The oriental armyworm, Mythimna separata (Walker, 1865) (Lepidoptera: Noctuidae), is a serious global migratory insect pest of grain crops. Although its migratory biology has been studied for a long history, the factors affecting wingbeat frequency (WBF), which is closely related to the flight activity of the insect, remain unclear. In this study, the WBFs of both cultured and migrating moths were tested under different conditions in the laboratory using a stroboscope. The results indicated that age and mating status significantly influenced WBF. One day old adults had the lowest WBF, and unmated females had a significantly higher WBF than that of mated females. In general, the WBF of males was significantly higher than that of female individuals. The WBF decreased gradually with increasing environmental humidity, and WBF had a significant negative binomial regression relationship with temperature change. The WBF of moths that fed on hydromel was much higher than those of the controls that fed on water or without diet. However, wind speed and air pressure had no significant effects on the moth WBF in the test environments. These findings provide a deeper understanding of factors that affect flight ability in M. separata , which will be helpful for developing a regional migratory monitoring and warning system of the pest, such as identifying target insect species based on the WBF from radar observation. The codling moth Cydia pomonella is a major pest of global significance impacting pome fruits and walnuts. It threatens the apple industry in the Loess Plateau and Bohai Bay in China. Sterile insect technique (SIT) could overcome the limitations set by environmentally compatible area-wide integrated pest management (AW-IPM) approaches such as mating disruption and attract-kill that are difficult to suppress in a high-density pest population, as well as the development of insecticide resistance. In this study, we investigated the effects of X-ray irradiation (183, 366, 549 Gy) on the fecundity and fertility of a laboratory strain of C. pomonella , using a newly developed irradiator, to evaluate the possibility of X-rays as a replacement for Cobalt60 (60Co- γ) and the expanded future role of this approach in codling moth control. Results show that the 8th-day is the optimal age for irradiation of male pupae. The fecundity decreased significantly as the dosage of radiation increased. The mating ratio and mating number were not influenced. However, treated females were sub-sterile at a radiation dose of 183 Gy (20.93%), and were almost 100% sterile at a radiation dose of 366 Gy or higher. Although exposure to a radiation dose of 366 Gy resulted in a significant reduction in the mating competitiveness of male moths, our radiation biology results suggest that this new generation of X-ray irradiator has potential applications in SIT programs for future codling moth control. Simple Summary: The invasive pest Spodoptera frugiperda has become a global problem. Sterile insect technology is an effective means to prevent invasive insects, but at present, the research on this pest is not perfect. Our results showed that egg sterility in offspring reached 74% when the ratio of those given a 250 Gy dose of X-ray radiation to the non-irradiated males was 12:1, and there was no significant difference in mating competitiveness between irradiated males of different ages. Additional cornfield cage studies confirmed this finding by showing that release ratios from 12:1 to 20:1 significantly reduced larval populations in the field, with a 48 to 69% leaf protection effect and a 58 to 83% insect population reduction. This study provides fundamental information for the management of S. frugiperda through the use of sterile insect techniques. Spodoptera frugiperda, an invasive pest, has a huge impact on food production in Asia and Africa. The potential and advantages of sterile insect techniques for the permanent control of S. frugiperda have been demonstrated, but the methods for their field application are still unavailable. For the purposes of this study, male pupae of S. frugiperda were irradiated with an X-ray dose of 250 Gy to examine the effects of both the release ratio and the age of the irradiated males on the sterility of their offspring. The control effect of the irradiated male release ratio on S. frugiperda was evaluated using field-cage experiments in a cornfield. The results showed that when the ratio of irradiated males to non-irradiated males reached 12:1, the egg-hatching rate of the offspring of S. frugiperda decreased to less than 26%, and there was also no significant difference in mating competitiveness among the different ages. Field-cage testing showed that when irradiated males were released at ratios of 12:1–20:1 to normal males, the leaf protection effect for the corn reached 48–69% and the