Puccinia striiformis f. sp. tritici (Pst) is a devastating obligate fungal biotroph that causes stripe rust disease in wheat in temperate and cooler regions, especially in Kazakhstan and Turkey. Chemical control of Pst is not a sustainable or economic strategy in these countries. Therefore, a suitable alternative to these pesticides, such as the development of resistant wheat cultivars, is needed. During the present investigation, a large set of wheat germplasms was characterized to determine the presence of the broad-spectrum resistance Yr15 gene using Y15K1_F2/uhw30_1R primers. This collection contained 360 accessions including 233 durum and bread wheat collected from KASIB cultivars and advanced lines that were developed by eight breeding programs within the Kazakhstan-Siberia Wheat Improvement Network and Turkey, and 127 wild emmer collected from Turkey only. The positive control was wild emmer IDs WEW 83-1, WEW 84-1, and WEW-85, which previously showed positive amplification of the Yr15 gene with these primers. Yr15, a broad-spectrum R-gene derived from wild emmer wheat encoding a putative kinase-pseudokinase protein designated as wheat tandem kinase 1, comprises a unique R-gene structure in wheat against the yellow (stripe) rust. None of the tested cultivated wheat germplasms from 174 durum wheat of the KASIB Network or 59 of bread wheat from Turkey showed positive amplification, but 89 of 127 (70.07%) wild Triticum dicoccoides showed positive amplification. The collection from the Karacadağ regions showed the maximum number of genotypes (46) with positive amplification for Yr15, followed by Eruh (10), Şırnak (5), Malazgir-Tunceli (4), Tunceli (4), Şırnak-Uludere yol (3), Tunceli-Pülümür (3), and Karacadağ/Kerteş Köyü –Siverek (3). We conclude that wild wheat cultivars are a good source of the Yr15 gene and that introgression of this gene into the cultivated wheat germplasm of both countries should be performed to manage the Pst pathogen in the future. This study examined the effects of an extract of the green algae Codium fragile (hereafter referred to as CFE) on dextran sulfate sodium (DSS)-induced colitis. As the administration of CFE increased, the proliferation of Akkermansia muciniphila, which is a key player in metabolic and gastrointestinal disorders, also increased. After CFE administration for 10 weeks, acetic acid was identified as the major metabolite in mouse cecum and β-glucuronidase activity in mouse fecesdecreased. Further, CFE significantly alleviated the acute intestinal injury induced by DSS administration, including DAI score, colon length, and histological score. The experimental group also displayed indications of significantly lower neutrophil activity and inflammation. The current study was carried out to screen 10 isolates (ARS-01–ARS-10) of Rhizoctonia. solani from potato tubers cv. Kuroda, which were collected from various potato fields in Multan, Pakistan. The isolates were found to be morphologically identical, as the hyphae exhibit the production of branches at right angles and acute angles often accompanied by septum near the emerging branches. Anastomosis grouping showed that these isolates belonged to AG-3. A pathogenicity test was performed against the susceptible Kuroda variety and among the isolates, ARS-05 exhibited the highest mean severity score of approximately 5.43, followed by ARS-09, which showed a mean severity score of about 3.67, indicating a moderate level of severity. On the lower end of the severity scale, isolates ARS-06 and ARS-07 displayed mean severity scores of approximately 0.53 and 0.57, respectively, suggesting minimal symptom severity. These mean severity scores offer insights into the varying degrees of symptom expression among the different isolates of R. solani under examination. PCoA indicates that the severe isolate causing black scurf on the Kuroda variety was AG-3. A comprehensive analysis of the distribution, genetic variability, and phylogenetic relationships of R. solani anastomosis groups (AGs) related to potato crops across diverse geographic regions was also performed to examine AG prevalence in various countries. AG-3 was identified as the most widespread group, prevalent in Sweden, China, and the USA. AG-5 showed prominence in Sweden and the USA, while AG-2-1 exhibited prevalence in China and Japan. The phylogenetic analysis unveiled two different clades: Clade I comprising AG-3 and Clade II encompassing AG-2, AG-4, and AG-5, further subdivided into three subclades. Although AGs clustered together regardless of origin, their genetic diversity revealed complex evolutionary patterns. Bioactive compounds are secondary metabolites of plants. They offer diverse pharmacological properties. Peganum harmala is reported to have pharmaceutical effects like insecticidal, antitumor, curing malaria, anti-spasmodic, vasorelaxant, antihistaminic effect. Rosa brunonii has medicinal importance in its flower and fruits effective against different diseases and juice of leaf is reported to be applied externally to cure wounds and cuts. Dryopteris ramosa aqueous leaf extract is used to treat stomach ulcers and stomachaches. Each of these three medicinal plants have been indicated to have anticancer, antiviral, antioxidant, cytotoxic and antifungal effects but efficacy of their bioactive compounds remained unexplored. Study was aimed to explore In-vitro and In-silico anticancer, antiviral, antioxidant, cytotoxic and antifungal effects of bioactive compounds of above three medicinal plants. DPPH and ABTS assay were applied for assessment of antioxidant properties of compounds. Antibacterial properties of compounds were checked by agar well diffusion method. Brine shrimp lethality assay was performed to check cytotoxic effect of compounds. Molecular docking was conducted to investigate the binding efficacy between isolated compounds and targeted proteins. The compound isomangiferrin and tiliroside presented strong antioxidant potential 78.32% (± 0.213) and 77.77% (± 0.211) respectively in DPPH assay while harmaline showed 80.71% (± 0.072) at 200 µg/mL in ABTS assay. The compound harmine, harmaline and PH-HM 17 exhibited highest zone of inhibition 22 mm, 23 mm, 22 mm respectively against Xanthomonas while Irriflophenone-3-C-β- D-glucopyranoside showed maximum zone of inhibition 34 mm against E. coli. The compound isomangiferrin and