ABSTRACT Wheel slipping and fuel consumption are variables influenced by tire pressure, speed, ballasting, and soil surface. Once the tractor is present in all agricultural operations, knowledge of these variables is essential to achieving the most operational efficiency. This study aimed to develop an embedded system to collect data and show tractor slipping and fuel consumption in real-time via smartphone. The system was developed using prototyping platforms and validated through randomized field tests under four speeds and two terrain conditions. Three methods for determining slippage under different conditions were compared. The developed embedded system determined an agricultural tractor's slipping and fuel consumption conditions. The slip and fuel consumption rates varied depending on the type of rolling surface (ground preparations) and the tractor's operating speeds. Conventional tillage surface increased fuel demand by 6% and slippage by 17% compared to no-till rates. The proposed embedded system found similar results and could present the slipping values in the developed Android to real-time operation compared to a conventional method. This application shows both slipping and fuel consumption values over time on graphs, which can be very useful during machine operation, helping to define better configuration and reduce environmental impacts.

The evolving prevalence of anabolic androgenic steroids (AAS) abuse among nonathletes is alarming because of the known harm to an individual’s health. Among the adverse effects of AAS abuse, male infertility and sexual dysfunction have been often reported in the literature, but little is known regarding its actual prevalence, possible underpinning mechanisms, and potential treatments either during or post-AAS usage. Thus, the current narrative review summarizes the state-of-art regarding the effects of AAS on male fertility and sexual function. Evidence was gathered from the latest reviews and recent original studies, specifically from prospective cohorts and clinical trials, ultimately resulting in five main topics of discussion. First, AAS usage is briefly characterized by its historical background, main physiological mechanisms, and the most frequently used AAS substances. Second, data on the prevalence of AAS-induced male infertility and sexual dysfunction are described. Third, some new insights on possible underpinning mechanisms of AAS-induced male infertility and sexual dysfunction are thoroughly discussed, with particular attention to histological data derived from animal models and the latest insights from prospective cohorts in humans. Fourth, the potential treatments during and after the AAS usage are presented, highlighting the odds of resolving male infertility and sexual dysfunction. Fifth, future directions on this topic are discussed, focusing on the methodological robustness of scientific studies.

This study presents the dielectric properties of a barium titanate–gadolinium ferrite composite material, obtained through a solid-state reaction method. The aim of this research was to create a composite material with enhanced dielectric properties compared to each individual component, and to investigate the electrical properties of the composites, using impedance spectroscopy. The structural and morphologic properties were analyzed using X-ray diffraction and scanning electron microscopy, respectively. Impedance spectroscopy measurements were performed over a wide frequency range (100–0.1 GHz) and temperature (45–170 °C) to evaluate the electrical behavior of the material. The dielectric relaxations were analyzed using the Havriliak–Negami function, and the key electrical parameters such as relaxation frequency, dielectric strength, and electrical conductivity were extracted. Several relaxation processes were identified, which depend on the mixture of the initial titanate and ferrite materials, and a correlation between structural, morphologic, and electrical properties was exposed. The sample with the highest dielectric constant was the 25 wt% gadolinium ferrite composite, with ε′ close to 240 and loss tangent values below 0.1, affording it the more appropriate composition for energy storage devices such as lead-free dielectric capacitors.

BackgroundThe co-circulation of flaviviruses in tropical regions has led to the hypothesis that immunity generated by a previous dengue infection could promote severe disease outcomes in subsequent infections by heterologous serotypes. This study investigated the influence of antibodies generated by previous Zika infection on the clinical outcomes of dengue infection.Methodology/principal findingsWe enrolled 1,043 laboratory confirmed dengue patients and investigated their prior infection to Zika or dengue. Severe forms of dengue disease were more frequent in patients with previous Zika infection, but not in those previously exposed to dengue.Conclusions/significanceOur findings suggest that previous Zika infection may represent a risk factor for subsequent severe dengue disease, but we did not find evidence of antibody-dependent enhancement (higher viral titer or pro-inflammatory cytokine overexpression) contributing to exacerbation of the subsequent dengue infection.