reduction in the insect population reached 58–83%. In this study, an appropriate release ratio is suggested, and the mating competitiveness of irradiated and non-irradiated males of S. frugiperda is investigated, thus providing a theoretical basis for the use of sterile insect techniques to control S. frugiperda. Background: The fall armyworm (FAW) Spodoptera frugiperda Smith (Lepidoptera: Noctuidae) invaded Myanmar and China in 2018 and greatly impacted agricultural production and ecosystem balance in these areas. FAW is a migratory insect, but its seasonal migration pattern between the two countries has been largely unknown. From 2019 to 2021, we monitored the seasonal migration of FAW in the China–Myanmar border area using a searchlight trap, assessed the reproductive development status of female migrants and traced the migratory routes by trajectory simulation. Results: FAW moths were trapped by the searchlight trap in Lancang County (Yunnan, China) all year, with obvious seasonal differences in the number caught. There were small‐scale persistent trapping peaks in spring and summer, and obvious peaks in autumn; only a small number of moths were trapped in winter. Examination of the ovaries of female moths collected in different seasons showed that most females had matured, indicating that the moths were migrating and did not take off from the local area. In the migration trajectory simulation, FAW mainly migrated from Myanmar to Southwest China in spring and summer and back to Myanmar in autumn. Conclusion: Our findings indicate that FAW migrates between China and Myanmar according to the monsoon circulation, which will help guide cross‐border regional monitoring and management strategies against this pest. © 2022 Society of Chemical Industry. BACKGROUND: Spodoptera frugiperda has spread to Africa, Asia, and Oceania, posing a serious threat to global agriculture. We estimated the appropriate dose of X‐ray sterilization for S. frugiperda using an X‐ray irradiation instrument to investigate environmentally acceptable control techniques, laying the framework for future applications of sterile insect technology (SIT) to manage the pest environmentally‐friendly. RESULTS: This study is the first to investigate the effects of X‐ray irradiation on the growth, development, survival, reproduction, and flight of S. frugiperda. The results showed that irradiation with 50–400 Gy had no significant effect on pupal eclosion, but females were more sensitive than males in terms of reproductive parameters, especially when doses of radiation were > 350 Gy. After irradiation with a sub‐sterilizing dose of 250 Gy, the parental sterility rate was > 85%, and the sterility traits could be passed on to their offspring, resulting in a continuous decrease in the population of F1 and F2 generations. CONCLUSION: Our laboratory experiments theoretically confirmed the feasibility of SIT for controlling S. frugiperda in the field using X‐ray radiation. The fall armyworm Spodoptera frugiperda, an invasive insect pest native to the Americas, has established populations throughout eastern China. The North China Plain—a key corn‐producing area in East China with a unique topography—was invaded by fall armyworm in 2019 and is seriously threatened by this migratory pest. However, the spatiotemporal extent of the migratory movements of fall armyworm from the North China Plain remains poorly understood. Using an air transport‐based trajectory modeling approach that incorporates flight behavior, we simulated the potential nocturnal migration trajectories of fall armyworm from the North China Plain based on historical meteorological data from June to October of 2015–2019, and examined the night‐time atmospheric conditions associated with their possible flights. The emigration patterns showed monthly variation in the main landing area and common migration direction. The displacement of newly emerged moths from the North China Plain was concentrated in the Northeast China Plain (including Liaoning, Jilin and Heilongjiang provinces) before late summer, after which they were most likely to undertake return flights to the south (especially into Hubei, Anhui and Hunan provinces). This southwest‐northeast aerial migration corridor follows the topography of East China and is affected by the East Asian monsoon. These topographic‐atmospheric conditions have resulted in the North China Plain becoming a key stopover for fall armyworm populations engaging in multigenerational long‐distance migration across East China. These findings contribute to our knowledge of fall armyworm migration and will aid in the implementation of management and control strategies against this highly migratory agricultural pest. Although hepatocellular carcinoma (HCC) is one of the deadliest health burdens worldwide, few drugs are available for its clinical