1. Evaluation of multi-season convection-permitting atmosphere – mixed-layer ocean simulations of the Maritime Continent

Abstract: A multi-season convection-permitting regional climate simulation of the Maritime Continent (MC) using the Met Office Unified Model (MetUM) with 2.2 km grid spacing is presented and evaluated. The simulations pioneer the use of atmosphere–ocean coupling with the multi-column K profile parametrisation (KPP) mixed-layer ocean model in atmospheric convection-permitting climate simulations. Comparisons are made against a convection-parametrised simulation in which it is nested and which in turn derives boundary conditions from the ERA5 reanalysis. This paper describes the configuration, performance of the mean state and variability in the two simulations compared against observational datasets. The models have both minor sea surface temperature (SST) and wet precipitation biases. The diurnal cycle, representation of equatorial waves, and relationship between SST and precipitation are all improved in the convection-permitting model compared to the convection-parametrised model. The Madden–Julian oscillation (MJO) is present in both models with a faster-than-observed propagation speed. However, it is unclear whether fidelity of the MJO simulation is inherent to the model or whether it predominantly arises from the forcing at the boundaries.

2. Spatial and temporal variability of solar penetration depths in the Bay of Bengal and its impact on sea surface temperature (SST) during the summer monsoon

Abstract: Chlorophyll has long been known to influence air–sea gas exchange and CO2 drawdown. But chlorophyll also influences regional climate through its effect on solar radiation absorption and thus sea surface temperature (SST). In the Bay of Bengal, the effect of chlorophyll on SST has been demonstrated to have a significant impact on the Indian summer (southwest) monsoon. However, little is known about the drivers and impacts of chlorophyll variability in the Bay of Bengal during the southwest monsoon. Here we use observations of downwelling irradiance measured by an ocean glider and three profiling floats to determine the spatial and temporal variability of solar absorption across the southern Bay of Bengal during the 2016 summer monsoon. A two-band exponential solar absorption scheme is fitted to vertical profiles of photosynthetically active radiation to determine the effective scale depth of blue light. Scale depths of blue light are found to vary from 12 m during the highest (0.3–0.5 mg m−3) mixed-layer chlorophyll concentrations to over 25 m when the mixed-layer chlorophyll concentrations are below 0.1 mg m−3. The Southwest Monsoon Current and coastal regions of the Bay of Bengal are observed to have higher mixed-layer chlorophyll concentrations and shallower solar penetration depths than other regions of the southern Bay of Bengal. Substantial sub-daily variability in solar radiation absorption is observed, which highlights the importance of near-surface ocean processes in modulating mixed-layer chlorophyll. Simulations using a one-dimensional K-profile parameterization ocean mixed-layer model with observed surface forcing from July 2016 show that a 0.3 mg m−3 increase in chlorophyll concentration increases sea surface temperature by 0.35 ∘C in 1 month, with SST differences growing rapidly during calm and sunny conditions. This has the potential to influence monsoon rainfall around the Bay of Bengal and its intraseasonal variability.

3. The role of geomorphology, rainfall and soil moisture in the occurrence of landslides triggered by 2018 Typhoon Mangkhut in the Philippines

Abstract: In 2018 Typhoon Mangkhut (locally known as Typhoon Ompong) triggered thousands of landslides in the Itogon region of the Philippines. A landslide inventory of the affected region is compiled for the first time, comprising 1101 landslides over a 570 km2 area. The inventory is used to study the geomorphological characteristics and land cover more prone to landsliding as well as the hydrometeorological conditions that led to widespread failure. The results showed that landslides mostly occurred on grassland and wooded slopes of clay superficial geology, predominantly facing east-southeast. Rainfall (Integrated Multi-satellitE Retrievals for Global Precipitation Measurement, IMERG GPM) associated with Typhoon Mangkhut is compared with 33 high-intensity rainfall events that did not trigger regional landslide events in 2018. Results show that landslides occurred during high-intensity rainfall that coincided with the highest soil moisture values (estimated clays saturation point), according to Soil Moisture Active Passive level 4 (SMAP-L4) data. Our results demonstrate the potential of SMAP-L4 and GPM IMERG data for landslide hazard assessment and early warning where ground-based data are scarce. However, other rainfall events in the months leading up to Typhoon Mangkhut that had similar or higher rainfall intensities and also occurred when soils were saturated did not trigger widespread landsliding, highlighting the need for further research into the conditions that trigger landslides in typhoons.

4. The effect of seasonally and spatially varying chlorophyll on Bay of Bengal surface ocean properties and the South Asian monsoon

Abstract: Chlorophyll absorbs solar radiation in the upper ocean, increasing the mixed layer radiative heating and sea surface temperatures (SST). Although the influence of chlorophyll distributions in the Arabian Sea on the southwest monsoon has been demonstrated, there is a current knowledge gap regarding how chlorophyll distributions in the Bay of Bengal influence the southwest monsoon. The solar absorption caused by chlorophyll can be parameterized as an optical parameter, h2, which expresses the scale depth of the absorption of blue light. Seasonally and spatially varying h2 fields in the Bay of Bengal were imposed in a 30-year simulation using an atmospheric general circulation model coupled to a mixed layer thermodynamic ocean model in order to investigate the effect of chlorophyll distributions on regional SST, the southwest monsoon circulation, and precipitation. There are both direct local upper-ocean effects, through changes in solar radiation absorption, and indirect remote atmospheric responses. The depth of the mixed layer relative to the perturbed solar penetration depths modulates the response of the SST to chlorophyll. The largest SST response of 0.5 ∘C to chlorophyll forcing occurs in coastal regions, where chlorophyll concentrations are high (> 1 mg m−3), and when climatological mixed layer depths shoal during the inter-monsoon periods. Precipitation increases significantly (by up to 3 mm d−1) across coastal Myanmar during the southwest monsoon onset and over northeast India and Bangladesh during the Autumn inter-monsoon period, decreasing model biases.

