The globally important carbon sink of intact, old-growth tropical humid forests is declining because of climate change, deforestation and degradation from fire and logging1–3. Recovering tropical secondary and degraded forests now cover about 10% of the tropical forest area4, but how much carbon they accumulate remains uncertain. Here we quantify the aboveground carbon (AGC) sink of recovering forests across three main continuous tropical humid regions: the Amazon, Borneo and Central Africa5,6. On the basis of satellite data products4,7, our analysis encompasses the heterogeneous spatial and temporal patterns of growth in degraded and secondary forests, influenced by key environmental and anthropogenic drivers. In the first 20 years of recovery, regrowth rates in Borneo were up to 45% and 58% higher than in Central Africa and the Amazon, respectively. This is due to variables such as temperature, water deficit and disturbance regimes. We find that regrowing degraded and secondary forests accumulated 107 Tg C year−1 (90–130 Tg C year−1) between 1984 and 2018, counterbalancing 26% (21–34%) of carbon emissions from humid tropical forest loss during the same period. Protecting old-growth forests is therefore a priority. Furthermore, we estimate that conserving recovering degraded and secondary forests can have a feasible future carbon sink potential of 53 Tg C year−1 (44–62 Tg C year−1) across the main tropical regions studied.Analysis of satellite-based data on recovering degraded and secondary forests in three tropical moist forest regions quantifies the amount of aboveground carbon accumulated, which counterbalanced one quarter of carbon emissions from old-growth forest loss between 1984 and 2018. Complex organ development depends on single lumen formation and its expansion during tubulogenesis. This can be achieved by correct mitotic spindle orientation during cell division, combined with luminal fluid filling that generates hydrostatic pressure. Using a human 3D cell culture model, we have identified two regulators of these processes. We find that pleckstrin homology leucine-rich repeat protein phosphatase (PHLPP) 2 regulates mitotic spindle orientation, and thereby midbody positioning and maintenance of a single lumen. Silencing the sole PHLPP family phosphatase in Drosophila melanogaster, phlpp, resulted in defective spindle orientation in Drosophila neuroblasts. Importantly, cystic fibrosis transmembrane conductance regulator (CFTR) is the main channel regulating fluid transport in this system, stimulated by phosphorylation by protein kinase A and inhibited by the AMP-activated protein kinase AMPK. During lumen expansion, CFTR remains open through the action of PHLPP1, which stops activated AMPK from inhibiting ion transport through CFTR. In the absence of PHLPP1, the restraint on AMPK activity is lost and this tips the balance in the favour of channel closing, resulting in the lack of lumen expansion and accumulation of mucus. This study assessed the impact of dietary supplementation of single‐ and multi‐strain Bacillus‐based probiotics on growth performance, intestinal morphometry, blood parameters, immune‐oxidative status and transcriptomic response of Nile tilapia for 60 days. Fish (N = 150, 1.632 ± 0.035 g) were allocated into five experimental groups: (1) Control group (CG) that was fed on a basal diet; (2) Eco. 0.01%, received basal diet with 0.1 g/kg Ecobiol; (3) Eco. 0.02%, received basal diet with 0.2 g/kg Ecobiol; (4) Pro‐F FMC 0.001%, received basal diet with 0.001 g/kg Sanolife PRO‐F FMC; (5) Pro‐F FMC 0.002%, received basal diet with 0.002 g/kg Pro‐F‐FMC. Fish received Pro‐F FMC 0.002% displayed higher growth performance than CG (p < 0.05), while Pro‐F FMC 0.001% exhibited the highest intestinal weight. Sanolife PRO‐F FMC supplementation improved intestinal villi length and goblet cell number while decreased inter‐villi space (p < 0.05). The best haemoglobin, haematocrit and monocyte per cent were observed in Pro‐F FMC 0.001%, whereas Pro‐F FMC 0.002% revealed the best leukocyte, lymphocyte and neutrophil contents. Pro‐F FMC‐supplemented groups showed better phagocytic activity, phagocytic index and lysozyme activity than Ecobiol‐supplemented groups and CG. Pro‐F FMC 0.001% enrichment induced the highest upregulation level of NPYα, AgRp‐2, ghrelin and GH‐1, whereas Pro‐F FMC 0.002% significantly upregulated IGF‐1 and IGF‐2 gene expression. Eco. 0.02% induced the highest upregulation level of orexin and galanin gene expression. Conclusively, dietary enrichment with single‐ or multi‐strain Bacillus‐based probiotics is likely to improve growth, immunological parameters and transcriptomic response of Nile tilapia. The present research was planned to assess the harmful effects of heavy metals as well as antioxidant alterations of Oreochromis niloticus collected from two polluted fish farms located in Abbassa and Manzala (areas A and B, respectively) in Egypt. They were compared with reference site in central laboratory for aquaculture researchers during the summer season, 2020. A total number of 75 fish collected from all sites was subjected to clinical, postmortem, and bacteriological examination. Water parameters, as well as heavy metals (arsenic, manganese, and nickel) in water, serum, and tissues (muscle, gills, and liver), were evaluated. In addition, antioxidant enzymes (SOD and CAT) and (MDA) were assessed in the serum of O. niloticus. Bacterial