Capillaries (25-and 50-μm inner diameter) coated with a double-alkyl-chain cationic surfactant N, N-didodecyl- N, N-dimethylammonium bromide (DDAB) were used for the separation of four basic standard proteins in buffers of pH 4 at various ionic strengths. The choice of buffer is critical for the analytical performance. Ammonium ions must be avoided in the buffer used in the non-covalent coating procedure owing to competition with the surfactant. Phosphate buffer gave a better separation performance than some volatile buffers; the reason seems to be a complex formation between the proteins and dihydrogenphosphate ions, which decreases tendencies for adsorption to the capillary surface. The DDAB coating was easy to produce and stable enough to permit, without recoating, consecutive separations of the proteins for up to 100 min with good precision in migration times and peak areas. A strong electroosmotic flow gives rapid separations, which is of special importance when commercial instruments are used, since the choice of the length of the capillary is restricted. The enantioselectivity, resolution factor and migration times for some local anaesthetic drugs were evaluated at different selector concentrations and plug lengths with different cyclodextrins as chiral selectors using the partial filling technique. The resolution remained constant when equal amounts of the chiral selector at a concentration below the optimal concentration were introduced in the capillary. An HPLC method based on a prodigy ODS2 column with a mobile phase contianging phosphate buffer, tetrabutylammonium hydrogen sulfate (TBA) and an acetonitrile gradient was optimized for the determination of erythromycin in delayed-release capsules. A central composite face-centered (CCF) factorial design was used with the factors pH, concentration of TBA and gradient slope. The method could separate erythromycin A from 7 related erythromycins. The partial filling technique (PFT) in capillary electrophoresis (CE) is an efficient system where, only 50–800 nanolitres of a chiral selector solution needs to be added to each run. PFT is especially applicable when these additives to the background electrolyte (BGE) are expensive or absorb UV light. The selector dissolved in the BGE is applied to the capillary as a plug, shorter than the effective length of the capillary, prior to application of the analyte. During the run both ends of the capillary are connected to the BGE. The applied plug and the analyte may move in opposite directions or in the same direction at different velocities depending on their electrophoretic mobilities. Thus the final plug length is either longer or shorter than the original length. The technique has been successfully applied in a number of studies including enantiomeric separation with a variety of selectors, and for the determination of conditional association constants. The relative bioavailabilities of single oral doses of ampicillin, amoxycillin, and bacampicillin were compared with and without concomitant administration of a six-times higher molar dose of cyclacillin. As the absorption of cyclacillin has been shown to involve a capacity-limited transport system in animals, it was selected as the reference compound for the study. The treatments were given to 14 fasting volunteers using a randomized, complete crossover design. The drugs in plasma and urine were determined by liquid chromatography. Renal clearance was 17%, 10% and 19% lower when ampicillin, amoxycillin, and bacampicillin were given together with cyclacillin. Consequently, differences in the relative bioavailability were based on urinary recoveries assuming constant non-renal clearance. When amoxycillin was given with cyclacillin there was a 67% delay in the time of the plasma peak concentration, and an 8% lower urinary recovery than when it was given alone. There was a 50% and 33% delay in the t of ampicillin and bacampicillin when combined with cyclacillin; the urinary recovery of ampicillin in the combination was 10% lower but that of bacampicillin was similar. There was also a 20% delay in the t of cyclacillin when combined with amoxycillin. The differences in renal clearance indicate an interaction in the renal elimination of the drugs, but the effect was probably not the explanation for the marked shift in time of the absorption of these rapidly absorbed drugs. The pharmacokinetics of bacmecillinam and pivmecillinam were studied in healthy fasting volunteers given tablets in a cross-over, randomized order. The mean (±SD) peak levels of plasma mecillinam were 1.43±0.34, 2.73±0.43, and 4.62±1.41 mg/l after bacmecillinam 100, 200, and 400 mg and 2.38±0.65 mg/l after pivmecillinam 400 mg. The corresponding areas under plasma Vs time curves (AUC) were 2.21±0.19, 3.99±0.63, and 7.74±1.38 mg·h·l for bacmecillinam and 5.35±0.93 mg·h·l for pivmecillinam. The elimination half-lives were 0.8-1.1h for bacmecillinam and 0.7h for pivmecillinam. The 12 h urinary recovery of unchanged mecillinam after the 400 mg doses was 41% for bacmecillinam and 30% for pivmecillinam. The 400 mg dose of bacmecillinam gave