5. Vertical distribution of chlorophyll in dynamically distinct regions of the southern Bay of Bengal

Abstract: The Bay of Bengal (BoB) generally exhibits surface oligotrophy due to nutrient limitation induced by strong salinity stratification. Nevertheless, there are hotspots of high chlorophyll in the BoB where the monsoonal forcings are strong enough to break the stratification; one such region is the southern BoB, east of Sri Lanka. A recent field programme conducted during the summer monsoon of 2016, as a part of the Bay of Bengal Boundary Layer Experiment (BoBBLE), provides a unique high-resolution dataset of the vertical distribution of chlorophyll in the southern BoB using ocean gliders along with shipboard conductivity–temperature–depth (CTD) measurements. Observations were carried out for a duration of 12–20 days, covering the dynamically active regions of the Sri Lanka Dome (SLD) and the Southwest Monsoon Current (SMC). Mixing and upwelling induced by the monsoonal wind forcing enhanced surface chlorophyll concentrations (0.3–0.7 mg m−3). Prominent deep chlorophyll maxima (DCM; 0.3–1.2 mg m−3) existed at intermediate depths (20–50 m), signifying the contribution of subsurface productivity to the biological carbon cycling in the BoB. The shape of chlorophyll profiles varied in different dynamical regimes; upwelling was associated with sharp and intense DCM, whereas mixing resulted in a diffuse and weaker DCM. Within the SLD, open-ocean Ekman suction favoured a substantial increase in chlorophyll. Farther east, where the thermocline was deeper, enhanced surface chlorophyll was associated with intermittent mixing events. Remote forcing by the westward propagating Rossby waves influenced the upper-ocean dynamics and chlorophyll distribution in the southern BoB. Stabilizing surface freshening events and barrier-layer formation often inhibited the generation of surface chlorophyll. The pathway of the SMC intrusion was marked by a distinct band of chlorophyll, indicating the advective effect of biologically rich Arabian Sea waters. The region of the monsoon current exhibited the strongest DCM as well as the highest column-integrated chlorophyll. Observations suggest that the persistence of DCM in the southern BoB is promoted by surface oligotrophy and shallow mixed layers. Results from a coupled physical–ecosystem model substantiate the dominant role of mixed layer processes associated with the monsoon in controlling the nutrient distribution and biological productivity in the southern BoB. The present study provides new insights into the vertical distribution of chlorophyll in the BoB, emphasizing the need for extensive in situ sampling and ecosystem model-based efforts for a better understanding of the biophysical interactions and the potential climatic feedbacks.

6. Comparisons of Core Temperature Between a Telemetric Pill and Heart Rate Estimated Core Temperature in Firefighters

Abstract: Background: Firefighters may experience high environmental temperatures or carry out intensive physical tasks, or both, which leads to increased core body temperature and risk of fatalities. Hence there is a need to remotely and non-invasively monitor core body temperature. Methods: Estimated (heart rate algorithm) and actual core body temperature (ingested telemetric pill) measures were collected simultaneously for comparison during training exercises on 44 firefighter volunteers. Results: Prediction of core body temperature varied, with no specific identifiable pattern between the algorithm values and directly measured body core temperatures. Group agreement of Lin's Concordance of 0.74 (95% Upper 0.75, lower CI 0.73), was deemed poor. Conclusion: From individual agreement data Lin's Concordance was variable (Min 0.11, CI 0.13–0.01; Max 0.83, CI 0.86–0.80), indicating that the heart rate algorithm approach was not suitable for core body temperature monitoring in this population group, especially at the higher more critical core body temperatures seen.

7. Fine-grain beta diversity in Palaearctic open vegetation: variability within and between biomes and vegetation types

Abstract: Aims: To quantify how fine-grain (within-plot) beta diversity differs among biomes and vegetation types. Study area: Palaearctic biogeographic realm. Methods: We extracted 4,654 nested-plot series with at least four different grain sizes between 0.0001 m² and 1,024 m² from the GrassPlot database spanning broad geographic and ecological gradients. Next, we calculated the slope parameter (z-value) of the power-law species–area relationship (SAR) to use as a measure of multiplicative beta diversity. We did this separately for vascular plants, bryophytes and lichens and for the three groups combined (complete vegetation). We then tested whether z-values differed between biomes, ecological-physiognomic vegetation types at coarse and fine levels and phytosociological classes. Results: We found that z-values varied significantly among biomes and vegetation types. The explanatory power of area for species richness was highest for vascular plants, followed by complete vegetation, bryophytes and lichens. Within each species group, the explained variance increased with typological resolution. In vascular plants, adjusted R2 was 0.14 for biomes, but reached 0.50 for phytosociological classes. Among the biomes, mean z-values were particularly high in the Subtropics with winter rain (Mediterranean biome) and the Dry tropics and subtropics. Natural grasslands had higher z-values than secondary grasslands. Alpine and Mediterranean vegetation types had particularly high z-values whereas managed grasslands with benign soil and climate conditions and saline communities were characterised by particularly low z-values. Conclusions: In this study relating fine-grain beta diversity to typological units, we found distinct patterns. As we explain in a conceptual figure, these can be related to ultimate drivers, such as productivity, stress and disturbance, which can influence z-values via multiple pathways. The provided means, medians and quantiles of z-values for a wide range of typological entities provide benchmarks for local to continental studies, while calling for additional data from under-represented units. Abstract: Abstract We present a measurement of the cosmic-ray spectrum above 100 PeV using the part of the surface detector of the Pierre Auger Observatory that has a spacing of 750 m. An inflection of the spectrum is observed, confirming the presence of the so-called second-knee feature. The spectrum is then combined with that of the 1500 m array to produce a single measurement of the flux, linking this spectral feature with the three additional breaks at the highest energies. The combined spectrum, with an energy scale set calorimetrically via fluorescence telescopes and using a single detector type, results in the most statistically and systematically precise measurement of spectral breaks yet obtained. These measurements are critical for furthering our understanding of the highest energy cosmic rays.

9. Bromodomain Inhibitors Modulate FcγR-Mediated Mononuclear Phagocyte Activation and Chemotaxis

Abstract: IgG antibodies form immune complexes (IC) that propagate inflammation and tissue damage in autoimmune diseases such as systemic lupus erythematosus. IgG IC engage Fcγ receptors (FcγR) on mononuclear phagocytes (MNP), leading to widespread changes in gene expression that mediate antibody effector function. Bromodomain and extra-terminal domain (BET) proteins are involved in governing gene transcription. We investigated the capacity of BET protein inhibitors (iBET) to alter IgG FcγR-mediated MNP activation. We found that iBET dampened IgG IC-induced pro-inflammatory gene expression and decreased activating FcγR expression on MNPs, reducing their ability to respond to IgG IC. Despite FcγR downregulation, iBET-treated macrophages demonstrated increased phagocytosis of protein antigen, IgG IC, and apoptotic cells. iBET also altered cell morphology, generating more amoeboid MNPs with reduced adhesion. iBET treatment impaired chemotaxis towards a CCL19 gradient in IC-stimulated dendritic cells (DC) in vitro, and inhibited IC-induced DC migration to draining lymph nodes in vivo, in a DC-intrinsic manner. Altogether, our data show that iBET modulates FcγR-mediated MNP activation and migration, revealing the therapeutic potential of BET protein inhibition in antibody-mediated diseases.

Abstract: Two proteins of the Escherichia coli membrane protein complex, CsgG and CsgF, are studied as proteinaceous nanopores for DNA sequencing. It is highly desirable to control the DNA as it moves through the pores, this requires characterisation of DNA translocation and subsequent optimization of the pores. In order to inform protein engineering to improve the pores, we have conducted a series of molecular dynamics simulations to characterise the mechanical strength and conformational dynamics of CsgG and the CsgG-CsgF complex and how these impact ssDNA, water and ion movement. We find that the barrel of CsgG is more susceptible to damage from external electric fields compared to the protein vestibule. Furthermore, the presence of CsgF within the CsgG-CsgF complex enables the complex to withstand higher electric fields. We find that the eyelet loops of CsgG play a key role in both slowing the translocation rate of DNA and modulating the conductance of the pore. CsgF also impacts the DNA translocation rate, but to a lesser degree than CsgG.