isolates, Aeromonas hydrophila, were identified in fish at both localities, and the recorded mortality rate was about 30% and 5% in areas A and B, respectively. The levels of As in water samples from both polluted areas were higher than the permissible limits. In addition, serum of O. niloticus showed a significant decrease in the concentrations of Ni in both polluted areas. In addition, a significant decrease in the mean levels of Ni and As was found in the gills and muscles of O. niloticus in both polluted areas. Tropical secondary forests sequester carbon up to 20 times faster than old-growth forests. This rate does not capture spatial regrowth patterns due to environmental and disturbance drivers. Here we quantify the influence of such drivers on the rate and spatial patterns of regrowth in the Brazilian Amazon using satellite data. Carbon sequestration rates of young secondary forests (<20 years) in the west are ~60% higher (3.0 ± 1.0 Mg C ha−1 yr−1) compared to those in the east (1.3 ± 0.3 Mg C ha−1 yr−1). Disturbances reduce regrowth rates by 8–55%. The 2017 secondary forest carbon stock, of 294 Tg C, could be 8% higher by avoiding fires and repeated deforestation. Maintaining the 2017 secondary forest area has the potential to accumulate ~19.0 Tg C yr−1 until 2030, contributing ~5.5% to Brazil's 2030 net emissions reduction target. Implementing legal mechanisms to protect and expand secondary forests whilst supporting old-growth conservation is, therefore, key to realising their potential as a nature-based climate solution. This study uses regional and global remote sensing data to assess the regrowth of secondary forests in the Brazilian Amazon biome. Objectives. To describe methods employed to track infants enrolled in the New York State Zika Pregnancy and Infant Registry (NYSZPIR) and demonstrate the benefits of population databases to improve the process. Methods. We used patient medical records and provider outreach, New York State Immunization Information System (NYSIIS), and New York State Early Hearing Detection and Intervention Information System (NYEHDI-IS) to gather medical information. We used descriptive statistics to summarize variables and the McNemar test to determine statistical significance (P <.05). Results. We identified 109 live births from NYSZPIR mothers. Provider information was documented for 106 (97.2%) infants in NYSIIS compared with 72 (66.1%) through chart review. Collected results of newborn hearing screening increased from 82 (75.2%) to 106 (97.2%) using NYEHDI-IS. The amount of data obtained was significantly higher (P <.001) when including NYSIIS and NYEHDI-IS compared with using medical records alone. Conclusions. Public health surveillance systems can be used to track infants using data sources such as NYSIIS and NYEHDI-IS in addition to traditional methods. One of the top challenges in education and neuroscience consists in translating laboratory results into strategies to improve learning and memory in teaching environments. In that sense, during the last two decades, researchers have discovered specific temporal windows around learning, during which the intervention with some experiences induces modulatory effects on the formation and/or persistence of memory. Based on these results, the aim of the present study was to design a specific strategy to improve the memory of students in a high-school scenario, by assessing the effect of a novel situation experienced close to learning. We found that the long-term memory about a geometrical figure was more precise in the group of students that faced a novel situation 1 h before or after learning the figure than the control group of students who did not face the novelty. This enhancement was probably triggered by processes acting on memory formation mechanisms that remained evident 45 days after learning, indicating that the improvement was sustained over time. In addition, our results showed that novelty no longer improved the memory if it was experienced 4 h before or after learning. However, far beyond this window of efficacy, when it was faced around 10 h after learning, the novel experience improved the memory persistence tested 7 days later. In summary, our findings characterized different temporal windows of the effectiveness of novelty acting on memory processing, providing a simple and inexpensive strategy that could be used to improve memory formation and persistence in high-school students. Wind energy generation has become an important means to reduce reliance on fossil fuels and mitigate against human‐induced climate change, but could also represent a significant human–wildlife conflict. Airborne taxa such as birds may be particularly sensitive to collision mortality with wind turbines, yet the relative vulnerability of species' populations across their annual life cycles has not been evaluated.Using GPS telemetry, we studied the movements of lesser black‐backed gulls Larus fuscus from three UK breeding colonies through their annual cycle. We modelled the distance travelled by birds at altitudes between the minimum and maximum rotor sweep zone of turbines, combined with the probability of collision, to estimate sensitivity to collision. Sensitivity was then combined with turbine density (exposure) to evaluate spatio‐temporal vulnerability.Sensitivity was highest near to colonies during the breeding season, where a greater distance travelled by