Filtering processes across environmental gradients can structure patterns of trait variation within communities. The community‐weighted mean (CWM) is a metric that is commonly used to indicate the directionality of such filtering processes and the optimal adaptive strategy of taxa within community. Proximity to the CWM indicates higher fitness, and deviations from this optimal value result in changes in the relative abundances of coexisting species.We investigated patterns of intraspecific trait variation in four coexisting carrion beetle species (Coleoptera: Silphidae) across elevational gradients. The study was conducted in temperate forest ecosystems with distinct natural vegetation zones ranging from 950 m to 1700 m above sea level.Of the 12 traits measured, we found that intraspecific variation ranged from 34% (body length) to 100% (ratio of elytra length, head length and head width to body length) and accounted for a larger proportion of variation than interspecific variation in 7 traits. For most traits, trait range, which indicates the niche breadth of species at a given elevation, was positively correlated with relative abundance. The CWMs of traits associated with long‐distance dispersal decreased with elevation, whereas those associated with microhabitat use showed the opposite trend. Soil temperature influenced tibia length after controlling for the effects of species identity, but soil water content had no effect on trait variation. Patterns of variation in body width and thorax width of two beetle species supported the CWM‐optimality hypothesis; however, patterns of trait variation in body width and thorax length of another species showed an opposite trend.Our study identifies several traits that can be highly variable within carrion beetle species. The metacommunity theory enhances our understanding of how ecological processes regulate community structure. Yet, unraveling the complexities of soil nematode metacommunity structures across various spatial scales and determining the factors influencing these patterns remains challenging. Therefore, we conducted an investigation on soil nematode metacommunities spanning from north to south in the Northeastern China. Our aim was to test whether nematode metacommunities were structured by different drivers under three land covers (i.e., farmland, grassland and woodland) at the local and regional scales. The results revealed that the Clementsian, Gleasonian and their quasi‐structures of soil nematodes collectively accounted for 93% of the variation across the three land covers at the local and regional scales. These structures suggest that the soil nematode metacommunities in the Northeast China responded to fluctuations in environmental gradients. At the local scale, metacommunities were primarily shaped by biological interactions. At the regional scale, environmental heterogeneity, dispersal limitation and biological interactions all contributed to nematode metacommunities. Meanwhile, biological interactions under three land covers were represented within different trophic groups, with plant parasites predominant in farmlands and bacterivores in grasslands and woodlands. In conclusion, the metacommunity structures of soil nematodes remain stable at different spatial scales and land covers. Biological interactions are widespread among nematodes regardless of changes in spatial scales and land covers. This study reveals the importance of nematode sensitivity to the environment and biological interactions in shaping the nematode metacommunities, potentially enhancing our understanding of the spatial patterns of nematode metacommunities. Climate change will likely increase habitat loss of endemic tree species and drives forest conversion in mountainous forests. Elevation gradients provide the opportunity to predict possible consequences of such changes. While species compositions of various taxa have been investigated along elevation gradients, data on trophic changes in soil-dwelling organisms are scarce. Here, we investigated trophic changes of the Collembola communities along the northern slope of Changbai Mountain, China. We sampled Collembola in primary forests at seven elevations (800–1700 m asl). We measured individual body lengths and bulk stable isotopes on species level. We further categorized Collembola species into life forms. The community-weighted means of Δ15N and Δ13C values as well as minimum Δ15N values and isotopic uniqueness of Collembola communities increased with increasing elevation, while the range of Δ15N values decreased. Maximum and minimum of Δ13C values differed between elevations but showed no linear trend. Further, Δ15N values of Collembola species occurring across all elevations increased with elevation. Changes