In this paper, we demonstrate a tunable mid-infrared (mid-IR) optical parametric oscillator (OPO) using an MgO: PPLN crystal with a polarization period of 29.8 μm, which was end-pumped by a 1064 nm electro-optic cavity-dumped all-solid-state laser (1064 nm EOCD-laser) with a pulse repetition frequency range of 100 Hz to 1 kHz. Thanks to the use of 808 nm pulsed laser diode (LD) side-pumping Nd: YAG crystal, transverse electro-optic modulating (TEOM) MgO: LN crystal, and birefringent crystal (BC) filtering in the 1064 nm cavity-dumped cavity, both the 1064 nm EOCD-laser and the idler light output by the OPO exhibited high peak power and low peak-to-peak instability in both pulse width and single pulse energy. By combining temperature tuning of the MgO: PPLN crystal from 30 °C to 200 °C, wavelength tuning of the idler light from 3705.2 to 3424.3 nm was achieved. For the first time, a pulsed idler light at 3705.2 nm with a high peak power of 1.224 MW, a narrow pulse width of 4.97 ns, and an optical-to-optical (1064 nm to 3705.2 nm) conversion efficiency of 38.1% was obtained, corresponding to a tuning temperature of 30 °C and a pulse repetition frequency of 100 Hz. The effect of temperature on the corrosion resistance performance of the passivation film (PF) formed at 20 °C to 60 °C was investigated systematically in Cl
− ion or/and SO4 2− ion solution. It is found that the temperature accelerates the growth rate of PF and increases the thickness of PF. In addition, the critical chloride ion concentration of PF at 20 °C to 40 °C, and that of PF at 50 °C and 60 °C, which is increased by 15.8%. In the single SO4 2− , all the PF were not depassivated with the increased concentration of SO4 2− . However, at the presence of Cl− , the PF were depassivated once the concentration of SO4 2− reaches 0.18 mol/L. These results show that in a single ion environment, the corrosion resistance of PF increased with the passivation temperature, while the impact of temperature on the corrosion resistance of PF is negligible in the combine solution of 3.5 wt% Cl− and SO4 2− ions. Hepatocellular carcinoma (HCC) is one of the most common primary neoplasms of the liver and one of the most common solid tumors in the world. Its global incidence is increasing and it has become the third leading cause of cancer-related deaths. There is growing evidence that chemokines play an important role in the tumor microenvironment, regulating the migration and localization of immune cells in tissues and are critical for the function of the immune system. This review comprehensively analyses the expression and activity of chemokines in the TME of HCC and describes their interrelationship with hepatocarcinogenesis and progression. Special attention is given to the role of chemokine-chemokine receptors in the regulation of immune cell accumulation in the TME. Therapeutic strategies targeting tumor-promoting chemokines or the induction/ release of beneficial chemokines are reviewed, highlighting the potential value of natural products inmodulating chemokines and their receptors in the treatment of HCC. Progress is made in the advancement of efficient and durable electrocatalysts for the ethylene glycol oxidation reaction (EGOR), which is a critical breakthrough in the conversion of waste plastic into high‐value‐added chemicals. Here, an organic ligand‐modified PdNi bimetallenes (PdNi O‐BMs) catalyst is designed and constructed through the one‐pot method. Benefiting from the frizzy and porous surface structure and inorganic–organic hybrid interface modification, PdNi O‐BMs undergo electrocatalytic reforming of ethylene glycol (EG) derived from polyethylene terephthalate (PET) to form formic acid. The unique structural advantages result in high EGOR performance. Meanwhile, the influence of Ni and organic ligand on the catalyst performance is confirmed through density functional theory calculations. This study reveals an advanced strategy for the synthesis of inorganic–organic hybrid bimetallenes, providing a green and sustainable new approach for PET value‐added transformation. Background: Patients with frailty are more prone to have perioperative adverse events, and enhanced recovery after surgery (ERAS) has been widely adopted to improve perioperative outcomes. The purpose of this study was to assess the impact of improved compliance with ERAS on perioperative outcomes in frail patients. Methods: Geriatric patients (over 65 years) who underwent multi-level lumbar fusion surgery between June 2017 and June 2022 were included. The patients were divided into two groups according to their degree of compliance with the ERAS. Stepwise nearest-neighbor propensity score matching 1:1 cohorts for age, gender, body mass index (BMI), American Society of Anesthesiologists (ASA) classfication and Charlson Comorbidity Index (CCI) was performed between groups, namely frail-compliant (FC), frail-noncompliant (FN). Further length of stay (LOS), complications and clinical efficacy were compared between groups. Results: There were 83 pairs of well-balanced patients with comparable clinical baseline data. It was worth noting that patients in FC group has significant lower overall complications (20.5% in the FC group vs 39.8% in the FN group, P = 0.007), major complications (7.2% in the FC group vs 19.3% in the FN group, P = 0.022) and shorter LOS (11.18 ± 5.32 in the FC group vs 14.45 ± 4.68 in the FN group, P < 0.001) than patients in FN group. In addition, the initial occurrence of ambulation (2.14 ± 1.21 in FC group vs 3.18 ± 1.73 in FN group, P = 0.012) and bowel movement (3.68 ± 1.24 in FC group vs 4.17 ± 1.32 in FN group, P = 0.031) were earlier for patients in FC group than patients in FN group. With regard to clinical efficacy, there were no significant difference between FC and FN group in terms of patients who meet minimal clinical important difference (MCID) for Oswestry Disability Index (ODI) at postoperative day (POD) 30, Visual Analog Scale (VAS) for back at POD 30-90 and VAS for legs at POD 30, 90, and 180 follow-up intervals. However, there were significant more patients meeting MCID for ODI at POD 90 and180, and VAS for back at POD 180 between FC and FN group. Conclusions: In this retrospective cohort study, we found that frail patients with higher ERAS adherence group had a lower incidence of overall complication, mjor complications, and a shorter LOS than their lower ERAS adherence counterparts. In addition, frail patients with higher ERAS adherence had earlier ambulatioin and bowel movement. More importantly, we found there were significant more patients meeting MCID for ODI at POD 90 and180, and VAS for back at POD 180 in higher ERAS adherence than their lower counterparts. Precise diagnostic biomarkers of anticitrullination protein antibody (ACPA)‐negative and early‐stage RA are still to be improved. We aimed to screen autoantibodies in ACPA‐negative patients and evaluated their diagnostic performance. The human genome‐wide protein arrays (HuProt arrays) were used to define specific autoantibodies from the sera of 182 RA patients and 261 disease and healthy controls. Statistical analysis was performed with SPSS 17.0. In Phase I study, 51 out of 19,275 recombinant proteins covering the whole human genome were selected. In Phase II validation study, anti‐ANAPC15 (anaphase promoting complex subunit 15) exhibited 41.8% sensitivity and 91.5% specificity among total RA patients. There were five autoantibodies increased in ACPA‐negative RA, including anti‐ANAPC15, anti‐LSP1, anti‐APBB1, anti‐parathymosin, and anti‐UBL7. Anti‐parathymosin showed the highest prevalence of 46.2% (p = 0.016) in ACPA‐negative early stage (<2 years) RA. To further improve the diagnostic efficacy, a prediction model was constructed with 44 autoantibodies. With increased threshold for RA calling, the specificity of the model is 90.8%, while the sensitivity is 66.1% (87.8% in ACPA‐positive RA and 23.8% in ACPA‐negative RA) in independent testing patients. Therefore, HuProt arrays identified RA‐associated autoantibodies that might become possible diagnostic markers, especially in early stage ACPA‐negative RA. [ABSTRACT FROM AUTHOR]

