because of their strong light-confining abilities. These structures have a wide range of potential applications, for example, in sensing, bioimaging, and surface-enhanced Raman scattering. Au nanoparticles, which are important plasmonic materials with high photothermal conversion efficiencies in the visible to near-infrared region, have contributed greatly to the development of photothermal catalysis. However, the existing methods for synthesizing nanostructures with tips need the assistance of poly(vinylpyrrolidone), thiols, or biomolecules. This greatly hinders signal detection because of stubborn residues. Here, we propose an efficient binary surfactant–mediated method for controlling nanotip growth on Au nanoparticle surfaces. This avoids the effects of surfactants and can be used with other Au nanostructures. The Au architecture tip growth process can be controlled well by adjusting the ratio of hexadecyltrimethylammonium bromide to hexadecyltrimethylammonium chloride. This is due to the different levels of attraction between Br−/Cl− and Au3+ ions. The surface-enhanced Raman scattering and catalytic abilities of the synthesized nanoparticles with tips were evaluated by electromagnetic simulation and photothermal catalysis experiments (with 4-nitrothiophenol). The results show good potential for use in surface-enhanced Raman scattering applications. This method provides a new strategy for designing plasmonic photothermal nanostructures for chemical and biological applications.

As the predominant factor influencing the service performance and life cycle of finished parts, surface quality is always improved by polishing technology in the machining process of aero-engine blade. The reasonable selection of polishing process parameter intervals have significant impact on surface quality, which is critically bound up with the performances of finished blade. In this paper, two different characteristic values of surface quality, including morphology feature (surface roughness) and stress property (residual stress), were analyzed respectively. To obtain the global optimal parameter intervals for surface quality improvement in Ti-6Al-4V blade polishing process, the comprehensive analysis methods were presented in this paper. In order to develop the prediction models of surface roughness and residual stress, regression analysis was conducted based on the results of Central-Composite designed polishing experiments. The significance and adequacy of the developed model was verified by using the analysis of variance (ANOVA). The sensitivity analysis was carried out to evaluate the influence of polishing process parameters on surface quality. Subsequently, stability interval and instability interval were divided according to sensitivity curves. Finally, the optimization method of process parameter intervals was proposed and the comprehensive optimal intervals were obtained. The experiments were carried out to verify the developed prediction models and the obtained global optimal parameter intervals. The results of polishing confirmation experiments indicated that the surface-quality-oriented optimal intervals of polishing process parameters were dependable. Context: The possible pathophysiological mechanisms of no-reflow are ischemic injury, reperfusion injury, endothelial injury, distal thromboembolism, etc., Moreover, no-reflow can also remarkably increase the risks of acute myocardial infarction, heart failure, malignant arrhythmia, pericardial tamponade, and even death. Aims: The aim was to evaluate the efficacy of sodium nitroprusside combined with verapamil in the treatment of no-reflow. Settings and Design: From December 2016 to December 2017, 100 patients with no-reflow during coronary intervention in our hospital were selected and randomly divided into two groups: treatment group (50 cases) treated by sodium nitroprusside combined with verapamil and control group (50 cases) treated with sodium nitroprusside treatment alone. Subjects and Methods: The blood flow grading of the two groups according to a thrombolysis test in myocardial infarction (TIMI) was compared and analyzed, and the incidence of cardiovascular adverse events was compared in the 6-month period following the operation. Statistical Analysis Used: SPSS 17.0 (IBM, America) was employed in all calculations. Quantitative data were expressed as mean ± standard deviation, and intergroup comparison was calculated using a two-tailed t-test. Qualitative data were presented in percentages, and intergroup comparison was performed with Chi-square test.P < 0.05 was considered statistically significant. Results: The patients with TIMI Grade III blood flow in the treatment group were significantly more than those in the control group after medication, and the difference was statistically significant (P < 0.05). In the following 6-month period after the operation, the incidence of cardiovascular adverse events in the treatment group was 6.00% (3/50), which was lower than that in the control group 18.0% (9/50), with statistically significant difference (P < 0.05). Conclusions: Sodium nitroprusside combined with verapamil can significantly improve the no-reflow following coronary interventional therapy and reduce the incidence of adverse cardiovascular events.

