The research in this field primarily centers on investigating the alterations and associations of GM in MS or EAE, the molecular immunological mechanisms, and the potential of GM-based interventions to provide beneficial effects in MS or EAE. The Keywords co-occurrence network reveals five primary research directions in this field. The most frequently occurring keywords are inflammation, probiotics, diet, dysbiosis, and tryptophan. In recent years, neurodegeneration and neuropsychiatric disorders have been prominent, indicating that the investigation of the mechanisms and practical applications of GM in MS has emerged as a current research focus. Moreover, GM research is progressively extending into the realm of neurodegenerative and psychiatric diseases, potentially becoming future research hotspots. Background: Our study aims to enhance the understanding of lymph node venous networks (LNVNs) by summarising their anatomical, sonographic features, and reflux patterns. Method: We examined 241 legs from 141 patients with primary chronic venous disease (CVD) using duplex ultrasound. Results: The findings indicated variations in the shape, size, vascularity, and echogenicity of LNVN. The superficial inguinal lymph node with reflux appeared slightly larger, exhibiting higher velocities in the hilar artery. Regarding connections, venous flow within LNVN was predominantly drained through the saphenofemoral junction (SFJ), anterior accessory great saphenous vein (AAGSV), and great saphenous vein (GSV). A significant number of LNVNs were observed to be associated with anterolateral thigh tributaries. The study also identified valve cusps within LNVN. Conclusion: This study revealed a 12% prevalence of primary LNVN. Accurate prediction of allograft survival after kidney transplantation allows early Identification of at-risk recipients for adverse outcomes and initiation of preventive interventions to optimize post-transplant care. Many prediction algorithms do not model cohort heterogeneity and may lead to inaccurate assessment of longer-term graft outcomes among minority groups. Using data from a national Australian kidney transplant cohort (2008-2017) as the derivation set, we developed P-Cube, a multi-step precision prediction pathway model for predicting overall graft survival in three ethnic subgroups: European Australians, Asian Australians and Aboriginal and Torres Strait Islander Peoples. The concordance index for the European Australians, Asian Australians, and Aboriginal and Torres Strait Islander Peoples subpopulations were 0.99 (0.98-0.99), 0.93 (0.92-0.94) and 0.92 (0.91-0.93), respectively. Similar findings were observed when validating P-cube using an external dataset [Scientific Registry of Transplant Recipient Registry (2006-2020)]. Six sub-categories of recipients with distinct risk factor profiles were identified. Some factors such as blood group compatibility were considered important across the entire transplant population. Other factors such as human leukocyte antigen (HLA)-DR mismatches were unique to older recipients. Introduction: Early and effective application of antimicrobial medication has been evidenced to improve outcomes of patients with bloodstream infection (BSI). However, conventional microbiological tests (CMTs) have a number of limitations that hamper a rapid diagnosis. Methods: We retrospectively collected 162 cases suspected BSI from intensive care unit with blood metagenomics next-generation sequencing (mNGS) results, to comparatively evaluate the diagnostic performance and the clinical impact on antibiotics usage of mNGS. Results and discussion: Results showed that compared with blood culture, mNGS detected a greater number of pathogens, especially for Aspergillus spp, and yielded a significantly higher positive rate. With the final clinical diagnosis as the standard, the sensitivity of mNGS (excluding viruses) was 58.06%, significantly higher than that of blood culture (34.68%, P<0.001). Combing blood mNGS and culture results, the sensitivity improved to 72.58%. Forty-six patients had infected by mixed pathogens, among which Klebsiella pneumoniae and Acinetobacter baumannii contributed most. Compared to monomicrobial, cases with polymicrobial BSI exhibited dramatically higher level of SOFA, AST, hospitalized mortality and 90-day mortality (P<0.05). A total of 101 patients underwent antibiotics adjustment, among which 85 were adjusted according to microbiological results, including 45 cases based on the mNGS results (40 cases escalation and 5 cases de-escalation) and 32 cases on blood culture. Collectively, for patients suspected BSI in critical condition, mNGS results can provide valuable diagnostic information and contribute to the optimizing of antibiotic treatment. CONSTANS (CO) is a central regulator of floral initiation in response to photoperiod. In this study, we show that the GSK3 kinase BIN2 physically interacts with CO and the gain-of-function mutant bin2-1 displays late flowering phenotype through down-regulation of FT transcription. Genetic analyses show that BIN2 genetically acts upstream of CO in regulating flowering time. Further, we illustrate that BIN2 phosphorylates the Thr280 residue of CO. Importantly, the BIN2 phosphorylation of Thr280 residue restricts the function of CO in promoting flowering through affecting its DNA-binding activity. Moreover, we reveal that the N-terminal part of CO harboring the B-Box domain mediates the interaction of both CO-CO and BIN2-CO. We find that BIN2 inhibits the formation of CO dimer/ oligomer. Taken together, this study reveals that BIN2 regulates flowering time through phosphorylating the Thr280 of CO and inhibiting the CO-CO interaction in Arabidopsis. [ABSTRACT FROM AUTHOR]

