Our recently published [Weike et al., J. Chem. Phys. 159, 244119 (2023)] spin–orbit coupled diabatic potential energy model for HI is used in a thorough analysis of bound and quasi-bound states as well as elastic and inelastic processes in H + I collisions. The potential energy model, designed explicitly for studying scattering, accurately describes the various couplings in the system, which lead to complex dynamics. Ro-vibronic bound and quasi-bound states related to the adiabatic electronic ground state and an excited electronic state are analyzed. Calculations using the full 104 × 104 diabatic matrix model or a single adiabatic state are compared in order to investigate approximations in the latter. Elastic and inelastic scattering cross sections as well as thermal rates between the ground and first excited fine structure levels of iodine are computed for collision energies up to 12 500 cm−1. Resonances related to the quasi-bound states are analyzed in terms of their energy, width, lifetime, and decay probabilities. The effect of different resonances on the thermal rates is discussed. Resonances between 30 000 and 40 000 cm−1 are also studied for selected values of the total angular momentum, in particular their decay probabilities into different final states of iodine and hence their potential effect on branching ratios in photodissociation of HI. The scattering of H by I is a prototypical model system for light-heavy scattering in which relativistic coupling effects must be taken into account. Scattering calculations depend strongly on the accuracy of the potential energy surface (PES) model. The methodology to obtain such an accurate PES model suitable for scattering calculations is presented, which includes spin–orbit (SO) coupling within the Effective Relativistic Coupling by Asymptotic Representation (ERCAR) approach. In this approach, the SO coupling is determined only for the atomic states of the heavy atom, and the geometry dependence of the SO effect is accounted for by a diabatization with respect to asymptotic states. The accuracy of the full model, composed of a Coulomb part and the SO model, is achieved in the following ways. For the SO model, the extended ERCAR approach is applied, which accounts for both intra-state and inter-state SO coupling, and an extended number of diabatic states are included. The corresponding coupling constants for the SO operator are obtained from experiments, which are more accurate than computed values. In the Coulomb Hamiltonian model, special attention is paid to the long range behavior and accurate c6 dispersion coefficients. The flexibility and accuracy of this Coulomb model are achieved by combining partial models for three different regions. These are merged via artificial neural networks, which also refine the model further. The nitrate radical NO3 plays an important role in atmospheric chemistry, yet many aspects of its coupled and anharmonic vibronic structure remain elusive. Here, using an accurate, coupled full-dimensional diabatic potential that includes five electronic states, we revisit the vibronic spectrum associated with the electronic state. Using recently developed tensor network state methods, we are able to compute more than 2500 vibronic states, thereby increasing the number of computed full-dimensional states by a factor of 50, compared to previous work. While we obtain good agreement with experiment for most of the assigned vibronic levels, for several others, we observe striking disagreement. Further, for the antisymmetric bending motion we find remarkably large symmetry-induced level splittings that are larger than the zero-order reference. We discuss non-negligible nonadiabatic effects and show that the Born–Oppenheimer approximation leads to significant errors in the spectrum. [ABSTRACT FROM AUTHOR]

Microcephalic osteodysplastic primordial dwarfism type I (MOPDI) is a very rare and severe autosomal recessive disorder characterized by marked intrauterine growth retardation, skeletal dysplasia, microcephaly and brain malformations. MOPDI is caused by biallelic mutations in RNU4ATAC, a non‐coding gene involved in U12‐type splicing of 1% of the introns in the genome, which are recognized by their specific splicing consensus sequences. Here, we describe a unique observation of immunodeficiency in twin sisters with mild MOPDI, who harbor a novel n.108_126del mutation, encompassing part of the U4atac snRNA 3′ stem‐loop and Sm protein binding site, and the previously reported n.111G>A mutation. Interestingly, both twin sisters show mild B‐cell anomalies, including low naive B‐cell counts and increased memory B‐cell and plasmablasts counts, suggesting partial and transitory blockage of B‐cell maturation and/or excessive activation of naive B‐cells. Hence, the localization of a mutation in stem II of U4atac snRNA, as observed in another RNU4ATAC‐opathy with immunodeficiency, that is, Roifman syndrome (RFMN), is not required for the occurrence of an immune deficiency. Background: Identifying valid and accessible tools for monitoring and improving physical activity levels is essential for promoting functional ability and healthy aging. The Physical Activity Scale for the Elderly (PASE) is a commonly used and recommended self-report measure of physical activity in older adults. The objective