Plain language summary SARS-CoV-2, the virus responsible for COVID-19, has evolved into different variants, virus lineages with a slightly different genetic material. Here, we evaluated the introductions of SARS-CoV-2 lineages in a Brazilian city with the third-highest number of confirmed COVID-19 cases in the state of São Paulo. Our analyses revealed several SARS-CoV-2 lineages first detected in the city, but after the introduction of the Gamma lineage, the majority of them were replaced by this variant that became the most common. At the same time, there was an increase in the number of infections, severe disease cases, and deaths, mainly in individuals under 70 years old, the age group that was not fully vaccinated at the time. Our findings further support the evidence that vaccination effectively decreases SARS-CoV-2 spread and transmission.

Abstract Background The purpose of this systematic review was to examine the associations between school-related sedentary behaviours and indicators of health and well-being in children and youth (~ 5–18 years) attending school. Methods This review was conducted to inform the development of School-Related Sedentary Behaviour Recommendations. Peer-reviewed, published, or in-press articles in English were included. Reviews, meta-analyses, and case studies were excluded; all other study designs were eligible. Further, articles had to meet the a priori study criteria for population, intervention, comparator (PROSPERO ID: CRD42021227600). Embase, MEDLINE® ALL, and PsycINFO were searched. Risk of bias was assessed for individual experimental studies using the Cochrane risk of bias assessment tool, and in observational studies based on the GRADE framework and in line with previous systematic reviews examining sedentary behaviours in children. Overall quality of evidence was assessed using the GRADE framework for each outcome category and study design. Results were synthesized narratively, grouped by study design and outcome category. Further, several high-level summaries were conducted to help interpret results. Results Evidence was synthesized from 116 reports, including 1,385,038 participants and 1173 extracted associations. More school-related sedentary behaviour was favourably associated with nearly one-third of extracted associations for cognitive (33%) and social-emotional (32%) indicators (e.g., less anxiety), but unfavourably associated with other movement behaviours (e.g., less physical activity) (35%). Active lessons were favourable (72%), compared to more school-related sedentary behaviours, when examining associations for all health and well-being indicators. More homework was favourable across all health and well-being indicators in 4% of extracted associations for primary school children, and 25% of extracted associations for secondary school children. However, ≥2 h/day of homework appeared to be unfavourable for health and well-being. Limitations for synthesized studies included generally low quality of evidence and a lack of studies in South American, African, or low-middle income countries. Conclusions Findings can help inform policy makers, schools, and teachers, regarding the amount of homework assigned and the introduction of active lessons into the classroom to enhance health and well-being of children. More research is needed examining school-related sedentary behaviours and indicators of health and well-being in low- and middle-income countries.

Abstract Background Existing sedentary behaviour guidelines for children and youth target overall sedentary behaviour and recreational screen time, without any specific recommendations regarding school-related sedentary behaviours (i.e., sedentary behaviours performed during the school day, or within the influence of school). The purpose of this paper is to describe the development of international evidence-based recommendations for school-related sedentary behaviours for children and youth, led by the Sedentary Behaviour Research Network (SBRN). Methods A panel of international experts was convened by SBRN in November 2020 to guide the development of these recommendations for children and youth aged ~ 5–18 years. The recommendations were informed by 1) age-relevant existing sedentary behaviour guidelines, 2) published research on the relationship between overall sedentary behaviour and health, 3) a de novo systematic review on the relationship between school-related sedentary behaviours and health and/or academic outcomes, and 4) a de novo environmental scan of the grey literature to identify existing recommendations for school-related sedentary behaviours. Draft recommendations were presented to the Expert Panel in June 2021. Following thorough discussion and modifications, updated recommendations were distributed for stakeholder feedback from July 9–26. Feedback was received from 148 stakeholders across 23 countries, leading to additional updates to the recommendations. Following further rounds of discussion and updates with the Expert Panel in August and September 2021, consensus was achieved on the final recommendations. Results A healthy day includes breaking up extended periods of sedentary behaviour and incorporating different types of movement into homework whenever possible, while limiting sedentary homework. School-related screen time should be meaningful, mentally or physically active, and serve a specific pedagogical purpose that enhances learning. Replacing sedentary learning activities with movement-based learning activities, and replacing screen-based learning activities with non-screen-based learning activities, can further support students’ health and wellbeing. Discussion This paper presents the first evidence-based recommendations for school-related sedentary behaviours for children and youth. These recommendations will support the work of parents, caregivers, educators, school system administrators, policy makers, researchers and healthcare providers interested in promoting student health and academic success.