Abstract: Abstract The sensitivity of the Deep Underground Neutrino Experiment (DUNE) to neutrino oscillation is determined, based on a full simulation, reconstruction, and event selection of the far detector and a full simulation and parameterized analysis of the near detector. Detailed uncertainties due to the flux prediction, neutrino interaction model, and detector effects are included. DUNE will resolve the neutrino mass ordering to a precision of 5 $$\sigma $$ σ , for all $$\delta _{\mathrm{CP}}$$ δ CP values, after 2 years of running with the nominal detector design and beam configuration. It has the potential to observe charge-parity violation in the neutrino sector to a precision of 3 $$\sigma $$ σ (5 $$\sigma $$ σ ) after an exposure of 5 (10) years, for 50% of all $$\delta _{\mathrm{CP}}$$ δ CP values. It will also make precise measurements of other parameters governing long-baseline neutrino oscillation, and after an exposure of 15 years will achieve a similar sensitivity to $$\sin ^{2} 2\theta _{13}$$ sin 2 2 θ 13 to current reactor experiments.

Abstract: Abstract The hybrid design of the Pierre Auger Observatory allows for the measurement of the properties of extensive air showers initiated by ultra-high energy cosmic rays with unprecedented precision. By using an array of prototype underground muon detectors, we have performed the first direct measurement, by the Auger Collaboration, of the muon content of air showers between $$2\times 10^{17}$$ 2×1017 and $$2\times 10^{18}$$ 2×1018 eV. We have studied the energy evolution of the attenuation-corrected muon density, and compared it to predictions from air shower simulations. The observed densities are found to be larger than those predicted by models. We quantify this discrepancy by combining the measurements from the muon detector with those from the Auger fluorescence detector at $$10^{{17.5}}\, {\mathrm{eV}} $$ 1017.5eV and $$10^{{18}}\, {\mathrm{eV}} $$ 1018eV . We find that, for the models to explain the data, an increase in the muon density of $$38\%$$ 38% $$\pm 4\% (12\%)$$ ±4%(12%) $$\pm {}^{21\%}_{18\%}$$ ±18%21% for EPOS-LHC, and of $$50\% (53\%)$$ 50%(53%) $$\pm 4\% (13\%)$$ ±4%(13%) $$\pm {}^{23\%}_{20\%}$$ ±20%23% for QGSJetII-04, is respectively needed.

15. Impact of 3D genome organization, guided by cohesin and CTCF looping, on sex-biased chromatin interactions and gene expression in mouse liver

Abstract: Abstract Several thousand sex-differential distal enhancers have been identified in mouse liver; however, their links to sex-biased genes and the impact of any sex-differences in nuclear organization and chromatin interactions are unknown. To address these issues, we first characterized 1847 mouse liver genomic regions showing significant sex differential occupancy by cohesin and CTCF, two key 3D nuclear organizing factors. These sex-differential binding sites were primarily distal to sex-biased genes but rarely generated sex-differential TAD (topologically associating domain) or intra-TAD loop anchors, and were sometimes found in TADs without sex-biased genes. A substantial subset of sex-biased cohesin-non-CTCF binding sites, but not sex-biased cohesin-and-CTCF binding sites, overlapped sex-biased enhancers. Cohesin depletion reduced the expression of male-biased genes with distal, but not proximal, sex-biased enhancers by >10-fold, implicating cohesin in long-range enhancer interactions regulating sex-biased genes. Using circularized chromosome conformation capture-based sequencing (4C-seq), we showed that sex differences in distal sex-biased enhancer–promoter interactions are common. Intra-TAD loops with sex-independent cohesin-and-CTCF anchors conferred sex specificity to chromatin interactions indirectly, by insulating sex-biased enhancer–promoter contacts and by bringing sex-biased genes into closer proximity to sex-biased enhancers. Furthermore, sex-differential chromatin interactions involving sex-biased gene promoters, enhancers, and lncRNAs were associated with sex-biased binding of cohesin and/or CTCF. These studies elucidate how 3D genome organization impacts sex-biased gene expression in a non-reproductive tissue through both direct and indirect effects of cohesin and CTCF looping on distal enhancer interactions with sex-differentially expressed genes.

18. Harnessing natural variation to identify cis regulators of sex-biased gene expression in a multi-strain mouse liver model.

Abstract: Sex differences in gene expression are widespread in the liver, where many autosomal factors act in tandem with growth hormone signaling to regulate individual variability of sex differences in liver metabolism and disease. Here, we compare hepatic transcriptomic and epigenetic profiles of mouse strains C57BL/6J and CAST/EiJ, representing two subspecies separated by 0.5-1 million years of evolution, to elucidate the actions of genetic factors regulating liver sex differences. We identify 144 protein coding genes and 78 lncRNAs showing strain-conserved sex bias; many have gene ontologies relevant to liver function, are more highly liver-specific and show greater sex bias, and are more proximally regulated than genes whose sex bias is strain-dependent. The strain-conserved genes include key growth hormone-dependent transcriptional regulators of liver sex bias; however, three other transcription factors, Trim24, Tox, and Zfp809, lose their sex-biased expression in CAST/EiJ mouse liver. To elucidate the observed strain specificities in expression, we characterized the strain-dependence of sex-biased chromatin opening and enhancer marks at cis regulatory elements (CREs) within expression quantitative trait loci (eQTL) regulating liver sex-biased genes. Strikingly, 208 of 286 eQTLs with strain-specific, sex-differential effects on expression were associated with a complete gain, loss, or reversal of the sex differences in expression between strains. Moreover, 166 of the 286 eQTLs were linked to the strain-dependent gain or loss of localized sex-biased CREs. Remarkably, a subset of these CREs apparently lacked strain-specific genetic variants yet showed coordinated, strain-dependent sex-biased epigenetic regulation. Thus, we directly link hundreds of strain-specific genetic variants to the high variability in CRE activity and expression of sex-biased genes and uncover underlying genetically-determined epigenetic states controlling liver sex bias in genetically diverse mouse populations.

Abstract: We consider the problem of classifying curves when they are observed only partially on their parameter domains. We propose computational methods for (i) completion of partially observed curves; (ii) assessment of completion variability through a nonparametric multiple imputation procedure; (iii) development of nearest neighbor classifiers compatible with the completion techniques. Our contributions are founded on exploiting the geometric notion of shape of a curve, defined as those aspects of a curve that remain unchanged under translations, rotations and reparameterizations. Explicit incorporation of shape information into the computational methods plays the dual role of limiting the set of all possible completions of a curve to those with similar shape while simultaneously enabling more efficient use of training data in the classifier through shape-informed neighborhoods. Our methods are then used for taxonomic classification of partially observed curves arising from images of fossilized Bovidae teeth, obtained from a novel anthropological application concerning paleoenvironmental reconstruction.

Abstract: Many low-threshold experiments observe sharply rising event rates of yet unknown origins below a few hundred eV, and larger than expected from known backgrounds. Due to the significant impact of this excess on the dark matter or neutrino sensitivity of these experiments, a collective effort has been started to share the knowledge about the individual observations. For this, the EXCESS Workshop was initiated. In its first iteration in June 2021, ten rare event search collaborations contributed to this initiative via talks and discussions. The contributing collaborations were CONNIE, CRESST, DAMIC, EDELWEISS, MINER, NEWS-G, NUCLEUS, RICOCHET, SENSEI and SuperCDMS. They presented data about their observed energy spectra and known backgrounds together with details about the respective measurements. In this paper, we summarize the presented information and give a comprehensive overview of the similarities and differences between the distinct measurements. The provided data is furthermore publicly available on the workshop’s data repository together with a plotting tool for visualization.