treatment. However, in recent years, major breakthroughs have been made in the development of new drugs due to intensive fundamental research and numerous clinical trials in HCC. Traditional systemic therapy schemes and emerging immunotherapy strategies have both advanced. Between 2017 and 2020, the United States Food and Drug Administration (FDA) approved a variety of drugs for the treatment of HCC, including multikinase inhibitors (regorafenib, lenvatinib, cabozantinib, and ramucirumab), immune checkpoint inhibitors (nivolumab and pembrolizumab), and bevacizumab combined with atezolizumab. Currently, there are more than 1000 ongoing clinical trials involving HCC, which represents a vibrant atmosphere in the HCC drug research and development field. Additionally, traditional Chinese medicine approaches are being gradually optimized. This review summarizes FDA-approved agents for HCC, elucidates promising agents evaluated in clinical phase I/II/III trials and identifies emerging targets for HCC treatment. In addition, we introduce the development of HCC drugs in China. Finally, we discuss potential problems in HCC drug therapy and possible future solutions and indicate future directions for the development of drugs for HCC treatment. [ABSTRACT FROM AUTHOR]

Aphidophagous syrphids (Diptera: Syrphidae) are important insects in agroecosystems for pollination and biological control. Insect chemoreception is essential for these processes and for insect survival and reproduction; however, molecular determinants is not well understood for these beneficial insects. Here, we used recent transcriptome data for the common hoverfly, Episyrphus balteatus , to characterize key molecular components of chemoreception: odorant-binding proteins (OBPs) and chemosensory proteins (CSPs). Six EbalCSPs and 44 EbalOBPs were cloned from this species, and sequence analysis showed that most share the characteristic hallmarks of their protein family, including a signal peptide and conserved cysteine signature. Some regular patterns and key conserved motifs of OBPs and CSPs in Diptera were identified using the online tool MEME. Motifs were also compared among the three OBP subgroups. Quantitative real-time PCR (qRT-PCR) showed that most of these chemosensory genes were expressed in chemosensory organs, suggesting these genes have chemoreceptive functions. An overall comparison of the Ka/Ks values of orthologous genes in E. balteatus and another predatory hoverfly species to analyze the evolution of these olfactory genes showed that OBPs and CSPs are under strong purifying selection. Overall, our results provide a molecular basis for further exploring the chemosensory mechanisms of E. balteatus , and consequently, may help us to understand the tritrophic interactions among plants, herbivorous insects, and natural enemies. Helicoverpa armigera, cotton bollworm, is one of the most disastrous pests worldwide, threatening various food and economic crops. Functional genomic tools may provide efficient approaches for its management. The clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR‐associated protein 9 (Cas9) system, dependent on a single guide RNA (sgRNA), has been used to induce indels for targeted mutagenesis in cotton bollworm. However, genomic deletions may be more desirable to disrupt the function of noncoding genes or regulatory sequences. By injecting two sgRNAs with Cas9 protein targeting different exons, we obtained predictable genomic deletions of several hundred bases. We achieved this type of modification with different combinations of sgRNA pairs, including HaCad and HaABCC2. Our finding indicated that CRISPR/Cas9 can be used as an efficient tool to engineer genomes with chromosomal deletion in H. armigera. [ABSTRACT FROM AUTHOR]

Since the foundation of The People's Republic of China 70 years ago, continuous improvements in the field of agricultural science and technology have led to significant progress in agricultural pest control strategies, which have been upgraded from primary agronomic measures in the 1950's to the current more efficient, scientific and environmental-friendly, green pest-management strategies. Chinese agricultural entomologists have successively clarified the outbreak principles of the major pests of rice, wheat, corn, cotton, vegetables and fruit crops, and have also innovated a number of monitoring, forecasting, biological and physical control, and eco-chemical regulation, technologies. They have also developed resistance management strategies, and insect-resistant crop breeding techniques, and have constructed a comprehensive control technology system for both important crops and major pests. Pest management models mainly driven by agronomic measures, source population monitoring and management, chemical pesticides, biological measures, and transgenic technology, have been used