Physical activity (PA) and sedentary behaviour (SED) have been linked to the mental health of children and adolescents, yet the timing of behaviours may play a role in this relationship and clarifying this could inform interventions. We explored cross-sectional associations of PA and SED in varying time segments throughout the school day with the mental health of school-aged children and adolescents from rural Northeastern Ontario, Canada. A total of 161 students (56% female, M = 10.3 years old) wore accelerometers for 8 days (7 nights) and completed a self-report survey (parent reported for children younger than 11). Mental health was measured using the Strengths and Difficulties Questionnaire. Accelerometer-measured SED, light PA (LPA), and moderate and vigorous PA (MVPA) were estimated in the time-segments before school (06:00–08:44), school time (08:45–15:04), after school (15:05–16:59), and evenings (17:00–21:59). Associations were tested with multilevel linear regressions while adjusting for confounding factors. Students spent 72.6 min in MVPA, 209.0 min in LPA, and 621.0 min in SED per day. Dengue is a serious mosquito-transmitted disease caused by the dengue virus (DENV). Rapid and reliable diagnosis of DENV infection is urgently needed in dengue-endemic regions. We describe here the performance evaluation of the CE-marked VIDAS® dengue immunoassays developed for the automated detection of DENV NS1 antigen and anti-DENV IgM and IgG antibodies. A multicenter concordance study was conducted in 1296 patients from dengue-endemic regions in Asia, Latin America, and Africa. VIDAS® dengue results were compared to those of competitor enzyme-linked immunosorbent assays (ELISA). The VIDAS® dengue assays showed high precision (CV ≤ 10.7%) and limited cross-reactivity (≤15.4%) with other infections. VIDAS® DENGUE NS1 Ag showed high positive and negative percent agreement (92.8% PPA and 91.7% NPA) in acute patients within 0–5 days of symptom onset. VIDAS® Anti-DENGUE IgM and IgG showed a moderate-to-high concordance with ELISA (74.8% to 90.6%) in post-acute and recovery patients. PPA was further improved in combined VIDAS® NS1/IgM (96.4% in 0–5 days acute patients) and IgM/IgG (91.9% in post-acute patients) tests. Altogether, the VIDAS® dengue NS1, IgM, and IgG assays performed well, either alone or in combination, and should be suitable for the accurate diagnosis of DENV infection in dengue-endemic regions.

Automatized scalable healthcare support solutions allow real-time 24/7 health monitoring of patients, prioritizing medical treatment according to health conditions, reducing medical appointments in clinics and hospitals, and enabling easy exchange of information among healthcare professionals. With recent health safety guidelines due to the COVID-19 pandemic, protecting the elderly has become imperative. However, state-of-the-art health wearable device platforms present limitations in hardware, parameter estimation algorithms, and software architecture. This paper proposes a complete framework for health systems composed of multi-sensor wearable health devices (MWHD), high-resolution parameter estimation, and real-time monitoring applications. The framework is appropriate for real-time monitoring of elderly patients' health without physical contact with healthcare professionals, maintaining safety standards. The hardware includes sensors for monitoring steps, pulse oximetry, heart rate (HR), and temperature using low-power wireless communication. In terms of parameter estimation, the embedded circuit uses high-resolution signal processing algorithms that result in an improved measure of the HR. The proposed high-resolution signal processing-based approach outperforms state-of-the-art HR estimation measurements using the photoplethysmography (PPG) sensor.