of this scoping review was to map the nature and extent to which the PASE has been used in the literature on community-dwelling older adults, including the evidence for its psychometric properties. Methods: Seven electronic databases (MEDLINE (Ovid), Embase (Ovid), AMED (Ovid), Emcare (Ovid), CINAHL (EBSCO), Ageline (EBSCO)) were searched from inception to January 25, 2023. Studies were included if physical activity was part of the aim(s) and measured using the PASE, participants had a mean age of 60 years or older and lived in the community, and papers were peer-reviewed journal articles published in English. Pairs of independent reviewers screened abstracts, full-texts, and extracted data. Where possible, weighted mean PASE scores were calculated for different subgroups based on age, sex, and clinical population. Results: From 4,124 studies screened, 232 articles from 35 countries met the inclusion criteria. Most studies were cross-sectional (60.78%), completed in high-income countries (86.4%) and in North America (49.57%). A variety of clinical conditions were included (n = 21), with the most common populations being osteoarthritis (n = 13), Parkinson’s disease (n = 11), and cognitive impairment (n = 7). Psychometric properties of ten versions of the PASE were found. All versions demonstrated acceptable test-retest reliability. Evidence for construct validity showed moderate correlations with self-reported physical activity, fair to moderate with accelerometry derived activity and fair relationships with physical function and self-reported health. Pooled means were reported in graphs and forest plots for males, females, age groups, and several clinical populations. Conclusion: The PASE was widely used in a variety of clinical populations and geographical locations. The PASE has been culturally adapted to several populations and evaluated for its reliability and convergent validity; however, further research is required to examine responsiveness and predictive validity. Prenatal and infant exposure to drugs of abuse is an emerging social and public health problem affecting children health and which may relate to child abuse and neglect. Exposure to drugs of abuse may occur through different routes, including intrauterine, breastfeeding, accidental intake, passive inhalation, and intentional administration. Currently, cases of suspected exposure can be investigated by hair toxicological analysis, the interpretation of which is, however, often difficult, leading to consequent difficulties in the management of such cases. In order to provide a contribution in terms of interpretation of the analytical results, this study aimed to search for the possible existence of elements, from a toxicological point of view, indicative towards the route of exposure. A retrospective study was performed on cases of suspected exposure to drugs of abuse in children aged 0–1 year, evaluated at a University Hospital between 2018 and 2022. Data of children hair toxicological analysis were analyzed and then compared with those of their mothers, when available; 41.6% children tested positive for cocaine. The study found a significant correlation between cocaine and benzoylecgonine concentrations, and a benzoylecgonine/cocaine ratio that tends to decrease as the age of children increases. From the comparison with mothers, a child/mother cocaine concentration ratio lower than 1 was found in all cases of hair sampled within the first week of life, and a ratio greater than or equal to 1 in all cases in which the sampling was performed later. These results, if confirmed in a larger cohort, could represent a contribution in the interpretation of cases of infant exposure to drugs of abuse and be integrated in the context of their multidisciplinary evaluation. Novel therapeutic strategies against difficult-to-treat bacterial infections are desperately needed, and the faster and cheaper way to get them might be by repurposing existing antibiotics. Nanodelivery systems enhance the efficacy of antibiotics by guiding them to their targets, increasing the local concentration at the site of infection. While recently described nanodelivery systems are promising, they are generally not easy to adapt to different targets, and lack biocompatibility or specificity. Here, nanodelivery systems are created that source their targeting proteins from bacteriophages. Bacteriophage receptor-binding proteins and cell-wall binding domains are conjugated to nanoparticles, for the targeted delivery of rifampicin, imipenem, and ampicillin against bacterial pathogens. They show excellent specificity against their targets, and accumulate at the site of infection to deliver their antibiotic payload. Moreover, the nanodelivery systems suppress pathogen infections more effectively than 16 to 32-fold higher doses of free antibiotics. This study demonstrates that bacteriophage sourced targeting proteins are promising candidates to guide nanodelivery systems. Their specificity, availability, and biocompatibility make them great options to guide the antibiotic nanodelivery systems that are desperately needed to combat difficult-to-treat infections. Zhao et al. demonstrate a strategy that delivers antibiotics to the site of infection using proteins from bacteriophages, which naturally target bacteria. Purpose: We compared 