a significantly higher plasma peak ( p<0.001), AUC ( p<0.001) and urinary recovery ( p<0.001) than did pivmecillinam 400 mg. The plasma peaks appeared earlier and the rate of absorption was higher after bacmecillinam than after pivmecillinam ( p<0.05). In conclusion, bacmecillinam had a better bioavailability than pivmecillinam in the tablet formulations studied. The AUC increased linearly with increasing doses of bacmecillinam. The systemic availability of a new antidepressant, zimelidine, and of its pharmacologically active metabolite, norzimelidine, was studied in six healthy male volunteers. Three single doses of zimelidine (25 mg and 100 mg orally and 25 mg i.v.) and two single doses of norzimelidine (25 mg orally and i. v.) were given to each volunteer allowing at least seven days between administrations. Plasma concentrations of zimelidine and norzimelidine were determined in serial blood samples by HPLC. Following oral zimelidine peak plasma concentrations of the metabolite were attained about 3 h after dosing. Oral administration of norzimelidine itself resulted in a plasma concentration profile for this compound that was similar to that observed after oral zimelidine. Utilising the plasma concentration data following intravenous infusion of each compound, the elimination half-lives for zimelidine and norzimelidine were calculated to be 5.1 h (range 4.3-6.0) and 15.5 h (range 10.6-22.9) respectively. The total body clearances of the 2 compounds were similar at 0.52 l · min (range 0.26-0.70) for zimelidine and 0.56 l · min (range 0.28-0.83) for norzimelidine. The substantially longer elimination half-life of norzimelidine was apparently the result of a larger volume of distribution (9.4 l · kg; range 7.8-11.4) for this metabolite, as compared to zimelidine (3.21 · kg; range 1.6-4.9). The calculated bioavailability of zimelidine was 26% (range 9.1-39) after the 25 mg oral dose, and 29% (range 14-46) after the 100 mg dose. The bioavailability of norzimelidine was 66% (range 36-91). However, oral administration of zimelidine resulted in as much or more norzimelidine reaching the systemic circulation, as the oral administration of norzimelidine itself. The selective inhibitors of neuronal 5-hydroxytryptamine (5-HT) and noradrenaline (NA) uptake, zimelidine and desipramine, were compared in a double blind crossover study of 40 inpatients with endogenous depression. The clinical effects of these two drugs and some biochemical variables related to the monoamine systems were studied during 4 weeks' treatment. Patients with a low pretreatment level of 5-hydroxyindoleacetic acid (5-HIAA) in the cerebrospinal fluid (CSF) (<20 ng/ml) responded significantly better to zimelidine treatment than those with a high pretreatment level (>20 ng/ml). In the group treated with desipramine no difference in therapeutic outcome was obtained between patients with low and high pretreatments levels of 5-HIAA in the CSF. Attempts to correlate the steady state plasma concentrations of zimelidine, norzimelidine and desipramine with the therapeutic effect were unsuccessful. The plasma concentration of norzimelidine demonstrated a singificant ( P<0.05) positive correlation with age. The mean value of the uptake of C-5-HT in the patients' platelets, when measured before the treatment, was significantly ( P<0.05) lower than found in a control group. Zimelidine, mainly via its metabolite norzimelidine, caused a pronounced inhibition of uptake of C-5-HT in platelets, decrease in whole blood 5-HT and inhibition of accumulation of C-5-HT in rat hypothalamic synaptosomes incubated in the patients plasma. Desipramine produced a slight inhibition of accumulation of C-5-HT in rat synaptosomes, but a marked inhibition of uptake of C-5-HT in the patient's platelets and a decrease in whole blood 5-HT. The accumulation of H-NA in rat synaptosomes incubated in the patients' plasma was strongly inhibited by desipramine treatment but only slighty affected by zimelidine. [ABSTRACT FROM AUTHOR]

Different approaches how to handle the electromigration dispersion process that occurs in separation and determination of enantiomers are presented. The use of cyclodextrins as chiral selectors in resolution enantiomers involves the possibility to tune the conductivity of the sample band in order to obtain symmetrical and efficient peaks. Determination of impurities that migrate in the rear part of an overloaded main peak can be accomplished if the conductivity of the background electrolyte (BGE) is adapted to the conductivity of the sample band. This strategy was shown in determination of the content of D-sotalol in a mixture of L and D-sotalol. The efficiency and the symmetry of the overloaded L-sotalol peak was substantially improved by substitution of tetrabutylammonium ions for tetrapentylammonium ions as co-ions in the BGE. In this system it was possible to determine 0.2% w/w of the chiral impurity D-sotalol. A resolution model is presented and used qualitatively in the study where the complexation between the tetraalkyllammonium ions and the cyclodextrins is taken into account. [ABSTRACT FROM AUTHOR]