12C fusion reactions are among the most important in stellar evolution since they determine the destiny of massive stars. Over the past fifty years, massive efforts have been done to measure these reactions at low energies. However, existing data present several discrepancies between sets and large uncertainties specially at the lowest energies. Factors such as beam/environmental backgrounds, extremely low cross sections and insufficient knowledge of the reaction mechanism contribute to these problems. Recently, the ERNA collaboration measured the 12C+12C reactions at Ec.m. = 2.51 - 4.36 MeV with energy steps between 10 and 25 keV in the centre of mass. Representing the smallest energy steps to date. In these measurements, beam induced background was minimised and S-factors for the proton and alpha channels were calculated. Results indicate that a possible explanation for the discrepancies between data sets is the wrongly assumed constant branching ratios and isotropical angular distributions. Given the excellent performance of the detectors for low energy measurements, a collaboration with the LUNA group (LNGS) has started. Background measurements underground are being performed and results indicate it could be possible to measure the 12C+12C reactions directly into the Gamow Window.

Hydrogels are smart-swelling 3D structures capable of incorporating/expelling water while maintaining their structures. When combined with electroactive materials, such as conducting polymers, the resulting composite may present tunable properties. Herein, the preparation and characterization of alginate-polypyrrole composite hydrogels is described using chemical polymerization to form polypyrrole inside and around alginate beads, employing two simple protocols. These materials were qualitatively tested as extraction phases, using the solid-phase extraction technique, for the pre-concentration of contaminants of emerging concern (atrazine, caffeine, and progesterone). Compared to alginate alone, the composite materials showed a modified extraction capacity, especially for the extraction of progesterone. It was shown that the alginate matrix also contributes to the extraction, not only acting as a support but also as an active extraction media, evidencing a good combination of materials.

Arbovirus infections are increasingly important causes of disease, whose spectrum of neurological manifestations are not fully known. This study sought to retrospectively assess the incidence of arboviruses in cerebrospinal fluid samples of patients with neurological symptoms to inform diagnosis of central and peripheral nervous system disorders. A total of 255 cerebrospinal fluid (CSF) samples collected from January 2016 to December 2017 were tested for dengue virus (DENV 1-4), Zika virus (ZIKV), and Chikungunya virus (CHIKV) in addition to other neurotropic arboviruses of interest, using genetic and serologic assays. Of the 255 CSF samples analyzed, 3.53% (09/255) were positive for arboviruses presenting mainly as meningitis, encephalitis, and cerebrovascular events, of which ZIKV was detected in 2.74% (7/255), DENV in 0.78% (2/255), in addition to an identified ILHV infection that was described previously. All the cases were detected in adults aged 18 to 74 years old. Our findings highlight the scientific and clinical importance of neurological syndromes associated with arboviruses and demonstrate the relevance of specific laboratory methods to achieve accurate diagnoses as well as highlight the true dimension of these diseases to ultimately improve public health planning and medical case management.