Roses, a popular ornamental crop, often face various abiotic stresses during growth and development, such as cold, drought, and salinity. Rosa multiflora is a commonly used rootstock and exhibits strong resistance to both biotic and abiotic stresses, making it an ideal material for studying mechanisms for resistance. Among the largest plant families, MYB transcription factors play a crucial role in plant abiotic stresses. Our previous research has indicated that RmMYB44 could be involved in the low-temperature response of R. multiflora. This study further investigated RmMYB44, revealing that its expression levels were upregulated in response to chilling, drought, and salt stress. The results suggested its potential role as a key transcription factor in plant resistance to abiotic stresses. Additionally, RmMYB44 encoded a nuclear-localized protein without the self-activating function. The overexpression of RmMYB44 in tobacco plants enhanced the resistance to cold, drought, and salt stresses, as evidenced by the improved growth compared to wild-type (WT) plants under conditions of 4 °C, 30% water-holding capacity, and 200 mM of NaCl, respectively. Moreover, in overexpression tobacco plants, the levels of hydrogen peroxide and malondialdehyde (MDA) were significantly reduced; and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT); as well as the proline content and the expression levels of NtPOD, NtCAT, and NtCBF; were significantly elevated under abiotic stresses. We assumed that the resistance to abiotic stress in plants conferred by RmMYB44 was associated with the regulation of cell membrane integrity. This study aimed to elucidate the role of the RmMYB44 gene in the resistance mechanism of R. multiflora against abiotic stress, thereby providing a candidate gene for the molecular breeding of abiotic stress resistance in roses and related species. Deep learning is applied to many complex tasks in the field of wireless communication, such as modulation recognition of spectrum waveforms, because of its convenience and efficiency. This leads to the problem of a malicious third party using a deep learning model to easily recognize the modulation format of the transmitted waveform. Some existing works address this problem directly using the concept of adversarial examples in the computer vision field without fully considering the characteristics of the waveform transmission in the physical world. Therefore, we propose two low‐interception waveforms (LIWs) generation methods, the LIW and ULIW algorithms, which can reduce the probability of the modulation being recognized by a third party without affecting the reliable communication of the friendly party. Among them, ULIW improves LIW algorithm by simulating channel noise during training cycle, and substantially reduces the perturbation magnitude while maintaining low interception accuracy. Our LIW and ULIW exhibit significant low‐interception performance in both numerical simulations and hardware experiments. UvrC is a subunit of excinuclease ABC, which mediates nucleotide excision repair (NER) in bacteria. Our previous studies showed that transposon Tn4531 insertion in the UvrC encoding gene Riean_1413 results in reduced biofilm formation by Riemerella anatipestifer strain CH3 and attenuates virulence of strain YZb1. In this study, whether R. anatipestifer UvrC has some biological functions other than NER was investigated. Firstly, the uvrC of R. anatipestifer strain Yb2 was in-frame deleted by homologous recombination, generating deletion mutant ΔuvrC, and its complemented strain cΔuvrC was constructed based on Escherichia coli – R. anatipestifer shuttle plasmid pRES. Compared to the wild-type (WT) R. anatipestifer strain Yb2, uvrC deleted mutant ΔuvrC significantly reduced biofilm formation, tolerance to H
2 O2 - and HOCl-induced oxidative stress, iron utilization, and adhesion to and invasion of duck embryonic hepatocytes, but not its growth curve and proteolytic activity. In addition, animal experiments showed that the LD50 value of ΔuvrC in ducklings was about 13-fold higher than that of the WT, and the bacterial loads in ΔuvrC infected ducklings were significantly lower than those in Yb2-infected ducklings, indicating uvrC deletion in R. anatipestifer attenuated virulence. Taken together, the results of this study indicate that R. anatipestifer UvrC is required for iron utilization, biofilm formation, oxidative stress tolerance and virulence of strain Yb2, demonstrating multiple functions of UvrC. RESEARCH HIGHLIGHTS Deletion of uvrC in R. anatipestfer Yb2 significantly reduced its biofilm formation. uvrC deletion led to reduced tolerance to H2 O2 - and HOCl-induced oxidative stress. Background: PD-1/PD-L1 blockade has become a powerful method to treat malignant tumors. However, a large proportion of patients still do not benefit from this treatment, due to low tumor immunogenicity and low tumor penetration of the agents. Recently, neutrophil elastase has been shown to induce robust tumor immunogenicity, while the insufficient enzyme activity at the tumor site restricted its anti-tumor application. Here, we designed polyethyleneimine-modified neutrophil elastase (PEI-elastase) loaded with PD-L1small interfering RNA (PD-L1 siRNA) for improving enzymatic activity and delivering siRNA to tumor, which was expected to solve the above-mentioned problems. Results: We first demonstrated that PEI-elastase possessed high enzymatic activity, which was also identified as an excellent gene-delivery material. Then, we synthesized anti-tumor lipopolymer (P-E/S Lip) by encapsulating PEI-elastase and PD-L1siRNA with pH-responsive anionic liposomes. The P-E/S Lip could be rapidly cleaved in tumor acidic environment, leading to exposure of the PEI-elastase/PD-L1 siRNA. Consequently, PEI-elastase induced powerful tumor immunogenicity upon direct tumor killing with minimal toxicity to normal cells. In parallel, PEI-elastase delivered PD-L1siRNA into the tumor and reduced PD-L1 expression. Orthotopic tumor administration of P-E/S Lip not only attenuated primary tumor growth, but also produced systemic anti-tumor immune response to inhibit growth of distant tumors and metastasis. Moreover, intravenous administration of P-E/S Lip into mice bearing subcutaneous tumors leaded to an effective inhibition of established B16-F10 tumor and 4T1 tumor, with histological analyses indicating an absence of detectable toxicity. Conclusions: In our study, a protease-based nanoplatform was used to cooperatively provoke robust tumor immunogenicity and down-regulate PD-L1 expression, which exhibited great potential as a combination therapy for precisely treating solid tumors. The distribution coefficient (K
d ) of radionuclides is a crucial parameter in assessing the safety of high-level radioactive waste (HLW) geological repository. It is determined in the laboratory through batch and column experiments. However, differences in obtained Kd values from distinct experiments have not been thoroughly assessed and compared. This study evaluated strontium (Sr) sorption on different granite materials using static batch and dynamic experiments (column and core-flooding experiments). The results from batch sorption experiments showed higher Sr sorption on granite under acidic and strongly alkaline conditions, low solid–liquid ratios, and low ionic strength. In column experiments, a two-site sorption model was used to simulate Sr transport in crushed granite and mixed pure minerals. The sorption of Sr on crushed granite exhibited a higher affinity than that of mixed pure minerals. The dual-porosity transport model was employed to investigate Sr transport behavior in fractured granite in the core-flooding experiment. Kd obtained from batch sorption experiments are four to twenty times higher than those from column experiments, and two to three orders of magnitude higher than that from a core-flooding experiment. 14. Discharge teaching quality positively predicts quality of life in colorectal cancer patients with temporary enterostomy: The mediating role of readiness for hospital discharge and stoma self-efficacy.