in Δ15N values with elevation were most pronounced in hemiedaphic species, while Δ13C values increased strongest with elevation in euedaphic species. Δ15N values increased with decreasing body size in hemiedaphic and euedaphic species. Overall, the results suggest that Collembola species functioning as primary decomposers at lower elevations shift towards functioning as secondary decomposers or even predators or scavengers at higher elevation forests. Mountain forests are at risk as the consequences of climate change will likely lead to altered tree species boundaries. Characterizing food webs along elevation gradients in primary forests may help to predict the potential consequences of such changes, for example with regard to the decomposition of dead organic matter. Here, for the first time, we studied trophic variations in two species‐rich microarthropod taxa, Collembola and Oribatida, along an elevation gradient of primary forest at Changbai Mountain, China. Samples were taken at seven elevations of 150‐m elevational difference between 800 and 1700 m. At each elevation, Collembola and Oribatida were extracted from litter samples of eight subplots. We applied three state‐of‐the‐art methods to elucidate trophic positions and basal resource use at community level: Bulk stable isotope analysis of nitrogen (Δ15Nbulk) and carbon (Δ13Cbulk), compound‐specific stable isotope analysis of amino acids (CSIA‐AA), and dietary routing of neutral lipid fatty acids (NLFAs). Trophic positions calculated using Δ15Nbulk and CSIA‐AA (TPCSIA) in both taxa increased similarly with elevation by about half and one third of a trophic position, respectively. Stable isotope mixing models and linear discriminant analysis bootstrapping using δ13C of essential amino acids indicated fungi as the most important resource at all elevations for both taxa. Also, proportions of marker NLFAs changed little across elevations in both taxa; overall high proportions of linoleic acid indicated high fungal contributions, but in Collembola the contribution of bacterial markers was generally higher than in Oribatida. Δ13Cbulk did not respond linearly to the elevation gradient; however, changes in elevation differed between Collembola and Oribatida. A strong linear relationship between δ15N of phenylalanine and δ15N of litter indicated litter as the basis of energy channels in both taxa. Overall, food web functioning likely changes with changing forest types along elevation gradients, with microarthropods switching from feeding closer to the base of the food web at lower elevations to feeding at higher trophic levels at higher elevations, potentially compromising their role in litter decomposition and nutrient cycling. Simple Summary: Global warming could affect Collembola and related decomposition processes within soil ecosystems in permafrost wetlands. Open top chambers (OTCs) were used to simulate climate warming in a cold temperate monsoon climate zone in the Great Hing'an Mountains of Northeast China. Collembola were captured using an aspirator after five years of simulated warming. We found that warming treatment increased the species richness and abundance of Collembola in most of the different seasons, except in May. Species composition differed significantly in the control and warming treatment in May and September. The Collembola species composition in permafrost wetlands was mainly determined by air humidity, indicating different responses of Collembola species to the indirect effect of warming on water availability. Mountain forests are important carbon stocks and biodiversity hotspots but are threatened by increased insect outbreaks and climate‐driven forest conversion. Soil microorganisms play an eminent role in nutrient cycling in forest habitats and form the basis of soil food webs. Uncovering the driving factors shaping microbial communities and functioning at mountainsides across the world is of eminent importance to better understand their dynamics at local and global scales. We investigated microbial communities and their climatic and local soil‐related drivers along an elevational gradient (800–1700 m asl) of primary forests at Changbai Mountain, China. We analyzed substrate‐induced respiration and phospholipid fatty acids (PLFA) in litter and two soil layers at seven sites. Microbial biomass (Cmic) peaked in the litter layer and increased towards higher elevations. In the litter layer, the increase in Cmic and in stress indicator ratios was negatively correlated with Ca concentrations indicating increased nutritional stress in high microbial biomass communities at sites with lower Ca availability. PLFA profiles in the litter layer separated low and high elevations, but this was less pronounced in soil, suggesting