Global climate models struggle to simulate both the convection and cloud ice fundamental to lightning formation. We use the first convection‐permitting, future climate simulations for the lightning hot spot of Africa, at the same time utilizing an ice‐based lightning parametrization. Both the model and observations show that lightning over Africa's drier areas, as well as the moist Congo, have more lightning per rainfall than other regions. Contrary to results in the literature, the future projection shows little increase in total lightning (~107 flashes (or 2%) per degree warming). This is a consequence of increased stability reducing the number of lightning days, largely offsetting the increased graupel and updraft velocity driving an increase in lightning per lightning day. The next step is to establish if these results are robust across other models and, if combined with parametrized‐convection models, whether ensemble‐based information on the possible responses of lightning to climate change can be investigated. Plain Language Summary: Lightning depends on ascending air in thunderstorms and the collision of cloud ice particles, which charge the thundercloud. Many climate models have too coarse a resolution to reliably capture these processes. We focus on Africa, which has some of the most frequent lightning in the world. We use a model that is much higher resolution than usual, and this allows us to explicitly simulate the deep convection associated with thunderstorms as well as provide more detailed representation of the distribution of cloud ice particles. Our results show that in drier regions, as well as the much wetter Congo, there is relatively more lightning per kilogram of surface rainfall than there is in other parts of the continent. Lightning does increase across the continent under climate change, but by a relatively small amount. This is despite the number of days with lightning decreasing as the lower atmosphere becomes more stable. On days with lightning, there are more lightning flashes because there is an increase in cloud ice and intensity of convection. This study gives much more detailed information about African lightning than previous work. However, it is a single simulation. Future research should look at these results across other climate models. Osteoarthritis affects the morphological properties of the femoral head. The goal of this study was to develop a method to elucidate whether these changes are localised to discrete regions, or if the reported trends in microstructural changes may be identified throughout the subchondral bone of the human femoral head. Whole femoral heads extracted from osteoarthritic (n = 5) and healthy controls (n = 5) underwent microCT imaging 39 μm voxel size. The subchondral bone plate was virtually isolated to evaluate the plate thickness and plate porosity. The trabecular bone region was divided into 37 volumes of interest spatially distributed in the femoral head, and bone morphometric properties were determined in each region. The study showed how the developed approach can be used to study the heterogeneous properties of the human femoral head affected by a disease such as osteoarthritis. As example, in the superior femoral head osteoarthritic specimens exhibited a more heterogeneous micro-architecture, with trends towards thicker cortical bone plate, higher trabecular connectivity density, higher trabecular bone density and thicker structures, something that could only be observed with the newly developed approach. Bone cysts were mostly confined to the postero-lateral quadrants extending from the subchondral region into the mid trabecular region. Nevertheless, in order to generalise these findings, a larger sample size should be analysed in the future. This novel method allowed a comprehensive evaluation of the heterogeneous micro-architectural properties of the human femoral head, highlighting effects of OA in the superior subchondral cortical and trabecular bone. Abstract: High‐resolution (grid spacing 10 km in midlatitudes) model simulations using explicitly resolved convection in the Met Office Unified Model, as part of the Horizon 2020 PRIMAVERA project, are used to provide a global lightning climatology. The results show for the first time that global simulations can capture the strong diurnal flash rate variation as well as the seasonal variation. The lightning parametrization uses information about the graupel and ice water path to estimate a total lightning flash rate. Comparisons are made with the World Lightning Location Network (that mainly detects cloud to ground lightning) and combined Lightning Imaging Sensor and Optical Transients Detector data set (that provides an estimate of total flash rate). The model results generally capture the temporal behavior and spatial distribution of the lightning over land. Over the ocean, the lightning in the Intertropical Convergence Zone appears excessive. The drive to increase the output of animal product in some sectors of ruminant livestock production has led to greater use of feeds such as cereal grains and soyabean meal that are potentially human-edible. This trend has caused concern since, by so doing, ruminants compete not only with monogastric livestock but also with the human population for a limited global area of cultivatable land on which to produce grain crops. Reasons for using potentially human-edible feeds in ruminant diets include increased total daily energy intake, greater supply of essential amino acids and improved ruminal balance between fermentable energy and degradable protein. Soyabean meal, produced on land that has been in arable cultivation for many years can fulfil a useful role as a supplier of undegraded dietary protein in diets for high-yielding dairy