Objectives: This study aimed to examine the mediating role of readiness for hospital discharge (RHD) and stoma self-efficacy (SSE) in the relationship between quality of discharge teaching (QDT) and health-related quality of life (HRQOL) in colorectal cancer patients with temporary enterostomy, and the gender difference of mediating effect. Background: It is not clear how RHD, QDT, SSE and HRQOL interact in colorectal cancer patients with temporary enterostomy. Methods: This was a prospective follow-up survey. 221 colorectal cancer patients with temporary enterostomy were conveniently recruited from a general hospital in Southeast China. The Quality of Discharge Teaching Scale, Readiness for Hospital Discharge Scale, Stoma Self-Efficacy Scale, and Stoma Quality of Life Scale were used to collect data. Pearson's correlation and structural equation models were used to analyze the data. SPSS 26.0 and Amos 28.0 software were used for analysis the collected data. Results: Regarding the relationship of QDT and HRQOL, only QDT-T had a direct effect among colorectal cancer patients with stomas (b = 0.233, P<0.001, percentile 95% CI = [0.145, 0.314]). However, both QDT-T and QDT-R can predict HRQOL indirectly through three paths: (1) the mediating role of SSE (b = 0.050, P = 0.009, percentile 95% CI = [0.013, 0.098]; b = 0.077, P = 0.008, percentile 95% CI = [0.021, 0.164]), (2) the mediating role of RHD (b = 0.044, P = 0.004, percentile 95% CI = [0.014, 0.085]; b = 0.044, P = 0.005, percentile 95% CI = [0.010, 0.102]), and (3) the chain mediating role of SSE and RHD (b = 0.030, P = 0.003, percentile 95% CI = [0.011, 0.059]; b = 0.047, P = 0.003, percentile 95% CI = [0.015, 0.103]). The similar chain mediating effect in male stoma patients was also found (b = 0.041, P = 0.002, percentile 95% CI = [0.016, 0.080]; b = 0.046, P = 0.004, percentile 95% CI = [0.011, 0.114]). Conclusions: Stoma self-efficacy and readiness for hospital discharge played important intermediary roles in the relationship between quality of discharge teaching and health-related quality of life in colorectal cancer patients with stomas. 15. Insulin attenuates LPS‐induced cognitive impairment and ferroptosis through regulation of glucose metabolism in hippocampus.

Aims: Neuroinflammation is a recognized contributor to cognitive disorders like Alzheimer's disease, with ferroptosis emerging as a novel mechanism underlying cognitive dysfunction associated with neuroinflammation. Insulin, pivotal in the central nervous system, holds promise for cognitive function enhancement. This study aimed to establish a cognitive impairment model through intracerebroventricular injection of lipopolysaccharide (LPS) and explore the impact of intracerebroventricular insulin injection on cognitive function in mice. Methods: We employed diverse experimental techniques, including animal behavior testing, molecular assays, targeted metabolomics, nuclear medicine, and electron microscopy, to assess neurodegenerative changes, brain insulin resistance (IR), glucose uptake and metabolism, and ferroptosis. The model of cognitive impairment was induced via intracerebroventricular injection of LPS, followed by intracerebroventricular administration of insulin to evaluate its effects. Results: Insulin treatment effectively mitigated LPS‐induced cognitive decline and safeguarded against neuronal degeneration. Furthermore, insulin alleviated LPS‐induced insulin resistance, enhanced glucose uptake in the hippocampus, and promoted the Pentose Phosphate Pathway (PPP) and nicotinamide adenine dinucleotide phosphate (NADPH) production. Additionally, insulin activated the glutathione (GSH)‐glutathione peroxidase 4 (GPX4) pathway, reducing lipid peroxidation, and mitochondrial damage characteristic of LPS‐induced ferroptosis in the hippocampus. Conclusion: Our findings underscore the therapeutic potential of insulin in alleviating LPS‐induced cognitive impairment and ferroptosis by modulating glucose metabolism. In order to meet the increasing requirements of speed and distance, an advanced digital signal processing (DSP) algorithm is preferred without changing the system structure in intensity modulation and the direct detection (IM/DD) system. As the transmission distance increases, the power fading induced by dispersion must be mitigated. In addition, linear and nonlinear inter symbol interference (ISI) introduced by bandwidth limitation and device imperfections becomes an obstacle to achieving higher capacity. The Gerchberg–Saxton (GS) algorithm was recently used to compensate for dispersion. In this paper, GS-based pre- and post-compensation schemes in the IM/DD system with nonlinearity were investigated. We investigated and compared the performance of the GS-based pre- and post-compensation algorithm in a 28 GB aud four-level pulse amplitude modulation (PAM-4) transmission over 40 km standard single-mode fiber (SSMF). The bit error rate (BER) achieved a threshold of 3.8 × 10
−3 using look-up-table (LUT), FFE, and the GS-based pre-compensation algorithm without iterations. Turning to the GS-based post-compensation scheme, 80 iterations are needed. However, the demand for FFE is reduced. The algorithm selection depends on the tolerance of the transmitter or receiver complexity in specific scenarios. Driver's risk perception ability is of great significance to traffic safety. Driver's risk perception ability is reflected in the risk handling in traffic situation and has a great impact on driving decisions, especially speed decisions. However, the mechanism of the influence of risk perception on driving decisions under risk situations is still unclear. According to standardized risk perception test, we divided drivers into high risk perception ability (H-RPA) group and low risk perception ability (L-RPA) group. Participants were shown traffic pictures with different levels of risk and asked to make decisions about whether to slow down, while their electrical brain responses were recorded. All participants were interviewed after the experiment. The results showed that in the low-risk traffic situation, compared to the drivers with H-RPA, the drivers with L-RPA made less deceleration decisions and evoked larger N1 and smaller P1 amplitude. 93.75% of the drivers with L-RPA stated in interviews that they did not notice the traffic risks on the road. The results show that risk perception affects the allocation of attention to risks in traffic situations, leading to a decrease in the safety of driving decisions. With the rapid development of modern science and technology, navigation technology provides great convenience for people's life, but the problem of inaccurate localization in complex environments has always been a challenge that navigation technology needs to be solved urgently. To address this challenge, this paper proposes an augmented reality navigation method that combines image segmentation and multi-sensor fusion tracking registration. The method optimizes the image processing process through the GA-OTSU-Canny algorithm and combines high-precision multi-sensor information in order to achieve accurate tracking of positioning and guidance in complex environments. Experimental results show that the GA-OTSU-Canny algorithm has a faster image edge segmentation rate, and the fastest start speed is only 1.8 s, and the fastest intersection selection time is 1.2 s. The navigation system combining the image segmentation and sensor tracking and registration techniques has a highly efficient performance in real-world navigation, and its building recognition rates are all above 99%. The augmented reality navigation system not only improves the navigation accuracy in high-rise and urban canyon environments, but also significantly outperforms traditional navigation solutions in