to control Locusta migratoria, Mythimna separata, the rice planthopper, Ostrinia furnacalis and Helicoverpa armigera, respectively, which have now become classic examples of agricultural pest control strategies worldwide. The rapid development of information technology, artificial intelligence, agricultural biotechnology, genome technology and other high throughput technologies, will promote continuous innovation in the fields of intelligent pest identification and forecasting, genetic diagnosis and rapid detection, targeted pesticide application, pest population genetics management, regional ecological control and transgenic technology. Many insects such as the crop pest Agrotis ipsilon (black cutworm) migrate seasonally to exploit suitable breeding and winter habitats. The migration of a crop pest also increases the risk of sudden outbreaks. Knowledge about insect migration thus is important for developing a forecasting and warning system for migratory pests. Here we used directional flight devices to study the orientation behavior of A. ipsilon in China during the northward and southward migratory periods and showed that the insects during each period exhibited a common cardinal orientation. Northward migratory moths were oriented at an orientation angle mean of 24.4° and the southward at 205.17°. Expression levels of cryptochrome and magnetoreceptor genes in migratory populations were higher than in a reared population and higher in the southward migratory population than in the northward. Wide planting of transgenic Bt cotton in China since 1997 to control cotton bollworm ( Helicoverpa armigera) has increased yields and decreased insecticide use, but the evolution of resistance to Bt cotton by H. armigera remains a challenge. Toward developing a new generation of insect-resistant transgenic crops, a chimeric protein of Vip3Aa1 and Vip3Ac1, named Vip3AcAa, having a broader insecticidal spectrum, was specifically created previously in our laboratory. In this study, we investigated cross resistance and interactions between Vip3AcAa and Cry1Ac with three H. armigera strains, one that is susceptible and two that are Cry1Ac-resistant, to determine if Vip3AcAa is a good candidate for development the pyramid cotton with Cry1Ac toxin. Our results showed that evolution of insect resistance to Cry1Ac toxin did not influence the sensitivity of Cry1Ac-resistant strains to Vip3AcAa. For the strains examined, observed mortality was equivalent to the expected mortality for all the combinations of Vip3AcAa and Cry1Ac tested, reflecting independent activity between these two toxins. When this chimeric vip3AcAa gene and the cry1Ac gene were introduced into cotton, mortality rates of Cry1Ac resistant H. armigera larvae strains that fed on this new cotton increased significantly compared with larvae fed on non-Bt cotton and cotton producing only Cry1Ac. Aphis gossypii Glover (Hemiptera: Aphididae) is a key pest in cotton crops, notably owing to its increasing resistance to commonly used pesticides. Such resistance prompts for the development of integrated pest management (IPM) programs that include novel pesticides being effective against the aphid. In the present study, we assessed lethal and sublethal effects of cycloxaprid, a novel chiral neonicotinoid pesticide developed in China, on A. gossypii. The lethal concentration at 50% (LC50) value of cycloxaprid on A. gossypii was estimated, using the dipping method, at 7.73 mg/L. The impact of a sublethal concentration (LC10) and a lethal concentration (LC40) of cycloxaprid on A. gossypii population growth and feeding behavior (using electrical penetration graph technique [EPG]), and its transgenerational effect were further assessed. Adult longevity and fecundity significantly decreased after exposure to LC40 or LC10 of cycloxaprid. Cycloxaprid with sublethal concentrations (especially LC40) had negative effects on phloem ingestion by A. gossypii. Additionally, the offspring of the adults exposed to LC40 of cycloxaprid had shorter nymphal development duration and adult longevity than the control, and those from LC10 and LC40 treatments had lower adult fecundity and net productive rate. We demonstrated that cycloxaprid is a pesticide showing both lethal and sublethal activities, and transgenerational effects on A. gossypii; it may be useful for implementation in IPM programs against this aphid pest. In northern China, due to the large-scale adoption of transgenic Bacillus thuringiensis Berliner (Bt) cotton, the number of Adelphocoris lineolatus ( Goeze) ( Hemiptera: Miridae) has increased quickly, causing significant loss in cotton and alfalfa production. One of the environmentally safe strategies without use of pesticides is the application of insect pheromone for mating disruption. In our study, we aim to identify the active components in sexually mature