cows. However, in the context of sustaining the production of high-quality foods from livestock to meet the demands of a growing human population, the use of potentially human-edible feed resources by livestock should be restricted to livestock with the highest daily nutrient requirements; that is, potentially human-edible feed inputs should be constrained to meeting requirements for energy and protein and to rectifying imbalances in nutrient supply from pastures and forage crops such as high concentrations of nitrogen (N). There is therefore a role for human-edible feeds in milk production because forage-only systems are associated with relatively low output per head and also low N use efficiency compared with systems with greater reliance on human-edible feeds. Profitability on farm is driven by control of input costs as well as product value and examples are given of low-cost bovine milk and meat production with little or no reliance on potentially human-edible feeds. In beef production, the forage-only systems currently under detailed real-time life-cycle analysis at the North Wyke Farm Platform, can sustain high levels of animal growth at low feed cost. The potential of all-forage diets should be demonstrated for a wide range of ruminant milk and meat production systems. The challenge for the future development of ruminant systems is to ensure that potentially human-edible feeds, or preferably human-inedible by-products if available locally, are used to complement pastures and forage crops strategically rather than replace them. Abstract: Highlights of progress in the production of silage over the past 50 years include the introduction of improved hybrids of maize (Zea mays L.), the forage harvester, the big baler, polyethylene covering for horizontal silos, stretch‐wrap film for bales and novel additives designed to improve the fermentation and aerobic stability of silage. The key biochemical pathways in the silage fermentation have been described together with the effects of microbial and chemical additives on fermentation and aerobic stability during the feed‐out phase. The significance of oxygen and water in silage fermentation has been quantified and efficacy of covering silos has been established, with recent progress in the development of oxygen barrier film. Future perspectives include improving food safety and animal health by increasing the hygienic quality of silage, reducing the environmental impact of silage by decreasing loss of nitrogen to soil and atmosphere, reducing methanogenesis in the rumen and increasing methane yield from silage as biofuel, and the use of silages as feedstocks for multiple end uses in biorefineries. Losses of organic matter in the outer layers of bunker silos covered with conventional polyethylene ( PE) plastic can be substantial due to oxygen transmission through the plastic top-covering film during the post-ensiling storage period. The effect of two silo covering materials, oxygen barrier ( OB) film (45 μm thickness) and clear PE film (50 μm thickness), as underlays to a white-on-black PE plastic top cover (120 μm thickness), was assessed in the outer layer of whole-plant maize silage stored in three large bunker silos in the People's Republic of China. Samples of the crop at harvest and of silage from the upper 45 cm layer at 5 months post-ensiling, prior to removal of silage for feed-out, were analysed for DM, fermentation profile and chemical composition. Loss of OM was estimated from concentrations of ash in the crop at harvest and in the silage. Differences between underlay films in silage fermentation profile were small. Silage protected with OB underlay film had higher mean concentration of starch ( p < .008) and higher mean NDF digestibility ( p < .003) than silage under PE underlay film. Concentrations of ash were lower ( p < .001) for silage covered with OB film than for PE film in all three trials. Mean estimated losses of OM were 170 g/kg for OB underlay film and 232 g/kg for PE underlay film ( p < .001), and whole-silo estimated net economic benefits to OB underlay film ranged from 0.17 to 0.74 US $ per tonne fresh crop ensiled. Implications of silage hygienic quality for animal production were investigated on forty-five dairy farms in South West England. Samples of grass and maize silages and of total mixed rations ( TMR) were obtained together with information on silage technology, herd size and animal production. Samples were analysed for mycotoxins, bacteria, yeasts, moulds and chemical composition. Thirteen mycotoxins were assayed, but none were detected in the samples of grass silage. However, mycotoxins were found in 0·9 of all maize and other silage samples, with deoxynivalenol and zearalenone predominating. There was no relationship between total mycotoxin concentration and mean lactation milk yield per cow. Enterobacteria counts tended to be higher in maize silage than in grass silage and higher still in TMR - a cause for concern. There were no relationships between mould counts and mycotoxin concentrations in silages, implying that mycotoxins may have been produced in the field pre-ensiling. [ABSTRACT FROM AUTHOR]