terms of navigation startup time and target building recognition accuracy. In summary, this research not only provides a new framework for the theoretical integration of image processing and multi-sensor data, but also brings innovative technical solutions for the development and application of practical navigation systems. The innovative aquaculture equipment known as high-density polyethylene (HDPE) floating rafts has gained popularity among fishermen in the southeast coastal regions of China. Compared to deep-water anti-wave fish cages, the construction costs of HDPE floating rafts are 50% to 75% less. There is a dearth of comprehensive publicly available records of HDPE floating rafts sea trial data, despite substantial numerical studies on the motion response of aquaculture fish cages and scale model experiments under controlled-wave conditions. This study involves sea trial techniques under operational and extreme environmental conditions for motion responses of HDPE floating rafts, presents a comprehensive procedure for sea trials of HDPE floating rafts, summarizes the issues encountered during the trials, and suggests solutions. Using MATLAB for independent programming, motion videos and photos collected from the sea trials are processed for image capture, yielding the original time history curve of vertical displacement. Based on the sea trials' data, including motion displacement, acceleration, mooring line force, overall deformation patterns, and current and wave data, recommendations are provided for the design and layout of HDPE floating rafts. Based on the Fast Fourier Transform (FFT) method for spectral analysis, the influence of interference items on the observational data is eliminated; the rationality of the observational data is verified in conjunction with the results of the Gabor Transform. This study offers a scientific analytical method for the structural design and safe operation of HDPE floating rafts and provides a reference for subsequent numerical simulations. - Abstract
The airport waiting chair frames, as an important part of the overall seating, must be designed to provide comfort, safety, and aesthetic appeal. While the airport furniture industry has made progress in terms of sustainability, more efforts are needed to improve material selection, manufacturing processes, and supply chain management to support the development of sustainable furniture. This study proposes innovative ideas for the lightweight design of the frame, based on the limitations of the existing design. Firstly, structural innovations are discussed, non-traditional mesh panels and curved rounded designs are discussed, and non-introduced mesh panels and curved designs are used to enhance the strength and stability of airport waiting chairs and enhance their overall performance. Secondly, innovations in lightweighting have focused on adjusting the thickness dimensions to enhance comfort, material utilization, and sustainability as well as to achieve a lightweight and thin appearance effect. In order to determine the optimal ranges of values for the thickness of the seat surface support strip (P5), the thickness of the backrest strip (P3), and the thickness of the seat panel (P1), nine groups of chairs with different frame sizes were tested using an orthogonal experimental method. Based on the experimental results for size and topology optimization, NX2312 software modeling will be imported into ANSYS Workbench for static analysis. Using the optimized results, the use of 2.842 kg of steel was successfully reduced by 34.8% to ensure the seat's stability. This provides a reference and idea for the digital and standardized innovative design of airport waiting chair furniture structure in the future. Huangshui is a typical organic wastewater in Chinese Baijiu production, with high pollution and valuable ingredients. Conventional wastewater treatment leads to resource-wasting and environmental pollution. It is urgent that the demand for effective Huangshui treatment with the development of the Baijiu-making industry. This review systematically summarizes recent studies, revealing the main characteristics and application of Huangshui, focusing on the application of the rich microbial resources and flavor substances, which provides a practical approach to cascade and full use of Huangshui in medicine, cosmetic, functional food, fertilizer, and wastewater treatment fields. Further research suggested that Huangshui can also be used as an external carbon source for the denitrification system or as an organic liquid water-soluble fertilizer for more fruits and grains. The applications favor improving production efficiency and lowering pollutant emissions and introduce novel concepts for the sustainable development of related industries. Dwarf apple is one of the most important forms of garden economy, which has become a new engine for rural revitalization. The effective detection of coloring and ripening apples in complex environments is important for the sustainable development of smart agricultural operations. Addressing the issues of low detection efficiency in the greenhouse and the challenges associated with deploying complex target detection algorithms on low-cost equipment, we propose an enhanced lightweight model rooted in YOLOv7. Firstly, we enhance the model training performance by incorporating the Squeeze-and-Excite attention mechanism, which can enhance feature extraction capability. Then, an SCYLLA-IoU (SIoU) loss function is introduced to improve the ability of extracting occluded objects in complex environments. Finally, the model was simplified by introducing depthwise separable convolution and adding a ghost module after up-sampling layers. The improved YOLOv7 model has the highest AP value, which is 10.00%, 5.61%, and 6.00% higher compared to YOLOv5, YOLOv7, and YOLOX, respectively. The improved YOLOv7 model has an MAP value of 95.65%, which provides higher apple detection accuracy compared to other detection models and is suitable for potted dwarf anvil apple identification and detection. Background: The protein kinase domain containing cytoplasmic (PKDCC) gene (OMIM#618821) is associated with bone development. Biallelic variants in the PKDCC gene can cause rhizomelic limb shortening with dysmorphic features. Case Report: A fetus was found to be rhizomelic limb shortening at 16 weeks of gestation and amniocentesis was performed at 19 weeks of gestation. Genomic DNA extracted from the amniotic fluid was subjected to chromosomal microarray analysis (CMA), and Trio‐total whole‐exome sequencing (Trio‐WES). Sanger sequencing was used to verify the candidate pathogenic variants. CMA was normal, while Trio‐WES identified two compound heterozygous variants in the PKDCC gene, namely c.417_c.423delCGGCGCG insTCATGGGCTCAGTACAC(p.G140fs*35) and c.345G>A (p.W115*,379). Then the fetus was aborted and the development of its bone cells were compared with that of a normal fetus of similar gestational age by histopathological examination. Clinical findings of the fetus were shortening humerus and femur, synophrys, much hair on the side face, simian line on the right palm, etc. Histopathological examination showed that the affected fetus had increased proliferative chondrocytes, widened proliferative bands, and delayed bone mineralization. Conclusions: We reported a prenatal case of rhizomelic shortening of limbs caused by compound heterozygous variants in the PKDCC gene, which emphasized the important role of Trio‐WES for diagnosis of skeletal dysplasia in fetuses. Background: Myocardial injury after non-cardiac surgery (MINS) is a common and insidious postoperative complication. This study aimed to evaluate the relationship between the triglyceride-glucose index (TyG) and MINS in advanced-age patients. Methods: We performed a single-center retrospective study including patients ≥ 65 years of age who underwent non-cardiac surgery. The relationship between TyG and MINS was investigated using univariate and multivariate logistic regression analyses. Multivariate logistic regression analysis involved three models: Model I adjusted for preoperative factors, Model II adjusted for surgery-related factors, and Model III adjusted for both preoperative and surgery-related factors. Propensity score matching (PSM) was used to reduce the confounding effects of covariates. Subgroup analyses were then performed to evaluate the relationship between TyG and MINS in various subsamples. Results: A total of 7789 patients were studied, among whom 481 (6.2%) developed MINS. A cut-off value of TyG of 8.57 was determined using a receiver operating characteristic (ROC) curve to be associated with the best predictive performance. Participants with TyG ≥ 8.57 were at a higher risk of developing MINS than those with TyG < 8.57 [n = 273 (7.6%) vs. n = 208 (4.9%), respectively; p < 0.001]. The univariate analysis showed that TyG ≥ 8.57 was significantly associated with MINS in elderly patients [odds ratio (OR): 1.58; 95% confidence interval (95%CI): 1.32–1.91; p < 0.001)]. In multivariate logistic regression, adjustments were made for risk factors including age, sex, body mass index (BMI), hypertension, coronary heart disease, and duration of surgery, etc. The adjusted ORs for TyG ≥ 8.57 were 1.46 (95%CI: 1.17–1.82), p = 0.001; 1.46 (95%CI: 1.19–1.77), p < 0.001; and 1.43 (95%CI: 1.13–1.81), p = 0.003, in the three multivariate models, respectively. The relationship remained after PSM (adjusted OR: 1.35, 95% CI: 1.03–1.78, p = 0.029). Furthermore, the relationship between TyG and MINS remained in a number of subgroups in the sensitivity analyses, but not in participants with peripheral vascular stenosis. Conclusions: A preoperative high TyG (≥ 8.57) is associated with a higher risk of MINS in advanced-age patients undergoing non-cardiac surgery. [ABSTRACT FROM AUTHOR]

Background: Bawei Chenxiang Wan (BCW) is among the most effective and widely used therapies for coronary heart disease and angina pectoris in Tibet. However, whether it confers protection through a right-ventricle (RV) myocardial metabolic mechanism is unknown. Methods: Male Sprague–Dawley rats were orally administrated with BCW, which was injected concurrently with a bolus of Sugen5416, and subjected to hypoxia exposure (SuHx; 5000 m altitude) for 4 weeks. Right ventricular hypertrophy (RVH) in high-altitude heart disease (HAHD) was assessed using Fulton's index (FI; ratio of RV to left ventricle + septum weights) and heart-weight-to-body-weight ratio (HW/BW). The effect of therapeutic administration of BCW on the RVH hemodynamics was assessed through catheterization (mean right ventricular pressure and mean pulmonary artery pressure (mRVP and mPAP, respectively)). Tissue samples were used to perform histological staining, and confirmatory analyses of mRNA and protein levels were conducted to detect alterations in the mechanisms of RVH in HAHD. The protective mechanism of BCW was further verified via cell culture. Results: BCW considerably reduced SuHx-associated RVH, as indicated by macro morphology, HW/BW ratio, FI, mPAP, mRVP, hypertrophy markers, heart function, pathological structure, and myocardial enzymes. Moreover, BCW can alleviate the disorder of glucose and fatty acid metabolism through upregulation of carnitine palmitoyltransferase1ɑ, citrate synthase, and acetyl-CoA and downregulation of glucose transport-4, phosphofructokinase, and pyruvate, which resulted in the reduced levels of free fatty acid and lactic acid and increased aerobic oxidation. This process may be mediated via the regulation of sirtuin 3 (SIRT3)-hypoxia-inducible factor 1α (HIF1α)-pyruvate dehydrogenase kinase (PDK)/pyruvate dehydrogenase (PDH) signaling pathway. Subsequently, the inhibition of SIRT3 expression by 3-TYP (a selective inhibitor of SIRT3) can reverse substantially the anti-RVH effect of BCW in HAHD, as indicated by hypertrophy marker and serum myocardial enzyme levels. Conclusions: BCW prevented SuHx-induced RVH in HAHD via the SIRT3-HIF1ɑ-PDK/PDH signaling pathway to alleviate the disturbance in fatty acid and glucose metabolism. A new approach is proposed that identifies three different zones of the Si-rich network structure (the cellular structure) in laser powder bed fused (LPBF) AlSi10Mg alloy, based on the variation in morphology, grain growth transition, and melt pool solidification conditions. The three identified zones are denoted in the present work as the liquid solidification zone (LSZ), the mushy solidification zone (MSZ), and the heat affected zone (HAZ). The LSZ is the result of liquid–solid transformation, showing small planar growth at the boundary and large cellular growth in the center, while the MSZ is related to a semisolid reaction, and the HAZ arises from a short-time aging process. The boundary between the LSZ and MSZ is identified by the change of grain growth direction and the Si-rich network advancing direction. The boundary between MSZ and HAZ is identified by the start of the breakdown of the Si-rich network. Cymbidium ensifolium L. is an economically important terrestrial orchid. Leaf color is a key horticultural trait with aesthetic and ornamental significance. However, the molecular basis of leaf color change in C. ensifolium remains to be fully understood. Here, different color leaves of C. ensifolium were collected and classified into two groups, namely, leaf buds that appear red in the juvenile stage (RL) and those that transform green in the mature stage (GL). Significantly higher anthocyanin content was observed in RL than GL. RNA-seq data showed a total of 3955 differentially expressed unigenes (DEUs) were screened, and 381 differentially accumulated metabolites (DAMs) were detected by metabolome analysis in RL vs GL. Further analysis revealed that the anthocyanin biosynthesis pathway (ko00942) is essential for leaf color change. Cyanidin-3-O-glucoside, petunidin-3-O-(6″-O-p-coumaroyl) rutinoside, and peonidin-3-O-glucoside were identified as key anthocyanins affecting leaf color change. Meanwhile, we identified key structural unigenes associated with leaf color change, including CHS, CHI, DFR, F3H, F3′H, and ANS. Correlation analysis revealed 77 DEUs associated with transcription factors to exhibit strong correlations with both DEUs and DAMs involved in anthocyanin biosynthesis. Chicken single‐chain fragment variable (IgY‐scFv) is a functional fragment and an emerging development in genetically engineered antibodies with a wide range of biomedical applications. However, scFvs have considerably shorter serum half‐life due to the absence of antibody Fc region compared with the full‐length antibody, and usually requires continuous intravenous administration for efficacy. A promising approach to overcome this limitation is to fuse scFv with immunoglobulin G (IgG) Fc region, for better recognition and mediation by the neonatal Fc receptor (FcRn) in the host. In this study, engineered mammalian ΔFc domains (CH2, CH3, and intact Fc region) were fused with anti‐canine parvovirus‐like particles avian IgY‐scFv to produce chimeric antibodies and expressed in the HEK293 cell expression system. The obtained scFv‐CH2, scFv‐CH3, and scFv‐Fc can bind with antigen specifically and dose‐dependently. Surface plasmon resonance investigation confirmed that scFv‐CH2, scFv‐CH3, and scFv‐Fc had different degrees of binding to FcRn, with scFv‐Fc showing the highest affinity. scFv‐Fc had a significantly longer half‐life in mice compared with the unfused scFv. The identified ΔFcs are promising for the development of engineered Fc‐based therapeutic antibodies