that the litter layer functions as a buffer for soil microbial communities. Annual variations in temperature correlated with PLFA profiles in all three layers, while annual variations in precipitation correlated with PLFA profiles in upper soil only. Furthermore, the availability of resources, soil moisture, Ca concentrations, and pH structured the microbial communities. Pronounced changes in Cmic and stress indicator ratios in the litter layer between pine‐dominated (800–1100 m) and spruce‐dominated (1250–1700 m) forests indicated a shift in the structure and functioning of microbial communities between forest types along the elevational gradient. The study highlights strong changes in microbial community structure and functioning along elevational gradients, but also shows that these changes and their driving factors vary between soil layers. Duck hepatitis A virus type 1 (DHAV-1) is a highly lethal virus that severely affects the duck industry worldwide. Long noncoding RNAs (lncRNAs) exert crucial roles in pathogen attacks. Here, we conducted deep transcriptome analysis to investigate the dynamic changes of host lncRNAs profiles in DHAV-1-infected duck embryo fibroblasts. We identified 16,589 lncRNAs in total and characterized their genomic features. Moreover, 772 and 616 differentially expressed lncRNAs (DELs) were screened at 12 and 24 h post-infection. Additionally, we predicted the DELs' cis- and trans-target genes and constructed lncRNA-target genes regulatory networks. Functional annotation analyses indicated that the putative target genes of DELs participated in diverse vital biological processed, including immune responses, cellular metabolism, and autophagy. For example, we confirmed the dysregulation of pattern recognition receptors (TLR3, RIG-I, MDA5, LGP2, cGAS), signal transducers (STAT1), transcription factors (IRF7), immune response mediators (IL6, IL10, TRIM25, TRIM35, TRIM60, IFITM1, IFITM3, IFITM5), and autophagy-related genes (ULK1, ULK2, EIF4EBP2) using RT-qPCR. Finally, we confirmed that one DHAV-1 induced lncRNA-XR_003496198 is likely to inhibit DHAV-1 replication in DEFs. Our study comprehensively analyzed the lncRNA profiles upon DHAV-1 infection and screened the target genes involved in the innate immune response and autophagy signaling pathway, thereby revealing the essential roles of duck lncRNAs and broadening our understanding of host-virus interactions. Aim: Both ecological and evolutionary processes shape biological communities along elevational gradients. Compared to above‐ground taxa, elevational patterns and processes of below‐ground animals are little studied. Here, we investigated how environmental gradients across elevation may affect species divergence in the past and act as filters of contemporary assembly of soil detritivores via traits. We asked: (1) Are environmental filtering processes due to elevation, vegetation or microhabitat‐related factors driving isotomid springtail community assembly across elevation; (2) did species divergences occur after mountain uplift; and (3) are there environmental factors related to species divergences in the past. Location: Changbai Mountain, north‐east China. Taxon Isotomidae, Collembola. Methods: We collected isotomid springtails from 800 to 2150 m above sea level and reconstructed the phylogeny using mitochondrial genome sequencing. We inspected the body length, number of ommatidia and pigmentation, and determined their evolutionary patterns. We then derived community parameters using trait‐based and metacommunity phylogenetic approaches and used linear mixed‐effects models to identify environmental variables that allow predicting community trait and phylogenetic parameters. Results: Isotomid springtails differed more in traits when co‐occurring in environments with lower soil nitrogen, typically at higher elevations. Most communities exhibited phylogenetic clustering, but this pattern was not related to elevation or any environmental variables. These isotomid species mainly diverged in the Mesozoic with some divergence events associated with soil N and pH, as well as other factors covarying with the contemporary elevational gradient studied. The current communities comprised old phylogenetic lineages possessing traits that have undergone evolutionary drift. Main conclusions: While environmental gradients act as filters for below‐ground detritivores, evolution of traits preconditions their assembly. Species divergence likely resulted from filtering processes of past environments resembling the present day. Altitudinal changes in the diversity of plants and animals have been well documented; however, soil animals received little attention in this context and it is unclear whether their diversity follows general