12-month outcomes of eyes with polypoidal choroidal vasculopathy (PCV) with or without complete regression of polyps observed one month after three monthly intravitreal administrations (loading phase) of aflibercept (2.0 mg/0.05 mL) or brolucizumab (6.0 mg/0.05 mL). Methods: All patients underwent indocyanine green angiography at both baseline and 3 months after initial injection and were followed up monthly with an as-needed regimen for up to 12 months. A total of 62 patients with PCV were included: 30 eyes were treated with brolucizumab, and 32 were treated with aflibercept. Eyes with complete regression of polyps (regression group) had significantly smaller maximum polyp diameter and were more frequently treated with brolucizumab than those without complete regression (non-regression) group. Results: Best corrected visual acuity was comparable between the two groups at 12 months. Although the 12-month retreatment-free proportion was comparable between the two groups (33.0% versus 27.0%, p = 0.59), a retreatment-free period was significantly longer in the regression group than in the non-regression group (8.3 ± 3.3 versus 6.5 ± 3.6 months, p = 0.022), and the number of additional injections was significantly fewer in the regression group than in the non-regression group (1.2 ± 1.2 versus 3.0 ± 2.6, p = 0.007). Conclusions: Complete regression of polyps observed after the initial phase possibly prolongs the retreatment-free period and reduces the number of additional injections irrespective of aflibercept or brolucizumab. The primary source of hard fiber globally is Agave sisalana Perrine, also known as sisal. In areas where sisal is grown, the roots of the plant are usually left in the field after it has stopped producing, which leads to soil degradation and decreased sisal productivity. It is, therefore, critical to find alternatives to reuse this waste. This study explores the potential use of sisal waste in the cosmetic industry by incorporating a hydroethanolic extract (HER) into a cream–gel formulation, taking advantage of the plant's recognized ethnopharmacological value. The study involves analyzing the extract's phytochemical composition (flavonoids) and evaluating its cytotoxicity. Subsequently, the antioxidant and antiglycation activities of the extract and cream–gel are evaluated, as well as ex vivo ocular toxicity, photoprotective activity, and preliminary stability analyses. The HER extract showed a flavonoid composition (catechin, kaempferol, isorhamnetin, and chrysin) and maintained cell viability above 70% throughout all time points analyzed in the MTT assay. Furthermore, the extract and the formulation demonstrated proven antioxidant and antiglycation activities. The cream–gel's UVB and UVA protection effectiveness with the HER was comparable to that of synthetic UVB/UVA sunscreens, with the samples proving nonirritating and stable. In conclusion, the extract has a significant presence of flavonoids, and the cream–gel developed with it did not present cytotoxicity and met the stability requirements, indicating phytocosmetic potential with antioxidant, antiglycation, and photoprotective properties. Context. The collisional (de-)excitation of H2 by He plays an important role in the thermal balance and chemistry of various astro-physical environments, making accurate rate coefficients essential for interpreting observations of the interstellar medium. Aims. Our goal is to utilize a state-of-the-art potential energy surface (PES) to provide comprehensive state-to-state rate coefficients for He-induced transitions among rovibrational levels of H2. Methods. We performed quantum scattering calculations for the H2-He system. Thus, we were able to provide state-to-state rate coefficients for 1059 transitions between rovibrational levels of H2, with internal energies up to ≃15 000 cm−1, for temperatures ranging from 20 to 8000 K. Results. Our results demonstrate a good agreement with previous calculations for pure rotational transitions between low-lying rotational levels. However, we do find significant discrepancies for rovibrational processes involving highly-excited rotational and vibrational states. We attribute these differences to two key factors: (1) the broader range of intramolecular distances covered by ab initio points and (2) the superior accuracy of the PES, resulting from the utilization of the state-of-the-art quantum chemistry methods, compared to previous lower-level calculations. Radiative transfer calculations performed with the new collisional data indicate that the population of rotational levels in excited vibrational states experiences significant modifications, highlighting the critical need for this updated dataset in models of high-temperature astrophysical environments. Therapeutic applications of nuclease-based genome editing would benefit from improved methods for transgene integration via homology-directed repair (HDR). To improve HDR efficiency, we screened six small-molecule inhibitors of DNA-dependent protein kinase catalytic subunit (DNA-PKcs), a key protein in the alternative repair pathway of non-homologous end joining (NHEJ), which generates genomic insertions/deletions (INDELs). From this screen, we identified AZD7648 as the most potent compound. The use of AZD7648 significantly increased HDR (up to 