and proteins with longer half‐lives. The avian IgY‐scFv‐mammalian IgG Fc region opens up new avenues for antibody engineering, and it is a novel strategy to enhance the rapid development and screening of functional antibodies in veterinary and human medicine. Developmental delay (DD), or intellectual disability (ID) is a very large group of early onset disorders that affects 1–2% of children worldwide, which have diverse genetic causes that should be identified. Genetic studies can elucidate the pathogenesis underlying DD/ID. In this study, whole-exome sequencing (WES) was performed on 225 Chinese DD/ID children (208 cases were sequenced as proband-parent trio) who were classified into seven phenotype subgroups. The phenotype and genomic data of patients with DD/ID were further retrospectively analyzed. There were 96/225 (42.67%; 95% confidence interval [CI] 36.15–49.18%) patients were found to have causative single nucleotide variants (SNVs) and small insertions/deletions (Indels) associated with DD/ID based on WES data. The diagnostic yields among the seven subgroups ranged from 31.25 to 71.43%. Three specific clinical features, hearing loss, visual loss, and facial dysmorphism, can significantly increase the diagnostic yield of WES in patients with DD/ID (P = 0.005, P = 0.005, and P = 0.039, respectively). Of note, hearing loss (odds ratio [OR] = 1.86%; 95% CI = 1.00-3.46, P = 0.046) or abnormal brainstem auditory evoked potential (BAEP) (OR = 1.91, 95% CI = 1.02–3.50, P = 0.042) was independently associated with causative genetic variants in DD/ID children. Our findings enrich the variation spectrums of SNVs/Indels associated with DD/ID, highlight the value genetic testing for DD/ID children, stress the importance of BAEP screen in DD/ID children, and help to facilitate early diagnose, clinical management and reproductive decisions, improve therapeutic response to medical treatment. 32. Cascaded bursting oscillations in a Mathieu–van der Pol–Duffing oscillator excited by low-frequency excitations.

- Abstract
This paper focuses on bursting behaviors of a Mathieu–van der Pol oscillator excited by low-frequency parametric and external excitations, where two novel bursting modes (namely cascaded bursting oscillations and non-cascaded bursting oscillations) are presented. In particular, the folds induced by the extreme values of the slow variable and the shape of the equilibrium branches are found on the quiescent state vibrations in two bursting patterns. We find that the spiking state vibrations can be induced by the subcritical Hopf bifurcations, supercritical Hopf bifurcations, and limit cycle fold bifurcations, and these three different types of the spiking state attractors can be found simultaneously with the variation of the positive integer ratio n. For the cascaded bursting, more and more large-amplitude limit cycles can be observed with the increase of n, leading to a greater number of the spiking states, while for the non-cascaded bursting, only one large-amplitude limit cycle can be found. In addition, we show that the spiking state attractors cannot be created by the small-amplitude limit cycles due to the Hopf bifurcation delay. [ABSTRACT FROM AUTHOR]

The aging process can affect the physical and chemical properties as well as adsorption capacity of biochar. This study focuses on the heavy metal cadmium (Cd) as the research object, and artificially ages biochar prepared from rice straw and corn straw through accelerated freeze–thaw cycles, alternating dry wet cycles, and ultraviolet light treatment, in order to evaluate the effects of different aging conditions on the physical and chemical properties of the two different types of biochar and on their adsorption capacities for Cd. After aging, the pH of rice and corn biochar decreased to varying degrees, respectively. The surface structure was ruptured, the average pore diameter was decreased, and the specific surface area was increased by 27.3%, 21.9%, and 9.8% (rice) and 95.4%, 27.7%, and 13.4% (corn). Ultraviolet light aging has the most significant impact on the elemental content of biochar, and the C content was decreased by 12.4% (rice) and 9.3% (corn). The O content was increased by 11.2% (rice) and 44.1% (corn), and the numbers of O/C, H/C, (O + N)/C, and oxygen-containing functional groups were increased. These results demonstrate that the aging process reduces the degree of aromatization of biochar, while enhancing its polarity and Cd adsorption capacity. Rice straw biochar (RSB) has a greater ability to adsorb Cd than corn straw biochar (CSB). In addition, ultraviolet light aging is particularly effective in increasing heavy metal adsorption. Ulcerative colitis (UC) is a chronic, recurrent inflammatory bowel disease. UC confronts with severe challenges including the unclear pathogenesis and lack of specific diagnostic markers, demanding for identifying predictive biomarkers for UC diagnosis and treatment. We perform immune infiltration and weighted gene co-expression network analysis on gene expression profiles of active UC, inactive UC, and normal controls to identify UC related immune cell and hub genes. Neutrophils, M1 macrophages, activated dendritic cells, and activated mast cells are significantly enriched in active UC. MMP-9, CHI3L1, CXCL9, CXCL10, CXCR2 and S100A9 are identified as hub genes in active UC. Specifically, S100A9 is significantly overexpressed in mice with colitis. The receiver operating characteristic curve demonstrates the excellent performance of S100A9 expression in diagnosing active UC. Inhibition of S100A9 expression reduces DSS-induced colonic inflammation. These identified biomarkers associated with activity in UC patients enlighten the new insights of UC diagnosis and treatment. Bronchopulmonary dysplasia (BPD) is a prevalent lung disease in neonates that is associated with numerous complications and high mortality. The promising approach to treat BPD is the use of mesenchymal stem cells (MSCs), However, the current treatment of MSCs presents safety concerns, including occlusion of blood vessels and tumorigenicity. In this study, relevant publications from the Web of Science Core Collection were downloaded in January 2023. The acquired data were analyzed and predicted for trends and hotspots in this field using CiteSpace software. Results revealed that in recent years, the focus of co-cited references has been primarily on the clinical studies of MSCs and the application of MSCs derivatives for treating BPD models. The keywords that have gained attention are extracellular vesicles and exosomes. The United States has emerged as the most influential co-authoring country in this field. Among the co-cited journals, the American Journal of Respiratory and Critical Care Medicine holds the highest influence. Thus, this study provides trends in publications, collaboration, research interests, and hotspots, and provides clues for novel ideas and strategies in to further MSCs treatments for BPD. Antimicrobial peptide-mimicking antibacterial polymers represent a practical strategy to conquer the ever-growing threat of antimicrobial resistance. Herein, we report the syntheses and antibacterial performance of degradable amphiphilic cationic polyesters containing pendent quaternary ammonium motifs and hydrophobic alkyl or fluoroalkyl groups. These polyesters were conveniently prepared from poly(3-methylene-1,5-dioxepan-2-one) via highly efficient one-pot successive thiol-Michael addition reactions. The antibacterial activity of these polyesters against S. aureus and E. coli and their hemolytic activity toward red blood cells were evaluated; some of them showed moderate antibacterial activity and selectivity against Gram-positive S. aureus. The membrane disruption mechanism of these cationic polyesters