Schools have been the main context for physical activity (PA) and sedentary behavior (SB) interventions among adolescents, but there is inconsistent evidence on whether they also improve dimensions of the health−related quality of life (HRQoL). The aim of this study was to evaluate the effects of a school-based active lifestyle intervention on dimensions of HRQoL. A secondary aim was to verify whether sex, age, and HRQoL at baseline were moderators of the intervention effect. A cluster-randomized controlled trial was conducted at three control and three intervention schools in Florianopolis, Brazil. All students from 7th to 9th grade were invited to participate. A school year intervention, designed primarily to increase PA and reduce SB, included strategies focused on (i) teacher training on PA, SB, and nutrition, and availability of teaching materials related to these contents; (ii) environmental improvements (i.e., creation and revitalization of spaces for the practice of PA in school); and (iii) education strategies, with the availability of folders and posters regarding PA, SB, and nutrition. Participants and the research staffs were not blinded to group assignment, but a standardized evaluation protocol was applied at baseline and after the intervention (March and November 2017) using the KIDSCREEN−27 to assess HRQoL across five dimensions. Mixed linear models were performed to evaluate the effect of the Movimente intervention on the five HRQoL dimensions. Of the 921 students who answered the questionnaire at baseline, 300 and 434 completed the study in control and intervention groups, respectively (dropouts: 20%). The results revealed no significant effects of the intervention on any HRQoL dimensions. A reduction of the school environment dimension was observed in both the control (-2.44; 95% CI: -3.41 to -1.48) and intervention groups (-2.09; 95% CI: -2.89 to -1.30). Sensitivity analyses showed that students in the highest baseline tertiles of HRQoL in any dimension had a reduction in their respective scores from pre- to post-intervention in both school groups. In conclusion, our results demonstrated no intervention effect on HRQoL dimensions and those students with the highest levels of HRQoL at baseline on all dimensions reduced from pre to post-intervention.Clinical Trial RegistrationThe trial is registered at the Clinical Trial Registry (Trial ID: NCT02944318; date of registration: October 18, 2016).

The process of identifying and managing Information and Communication Technology (ICT) risks has become a concern and a challenge for public and private organizations. In this context, risk management methodologies within the Brazilian Federal Public Administration organizations have become indispensable to help the managers of these organizations in decision making, especially in the distribution of public funds, elaboration of public policies focused on transparency, social actions contemplating indemnities, and social benefits, among others. In addition, the various ICT projects controlled by the public administration need a methodology to perform their management of ICT resources. In this article, we present the Governance and Risk Management methodology used to model the Administrative Council for Economic Defense (CADE) macro processes. The proposed methodology used the risk management process aligned to the ISO 31000 standards. This alignment was necessary for mapping CADE’s risk events, regardless of their complexity. The modeled ICT risk processes will support the organization’s managers in decision making and may be used or customized by any other organization of the Brazilian Federal Public Administration.

BackgroundThe present cross-sectional study aimed to determine the proportion of adolescents meeting the 24-hour movement guidelines, and investigate sociodemographic factors associated with meeting them.MethodsSelf-reported (average daily volume of MVPA, sleep duration, and time watching videos and playing videogames) and accelerometer-measured (MVPA and sleep duration) 24-hour movement behaviors were classified according to recommendations, and sex, age, socioeconomic status (SES), family structure, parental education, and number of people in the household were tested as correlates of meeting recommendations using multilevel logistic regressions.ResultsThe proportion of adolescents (n = 867, mean age: 16.4 years, 50.3% girls) meeting the MVPA, ST, and sleep duration guidelines was of 25%, 28%, and 41%, respectively, for self-reported data. From accelerometer data (n = 688), 7.1% met MVPA and 31.7% met sleep duration recommendations. Adherence to all three recommendations was 3% with self-report and 0.2% with accelerometer data. Boys were more likely to meet MVPA, but not ST and sleep-duration recommendations. A positive relationship was observed between age and meeting the ST recommendation.ConclusionsAdherence to the sleep duration recommendation was higher than to the screen-time and MVPA recommendations and few in this sample of Brazilian adolescents achieved the 24-hour guidelines. Efforts are needed to improve 24-hour movement behaviors.