birds was in concentrated areas where they were exposed to turbines.Consequently, vulnerability was high near to colonies but was also high at some migration bottlenecks and wintering sites where, despite a reduced sensitivity, exposure to turbines was greatest.Synthesis and applications. Our framework combines bird‐borne telemetry and spatial data on the location of wind turbines to identify potential areas of conflict for migratory populations throughout their annual cycle. This approach can aid the wind farm planning process by: (a) providing sensitivity maps to inform wind farm placement, helping minimize impacts; (b) identifying areas of high vulnerability where mitigation warrants exploration; (c) highlighting potential cumulative impacts of developments over international boundaries and (d) informing the conservation status of species at protected sites. ABSTRACT Conflicts in resource use between humans and wildlife populations are increasingly determined through quantitative approaches. To better understand interactions between birds and human activities in the marine environment, telemetry is routinely used to characterize the area use of species, but evaluations are often based on a small number of individuals taken as representative of a local population studied. Furthermore, the relative importance of the number of animals required and for what duration they should be tracked has received little attention. We examined the central-place foraging movements of 24 lesser black-backed gulls ( Larus fuscus) from a protected population from 1 March to 31 August during 2010-2013. Using bootstrapping and non-linear modeling, we investigated whether sample sizes were sufficient to characterize offshore area use by considering the cumulative area use for an increasing number of birds and duration of tracking. Box-and-whisker analysis suggested a minimum of 13 birds and a precautionary upper maximum of 41 birds were needed to describe 95% of the estimated area use of the population (defined by 100% occupancy). Tracking fewer birds for longer was more important than tracking more birds for less time. A period of 145 days was required to characterize area use for 13-41 birds; however, offshore areas were used primarily after May, meaning that a 97-day tracking period from May onwards was also representative. Predicted and observed areas were strongly correlated, and the predicted area of 15 birds for 151 days was 91% of the total estimated for the population. These findings suggest that the data were suitable for determining interaction with offshore developments, and were characteristic of the population. This study has revealed the power of a long-term tracking dataset, and has uncovered further complexities surrounding study design and analysis that may shape conclusions drawn. Wind energy generation is increasing globally, and associated environmental impacts must be considered. The risk of seabirds colliding with offshore wind turbines is influenced by flight height, and flight height data usually come from observers on boats, making estimates in daylight in fine weather. GPS tracking provides an alternative and generates flight height information in a range of conditions, but the raw data have associated error., Here, we present a novel analytical solution for accommodating GPS error. We use Bayesian state-space models to describe the flight height distributions and the error in altitude measured by GPS for lesser black-backed gulls and great skuas, tracked throughout the breeding season. We also examine how location and light levels influence flight height., Lesser black-backed gulls flew lower by night than by day, indicating that this species would be less likely to encounter turbine blades at night, when birds' ability to detect and avoid them might be reduced. Gulls flew highest over land and lowest near the coast. For great skuas, no significant relationships were found between flight height, time of day and location., We consider four 'collision risk windows', corresponding to the airspace swept by rotor blades for different offshore wind turbine designs. We found the highest proportion of birds at risk for a 22-250 m turbine (up to 9% for great skuas and 34% for lesser black-backed gulls) and the lowest for a 30-258 m turbine. Our results suggest lesser black-backed gulls are at greater risk of collision than great skuas, especially by day., Synthesis and applications. Our novel modelling approach is an effective way of resolving the error associated with GPS tracking data. We demonstrate its use on GPS measurements of altitude, generating important information on how breeding seabirds use their environment. This approach and the associated data also provide information to improve avian collision risk assessments for offshore wind farms. Our modelling approach could be applied to other GPS data sets to help manage the ecological needs of seabirds and other species at a time when the pressures on the marine environment are growing. Recent developments in GPS tracking technology allow the movements of bird species to be followed in ever-greater detail. Seabird research is benefiting greatly, due to the challenges of tracking species that often roam widely out at sea. Amongst the gulls, one of the pressing issues is to understand the ecology of the relatively recent urban colonists and how they differ from their counterparts in traditional rural