Listeriosis is highly prevalent in the animal farming industry, with Listeria monocytogenes as the causative pathogen. To identify potential therapeutic targets for LM infection, we investigated the mechanisms of LM infection in goat uteri. We inoculated a group of goats with LM via jugular vein injection, isolated and raised them, and subsequently collected sterile samples of their uterine tissue after they exhibited clinical symptoms of LM infection. We used Giemsa staining, immunohistochemical staining, real-time qPCR, and Western blotting as experimental methods.First, we investigated the mechanism of Listeria monocytogenes (LM) infection in the goat uterus by examining the expression levels of listeriolysin O, E-cadherin, and tyrosine kinase c-Met in the uterus. Furthermore, we investigated the impact of LM infection on uterine autophagy and cell apoptosis. The results indicate that the injection of LM into the goats' jugular veins leads to LM infection in the goats' uteri. During LM survival inside the goat uterine cells, there is a significant increase in the expression levels of LLO, E-cadherin, and c-Met in the host uterine tissue. This suggests that LM may potentially infect goat uteri through the InlA/E-cadherin and InlB/c-Met pathways. Furthermore, LM infection increases the levels of apoptosis and autophagy in goat uteri. Apoptosis genes Bcl-2 and Bax, as well as autophagyrelated genes LC3B, PINK1, and Parkin, exhibit varying degrees of changes in localization and expression in goat uteri, mediating the occurrence of apoptotic and autophagic responses. Recently, some scholars have proposed that the South China Block (SCB) was controlled by a compressive tectonic regime in the middle-late Early Cretaceous, challenging the belief that the SCB was under an extensional setting during the Cretaceous. The Early Cretaceous tectonic setting constraint in the SCB can offer vital insight to clarify the Mesozoic subduction history of the Paleo-Pacific. Therefore, to determine the SCB tectonic regime during the Early Cretaceous, this study investigated sedimentary rocks from the Lower Cretaceous Heshui Formation in the Xingning Basin, a foreland basin located in the southeastern SCB. Provenance analysis was performed using sandstone modal analysis, sandstone geochemical characteristics, and detrital zircon geochronology. Based on the results, we discussed basin sediment sources and the SCB tectonic regime during the Early Cretaceous. The results showed that the maximum Heshui Formation depositional age was 103 Ma ± 1.6 Ma in the Early Cretaceous Albian. Detrital framework modes and geochemical characteristics of sandstone indicated that Heshui Formation's source rocks were granites and sedimentary rocks. The detrital zircon U-Pb ages could be classified into two major and four subordinate age populations. The Wuyi Terrane to the north and southeast coastal regions to the east were the primary potential Heshui Formation source areas. However, the lower and upper sandstones are different in the peak ages, 437 and 146 to 104 Ma, respectively, indicating that the major source area shifted from the Wuyi Terrane to the southeastern coastal regions during the late Early Cretaceous. The sandstone modal analysis results indicated that the source area comprised mainly collisional-orogenic material. Understanding how blood lipid levels change with age in the general population is a precondition to defining dyslipidemia. To explore age-related trends in LDL-C and non-HDL-C levels in the general population, a large-scale cross-sectional study with 49,201 males and 35,084 females was adopted. Trends of non- HDL-C and LDL-C levels were plotted against each age (18 to 85 years old, one-year increments); the trends, as well as the influence of confounding factors on the trends, were validated and adjusted by linear regression modeling. The trajectory of LDL-C and non-HDL-C levels by age displayed a nonlinear correlation trend. Further multivariate linear regression modeling that incorporated sex-specific age phases showed that age was positively associated with LDL-C and non-HDL-C levels, with coefficients of 0.018 and 0.031, respectively, in females aged ≥18 to ≤56 years and negatively associated with LDL-C and non-HDL-C levels, with coefficients of -0⋅013 and -0.015, respectively, in females aged ≥57 years. The LDL-C and non-HDL-C levels increased with age in males ≥18 to ≤33 years of age, with coefficients of 0.025 and 0.053, respectively; the lipid levels plateaued at ≥34 to ≤56 years of age and subsequently decreased in those ≥57 years of age, with coefficients of -0.008 and -0.018, respectively. In contrast, pooled analyses without age stratification concealed these details. In conclusion, fluctuating increasing and decreasing lipid levels occurred with phases of aging in both sexes. The present study aimed to investigate the association between breast cancer susceptibility gene 1 (BRCA1) expression and drug resistance in colon cancer, with the specific aim of elucidating the underlying molecular mechanisms of vincristine (VCR) resistance in tumor cells. The HCT‑8 human colon cancer cell line was used to establish the VCR‑resistant HCT‑8/V line by gradually increasing the concentration of VCR during cell culture. The relative mRNA and protein expression levels of BRCA1 in these colon cancer cell lines was assessed by reverse transcriptase‑quantitative polymerase chain reaction (RT‑qPCR) analysis and western blotting, respectively. Resistance to VCR was established in the HCT‑8/V colon cancer cells, and RT‑qPCR and western blot analysis revealed the expression of BRCA1 to be significantly higher in the VCR‑resistant cells compared with their drug‑sensitive counterparts (P<0.05). The decreased BRCA1 expression in these VCR‑resistant cells may be associated with the drug resistance frequently observed in colon cancer. [ABSTRACT FROM AUTHOR]