Abstract: Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Methods and results: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012–2016). 23. Long-term effect of botulinum toxin A on the hip and spine in cerebral palsy: A national retrospective cohort study in Taiwan.

Abstract: ObjectivesTo investigate the effect of botulinum toxin A (BTA) on the development of hip dislocation and scoliosis, surgical rates for hip and spine, and mortality in cerebral palsy (CP).Study designA cohort study was conducted using CP data from a Taiwan National Insurance Health Research Database. Diagnoses were defined using the International Classification of Diseases codes, 9th revision. Adjusted hazard ratios for outcomes were calculated using Cox regression analysis and adjusted for the following variables: BTA injection, sex, age, severities of CP, comorbidities, location, urbanization level, and level of care.ResultsA total of 1,405 CP children (670 female vs. 735 male), 281 in the BTA group and 1,124 in the controls, were followed-up for a mean of 5 years 4 months. There were no significant differences in the outcomes in both groups, in the incidence rates of hip dislocation and scoliosis, nor in the surgical rates for hip and spine surgery. Mortality rate in the BTA group was 0.49 times lower than that in the controls (p = 0.001). Moderate to severe types of CP had higher incidence rates of hip dislocation, scoliosis, hip surgery, spine surgery, and mortality.ConclusionModerate to severe types of CP had poorer outcomes in all aspects, including a higher risk of hip dislocation, scoliosis, surgical rate for hip and spine, and mortality. Although BTA injection in children with CP proved to not significantly reduce hip dislocation and scoliosis, it is considered safe as an anti-spasticity treatment and may be beneficial for survival.

Abstract: Metal parts produced by laser powder bed fusion (LPBF) additive manufacturing exhibit characteristic microstructures comparable to those observed in laser welding. The primary cause of this characteristic microstructure is rapid, localized heating and cooling cycles, which result in extreme thermal gradients where material solidification is followed by fast cooling in the solid state. The final microstructure and mechanical performance are also influenced by pore formation caused by melt pool fluid dynamics. Here, we use high speed, in situ X-ray diffraction to probe the kinetics of cooling and solid-solid phase transitions after laser melting in two aerospace titanium alloys: Ti-6Al-4V, an α + β alloy; and Ti-5Al-5V-5Mo-3Cr, a near-β alloy. We complement these diffraction studies with in situ X-ray imaging to probe melt pool dynamics and pore formation. From these two complementary probes, we quantify pore formation during melting and the subsequent microstructural evolution as the material rapidly cools after solidification. These results are critical for understanding defect formation and residual stress development in different titanium alloys under LPBF conditions and can help inform process models to predict final part performance.

Abstract: Abstract Elves are a class of transient luminous events, with a radial extent typically greater than 250 km, that occur in the lower ionosphere above strong electrical storms. We report the observation of 1,598 elves, from 2014 to 2016, recorded with unprecedented time resolution (100 ns) using the fluorescence detector (FD) of the Pierre Auger Cosmic‐Ray Observatory. The Auger Observatory is located in the Mendoza province of Argentina with a viewing footprint for elve observations of 3·106 km 2, reaching areas above the Pacific and Atlantic Oceans, as well as the Córdoba region, which is known for severe convective thunderstorms. Primarily designed for ultrahigh energy cosmic‐ray observations, the Auger FD turns out to be very sensitive to the ultraviolet emission in elves. The detector features modified Schmidt optics with large apertures resulting in a field of view that spans the horizon, and year‐round operation on dark nights with low moonlight background, when the local weather is favorable. The measured light profiles of 18% of the elve events have more than one peak, compatible with intracloud activity. Within the 3‐year sample, 72% of the elves correlate with the far‐field radiation measurements of the World Wide Lightning Location Network. The Auger Observatory plans to continue operations until at least 2025, including elve observations and analysis. To the best of our knowledge, this observatory is the only facility on Earth that measures elves with year‐round operation and full horizon coverage.

26. The influence of carbonate platform interactions with subduction zone volcanism on palaeo-atmospheric CO2 since the Devonian

Abstract: The CO2 liberated along subduction zones through intrusive/extrusive magmatic activity and the resulting active and diffuse outgassing influences global atmospheric CO2. However, when melts derived from subduction zones intersect buried carbonate platforms, decarbonation reactions may cause the contribution to atmospheric CO2 to be far greater than segments of the active margin that lacks buried carbon-rich rocks and carbonate platforms. This study investigates the contribution of carbonate-intersecting subduction zones (CISZs) to palaeo-atmospheric CO2 levels over the past 410 million years by integrating a plate motion and plate boundary evolution model with carbonate platform development through time. Our model of carbonate platform development has the potential to capture a broader range of degassing mechanisms than approaches that only account for continental arcs.Continuous and cross-wavelet analyses as well as wavelet coherence are used to evaluate trends between the evolving lengths of carbonate-intersecting subduction zones, non-carbonate-intersecting subduction zones and global subduction zones, and are examined for periodic, linked behaviour with the proxy CO2 record between 410 Ma and the present. Wavelet analysis reveals significant linked periodic behaviour between 60 and 40 Ma, when CISZ lengths are relatively high and are correlated with peaks in palaeo-atmospheric CO2, characterised by a 32–48 Myr periodicity and a ∼ 8–12 Myr lag of CO2 peaks following CISZ length peaks. The linked behaviour suggests that the relative abundance of CISZs played a role in affecting global climate during the Palaeogene. In the 200–100 Ma period, peaks in CISZ lengths align with peaks in palaeo-atmospheric CO2, but CISZ lengths alone cannot be determined as the cause of a warmer Cretaceous–Jurassic climate. Nevertheless, across the majority of the Phanerozoic, feedback mechanisms between the geosphere, atmosphere and biosphere likely played dominant roles in modulating climate. Our modelled subduction zone lengths and carbonate-intersecting subduction zone lengths approximate magmatic activity through time, and can be used as input into fully coupled models of CO2 flux between deep and shallow carbon reservoirs.

Abstract: Obesity is a major human health crisis that promotes insulin resistance and, ultimately, type 2 diabetes. The molecular mechanisms that mediate this response occur across many highly complex biological regulatory levels that are incompletely understood. Here, we present a comprehensive molecular systems biology study of hepatic responses to high-fat feeding in mice. We interrogated diet-induced epigenomic, transcriptomic, proteomic, and metabolomic alterations using high-throughput omic methods and used a network modeling approach to integrate these diverse molecular signals. Our model indicated that disruption of hepatic architecture and enhanced hepatocyte apoptosis are among the numerous biological processes that contribute to early liver dysfunction and low-grade inflammation during the development of diet-induced metabolic syndrome. We validated these model findings with additional experiments on mouse liver sections. In total, we present an integrative systems biology study of diet-induced hepatic insulin resistance that uncovered molecular features promoting the development and maintenance of metabolic disease.