vasicine contained strong antibacterial activity 32 mm and 22 mm respectively against S. aureus. The compound mangiferrin, astragalin, tiliroside, quercitin-3-O-rhamnoside showed maximum inhibitory zone 32 mm, 26 mm, 24 mm and 22 mm respectively against Klebsiella pneumoniae. Highest cytotoxic effect was observed by compound tiliroside i.e. 95% with LD50 value 73.59 µg/mL. The compound tiliroside showed the best binding mode of interaction to all targeted proteins presenting maximum hydrophobic interactions and hydrogen bonds. The binding affinity of tiliroside was − 17.9, − 14.9, − 14.6, − 13.8, − 12.8 against different proteins 6VAR, 5C5S, IEA3, 2XV7 and 6LUS respectively. Bioactive compounds are significant natural antioxidants, which could help to prevent the progression of various diseases caused by free radicals. Terminalia chebula Retz. (Fam. Combretaceae), commonly called black or chebulic myrobalan, is a species of Terminalia. In Tibet, T. chebula is called the "King of Medicine". It is well known as 'Haritaki' since it can be used to cure all kinds of diseases and is considered sacred to God Siva (Hara). The whole plant possesses high medicinal value. The aim of this article is to review the available scientific information regarding traditional uses, bioactive chemical constituents, and pharmacological activities of T. chebula based on in vitro and in vivo studies. First, in this paper, the studies published on T. chebula from 1980 to 2022 were reviewed, and the biological activity and mechanisms of action of T. chebula were evaluated based on in vitro and in vivo studies. T. chebula presented anti-diabetic, anti-hyperlipidemic, antioxidant, hepatoprotective, neuroprotective, anti-inflammatory, anti-arthritic, gastroprotective, anti-microbial, antiparasitic, wound healing, and anti-aging activities. To reach the sustainable development goals on health management in Litopenaeus vannamei shrimp culture, Pediococcus pentosaceus AB01 was supplemented in shrimp diet. In this study, the control diet and three experimental diets containing P. pentosaceus AB01 (108, 109, 1010 CFU/g) were separately introduced to L. vannamei for a 28 days feeding trial. After the feeding trial, percent weight gain, feeding efficiency, and feed conversion ratio (FCR) were significantly elevated in L. vannamei administered with P. pentosaceus AB01 at 109 and 1010 colony‐forming unit (CFU)/g. Protease, amylase, and trypsin were found at higher levels in the probiotic‐supplied groups. The feeding of shrimps with P. pentosaceus AB01 significantly increased innate immune response and levels of related biochemical parameters in the haemolymph. After the white spot syndrome virus (WSSV) challenge, supplementation of P. pentosaceus AB01 had significant positive effects (p <.05) on survival rate, compared to that of the control diet. The higher resistance of L. vannamei to WSSV might have been due to alterations in the gut microbiome composition and upregulation of the Toll‐Like Receptor (TLR) signalling pathway. Optimum growing temperature is necessary for maximum yield-potential in any crop. The global atmospheric temperature is changing more rapidly and irregularly every year. High temperature at the flowering/reproductive stage in rice causes partial to complete pollen sterility, resulting in significant reduction in grain yield. Green Super Rice (GSR) is an effort to develop an elite rice type that can withstand multiple environmental stresses and maintain yield in different agro-ecological zones. The current study was performed to assess the effect of heat stress on agronomic and physiological attributes of GSR at flowering stage. Twenty-two GSR lines and four local checks were evaluated under normal and heat-stress conditions for different agro-physiological parameters, including plant height (PH), tillers per plant (TPP), grain yield per plant (GY), straw yield per plant (SY), harvest index (HI), 1000-grain weight (GW), grain length (GL), cell membrane stability (CMS), normalized difference vegetative index (NDVI), and pollen fertility percentage (PFP). Genotypes showed high significant variations for all the studied parameters except NDVI. Association and principal component analysis (PCA) explained the genetic diversity of the genotypes, and relationship between the particular parameters and grain yield. We found that GY, along with other agronomic traits, such as TPP, SY, HI, and CMS, were greatly affected by heat stress in most of the genotypes, while PH, GW, GL, PFP, and NDVI were affected only in a few genotypes. Outperforming NGSR-16 and NGSR-18 in heat stress could be utilized as a parent for the development of heat-tolerant rice. The abundance and structural composition of nitrogen (N) transformation-related microbial communities under certain environmental conditions provide sufficient information about N cycle under different soil conditions. This study aims to explore the major challenge of low N use efficiency (NUE) and N dynamics in aerobic rice systems and reveal the agronomic-adjustive measures to increase NUE through insights into the ecophysiology of ammonia oxidizers. Water-saving practices, like alternate wetting and drying (AWD), dry direct seeded rice (DDSR), wet direct seeding, and saturated soil culture (SSC), have been evaluated in lowland rice; however, only few studies have been conducted on N dynamics in aerobic rice systems. Biological ammonia oxidation is majorly conducted by two types of microorganisms, ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB). This review focuses on how diversified are ammonia oxidizers (AOA and AOB), whose factors affect their activities and abundance under different soil conditions. It summarizes findings on pathways of N cycle, rationalize recent research on ammonia oxidizers in N-cycle, and thereby suggests adjustive agronomic measures to reduce N losses. This review also suggests that variations in soil properties significantly impact the structural composition and abundance of ammonia oxidizers. Nitrification inhibitors (NIs) especially nitrapyrin, reduce the nitrification rate and inhibit the abundance of bacterial amoA without impacting archaeal amoA. In contrast, some NIs confine the hydrolysis of synthetic N and, therefore, keep low NH4+-N concentrations that exhibit no or very slight impact on ammonia oxidizers. Variations in soil