virgin female A. lineolatus, and activity and optimal ratio of these components. By using gas chromatography-mass spectrometry ( GC- MS) and gas chromatography-electroantennographic detection ( GC- EAD), we identified three active compounds: hexyl butyrate ( HB), ( E)-2-hexenyl butyrate (E2 HB), and ( E)-4-oxo-2-hexenal (4- OHE). We examined the release rate of septum and tube dispensers in a wind tunnel. In a field experiment, deletion of 4- OHE or E2 HB resulted in significant suppression of male trap catches, indicating that E2 HB or 4- OHE may be the active pheromone components. Traps baited with a blend of 4- OHE and E2 HB at 2:5, 3:4, and 4:3 caught significantly more males, suggesting that the optimal ratio of 4- OHE and E2 HB could be in the range of 1:1 to 1:2. The addition of a large amount of HB could strongly reduce the attractive activity of both virgin females and artificial lures. We also compared the attractiveness of septum lures and tube lures in field experiments. The septum lures attracted males in the first 3 days. The tube lures caught more males after 3 days and the attractive effects existed up to 5 weeks, suggesting them as a tool for long-term monitoring and control of plant bugs. [ABSTRACT FROM AUTHOR]

Insects use their sensitive and selective olfactory system to detect outside chemical odorants, such as female sex pheromones and host plant volatiles. Several groups of olfactory proteins participate in the odorant detection process, including odorant binding proteins (OBPs), chemosensory proteins (CSPs), odorant receptors (ORs), ionotropic receptors (IRs) and sensory neuron membrane proteins (SNMPs). The identification and functional characterization of these olfactory proteins will enhance our knowledge of the molecular basis of insect chemoreception. In this study, we report the identification and differential expression profiles of these olfactory genes in the black cutworm moth Agrotis ipsilon. In total, 33 OBPs, 12 CSPs, 42 ORs, 24 IRs, 2 SNMPs and 1 gustatory receptor (GR) were annotated from the A. ipsilon antennal transcriptomes, and further RT-PCR and RT-qPCR revealed that 22 OBPs, 3 CSPs, 35 ORs, 14 IRs and the 2 SNMPs are uniquely or primarily expressed in the male and female antennae. Furthermore, one OBP (AipsOBP6) and one CSP (AipsCSP2) were exclusively expressed in the female sex pheromone gland. These antennae-enriched OBPs, CSPs, ORs, IRs and SNMPs were suggested to be responsible for pheromone and general odorant detection and thus could be meaningful target genes for us to study their biological functions in vivo and in vitro. In assessing an insect-resistant genetically engineered (IRGE) crop before its commercialization, researchers normally use so-called 'Tier-1 assays' as the initial step to determine the effects of the crop on non-target organisms. In these tests, the insecticidal proteins (IPs) produced by the IRGEs are added to the diets of test organisms in the laboratory. Test organisms in such assays can be directly exposed to much higher concentrations of the test IPs than they would encounter in the field. The results of Tier-1 assays are thus more conservative than those generated in studies in which the organisms are exposed to the IPs by feeding on IRGE plant tissue or in the case of predators or parasites, by feeding on invertebrate prey or hosts that have fed on IRGE plant tissue. In this report, we consider three important factors that must be considered in Tier-1 assays: (i) methods for delivery of the IP to the test organisms; (ii) the need for and selection of compounds used as positive controls; and (iii) methods for monitoring the concentration, stability and bioactivity of the IP during the assay. We also analyze the existing data from Tier-1 assays regarding the toxicity of Bt Cry proteins to non-target arthropod species. Odorant receptors are thought to play critical roles in the perception of chemosensory stimuli by insects. The primary method to address the functions of odorant receptors in insects is to use in vitro binding assays between the receptors and potential chemical stimuli. We injected MmedOrco dsRNA into the abdominal cavity of a braconid wasp, Microplitis mediator, and assayed for expression of this gene 72 h after treatment (RNAi). Quantitative real-time PCR demonstrated that the level of mRNA expression in MmedOrco dsRNA-treated M. mediator was significantly reduced (>90%) when compared with water-treated controls. Furthermore, electroantennogram (EAG) responses of M. mediator to two chemical attractants, nonanal and farnesene, were also reduced significantly (~70%) in RNAi-treated M. mediator when compared to controls. RNAi-treated M. mediator also responded by walking/flying at a lower rate to both chemicals when compared with controls in a Y-tube olfactometer bioassay, which provides direct evidence