Nutrition affects the environmental impact of milk production through its direct influence on enteric fermentation and excretion of undigested and excess nutrients, especially nitrogenous compounds, in manure and its indirect influence on daily level of milk output per cow and fertility. The higher the level of milk production and fertility, the lower the emissions intensity, or carbon footprint, of a dairy herd. Increasing feed efficiency reduces greenhouse gas emissions per litre of milk because emissions are spread over higher product output per cow. Nitrogen use efficiency and cow fertility can be increased and diet carbon footprint reduced by altering diet formulation to utilise by-product raw material feeds of low carbon footprint. A range of options was explored to test the hypothesis that diets for dairy cows could be formulated to reduce the carbon footprint (CFP) of feed, increase efficiency of conversion of potentially human-edible feed into milk, increase nitrogen use efficiency (NUE) and reduce methane (CH4) emissions per kg milk. Diets based on grazed grass, grass silage, maize silage or straw, supplemented with raw material feeds, were formulated to meet requirements for metabolizable energy and metabolizable protein for a range of daily milk yields. At similar levels of milk yield, NUE, predicted CH4 emissions and diet CFP were generally higher for diets based on maize silage than for those based on grazed grass, grass silage or straw. Predicted CH4 emissions and human-edible proportion decreased, while NUE increased with the increasing level of milk yield. It is concluded that there is potential to reduce the environmental impact of milk production by altering diet formulation, but the extent to which this might occur is likely to depend on availability of raw material feeds with low CFPs. The concentration of crude protein in grazed New Zealand pasture is typically in excess of 200 g/kg DM, which exceeds the requirement of the dairy cow and is reflected in elevated levels of daily urinary nitrogen (N) excretion, estimated to average 262 g N/head or 751 kg N/ha. This has adverse environmental consequences, including leaching of nitrate from soil into water courses and atmospheric emissions of nitrous oxide. Problems associated with pasture as the sole feed for dairy cows include weight loss in early lactation, poor fertility and reduced lactation length. Low-protein supplements can reduce N excretion rates and increase N use efficiency. A simple feeding strategy is proposed in which pasture is supplemented with maize or whole-crop wheat silage at 5 kg DM/cow per day from the start of the pre-calving dry period on winter run-off pasture to 100 days post-calving, and from 250 days post-calving to the end of lactation. The expected response, at an assumed substitution rate of 0.7 kg decrease in pasture intake per kg silage DM, is an increase in metabolisable energy of 10 to 15 MJ/cow/day, equivalent to 33 to 48 g milk solids (MS)/kg DM of supplement. This strategy is expected to result in significantly lower urinary N excretion by the cow. Actual responses in daily milk output, from published experiments where grazing stocking rates were increased to take account of reduced herbage intake, ranged from 50 to 100 g MS/kg DM of supplement. Other benefits include early lactation bodyweight maintenance, higher percentage of cows calved which are pregnant at 150 days in milk and increased lactation period. Constraints for farmers to implement such changes include cost of silage, value of milk sold, failure to integrate forage maize or wheat in rotational cropping with pasture, and the relatively poor aerobic stability of maize and whole-crop wheat silages. Silage making involves preserving forage crops by fermentation of water-soluble carbohydrates (sugars) to organic acids. The fermentation is relatively uncontrolled and inadequate acidification can allow the growth of undesirable microorganisms such as enterobacteria and clostridia. Ingress of oxygen to the outer layer of the silo or bale during the storage period encourages growth of yeasts and moulds. Mycobacterium bovis may survive in silages with pH above 5.0. Outgrowth of clostridia in aerobically deteriorating silage can lead to high counts of butyric acid spores in silage that can contaminate milk and cheese. Milk may be contaminated with Penicillium toxins and be rejected for containing antibiotics. Specific disease risks to livestock from poorly preserved silage include bacterial endotoxicosis, listeriosis, botulism, mycotoxicosis and nitric acid poisoning in addition to reduced feed value. Rapid acidification is essential once the crop is ensiled to restrict the growth of enterobacteria, listeria and clostridia. Oxygen barrier film, in addition to reducing loss of dry matter during the storage period, prevents clostridial outgrowth in the outer layer of the silo and is also likely to reduce the risk of listeriosis. A physically based method for parameterizing the role of subgrid-scale turbulence in the production and maintenance of supercooled liquid water and mixed-phase clouds is presented. The approach used is to simplify the dynamics of supersaturation fluctuations to a stochastic differential equation that can be solved analytically, giving increments to the prognostic liquid cloud fraction and liquid water content fields in a general circulation model (GCM). Elsewhere, it has been demonstrated that the approach captures the properties of decameter-resolution large-eddy simulations of a turbulent mixed-phase environment. In this paper, it is shown that it can be implemented in a GCM, and the effects that this has on Southern Ocean biases and on Arctic stratus are investigated. [ABSTRACT FROM AUTHOR]

