Introduction: The intestinal tract of animals is a complex and dynamic microecosystem that is inextricably linked to the health of the host organism. Takin (Budorcas taxicolor) is a threatened species, and its gut microbiome is poorly understood. Therefore, this study aimed to analyze the microbial community structure and potential pathogens of takin. Methods: Takin fecal samples were collected from five sites in a nature reserve to ensure the uniformity of sample collection, determine the effects of different geographical locations on gut microbes, and analyze the differences in microbial communities between sites. Subsequently, high-throughput 16S rDNA gene sequencing was performed to analyze the microbial diversity and potential pathogens in the gut; the findings were verified by isolating and culturing bacteria and metagenomic sequencing. Results and discussion: The takin gut microflora consisted mainly of four phyla: Firmicutes (69.72%), Bacteroidota (13.55%), Proteobacteria (9.02%), and Verrucomicrobiota (3.77%), representing 96.07% of all microorganisms. The main genera were UCG-005 (20.25%), UCG-010_unclassified (12.35%), Firmicus_unclassified (4.03%), and Rumino coccsea_unclassified (3.49%), while the main species were assigned to Bacteria_unclassified. Potential pathogens were also detected, which could be used as a reference for the protection of takin. Pseudomonas presented the highest abundance at Shuichiping and may represent the main pathogen responsible for the death of takin at the site. This study aimed to explore the relationship between job-related stress and township teachers' professional well-being. Based on Job Demand-Resource Model, this study examined the mediating role of teachers' professional identity and the moderating role of perceived organizational support in this relationship. A total of 24,276 township teachers in China responded to the Teacher Stress Scale, the Teachers' Professional Identity Scale, the Teachers' Professional Well-Being Structure Questionnaire, and the Perceived Organizational Support Scale. Results showed that: (1) the professional wellbeing of township teachers differed significantly in terms of school type and demographic characteristics like age and gender; (2) job-related stress negatively predicted township teachers' professional well-being, with teachers' professional identity playing a mediating role; (3) the relation between job-related stress and teachers' professional identity was moderated by perceived organizational support; and (4) in the moderated mediation analysis, job-related stress positively predicted township teachers' professional well-being. These findings indicated that township teachers' professional well-being was influenced by both organizational and individual factors, which provided a theoretical basis and intervention pathways for improving township teachers' professional well-being. Background: Glioma as the most frequently discovered tumor affecting the brain shows significant morbidity and fatality rates with unfavorable prognosis. There is an urgent need to find novel therapeutic targets to overcome the low chemotherapeutic efficacy of glioma. This research examined whether the copper-metabolism-domain protein, COMMD4, had predictive and therapeutic significance in glioma. Methods: Using the freely accessible CGGA (The Chinese Glioma Atlas) and TCGA (The Cancer Genome Atlas) databases, we examined the function of COMMD4 in GBM and LGG. CIBERSORT and TIMER were utilized to assess the associations between COMMD4 and immune cells. The Gene Set Enrichment Analysis (GSEA) was employed to examine the functional data. Furthermore, the link between COMMD4 expression and predicted treatment response was evaluated via CellMiner Cross-Database. Meanwhile, qRT-PCR was conducted to examine COMMD4 expression in human glioma. Finally, Migration and invasion of glioma cells (U-87, U-251)were assessed using transwell assays. R was used to analyze the statistical data. Results: According to our findings, COMMD4 expression level was higher in patients having grade-dependent glioma who also showed an unfavorable prognosis. Furthermore, qRT-PCR confirmed the high expression of COMMD4 in glioma tissues and cells. Additionally, using integrated correlation analysis, we acquired significant prognostic findings between isocitrate dehydrogenase 1(IDH1) and COMMD4. Meanwhile, a link between COMMD4 and many tumor-infiltrating immune cells was observed. GSEA and drug response analysis revealed the potential mechanism of COMMD4 in drug resistance of glioma. Conclusion: The current findings validated COMMD4 as a novel biological marker, which might offer insights into the possible drug resistance mechanisms and the impact of the immune microenvironment on glioma. Background: Copper ions are essential for cellular physiology. Cuproptosis is a novel method of copper-dependent cell death, and the cuproptosis-based signature for glioma remains less studied. Methods: Several glioma datasets with clinicopathological information were collected from TCGA, GEO and CGGA. Robust Multichip Average (RMA) algorithm was used for background correction and normalization, cuproptosis-related genes (CRGs) were then collected. The TCGA-glioma cohort was clustered using ConsensusClusterPlus. Univariate Cox regression analysis and the Random Survival Forest model were performed on the differentially expressed genes to identify prognostic genes. The cuproptosissignature was constructed by calculating CuproptosisScore using Multivariate Cox regression analysis. Differences in terms of genomic mutation, tumor microenvironment, and