properties are more influential in the community structure and abundance of ammonia oxidizers than application of synthetic N fertilizers and NIs. Biological nitrification inhibitors (BNIs) are natural bioactive compounds released from roots of certain plant species, such as sorghum, and could be commercialized to suppress the capacity of nitrifying soil microbes. Mixed application of synthetic and organic N fertilizers enhances NUE and plant N-uptake by reducing ammonia N losses. High salt concentration promotes community abundance while limiting the diversity of AOB and vice versa for AOA, whereas AOA have lower rate for potential nitrification than AOB, and denitrification accounts for higher N2 production. Archaeal abundance, diversity, and structural composition change along an elevation gradient and mainly depend on various soil factors, such as soil saturation, availability of NH4+, and organic matter contents. Microbial abundance and structural analyses revealed that the structural composition of AOA was not highly responsive to changes in soil conditions or N amendment. Further studies are suggested to cultivate AOA and AOB in controlled-environment experiments to understand the mechanisms of AOA and AOB under different conditions. Together, this evaluation will better facilitate the projections and interpretations of ammonia oxidizer community structural composition with provision of a strong basis to establish robust testable hypotheses on the competitiveness between AOB and AOA. Heavy metal toxicity reduces the growth and development of crop plants growing in metal-contaminated regions. Disposal of industrial waste in agricultural areas has negative effects on the physiochemical activities of plants. This research aimed to examine the fulvic acid (FA)-mediated efficacy of Brassica napus L. regarding stress tolerance in soil amended with paper sludge (PS). For this purpose, plants were grown for 90 days under greenhouse conditions at various concentrations of PS-amended soils (0, 5, 10, and 15%) being irrigated with water containing FA (0, 10, and 20%). All the physicochemical parameters of PS were carried out before and after plant transplantation. Paper sludge toxicity reduced the growth (shoot/root length, fresh/dry weight of shoot/root, numbers of flowers and leaves) and physicochemical characteristics of exposed B. napus plants. In comparison, FA application improved growth by reducing the metal uptake in the shoot of plants grown at various concentrations of PS. An increasing trend in antioxidant enzyme activity was observed by increasing the FA concentration (0%-10% and 20%). Post-harvest analysis indicated that the amount of tested metals was significantly reduced at all PS concentrations. Minimum metal uptake was observed at 0% concentration and maximum at 15% concentration of paper sludge. Additionally, FA application at 20% concentration reduced Chromium (Cr), Cadmium (Cd), and Lead (Pb) uptake in the shoot from 6.08, 34.42, and 20.6 mgkg−1 to 3.62, 17.33, and 15.22 mgkg−1, respectively. At this concentration of paper sludge in the root, 20% FA reduced Cr, Cd, and Pb uptake from 11.19, 44.11, and 35.5 mgkg−1 to 7.88, 27.01, and 24.02 mgkg−1, respectively. Thus, FA at 20% concentration was found to be an effective stimulant to mitigate the metal stress in B. napus grown in paper sludge-polluted soil by reducing metal uptake and translocation to various plant parts. Scrophularia buergeriana (SB) was once thought to be an herbal remedy with a wide range of uses. Lactobacillus transformation improves the medicinal benefits of biological substances. However, the effect of fermented SBE in macrophages on inflammatory expression is unknown. Furthermore, the synergistic effects of SBE and lactic acid bacteria (LAB) have not been previously investigated. Therefore, in the present study, we investigated the mixture of SB fermented with different lactic acid bacteria (LAB) strains, and analyzed their characteristics before and after fermentation. The results showed that LAB activities increased the levels of phenolic compounds, flavonoids, and iridoids, and SB extract (SBE) stimulated the growth of Lactobacillus strains. Moreover, we also determined the anti-inflammatory effects of the SBE fermented sample in lipopolysaccharide-treated RAW264.7 cells. It was observed that the treatment of SBE fermented sample inhibited nitric oxide (NO) production than SBE and also decreased the RNA expression of iNOS, IL-1β, IL-6, TNF-α, and COX-2. Abioticstress such as drought is a potential threat posing a severe challenge to wheat production across the globe. The current study comparatively delineated the performance of elite Pakistani bread wheat genotypes at physiological (chlorophyll, canopy temperature, cell membrane percentage stability and leaf relative water content), agronomic (plant height, tillers plant−1, flag leaf area, spike length, spikelets spike−1, grains spike−1, grain yield spike−1, thousand grain weight and plant biomass) and genetic (TaDREB1A, TaGROS-A, TaLEA3, TaHSFA1a, TaWRKY44 and TaEXPA2) levels. Atri-replicate experiment was conducted in a two factorial arrangement using RCBD, and data were analyzed statistically using the computer-based programsStatistix8.1 and R-studio. In general, all wheat genotypes illustrated significant (p ≤ 0.05) alterations in physiological and agronomic traits under drought stress as compared to the control; however, this alteration was significantly (p ≤ 0.05) different among all genotypes owing to their varying genetic potential. Furthermore, these genotypes were evaluated for the extent of the association of physiological and agronomic traits using PCA, correlation and heatmap analysis, which proved statistically significant variation in the paired association of traits among all genotypes during drought stress as compared to the control. In addition, based on statistical evaluations, the genotypes Pakistan-13, Shahkar-13, AAS-11, Chakwal-86, Chakwal-50 and AUR-09 were found to be tolerant, while genotypes Anmol-97, Chakwal-97, Bhakkar-02 and BWP-97 were comparatively susceptible. Furthermore, these screened genotypes showed differential expression of drought-related genes, with relatively high expression in tolerant genotypes compared to susceptible genotypes. The current study concluded that physiological, agronomic and molecular