A new restricted access media (RAM) type of precolumn, Bio Trap 500 C18, for direct injection of plasma samples in column-switching systems was evaluated with respect to the elution of plasma proteins in different mobile phases, the loading capacity of plasma samples, the chromatographic behavior during plasma injections and protein contamination of the packing and sealings. More than 95% of plasma proteins could be excluded from the precolumn within three minutes for all selected mobile phases. Quantitative analyte recoveries could be obtained by injecting plasma samples ranging from 5 to 500 μL with the analyte mass>150 ng onto a BioTrap 500 C18 column (20×4 mm I.D.). One precolumn tolerated about 15 mL of plasma injection without out noticeable change in retention and pressure. Clogging of the precolumn was encountered (≥45 mL of plasma) due mainly to the adsorption of proteins on the packing. The performance of the analytical column (Kromasil C18) was also examined. A simultaneous optimization of resolution, efficiency and migration times of enkephalin-related peptides in micellar electrokinetic capillary chromatography (MEKC) was performed. Six experimental variables; the surfactant concentration, the percentage of organic modifier, the ionic strength of the buffer, the injected plug length, the applied temperature and the sample solution composition were studied via central composite design (CCD). Large differences in separation performance were observed at a commonly used level of organic modifier in the background electrolyte. Partial least squares regression of the responses revealed that the experimental domain was too large and complicated to be explained by the model. A new CCD model was obtained with improved prediction ability at narrower ranges of the experimental factors, especially of the organic modifier. A conflict between maximum resolution and efficiency within the shortest analysis time was observed. Therefore, constraints were set on maximal resolution and analysis time, while solving for maximum efficiency. Optimal operating conditions were found at 35 mM sodium dodecyl sulfate (SDS), 5% v/v of acetonitrile, 1.9 mm injected plug length, 35°C and sample solution with no added micelles, giving high efficiency and resolution at short analysis time. Critical parameters for the separation of enkephalin related peptides by micellar electrokinetic chromatography were identified by using statistical experimental design. Nine experimental variables: micelle concentration, pH of the background electrolyte, addition of organic modifier to the background electrolyte, injection length, temperature of the capillary, applied voltage, ionic strength of the buffer, the composition of the injected solution and stacking were investigated in a two-level fractional factorial design. The pH was found to be of fundamental importance but a great variation in separation performance was observed, subsequent modeling revealed two subgroups with respect to pH. For further screening of significant factors, the pH was kept constant and by using D-optimal design it was possible to study six interaction terms together with eight main effects in only 20 experiments. The effect of the interaction terms were often found to be as high as the main effects, thus exemplifying the complexity of the system. The effect of sodium dodecyl sulfate (SDS) concentration on efficiency and resolution was highly dependent on the amount of organic modifier, temperature and ionic strength. The migration factor calculated from obtained migration times was found to be a non-linear function of the factors involved. This paper describes a strategy where a major part of a single capillary is filled with sample which when is isotachophoretically (ITP) preconcentrated while a hydrodynamic backpressure is applied to keep the analytes in the capillary. In the subsequent analysis of the test compounds, amitriptyline and metoprolol, capillary zone electrophoresis is used. The concentration limit of detection is lowered at least 170 times. Preliminary results where an ITP-preconcentration is combined with micellar electrokinetic chromatography with a neutral micellar agent is also presented. [ABSTRACT FROM AUTHOR]

An isocratic HPLC technique for separation of phenolic compounds and corresponding glucuronides in urine is developed. Sample pre-treatment, often a tedious and rate-limiting factor, was eliminated by use of a coupled column system. Spiked urine samples were injected directly into a C4 precolumn and a selected fraction was transferred on-line from the precolumn to a silanized C18 analytical column in the backflush mode. Analyte peak enrichment was attained by employing mobile phases of different elution strengths. The weaker mobile phase (7% v/v acetonitrile) was used to strongly retain the analytes on the precolumn while most of the polar endogenous compounds were washed to waste. Elution and transfer of the trapped analytes from the precolumn to the analytical column was achieved by introducing a stronger mobile phase (20% v/v acetonitrile) with the aid of a