Losses of dry matter and metabolisable energy (ME) occur in silage making due to delayed harvest and also between field and feed trough due to fermentation and aerobic spoilage. Farmers should aim to exert greater control of silage making operations to improve timeliness of harvest and to minimise losses of nutrients. Concentrate input per unit of milk production is often higher than optimal which is more often a reflection of low silage quality than of wasteful overfeeding. A week's delayed harvest of grass is estimated to incur a net cost of 13 pence per cow per day of winter feeding. Mowing a dry crop and spreading it out to maximise rate of water loss during the wilting period can reduce losses in the field. Consolidation of crop in the silo to achieve a minimum fresh weight density of 700 kg/m3 and dry matter (DM) density of 210 kg/m3 will help to minimise losses during the storage period, paying particular attention to the outermost layer which should be covered with an oxygen barrier film. A feed-out progression rate of at least 1 metre per week in winter and 1.5 to 2 metres per week in summer, coupled with harvesting a clean crop with low yeast and mould counts will help to reduce losses during feed-out. Many bunker silos are too old, too small, have no safety rail and are over-filled. Safety issues should be considered at all stages of silage making. A meta-analysis was undertaken of 51 comparisons of standard polyethylene film with oxygen barrier ( OB) film in covering systems for bunker silos, unwalled clamp silos and bales. Mean losses of DM or OM during storage from the top 10 to 60 cm of bunker and clamp silos were 195 g kg−1 for standard film and 114 g kg−1 for OB film systems (41 sets of data, P < 0·001), while mean total losses of DM from baled silage were 76·8 g kg−1 for standard film and 45·6 g kg−1 for OB film systems (10 sets of data, P < 0·001). Top surface silage judged subjectively to be inedible was 107 and 29·6 g kg−1 for standard film and OB film systems respectively (5 sets of data, P = 0·02). Aerobic stability was 75 h for silage stored under standard film system and 135 h for silage stored under OB film system (11 sets of data, P = 0·001). It is concluded that the OB film system reduces losses from the outer layers of silos and from bales and increases the aerobic stability of silage in the outer layers of silos. Introduction. One in eight of all total hip replacements requires revision within 10 years, 60% because of wear-related complications. The bearing surfaces may be made of cobalt/chromium, stainless steel, ceramic, or polyethylene. Friction between bearing surfaces and corrosion of non-moving parts can result in increased local and systemic metal concentrations. Objectives. To identify and systematically review published reports of systemic toxicity attributed to metal released from hip implants and to propose criteria for the assessment of these patients. Methods. Medline (from 1950) and Embase (from 1980) were searched to 28 February 2014 using the search terms (text/abstract) chrom* or cobalt* and [toxic* or intox* or poison* or adverse effect or complication] and [prosthes* or 'joint replacement' or hip or arthroplast*] and PubMed (all available years) was searched using the search term (('Chromium/adverse effects'[Mesh] OR 'Chromium/poisoning'[Mesh] OR 'Chromium/toxicity'[Mesh]) OR ('Cobalt/adverse effects'[Mesh] OR 'Cobalt/poisoning'[Mesh] OR 'Cobalt/toxicity'[Mesh])) AND ('Arthroplasty, Replacement, Hip'[Mesh] OR 'Hip Prosthesis'[Mesh]). These searches identified 281 unique references, of which 23 contained original case data. Three further reports were identified from the bibliographies of these papers. As some cases were reported repeatedly the 26 papers described only 18 individual cases. Systemic toxicity. Ten of these eighteen patients had undergone revision from a ceramic-containing bearing to one containing a metal component. The other eight had metal-on-metal prostheses. Systemic toxicity was first manifest months and often several years after placement of the metal-containing joint. The reported systemic features fell into three main categories: neuro-ocular toxicity (14 patients), cardiotoxicity (11 patients) and thyroid toxicity (9 patients). Neurotoxicity was manifest as peripheral neuropathy (8 cases), sensorineural hearing loss (7) and cognitive decline (5); ocular toxicity presented as visual impairment (6). All these neurological features, except cognitive decline, have been associated with cobalt poisoning previously. Type of prosthesis and blood metal concentrations. Where blood or serum metal concentrations were reported ( n = 17 for cobalt and n = 14 for chromium), the median cobalt concentration was 398 (range, 13.6-6521) μg/L and the median chromium concentration was 48 μg/L (in whole blood) (range, 4.1-221 μg/L including serum and blood values). Those patients reported to have systemic features who had received a metal-on-metal prosthesis ( n = 8) had a median peak blood cobalt concentration of 34.5 (range, 13.6-398.6) μg/L; those with a metal-containing revision of a failed ceramic prosthesis ( n = 10) had a median blood cobalt concentration of 506 (range, 353-6521) μg/L. Management. The most common treatment was removal of the metal-containing prosthesis, undertaken in all but 2 patients. This was usually associated with a fall in circulating cobalt concentration and improvement in some or all features. Clinical and toxicological assessment of systemic features. We propose the following criteria for assessing the likelihood that clinical features are related to cobalt toxicity: clinical effects consistent with the known neurological, cardiac, or thyroidal effects of cobalt, and for which any other explanation is less likely; increased blood cobalt concentrations (substantially higher than those in patients with well-functioning prostheses) several months after hip replacement; a fall in the blood cobalt concentration, usually accompanied by signs of improvement in features. When judged by these criteria, the systemic features in 10 of the reported cases are likely to be related to cobalt exposure from a metal-containing hip prosthesis. Conclusions. Rarely, patients exposed to high circulating concentrations of cobalt from failed hip replacements develop neurological damage, hypothyroidism and/or cardiomyopathy, which may not resolve completely even after removal of the prosthesis. The