was briefly explored by monitoring the bacteria killing kinetics and SEM observations. Moreover, the effects of cationic/hydrophobic ratio and the incorporation of fluoroalkyl groups on the antibacterial activity and selectivity of the polyesters were demonstrated. Drinking groundwater contamination by pathogenic viruses represents a serious risk to worldwide public health, particularly for enteric viruses, which exhibit high prevalence and occurrence during outbreaks. Understanding how enteric viruses adsorb in groundwater is essential to protecting human health and ensuring the sustainable use of water resources. The adsorption properties of Coxsackievirus A16 (CA16), a common gastrointestinal virus that spreads through groundwater, were investigated in this work. A typical batch equilibrium approach was used to investigate CA16 adsorption and factors that influence it. In a laboratory recognized nationally as a biosafety level 2 facility, stringent research protocols were followed to guarantee compliance with experimental standards. The variables that were investigated included the size of the sediment particles, the starting concentration of the virus, temperature, pH level, and humic acid content. The findings showed that the CA16 virus was more strongly attracted to finer sediment particles and that its adsorption increased as the size of the sediment particle decreased. Furthermore, it was discovered that higher temperatures improved the CA16 virus's ability to bind to sediment particles. The pH of the aqueous environment has a significant effect on the effectiveness of virus adsorption; higher effectiveness was seen in acidic environments. Furthermore, it was found that the presence of humic acid decreased the ability of clay to adsorb CA16, suggesting that humic acid has a detrimental influence on clay's ability to adsorb viruses. The examination of kinetic models demonstrated that, in every scenario examined, the adsorption process of CA16 adhered to the pseudo-second-order kinetics model. Additionally, the Langmuir and Freundlich isotherm models were used to assess the equilibrium data that were collected in this investigation. The outcomes amply proved that the most accurate representation of the adsorption equilibrium was given by the Langmuir isotherm model. 43. Characteristics of patients with initial diagnosis of systemic lupus erythematosus in emergency department and their outcomes: a retrospective single-center study.

Study objective: This study aims to investigate the characteristics of patients with an initial diagnosis of systemic lupus erythematosus (SLE) in an emergency department (ED) and their outcomes. Methods: A total of 147 SLE patients (119 females and 28 males, mean age 26 ± 19 years) who visited the ED of the Peking University People's Hospital between January 2017 and June 2022 were enrolled in the study. Data on demographic information, clinical characteristics, comorbidities, therapy, and outcomes were collected. Results: Most patients visit ED because of symptoms related to SLE (74.8%, 110/147). The remaining 37 patients (25.2%) visited ED due to infection (43.2%, 16/37), gastrointestinal bleeding (10.8%, 4/37), coronary heart or cerebrovascular disease (18.9%, 7/37), macrophage activation syndrome or thrombotic microangiopathy (18.9%, 7/37), leukemia (5.4%, 2/37), and hepatic encephalopathy (2.7%, 1/37). Of the patients, 54.4% (80/147) were first diagnosed with SLE at the time of their ED visit. Thrombocytopenia events occurred significantly more frequently in this group of patients (OR 3.664, 95% CI 1.586–8.464, p = 0.002). Pulse steroid therapy was administered to 32.5% (26/80) of the patients with an initial diagnosis of SLE, and 26.3% (21/80) of these patients also received IVIG therapy during their ED visit. SLEDAI scores were significantly decreased after 6 months of therapy. The rate of mortality was 6.8% (10/147) in the 6-month follow-up period, and all the ten deaths happened in patients with disease-established SLE. The main causes of death were infections (two patients) and SLE flare (four patients). Conclusion: Understanding disease patterns can contribute to physicians providing accurate diagnosis and efficient care for SLE patients in ED. Key Points • Systemic lupus erythematosus, a complex autoimmune disorder, can have either a chronic or a relapsing and remitting disease course. The disease can involve acute events or severe comorbidities, and frequent visits to the emergency department (ED) are inevitable. • It is essential to better understand which comorbidities can lead to emergency department visits. Accurate clinical diagnosis and appropriate interventions from ED physicians can have a strong impact on the prognosis of the disease. • Hematologic compromise attributed to SLE flare is the most common reason for ED visits. 44. Molecular engineering of N,S-heteroarene-based small-molecule acceptors: effects of side chains, backbone extension and end groups on structure, optoelectronic properties and solar cell performance.

Studying the structure–property correlation of small-molecule acceptors (SMAs) is beneficial in optimizing their optoelectronic properties and enhancing photovoltaic performance. Here, we designed and synthesized six N,S-heteroarene (axa-indacenodithiophene, NIT)-based SMAs through molecular engineering (side chain, backbone extension and end group), which are defined as the pentacyclic central core series (NIT68, NIT810, and NIT810-F) and the backbone extension series (T-NIT68, NIT810-T, and NIT810-T-F), respectively. The results reveal that end group engineering (fluorination vs. non-fluorination) has a greater effect on the absorption properties and molecular energy levels as compared with side chain (2-hexyldecyl vs. 2-octyldodecyl) and backbone extension engineering. Theoretical calculation suggests that molecular engineering has a tiny impact on the backbone planarity of these NIT-based SMAs, but has a certain effect on the geometric configuration and electronic structure of molecules. Organic solar cells (OSCs) based on these SMAs were fabricated and device performance was optimized. OSCs based on the backbone extension series showed a higher open-circuit voltage (V
oc ) than those of the pentacyclic central core series, because of upshifted LUMOs. PBDB-T:NIT810-based OSCs exhibited the best power conversion efficiency (PCE) of 7.25% among these six SMAs. However, when PBDB-T was replaced by fluorinated PM6, the fluorinated NIT810-F and NIT810-T-F-based OSCs exhibited an improved PCE of up to 7.78% and 9.51%, respectively, due to the obviously increased fill factor (FF) and short-circuit current density (Jsc ). The improved FF and Jsc should be primarily attributed to the increased exciton dissociation and charge transport properties, suppressed bimolecular recombination, and optimized phase separation. Our work focusing on the effect of molecular engineering on molecular structure, optoelectronic and photovoltaic properties will certainly provide some significant guidance for the molecular design of high-performance SMAs. Observational studies have confirmed that 25‐hydroxyvitamin D (25(OH)D) is associated with pulmonary hypertension (PH), but the causal association between each other is unclear. Therefore, Mendelian randomization (MR) method was performed to validate the causal association between PH and serum 25(OH)D levels. The summary data for 25(OH)D and PH were from the National Human Genome Research Institute‐European Bioinformatics Institute. Catalog of human genome‐wide association studies and FinnGen biobank consortium. MR analysis was utilized to explore the potential causal association between PH and 25(OH)D. To evaluate this association, inverse variance weighting was considered as the primary method. Cochran's