Abstract: The Deep Underground Neutrino Experiment (DUNE) is an international, world-class experiment aimed at exploring fundamental questions about the universe that are at the forefront of astrophysics and particle physics research. DUNE will study questions pertaining to the preponderance of matter over antimatter in the early universe, the dynamics of supernovae, the subtleties of neutrino interaction physics, and a number of beyond the Standard Model topics accessible in a powerful neutrino beam. A critical component of the DUNE physics program involves the study of changes in a powerful beam of neutrinos, i.e., neutrino oscillations, as the neutrinos propagate a long distance. The experiment consists of a near detector, sited close to the source of the beam, and a far detector, sited along the beam at a large distance. This document, the DUNE Near Detector Conceptual Design Report (CDR), describes the design of the DUNE near detector and the science program that drives the design and technology choices. The goals and requirements underlying the design, along with projected performance are given. It serves as a starting point for a more detailed design that will be described in future documents.

29. Metal vapor micro-jet controls material redistribution in laser powder bed fusion additive manufacturing

Abstract: Abstract The results of detailed experiments and finite element modeling of metal micro-droplet motion associated with metal additive manufacturing (AM) processes are presented. Ultra high speed imaging of melt pool dynamics reveals that the dominant mechanism leading to micro-droplet ejection in a laser powder bed fusion AM is not from laser induced recoil pressure as is widely believed and found in laser welding processes, but rather from vapor driven entrainment of micro-particles by an ambient gas flow. The physics of droplet ejection under strong evaporative flow is described using simulations of the laser powder bed interactions to elucidate the experimental results. Hydrodynamic drag analysis is used to augment the single phase flow model and explain the entrainment phenomenon for 316 L stainless steel and Ti-6Al-4V powder layers. The relevance of vapor driven entrainment of metal micro-particles to similar fluid dynamic studies in other fields of science will be discussed.

30. Hyper- and hypo- nutrition studies of the hepatic transcriptome and epigenome suggest that PPARα regulates anaerobic glycolysis

Abstract: Abstract Diet plays a crucial role in shaping human health and disease. Diets promoting obesity and insulin resistance can lead to severe metabolic diseases, while calorie-restricted (CR) diets can improve health and extend lifespan. In this work, we fed mice either a chow diet (CD), a 16 week high-fat diet (HFD), or a CR diet to compare and contrast the effects of these diets on mouse liver biology. We collected transcriptomic and epigenomic datasets from these mice using RNA-Seq and DNase-Seq. We found that both CR and HFD induce extensive transcriptional changes, in some cases altering the same genes in the same direction. We used our epigenomic data to infer transcriptional regulatory proteins bound near these genes that likely influence their expression levels. In particular, we found evidence for critical roles played by PPARα and RXRα. We used ChIP-Seq to profile the binding locations for these factors in HFD and CR livers. We found extensive binding of PPARα near genes involved in glycolysis/gluconeogenesis and uncovered a role for this factor in regulating anaerobic glycolysis. Overall, we generated extensive transcriptional and epigenomic datasets from livers of mice fed these diets and uncovered new functions and gene targets for PPARα.

Abstract: We report a first measurement for ultrahigh energy cosmic rays of the correlation between the depth of shower maximum and the signal in the water Cherenkov stations of air-showers registered simultaneously by the fluorescence and the surface detectors of the Pierre Auger Observatory. Such a correlation measurement is a unique feature of a hybrid air-shower observatory with sensitivity to both the electromagnetic and muonic components. It allows an accurate determination of the spread of primary masses in the cosmic-ray flux. Up till now, constraints on the spread of primary masses have been dominated by systematic uncertainties. The present correlation measurement is not affected by systematics in the measurement of the depth of shower maximum or the signal in the water Cherenkov stations. The analysis relies on general characteristics of air showers and is thus robust also with respect to uncertainties in hadronic event generators. The observed correlation in the energy range around the ‘ankle’ at lg⁡(E/eV)=18.5–19.0 differs significantly from expectations for pure primary cosmic-ray compositions. A light composition made up of proton and helium only is equally inconsistent with observations. The data are explained well by a mixed composition including nuclei with mass A>4. Scenarios such as the proton dip model, with almost pure compositions, are thus disfavored as the sole explanation of the ultrahigh-energy cosmic-ray flux at Earth. Abstract: Age-related hearing loss is a chronic degenerative disorder affecting one in two individuals above the age of 75. Current population projections predict a steady climb in the number of older individuals making the search for interventions to prevent or reverse this disorder even more critical. There is growing acceptance that aberrant activity of resident or infiltrating immune cells, such as macrophages, is a major factor contributing to the onset and progression of age-related degenerative diseases. However, how macrophage populations and their functionally-driven morphological characteristics change with age in the human cochlea remains largely unknown. In this study, we employed immunohistochemical approaches along with confocal and super-resolution imaging, three-dimensional reconstructions, and quantitative analysis to determine age-related changes in macrophage numbers and morphology as well as interactions with other cell-types and structures of the auditory nerve and lateral wall in the human cochlea. In the cochlea of human ears from young and middle aged adults those macrophages in the auditory nerve assumed a worm-like structure in contrast to those in the spiral ligament or associated with the dense microvascular network in the stria vascularis which exhibited a highly ramified morphology. Macrophages in both the auditory nerve and cochlear lateral wall showed morphological alterations with age. The population of activated macrophages in the auditory nerve increased in cochleas obtained from older donors. Dual-immunohistochemical staining with macrophage, myelin, and neuronal markers revealed increased interactions of macrophages with the glial and neuronal components of the aged auditory nerve. These findings implicate the involvement of abnormal macrophage-glia interactions in age-related physiological and pathological alterations in the human cochlea. There is clearly a need to further investigate the contribution of macrophage-associated inflammatory dysregulation in human presbyacusis.

33. An analysis of the effect of tooth wear on bovid identification

Abstract: Previous research provides a method for reducing the subjectivity in taxonomic identification of species in the family Bovidae by quantifying the occlusal surface of molar teeth using elliptical Fourier analysis. In this current study, we specifically test what effect medium to late tooth wear has on the identification of bovids when using the form (size and shape) of the occlusal surface to classify specimens. To achieve this, the classification results of teeth with ≥85% of their occlusal surface (training data set) were compared with the results of teeth with 87% of the time with both Alcelaphini and Reduncini classifying correctly >90% of the time. The worn teeth in the test data set classified correctly at lower rates, but all tribes still collectively had a good classification accuracy (>60%) with classification of Alcelaphini and Tragelaphini at >75% and Reduncini at >65%. Hippotragini classified correctly 50% of the time and Neotragini 42%. The one worn Bovini tooth in the test data set was classified as that of a Hippotragini. The classification rates of teeth with medium to late wear were lower than the unworn teeth, but not so low as to suggest that this methodology for identifying bovid teeth is inappropriate for worn teeth, especially when supplementary to other methods. Significance: • Worn teeth can be taxonomically identified by performing elliptical Fourier analysis on the occlusal surface of their teeth. • While teeth with medium to late wear correctly classify at lower rates than relatively unworn ones, the methodology is a valuable supplement to other methods for taxonomically identifying bovid teeth. Abstract: An overview of the multi-messenger capabilities of the Pierre Auger Observatory is presented. The techniques and performance of searching for Ultra-High Energy neutrinos, photons and neutrons are described. Some of the most relevant results are reviewed, such as stringent upper bounds that were placed to a flux of diffuse cosmogenic neutrinos and photons, bounds placed on neutrinos emitted from compact binary mergers that were detected by LIGO and Virgo during their first and second observing runs, as well as searches for high energy photons and neutrons from the Galactic center that constrain the properties of the putative Galactic PeVatron, observed by the H.E.S.S. collaboration. The observation of directional correlations between ultra-high energy cosmic rays and either high energy astrophysical neutrinos or specific source populations, weighted by their electromagnetic radiation, are also discussed. They constitute additional multi-messenger approaches aimed at identifying the sources of high energy cosmic rays.