enrichment pathways were evaluated between highor low-CuproptosisScore. Furthermore, drug response prediction was carried out utilizing pRRophetic. Results: Two subclusters based on CRGs were identified. Patients in cluster2 had better clinical outcomes. The cuproptosis-signature was constructed based on CuproptosisScore. Patients with higher CuproptosisScore had higher WHO grades and worse prognosis, while patients with lower grades were more likely to develop IDH mutations or MGMT methylation. Univariate and Multivariate Cox regression analysis demonstrated CuproptosisScore was an independent prognostic factor. The accuracy of the signature in prognostic prediction was further confirmed in 11 external validation datasets. In groups with high-CuproptosisScore, PIK3CA, MUC16, NF1, TTN, TP53, PTEN, and EGFR showed high mutation frequency. IDH1, TP53, ATRX, CIC, and FUBP1 demonstrated high mutation frequency in low-CuproptosisScore group. The level of immune infiltration increased as CuproptosisScore increased. SubMap analysis revealed patients with high-CuproptosisScore may respond to anti-PD-1 therapy. The IC50 values of Bexarotene, Bicalutamide, Bortezomib, and Cytarabine were lower in the high-CuproptosisScore group than those in the low-CuproptosisScore group. Finally, the importance of IGFBP2 in TCGAglioma cohort was confirmed. Conclusion: The current study revealed the novel cuproptosis-based signature might help predict the prognosis, biological features, and appropriate treatment for patients with glioma. This paper uses radial colour profiles to infer the distributions of dust, gas and star formation in z = 0.4-1.4 star-forming main-sequence galaxies. We start with the standard UVJ-based method to estimate dust extinction and specific star formation rate (sSFR). By replacing J with I band, a new calibration method suitable for use with ACS+WFC3 data is created (i.e. UVI diagram). Using a multi-wavelength multi-aperture photometry catalogue based on CANDELS (Cosmic Assembly Near-infrared Deep Extragalactic Legacy Survey), UVI colour profiles of 1328 galaxies are stacked in stellar mass and redshift bins. The resulting colour gradients, covering a radial range of 0.2-2.0 effective radii, increase strongly with galaxy mass and with global AV. Colour gradient directions are nearly parallel to the Calzetti extinction vector, indicating that dust plays a more important role than stellar population variations. With our calibration, the resulting AV profiles fall much more slowly than stellar mass profiles over the measured radial range. sSFR gradients are nearly flat without central quenching signatures, except for M⋆ > 1010.5M⊙, where central declines of 20-25 per cent are observed. Both sets of profiles agree well with previous radial sSFR and (continuum) AV measurements. They are also consistent with the sSFR profiles and, if assuming a radially constant gas-to-dust ratio, gas profiles in recent hydrodynamic models. We finally discuss the striking findings that SFR scales with stellar mass density in the inner parts of galaxies, and that dust content is high in the outer parts despite low stellar mass surface densities there. Light harvesting is the first step of photovoltaic process in polymer solar cells. However, such donor: acceptor bulk junction layers are usually featured with vertical phase segregation as well as film-depth-dependent molecular aggregation, chain orientation and crystallinity, leading to a significant variation of photon absorption and exciton generation at different film-depths. We propose an experimentally and numerically accessible method to investigate the depth-dependent light harvesting behaviors in the active layer in polymer solar cells. A low-pressure oxygen plasma is utilized to etch the active layer gradually which is monitored by a light absorption spectrometer. Including the obtained sublayer absorption spectra into transfer matrix optical model yields depth-dependent optical properties and exciton generation profiles, which contribute to quantum efficiency and short-circuit current. Compared with the one immobilized in the conventional mesoporous microspheres, the enzyme immobilized in gigaporous microspheres showed much higher activity and better stability. To gain a deeper understanding, we herein selected lipase as a prototype to comparatively analyze the adsorption behavior of lipase at interfaces in gigaporous and mesoporous polystyrene microspheres at very low lipase concentration, and further compared with the adsorption on a completely flat surface (a chip). Owing to the limited space of narrow pores, lipase molecules were inclined to be adsorbed as a monolayer in mesoporous microspheres. During this process, the interaction between lipase molecules and the interface was stronger, which could result in the structural change of lipase molecular and compromised specific activity. In addition to monolayer adsorption, more multilayer adsorption of enzyme molecules also occurred in gigaporous microspheres. Besides the adsorption state, the pore curvature also affected the lipase adsorption. Due to the multilayer adsorption, the excellent mass transfer properties for the substrate and the product in the large pores, and the small pore curvature, lipase immobilized in gigaporous microspheres showed better behaviors. However, as a key component of NADPH oxidase, how p47phox regulates NADPH oxidases activity, ROS production and adventitial fibroblasts (AFs) function remains unclear. Methods: p47phox in aortic arteries of apoE(-/-) mice fed with hyperlipid diet was detected by immunohistochemistry. NADPH oxidase