12C+12C reactions are crucial in the evolution of massive stars and explosive scenarios. The measurement of these reactions at astrophysical energies is very challenging due to their extremely small cross sections, and the presence of beam induced background originated by the natural 1,2H contaminants in the C targets. In addition, the many discrepancies between different data sets and the complicated resonant structure of the cross sections make the extrapolation to low energies very uncertain. Recently, we performed a direct measurement of the 12C+12C reactions at the CIRCE Laboratory in Italy. Results from a study on target contamination were used, allowing us to measure cross sections at Ec.m. =2.51 − 4.36 MeV with 10-25 keV energy steps. Two stage ΔE-Erest detectors were used for unambiguous particle identification. Branching ratios of individual particle groups were found to vary significantly with energy and angular distributions were also found to be anisotropic, which could be a potential explanation for the discrepancies observed among different data sets.

Low-code is an emerging concept that transforms visual representations into functional software, allowing anyone to be a developer. However, building a low-code platform from scratch can be challenging concerning the scarce available literature about the topic. In this sense, this paper proposes an Open Low-Code Platform (OLP), a low-code solution that enables regular users to create applications. Furthermore, it presents low-code’s functional and nonfunctional requirements, as well as its similarities and its differences with the no-code concept. The experience obtained while developing OLP was translated into a pipeline that details how code was transformed from the visual representations into a fully fledged application. The paper demonstrates the solution’s viability and is especially useful for building a low-code platform from scratch or improving an existing one.

Swainsonine—a cytotoxic fungal alkaloid and a potential cancer therapy drug—is produced by the insect pathogen and plant symbiont Metarhizium robertsii, the clover pathogen Slafractonia leguminicola, locoweed symbionts belonging to Alternaria sect. Undifilum, and a recently discovered morning glory symbiont belonging to order Chaetothyriales. Genome sequence analyses revealed that these fungi share orthologous gene clusters, designated “SWN,” which included a multifunctional swnK gene comprising predicted adenylylation and acyltransferase domains with their associated thiolation domains, a β-ketoacyl synthase domain, and two reductase domains. The role of swnK was demonstrated by inactivating it in M. robertsii through homologous gene replacement to give a ∆swnK mutant that produced no detectable swainsonine, then complementing the mutant with the wild-type gene to restore swainsonine biosynthesis. Other SWN cluster genes were predicted to encode two putative hydroxylases and two reductases, as expected to complete biosynthesis of swainsonine from the predicted SwnK product. SWN gene clusters were identified in six out of seven sequenced genomes of Metarhzium species, and in all 15 sequenced genomes of Arthrodermataceae, a family of fungi that cause athlete’s foot and ringworm diseases in humans and other mammals. Representative isolates of all of these species were cultured, and all Metarhizium spp. with SWN clusters, as well as all but one of the Arthrodermataceae, produced swainsonine. These results suggest a new biosynthetic hypothesis for this alkaloid, extending the known taxonomic breadth of swainsonine producers to at least four orders of Ascomycota, and suggest that swainsonine has roles in mutualistic symbioses and diseases of plants and animals.

Background and purpose — The length of stay after total hip arthroplasty has been reduced to 2–4 days after implementing fast-track surgery. We investigated whether a new time-based patient-centered primary direct anterior approach (DAA) total hip arthroplasty (THA) treatment protocol in a specialized clinic, with a planned length of stay of about 24 hours, could be achieved in all patients or only in a selected group of patients. Patients and methods — We analyzed prospectively collected data in a cohort of 378 consecutive patients who underwent a primary direct anterior THA as a patient-centered time-based procedure between March 1, 2012 and December 31, 2015. Patients with complicated medical comorbidity and those over the age of 85 were excluded from the study. The average length of stay was recorded and all complications, re-admissions, and reoperations were registered and analyzed. The primary outcome measures were length of stay and complication rate, at discharge and 90 days postoperatively. Results — The average length of stay for all patients was 26 hours. All patients were discharged from the clinic on the day after the operation and were able to continue their recovery at home or in a rehabilitation facility. The overall complication rate within 3 months of surgery was 6%. The 3-month re-admission rate and the 3-month reoperation rate were both 2%. Interpretation — Performing a time-based, patient-centered fast-track program for DAA total hip arthroplasty can result in a standardized length of stay of about 24 hours and a high level of patient satisfaction with few complications, re-admissions, and reoperations.