greatest risk of systemic cobalt toxicity seems to result from accelerated wear of a cobalt-containing revision of a failed ceramic prosthesis, rather than from primary failure of a metal-on-metal prosthesis. Extraction of bone contours from radiographs plays an important role in disease diagnosis, preoperative planning, and treatment analysis. We present a fully automatic method to accurately segment the proximal femur in anteroposterior pelvic radiographs. A number of candidate positions are produced by a global search with a detector. Each is then refined using a statistical shape model together with local detectors for each model point. Both global and local models use Random Forest regression to vote for the optimal positions, leading to robust and accurate results. The performance of the system is evaluated using a set of 839 images of mixed quality. We show that the local search significantly outperforms a range of alternative matching techniques, and that the fully automated system is able to achieve a mean point-to-curve error of less than 0.9 mm for 99% of all 839 images. When silage is exposed to air on opening the silo, or after its removal from the silo, fermentation acids and other substrates are oxidized by aerobic bacteria, yeasts and moulds. The aerobic stability of silage is a key factor in ensuring that silage provides well-preserved nutrients to the animal with minimal amounts of mould spores and toxins. In this paper, key findings and recent developments are reviewed, and findings of recent research are integrated in terms of four themes: (i) the most significant biochemical and microbiological factors, (ii) physical and management factors, (iii) type of additive and (iv) silo sealing. The development of yeasts and moulds during plant growth, and during field wilting or storage, and the concentration of undissociated acetic acid in silage are important microbiological and biochemical factors affecting aerobic stability. Silage density and porosity are key physical factors that affect the rate of ingress of oxygen into the silage mass during the feed-out period. A target for potential silage aerobic stability is 7 d including time in the feed trough. To achieve this target, speed of harvest should be coordinated with packing tractor weight to achieve a minimum silage density by the time of feed-out of 210 kg DM m−3, maximum proportional porosity of 0·4 and a rate of silage removal, which matches or exceeds the depth of air penetration into the silo. The use of additives to increase aerobic stability is advisable when there is the risk of these objectives not being met. It is difficult to obtain phenotypic data on disease susceptibility directly from swine in an industry setting. The magnitude of the proliferative response of peripheral blood mononuclear cells (PBMC) to the T cell mitogen concanavalin A (Con A) has long been used as an indirect measure of the responsiveness of the immune system to antigenic stimulation. This trait is known to exhibit moderate heritability in swine, but little is known about the identity of the genes that control the response. In this study, we carried out a time-course microarray experiment to measure gene expression at 3 different stages (3, 20, and 68 h) poststimulation of PBMC with Con A. A total of 46, 452, and 418 differentially expressed (DifEx) genes were identified at each time point, respectively. Expression changes for a subset of these genes were subsequently confirmed by real-time PCR. Functional annotation analyses of the microarray results successfully identified sets of genes involved in processes associated with multiple aspects of cell division, such as DNA and protein synthesis, and control of mitosis. However, the discovery of genes that controlled the response of PBMC to mitogen was limited with this approach, because the drastic changes in the transcriptional program necessitated by cells undergoing division masked changes in smaller immune response gene sets. Pathway and network analyses that focused on immune cells proved to be a more effective strategy for the identification of genes that coordinate aspects of the mitogenic response that are specific to PBMC. The cytokine gene IL15 was shown to be central to the highest scoring network at 20 h and affect the expression of 16 other DifEx genes, including some genes known to regulate T cell activation, such as IL7R, JUN, TNFRSF9, and ZAP70. The IL15 gene maps to a previously identified QTL interval for immune responsiveness to Con A on SSC 8, which also contains the related IL2 gene. At 68 h, a distinct downregulation of major histocompatibility complex class II antigen presentation genes was observed. Overall, the gene expression profile of the Con A-stimulated porcine PBMC points to a Th, bias in immune activation. Further work is required to determine whether polymorphisms linked to genes identified in this study affect this immune response trait in pig populations and whether the trait itself correlates with decreased susceptibility to intracellular pathogens in swine. The article explores the Global Atmosphere 3.0 (GA3.0) of the Met Office Unified Model (MetUM) climate research and the Global Land 3.0 (GL3.0) of JULES community land surface model. An overview of the Global Atmosphere configuration principle as an atmospheric component applied to MetUM, JULES Global Land, and NEMO ocean model is provided. It highlights aspects of GA3.0 and GL3.0 including on formulation and discretization, solar and terrestrial radiation, and orographic gravity.