characteristics are significantly interconnected, and these associations determine the end productivity of wheat genotypes during abiotic stress. We evaluated the effectiveness of Scrophularia buergeriana extract (Brainon) on cognitive dysfunction and determined its underlying mechanisms in a scopolamine (SCO)-treated mouse model of memory impairment. Brainon treatment for 28 days ameliorated the symptoms of memory impairment as indicated by the results of both passive avoidance performance and the Morris water mazes. Brainon lowered acetylcholinesterase activity and raised acetylcholine levels in the hippocampus. The treatment elevated the protein levels of brain-derived neurotrophic factor (BDNF) and phosphorylated cAMP response element-binding (CREB). Additionally, the excessive generation of SCO-induced reactive oxygen species (ROS) and subsequent oxidative stress were suppressed by the enhancement of superoxide dismutase (SOD)-1 and SOD-2 proteins. mRNA levels of upregulated interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α, as well as the apoptotic protein Bcl-2-associated X protein (Bax), cleaved caspase-9, and cleaved poly adenosine diphosphate-ribose polymerase (PARP) expression after SCO injection were downregulated by Brainon treatment. Collectively, these findings suggested that Brainon possesses anti-amnesic effects through the CREB-BDNF pathway. East Asian region is home to wild soybean and has high topographic complexity. Measuring genetic diversity and geographical distribution patterns are essential steps to expand the germplasm base. We used SoySNP6K to determine the genetic diversity and population genetic structure of the wild soybean populations from four East-Asian geographies i.e. China, Japan, Korea, and Russian Far East. The wild soybean populations from China, Japan, and Korea are distinct from one another based on genetic structure, whereas the one from Russian Far East was not distinguishable from the Chinese wild soybeans. These populations share three ancestral populations and have a large number of common SNPs. Despite the low genetic diversity scores between the populations, the Chinese accessions had the highest genetic diversity values while the accession from Russian Far East had the lowest. Our analysis also showed that the population is showing a departure from the Hardy–Weinberg equilibrium and there is a possibility of inbreeding for most of the loci. The results showed that the genetic variation follows the geographic distribution within each geographically distinct wild soybeans population. Therefore, a diverse collection of wild soybeans representing all-natural habitats should include representatives from each of these sub-centers. This population would provide important breeding materials to expand the allelic pool of cultivated soybeans through introgression. Based on these criteria, the Chung's wild legume germplasm collection from Korea provides a good representation of the genetic diversity in wild soybeans for use in soybean breeding programs. Galactan is a major sulfate polysaccharide in the red seaweed, Pyropia yezoensis. In the present study, the prebiotic potential and anticancer activities of three sulfate galactans with different molecular weights (GPYcrude, GPY300, and GPY10) were evaluated in vitro. Monosaccharide composition and structural analyses of these sulfate galactans showed that all polysaccharide fractions contained the same monosaccharides but had different molecular weights. All the polysaccharides tested displayed potential prebiotic activity, with GPY10 noticeably having the best potential compared to others, especially for Bifidobacterium spp., which highlights possible correlations between structural galactan features and prebiotic activity. Furthermore, all three sulfate galactans were cytotoxic to DU145 and PC-3 prostate cancer cells, with GPY10 exhibiting the greatest inhibition in DU145 cells. Moreover, GPY10 induced modulates the production of intracellular reactive oxygen species (ROS), with subsequent increased apoptosis of DU145 prostate cancer cell and modulating the PI3K/AKT/mTOR signaling pathway. GPY10 increased expression of Bax, executor caspase-3, and initiator caspases 8 and 9. Osteoarthritis (OA) is a general joint illness caused by the destruction of joint cartilage, and is common in the population of old people. Its occurrence is related to inflammatory reactions and cartilage degradation. AyuFlex® is an aqueous extract of Terminalia chebula fruit, and T. chebula has been utilized extensively in several traditional oriental medications for the management of diverse diseases. Pre-clinical and clinical research has shown its antioxidant and anti-inflammatory effectiveness. Nevertheless, the mechanism underlying the anti-arthritic effects of AyuFlex® remains unclear. In the current research, we proposed the ameliorating effects of AyuFlex® with respect to the incidence of OA and described the latent signalization in interleukin (IL)-1β-treated chondrocytes and MIA-incurred OA in a rat model. In vitro, AyuFlex® decreased oxidative stress and induction of pro-inflammatory cytokines and mediators as well as matrix metalloproteinases (MMPs), while also increasing the levels of collagen synthesis-related proteins. Mechanistically, we identified that AyuFlex® disrupted nuclear factor kappa B (NF-κB) and mitogen-activated protein kinase (MAPK) activation via the inhibition of NF-κB p65 and extracellular regulated protein kinase (ERK) phosphorylation. The ameliorating effects of AyuFlex® were also observed in vivo. AyuFlex® significantly inhibited the MIA-incurred increase in OA symptoms such as oxidative stress, cartilage damage, and changes in cytokines and MMPs revelation in arthrodial cartilage. Therefore, our results suggest that AyuFlex® attenuates OA progression in vivo, indicating that AyuFlex® can be suggested as an excellent therapeutic remedy for the care of OA. Scrophularia buergeriana (SB) Miq. (Scrophulariaceae) has been used to help cure swelling and fever and has reported antioxidant and neuro-protective effects. However, few mechanism–based studies have evaluated the memory-improving effects in a beta-amyloid induced memory loss model. As a result of Scrophularia buergeriana extract (SBE) administration (30 and 100 mg/kg) for 28 days significantly recovered beta-amyloid-induced amnesia in the passive avoidance test and improved the impairment of spatial memory in the Morris Water Maze (MWM) task. Furthermore, SBE up-regulated superoxide dismutase-1 (SOD)-1, SOD-2, glutathione peroxidase-1, and B-cell lymphoma (Bcl)-2 protein expression levels. Additionally, SBE downregulated Bcl-2-associated X protein, cleaved caspase-9, cleaved poly (adenosine diphosphate-ribose) polymerase, and Aβ protein expression levels and inhibited the phosphorylation of the tau protein of Aβ-treated mice hippocampus. These results demonstrate that SBE improved memory impairment by reducing beta-amyloid induced neurotoxicity and regulated oxidative stress, anti-apoptotic pathways. [ABSTRACT FROM AUTHOR]