Ilheus virus is an arbovirus with the potential for central nervous system involvement. Accurate diagnosis is a challenge due to similar clinical symptoms and serologic cross-reactivity with other flaviviruses. Here, we describe the first documented case of a fatal outcome following the identification of Ilheus virus in the cerebrospinal fluid (CSF) of a patient with cerebral encephalitis in Brazil.

Arthropod-borne viruses (arboviruses) of the genus Flavivirus are distributed globally and cause significant human disease and mortality annually. Flavivirus infections present a spectrum of clinical manifestations, ranging from asymptomatic to severe manifestations, including hemorrhage, encephalitis and death. Herein, we describe 3 case reports of cerebrovascular involvement in patients infected by dengue and Zika viruses in Sao Jose do Rio Preto, São Paulo State, Brazil, a hyperendemic area for arbovirus circulation, including dengue, yellow fever, chikungunya and Saint Louis encephalitis viruses. Our findings highlight the potential threat that unusual clinical manifestations may pose to arbovirus disease management and recovery.

Os processos de Gerenciamento Costeiro (GC) nas últimas décadas vêm evoluindo, apresentando diferentes métodos de gestão, sendo que uma nova fronteira se encontra na Gestão com Base Ecossistêmica (GBE). No entanto, para colocar em prática a GBE, a qual leva em consideração as funções, os processos e os serviços ecossistêmicos dos ambientes costeiros e marinhos, entendendo-os como um conjunto de ecossistemas compostos por elementos ecológicos (naturais), econômicos e sociais, se faz necessária a base de informação ecossistêmica. O presente trabalho propõe, apresentando resultados aplicados, um roteiro metodológico de seis etapas: 1. Identificar os ecossistemas como “Unidades de Gestão”; 2. Mapear, modelar e simular os ecossistemas; 3. Identificar e classificar os serviços ecossistêmicos; 4. Definir os valores e a qualidade dos serviços; 5. Identificar os espaços de gestão; e 6. Integrar com políticas e demais instrumentos de gestão e legais. As aplicações práticas apresentadas vão desde trabalhos acadêmicos de identificação e caracterização de ambientes costeiros e marinhos a aplicações nos processos de gestão ambiental portuários, passando também pelo desenvolvimento de Zoneamentos Ecológico-Econômicos (ZEEs) em nível regional, por exemplo. Na apreciação integral do conjunto de exemplos e iniciativas para todas as etapas do modelo, depreende-se que a sua aplicação é ampla, variada e consideravelmente simples. Da mesma forma, a multiplicidade de possíveis aplicações do modelo em ações voltadas ao suporte de uma GBE sugere que seu uso pode crescer e buscar iniciativas inovadoras. A expectativa dos autores é de que tal ferramenta possa, de fato, delinear e estimular pesquisas e aplicações com base ecossistêmica na busca da sustentabilidade da costa e do bem-estar de seus atores sociais.

Os imbróglios que permeiam a educação não se esgotam nos já conhecidos embates entre o poder público e a classe educacional. Uma das dimensões mais danosas da miséria à qual a educação está submetida reside na educação menor, no papel do professor, na sua formação e na sua postura perante os outros, no caso seus alunos. Para a promoção de alguma transformação nas relações que orbitam esse pequeno universo, lança-se mão de uma possível sintonia entre o Anarquismo Epistemológico de Paul Feyerabend e o Rizoma de Gilles Deleuze e Félix Guattari. Tendo a Interdisciplinaridade como fim e as concepções de Feyerabend, Deleuze e Guattari como meio, este trabalho lança um novo desafio: fazer uma reflexão sobre os problemas educacionais tendo como ponto de partida e como ponto de chegada o professor. A formação de uma consciência de que a forma (método) e o conteúdo (conhecimento) não devem acorrentar o sujeito, mas serem superados quando se impõem como obstáculos à ação de espíritos livres.