Erysipelothrix rhusiopathiae is a causative agent of erysipelas in animals and erysipeloid in humans. However, current information regarding E. rhusiopathiae pathogenesis remains limited. Previously, we identified two E. rhusiopathiae strains, SE38 and G4T10, which were virulent and avirulent in pigs, respectively. Here, to further study the pathogenic mechanism of E. rhusiopathiae, we sequenced and assembled the genomes of strains SE38 and G4T10, and performed a comparative genomic analysis to identify differences or mutations in virulence-associated genes. Next, we comparatively analyzed 25 E. rhusiopathiae virulence-associated genes in SE38 and G4T10. Compared with that of SE38, the spaA gene of the G4T10 strain lacked 120 bp, encoding repeat units at the C-terminal of SpaA. To examine whether these deletions or splits influence E. rhusiopathiae virulence, these 120 bp were successfully deleted from the spaA gene in strain SE38 by homologous recombination. The mutant strain ΔspaA displayed attenuated virulence in mice and decreased adhesion to porcine iliac artery endothelial cells, which was also observed using the corresponding mutant protein SpaA’. Our results demonstrate that SpaA-mediated adhesion between E. rhusiopathiae and host cells is dependent on its C-terminal repeat units.

Gastrointestinal toxicity (GIT) is a debilitating side effect of Irinotecan (CPT-11) and limits its clinical utility. Gut dysbiosis has been shown to mediate this side effect of CPT-11 by increasing gut bacterial β-glucuronidase (GUSB) activity and impairing the intestinal mucosal barrier (IMB). We have recently shown the opposing effects of omega-6 (n-6) and omega-3 (n-3) polyunsaturated fatty acids (PUFA) on the gut microbiome. We hypothesized that elevated levels of tissue n-3 PUFA with a decreased n-6/n-3 PUFA ratio would reduce CPT-11-induced GIT and associated changes in the gut microbiome. Using a unique transgenic mouse (FAT-1) model combined with dietary supplementation experiments, we demonstrate that an elevated tissue n-3 PUFA status with a decreased n-6/n-3 PUFA ratio significantly reduces CPT-11-induced weight loss, bloody diarrhea, gut pathological changes, and mortality. Gut microbiome analysis by 16S rRNA gene sequencing and QIIME2 revealed that improvements in GIT were associated with the reduction in the CPT-11-induced increase in both GUSB-producing bacteria (e.g., Enterobacteriaceae) and GUSB enzyme activity, decrease in IMB-maintaining bacteria (e.g., Bifidobacterium), IMB dysfunction and systemic endotoxemia. These results uncover a host–microbiome interaction approach to the management of drug-induced gut toxicity. The prevention of CPT-11-induced gut microbiome changes by decreasing the tissue n-6/n-3 PUFA ratio could be a novel strategy to prevent chemotherapy-induced GIT.

Basic leucine zipper (bZIP), a conserved transcription factor widely found in eukaryotes, has important regulatory roles in plant growth. To understand the information related to the bZIP gene family in walnut, 88 JrbZIP genes were identified at the genome-wide level and classified into 13 subfamilies (A, B, C, D, E, F, G, H, I, J, K, M, and S) using a bioinformatic approach. The number of exons in JrbZIPs ranged from 1 to 12, the number of amino acids in JrbZIP proteins ranged from 145 to 783, and the isoelectric point ranged from 4.85 to 10.05. The majority of JrbZIP genes were localized in the nucleus. The promoter prediction results indicated that the walnut bZIP gene contains a large number of light-responsive and jasmonate-responsive action elements. The 88 JrbZIP genes were involved in DNA binding and nucleus and RNA biosynthetic processes of three ontological categories, molecular functions, cellular components and biological processes. The codon preference analysis showed that the bZIP gene family has a stronger bias for AGA, AGG, UUG, GCU, GUU, and UCU than other codons. Moreover, the transcriptomic data showed that JrbZIP genes might play an important role in floral bud differentiation. The results of a protein interaction network map and kegg enrichment analysis indicated that bZIP genes were mainly involved in phytohormone signaling, anthocyanin synthesis and flowering regulation. qRT-PCR demonstrated the role of the bZIP gene family in floral bud differentiation. Co-expression network maps were constructed for 29 walnut bZIP genes and 6 flowering genes, and JrCO (a homolog of AtCO) was significantly correlated (p < 0.05) with 13 JrbZIP genes in the level of floral bud differentiation expression, including JrbZIP31 (homolog of AtFD), and JrLFY was significantly and positively correlated with JrbZIP10,11,51,59,67 (p < 0.05), and the above results suggest that bZIP family genes may act together with flowering genes to regulate flower bud differentiation in walnut. This study was the first genome-wide report of the walnut bZIP gene family, which could improve our understanding of walnut bZIP proteins and provide a solid foundation for future cloning and functional analyses of this gene family.