Understanding and enhancing switchgrass (Panicum virgatum L.) photosynthesis will help to improve yield and quality for bio-industrial applications on cellulosic biofuel production. In the present study, leaf anatomical traits and physiological characteristics related to photosynthetic capacity of both lowland and upland switchgrass were recorded from four varieties across the vegetative, elongation and reproductive growth stages. Compared with the upland varieties, the lowland switchgrass showed 37-59, 22-64 and 27-73% higher performance on height, stem and leaf over all three growth stages. Leaf anatomical traits indicated that the leaves of lowland varieties provided more space for carbon assimilation and transportation caused by enhanced cell proliferation with more bundles sheath cells and larger contact areas between the bundle sheath and mesophyll cells (CAMB), which lead to the 32-72%higher photosynthetic capacity found in the lowland varieties during vegetative and elongation growth. However, photosynthetic capacity became 22-51% higher in the upland varieties during the reproductive stage, which is attributed to more photosynthetic pigment. In conclusion, lowland varieties gain a photosynthetic advantage with enhanced bundle sheath cell proliferation, while the upland varieties preserved more photosynthetic pigments. Switchgrass (Panicum virgatum) has been developed into a model lignocellulosic bioenergy crop. Downregulation of caffeic acid O-methyltransferase (COMT), a key enzyme in lignin biosynthesis, has been shown to alter lignification and increase biofuel yield in switchgrass. Methylenetetrahydrofolate reductase (MTHFR) mediates C1 metabolism and provides methyl units consumed by COMT. It was predicted that co-silencing of MTHFR and COMT would impact lignification even more than either of the single genes. However, our results showed that strong downregulation of MTHFR in a COMT-deficient background led to altered plant growth and development, but no significant change in lignin content or composition was found when compared with COMT plants. Another unexpected finding was that the double MTHFR/COMT downregulated plants showed a novel lesion-mimic leaf phenotype. Molecular analyses revealed that the lesion-mimic phenotype was caused by the synergistic effect of MTHFR and COMT genes, with MTHFR playing a predominant role. Microarray analysis showed significant induction of genes related to oxidative and defense responses. The results demonstrated the lack of additive effects of MTHFR and COMT on lignification. Furthermore, this research revealed an unexpected role of the two genes in the modulation of lesion-mimic cell death as well as their synergistic effects on agronomic performance. Switchgrass (Panicum virgatum L.) has been increasingly recognized as one of the most valuable perennial bioenergy crop. To improve its biomass production, especially under salt stress, we isolated a putative vacuolar Na+ (K+)/H+ antiporter gene from switchgrass and designated as PvNHX1. Subcellular localization revealed that this protein was localized mainly on the vacuole membrane. The PvNHX1 was found to be expressed throughout the entire growth period of switchgrass, exhibited preferentially expressed in the leaf tissue, and highly induced by salt stress. Transgenic switchgrass overexpressing PvNHX1 showed obvious advantages with respect to plant height and leaf development compared to the wild-type (WT) and transgenic control (EV, expressing the empty vector only) plants, suggesting PvNHX1 may serve as a promoter in switchgrass growth and development. Moreover, transgenic switchgrass were more tolerant than control plants with better growth-related phenotypes (higher shoot height, larger stem diameter, longer leaf length, and width) and physiological capacities (increased proline accumulation, reduced malondialdehyde production, preserved cell membrane integrity, etc.) under high salinity stress. Furthermore, the genes related to cell growth, flowering, and potassium transporters in transgenic switchgrass exhibited a different expression profiles when compared to the control plants, indicating a pivotal function of PvNHX1 in cell expansion and K+ homeostasis. Taken together, PvNHX1 is essential for normal plant growth and development, and play an important role in the response to salt stress by improving K+ accumulation. Melatonin serves pleiotropic