Livestock, particularly ruminants, can eat a wider range of biomass than humans. In the drive for greater efficiency, intensive systems of livestock production have evolved to compete with humans for high-energy crops such as cereals. Feeds consumed by livestock were analysed in terms of the quantities used and efficiency of conversion of grassland, human-edible (‘edible’) crops and crop by-products into milk, meat and eggs, using the United Kingdom as an example of a developed livestock industry. Some 42 million tonnes of forage dry matter were consumed from 2008 to 2009 by the UK ruminant livestock population of which 0.7 was grazed pasture and 0.3 million tonnes was conserved forage. In addition, almost 13 million tonnes of raw material concentrate feeds were used in the UK animal feed industry from 2008 to 2009 of which cereal grains comprised 5.3 and soyabean meal 1.9 million tonnes. The proportion of edible feed in typical UK concentrate formulations ranged from 0.36 for milk production to 0.75 for poultry meat production. Example systems of livestock production were used to calculate feed conversion ratios (FCR – feed input per unit of fresh product). FCR for concentrate feeds was lowest for milk at 0.27 and for the meat systems ranged from 2.3 for poultry meat to 8.8 for cereal beef. Differences in FCR between systems of meat production were smaller when efficiency was calculated on an edible input/output basis, where spring-calving/grass finishing upland suckler beef and lowland lamb production were more efficient than pig and poultry meat production. With the exception of milk and upland suckler beef, FCR for edible feed protein into edible animal protein were >1.0. Edible protein/animal protein FCR of 1.0 may be possible by replacing cereal grain and soyabean meal with cereal by-products in concentrate formulations. It is concluded that by accounting for the proportions of human-edible and inedible feeds used in typical livestock production systems, a more realistic estimate of efficiency can be made for comparisons between systems. Objectives The genetic aetiology of osteoarthritis has not yet been elucidated. To enable a well-powered genome-wide association study (GWAS) for osteoarthritis, the authors have formed the arcOGEN Consortium, a UK-wide collaborative effort aiming to scan genome-wide over 7500 osteoarthritis cases in a two-stage genome-wide association scan. Here the authors report the findings of the stage 1 interim analysis. Methods The authors have performed a genome-wide association scan for knee and hip osteoarthritis in 3177 cases and 4894 population-based controls from the UK. Replication of promising signals was carried out in silico in five further scans (44 449 individuals), and de novo in 14 534 independent samples, all of European descent. Results None of the association signals the authors identified reach genome-wide levels of statistical significance, therefore stressing the need for corroboration in sample sets of a larger size. Application of analytical approaches to examine the allelic architecture of disease to the stage 1 genome-wide association scan data suggests that osteoarthritis is a highly polygenic disease with multiple risk variants conferring small effects. A variety of assays were utilized to determine the effects of Leptospermum petersonii essential oil on both Candida albicans and Aspergillus fumigatus. Hyphal morphology, susceptibility of spheroplasts and uptake of propidium iodide following exposure to the oil suggest that the mode of action of L. petersonii essential oil is through direct disturbance of the fungal cell membrane. Data also confirms that the volatile component of the oil is highly antifungal, independent of direct contact between the liquid oil and the fungal membrane. The degree of inhibition was greater when fungi were directly exposed to oil volatiles compared to pre-inoculation exposure of oil volatiles into the agar. Livestock contribute directly (i.e. as methane and nitrous oxide (N2O)) to about 9% of global anthropogenic greenhouse gas (GHG) emissions and around 3% of UK emissions. If all parts of the livestock production lifecycle are included (fossil fuels used to produce mineral fertilizers used in feed production and N2O emissions from fertilizer use; methane release from the breakdown of fertilizers and from animal manure; land-use changes for feed production and for grazing; land degradation; fossil fuel use during feed and animal production; fossil fuel use in production and transport of processed and refrigerated animal products), livestock are estimated to account for 18% of global anthropogenic emissions, but less than 8% in the UK. In terms of GHG emissions per unit of livestock product, monogastric livestock are more efficient than ruminants; thus in the UK, while sheep and cattle accounted for 32% of meat production in 2006, they accounted for ~48% of GHG emissions associated with meat production. More efficient management of grazing lands and of manure can have a direct impact in decreasing emissions. Improving efficiency of livestock production through better breeding, health interventions or improving fertility can also decrease GHG emissions through decreasing the number of livestock required per unit product. Increasing the energy density of the diet has a dual effect, decreasing both direct emissions and the numbers of livestock per unit product, but, as the demands for food increase in response to increasing human population and a better diet in some developing countries, there is increasing competition for land for food v. energy-dense feed crops. Recalculating efficiencies of energy and protein production on the basis of human-edible food produced per unit of human-edible feed consumed gave higher efficiencies for ruminants than for monogastric animals. The policy community thus have difficult decisions to make in balancing the negative contribution of livestock to the environment against the positive benefit in terms of food security. A retrospective study has been undertaken in an attempt to identify physical parameters that could confidently be used to predict an enhanced risk of bladder morbidity following intracavitary brachytherapy. 