Lactobacillus fermentum MJM60397 was subjected to in vitro safety tests and in vivo probiotic characterization. The MJM60397 strain was susceptible to antibiotics and was found to be non-mucinolytic and non-hemolytic, and does not produce bioamines. In addition, MJM60397 tolerated simulated oro-gastrointestinal conditions and adhered to Caco-2 cells. MJM60397 also exhibited bile salt hydrolase activity and could deconjugate bile acids. The hypocholesterolemic effects of strain MJM60397 were studied in high-fat diet-induced hypercholesterolemic male ICR mice. The mice were fed a high-cholesterol diet (HCD) and were divided into the following three experimental groups: HCD-control (HCD-Con), mice fed with HCD + L. fermentum MJM60397 (HCD-MJM60397), and mice fed with HCD + L. acidophilus ATCC 43121 (HCD-L.ac) as the positive control. Simultaneously, a normal control diet (NCD) group was maintained. After 7 weeks, the total cholesterol and low-density lipoprotein (LDL) cholesterol levels were significantly reduced in the livers of the HCD-MJM60397 mice when compared to those in the HCD-Con and HCD-L.ac mice. Fecal total bile acid content was significantly (P < 0.05) higher in the HCD-MJM60397 group than in the NCD, HCD-Con, and HCD-L.ac groups. Analysis of gene expression revealed higher expression of LDLR gene in the livers of the HCD-MJM60397 and HCD-L.ac mice than in the livers of the HCD-Con mice. These findings show that the hypocholesterolemic effects of the MJM60397 strain were attributable to its bile salt deconjugating activity, which resulted in decreased bile acid absorption and increased excretion of bile acids in the feces. The study was aimed to characterize the probiotic properties of Lactobacillus paracasei subsp. paracasei strain KNI9 and its antagonistic activity against Yersinia enterocolitica subsp. enterocolitica. The strain KNI9 was susceptible to antibiotics such as chloramphenicol, tetracycline, erythromycin, and streptomycin recommended by European food safety authority (EFSA). Strain KNI9 exhibited tolerance to simulated oro-gastrointestinal (OGT) condition, adherence to Caco-2 cells, and antimicrobial activity against intestinal enteric pathogens such as Yersinia enterocolitica subsp. enterocolitica, Shigella boydii, and Listeria monocytogenes. Furthermore, the strain KNI9 inhibited the adherence and invasiveness of Y. enterocolitica subsp. enterocolitica to Caco-2 cell line. These results indicate that the L. paracasei subsp. paracasei KNI9 could be further developed into a potential probiotic strain after appropriate in vivo studies. [ABSTRACT FROM AUTHOR]

The aim of the present study was to evaluate the neuroprotective effect of Citrus aurantium extract (CAE) and nobiletin against amyloid β 1–42 (Aβ 1–42)-induced spatial learning and memory impairment in mice. After injecting Aβ 1–42 (5 µl/2.5 min, intracerebroventricular injection), amnesic mice were orally administered CAE and nobiletin for 28 days. Memory, spatial and cognitive ability were measured using passive avoidance and a Morris water maze task. Acetylcholinesterase (AchE) activity was investigated in the hippocampus and cortex using commercial kits and the analysis of Bax, Bcl-2, and cleaved caspase-3 protein expression by western blot assays was used to confirm the anti-apoptotic mechanism of CAE and nobiletin. The present study confirmed impairments in learning and memory in the Aβ-induced neurodegenerative mice with increased AchE activity in the brain. However, the daily administration of CAE and nobiletin reduced the spatial learning deficits and increased the AchE activity in the cortex and hippocampus. Furthermore, CAE and nobiletin significantly downregulated the Bax and cleaved caspase-3 protein expression and upregulated the Bcl-2 and Bcl-2/Bax expression in the cortex and hippocampus of Aβ-treated mice. These results suggest that CAE and nobiletin exert a neuroprotective effect by regulating anti-apoptotic mechanisms, including reduced AchE activity in the cortex and hippocampus of the cognitive deficit mouse model. Cholesterol-lowering activity is an important health benefit of lactic acid bacteria (LAB). This study aimed to screen LAB strains with cholesterol-lowering activities from salted fermented shrimp and evaluate probiotic characteristics and cholesterol-lowering potentials of these LAB isolates. Among 191 lactic acid strains isolated from traditional salted-fermented shrimp food, FB003 isolate showed the highest cholesterol-lowering activity and investigated as probiotics with cholesterol-lowering ability. Biochemical analysis and 16S rRNA sequencing revealed that this LAB isolate was Lactobacillus plantarum FB003. To screen probiotic trait, L. plantarum FB003 was found to be susceptible to six antibiotics tested and broad-spectrum antimicrobial activity. It also produced various enzymes such as galactosidase, glucosidase, and mannosidase. In addition, this strain showed autoaggregation, and coaggregation capacity for various pathogens. Moreover, it could adhere to Caco-2 cells and be exerted lowering cholesterol effects in Caco-2 cells via an upregulation of PPARα to inhibit NPC1L1 mRNA expression. Strain L. plantarum FB003 might be effective as a candidate probiotic with high cholesterol-lowering activity. The results of the present study suggest that L. plantarum FB003 have an impact on preventing high cholesterol level and may be used as starter culture for shrimp fermentation. An exponential increase of heavy metal toxicant in the human body is a growing health‐related problem. In the present study, Pediococcus pentosaceus FB145 and FB181 strains were isolated from fermented seafood