Compared with 454 sequencing technology, short-read sequencing (e.g., Illumina) technology generates sequences of high accuracy, but limited length (<500 bp). Such a limitation can prove that studying a target gene using a large amplicon (>500 bp) is challenging. The ammonia monooxygenase subunit A (amoA) gene of ammonia-oxidizing archaea (AOA), which plays a crucial part in the nitrification process, is such a gene. By providing a full overview of the community of a functional microbial guild, 16S ribosomal ribonucleic acid (rRNA) gene sequencing could overcome this problem. However, it remains unclear how 16S rRNA primer selection influences the quantification of relative abundance and the identification of community composition of nitrifiers, especially AOA. In the present study, a comparison was made between the performance of primer pairs 338F-806R, 515F-806R, and 515F-907R to a shotgun metagenome approach. The structure of nitrifier communities subjected to different long-term organic matter amendment and water management protocols was assessed. Overall, we observed higher Chao1 richness diversity of soil total bacteria by using 515F-806R compared to 338F-806R and 515F-907R, while higher Pielou's evenness diversity was observed by using 515F-806R and 515F-907R compared to 338F-806R. The studied primer pairs revealed different performances on the relative abundance of Thaumarchaeota, AOB, and NOB. The Thaumarchaeota 16S rRNA sequence was rarely detected using 338F-806R, while the relative abundances of Thaumarchaeota detected using 515F-806R were higher than those detected by using 515F-907R. AOB showed higher proportions in the 338F-806R and 515F-907R data, than in 515F-806R data. Different primers pairs showed significant change in relative proportion of NOB. Nonetheless, we found consistent patterns of the phylotype distribution of nitrifiers in different treatments. Nitrosopumilales (NP) and Nitrososphaerales (NS) clades were the dominant members of the AOA community in soils subject to controlled irrigation, whereas Ca. Nitrosotaleales (NT) and NS clades dominated the AOA community in soils subject to flooding irrigation. Nitrospira lineage II was the dominant NOB phylotype in all samples. Overall, ideal 16S rRNA primer pairs were identified for the analysis of nitrifier communities. An artificial semisynthetic material can be derived from artemisinin (ART) called dihydroartemisinin (DHA). Although DHA has enhanced antigiardial potential, its clinical application is limited because of its poor selectivity and low solubility. The drug's absorption has a direct impact on the cell, and mechanism research is limited to its destruction of the cytoskeleton. In this study, we used the zeolitic imidazolate framework-8 and loaded it with DHA (DHA@Zif-8) to improve its antigiardial potential. DHA@Zif-8 can enhance cellular uptake, increase antigiardial proliferation and encystation, and expand the endoplasmic reticulum compared with the DHA-treated group. We used RNA sequencing (RNA-seq) to investigate the antigiardial mechanism. We found that 126 genes were downregulated and 123 genes were upregulated. According to the KEGG and GO pathway analysis, the metabolic functions in G. lamblia are affected by DHA@Zif-8 NPs. We used real-time quantitative reverse transcription polymerase chain reaction to verify our results using the RNA-seq data. DHA@ Zif-8 NPs significantly enhanced the eradication of the parasite from the stool in vivo. In addition, the intestinal mucosal injury caused by G. lamblia trophozoites markedly improved in the intestine. In this paper, the dynamics of an acoustic bubble with a constant charge in compressible liquid are investigated numerically, which is based on the Gilmore-NASG model to estimate the radial oscillations. The cavitation effects are enhanced due to the presence of the charge on the bubble surface. The obtained results from the present model are compared with that calculated by the previous model within a wide range of parameters (e.g., charge, acoustic pressure amplitude, ultrasound frequency, and liquid temperature). The similar influences of these parameters on bubble collapse intensity can be observed from both models. Since the present model fully considers the compressibility of gas and liquid, it can be applied to a wider parameter range and leads to the larger predicted values. The research in this paper can provide important insights about the effects of charge on bubble dynamics and the acoustic cavitation applications (e.g., sonochemistry, water treatment, and food industry). Pollution flashover is one of themajor insulation accidents that can occur inmodern power systems. If the local arc is not sufficiently elongated by the creepage distance, the flashover performance of the insulator will decrease. The insulator structure and equivalent salt deposition density (ESDD) are the main factors affecting the arc path and length. To