that MmedOrco plays an important role in perception of nonanal and farnesene in M. mediator. (Corresponding author) The application of recombinant DNA technology has resulted in many insect‐resistant varieties by genetic engineering (GE). Crops expressing Cry toxins derived from Bacillus thuringiensis (Bt) have been planted worldwide, and are an effective tool for pest control. However, one ecological concern regarding the potential effects of insect‐resistant GE plants on non‐target organisms (NTOs) has been continually debated. In the present study, we briefly summarize the data regarding the development and commercial use of transgenic Bt varieties, elaborate on the procedure and methods for assessing the non‐target effects of insect‐resistant GE plants, and synthetically analyze the related research results, mostly those published between 2005 and 2010. A mass of laboratory and field studies have shown that the currently available Bt crops have no direct detrimental effects on NTOs due to their narrow spectrum of activity, and Bt crops are increasing the abundance of some beneficial insects and improving the natural control of specific pests. The use of Bt crops, such as Bt maize and Bt cotton, results in significant reductions of insecticide application and clear benefits on the environment and farmer health. The objective of this study was to identify rice genes that are in response to the striped stem borer (SSB) ( Chilo suppressalis Walker) feeding at the first to second larval stage. Using combined suppression subtractive hybridization (SSH) and dot blot approaches, we analyzed the induced defense genes that took place during the first 72 h of infesting intact rice ( Oryza sativa L.) plants in sheath tissues with SSB larvae. By sequencing the whole SSH library, 39 expressed sequence tags involved in disease stress, insect stress or other stress responses were identified to be up-regulated by SSB larvae feeding. Among these genes, rice allene oxide cyclase (AOC), terpene synthase (TPS) and four proteinase inhibitor (PI) genes were up-regulated by SSB larvae feeding. Real-time quantitative reverse transcription polymerase chain reaction analysis showed that four rice PI genes were already up-regulated at 6 h, and reached peaks between 6 h to 12 h. In addition, the transcription of gene involving in jasmonate signaling pathway such as allene oxide cyclase (AOC) concerning rice early defense response to SSB feeding was activated after rice feeding by SSB for 2 h. Although the expression of rice terpene synthase (TPS) gene, involved in the biosynthesis of monoterpenes or diterpenes, was already up-regulated at 7 h, a significant increase in the expression was delayed until 12 h and reached its peak at 24 h. The rice stem borer, Chilo suppressalis Walker is one of the most important insect pests on rice in Asia, north Africa and southern Europe. Transgenic Bt rice has been developed in the laboratory with good resistance to this pest and other Lepidopteran insects, which will provide a possible alternative tool for this pest control. The full-length cDNAs encoding an aminopeptidase N (CsAPN) and a cadherin (CsCad) were cloned from C. suppressalis. CsAPN showed common features of, and high identities to, other insect APNs in its deduced amino acid sequence. Although a full-length cDNA encoding cadherin-like protein has been reported in GenBank, the newly isolated cadherin here (CsCad) showed some differences in its amino acid sequence, especially at the 7th cadherin repeat region (CR7), which indicated the newly isolated CsCad might be another allele. CsAPN and CsCad were successfully expressed in insect Tn cells, and the blot analysis showed these two proteins could bind Bt toxin Cry1Ab. Transgenic cotton expressing the Bacillus thuringiensis (Bt) Cry1Ac toxin has been commercially cultivated in China since 1997, and by 2000 Bt cotton had almost completely replaced non-transgenic cotton cultivars. To evaluate the impact of Bt cotton planting on the seasonal population patterns of cotton bollworm, Helicoverpa armigera, the dynamics of H. armigera moths were monitored with light traps from four locations (Xiajin, Linqing and Dingtao of Shandong Province; Guantao of Hebei Province) in high Bt density region and five locations (Anci and Xinji of Hebei Province; Dancheng and Fengqiu of Henan Province; Gaomi of Shandong Province) in low Bt density region from 1996 to 2008. A negative correlation was found between moth densities of H. armigera and the planting years of Bt cotton in both high and low Bt density areas. These data indicate that the moth population density of H. armigera was reduced with the introduction of Bt cotton in northern China. Three generations of moths occurred between early June and late September in the cotton regions. Interestingly, second-generation moths decreased