Abstract Background Understanding the mechanism of the sexual dimorphism in susceptibility to obesity and metabolic syndrome (MS) is important for the development of effective interventions for MS. Results Here we show that gut microbiome mediates the preventive effect of estrogen (17β-estradiol) on metabolic endotoxemia (ME) and low-grade chronic inflammation (LGCI), the underlying causes of MS and chronic diseases. The characteristic profiles of gut microbiome observed in female and 17β-estradiol-treated male and ovariectomized mice, such as decreased Proteobacteria and lipopolysaccharide biosynthesis, were associated with a lower susceptibility to ME, LGCI, and MS in these animals. Interestingly, fecal microbiota-transplant from male mice transferred the MS phenotype to female mice, while antibiotic treatment eliminated the sexual dimorphism in MS, suggesting a causative role of the gut microbiome in this condition. Moreover, estrogenic compounds such as isoflavones exerted microbiome-modulating effects similar to those of 17β-estradiol and reversed symptoms of MS in the male mice. Finally, both expression and activity of intestinal alkaline phosphatase (IAP), a gut microbiota-modifying non-classical anti-microbial peptide, were upregulated by 17β-estradiol and isoflavones, whereas inhibition of IAP induced ME and LGCI in female mice, indicating a critical role of IAP in mediating the effects of estrogen on these parameters. Conclusions In summary, we have identified a previously uncharacterized microbiome-based mechanism that sheds light upon sexual dimorphism in the incidence of MS and that suggests novel therapeutic targets and strategies for the management of obesity and MS in males and postmenopausal women.

Colorectal cancer: Beneficial bacteria prompt anti-tumor gene response Boosting specific beneficial bacteria in the gut may enhance expression levels of a tumor-suppressing gene in colorectal cancer (CRC). Both genetic factors and the bacteria present in the gut play vital roles in CRC development. However, it is unclear exactly how genes interact with the bacteria to affect tumor growth. Man-tian Mi and co-workers at the Third Military Medical University in Chongqing, China, examined the role of a gene called Sirt-3 in CRC development. Mice lacking the Sirt-3 gene suffered severe chronic inflammation and developed tumors due to altered signalling pathways and reduced intestinal integrity. Further, the guts of the mice harboured more pathogenic bacteria than wild-type mice. The team also found lower levels of two key types of beneficial bacteria that would ordinarily prevent reduced Sirt-3 expression.

Erysipelothrix rhusiopathiae can cause erysipelas in animals and erysipeloid in humans. Since its recurrence in 2012, swine erysipelas has caused serious losses within the pig industry in China. The aim of this study was to perform multilocus sequence typing and understand the virulence and antimicrobial susceptibility of E. rhusiopathiae isolates in China. Multilocus sequence typing (MLST) of a total of 120 strains was performed, and as a result, three different sequence types were identified, of which ST48 was the main one. Five isolates of each MLST type were randomly selected to be used to challenge mice. ST48 was associated with a higher virulence. Antimicrobial susceptibility was tested using a microdilution technique and, to analyze the resistance mechanism, six strains were selected for genome sequencing. A comparison of the six genomes indicated the presence of a suspected macrolide resistance gene, namely, Erm(A)-like, in erythromycin-resistant strains, which increased the minimum inhibitory concentration (MIC) of erythromycin against E. coli C600 at least four-fold. In addition, three mutations (gyrA86T-I, gyrA90D-N, and parC81S-I) were observed in the quinolone resistance-determining regions (QRDRs) of gyrA and parC in quinolone-resistant strains. After the gyrA gene with the 86T-I mutation or the parC gene with the 81S-I mutation was transfected into E. coli C600, the MIC of enrofloxacin against this strain increased at least two-fold. Our findings provide a theoretical basis for developing antibacterial drugs and may contribute to the clinical prevention and control of E. rhusiopathiae.

Chromatography is a powerful and generally applicable method for the analytical separation and quantification of the chemical constituents in complex mixtures because chromatographic separation can provide high selectivity by isolating all analytes from interferences. Multiway analysis based on the multilinear model is an increasingly widely used method for interference-free and fast determination of the chemical constituents also in complex mixtures because multilinear mathematical separation can provide high selectivity by extracting the pure signal of the analyte from the mixed signal of a real sample. By combining chromatographic separation with mathematical separation, multiway calibration method, multiway standard additions method, and multiway internal standard method can be established. Chromatography assisted by multiway analysis can reduce the requirements for complete chromatographic separation, save elution time, and decrease the consumption of the mobile phase, particularly when the peak coelution problem is difficult to solve. This review presents the fundamentals and analytical applications of multilinear mathematical separation in chromatography.