activity, superoxide anion (O2-) generation and p47phox expression were analyzed in primary AFs treated by diphenyleneiodonium (DPI). The proliferation and migration of AFs were also analyzed. Results: p47phox expression was low in the aortic adventitia but high in the site of intimal injury with continuous hyperlipidic diet. Compared to AFs from wild-type mice, AFs derived from apoE(-/-) mice exhibited elevated NADPH oxidase activity, O2- production and higher mRNA and protein levels of p47phox, correlated with increased capability of proliferation and migration. DPI inhibited NADPH oxidase activity and AFs proliferation and migration in a dose-dependent manner. In addition, siRNA mediated knockdown of p47phox attenuated the proliferation and migration of AFs derived from apoE(-/-) mice. Conclusion: p47phox plays a critical role in the regulation of adventitial fibroblast proliferation and migration and may be a new therapeutic target for neointimal hyperplasia. The majority of massive star-forming galaxies at z ∼ 2 have velocity gradients suggestive of rotation, in addition to large amounts of disordered motions. In this paper, we demonstrate that it is challenging to distinguish the regular rotation of a disk galaxy from the orbital motions of merging galaxies with seeing-limited data. However, the merger fractions at z ∼ 2 are likely too low for this to have a large effect on measurements of disk fractions. To determine how often mergers pass for disks, we look to galaxy formation simulations. We analyze ∼24,000 synthetic images and kinematic maps of 31 high-resolution simulations of isolated galaxies and mergers at z ∼ 2. We determine if the synthetic observations pass the criteria commonly used to identify disk galaxies and whether the results are consistent with their intrinsic dynamical states. Galaxies that are intrinsically mergers pass the disk criteria for anywhere from 0% to 100% of sightlines. The exact percentage depends strongly on the specific disk criteria adopted and weakly on the separation of the merging galaxies. Therefore, one cannot tell with certainty whether observations of an individual galaxy indicate a merger or a disk. To estimate the fraction of mergers passing as disks in current kinematics samples, we combine the probability that a merger will pass as a disk with theoretical merger fractions from a cosmological simulation. Taking the latter at face value, the observed disk fractions are overestimated by small amounts: at most by 5% at high stellar mass (1010–11M⊙) and 15% at low stellar mass (109–10M⊙). At intermediate and high redshifts, measurements of galaxy star formation rates are usually based on rest-frame ultraviolet (UV) data. A correction for dust attenuation, AUV, is needed for these measurements. This correction is typically inferred from UV spectral slopes (β) using an equation known as “Meurer’s Relation.” In this paper, we study this relation at a redshift of 1.5 using images and photometric measurements in the rest-frame UV (HST) through mid-infrared (Spitzer). It is shown that massive star-forming galaxies (above ) have dust corrections that are dependent on their inclination to the line of sight. Edge-on galaxies have higher AUV and infrared excess (IRX = L(IR)/L(UV)) than face-on galaxies at a given β. Interestingly, dust corrections for low-mass star-forming galaxies do not depend on inclination. This is likely because more massive galaxies have more disk-like shapes/kinematics, while low-mass galaxies are more prolate and have more disturbed kinematics. To account for an inclination-dependent dust correction, a modified Meurer’s Relation is derived: , where b/a is the galaxy axis ratio. This inclination dependence of AUV can be explained by a two-component model of the dust distribution inside galaxies. In such a model, the dust attenuation of edge-on galaxies has a higher contribution from a mixture component (dust uniformly mixed with stars in the diffuse interstellar medium), and a lower contribution from a birth cloud component (near-spherical dust shells surrounding young stars in H ii regions) than that of face-on galaxies. We have measured the radial profiles of isophotal ellipticity (ε) and disky/boxy parameter A4 out to radii of about three times the semimajor axes for ∼4600 star-forming galaxies (SFGs) between redshift 0.5 and 1.8 in the CANDELS/GOODS-S and UDS fields. Based on the average size–mass relation in each redshift bin, we divide our galaxies at a given mass into Small SFGs (SSFGs; smaller than the average) and Large SFGs (LSFGs; larger than the average). We show that, at low masses (), the SSFGs generally have nearly flat ε and A4 profiles in both edge-on and face-on views, especially at . Moreover, the median A4 values at all radii are almost zero. In contrast, the highly inclined low-mass LSFGs in the same mass-redshift bins generally have monotonically increasing ε profiles with radius and disky feature dominated in the intermediate regions. These findings imply that at these redshifts, the low-mass SSFGs are not disk-like, whereas the low-mass LSFGs likely harbour disk-like components flattened by significant rotations. At high masses (), both highly inclined SSFGs and LSFGs generally exhibit distinct trends in both ε and A4 profiles, which increase at lower radii, reach maxima, then decrease at larger radii. Such the feature is more prevalent for more massive () galaxies or at lower redshifts (). This feature can be simply explained if galaxies possess all three components: central bulges, disks in the intermediate regions, and halo-like stellar components in the outskirts.