50-fold) and concomitantly decreased INDELs across different genomic loci in various therapeutically relevant primary human cell types. In all cases, the ratio of HDR to INDELs markedly increased, and, in certain situations, INDEL-free high-frequency (>50%) targeted integration was achieved. This approach has the potential to improve the therapeutic efficacy of cell-based therapies and broaden the use of targeted integration as a research tool. The first aim of the study was to identify sex differences in the use of psychoactive substances among subjects with a previous driving under the influence (DUI) episode. The secondary objective was to propose specific strategies for medico-legal improvements. This was a retrospective observational study that took place between June 1, 2019, and August 31, 2023. It was conducted on DUI subjects examined for reinstatement of their driver's license using an integrated medico-legal and toxicological approach. Ethyl glucuronide (EtG) and illicit psychoactive substances were determined from hair samples. We performed descriptive statistical analyses for the entire sample as well as separately by sex. Additionally, we conducted binary logistic regression analyses separately for males and females to identify protective/risk factors associated with previous road accidents and judgments of unfitness to drive due to excessive alcohol consumption (EtG ≥ 30 pg/mg). The study included 2,221 subjects, comprising 1,970 men and 251 women. Men exhibited a higher prevalence of tobacco, alcohol, and illicit psychoactive substance use. Women were more frequently co-users of alcohol and psychoactive substances and involved in road accidents at the time of DUI. Among the men, being married or having a partner was found to be a protective factor concerning past traffic accidents. For both sexes, a DUI episode with a blood alcohol concentration (BAC) exceeding 1.5 g/L or the co-ingestion of alcohol and drugs was identified as a risk factor for road accident involvement. For men, smoking more than 20 cigarettes per day and, for women, having a DUI episode with a BAC over 1.5 g/L were the main factors indicating unfitness to drive, as determined through high hair EtG levels (> 30 pg/mg). Women with a previous history of road accidents were less likely to have EtG levels of 30 pg/mg or more. The study confirmed sex differences in subjects with a previous DUI episode. A BAC exceeding 1.5 g/L or the simultaneous use of alcohol and drugs at the time of DUI necessitate careful assessment of both men and women seeking driver's license reinstatement. In women, a BAC exceeding 1.5 g/L is considered a risk factor for a subsequent judgment of unfitness to drive. The medico-legal assessment should also involve a thorough investigation of smoking habits in men, as these habits could be related to an increased risk of excessive alcohol consumption. We present a comprehensive set of forecasts for the cross-correlation signal between 21 cm intensity mapping and galaxy redshift surveys. We focus on the data sets that will be provided by the SKAO for the 21 cm signal, DESI and Euclid for galaxy clustering. We build a likelihood which takes into account the effect of the beam for the radio observations, the Alcock–Paczynski effect, a simple parametrization of astrophysical nuisances, and fully exploit the tomographic power of such observations in the range z  = 0.7–1.8 at linear and mildly non-linear scales (k < 0.25 h  Mpc−1). The forecasted constraints, obtained with Monte Carlo Markov Chains techniques in a Bayesian framework, in terms of the six base parameters of the standard ΛCDM model, are promising. The predicted signal-to-noise ratio for the cross-correlation can reach ∼50 for z ∼ 1 and k ∼ 0.1 h  Mpc−1. When the cross-correlation signal is combined with current Cosmic Microwave Background (CMB) data from Planck, the error bar on |$\Omega _{\rm c}\, h^2$| and H 0 is reduced by factors 3 and 6, respectively, compared to CMB only data, due to the measurement of matter clustering provided by the two observables. The cross-correlation signal has a constraining power that is comparable to the autocorrelation one and combining all the clustering measurements a sub-per cent error bar of 0.33 per cent on H 0 can be achieved, which is about a factor 2 better than CMB only measurements. Finally, as a proof of concept, we test the full pipeline on the real data measured by the MeerKat collaboration (Cunnington et al. 2022) presenting some (weak) constraints on cosmological parameters. Alzheimer's disease (AD) is the most common neuronal disorder that leads to the development of dementia. Until nowadays, some therapies may alleviate the symptoms, but there is no pharmacological treatment. Microdosing lithium has been used to modify the pathological characteristics of the disease, with effects in both experimental and clinical conditions. The present work aimed to analyze the effects of this treatment on spatial memory, anxiety, and molecular mechanisms related to long-term memory formation during the aging process of a mouse model of accelerated aging (SAMP-8). Female SAMP-8 showed learning and memory impairments together with disruption of memory mechanisms, neuronal loss, and increased density of senile plaques compared to their natural control strain, the