Escherichia coli is a major etiologic agent of cow mastitis, a condition that results in huge economic losses. There is a lack of an oral vaccine for cow mastitis. Previous studies have confirmed that the outer membrane protein A (OmpA) of E. coli is immunogenic and can be used for vaccine design. In the present study, OmpA was encapsulated into nanoparticles (NP-OmpA) for an oral vaccine for cow mastitis. Methods: OmpA was purified with Ni-NTA flow resin and encapsulated with chitosan (CS) to prepare NP-OmpA nanoparticles. The gastrointestinal tract was simulated in vitro (PBS, pH 1.2) to measure the protein release rate. The optimal preparation conditions for NP-OmpA were determined by analyzing the concentrations of OmpA and CS, magnetic mixing speed, mixing time, and the ratio of tripolyphosphate (TPP)/CS (w/w). NP-OmpA safety was assessed by function factors and histopathological examination of livers and kidneys. The immune activity of NP-OmpA was determined using qRT-PCR to assess immune-related gene expression, leukocyte phagocytosis of Staphylococcus aureus, ELISA to evaluate antiserum titer and immune recognition of E. coli, and the organ index. The immune protection function of NP-OmpA was assessed by the protection rate of NP-OmpA to E. coli in mice, qRT-PCR for inflammation-related gene expression, assay kits for antioxidant factors, and visceral injury in the histopathological sections. Results: NP-OmpA nanoparticles had a diameter of about 700 nm, loading efficiency (LE) of 79.27%, and loading capacity (LC) of 20.31%. The release rate of NP-OmpA (0~96 h) was less than 50% in vitro. The optimal preparation conditions for NP-OmpAs were OmpA protein concentration of 2 mg/mL, CS concentration of 5 mg/mL, TPP/CS (w/w) of 1:1, magnetic mixing speed of 150 r/min, and mixing time of 15 min. Histopathological sections and clinical analytes of uric acid (UA), creatinine (Cr), alanine aminotransferase (ALT), aspartate transaminase (AST), catalase (CAT), glutathione (GSH), and malondialdehyde (MDA) showed NP-OmpA did not damage mice livers or kidneys. NP-OmpA could enhance the immune-related gene expression of IFN-γ and HSP70 in the spleen, liver, and kidney and the leukocyte phagocytosis of S. aureus. The antiserum titer (1:3200) was obtained from mice immunized with NP-OmpA, which had an immune recognition effect to E. coli. The immune protection rate of NP-OmpA was 71.43% (p < 0.05) to E. coli. NP-OmpA could down-regulate the inflammation-related gene expression of TNF-a, IL-6, and IL-10 in the spleen, liver, and kidney, and the antioxidant factors MDA and SOD in the liver, and reduce injury in the liver and kidney of mice induced by E. coli. Conclusions: A novel NP-OmpA nanoparticle was encapsulated, and the optimal preparation conditions were determined. The NP-OmpA was safe and had good immune functions. They are expected to induce a response that resists infection with the major etiologic agent (E. coli) of cow mastitis.

The fluorescent carbon dot is a novel type of carbon nanomaterial. In comparison with semiconductor quantum dots and fluorescence organic agents, it possesses significant advantages such as excellent photostability and biocompatibility, low cytotoxicity and easy surface functionalization, which endow it a wide application prospect in fields of bioimaging, chemical sensing, environmental monitoring, disease diagnosis and photocatalysis as well. Biomass waste is a good choice for the production of carbon dots owing to its abundance, wide availability, eco-friendly nature and a source of low cost renewable raw materials such as cellulose, hemicellulose, lignin, carbohydrates and proteins, etc. This paper reviews the main sources of biomass waste, the feasibility and superiority of adopting biomass waste as a carbon source for the synthesis of carbon dots, the synthetic approaches of carbon dots from biomass waste and their applications. The advantages and deficiencies of carbon dots from biomass waste and the major influencing factors on their photoluminescence characteristics are summarized and discussed. The challenges and perspectives in the synthesis of carbon dots from biomass wastes are also briefly outlined.