altitudinal distribution patterns. Changbai Mountain is one of few well‐conserved mountain regions comprising natural ecosystems on the Eurasian continent. Here, we present a comprehensive analysis of the diversity and community composition of Collembola along ten altitudinal sites representing five vegetation types from forest to alpine tundra. Among 7834 Collembola individuals, 84 morphospecies were identified. Species richness varied marginally significant with altitude and generally followed a unimodal relationship with altitude. By contrast, the density of Collembola did not change in a consistent way with altitude. Collembola communities changed gradually with altitude, with local habitat‐related factors (soil and litter carbon‐to‐nitrogen ratio, litter carbon content, and soil pH) and climatic variables (precipitation seasonality) identified as major drivers of changes in Collembola community composition. Notably, local habitat‐related factors explained more variation in Collembola assemblages than climatic variables. The results suggest that local habitat‐related factors including precipitation and temperature are the main drivers of changes in Collembola communities with altitude. Specifically, soil and litter carbon‐to‐nitrogen ratio correlated positively with Collembola communities at high altitudes, whereas soil pH correlated positively at low altitudes. This documents that altitudinal gradients provide unique opportunities for identifying factors driving the community composition of not only above‐ but also belowground invertebrates. Duck hepatitis A virus 1 (DHAV-1) is a highly contagious etiological agent that causes acute hepatitis in young ducklings. MicroRNAs (miRNAs) play important regulatory roles in response to pathogens. However, the interplay between DHAV-1 infection and miRNAs remains ambiguous. We characterized and compared miRNA and mRNA expression profiles in duck embryo fibroblasts cells (DEFs) infected with DHAV-1. In total, 36 and 96 differentially expressed (DE) miRNAs, and 4110 and 2595 DE mRNAs, were identified at 12 and 24 h after infection. In particular, 126 and 275 miRNA–mRNA pairs with a negative correlation were chosen to construct an interaction network. Subsequently, we identified the functional annotation of DE mRNAs and target genes of DE miRNAs enriched in diverse Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, which may be important for virus resistance, cell proliferation, and metabolism. Moreover, upregulated miR-222a could negatively regulate DHAV-1 replication in DEFs and downregulate integrin subunit beta 3 (ITGB3) expression by targeting the 3′ untranslated region (3′UTR), indicating that miR-222a may modulate DHAV-1 replication via interaction with ITGB3. In conclusion, the results reveal changes of mRNAs and miRNAs during DHAV-1 infection and suggest miR-222a as an antiviral factor against DHAV-1. [ABSTRACT FROM AUTHOR]

Duck hepatitis A virus (DHAV) causes a highly contagious and acute disease in ducklings younger than 3 weeks of age and spreads rapidly by horizontal transmission to all susceptible ducklings in the flock. To date, there is no evidence of vertical transmission of DHAV‐1. In a previous study, we identified a novel DHAV type 1 (DHAV‐1) isolate that could infect adult ducks and induce laying drop. In this study, 30 non‐embryonated duck eggs and 60 17‐day‐old embryos were collected from three breeding duck flocks with egg drop syndrome caused by DHAV‐1 in China, and 30 17‐day‐old embryos were randomly selected from the 60 embryos and allowed to hatch. DHAV‐1 RNA was detected by RT‐PCR in 10 of 30 non‐embryonated eggs, 9 of 30 17‐day‐old embryos, 5 of 7 dead embryos and 5 of 23 newly hatched ducklings. Overall, 29 of 90 (32.2%) eggs and embryos were positive for DHAV‐1. Three DHAV‐1 strains were isolated from the dead duck embryos of the three breeding duck flocks, respectively. Pathogenicity studies showed that the three DHAV‐1 isolates had median embryo lethal doses but were highly pathogenic to healthy ducklings. Compared with the DHAV reference strains, there were two specific amino acid mutation sites (F169 and S220) in VP1 of the three isolates. To the best of our knowledge, this is the first report that DHAV‐1 is isolated from duck embryos. Waterfowl parvoviruses (WPVs) including goose parvovirus (GPV), novel GPV-related virus (NGPV) and Muscovy duck parvovirus (MDPV) cause significant economic losses and epizootic threat to the waterfowl industries, and little is known about the B-cell epitopes of WPVs. In this study, a monoclonal antibody (mAb) 5B5 against the VP3 protein of NGPV was used to identify the possible epitope in the three kinds of WPVs. The mAb 5B5 had neutralizing activities to the three viruses, and