colonies. Here, we present what we believe are the first GPS results from roof-nesting gulls. Four adult Herring Gulls (two males, two females) were fitted with GPS tags in May 2014 in the seaside town of St Ives, Cornwall (breeding colony c 250 pairs), and tracked for c 100 days during the 2014 breeding season. We estimated the home ranges of the four individuals and how their movement behaviour varied through the 24-h period and across the breeding season. The results highlight how variable movement behaviour was among individuals: whilst one bird roamed widely (90% range estimate = 560 km2), heading >50 km offshore and often active at night or roosting at sea, two birds had small ranges (<10 km2), always attended the colony at night and rarely headed more than a few hundred metres offshore, with the fourth displaying intermediate behaviour. All of the birds regularly utilised a few key sites within the agricultural landscape south of St Ives. Whilst this study was too small to allow general conclusions to be drawn about urban Herring Gulls, it reinforces how variable individual behaviour can be amongst the large gulls and will be particularly interesting when applied to a larger sample of birds, especially in big urban gull colonies further inland. Telemetry has become an important method for studying the biology and ecology of animals. However, the impact of tracking devices and their method of attachment on different species across multiple temporal scales has seldom been assessed. We compared the behavioural and demographic responses of two species of seabird, Lesser Black-backed Gull Larus fuscus and Great Skua Stercorarius skua, to a GPS device attached using a crossover wing harness. We used telemetry information and monitoring of breeding colonies to compare birds equipped with a device and harness, and control birds without an attachment. We assessed whether tagged birds have lower short-term breeding productivity or lower longer term overwinter return rates (indicative of overwinter survival) than controls. For Great Skua, we also assessed whether territory attendance within the breeding season differed between tagged and control birds. As with previous studies on Lesser Black-backed Gull, we found no short-term impacts on breeding productivity or long-term impacts on overwinter return rates. For Great Skua, there was no evidence for impacts of the device and harness on territory attendance or breeding productivity. However, as found by a previous study of Great Skuas using a different (body) harness design, there was strong evidence of reduced overwinter return rates. Consequently, a device attached using a wing harness was considered suitable for long-term deployment on Lesser Black-backed Gulls, but not on Great Skuas. Tracking devices are useful in studying the movement and behaviour of birds throughout the year. However, the effects of device attachment must be monitored for bird welfare and the scientific veracity of information gathered. We report on a trial of three different harness types (leg-loop, body and wing) for attaching GPS devices to Lesser Black-backed Gulls Larus fuscus and of a leg-loop harness on Great Skuas Stercorarius skua. This trial evaluated the functionality of the device whilst deployed, harness retention and the effects of each harness on the birds. The leg-loop harness prevented the devices from charging, due to feathers overlapping their solar panels, and individuals of both species shed this attachment during the first breeding season. For Lesser Black-backed Gull, devices functioned properly when attached using body and wing harnesses; foraging behaviour and territory attendance of birds were similar to the findings of other studies. Despite small sample sizes, annual adult survival rates for Lesser Black-backed Gull were similar to published estimates, suggesting harnesses had no deleterious effects. The wing harness has a smaller amount of skin contact than the body harness, and a non-constricting fit that better accommodates changes in body size. Previously, we have shown that the phosphoinositidemetabolizing enzymes PIKfyve (phosphoinositide 5-kinase, FYVE finger containing) and MTMR3 (myotubularin-related protein 3), together with their lipid product PtdIns5P, are important for migration of normal human fibroblasts. As these proteins are a kinase and a phosphatase respectively, and thereby considered druggable, we wanted to test their involvement in cancer cell migration and invasion. First, we showed that PIKfyve and MTMR3 are expressed in most cancer cells. Next, we demonstrated that depletion of PIKfyve or MTMR3 resulted in decreased velocity in three different cancer cell lines by using new software for cell tracking. Inhibition of the enzymatic activity of PIKfyve by the inhibitor YM201636 also led to a strong reduction in cell velocity. Mechanistically, we show that PIKfyve and MTMR3 regulate the activation of the Rho family GTPase Rac1. Further experiments also implicated PtdIns5P in the activation of Rac1. The results suggest a model for the activation of Rac1 in cell migration where PIKfyve and MTMR3 produce PtdIns5P on cellular membranes which may then serve to recruit effectors to activate Rac1. Finally, in an invasion assay, we demonstrate that both PIKfyve and MTMR3 are implicated in invasive behaviour of cancer cells. Although phosphatidylinositol 5-phosphate (PtdIns5P) is present in many