Laser-induced rear surface breakdown process of sodium chloride (NaCl) optical window was investigated based on the time-resolved shadowgraphy and interferometry. Violent particle ejection behavior lasting from tens of nanoseconds to tens of microseconds after the breakdown was observed. Classified by the particle velocity and propagating direction, the ejection process can be divided into three phases: (1) high-speed ejection of liquid particles during the first 100-ns delay; (2) micron-sized material clusters ejection from ~100-ns to ~1-µs delay; (3) larger and slower solid-state particles ejection from ~1 µs to tens of microseconds delay. The moving directions of particles in the first and third phases are both perpendicular to the sample surface while particles ejected in the second phase exhibits angular ejection and present a V-like particle pattern. Mechanisms include explosive boiling, impact ejection, and shockwave ejection are discussed to explain this multiple phase ejection behavior. Our results highlight the significance of impact ejection induced by recoil pressure and backward propagating internal shockwave for laser-induced rear surface breakdown events of optical materials with low melting point. Di-2-pyridylketone-4,4,-dimethyl-3-thiosemicarbazone (Dp44mT) exhibits significant antitumor activity. However, the mechanism of its pharmacological interaction with human serum albumin (HSA) and DNA remains poorly understood. Here, we aimed to elucidate the interactions of Dp44mT with HSA and DNA using MTT assays, spectroscopic methods, and molecular docking analysis. Our results indicated that addition of HSA at a ratio of 1:1 did not alter the cytotoxicity of Dp44mT, but did affect the cytotoxicity of the Dp44mT-Cu complex. Data from fluorescence quenching and UV-VIS absorbance measurements demonstrated that Dp44mT could bind to HSA with a moderate affinity (Ka = approximately 104 M-1). CD spectra revealed that Dp44mT could slightly disrupt the secondary structure of HSA. Dp44mT could also interact with Ct-DNA, but had a moderate binding constant (KEB = approximately 104 M-1). Docking studies indicated that the IB site of HSA, but not the IIA and IIIA sites, could be favorable for Dp44mT and that binding of Dp44mT to HSA involved hydrogen bonds and hydrophobic force, consistent with thermodynamic results from spectral investigations. Thus, the moderate binding affinity of Dp44mT with HSA and DNA partially contributed to its antitumor activity and may be preferable in drug design approaches. [ABSTRACT FROM AUTHOR]

Employing a pump-probe polarization-based two-frame shadowgraphy setup, the formation of filamentary damage induced in bulk fused silica by a nanosecond pulse at 1064 nm is investigated with a picosecond probe. Three different phases are exhibited in the damage experiments. The first phase is the formation of a micrometric plasma channel along the laser direction during the beginning of the pulse likely caused by multi-photon ionization. This channel exhibits growth during 400 ps, and the newly grown plasma is discrete. Then, during the end of the pulse, this channel evolves into a tadpole-like morphology showing an elliptical head upstream the laser flux followed by a thin tail. This observed asymmetry is attributed to shielding effects caused by both the plasma and hot modified silica. Once the damage shows its almost final morphology, a last phase consists in the launch of a pressure wave enlarging it after the laser pulse. The physical mechanisms that might be involved in the formation of plasma channels are discussed. The experimental data are first confronted to the moving breakdown model which overestimates the filamentary damage length. Finally, taking into account the temporal shape of the laser pulses, the coupling between Kerr-induced self-focusing and stimulated Brillouin scattering is discussed to interpret the observations. Recently, topological insulator based heterostructures (HSs) have attracted tremendous research interest, due to their efficient carrier transfer features at the heterointerface induced by metallic surface states. Here, a novel HS comprising 0D perovskite CsPbBr3 quantum dots (QDs) and 2D material topological insulator Bi2Se3 film is proposed and experimentally investigated. Specifically, steady state and time-resolved photoluminescence (PL) measurements are employed, from which a significant quenching behaviour is observed in the HS, with an average quenching factor of 93.2 ± 0.8%. Additionally, time-resolved PL spectroscopy affirms that the carrier transfer efficiency can be up to 92.6 ± 0.2%. Furthermore, the dynamics of carrier transfer within the 0D-2D HS are characterized by utilizing femtosecond broadband transient absorption (TA) spectroscopy, revealing an ultrafast exciton transfer from photoexcited CsPbBr3 QDs to the Bi2Se3 film with a time-scale around 1.1 ± 0.2 ps. An alternative important finding is that the band renormalization is exhibited in CsPbBr3 QDs of the HS, with the dominant factor being the Coulomb screening effect.