reacted with the conserved linear B-cell epitopes of 438LHNPPP443 in VP3 protein of GPV, NGPV and MDPV. To the authors' best knowledge, this is the first report on identification of the common conserved neutralizing linear B-cell epitope on VP3 protein of three different WPVs, which would facilitate the development of a novel immunodiagnostic assay for rapid detection of WPV infection. Collembola are a basal group of Hexapoda renowned for both unique morphological characters and significant ecological roles. However, a robust and plausible phylogenetic relationship between its deeply divergent lineages has yet to be achieved. We carried out a mitophylogenomic study based on a so far the most comprehensive mitochondrial genome dataset. Our data matrix contained mitogenomes of 31 species from almost all major families of all four orders, with 16 mitogenomes newly sequenced and annotated. We compared the linear arrangements of genes along mitochondria across species. Then we conducted 13 analyses each under a different combination of character coding, partitioning scheme and heterotachy models, and assessed their performance in phylogenetic inference. Several hypothetical tree topologies were also tested. Mitogenomic structure comparison revealed that most species share the same gene order of putative ancestral pancrustacean pattern, while seven species from Onychiuridae, Poduridae and Symphypleona bear different levels of gene rearrangements, indicating phylogenetic signals. Tomoceroidea was robustly recovered for the first time in the presence of all its families and subfamilies. Monophyly of Onychiuroidea was supported using unpartitioned models alleviating LBA. Paronellidae was revealed polyphyletic with two subfamilies inserted independently into Entomobryidae. Although Entomobryomorpha has not been well supported, more than half of the analyses obtained convincing topologies by placing Tomoceroidea within or near remaining Entomobryomorpha. The relationship between elongate-shaped and spherical-shaped collembolans still remained ambiguous, but Neelipleona tend to occupy the basal position in most trees. This study showed that mitochondrial genomes could provide important information for reconstructing the relationships among Collembola when suitable analytical approaches are implemented. Of all the data refining and model selecting schemes used in this study, the combination of nucleotide sequences, partitioning model and exclusion of third codon positions performed better in generating more reliable tree topology and higher node supports than others. microRNAs (miRNAs) perform critical roles in various biological and metabolic processes by regulating gene expression at the post-transcriptional level. To investigate the functional roles of miRNAs in the lactating mammary gland of Capra hircus, a library was constructed from the lactating mammary glands of Laoshan dairy goats ( C. hircus) during early lactation. The miRNA expression profiles were systematically screened, and miRNAs were identified and characterized using Solexa deep-sequencing technology and bioinformatics. As a result, a total of 18,031,615 clean reads were obtained representing 305,711 unique sRNAs. A total of 12,086,616 sRNAs representing 3,701 unique sRNAs matched the known Bos taurus miRNA precursors in miRBase 17.0, and 300 known miRNAs and 15 miRNA* were discovered. In addition, 131 novel miRNAs sequences were also obtained, and 147,703 putative targets were predicted. GO and KEGG pathway analysis showed that the majority of targets were involved in cellular processes and metabolic pathways. The 290 known miRNAs, 14 miRNA* and 38 novel miRNAs were validated by sequencing a second library that was constructed from the same tissues as the first library. Our study provided the first large-scale identification and characterization of miRNAs in the mammary gland tissue of the dairy goat. The results indicate that the regulation of miRNA-mediated gene expression occurs during early lactation in dairy goats. This study significantly enriches the C. hircus miRNA repertoire and provides a reference for the elucidation of complex miRNA-mediated regulatory networks for gene expression in the physiology and developmental progression of the lactating mammary gland. Objectives Previous studies have reported several different plasmids that confer multidrug resistance (MDR) including resistance to aminoglycosides. In this study, we investigated the aminoglycoside resistance patterns for 224 Escherichia coli isolates from diseased chickens and ducks in China, characterized a novel MDR plasmid, and collected prevalence data on similar resistance plasmids. Methods Antibiotic susceptibilities were determined using disc diffusion and the