functions in prompting plant growth and resistance to various stresses. The accurate biosynthetic pathway of melatonin remains elusive in plant species, while the N-acetyltransferase and O-methyltransferase were considered to be the last two key enzymes during its biosynthesis. To investigate the biosynthesis and metabolic pathway of melatonin in plants, the RNA-seq profile of overexpression of the ovine HIOMT was analyzed and compared with the previous transcriptome of transgenic oAANAT gene in switchgrass, a model plant for cellulosic ethanol production. A total of 946, 405, and 807 differentially expressed unigenes were observed in AANAT vs. control, HIOMT vs. control, and AANAT vs. HIOMT, respectively. Two hundred and seventy-five upregulated and 130 downregulated unigenes were detected in transgenic oHIOMT line comparing with control, including the significantly upregulated (F-box/kelch-repeat protein, zinc finger BED domain-containing protein-3) genes, which were potentially correlated with enhanced phenotypes of shoot, stem and root growth in transgenic oHIOMT switchgrass. Several stress resistant related genes (SPX domain-containing membrane protein, copper transporter 1, late blight resistance protein homolog R1A-6 OS etc.) were specifically and significantly upregulated in transgenic oHIOMT only, while metabolism-related genes (phenylalanine-4-hydroxylase, tyrosine decarboxylase 1, protein disulfide-isomerase and galactinol synthase 2 etc.) were significantly upregulated in transgenic oAANAT only. Melatonin serves important functions in the promotion of growth and anti-stress regulation by efficient radical scavenging and regulation of antioxidant enzyme activity in various plants. To investigate its regulatory roles and metabolism pathways, the transcriptomic profile of overexpressing the ovine arylalkylamine N-acetyltransferase (oAANAT) gene, encoding the penultimate enzyme in melatonin biosynthesis, was compared with empty vector control using RNA-seq in switchgrass, a model plant of cellulosic ethanol conversion. The 85.22 million high quality reads that were assembled into 135,684 unigenes were generated by Illumina sequencing for transgenic oAANAT switchgrass with an average sequence length of 716 bp. A total of 946 differentially expression genes in transgenic line comparing to control switchgrass, including 737 up-regulated and 209 down-regulated genes, were mainly enriched with two main functional patterns of melatonin identifying by gene ontology analysis: the growth regulator and stress tolerance. Furthermore, KEGG maps indicated that the biosynthetic pathways of secondary metabolite (phenylpropanoids, flavonoids, steroids, stilbenoid, diarylheptanoid, and gingerol) and signaling pathways (MAPK signaling pathway, estrogen signaling pathway) were involved in melatonin metabolism. This study substantially expands the transcriptome information for switchgrass and provides valuable clues for identifying candidate genes involved in melatonin biosynthesis and elucidating the mechanism of melatonin metabolism. The objective of this study was to determine the effectiveness of microwave pretreatments on methane production from two switchgrass tissues (leaf vs. stem). The methane production from the leaf fraction was significantly affected by the microwave final temperature, while production from the stem fraction was affected by the combination of the microwave final temperature and heating rate. Thus, the highest methane yield from the leaf (134.81 mL CH4/g of volatile solids (VS)) was obtained at 100 °C, while the highest yield from the stem (99.35 mL CH4/g VS) was obtained at 150 °C, with a heating rate of 10 °C/min. Although methane production from the leaf fraction was merely enhanced by 9.1% after microwave pretreatments, the time required to reach 80% of ultimate methane production was reduced by 12 days. A physical model of the electric field induced by charged droplets taking account of the effect of space charged droplet emitted from the tip of cone-jet to the external electric field is proposed. Combining this model with the fluid flow equations and charge conservation equation, the evolution of the cone-jet is simulated. The diameter of droplets emitted from the cone-jet tip and current on cone-jet are predicted at various applied voltages and flow rates. The calculated droplet diameter agrees well with experimental measurement. For low conductivity liquid, the droplet diameter decreases with the increment of applied voltage, but decreases with the reduction of flow rate. The simulation result also indicates that the current on the cone-jet increases linearly with the applied voltage. The electric field induced by charged droplets results in the decrease of the cone angle and the presence of space charged droplets has a non-negligible effect on the operation parameters. Switchgrass is a perennial C4 plant with great potential as a bioenergy source and, thus, a high demand for establishment from seed. This research investigated the effects of ultrasound treatment on germination and