The correction of wavefront aberration plays a vital role in active optics. The traditional correction algorithms based on the deformation of the mirror cannot effectively deal with disturbances in the real system. In this study, a new algorithm called deep learning correction algorithm (DLCA) is proposed to compensate for wavefront aberrations and improve the correction capability. The DLCA consists of an actor network and a strategy unit. The actor network is utilized to establish the mapping of active optics systems with disturbances and provide a search basis for the strategy unit, which can increase the search speed; The strategy unit is used to optimize the correction force, which can improve the accuracy of the DLCA. Notably, a heuristic search algorithm is applied to reduce the search time in the strategy unit. The simulation results show that the DLCA can effectively improve correction capability and has good adaptability. Compared with the least square algorithm (LSA), the algorithm we proposed has better performance, indicating that the DLCA is more accurate and can be used in active optics. Moreover, the proposed approach can provide a new idea for further research of active optics.

When the dynamic characteristics of a bridge structure are analyzed though Hilbert–Huang transform (HHT), the noise contained in the bridge dynamic monitoring data may seriously affect the performance of the first natural frequency identification. A time-frequency analysis method that integrates wavelet threshold denoising and HHT is proposed to overcome this deficiency. The denoising effect of the experimental analysis on the simulated noisy signals proves the effectiveness of the proposed method. This method is used to perform denoising pre-processing on the dynamic monitoring data of Sutong Bridge, and the denoised results of different methods are compared and analyzed. Then, the best denoising data are selected as the input data of Hilbert spectrum analysis to identify the structural first natural frequency of the bridge. The results indicate that the wavelet-empirical mode decomposition (EMD) method effectively reduces the interference of random noise and eliminates useless intrinsic modal function (IMF) components, and the excellent properties of the signal evaluation index after denoising make the method suitable for processing non-stationary signals with noise. When Hilbert spectrum analysis is applied to the denoised data, the first natural frequency of the bridge structure can be identified clearly and is consistent with the theoretical calculation. The proposed method can effectively determine the natural vibration characteristics of the bridge structure.

As there is increasing usage of composite components with a closed-end shape, filament winding technology has been applied extensively in engineering practice. However, the residual stresses are readily caused by process parameters in composite manufacturing process. Hence, it is necessary to design the reasonable process parameters for obtaining the expected residual stresses. This study proposed a reverse derivation method to design the winding tension based on the residual stress model of the composite cylinder. To analyze the development of residual stresses, the thermal deformation behavior and a micromechanics model of resin shrinkage are considered to calculate thermal strains and instantaneous shrinkage strains of the hoop wound cylinder during the curing process. Combining the contribution of winding tension to the stresses, the simple model of residual stresses is established based on the superposition principle. Then inversely solving the analytic model, the design method of winding tension is illustrated to guarantee that the hoop wound cylinder with an inner liner has a uniform residual hoop stress. The model accuracy of residual stresses is validated by the destructive slitting experiment. The result indicates that the present model can be used to estimate the residual stresses and design the winding tension for the hoop wound cylinder.

ABSTRACT Metabolic endotoxemia originating from dysbiotic gut microbiota has been identified as a primary mediator for triggering the chronic low-grade inflammation (CLGI) responsible for the development of obesity. Capsaicin (CAP) is the major pungent bioactivator in chili peppers and has potent anti-obesity functions, yet the mechanisms linking this effect to gut microbiota remain obscure. Here we show that mice fed a high-fat diet (HFD) supplemented with CAP exhibit lower levels of metabolic endotoxemia and CLGI associated with lower body weight gain. High-resolution responses of the microbiota were examined by 16S rRNA sequencing, short-chain fatty acid (SCFA) measurements, and phylogenetic reconstruction of unobserved states (PICRUSt) analysis. The results showed, among others, that dietary CAP induced increased levels of butyrate-producing Ruminococcaceae and Lachnospiraceae, while it caused lower levels of members of the lipopolysaccharide (LPS)-producing family S24_7. Predicted function analysis (PICRUSt) showed depletion of genes involved in bacterial LPS synthesis in response to CAP. We further identified that inhibition of cannabinoid receptor type 1 (CB1) by CAP also contributes to prevention of HFD-induced gut barrier dysfunction. Importantly, fecal microbiota transplantation experiments conducted in germfree mice demonstrated that dietary CAP-induced protection against HFD-induced obesity is transferrable. Moreover, microbiota depletion by a cocktail of antibiotics was sufficient to block the CAP-induced protective phenotype against obesity, further suggesting the role of microbiota in this context. Together, our findings uncover an interaction between dietary CAP and gut microbiota as a novel mechanism for the anti-obesity effect of CAP acting through prevention of microbial dysbiosis, gut barrier dysfunction, and chronic low-grade inflammation. IMPORTANCE Metabolic endotoxemia due to gut microbial dysbiosis is a major contributor to the pathogenesis of chronic low-grade inflammation (CLGI), which primarily mediates the development of obesity. A dietary strategy to reduce endotoxemia appears to be an effective approach for addressing the issue of obesity. Capsaicin (CAP) is the major pungent component in red chili (genus Capsicum). Little is known about the role of gut microbiota in the anti-obesity effect of CAP. High-throughput 16S rRNA gene sequencing revealed that CAP significantly increased butyragenic bacteria and decreased LPS-producing bacteria (e.g., members of the S24-7 family) and LPS biosynthesis. By using antibiotics and microbiota transplantation, we prove that gut microbiota plays a causal role in dietary CAP-induced protective phenotype against high-fat-diet-induced CLGI and obesity. Moreover, CB1 inhibition was partially involved in the beneficial effect of CAP. Together, these data suggest that the gut microbiome is a critical factor for the anti-obesity effects of CAP.