35. Ultrafast dynamics of laser-metal interactions in additive manufacturing alloys captured by in situ X-ray imaging

Abstract: Advanced in situ characterization is essential for determining the underlying dynamics of laser-material interactions central to both laser welding and the rapidly expanding field of additive manufacturing. Traditional characterization techniques leave a critical experimental gap in understanding the complex subsurface fluid flow and metal evaporation dynamics inherent in laser-induced heating of the metal. Herein, in situ ultra-high-speed transmission X-ray imaging is revealed to be essential for bridging this information gap, particularly via comparison with and validation of advanced multiphysics simulations. Imaging on submicrosecond timescales enables correlation between dynamics of the laser-generated vapor–liquid interface and melt pool surface instabilities in industrially relevant alloys. X-ray imaging and complimentary simulations reveal vapor depression oscillations and rapid expansion due to reflection of the processing laser from the front surface of the vapor depression. Pore formation studies at steady state and during prompt removal of laser heating at the end of track reveal that the rapidly solidifying melt pool traps pores near the base of the vapor-filled depression. Moreover, pores within the melt pool are entrained by Marangoni convection which overcomes the force of buoyancy and forces the pores downward from the surface immediately before solidification. Observed solidification kinetics, consistent with previous results, give insight into surface morphology and porosity in the processed material. The information presented here is key for defining the physical models that describe laser-material interaction and ultimately increases our understanding of the emerging field of laser-based metal additive manufacturing. Abstract: Paneth cells are key epithelial cells that provide an antimicrobial barrier and maintain integrity of the small-intestinal stem cell niche. Paneth cell abnormalities are unfortunately detrimental to gut health and are often associated with digestive pathologies such as Crohn's disease or infections. Similar alterations are observed in individuals with impaired autophagy, a process that recycles cellular components. The direct effect of autophagy impairment on Paneth cells has not been analysed. To investigate this, we generated a mouse model lacking Atg16l1 specifically in intestinal epithelial cells, making these cells impaired in autophagy. Using three-dimensional intestinal organoids enriched for Paneth cells, we compared the proteomic profiles of wild-type and autophagy-impaired organoids. We used an integrated computational approach combining protein-protein interaction networks, autophagy-targeted proteins and functional information to identify the mechanistic link between autophagy impairment and disrupted pathways. Of the 284 altered proteins, 198 (70%) were more abundant in autophagy-impaired organoids, suggesting reduced protein degradation. Interestingly, these differentially abundant proteins comprised 116 proteins (41%) that are predicted targets of the selective autophagy proteins p62, LC3 and ATG16L1. Our integrative analysis revealed autophagy-mediated mechanisms that degrade key proteins in Paneth cell functions, such as exocytosis, apoptosis and DNA damage repair. Transcriptomic profiling of additional organoids confirmed that 90% of the observed changes upon autophagy alteration have effects at the protein level, not on gene expression. We performed further validation experiments showing differential lysozyme secretion, confirming our computationally inferred downregulation of exocytosis. Our observations could explain how protein-level alterations affect Paneth cell homeostatic functions upon autophagy impairment. Abstract: ABSTRACT A wide range of bacterial pathogens have been shown to form biofilms, which significantly increase their resistance to environmental stresses, such as antibiotics, and are thus of central importance in the context of bacterial diseases. One of the major structural components of these bacterial biofilms are amyloid fibrils, yet the mechanism of fibril assembly and its importance for biofilm formation are currently not fully understood. By studying fibril formation in vitro, in a model system of two common but unrelated biofilm-forming proteins, FapC from Pseudomonas fluorescens and CsgA from Escherichia coli, we found that the two proteins have a common aggregation mechanism. In both systems, fibril formation proceeds via nucleated growth of linear fibrils exhibiting similar measured rates of elongation, with negligible fibril self-replication. These similarities between two unrelated systems suggest that convergent evolution plays a key role in tuning the assembly kinetics of functional amyloid fibrils and indicates that only a narrow window of mechanisms and assembly rates allows for successful biofilm formation. Thus, the amyloid assembly reaction is likely to represent a means for controlling biofilm formation, both by the organism and by possible inhibitory drugs. IMPORTANCE Biofilms are generated by bacteria, embedded in the formed extracellular matrix. The biofilm's function is to improve the survival of a bacterial colony through, for example, increased resistance to antibiotics or other environmental stresses. Proteins secreted by the bacteria act as a major structural component of this extracellular matrix, as they self-assemble into highly stable amyloid fibrils, making the biofilm very difficult to degrade by physical and chemical means once formed. By studying the self-assembly mechanism of the fibrils from their monomeric precursors in two unrelated bacteria, our experimental and theoretical approaches shed light on the mechanism of functional amyloid assembly in the context of biofilm formation. Our results suggest that fibril formation may be a rate-limiting step in biofilm formation, which in turn has implications on the protein self-assembly reaction as a target for potential antibiotic drugs.

Abstract: Understanding patterns of community structure and the causes for their variation can be furthered by comparative biogeographic analyses of island biotas. We used woody plant data at the local scale to investigate variations in species rarity, alpha, beta, and gamma diversity within and between three islands from the oceanic archipelagoes of Azores, Canaries and Mascarene. We used standardized protocols to sample ten 50 m × 50 m forest plots in each of the three islands with contrasting climate and regional species pools: Terceira (Azores), Tenerife (Canaries), and Reunion (Mascarene Islands). Occupancy frequency distributions and species abundance distributions were used to investigate rarity. The partitioning of beta diversity in a distance-decay framework was used to test for spatial patterns of community composition. Rarity was much more pronounced in the highly diverse islands of Tenerife and Reunion than in the regionally poorer island of Terceira. The number of species rose faster with increasing sample area in both Tenerife and Reunion. The slope of the species rank abundance curve was steeper in Terceira whereas the richer island assemblages approached a lognormal model. Compositional changes according to spatial distance were mostly due to replacement of species in Terceira and Reunion. Our results point to important differences in the community structure of Terceira, which is the less diverse and temperate region in comparison to Tenerife and Reunion which are highly diverse.