seedling growth in switchgrass. Using an orthogonal matrix design, conditions for the ultrasound pretreatment in switchgrass seed, including sonication time (factor A), sonication temperature (factor B) and ultrasound output power (factor C), were optimized for germinating and stimulating seedling growth (indicated as plumular and radicular lengths) through modeling analysis. The results indicate that sonication temperature (B) was the most effective factor for germination, whereas output power (C) had the largest effect on seedling growth when ultrasound treatment was used. Combined with the analyses of range, variance and models, the final optimal ultrasonic treatment conditions were sonication for 22.5 min at 39.7°C and at an output power of 348 W, which provided the greatest germination percentage and best seedling growth. For this study, the orthogonal matrix design was an efficient method for optimizing the conditions of ultrasound seed treatment on switchgrass. The electrical conductivity of seed leachates in three experimental groups (control, soaked in water only, and ultrasound treatment) was determined to investigate the effects of ultrasound on seeds and eliminate the effect of water in the ultrasound treatments. The results showed that the electrical conductivity of seed leachates during either ultrasound treatment or water bath treatment was significantly higher than that of the control, and that the ultrasound treatment had positive effects on switchgrass seeds. SUMMARY A novel dynamic mixing length (DML) subgrid-scale model for large eddy simulations is proposed in this work to improve the cutoff length of the Smagorinsky model. The characteristic mixing length (or the characteristic wave number) is dynamically estimated for the subgrid-scale fluctuation of turbulence by the cutoff wave-number, kc, and the dissipation wave-number, kd. The dissipation wave number is derived from the kinetic energy spectrum equation and the dissipation spectrum equation. To prove the promise of the DML model, this model is used to simulate the lid-driven cubical cavity with max-velocity-based Reynolds numbers 8850 and 12,000, the channel flows with friction-velocity-based Reynolds numbers 180, 395, 590, and 950, and the turbulent flow past a square cylinder at the higher Reynolds number 21,400, respectively, compared with the Smagorinsky model and Germano et al.'s dynamic Smagorinsky model. Different numerical experiments with different Reynolds numbers show that the DML model can be used in simulations of flows with a wide range of Reynolds numbers without the occurrence of singular values. The DML model can alleviate the dissipation of the Smagorinsky model without the loss of its robustness. The DML model shows some advantages over Germano et al.'s dynamic Smagorinsky model in its high stability and simplicity of calculation because the coefficient of the DML model always stays positive. The characteristic mixing length in the DML model reflects the subgrid-scale fluctuation of turbulence in nature and thus the characteristic mixing length has a spatial and temporal distribution in turbulent flow. Switchgrass (Panicum virgatum L.) is a perennial grass native to the North American tallgrass prairie and savanna habitats and is broadly adapted to the central and eastern United States. Upland and lowland ecotypes represent the two major taxa within switchgrass, which have distinct but overlapping distributions. The purpose of this study was to survey a broad array of putative upland and lowland accessions for the possible presence of natural hybrids or hybrid derivatives and evidence of historic gene flow between the two ecotypes. All plants were classified as upland, lowland, or intermediate based on visual assessment of phenotype, using large nurseries of known upland or lowland plants as controls. A total of 480 plants were surveyed for 19 simple sequence repeat (SSR) markers and sequenced using five chloroplast DNA (cpDNA) segments. Genetic structure analysis revealed 21 individuals with strong evidence for intertaxa hybrid origin and another 25 individuals with moderate evidence for intertaxa hybrid origin. All but two of these individuals originated from remnant populations of the central or eastern Gulf Coast or along the Atlantic Seaboard, a region that is populated with significant quantities of both upland and lowland ecotypes. We propose the central and eastern Gulf Coast glacial refuge as the primary center of origin and diversity for switchgrass, with the western Gulf Coast as the secondary center of origin and diversity. The article discusses a study regarding all-trans retinoic acid (ATRA), which is an active metabolite of vitamin A that is used in the treatment of myelodysplastic syndrome and acute promyelocytic leukaemia. It says that the synthesised ATRA derivatives were characterised in terms of mass spectrometry, optical rotation, and nuclear magnetic resonance (NMR). Results show that the two derivatives of ATRA and all-trans retinoyl glutamate (RAE) have exhibited improved aqueous solubility.