cell types and its biogenesis is increased by diverse stimuli, its precise cellular function remains elusive. Here we show that PtdIns5P levels increase when cells are stimulated to move and we find PtdIns5P to promote cell migration in tissue culture and in a Drosophila in vivo model. First, class III phosphatidylinositol 3-kinase, which produces PtdIns3P, was shown to be involved in migration of fibroblasts. In a cell migration screen for proteins containing PtdIns3P-binding motifs, we identified the phosphoinositide 5-kinase PIKfyve and the phosphoinositide 3-phosphatase MTMR3, which together constitute a phosphoinositide loop that produces PtdIns5P via PtdIns(3,5)P2. The ability of PtdIns5P to stimulate cell migration was demonstrated directly with exogenous PtdIns5P and a PtdIns5P-producing bacterial enzyme. Background: Androgen receptor (AR) and the phosphatidylinositol-3 kinase (PI3K) signaling are two of the most important pathways implicated in prostate cancer. Previous work has shown that there is crosstalk between these two pathways; however, there are conflicting findings and the molecular mechanisms are not clear. Here we studied the AR–PI3K pathway crosstalk in prostate cancer cells in vitro as well as in vivo. Methods: Quantitative PCR, Western analysis, reporter assays, and proliferation analyses in vitro and in vivo were used to evaluate the effect of PI3K pathway inhibition on AR signaling and cell growth. Results: Transcriptional activity of AR was increased when the PI3K pathway was inhibited at different levels. In the androgen responsive prostate cancer cell line LNCaP, androgen and the mTOR inhibitor rapamycin synergistically activated androgen target genes. Despite increased androgen signaling, rapamycin treatment reduced LNCaP cell growth; the AR antagonist bicalutamide potentiated this effect. Furthermore, the rapamycin derivative CCI-779 reduced the growth of CWR22 prostate cancer xenografts while increasing AR target gene expression. Conclusion: These findings suggest that inhibition of the PI3K pathway activates AR signaling. Despite the increase in AR signaling which has proliferative effects, the result of PI3K pathway inhibition is antiproliferative. These findings suggest that the PI3K pathway is dominant over AR signaling in prostate cancer cells which should be considered in developing novel therapeutic strategies for prostate cancer. The article presents a study which examines the function of differently coloured bill tips, spots and stripes found on the beaks of breeding gulls. For the study, researchers analyzed continuous variables such as adult gull body length of both sexes and egg length. Through the use of the Integrated Taxonomic Information System, they gathered data for 45 species of birds. They found no significant differences in either body length or egg length among species with coloured bill tip, a black stripe or combined.

The ESCRT (endosomal sorting complex required for transport) machinery consists of four protein complexes that mediate sorting of ubiquitinated membrane proteins into the intraluminal vesicles of multivesicular endosomes, thereby targeting them for degradation in lysosomes. In the present paper, we review how ESCRT-mediated receptor down-regulation affects signalling downstream of Notch and growth factor receptors, and how ESCRTs may control cell proliferation, survival and cytoskeletal functions and contribute to tumour suppression. Rats exposed to a novel environment just prior to or 1-2 h, but not 4 or 6 h, before retention testing exhibited an enhanced retrieval of a one-trial inhibitory avoidance training. The bilateral intrahippocampal infusion of PD098059, an inhibitor of mitogenactivated protein kinase (MAPK), the specific upstream activator of p42 and p44 MAPKs, given 10 min before the exposure to the novel environment, blocked the enhancing effect of novelty on memory retrieval. In addition, prenovelty infusion of DL-2amino-5-phosphonovalerate (APV), an antagonist of glutamate NMDA receptors, produced similar effects. The exposure to the novel environment is associated with an activation of p42 and p44 MAPKs and an increase in the phosphorylation state of the transcription factor cAMP response element binding protein (CREB). No changes were observed in cAMP-dependent protein kinase (PKA) activity or in α-CAMKII activation. Taken together, our results indicate that novelty activates hippocampal MAPKs, which are necessary, along with glutamate NMDA receptors, for the enhancing effect of novelty on retrieval. The influence of a given event on long-term memory formation of another one has been a relevant topic of study in the neuroscience field in recent years. Students at school learn contents which are usually tested in exam format. However, exam elevates the arousal state of the students acting as a mild stressor that could influence another memory formation ongoing process. Thus, in this study we examine in high school students the effect of exams on long-term retention of unrelated information, learned at different times before or after the exams. Our results show that exams are not innocuous and that they could improve or reduce the retention of temporarily associated content. These effects did not show gender differences. Our findings should alert teachers about the side effects of exams on the learning of other content within the same school day. [ABSTRACT FROM AUTHOR]