and served as highly Cd‐resistant strains. The unchanged structural cells of P. pentosaceus strains after treatment with Cd cations were investigated using a scanning electron microscope energy dispersive X‐ray analysis. Furthermore, both P. pentosaceus strains showed strong human gastrointestinal tract resistance properties. The Cd biosorption results fit the pseudo–second‐order kinetics with capacities for P. pentosaceus FB145 and FB181 for Cd were 52.8 and 50.3 mg/g, respectively, whereas the maximum biosorption capacities were 52.65 and 50.35 mg/g, respectively. The equilibrium data were well fitted to the Freundlich isotherm model. The binding of Cd to bacterial cells may be caused by the presence of different functional groups such as carboxyl, amide, and phosphate on cell surface, which was confirmed by the Fourier transform infrared‐attenuated total reflectance spectra. Moreover, these strains decreased the Cd bioaccessibility by 44.7–46.8% in the in vitro digestion model. These findings indicate that P. pentosaceus FB145 and FB181 are novel potent biosorbent for preventing cadmium toxicity and reducing its absorption into the human body. [ABSTRACT FROM AUTHOR]

Cellulose synthase A (CESA) is a key enzyme involved in the complex process of plant cell wall biosynthesis, and it remains a productive subject for research. We employed systems biology approaches to explore structural diversity of eudicot CESAs by exon–intron organization, mode of duplication, synteny, and splice site analyses. Using a combined phylogenetics and comparative genomics approach coupled with co-expression networks we reconciled the evolution of cellulose synthase gene family in eudicots and found that the basic forms of CESA proteins are retained in angiosperms. Duplications have played an important role in expansion of CESA gene family members in eudicots. Co-expression networks showed that primary and secondary cell wall modules are duplicated in eudicots. We also identified 230 simple sequence repeat markers in 103 eudicot CESAs. The 13 identified conserved motifs in eudicots will provide a basis for gene identification and functional characterization in other plants. Furthermore, we characterized (in silico) eudicot CESAs against senescence and found that expression levels of CESAs decreased during leaf senescence. [ABSTRACT FROM AUTHOR]

Increase in the temperature of seawater due to global warming is one of the major stress factors that has reduced seaweed, in particular Pyropia yezoensis, mariculture worldwide. In this milieu, silicon application has been reported to protect and heal the cells of thallus under stress conditions. The present study investigated the effect of in vitro different temperature and irradiance levels in P. yezoensis gametophytes and the potential mitigating effect of silicic acid (50 μM). The results revealed that the effect of temperature stress on the relative growth rate, the optimum photochemical efficiency of PS II (Fv/Fm), and the net photosynthetic rate (Pn) were minimal in the presence of silicon. The silica content in the thallus increased with increase in temperature, thus upregulating the expression of antioxidant genes. However, the effect of silicon was not characterized in P. yezoensis under irradiance treatments. Legumes are the world's primary source of dietary protein and are particularly important for those in developing economies. However, the biofortification potential of legumes remains underexploited. Legumes offer a diversity of micronutrients and amino acids, exceeding or complementing the profiles of cereals. As such, the enhancement of legume nutritional composition presents an appealing target for addressing the "hidden hunger" of global micronutrient malnutrition. Affecting ~2 billion people, micronutrient malnutrition causes severe health effects ranging from stunted growth to reduced lifespan. An increased availability of micronutrient‐enriched legumes, particularly to those in socio‐economically deprived areas, would serve the dual functions of ameliorating hidden hunger and increasing the positive health effects associated with legumes. Here, we give an updated overview of breeding approaches for the nutritional improvement of legumes, and crucially, we highlight the importance of considering nutritional improvement in a wider ecological context. Specifically, we review the potential of the legume microbiome for agronomic trait improvement and highlight the need for increased genetic, biochemical, and environmental data resources. Finally, we state that such resources should be complemented by an international and multidisciplinary initiative that will drive crop improvement and, most importantly, ensure that research outcomes benefit those who need them most. Legumes are prime targets for biofortification, thus bear the potential to reduce micronutrient malnutrition. However, they are currently underexploited in global agriculture. Here, we provide an updated overview of the nutritional improvement of legumes and consider them in the context of their growth environment. Salinivibrio genus is commonly found in salted seafood products. In this study, chitinase produced by Salinivibrio sp. BAO‐1801 isolated from salted fermented shrimp was purified and subsequently characterized. The molecular weight of BAO‐1801 chitinase was approximately 94.2 kDa by SDS–PAGE analysis. It was classified as a chitinase C based on homology analysis of its N‐terminal amino acid residues. This strain BAO‐1801 chitinase was then used for synthesis of (GlcNAc)2. Degradation of colloidal chitin and N‐acetyl chitooligosaccharides by BAO‐1801 chitinase was then analyzed and (GlcNAc)2 was identified as the main product by thin layer chromatography and high‐performance liquid chromatography. Effects of temperature and pH on activity and stability of BAO‐1801 chitinase were also investigated. Furthermore, this enzyme inhibited fungal growth in a dose‐dependent manner. Taken together, these results suggest that this Salinivibrio or its chitinase can be used for the enzymatic degradation of chitin to produce chitobiose in industrial process. [ABSTRACT FROM AUTHOR]