Abstract: Background: An ethnobotanical study on local cuisine of Sasak tribe in Lombok Island was carried out, as a kind of effort of providing written record of culinary culture in some region of Indonesia. The cuisine studied included meals, snacks, and beverages that have been consumed by Sasak people from generation to generation. Objective: The aims of this study are to explore the local knowledge in utilising and managing plants resources in Sasak cuisine, and to analyze the perceptions and concepts related to food and eating of Sasak people. Methods: Data were collected through direct observation, participatory-observation, interviews and literature review. Results: In total 151 types of consumption were recorded, consisting of 69 meals, 71 snacks, and 11 beverages. These were prepared with 111 plants species belonging to 91 genera and 43 families. Fabaceae contributed the highest number of species to the cuisine. Cocos nucifera had the highest Index of Cultural Significance value and highest number of reported uses. Apparently traditional social and cultural values are still closely associated with Sasak food and eating. Conclusion: Sasak people interpret their food not only as a material for supporting life, but also as a means to maintain a good balance between humans, environment, and spiritual needs.

Abstract: MicroRNAs (miRNAs) regulate diverse biological processes by repressing mRNAs, but their modest effects on direct targets, together with their participation in larger regulatory networks, make it challenging to delineate miRNA-mediated effects. Here, we describe an approach to characterizing miRNA-regulatory networks by systematically profiling transcriptional, post-transcriptional and epigenetic activity in a pair of isogenic murine fibroblast cell lines with and without Dicer expression. By RNA sequencing (RNA-seq) and CLIP (crosslinking followed by immunoprecipitation) sequencing (CLIP-seq), we found that most of the changes induced by global miRNA loss occur at the level of transcription. We then introduced a network modeling approach that integrated these data with epigenetic data to identify specific miRNA-regulated transcription factors that explain the impact of miRNA perturbation on gene expression. In total, we demonstrate that combining multiple genome-wide datasets spanning diverse regulatory modes enables accurate delineation of the downstream miRNA-regulated transcriptional network and establishes a model for studying similar networks in other systems.

42. Transient-Evoked Otoacoustic Emissions Reflect Audiometric Patterns of Age-Related Hearing Loss

Abstract: Distinct forms of age-related hearing loss are hypothesized based on evidence from animal models of aging, which are identifiable in human audiograms. The Sensory phenotype results from damage (e.g., excessive noise or ototoxic drugs) to outer hair cells and sometimes inner hair cells, producing large threshold increases predominately at high frequencies. The Metabolic phenotype results from a decline in endocochlear potential that can reduce outer hair cell motility throughout the cochlea, producing gradually sloping thresholds from lower to higher frequencies. Finally, the combined Metabolic + Sensory phenotype results in low-frequency losses similar to the Metabolic phenotype and high-frequency losses similar to the Sensory phenotype. Because outer hair cell function appears to be affected differently in each phenotype, this study used audiograms from 618 adults aged 50 to 93 years ( n = 1,208 ears) to classify phenotypes and characterize differences in transient-evoked otoacoustic emission (TEOAE) data. Significant phenotype differences were observed in frequency-band TEOAEs and configuration (intercept and slope), including large and broadly distributed TEOAE reductions for Metabolic and Metabolic + Sensory ears and more focused high-frequency TEOAE reductions for Sensory ears. These findings are consistent with metabolic declines that reduce cochlear amplification across a broad range of frequencies and more basally situated, high-frequency declines in sensory hearing loss. The results provide further validation for the classification of age-related hearing loss phenotypes based on audiograms and show human TEOAE declines that are highly consistent with animal models.

43. Strength-Endurance Training Reduces the Hamstrings Strength Decline Following Simulated Football Competition in Female Players

Abstract: Hamstring strains are the most common injury in multiple sprint sports, with inadequate eccentric hamstring strength and fatigue identified as important risk factors. Resistance training interventions aimed at reducing injury risk typically focus on the development of maximum strength, while little is known about the impact of training on hamstring fatigue resistance. The present study compared the effects of strength endurance (SE) with a strength intervention (S) on the eccentric hamstring strength decline induced by a simulated soccer match. Twenty-one female soccer players were randomly assigned to a S group (n = 10) or a SE group (n = 11). Hamstrings and quadriceps isokinetic concentric and eccentric peak torque (PT) were assessed at 120°.s-1 and hamstrings-to-quadriceps ratio (HEcc:QCon) calculated, pre- and immediately post a 90-min simulated match (BEAST90). This was repeated following a 7-week intervention of either three to five sets of 6RM leg curl and stiff-leg deadlift with 3-min inter-set rest (S), or the same exercises performed using three sets of 12–20 RM with 45–90 s inter-set rest (SE). At baseline, the simulated match led to significant declines in hamstrings eccentric peak torque (EccPT) in both groups in both dominant (D) and non-dominant (ND) legs [SE: (D: -15.5, ND: -15.6%), P = 0.001 to 0.016; S: (D: -12.3%, ND: -15.5%), P = 0.001 to 0.018]. After the 7-week intervention, we observed a group∗intervention∗match interaction such that there was no significant decline in EccPT in the SE group following the simulated match (D: 5.3%, ND: 2.0%), but there remained significant declines in the S group (D: -14.2%, ND: -15.5%, P = 0.018–0.001). Similarly, in the SE group, there was a significant decrease in the HEcc:QCon in D before (-14.2%, P = 0.007), but not after the training intervention, whereas declines were observed in the S group both at baseline, and following the intervention (D: -13.9%, ND: -15.6%, P = 0.045). These results demonstrate that SE training can reduce the magnitude of the EccPT decline observed during soccer competition. As inadequate eccentric strength and fatigue are both risk factors for hamstring injury, SE training should be considered along with the development of peak eccentric strength, as a component of programs aimed at reducing injury risk in multiple-sprint sports.

Abstract: A cascade of alternative sigma factors directs developmental gene expression during spore formation by the bacterium Bacillus subtilis. As the spore develops, a tightly regulated switch occurs in which the early-acting sigma factor σF is replaced by the late-acting sigma factor σG. The gene encoding σG (sigG) is transcribed by σF and by σG itself in an autoregulatory loop; yet σG activity is not detected until σF-dependent gene expression is complete. This separation in σF and σG activities has been suggested to be due at least in part to a poorly understood intercellular checkpoint pathway that delays sigG expression by σF. Here we report the results of a careful examination of sigG expression during sporulation. Unexpectedly, our findings argue against the existence of a regulatory mechanism to delay sigG transcription by σF and instead support a model in which sigG is transcribed by σF with normal timing, but at levels that are very low. This low-level expression of sigG is the consequence of several intrinsic features of the sigG regulatory and coding sequence-promoter spacing, secondary structure potential of the mRNA, and start codon identity-that dampen its transcription and translation. Especially notable is the presence of a conserved hairpin in the 5' leader sequence of the sigG mRNA that occludes the ribosome-binding site, reducing translation by up to 4-fold. Finally, we demonstrate that misexpression of sigG from regulatory and coding sequences lacking these features triggers premature σG activity in the forespore during sporulation, as well as inappropriate σG activity during vegetative growth. Altogether, these data indicate that transcription and translation of the sigG gene is tuned to prevent vegetative expression of σG and to ensure the precise timing of the switch from σF to σG in the developing spore.