investigate the forms of arc paths under different insulator structures and their influence on flashover characteristics, a platformfor recording arcpaths is established. This studypresentsamethod to identify thepattern and distribution of insulator arc paths based on the effect of initial arc on electric field distribution. The results show that there are two main types of local arc paths between umbrellas--the cling-surface arc and the air-jump arc. Moreover, the air-jump arc includes two types--the bridging arc and the gap arc. With the increase in umbrella spacing and ESDD, the probability of arc propagation along the surface increases. The use of the creepage distance can be improved by changing the creepage coefficient to increase the distance between umbrellas. This method has over 90% accuracy. Salicylic acid (SA) and ethylene (ET) are crucial fruit senescence hormones. SA inhibited ET biosynthesis. However, the mechanism of SA delaying fruit senescence is less known. ETHYLENE INSENSITIVE 3 (EIN3), a key positive switch in ET perception, functions as a transcriptional activator and binds to the primary ET response element that is present in the promoter of the ETHYLENE RESPONSE FACTOR1 gene. In this study, a gene encoding putative EIN3 protein was cloned from sand pear and designated as PpEIN3a. The deduced PpEIN3a contains a conserved EIN3 domain. The evolutionary analysis results indicated that PpEIN3a belonged to the EIN3 superfamily. Real-time quantitative PCR analysis revealed that the accumulation of PpEIN3a transcripts were detected in all tissues of this pear. Moreover, PpEIN3a expression was regulated during fruit development. Interestingly, the expression of PpEIN3a was downregulated by SA but upregulated by ET, auxin, and glucose. Additionally, the contents of free and conjugated SA were higher than those of the control after SA treatment. While the content of ET and auxin (indole-3-acetic acid, IAA) dramatically decreased after SA treatment compared with control during fruit senescence. The content of glucose increased when fruit were treated by SA for 12 h and then there were no differences between SA treatment and control fruit during the shelf life. SA also delayed the decrease in sand pear (Pyrus pyrifolia Nakai. 'Whangkeumbae') fruit firmness. The soluble solid content remained relatively stable between the SA treated and control fruits. This study showed that SA plays an antagonistic role toward ET, auxin, and glucose in regulating the expression of PpEIN3a to delay fruit senescence. [ABSTRACT FROM AUTHOR]

Objective: To explore the long-term efficacy and safety of dexamethasone treatment via tympanic antrum catheterization (TAC) in intractable Meniere's disease (MD). Methods: In this retrospective analysis, 60 unilateral intractable MD patients treated with TAC in our hospital from January 2020 to August 2020 were followed for 2 years. Fifty patients who underwent endolymphatic sac decompression (ESD) and 50 patients who accepted intratympanic steroids (ITS) were established as the control groups. Vertigo control, hearing level, tinnitus, aural fullness and functional level were assessed during the study. Results: The effective vertigo control rate of intractable MD patients with TAC treatment was 76.7% (46/60) after 2 years follow-up, with a complete control rate of 58.3% (35/60) and a substantial control rate of 18.3% (11/60). The vertigo control rate of TAC was comparable to that of ESD (X² = 0.313, p > 0.05), and significantly higher than that of ITS (X² = 4.380, p < 0.05). The hearing loss rate of these patients was 10.8% (4/37), which was not significantly different from the control groups (X² = 2.452, p > 0.05). The tinnitus improvement rate of patients with TAC was 56.7% (34/60), which was significantly higher than that of patients with ESD (X² =11.962, p < 0.001) and ITS (X² =15.278, p < 0.001). The aural fullness improvement rate in the TAC group was 56.7% (34/60), which was significantly higher than that in the ESD (X² = 11.962, p < 0.001) and ITS groups (X² = 5.635, p < 0.05). The functional level improvement rate in the TAC group was 71.7% (43/60), which was much higher than that in the ITS group (X² = 17.256, p < 0.001), but there was no significant difference between TAC and ESD (X² = 0.410, p > 0.05). No patients had complications or adverse reactions following TAC treatment. Conclusion: Dexamethasone treatment via TAC can effectively control vertigo attacks and improve related symptoms of intractable MD patients, providing valuable new insights into the treatment of MD. Background: Migraine plays an important role in some subgroups of children with recurrent vertigo. Moreover, themigraine component varies fromdefinite to possibly absent as defined in this spectrum of three disorders--vestibular migraine of childhood (VMC), probable VMC (pVMC), and recurrent vertigo of childhood (RVC). However, studies on the sensory organization