A novel actinobacterium, strain SK68, was isolated from the rhizosphere of peanut plant and its salinity stress alleviation ability was studied using tomato (Solanum lycopersicum cv. Micro-Tom) plants. Based on 16S rDNA based phylogenetic analysis, strain SK68 has been identified as a Streptomyces sp. Strain SK68 had branched substrate mycelium bearing smooth surfaced spores and the spore colour is brownish grey on ISP4 medium. It exhibited enzyme activities such as xylanase, cellulase, amylase, and pectinase and degraded hypoxanthine, casein, and L-tyrosine. The strain SK68 differed in its banding pattern in BOX-PCR and RAPD fingerprinting compared to the closely matching type strains Streptomyces erythrochromogenes NBRC 3304T (AB184746), S. flavotricini NBRC 12770T (AB184132), S. racemochromogenes NBRC 12906T (AB184235), and S. polychromogenes NBRC 13072T (NR041109). Strain SK68 was evaluated for its salinity stress-alleviating activity in tomato plants with 180 mmol/L NaCl under gnotobiotic condition. Flavonoids are specialized plant secondary metabolites that are mainly present as glycoconjugates and function as attractants to pollinators and symbionts, UV protectants, allelochemicals, and have antimicrobial and antiherbivore activity for plant health. Because of the heterogeneity of UDP-glycosyltransferases (UGTs) for glycosylation in plants, their function in flavonoid glycosylation remains largely unknown in soybean and other legumes, particularly that of the UGT92 genes. Here, we identified 152 putative UGT92 genes across 48 plant species and elucidated their mode of duplication, expansion/deletion pattern, alignment, phylogenetic analysis, and genome-wide distribution. Two novel UGT-encoding genes Glyma14g04790 (UGT92G1) and Glyma15g03670 (UGT92G2) were isolated from soybean and their heterologous expression was optimized in Escherichia. coli. Both genes exhibited catalytic activity toward quercetin, kaempferol, and myricetin, with UDPglucose as the sugar donor and were characterized as flavanol-specific UGTs. High expression of both UGTs was observed in adaxial and abaxial parenchyma, suspensor cells, and adaxial and abaxial epidermis cells during seed development, suggesting that they are seed-specific flavanol glycosyltransferases in soybean. Co-expression analysis of UGT92 genes with their first and second neighborhood genes provided a basis for their network elucidation in soybean. We provide valuable information on the role of UGT92 in seed development via the glycosylation of multiple flavanols and the potential metabolic engineering of flavonoid compounds in both plants and E. coli. Light-emitting diode (LED) technology offers potential energy-efficient light sources for algal aquaculture. In this study, the growth rate and ulvan yield of Ulva pertusa used for broad commercial applications were enhanced in vitro. To investigate the response of Ulva to LEDs, algae were grown under white fluorescent, pure primary color LED, and mixed LED lights for 28 days. Ulva sp. under blue LED and white fluorescent light showed higher specific growth rate (SGR, 12% day−1) than other light sources, while the growth under mixed red-blue LED light was lower than that under white fluorescent lighting. Despite the differences in alga growth under LEDs and white fluorescent light, few differences are observed in N and P content among algae grown under any light source. U. pertusa showed tissue nitrogen and ulvan contents in the ranges of 4.4-6.7% and 20.4-25.1%, respectively, under all LED light treatments. The ulvan extracted from algae under blue LED showed high levels of antioxidant activity measured with 2,2-diphenylpicrylhydrazyl (DPPH) radical and hydrogen peroxide. Moreover, the ulvan obtained under different lights showed different monosaccharide compositions. This study investigated the effects of non-fermented Smilax china L. (SC) and fermented Smilax china L. leaf extract by Aspergillus oryzae (FSC) on lipogenesis and antioxidant activities in diet induced-obese mice. While principal bioactive constituents in SC were decreased by fermentation, diverse secondary metabolites were identified. Both SC and FSC supplementation reduced body weight and epididymal fat mass in mice fed a high-fat diet. Hepatic triglyceride and cholesterol levels were lowered by SC treatment and the degree of lipid reduction was higher in FSC-treated group due to hepatic upregulation of lipogenic genes. SC and FSC improved hepatic function by lowering the activity of serum biomarker enzymes. In contrast, the activities of antioxidant enzymes were elevated and cellular lipid peroxide were decreased in liver. Taken together, FSC is more potent in preventing obesity and ameliorating hepatic function by fermentation-mediated production of bioactive constituents that have higher antioxidant and anti-obese efficacy. Practical applications The studies revealing that specific mechanism regarding anti-obesity effect of consumption of SC leaf in animal models is limited. Extract of fermented SC by Aspergillus oryzae (FSC) could be used as a bioactive ingredient for functional food products that can have a health claim for reducing hepatic lipogenesis in the liver in high-fat diet induced-obese mice. New saponin components, Sg-6 saponins, have been recently reported from the seeds of wild soybean (Glycine soja) which may have specific health benefits. To evaluate the possible health benefits, a large amount of Sg-6 saponins are needed, but general group A acetyl saponins and new Sg-6 saponins are eluted in overlapping peaks by ordinal preparative high-performance liquid chromatography and/or open column methods. A new method is proposed in this report. This method includes (1) deacetylation of group A acetyl saponins in alkali condition with KOH, (2) precipitation of Sg-6 saponins in acid condition with HCl, (3) recovery of Sg-6 saponins with aqueous methanol from the precipitate, and (4) elution of Sg-6 saponins by preparative reverse-phase open column. With this method, from 450 g of wild soybean whole seed powder, about 1 g of Sg-6 saponins (mixture of six components) was clearly separated from other saponins with 61% recovery. Capsosiphon fulvescens is a filamentous green algae in the class Ulvophyceae. It has been consumed as food with unique flavor and soft texture to treat stomach disorders and hangovers, and its economic value justifies studying its nutritional and potential therapeutic effects. In contrast to these applications, only a few taxonomic studies have been conducted on C. fulvescens. In particular, classification