and seemed to vanish in recent years in high Bt density region, but this tendency was not found in low Bt density region. The data suggest that the planting of Bt cotton in high Bt density region was effective in controlling the population density of second-generation moths. The flight activity of Spodoptera litura in tethered conditions is evaluated using a computer-mediated flight-mill in the laboratory. The 3–4-day-old moths fly longer and farther than those of other ages. Male and female moths exhibit similar flight activity. Mating status does not influence the flight duration and distance of 2-day-old females. However, these two flight parameters with a 6-day-old mated female is significantly lower than that of unmated ones. The optimum temperature for flight ranged from 16–24°C, whereas the optimum RH ranged from 60%–100%. During 72-h period, the total flight duration and distance of 1-day-old male and female moths were 19.6 h (± 5.8) and 83.3 km (± 28.4), and 24.0 h (± 7.0) and 105.4 km (± 37.4), respectively. The pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), a cotton pest probably native to Indo-Pakistan, invaded China at the beginning of the 20th century. Chinese P. gossypiella have been assumed to be the result of indiscriminate introductions from Pakistan and America by transport of cotton seed. We tested this long-held hypothesis and genotyped a total of 527 individuals from 14 sites at 13 microsatellite loci. We analyzed these data with traditional statistics as well as with Bayesian methods. The loci were, for the most part, highly polymorphic. The allelic richness of Chinese populations at six loci was greater than those of the Pakistani and American populations. Significant deficits of heterozygotes were recorded for all 14 populations, and null alleles were the most probable factor contributing to these deficits. Pairwise FST estimates showed that there was significant differentiation among the pooled Chinese, Pakistani, and American populations, and there was structure within most of the Chinese populations. The Bayesian analysis revealed that the combined Chinese, American, and Pakistani populations formed separate clusters, and the nine Chinese populations were divided into two clusters. Allelic frequency distributions showed that private and shared alleles within Chinese P. gossypiella were derived only partly from the Pakistani and American populations. We developed eight polymorphic microsatellite markers for the green leaf bug, Lygus lucorum Meyer-Dür. The number of observed alleles per locus in 48 individuals ranged from three to 13, whereas the observed and expected heterozygosities ranged from 0.133 to 0.810 and from 0.236 to 0.886, respectively. Cross-species amplifications were conducted both within and between generas, and the amplification results indicated that these primers enriched for L. lucorum Meyer-Dür are species-specific. Mamestra brassicae L. is an important, regionally migratory pest of vegetable crops in Europe and Asia. Its migratory activity contributes significantly to population outbreaks, causing severe crop yield losses. Because an in-depth understanding of flight performance is key to revealing migratory patterns, here we used a computer-linked flight mill and stroboscope to study the flight ability and wingbeat frequency (WBF) of M. brassicae in relation to sex, age, temperature, and relative humidity (RH). The results showed that age significantly affected the flight ability and WBF of M. brassicae , and 3-d-old individuals performed the strongest performance (total flight distance: 45.6 ± 2.5 km; total flight duration: 9.3 ± 0.3 h; WBF: 44.0 ± 0.5 Hz at 24°C and 75% RH). The age for optimal flight was considered to be 2–3 d old. Temperature and RH also significantly affected flight ability and WBF; flight was optimal from 23°C to 25°C and 64–75% RH. Because M. brassicae thus has great potential to undertake long-distance migration, better knowledge of its flight behavior and migration will help establish a pest forecasting and early-warning system. The fall armyworm (FAW), native to the Americas, has rapidly invaded the whole of Southern China since January 2019. In addition, it can survive and breed in the key maize- and rice- growing area of the Yangtze River Valley. Furthermore, this pest is also likely to continue infiltrating other cropping regions in China, where food security is facing a severe threat. To understand the potential infestation area of newly-invaded FAW from the Yangtze River Valley, we simulated and predicted the possible flight pathways and range of the populations using a numerical trajectory modelling method combining meteorological data and self-powered flight behavior parameters of FAW. Our results indicate that the emigration of the first and second generations of newly-invaded FAW initiating from the