366 women received brachytherapy as all, or part, of their treatment for cervical cancer at the Christie Hospital in 1990 and 1991, and of these, 60 patients developed identifiable bladder morbidity (graded on a scale of 1-4 using the Franco-Italian glossary). These were age and stage matched with 60 asymptomatic women who were also treated for cervical cancer by brachytherapy during the same time period. The sizes of applicators used in the two groups were noted and compared. The two groups were also compared with respect to the heights of the applicator set above the symphysis pubis, the degree of anteversion or retroversion of the applicator sets and where possible, the doses at the International Commission on Radiation Units and Measurements (ICRU) bladder reference point. Where CT scans of the applications were available, these were reviewed to see if any differences in the size, shape or location of the bladder were apparent. No significant difference was found between the two groups of patients for any of the parameters investigated. The physical factors investigated in this study cannot be used to reliably predict bladder complications. Abstract The recycling of sewage sludge or biosolids to grassland is strategically important in the European Union (EU) and its use is tightly regulated to control the risk of pathogen transfer to animals and the food chain. Sewage sludges not only contain valuable concentrations of beneficial nutrients, but also elevated concentrations of potentially toxic metals (PTM) compared with average background concentrations in the soil. The EC and UK regulations refer to six PTM, Cd, Pb, Cu, Zn, Ni and Hg, with provisional regulations for Cr, that have to be controlled to prevent detrimental effects on soil and animal health. Despite these regulations, there is still a danger that grazing animals may ingest elevated concentrations of PTM. Biosolids may adhere to herbage after the surface application of sewage sludge to grassland. The repeated surface application of sewage sludge to grassland can lead to elevated concentrations of PTM at the soil surface that may be ingested, together with soil and herbage, by grazing ruminants. This may lead to accumulation of Cd or Pb in liver or kidney. The risk to the human food chain is considered to be low, but the impact on the environment is still unknown. There is little information, for example, on the amount of soil and PTMs that may become incorporated into conserved grass. At present EU and UK legislation and voluntary codes of practice have been developed to protect animal from pathogens in sewage sludge and to minimize any potential risks from accumulation of PTM. The health and welfare of dairy cows are dependent upon the stockpersons who handle, observe and monitor them. The attitudes and behaviour of these people have been little studied, although they are fundamental for the animals' well-being and performance. The results of a series of standardised interviews with 238 dairy stockpersons are discussed. Milking was considered both the most important and the most enjoyed task, and the most disliked task was cleaning the parlour and yards. These attitudes can affect the health of the herd. The stockpersons valued their interaction with animals; ‘a special event involving the cows’ was frequently mentioned as the component of a ‘good’ day, and a ‘bad event involving the cows’ was almost unanimously the factor associated with a ‘bad’ day. The extreme hydrologic response of gravelly, sandy soils in the Carnation Creek watershed is examined from observations at 12 standpipe piezometers. The nearly continuous piezometric data are reported as a time series of monthly maximum readings. Ten locations of measurement appear to exhibit an upper limit to the pore water pressure head that is independent of rainfall intensity and duration. Two locations exhibit artesian pressures that appear directly influenced by rainfall characteristics and may last for several hours. We found the impact of individual storms to be highly variable. Increasing amounts of sewage sludge will be applied to agricultural land over the next 10 years as a result of the prohibition of its disposal to the sea. The addition of potentially toxic elements (PTEs) to the soil via sewage sludge is controlled by European legislation designed to limit the accumulation of PTEs in soil which could give rise to toxicity to plants or livestock. However the possibility exists that direct ingestion of sewage sludge and soil together with grazed herbage may result in accumulation of PTEs in body tissues. To assess the extent of accumulation of PTEs by direct ingestion of soil and sewage sludge 12 groups of housed weaned lambs were given diets ad libitum comprising dried grass (900 g/kg total diet dry matter (DM)) and three different soils (100 glkg total diet DM). Soil was replaced by dried digested sewage sludge at levels ofO (level 0), 75 (level 1), 150 (level 2) and 300 g/kg soil DM (level 3). Voluntary intake of DM was markedly depressed by the inclusion of sewage sludge in the diet (on average by 24 g DM per g sewage sludge DM addition). There was no effect of sewage sludge on diet apparent digestibility. Live-weight gain was depressed (P < 0·001) by the addition of sewage sludge to the diet from 236 glday (level 0) to 141 glday (level 3). Liver and kidney weights were also reduced (P < 0·01). The apparent availability coefficients for Cd, Pb and Cu increased with increasing level of sewage sludge in the diet fP < 0·05), as did their concentrations in