and phylogenetic relationships of the C. fulvescens below the order level are controversial. To determine its phylogenetic position in the class, we used rbcL and 18S rDNA sequences as molecular markers to construct phylogenetic trees. The amplified rbcL and 18S rDNA sequences from 4 C. fulvescens isolates (Jindo, Jangheung, Wando, and Koheung, Korea) were used for phylogenetic analysis by employing three different phylogenetic methods: neighbor joining (NJ), maximum parsimony (MP), and maximum likelihood (ML). The rbcL phylogenetic tree showed that all taxa in the order Ulvales were clustered as a monophyletic group and resolved the phylogenetic position of C. fulvescens in the order Ulotrichales. The significance of our study is that the 18S rDNA phylogenetic tree shows the detailed taxonomic position of C. fulvescens. In our result, C. fulvescens is inferred as a member of Ulotrichaceae, along with Urospora and Acrosiphonia. [ABSTRACT FROM AUTHOR]

BACKGROUND It has been reported that Smilax china L. leaf ( SCL) provided various biological functions owing to polyphenols. The objective of the current study was to assess the enhancing effect of processing methods and microbial conversions on phenolic acid and flavonoid content and radical scavenging capacity of SCL for potential applications of diverse food products. RESULTS Targeted phenolic acid (chlorogenic acid) and flavonoids (piceid and quercetin) were identified in fresh SCL using liquid chromatography-mass spectrometry. The total amount of identified phenolic acid and flavonoids was highest in steamed SCL (12.70 ± 0.12 mg g−1 on a dry matter basis, dmb). A substantial amount of chlorogenic acid (5.81 ± 0.16 mg g−1 dmb), piceid (3.96 ± 0.04 mg g−1 dmb) and quercetin (6.06 ± 0.12 mg g−1 dmb) were quantified in SCL fermented by Bacillus species, roasted and steamed, respectively ( P < 0.05). The oxygen radical absorbance capacity ( ORAC) value was greater in microbial fermented SCL than in others, with the exception of Saccharomyces cerevisiae and Aspergillus oryzae. However, vitamin C equivalent antioxidant capacity ( VCEAC) was highest in SCL fermented by Aspergillus oryzae. Nobiletin and tangeretin are polymethoxy flavonoids (PMFs), found in rich quantities in the peel of citrus fruits. In the present study, we assessed the biological effect of the PMFs on liver damage using a mouse model of binge drinking. First, we extracted PMFs from the peels of Citrus aurantium to make Citrus aurantium extract (CAE). Male C57BL/6 mice were orally treated with silymarin and CAE (50, 100, and 200 mg/kg) for 3 days prior to ethanol (5 g/kg, total of 3 doses) oral gavage. Liver injury was observed in the ethanol alone group, as evidenced by increases in serum hepatic enzymes and histopathologic alteration, as well as by hepatic oxidative status disruption. CAE improved serum marker and hepatic structure and restored oxidative status by enhancing antioxidant enzyme levels and by reducing lipid peroxidation levels. In addition, CAE evidently suppressed inflammation and apoptosis in the livers of mice administered with ethanol, by 85% (tumor necrosis factor-α) and 44% compared to the control group, respectively. Furthermore, CAE activated lipid metabolism related signals and enhanced phosphorylation of AMP-activated protein kinase (AMPK) and nuclear factor E2-related factor 2 (Nrf2) with several cytoprotective proteins including heme oxygenase-1, NAD(P)H quinone oxidoreductase 1, and γ-glutamylcysteine synthetase. Taken together, the present study demonstrated that, CAE possesses antioxidant, anti-inflammatory, and antiapoptotic activity against ethanol-induced liver injury. [ABSTRACT FROM AUTHOR]

In addition, soyasaponins Aa and Ab suppressed the transcriptional activity of peroxisome proliferator-activated receptor γ (PPARγ) in HEK 293T cells. Furthermore, we confirmed that the expression of PPARγ and of CCAAT-enhancer-binding protein α (C/EBPα) was suppressed at both the mRNA and protein levels in 3T3-L1 adipocytes by treatment with soyasaponins Aa and Ab. Taken together, these findings indicate that soyasaponin Aa and Ab markedly inhibit adipocyte differentiation and expression of various adipogenic marker genes through the downregulation of the adipogenesis-related transcription factors PPARγ and C/EBPα in 3T3-L1 adipocytes. Glycemic variability comprises 'glucose variability' and 'hemoglobin A1c (HbA1c) variability.' Glucose variability relates to the within-day fluctuation of glucose level and may eventually reflect an increase in HbA1c level. Long-term glycemic variability, which is assessed by HbA1c fluctuation, reflects changes in glycemia over longer periods of time. Previous reports from several cohort studies have demonstrated that HbA1c variability is associated with the development or progression of microvascular complication in type 1 and 2 diabetes mellitus. However, data on the impact of HbA1c variability on macrovascular complications are controversial, and little is known about its impact on mortality. Further studies with a larger population including various ethnic groups are required to verify the impact of HbA1c variability on micro- and macrovascular complications of diabetes. Hypocotyl saponin composition of 1,198 accessions of wild soybean ( Glycine soja) collected from China, Korea, Japan and Russia Far East was analyzed by thin-layer chromatography to determine polymorphic variation and geographical distribution. Eight common distinguishable saponin phenotypes were identified: Aa, Ab, AaBc, AbBc, Aa+α, Ab+α, AaBc+α and AbBc+α. The latter four +α type were new. All eight types were identified in China. Type Ab+α was absent in Korea. Types Ab+α and AbBc+α, and Aa+α and Ab+α were not identified in Japan and Russia far east, respectively. Six new triterpene saponins were detected in +α type via LC-PDA/MS/MS analyses. They were, tentatively, designated as H-αg, H-αa, I-αg, I-αa, J-αg and J-αa. These saponins were inherited together by a single dominant allele. A gene symbol Sg- 6 was assigned. Hence, the new saponins were collectively named as Sg-6 saponins. The frequency of Sg- 6 allele was 17.6 % in Chinese, 10.0 % in Korean and 1.0 % in Japanese wild soybean. The wild soybeans having Sg-6 saponins can be utilized in soybean breeding programs as well as in saponin biosynthesis studies in soybean. [ABSTRACT FROM AUTHOR]