of balance control in these three disorders are rare. Objective: To explore the balance control of children with RVC, VMC, and pVMC, when the three sensory systems are challenged. Method: A retrospective analysis was performed on 125 children with VMC (18 female and 15male; aged 11.64 ± 2.74), pVMC (10 female and eightmale; aged 11.78 ± 2.51), and RVC (32 female and 42male; aged 11.10 ± 2.60). All children in each subtype were divided into groups of children aged ≤12 years old and 13-17 years old. Vestibular examination screening and assessment for postural control using the six conditions of the sensory organization test (SOT) were performed. The three primary outcomemeasures were: equilibriumscore (ES), strategy score (SS), and sensory analysis score of the SOT. Results: Equilibrium score under six different conditions and composite score increased with age (all P-values < 0.05). The somatosensory and visual scores also improved with growing (P-values < 0.05). However, vestibular scores did not increase significantly with age as the other senses did (P > 0.05). In the children ≤12 year-old group, children with VMC had a significantly higher visual preference score than those with pVMC and RVC (P < 0.05). There was an effect of age on the horizontal HIT. Ocular vestibular evoked myogenic potential (oVEMP), cervical vestibular evoked myogenic potential (cVEMP), and unilateral weakness (UW) values showed no significant difference among three diseases. Conclusion: Compared with patients at the age of 13-17 years old and with RVC and pVMC (both ≤12 years old), children with VMC had a higher degree of reliance on visual signals to maintain their balance and a poorer central integration of peripheral information before reaching 12 years of age. In addition, vision may predominate by weakening vestibular function based on visuo-vestibular interactions. The voltage-gated sodium channel Nav1.5 plays an essential role in the generation and propagation of action potential in cardiomyocytes. Mutations in Nav1.5 have been associated with LQT syndrome, Brugada syndrome, and sudden arrhythmia death syndrome. Genetic studies showed that Nav1.5 mutations vary across race-ethnic groups. Here we investigated an Asian-specific mutation Nav1.5-P1090L associated with LQT syndrome. We found that Nav1.5-P1090L mutation perturbed the sodium channel function. It altered the gating process of the channel and exhibited an enhanced window current. Treatment with mexiletine reversed the depolarization shift of the steady-state inactivation produced by P1090L. Mexiletine also modified the recovery from steady-state inactivation and the development of inactivation of P1090L. It rescued the dysfunctional inactivation of P1090L and reduced the P1090L channel's availability. [ABSTRACT FROM AUTHOR]

Straw strip mulching (SM) is a new mulching technology. From 2012 to 2018, SM's effects on soil moisture and temperature and production performances were compared with other mulching practices, using three treatments: full-cover plastic mulch (PM), no mulch with wheat sown in rows as the control (CK), and SM with 50% to 59% of the field area mulched. Compared with CK, on average over six growing seasons, SM and PM increased grain yield by 27.0% and 21.7%, straw yield by 21.6% and 22.6%, kernels ha-1 by 26.6% and 19.0%, net income by 29.8% and -25.0%, soil temperature at 5 cm by -1.5 °C and 0.2 °C from overwintering to maturity, and soil water storage at 0-200 cm by 25 and 22 mm, respectively. The increase in soil moisture in SM and PM was greater in the early period (overwintering to jointing) than in the later period (booting to maturity) and at 0 to 120 cm than at 120-200 cm in the early period. Although the mean evapotranspiration of whole growth period across six seasons was similar among treatments, SM and PM increased water consumption during the key formation period of yield components after overwintering by 16 and 32 mm, respectively, while reducing it before overwintering. Compared with CK, SM and PM had the effects of warming during overwintering and cooling after jointing. By increasing water consumption after overwintering and ratio of transpiration to evapotranspiration and providing favorable soil temperature for multiple growth stages and more sufficient soil moisture, SM and PM promoted vegetative growth and increased kernels ha-1, the main mechanisms by which SM and PM increased grain yield relative to CK. Increasing evidence suggests that microRNAs (miRNAs) play critical roles in bladder tumorigenesis and development by combining with the 3’ untranslated regions (3’-UTRs) of the corresponding mRNAs to negatively regulate gene expression. The role of miR-182-5p in bladder cancer (BC) remains unclear. Therefore, this study aimed to clarify the functional role of miR-182-5p in BC. We predicted candidate mRNAs for miR-182-5p via three databases (TarBase, ENCORI, and miRDB). Dual-luciferase