Q test, MR‐Egger intercept test, and "leave‐one‐out" sensitivity analyses were utilized to control the pleiotropy and heterogeneity in the study. Two‐sample MR analysis revealed an inverse causal relationship between 25(OH)D and PH (odds ratio: 0.376, 95% confidence interval: 0.162–0.876, p = 2.334 × 10−2). There was no significant heterogeneity and pleiotropy. The present study confirmed the inverse causal relationship between 25(OH)D and PH. This pathway may provide another treatment pathway in PH. We investigated possible protective effects of chlorogenic acid (CGA) against cyclophosphamide (CP) induced hepatic injury in mice. We measured aminotransferase alanine transaminase (ALT) and aspartate transaminase (AST) levels in the serum. We assayed catalase (CAT), superoxide dismutase (SOD), reduced glutathione (GSH), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) in hepatic tissue. We assessed expression of nuclear transcription factor 2 (Nrf2) and Kelch sample related protein-1 (keap1) proteins in hepatic tissues using immunohistochemistry. The relative mRNA expression levels of heme oxygenase-1 (HO-1), NADH quinone oxidoreductase 1 (NQO1), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were determined using quantitative real-time polymerase chain reaction (qRT-PCR). Hematoxylin & eosin staining was used to assess liver histopathology. We found that administration of CGA prior to induction of injury by CP decreased serum ALT, AST and MDA expressions in hepatic tissue, while CAT, SOD, GSH and GSH-Px concentrations were increased. We found that hepatocytes of animals administered CGA gradually returned to normal morphology. CGA increased the protein expression of Nrf2 in murine hepatic tissue. Administration of CGA up-regulated mRNA expression levels of HO-1, NQO1, TNF-α and IL-6 in hepatic tissue. CGA exhibited a marked protective effect on CP induced liver injury in mice. Purpose: As one of the most common gynecologic malignancies, endometrial cancer (EC) is driven by multiple genetic alterations that may be targeted for treatments. AT-rich interaction domain 1A (ARID1A) gene mutations were reported as early events in endometrial carcinogenesis. Methods: To explore the alterations of downstream molecular pathways caused by ARID1A mutations and the associated therapeutic implications, we edited ARID1A gene in human endometrial cancer cell line Ishikawa using the Clustered Regularly Interspaced Short Palindromic Repeats and CRISPR-Associated Proteins (CRISPR/Cas9) technology. We successfully constructed a stable Ishikawa cell line with a confirmed 10 bp deletion on the ARID1A gene, which resulted in a code-shift mutation and gene knockout. Results: Compared with unedited wild-type cells, ARID1A knockout (KO) led to reduced apoptosis, accelerated transformation from G0/G1 to S phase, and enhanced cell proliferation. ARID1A deficiency would reduce the protein levels of p21, caspase 7, and caspase 9 in Ishikawa endometrial cancer cells compared with the wild-type cells. In addition, ARID1A KO resulted in high levels of microsatellite instability (MSI-H). Moreover, transcriptomic analyses showed that ARID1A KO can lead to activated phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) signaling. Furthermore, experimental analyses demonstrated that ARID1A KO cells had reduced expression of genetic instability-associated markers mutL homologue 1 (MLH1) and progesterone receptor B (PR) and increased p-Akt expression. Conclusion: These findings support further exploration of ARID1A as a therapeutic target for EC and provide insight into developing more effective treatments in EC, such as the combinatory use of immune checkpoint inhibitors. Quinoa saponins are pentacyclic triterpene compounds composed of one triterpenoid glycoside and two different sugar chains. Previous studies have showed that natural quinoa saponins showed little or no antifungal activity, and there are few reports about their antifungal effects in recent decades. Fusarium wilt caused by Fusarium oxysporum f. sp. lycopersici (FOL) is the most serious for tomatoes in the field and under greenhouse conditions. The main objective of this study was to investigate the effectiveness of different concentrations and application modes of crude saponins from quinoa bran against the causal pathogen of tomato wilt under a greenhouse experiment. The results showed that the anti-FOL activity of quinoa saponins was weak in vitro, but significantly enhanced in vivo. Tomato seeds and seedlings treated with solution of quinoa saponins at 0.5 and 1.0 g/L significantly reduced the disease incidence (%) of tomato Fusarium wilt. The treatment types of saponin solution have influence on the preventive effects (%) of tomato seedlings against Fusarium wilt, among them, root soaking > foliar spray > seed soaking. The treatment of seed soaking with quinoa saponins inhibited germination of tomato seeds to some extent. However, the germination rate of tomato seeds after saponin soaking was comparable to the chemical pesticide (thiram carboxin); therefore, it could be used to control tomato wilt disease. This is due to the fact that the antifungal activity of quinoa saponins in vivo was much higher than that in vitro when the saponin concentration was between 0.5–1.0 g/L, indicating that the antifungal activity of quinoa saponins may be achieved mainly by inducing resistance. This investigation supports the potential use of quinoa saponins as a supplier of antifungal compounds, and could be the foundation for a future study examining the use of quinoa bran as a new resource against FOL. 49. Does the BRI contribute to poverty reduction in countries along the Belt and Road? A DID-based empirical test.

Most countries along the Belt and Road are developing countries that face pronounced challenges due to poverty. The Chinese government has committed to implementing the Belt and Road Initiative (BRI) to establish a 'Road to Poverty Reduction'. This study explores the impact of the BRI on poverty reduction in participating countries as well as the mechanisms underlying this impact. This research is of great significance to the continuing implementation of the BRI and global poverty governance. Based on a panel dataset extending from 2005 to 2019 and encompassing 151 countries, this article uses the difference-in-differences method to examine the Initiative's impact and conducts heterogeneity analysis to explore the mechanism underlying this impact in further detail. This approach results in four findings. First, joining the BRI can significantly reduce poverty rates in these countries. Second, the policy effect exhibits a time lag. In the long term, the poverty reduction effect becomes more significant. Third, the BRI can significantly reduce poverty in countries neighbouring China and in lower-middle-income countries. The Land Silk Road Economic Belt and the 21st-Century Maritime Silk Road distinctly reduce poverty. Fourth, the BRI contributes to poverty reduction in BRI countries by facilitating trade, enhancing financial and credit systems, increasing China's greenfield investment, and establishing digital infrastructure. Additionally, the BRI promotes poverty reduction through increased bilateral visits and technical cooperation, whereas knowledge sharing remains an area in need of further development. Through theoretical analysis and empirical tests, this article evaluates the policy effect of the BRI on poverty alleviation, tests the underlying mechanism, and responds to some concerns expressed by the international community. The findings of this article provide a feasible direction for improving the ability of the BRI to reduce poverty more effectively in the future and thus achieve the United Nations' sustainable development goals (SDGs).