A new extraction phase based on hydrogel disks of polyvinyl alcohol (PVOH) and pectin was proposed, characterized and evaluated for the extraction of six steroidal hormones (estriol, estrone, 17β-estradiol, 17α-ethinylestradiol, progesterone, and testosterone) in aqueous samples with subsequent determination by gas chromatography-tandem mass spectrometry (GC-MS/MS) after the derivatization procedure. The developed extraction procedure was based on the solid phase extraction (SPE) technique, but employed hydrogel as the sorbent phase. The effects of several parameters, including the amount and composition of the sorbent phase, pH, sample volume, flow rate, and gel swelling over the extraction efficiency, were evaluated. Gels with lower swelling indexes and larger amounts of sorbent ensured higher extraction yields of analytes. The main benefits of using the PVOH/pectin-based hydrogel as the extraction phase are the ease of synthesis, low-cost preparation, and the possibility of reusing the extraction disks. Limits of quantification of 0.5 μg L−1 for estrone and 17β-estradiol, and 1 μg L−1 for testosterone, 17α-ethinylestradiol, progesterone, and estriol were obtained. Accuracy values ranged from 80% to 110%, while the inter-assay precision ranged from 0.23% to 22.2% and the intra-assay from 0.55% to 12.3%. Since the sorbent phase has an amphiphilic character, the use of hydrogels is promising for the extraction of medium-to-high polarity compounds.

Abstract Background Nucleic acid aptamers have long demonstrated the capacity to bind viral envelope proteins and to inhibit the progression of pathogenic virus infections. Here we report on initial efforts to develop and screen DNA aptamers against recombinant envelope proteins or synthetic peptides and whole inactivated viruses from several virulent arboviruses including Chikungunya, Crimean-Congo hemorrhagic fever (CCHF), dengue, tickborne encephalitis and West Nile viruses. We also analyzed sequence data and secondary structures for commonalities that might reveal consensus binding sites among the various aptamers. Some of the highest affinity and most specific aptamers in the down-selected libraries were demonstrated to have diagnostic utility in lateral flow chromatographic assays and in a fluorescent aptamer-magnetic bead sandwich assay. Some of the reported aptamers may also be able to bind viral envelope proteins in vivo and therefore may have antiviral potential in passive immunity or prophylactic applications. Results Several arbovirus DNA aptamer sequences emerged multiple times in the various down selected aptamer libraries thereby suggesting some consensus sequences for binding arbovirus envelope proteins. Screening of aptamers by enzyme-linked aptamer sorbent assay (ELASA) was useful for ranking relative aptamer affinities against their cognate viral targets. Additional study of the aptamer sequences and secondary structures of top-ranked anti-arboviral aptamers suggest potential virus binding motifs exist within some of the key aptamers and are highlighted in the supplemental figures for this article. One sequence segment (ACGGGTCCGGACA) emerged 60 times in the anti-CCHF aptamer library, but nowhere else in the anti-arbovirus library and only a few other times in a larger library of aptamers known to bind bacteria and rickettsia or other targets. Diagnostic utility of some of the aptamers for arbovirus detection in lateral flow chromatographic assays and a fluorescent sandwich assay on the surface of magnetic microbeads is also demonstrated. Conclusions This article catalogues numerous DNA aptamer sequences which can bind various important pathogenic arboviruses and have, in some cases, already demonstrated diagnostic potential. These aptamer sequences are proprietary, patent-pending, and partially characterized. Therefore, they are offered to the scientific community for potential research use in diagnostic assays, biosensor applications or for possible passive immunity and prophylaxis against pathogenic viruses.