reporter assays and target prediction confirmed FOXF2 as a potential target of miR-182-5p. Quantitative RT-PCR (qRT-PCR) showed that endogenous miR-182-5p expression was significantly upregulated in BC cell lines and clinical samples of BC patients. IHC, western blotting, and qRT-PCR assays indicated that FOXF2 expression was concurrently downregulated in BC tissues and BC cell lines. Gain- and loss-of-function studies showed that the overexpression of miR-182-5p enhanced the proliferation and migration of BC cells, while the downregulation of miR-182-5p showed the opposite results. The effects induced by miR-182-5p were attenuated with the restoration of FOXF2 expression. In BC cells, the upregulation of miR-182-5p not only decreased FOXF2 expression but also markedly increased Sonic hedgehog (SHH) pathway levels. These findings suggested that FOXF2 directly binds to miR-182-5p and that miR-182-5p acts as a tumor promoter in BC genesis and metastasis by targeting FOXF2. The mechanical properties and magnetocaloric effect (MCE) of bonded La(Fe, Si)13 hydrides have been studied in detail. The mechanical strength increases with increasing the grade of epoxy resin from E-20 to E-51. This occurs because more pores and boundaries are filled with high grade resin since high epoxide content increases the degree of crosslinking and reduces the viscosity and shrinkage of resin. The compressive strength reaches 162 MPa for the bonded LaFe11.7Si1.3C0.2H1.8 with 3 wt.% E-51, which is 35% higher than that of bulk LaFe11.7Si1.3C0.2 compound (120 MPa). The mass ΔSM values remain almost same in bonded hydrides and are in a good agreement with the theoretical value. The maximum volumetric ΔSM values are 61.8, 58.0, and 54.7 mJ/cm³ K for bonded hydrides with epoxy resins E-20, E-44, and E-51, respectively, much higher than those of some magnetocaloric materials in same temperature range. The improved mechanical properties and large MCE indicate that bonded LaFe11.7Si1.3C0.2H1.8 is a promising material for room temperature magnetic refrigeration. [ABSTRACT FROM AUTHOR]

This paper discusses the implicit function of refusal expressions that has been used by Chinese native speakers when responding to people's offerings. By analyzing three conversations regarding how Chinese people have accepted people's offerings during different time periods (1960's, 1980's, and 2000's), the author argues that the verbal refusal in reacting to people's offerings (especially gifts) does not literally mean "No, I don't want it." Instead, it is a way to show humility, politeness, and respect to the gift giver, and it functions as an implicit form of acceptance. By referring to three excerpts chosen from The Book of Rites (Liji 禮記), the author demonstrates the Chinese cultural values and cultural themes, focusing on what Chinese people say and do to show respect and politeness when dealing with gifts or other friendliness related offerings. The paper finds two pedagogical implications:1) it is the instructor's responsibility to highlight the verbal and non-verbal behaviors that both native speakers and language learners do not ordinarily notice in language class, and 2) the repetitive contextualized performances in different levels of learning are necessary and play crucial roles in teaching and learning. A selective effect of motor expertise on mental rotation is revealed by a high correlation between the performance of sports experts and ability on a mental rotation task. Evidence is shown by studies involving a spatial factor, such as image interference or a movement constraint. Alternatively, the time constraint, as another performance factor, is considered critical in the effect of sport expertise on mental rotation. Three experiments were conducted to examine the role of time constraints in egocentric mental rotation and the stage performance of athletes (divers) and nonathletes. In Experiment 1, an egocentric mental rotation task in an untimed condition was conducted, and reaction times (RTs), error rates (ERs), RTs at 0 °, and mental rotation speed were assessed. The results indicated that divers outperformed nonathletes in terms of RT, as well as perceptual and decision stages and rotation stages. Experiments 2 and 3 added a relative time constraint (subtracting 1/2 SD of all the subjects' RT from the M of each group's RT) and an absolute time constraint (subtracting 1/2 SD of all the subjects' RT from the M of all the subjects' RT) to the task, respectively. Superior RT and lower ER were observed for the divers in the time constraint condition. Moreover, the results illustrated that divers were faster than nonathletes in both stages when facing time pressure. Besides soil microbial respiration, water-air replacement effect (CO2-riched soil air replaced by water) was considered as a potential factor causing CO2 pulse emission, but its contribution to the water induced CO2 pulse emission remains unclear. To