switching valve. The use of cetyltrimethylammonium bromide as counter ion and micellar agent in the mobile phase involved a high selectivity for the analytes relative to the urine matrix components and allowed simultaneous analysis of the glucuronides and parent compounds without the need of gradient elution. Various mobile phases including phosphate buffer, pure water and five kinds of biological buffers (pH around 7) were systematically studied in terms of their ability to clean-up plasma matrix on precolumns of restrictedaccess media in a column-switching system. The necessary washing time, buffer pH, type and content of organic modifier were evaluated with respect to plasma elution profiles on restricted-access media precolumns. The influence of different mobile phases on the recovery of plasma matrix from alkyl-diol silica precolumns was studied by means of a scanning spectrophotometer. Our results show that phosphate buffer near physiological pH with small amounts of 2-propanol or acetonitrile was prefereble for direct injection of large plasma volumes (500 μL). More than 93% of the proteins in a plasma matrix can be recovered within 3 min from the alkyl-diol silica C18 column (25 mx 4 mm I.D.) as measured at 280 nm for all selected mobile phases except for tris (hydroxymethyl) aminomethane buffer, from which only about 88% was obtained. In drug purity determinations by capillary electrophoresis using UV-detection generally a relative high concentration of the main compound has to be injected. Principles how to handle overloading effects are described. NXX-066 is a drug with a native fluorescence which made it possible to use of CE system equipped with a LIF-detection unit. Monitoring drug purity determination with LIF detection results in an improved limit of quantification of the impurities and/or opens a possibility to avoid an overloaded main peak. A frequency doubled (FRED) argon ion laser set at 244 nm was used as excitation source in drug purity determination with the LIF-detection unit and the chemicals used were therefore carefully chosen in order to minimize the background noise level, which easily is enhanced when fluorescence detection is performed in the UV-range. The separation of the fluorescent NXX-066 analogues proved to be a difficult task, but an adequate resolution was obtained when β- and γ-cyclodextrins was added as structural selectors and the separation window was expanded by the use of a repressed/reversed electroosmotic flow. A comparison of the separation capability for the UV-detection system and the laser induced fluorescence detection system was performed, showing that the overloading effects of the main peak can be removed using the CE-LIF system and thereby obtain a substantially improved resolution of the analogues. A comparison of LC-UV and CE-UV for drug purity determination of a crude product from, synthesis of NXX-066 is also included in this paper where a good correlation of the results were obtained. The aim of this study was to determine whether concentration-dependent renal clearance of ampicillin and amoxycillin occurs. The drugs were given as single 20 min i.v. infusions in doses ranging from 1.9 to 2.8 g to nine healthy volunteers using a cross-over design. Plasma and urinary concentrations were determined by a selective liquid chromatographic method using frequent sampling up to 10 and 30 h respectively after termination of the infusion. The renal clearance of the drugs was independent of the plasma concentration. The mean (s.d.) renal clearances of ampicillin and amoxycillin were 167 (24) and 157 (20) ml min-1 1.73 m-2 respectively. The net secretion was about 50% of the total renal clearance of both drugs. The plasma concentration and urinary excretion rate versus time curves indicated a polyexponential decline, which could be described by both a biexponential and a triexponential equation. The former proved to be more reliable, especially in the calculation of micro rate constants. There was a tendency to more sustained plasma concentrations after amoxycillin, also illustrated by a significantly lower mean (s.d.) plasma clearance of this drug, viz. 185 (30) ml min-1 1.73 m-2, as compared to ampicillin, 210 (24) ml min-1 1.73 m-2 (P less than 0.04). There were no major differences in the disposition rate constants and the distribution volumes of ampicillin and amoxycillin. The mean (s.d.) plasma half-life was 1.7 (0.3) h for both drugs. The urinary excretion rate indicated a slower terminal disposition rate however, with ampicillin and amoxycillin half-lives of 3.4 (2.0) and 3.9 (1.2) h respectively. The longer half-life in the terminal phase may be due to increased tubular reabsorption at low urinary concentrations. It was not possible to determine in this study whether the half-life was affected by changes in clearance or volume of distribution. The urinary solubility of the drugs was dependent on pH. This could explain the massive macroscopic crystalluria seen in one subject after amoxycillin. Three hours after termination of the infusion, crystals could no longer be found in the sediment.