Erysipelothrix rhusiopathiae (E. rhusiopathiae) is the causative agent of swine erysipelas. This microbe has caused great economic losses in China and in other countries. In this study, high-throughput cDNA microarray assays were employed to evaluate the host responses of porcine heart to E. rhusiopathiae and to gain additional insights into its pathogenesis. A total of 394 DE transcripts were detected in the active virulent E. rhusiopathiae infection group compared with the PBS group at 4 days post-infection. Moreover, 262 transcripts were upregulated and 132 transcripts were downregulated. Differentially expressed genes were involved in many vital functional classes, including inflammatory and immune responses, signal transduction, apoptosis, transport, protein phosphorylation and dephosphorylation, metabolic processes, chemotaxis, cell adhesion, and innate immune responses. Pathway analysis demonstrated that the most significant pathways were Chemokine signaling pathway, NF-kappa B signaling pathway, TLR pathway, CAMs, systemic lupus erythematosus, chemokine signaling pathway, Cytokine-cytokine receptor interaction, PI3K-Akt signaling pathway, Phagosome, HTLV-I infection, Measles, Rheumatoid arthritis and natural-killer-cell-mediated cytotoxicity. The reliability of our microarray data was verified by performing quantitative real-time PCR. This study is the first to document the response of piglet heart to E. rhusiopathiae infection. The observed gene expression profile could help screen potential host agents that can reduce the prevalence of E. rhusiopathiae. The profile might also provide insights into the underlying pathological changes that occur in pigs infected with E. rhusiopathiae.

The effectiveness of traditional Chinese medicine (TCM) against various diseases urges more low cost, speed and sensitive analytical methods for investigating the phamacology of TCM and providing a theoretical basis for clinical use. The potential of second-order calibration method was validated for the quantification of two effective ingredients of Schisandra chinensis in human plasma using spectrofluorimetry. The results obtained in the present study demonstrate the advantages of this strategy for multi-target determination in complex matrices. Although the spectra of the analytes are similar and a large number of interferences also exist, second-order calibration method could predict the accurate concentrations together with reasonable resolution of spectral profiles for analytes of interest owing to its âsecond-order advantageâ. Moreover, the method presented in this work allows one to simply experimental procedure as well as reduces the use of harmful chemical solvents. The composite tape winding process, which utilizes a tape winding machine and prepreg tapes, provides a promising way to improve the quality of composite products. Nevertheless, the process parameters of composite tape winding have crucial effects on the tensile strength and void content, which are closely related to the performances of the winding products. In this article, two different object values of winding products, including mechanical performance (tensile strength) and a physical property (void content), were respectively calculated. Thereafter, the paper presents an integrated methodology by combining multi-parameter relative sensitivity analysis and single-parameter sensitivity analysis to obtain the optimal intervals of the composite tape winding process. First, the global multi-parameter sensitivity analysis method was applied to investigate the sensitivity of each parameter in the tape winding processing. Then, the local single-parameter sensitivity analysis method was employed to calculate the sensitivity of a single parameter within the corresponding range. Finally, the stability and instability ranges of each parameter were distinguished. Meanwhile, the authors optimized the process parameter ranges and provided comprehensive optimized intervals of the winding parameters. The verification test validated that the optimized intervals of the process parameters were reliable and stable for winding products manufacturing.