quantify the contribution of water-air replacement effect to water-induced CO2 pulse fluxes, a laboratory experiment was conducted, the dynamic pattern of soil air emission and its CO2 concentrations were observed during watering process. Results indicated that the soil air emission rate varied synchronously with the water infiltration rate. The total air volumes emitted from soil were 89%-93% of infiltrated water volumes. CO2 concentrations in soil air were much higher than that in ambient air, and they increased over time, with a sudden increase during 24 to 36 minutes since the initial watering. It implied that soil microbial respiration may have a response to watering during this period. As a result of a large amount of replaced air volume and high CO2 concentrations, watering caused a large increase in soil CO2 fluxes during the watering process. The water-air replacement effect was confirmed as the main pathway of water induced CO2 pulse emission during the watering process, and its contribution was estimated as 50.2%-60.2% of total CO2 fluxes. The current results would help to understand the mechanism of water-induced pulse CO2 fluxes, and to accurately estimate CO2 emission from soil under the changing environment with more frequent rainfall or irrigation. Considering safe and continuous operation of the subway tunnel, apart from the conventional drainage system, the entrances or exits of tunnels generally are designed to install special devices to block the influx of either floods due to sudden storms or harmful gases caused by terrorist attack. Traditional devices include metal plates or sandbags. In recent years, researchers have invented textile-based capsule-like inflatable device to replace the traditional devices because the new one can be flexibly located in the tunnel, capable of blocking fluids completely, light-weight and low cost. However, the fabrication of capsule-like textile structure has been seldom discussed in the literature. The capsule-like textile structure is either created by fabric weaving followed by sewing for the capsule form, which may result in unevenness of stress distribution due to existence of seams in the structure, or produced by expensive and specially designed weaving equipment. This paper is to explore two different fabrications of capsule-like textile structure for the purpose that the device is evenly structured and can be produced in one step with conventional weaving loom. The techniques of fabrication with trapezoid-formed reed as well as the application of double-layered structure have been investigated. There are few approved drugs available for the treatment of muscle-invasive bladder cancer (MIBC). Recently, we have demonstrated that isorhapontigenin (ISO), a new derivative isolated from the Chinese herb Gnetum cleistostachyum, effectively induces cell-cycle arrest at the G0/G1 phase and inhibits anchorage-independent cell growth through the miR-137/Sp1/cyclin D1 axis in human MIBC cells. Herein, we found that treatment of bladder cancer (BC) cells with ISO resulted in a significant upregulation of p27, which was also observed in ISO-treated mouse BCs that were induced by N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN). Importantly, knockdown of p27 caused a decline in the ISO-induced G0-G1 growth arrest and reversed ISO suppression of anchorage-independent growth in BC cells. Mechanistic studies revealed that ISO promoted p27 expression at mRNA transcription level through increasing direct binding of forkhead box class O1 (FOXO1) to its promoter, while knockdown of FOXO1 attenuated ISO inhibition of BC cell growth. On the other hand, ISO upregulated the 3'-untranslated region (3'-UTR) activity of p27, which was accompanied by a reduction of miR-182 expression. In line with these observations, ectopic expression of miR-182 significantly blocked p27 3'-UTR activity, whereas mutation of the miR-182-binding site at p27 mRNA 3'-UTR effectively reversed this inhibition. Accordingly, ectopic expression of miR-182 also attenuated ISO upregulation of p27 expression and impaired ISO inhibition of BC cell growth. Our results not only provide novel insight into understanding of the underlying mechanism related to regulation of MIBC cell growth but also identify new roles and mechanisms underlying ISO inhibition of BC cell growth. Background: It is often difficult to obtain sufficient quantity of RNA molecules for gene expression profiling under many practical situations. Amplification from low-input samples may induce artificial signals. Results: We compared the expression measurements of low-input mRNA samples, from 25 pg to 1000 pg mRNA, which were amplified and profiled by Smart-seq, DP-seq and CEL-seq techniques using the Illumina HiSeq 2000 platform, with those of the paired high-input (50 ng) mRNA samples. Even with 1000 pg mRNA input, we found that thousands of genes had at least 2 folds-change of expression