microdilution method. The plasmid pXZ was analysed by restriction fragment length polymorphism (RFLP) with EcoRI and SalI, and sequenced. The prevalence of similar resistance plasmids was assessed by multiplex PCR and by RFLP analysis. Results Among the 224 E. coli isolates, 189 (84.4%) were resistant to streptomycin, 125 (55.8%) were resistant to kanamycin, 116 (51.8%) were resistant to gentamicin, 106 (47.3%) were resistant to neomycin and 98 (43.8%) were resistant to amikacin. Among the 224 E. coli isolates, 17 contained a plasmid with the MDR-encoding region of pXZ, which showed high-level resistance to aminoglycosides (MICs of gentamicin and amikacin ≥512 mg/L). The plasmid pXZ was digested into five fragments by EcoRI and six fragments by SalI. The plasmid pXZ was a circular DNA molecule of 76 635 bp with a 51.65% guanine + cytosine content and included four resistance genes (rmtB, fosA3, blaTEM-1 and blaCTX-M-24). Conclusions A novel MDR plasmid, pXZ, harbouring four resistance genes (rmtB, fosA3, blaTEM-1 and blaCTX-M) was identified. With the continuous development of duck farming and the increasing breeding density, the incidence of duck hepatitis A virus type 1 (DHAV-1) has been on the rise, seriously endangering the development of duck farming. To reduce the use of antibiotics in duck breeding, susceptibility risks and mortality, and avoid virulence recovery and immune failure risk, this study aims to develop a new type of mucosal immune probiotics and make full use of molecular biology techniques, on the level of genetic engineering, to modify Lactococcus lactis (L. lactis). In this study, a secretory recombinant L. lactis named MG1363-VP1 with an enhanced Green Fluorescent Protein (eGFP) and translation enhancer T7g10L was constructed, which could express the VP1-eGFP fusion protein of DHAV-1. The animal experiment in ducklings was performed to detect the immune response and protection effect of oral microecologics by recombinant L. lactis. The results showed that oral L. lactis MG1363-VP1 significantly induced the body's humoral immune system and mucosal immune system to produce specific anti-VP1 IgG antibodies and mucosal secretory immunoglobulin A (sIgA) for DHAV-1 in ducklings, and cytokines including interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10), and interferon gamma (IFN-γ). The mortality rate was monitored simultaneously by the natural infestation in the process of production and breeding; notably, the ducklings vaccinated with L. lactis MG1363-VP1 were effectively protected against the nature infection of DHAV-1. Simple Summary: Colistin has been used as a growth promotant in livestock feed for many years. To date, there are few reports about the prevalence and molecular characteristics of fecal Escherichia coli bearing mcr-1 in the meat ducks. In this study, among 120 fecal Escherichia coli strains isolated from healthy meat ducks, a total of nine mcr-1-containing E. coli strains were identified and two were identified as extra-intestinal pathogenic E. coli. The 9 mcr-1-bearing E. coli isolates were clonally unrelated, carried two different genetic contexts of mcr-1, and the colistin-resistant phenotype of them was successfully transferred to the recipient strains. These results highlight that healthy meat duck is a potential reservoir for multidrug resistant mcr-1-containing E. coli strains. To date, there are few reports about the prevalence and molecular characteristics of fecal E. coli bearing mcr-1 in the meat ducks. In this study, the prevalence of mcr-1 gene was investigated among 120 fecal E. coli strains isolated from healthy meat ducks in Shandong province of China between October 2017 and February 2018. A total of nine mcr-1-containing E. coli strains were identified and two were identified as extra-intestinal pathogenic E. coli (ExPEC) among them. The clonal relationship of the nine E. coli strains was determined by multilocus sequencing typing (MLST) and pulsed field gel electrophoresis (PFGE), and the results indicated that all mcr-1-carrying isolates were clonally unrelated. Two different genetic contexts of mcr-1 were identified among these isolates. Colistin-resistant phenotype of all the isolates was successfully transferred to the recipient strains by conjugation experiments and seven transconjugants carried a single plasmid. The mcr-1 was located on three replicon plasmids: IncI2 (n = 4), IncFII (n = 2) and IncN (n = 1). Complete sequence analysis of a representative plasmid pTA9 revealed that it was strikingly similar with plasmid pMCR1-IncI2 of E. coli, plasmid pHNSHP45 of E. coli, and plasmid pWF-5-19C of Cronobacter sakazakii, implying that pTA9-like plasmids may be epidemic plasmids that mediate the spread of mcr-1 among