Abstract: Taro, Colocasia esculenta (L.) Schott, is a vegetable and starchy root crop cultivated in Asia, Oceania, the Americas, Africa, and the Mediterranean. Very little is known about its early history in the Mediterranean, which previous authors have sought to trace through Classical (Greek and Latin) texts that record the name colocasia (including cognates) from the 3rd century BC onwards. In ancient literature, however, this name also refers to the sacred lotus, Nelumbo nucifera Gaertn. and its edible rhizome. Like taro, lotus is an alien introduction to the Mediterranean, and there has been considerable confusion regarding the true identity of plants referred to as colocasia in ancient literature. Another early name used to indicate taro was arum, a name already attested from the 4th century BC. Today, this name refers to Arum, an aroid genus native to West Asia, Europe, and the Mediterranean. Our aim is to explore historical references to taro in order to clarify when and through which routes this plant reached the Mediterranean. To investigate Greek and Latin texts, we performed a search using the Thesaurus Linguae Graecae (TLG) and the Thesaurus Linguae Latinae (TLL), plus commentaries and English and French translations of original texts. Results show that while in the early Greek and Latin literature the name kolokasia (Greek κολοκάσια) and its Latin equivalent colocasia refer to Nelumbo nucifera Gaertn., after the 4th century AD a poorly understood linguistic shift occurs, and colocasia becomes the name for taro. We also found that aron (Greek ἄρον) and its Latin equivalent arum are names used to indicate taro from the 3rd century BC and possibly earlier.

47. Verbal Learning and Memory in Cannabis and Alcohol Users: An Event-Related Potential Investigation

Abstract: Aims: Long-term heavy use of cannabis and alcohol are known to be associated with memory impairments. In this study, we used event-related potentials to examine verbal learning and memory processing in a commonly used behavioral task.Method: We conducted two studies: first, a small pilot study of adolescent males, comprising 13 Drug-Naive Controls (DNC), 12 heavy drinkers (HD) and 8 cannabis users (CU). Second, a larger study of young adults, comprising 45 DNC (20 female), 39 HD (16 female), and 20 CU (9 female). In both studies, participants completed a modified verbal learning task (the Rey Auditory Verbal Learning Test, RAVLT) while brain electrical activity was recorded. ERPs were calculated for words which were subsequently remembered vs. those which were not remembered, and for presentations of learnt words, previously seen words, and new words in a subsequent recognition test. Pre-planned principal components analyses (PCA) were used to quantify the ERP components in these recall and recognition phases separately for each study.Results: Memory performance overall was slightly lower than published norms using the standardized RAVLT delivery, but was generally similar and showed the expected changes over trials. Few differences in performance were observed between groups; a notable exception was markedly poorer delayed recall in HD relative to DNC (Study 2). PCA identified components expected from prior research using other memory tasks. At encoding, there were no between-group differences in the usual P2 recall effect (larger for recalled than not-recalled words). However, alcohol-related differences were observed in a larger P540 (indexing recollection) in HD than DNC, and cannabis-related differences were observed in a smaller N340 (indexing familiarity) and a lack of previously seen > new words effect for P540 in Study 2.Conclusions: This study is the first examination of ERPs in the RAVLT in healthy control participants, as well as substance-using individuals, and represents an important advance in methodology. The results indicate alterations in recognition memory processing, which even if not manifesting in overt behavioral impairment, underline the potential for brain dysfunction with early exposure to alcohol and cannabis.

Abstract: Abstract Invasive alien species (IAS) are species whose introduction or spread outside their native range threatens biological diversity, ecosystem functioning, economy, and/or public health. The recent European Union (EU) regulation on the management of IAS emphasizes the need for a consistent approach to alien species assessment that will underpin international measures for the early identification of newly introduced IAS followed by rapid action aimed at the prevention of introduction, spread, and negative impacts. The goals of the present study were (1) to present the risk classifications of 18 aquatic alien species for The Netherlands using the Invasive Species Environmental Impact Assessment protocol, (2) to compare these with available risk classifications made for countries spanning similar climatic and biogeographical regions to the EU, and (3) to provide explanations for inconsistencies between different risk classifications. Five species were classified as high risk: Carassius gibelio (Prussian carp), Cyprinus carpio (common carp), Sander lucioperca (pike‐perch), Cabomba caroliniana (fanwort), and Dreissena rostriformis bugensis (quagga mussel). Of the 14 species with existing risk classifications for countries spanning similar climatic and biogeographical regions to the EU, all but two of the assessed species (C. gibelio and D. rostriformis bugensis) were classified inconsistently. Reasons for these inconsistencies are the application of different risk assessment schemes, application on a national rather than biogeographical scale, differences in the definition and application of criteria, differences in habitat availability, and uncertainties that are intrinsic to risk assessment methodologies. Approaches that increase transparency by highlighting these methodological aspects, normative choices, and uncertainties are vital to the legitimacy of any risk assessment method and will increase acceptance among decision makers, nature managers, and stakeholders.

49. Examining the use of Amazon's Mechanical Turk for edge extraction of the occlusal surface of fossilized bovid teeth.

Abstract: In order to reconstruct environments associated with Plio-Pleistocene hominins in southern Africa, researchers frequently rely upon the animals associated with the hominins, in particular, animals in the Family Bovidae. Bovids in southern Africa are typically identified by their teeth. However, identifying the taxon of a bovid tooth is challenging due to various biasing factors. Furthermore, inaccurate identification of fossil bovids can have significant consequences on the reconstructed paleoenvironment. Recent research on the classification of bovid fossil teeth has relied on using elliptical Fourier analysis to summarize the shape of the outline of the occlusal surface of the tooth and the resulting harmonic amplitudes. Currently, an expert in the field must manually place landmarks around the edges of each tooth which is slow and time consuming. This study tests whether it is possible to crowdsource this task, while maintaining the necessary level of quality needed to perform a statistical analysis on each tooth. Amazon Mechanical Turk workers place landmarks on the edge of the tooth which is compared to the performance of an expert in the field. The results suggest that crowdsourcing the digitization process is reliable and replicable. With the technical aspects of digitization managed, researchers can concentrate on analyzing and interpreting the data.

50. Anti-MuSK-Positive Myasthenic Crisis in a 7-Year-Old Female

Abstract: A seven-year-old African American female with anti-MuSK-positive Juvenile Myasthenia Gravis collapsed while at school from progressively worsening weakness and dyspnea. On initial emergency department presentation, she required 15 liters per minute of supplemental oxygen to maintain oxygen saturation above 92%. Initial pulmonary function tests and venous blood gas led to the decision to place her on noninvasive positive pressure ventilation (NPPV) with BiPAP in the emergency department. Due to worsening hypercarbia, she later required mechanical intubation in the PICU and underwent IVIG therapy followed by plasmapheresis in order to achieve a stable discharge from the hospital. A respiratory virus panel PCR was positive for influenza A, influenza B, and rhinovirus, likely precipitating the respiratory failure and myasthenic crisis in this seven-year-old patient. Given the rarity of this condition, this case report is to provide further education to the clinician managing severe, prepubertal Juvenile Myasthenia Gravis and myasthenic crisis.