levels in the low-input samples compared with the corresponding paired high-input samples. Consequently, a transcriptional signature based on quantitative expression values and determined from high-input RNA samples cannot be applied to low-input samples, and vice versa. In contrast, the within-sample relative expression orderings (REOs) of approximately 90% of all the gene pair in the high-input samples were maintained in the paired low-input samples with 1000 pg input mRNA molecules. Similar results were observed in the low-input total RNA samples amplified and profiled by the Whole-Genome DASL technique using the Illumina HumanRef-8 v3.0 platform. As a proof of principle, we developed REOs-based signatures from high-input RNA samples for discriminating cancer tissues and showed that they can be robustly applied to low-input RNA samples. Conclusions: REOs-based signatures determined from the high-input RNA samples can be robustly applied to samples profiled with the low-input RNA samples, as low as the 1000 pg and 250 pg input samples but no longer stable in samples with less than 250 pg RNA input to a certain degree. Surgery followed by radiotherapy is the standard treatment for chordomas, which are a rare but low-grade type of bone cancer arising from remnants of the embryonic notochord. However, disease recurrence following radiotherapy is common, most likely due to endogenous DNA repair mechanisms that promote cell survival upon radiation strikes. The ataxia telangiectasia mutated/ataxia telangiectasia mutated and Rad3 related (ATM/ATR)-mediated pathway has a critical role in DNA repair mechanisms; however, it has rarely been investigated in chordomas. In the present study, the expression of signal molecules related to the ATM/ATR pathway in chordoma tissues and adjacent normal tissues were initially examined using immunohistochemistry and western blot analysis. Chordoma U-CH1 and U-CH2 cells were subsequently used to investigate cell responses to ionizing radiation and the potential protective actions mediated by the ATM/ATR pathway. Phosphorylated (p)-ATM, p-ATR, γ-H2A histone family, member X (H2AX) and RAD51 were significantly upregulated in chordoma tissues relative to adjacent normal tissues (P<0.05). No significant reductions were observed in the viability of U-CH1 and U-CH2 cells following exposure to low-dose (1 and 2 Gy) radiation. Radiation (1 and 2 Gy) triggered a significant upregulation in p-ATM, γ-H2AX and RAD51 expression in U-CH1 cells (P<0.05), as well as a significant upregulation in p-ATM, p-ATR and RAD51 levels in U-CH2 cells (P<0.05). RAD51 knockdown increased the responses of both U-CH1 and U-CH2 cells to 1 Gy radiation, as evidenced by the significantly decreased cell viability and increased apoptosis rate (P<0.05). Collectively, the results of the present study indicated that radioresistance of chordoma cells is associated with the ATM/ATR pathway, in which RAD51 serves as an important downstream effector. It is well established that motor expertise is linked to superior mental rotation ability, but few studies have attempted to explain the factors that influence the stages of mental rotation in sport experts. Some authors have argued that athletes are faster in the perceptual and decision stages but not in the rotation stages of object-based transformations; however, stimuli related to sport have not been used to test mental rotation with egocentric transformations. Therefore, 24 adolescent elite divers and 23 adolescent nonathletes completed mental rotation tasks with object-based and egocentric transformations. The results showed faster reaction times (RT s) for the motor experts in tasks with both types of transformations (object-based cube, object-based body, and egocentric body). Additionally, the differences in favour of motor experts in the perceptual and decision stages were confirmed. Interestingly, motor experts also outperformed nonathletes in the rotation stages in the egocentric transformations. Nonlinear boundary value problems arise frequently in physical and mechanical sciences. An effective analytic approach with two parameters is first proposed for solving nonlinear boundary value problems. It is demonstrated that solutions given by the two-parameter method are more accurate than solutions given by the Adomian decomposition method (ADM). It is further demonstrated that solutions given by the ADM can also be recovered from the solutions given by the two-parameter method. Background Adolescent idiopathic scoliosis exhibits high heritability and is one of the most common spinal deformities found in adolescent populations. However, little is known about the disease-causing genes in families with adolescent idiopathic scoliosis exhibiting Mendelian inheritance. Objective The aim of this study was to identify the causative gene in a family with adolescent idiopathic scoliosis. Methods Whole-exome sequencing was performed on