Enterobacteriaceae. Duck hepatitis A virus (DHAV), the major pathogen of duck virus hepatitis (DVH), causes severe diseases that threaten the duck industry worldwide. The VP1 protein, a major structural protein of DHAV, is able to induce neutralizing antibody in ducks. The purpose of this study was to identify the antigenic mimotope of DHAV by phage display technology. A monoclonal antibody (mAb) 4E6 against DHAV-1 and DHAV-3 was prepared, and a phage library prepared with the PhD-12 Phage Display Peptide Library Kit was screened with the mAb. A novel peptide, 1GLTWKLPPSM10 was identified with high affinity to the mAb and could specifically block mAb 4E6 from binding DHAV-1 and DHAV-3. Animal tests confirmed that the immunization of ducklings with the mimotope could inhibit the virus proliferation and protect the ducklings from DVH. In summary, the neutralizing conformational mimotope 1GLTWKLPPSM10 might be a promising vaccine candidate for the prevention of DHAV infection. [ABSTRACT FROM AUTHOR]

Autophagy is a tightly regulated catabolic process and is activated in cells in response to stress signals. Despite extensive study, the interplay between duck hepatitis A virus type 1 (DHAV-1) and the autophagy of host cells is not clear. In this study, we applied proteomics analysis to investigate the interaction mechanism between DHAV-1 and duck embryo fibroblast (DEF) cells. In total, 507 differentially expressed proteins (DEPs) were identified, with 171 upregulated proteins and 336 downregulated proteins. The protein expression level of heat shock proteins (Hsps) and their response to stimulus proteins and zinc finger proteins (ZFPs) were significantly increased while the same aspects of ribosome proteins declined. Bioinformatics analysis indicated that DEPs were mainly involved in the "response to stimulus", the "defense response to virus", and the "phagosome pathway". Furthermore, Western blot results showed that the conversion of microtubule-associated protein 1 light chain 3-I (LC3-I) to the lipidation form of LC3-II increased, and the conversion rate decreased when DEF cells were processed with 4-phenylbutyrate (4-PBA). These findings indicated that DHAV-1 infection could cause endoplasmic reticulum (ER) stress-induced autophagy in DEF cells, and that ER stress was an important regulatory factor in the activation of autophagy. Our data provide a new clue regarding the host cell response to DHAV-1 and identify proteins involved in the DHAV-1 infection process or the ER stress-induced autophagy process. Background: Reverse genetics systems enable the manipulation of viral genomes and therefore serve as robust reverse genetic tools to study RNA viruses. A DNA-launched rescue system initiates the transcription of viral genomic cDNA from eukaryotic promoter in transfected cells, generating homogenous RNA transcripts in vitro and thus enhancing virus rescue efficiency. As one of the hazardous pathogens to ducklings, the current knowledge of the pathogenesis of duck astrovirus type 1 (DAstV-1) is limited. The construction of a DNA-launched rescue system can help to accelerate the study of the virus pathogenesis. However, there is no report of such a system for DAstV-1. Methods: In this study, a DNA-launched infectious clone of DAstV-1 was constructed from a cDNA plasmid, which contains a viral cDNA sequence flanked by hammerhead ribozyme (HamRz) and a hepatitis delta virus ribozyme (HdvRz) sequence at both terminals of the viral genome. A silent nucleotide mutation creating a Bgl II site in the ORF2 gene was made to distinguish the rescued virus (rDAstV-1) from the parental virus (pDAstV-1). Immunofluorescence assay (IFA) and western blot were conducted for rescued virus identification in duck embryo fibroblast (DEF) cells pre-treated with trypsin. The growth characteristics of rDAstV-1 and pDAstV-1 in DEF cells and the tissue tropism in 2-day-old ducklings of rDAstV-1 and pDAstV-1 were determined. Results: The infectious DAstV-1 was successfully rescued from baby hamster kidney (BHK-21) cells and could propagate in DEF cells pre-treated with 1 μg/ml trypsin. Upon infection of DEF cells pre-treated with trypsin, DAstV-1 mRNA copies were identified after serial passaging, and the result showed that rDAstV-1 and pDAstV-1 shared similar replication kinetics. Animal experiment showed that the rDAstV-1 had an extensive tissue tropism, and the virus was capable of invading both the central and the peripheral immune organs in infected ducklings. Conclusions: An improved DNA-launched reverse genetics system for DAstV-1 was firstly constructed. Infectious virus recovered from BHK-21 cells could propagate in DEF cells pre-treated with trypsin. This is the first report of the successful in vitro cultivation of DAstV-1.