the liver and kidney (P < 0·01). Concentrations of Cd and Pb in liver increased from <0·07 mg/kg DM and <0·40 mg/kg DM (level 0) respectively to 0·61 mg/kg DM and 4·60 mg/kg DM (level 3) respectively at the end of the trial. Similarly the concentrations of Cd and Pb in kidneys increased from 0·19 mg/kg DM and <0·56 mg/kg DM (level 0) respectively to 0·80 mg/kg DM and 7·10 mg/kg DM (level 3) respectively by the end of the trial. No increases were observed in concentrations of Cd or Pb in muscle tissue. The main effect of type of soil on concentrations of PTEs in body tissues was not significant The daily rate of accumulation ofPb in kidney ranged from 1·1 to 51·5 uglg daily tissue DM growth during the first 57 days of the experiment and from 0·33 to 6·78 /xg/g daily tissue DM growth between day 57 and day 112. A decrease in the second period was also observed for Cd, with accumulation in kidney ranging from 0·31 to 4·44 fig/g daily tissue DM growth during the first 57 days and from 0·21 to 1·44 /jg/g daily tissue DM growth between day 57 and 112. Concentrations of Pb in liver of lambs given the highest level of sludge approached the statutory limit set for human food. The results indicate that in relation to accumulation of PTEs in liver and kidney there would appear to be little margin of safety with respect to the current United Kingdom statutory limits for the concentrations of Cd and Pb in sludge-amended soils. Current European Union legislation will increase the level of disposal of sewage sludge to agricultural land in the next 10 years. This increase may lead to an increase in ingestion, by grazing animals, of herbage with elevated levels of potentially toxic elements (PTEs) derived from sludge adhered to the herbage or sewage sludge-amended soil resulting in the accumulation of PTEs in body tissues, especially the liver and kidney. To assess the possible accumulation of PTEs from soils treated historically with sewage sludge, eight groups of housed weaned lambs were given either dried grass as a sole food (grass control), or diets comprising dried grass (0·9 of total diet dry matter (DM)) and soil (0·1 of total diet DM) from two experimental sites, plots within which had been treated 14 years previously with different quantities of sewage sludge. Soils from the two sites (Cassington and Royston) had contrasting physical characteristics, pH and contents of calcium. The soil from the site at Cassington was slightly acidic (pH 6·0) and had a concentration of calcium of 7·92 g/kg DM whereas the soil from the site at Royston was alkaline (pH 8·0) and had a high concentration of calcium (23·5 g/kg DM). Plots at each site had a range of concentrations of cadmium (Cd) in the soil up to three times the United Kingdom (UK) statutory limit of 3 mg Cd per kg DM. The soils from these two sites were designated control — no amendment with sewage sludge (0·69 mg Cd per kg DM), low (3·55 mg Cd per kg DM), medium (6·63 mg Cd per kg DM) and high (8·82 mg Cd per kg DM; Cassington soil only). Voluntary DM intake of diets by weaned lambs (mean 1436 glday) was not affected significantly by any dietary treatment. The concentrations of Cd in liver increased (P < 0·001) from 0·061 and 0·072 mg/kg DM (Royston and Cassington control, respectively) to 0·218 and 0·403 mg/kg DM (Royston and Cassington medium, respectively) and 0·500 mg/kg DM (Cassington high). The concentrations of Pb in liver increased (P < 0·001) from 0·733 and 0·627 mg/kg DM (Royston and Cassington control, respectively) to 118 and 1·25 mg/kg DM (Royston and Cassington medium, respectively) and 1·18 mg/kg DM (Cassington high). Similar changes were observed for concentrations of Cd and Pb in kidney. A depletion of the concentration of Cu in the liver was observed in all treatments containing soil in the diet. The rate of accumulation of Cd in the liver ranged from 0 to 0·35 μg/g daily tissue DM growth and in the kidney ranged from 0 to 0·44 μg/g daily tissue DM growth. The rate of accumulation of Pb in the liver ranged from 0 to 6·01 μg/g daily tissue DM growth and in the kidney ranged from 0 to 0·63 μg/g daily tissue DM growth. No significant accumulation of PTEs was observed in muscle tissue. It is concluded that the current UK statutory limits for the concentrations ofCd and Pb in soils treated with sewage sludge should be reviewed in the light of these results, though they require confirmation in the grazing situation. Although many human major histocompatibility genes have been identified, relatively few have been localized to the class I region. We searched for new class I region genes by sample sequencing, a process in which short stretches of random genomic sequence are generated from cosmids and then compared with sequences deposited in nucleotide databases. Four class I region cosmids were isolated for sample sequencing by screening a chromosome 6 specific cosmid library with probes derived from specific class I region genes or with overlapping class I region yeast artificial chromosomes. Cosmids were sonnicated to produce fragments of 0.5 – 1 kilobases, subcloned, and sequenced using an automated sequencer. Sequences were then compared with nucleotide sequences deposited in the GenBank databases using the BLASTN algorithm. A number of potential new class I region genes were identified, including a cDNA with similarity to the tre oncogene, the trans-activating factor SC1 ( TCF19), and a member of the interferon inducible 1 – 8 gene family. These observations suggest that sample sequencing is an efficient method for identifying new class I region genes, which can be applied to other regions of the genome and to other species, and support previous observations that the class I region contains a variety of genes other than those encoding HLA antigens.