this family to identify the candidate gene. Sanger sequencing was conducted to validate the candidate mutations and familial segregation. Real-time QPCR was used to measure the expression level of the possible causative gene. Results We identified the mutation c.2645A>C (p.E882A) within the AKAP2 gene, which cosegregated with the adolescent idiopathic scoliosis phenotypes. AKAP2 is located in a previously reported linkage locus (IS4) on chromosome 9q31.2-q34.2 and has been implicated in skeletal development. The mutation was absent in dbSNP144, ESP6500 and 503 ethnicity- matched controls. Real-time QPCR revealed that the mRNA expression level in the patients was increased significantly compared with the family controls (p<0.0001). Conclusions AKAP2 was therefore implicated as a novel gene mutated in a Chinese family with adolescent idiopathic scoliosis. The prevailing view that the evolution of cells in a tumor is driven by Darwinian selection has never been rigorously tested. Because selection greatly affects the level of intratumor genetic diversity, it is important to assess whether intratumor evolution follows the Darwinian or the non-Darwinian mode of evolution. To provide the statistical power, many regions in a single tumor need to be sampled and analyzed much more extensively than has been attempted in previous intratumor studies. Here, from a hepatocellular carcinoma (HCC) tumor, we evaluated multiregional samples from the tumor, using either whole-exome sequencing (WES) (n = 23 samples) or genotyping (n = 286) under both the infinite-site and infinite-allele models of population genetics. In addition to the many single-nucleotide variations (SNVs) present in all samples, there were 35 "polymorphic" SNVs among samples. High genetic diversity was evident as the 23 WES samples defined 20 unique cell clones. With all 286 samples genotyped, clonal diversity agreed well with the non-Darwinian model with no evidence of positive Darwinian selection. Under the non-Darwinian model, MALL (the number of coding region mutations in the entire tumor) was estimated to be greater than 100 million in this tumor. DNA sequences reveal local diversities in small patches of cells and validate the estimation. In contrast, the genetic diversity under a Darwinian model would generally be orders of magnitude smaller. Because the level of genetic diversity will have implications on therapeutic resistance, non- Darwinian evolution should be heeded in cancer treatments even for microscopic tumors. Chordoma is a rare, slow-growing primary malignant neoplasm of the axial skeleton, which arises from the remnants of the notochord. Emerging evidence suggests that microRNAs (miRs) are dysregulated in chordoma tissues and crucially involved in chordoma pathogenesis. In the present study, the expression of 11 candidate miRs were analyzed in chordoma tissues and miR-10a and miR-125a were found to be significantly downregulated compared with controls. Notably, the expression of the primary transcripts, pri-miR-125a and pri-miR-10a was unaltered, suggesting that disturbed microRNA expression may be induced by altered pri-miRNA processing. Previous studies have indicated that disturbed adenosine deaminase acting on RNA (ADAR) expression is able to alter mRNA and miRNA adenosine to inosine (A-to-I) levels associated with cancer pathogenesis. Therefore, the expression of ADAR1 and ADAR2 was analyzed in chordoma tissues. It was found that ADAR1 was significantly overexpressed, which was accompanied by enhanced pre-miR-10a and pri-miR-125a A-to-I editing. These findings suggest that ADAR2 overexpression causes enhanced pre-miR-10a and pri-miR-125a A-to-I editing, which alters mature miR-10a and miR-125a expression and may contribute to chordoma pathogenesis. Design and synthesis of two-dimensional (2D) materials with robust ferromagnetism and biocompatibility is highly desirable due to their potential applications in spintronics and biodevices. However, the hotly pursued 2D sheets including pristine graphene, monolayer BN, and layered transition metal dichalcogenides are nonmagnetic or weakly magnetic. Using biomimetic particle swarm optimization (PSO) technique combined with ab initio calculations we predict the existence of a 2D structure, a monolayer of rocksalt-structured CrN (100) surface, which is both ferromagnetic and biocompatible. Its dynamic, thermal and magnetic stabilities are confirmed by carrying out a variety of state-of-the-art theoretical calculations. Analyses of its band structure and density of states reveal that this material is half-metallic, and the origin of the ferromagnetism is due to p-d exchange interaction between the Cr and N atoms. We demonstrate that the displayed ferromagnetism is robust against thermal and mechanical perturbations. The corresponding Curie temperature is about 675 K which is higher than that of most previously studied 2D monolayers. [ABSTRACT FROM AUTHOR]