Yangtze River Valley started on 20 May 2019 and ended on 30 July 2019. The spread of migratory FAW benefitted from transport on the southerly summer monsoon so that FAW emigrants from the Yangtze River Valley can reach northern China. The maize-cropping areas of Northeastern China, the Korean Peninsula and Japan are at a high risk. To analyze genetic characters of migratory dragonflies, we used 10 microsatellite markers and a partial sequence of the mitochondrial gene Cytb to investigate genetic diversity and demographic history among 19 populations of P. flavescens in eastern the monsoon region of China. In a Bayesian clustering analysis of the microsatellite data, three distinct clades were present, and each population consisted of a mixture of individuals from the three clusters. An AMOVA of the data from both the microsatellite loci and Cytb revealed that genetic variation was mainly within each population. For the 543 individuals from the 19 regions, 77 unique haplotypes were obtained by DnaSP 4.0, and a median-joining network showed no obvious geographical pattern and displayed high gene flow and minimal population genetic structure among the 19 populations. According to a Mantel test, there was no significant association between genetic distance and geographic distribution and no isolation by distance. Mismatch distribution and neutrality tests showed no demographic expansion for the 19 populations. Microsatellite and mitochondrial DNA data suggested there was high gene flow and low differentiation among the populations. Cryptochromes act as photoreceptors or integral components of the circadian clock that involved in the regulation of circadian clock and regulation of migratory activity in many animals, and they may also act as magnetoreceptors that sensed the direction of the Earth's magnetic field for the purpose of navigation during animals' migration. Light is a major environmental signal for insect circadian rhythms, and it is also necessary for magnetic orientation. We identified the full-length cDNA encoding As-CRY1 and As-CRY2 in Agrotis segetum Denis and Schiffermaller (turnip moth (Lepidoptera: Noctuidae)). The DNA photolyase domain and flavin adenine dinucleotide-binding domain were found in both cry genes, and multiple alignments showed that those domains that are important for the circadian clock and magnetosensing were highly conserved among different animals. Quantitative polymerase chain reaction showed that cry genes were expressed in all examined body parts, with higher expression in adults during the developmental stages of the moths. Under a 14:10 (L:D) h cycle, the expression of cry genes showed a daily biological rhythm, and light can affect the expression levels of As-cry genes. The expression levels of cry genes were higher in the migratory population than in the reared population and higher in the emigration population than in the immigration population. These findings suggest that the two cryptochrome genes characterized in the turnip moth might be associated with the circadian clock and magnetosensing. Insects interact with their environment and respond to the changes in host plant conditions using semiochemicals. Such ecological interactions are facilitated by the olfactory sensilla and the use of olfactory recognition proteins. The cotton aphid Aphis gossypii can change its phenotype in response to ecological conditions. They reproduce mainly as wingless asexual morphs but develop wings to find mates or new plant hosts under the influence of environmental factors such as temperature, plant nutrition and population density. Two groups of small soluble proteins, odorant binding proteins (OBPs) and chemosensory proteins (CSPs) are believed to be involved in the initial biochemical recognition steps in semiochemical perception. However, the exact molecular roles that these proteins play in insect olfaction remain to be discovered. In this study, we compared the transcriptomes of three asexual developmental stages (wingless spring and summer morphs and winged adults) and characterised 9 OBP and 9 CSP genes. The gene structure analysis showed that the number and length of introns in these genes are much higher and this appears to be unique feature of aphid OBP and CSP genes in general. Another unique feature in aphids is a higher abundance of CSP transcripts than OBP transcripts, suggesting an important role of CSPs in aphid physiology and ecology. We showed that some of the transcripts are overexpressed in the antennae in comparison to the bodies and highly expressed in the winged aphids compared to wingless morphs, suggesting a role in host location. We examined the differential expression of these olfactory genes in ten aphid species and compared the expression profile with the RNA-seq analyses of 25 pea aphid transcriptome libraries hosted on AphidBase.