senescence-accelerated mouse resistant (SAMR-1). Chronic treatment with lithium promoted memory maintenance, reduction in anxiety, and maintenance of proteins related to memory formation and neuronal density. The density of senile plaques was also reduced. An increase in the density of gamma-aminobutyric acid A (GABAA) and α7 nicotinic cholinergic receptors was also observed and related to neuroprotection and anxiety reduction. In addition, this microdose of lithium inhibited the activation of glycogen synthase kinase-3beta (GSK-3β), the classical mechanism of lithium cell effects, which could contribute to the preservation of the memory mechanism and reduction in senile plaque formation. This work shows that lithium effects in neuroprotection along the aging process are not related to a unique cellular mechanism but produce multiple effects that slowly protect the brain along the aging process. The large majority of individuals who access substance use disorders (SUD) treatment do not receive evidence-based care. Little attention has been paid to the notion that the scale-up of evidence-based practices (EBPs) has been limited in large part due to a weakness in the "distribution system" for bringing new innovations to the attention of practitioners and into practice settings. This study explores the impact of the Training and Practice Implementation Institute (TPII; funded by the New York City Department of Health and Mental Hygiene), an intensive technical assistance initiative that offers external facilitation to outpatient SUD treatment providers via the incorporation of multiple evidence-based implementation strategies to enhance the practice of motivational interviewing (MI). Findings from this study show that staff completed a large majority (86%) of required training/technical assistance (TTA) activities across the 9-month implementation period, demonstrating a high level of engagement among staff and the feasibility of externally facilitated intensive TTA delivered to community-based organizations for the purpose of enhancing implementation of MI for SUDs. Results also show statistically significant improvements in the delivery of MI's technical components among staff, though did not reveal corresponding improvements in the delivery of MI's relational components. An understanding of the potential return on investment associated with externally facilitated TA, offers organizations an opportunity to inform the allocation of limited resources to areas where they will have the greatest impact and ultimately improve the quality and efficacy of services. Plastics are versatile materials that have the potential to propel humanity towards circularity and ultimate societal sustainability. However, the escalating concern surrounding plastic pollution has garnered significant attention, leading to widespread negative perceptions of these materials. Here, we question the role microbes may play in plastic pollution bioremediation by (i) defining polymer biodegradability (i.e., recalcitrant, hydrolysable and biodegradable polymers) and (ii) reviewing best practices for evaluating microbial biodegradation of plastics. We establish recommendations to facilitate the implementation of rigorous methodologies in future studies on plastic biodegradation, aiming to push this field towards the use of isotopic labelling to confirm plastic biodegradation and further determine the molecular mechanisms involved. Systemic lupus erythematosus (SLE) is a chronic autoimmune disease that is characterized by its large heterogeneity in terms of clinical presentation and severity. The pathophysiology of SLE involves an aberrant autoimmune response against various tissues, an excess of apoptotic bodies, and an overproduction of type-I interferon. The genetic contribution to the disease is supported by studies of monozygotic twins, familial clustering, and genome-wide association studies (GWAS) that have identified numerous risk loci. In the early 70s, complement deficiencies led to the description of familial forms of SLE caused by a single gene defect. High-throughput sequencing has recently identified an increasing number of monogenic defects associated with lupus, shaping the concept of monogenic lupus and enhancing our insights into immune tolerance mechanisms. Monogenic lupus (moSLE) should be suspected in patients with either early-onset lupus or syndromic lupus, in male, or in familial cases of lupus. This review discusses the genetic basis of monogenic SLE and proposes its classification based on disrupted pathways. These pathways include defects in the clearance of apoptotic cells or immune complexes, interferonopathies, JAK-STATopathies, TLRopathies, and T and B cell dysregulations. [ABSTRACT FROM AUTHOR]

Background: There are two major genetic types of Epstein-Barr Virus (EBV): type 1 (EBV-1) and type 2 (EBV-2). EBV functions by manipulating gene expression in host B cells, using virus-encoded gene regulatory proteins including Epstein-Barr Nuclear Antigen 2 (EBNA2). While type 1 EBNA2 is known to interact with human transcription factors (hTFs) such as RBPJ, EBF1, and SPI1 (PU.1), type 2 EBNA2 shares only ~ 50% amino acid identity with type 1 and thus may have distinct binding partners, human genome binding locations, and functions. Results: In this study, we examined genome-wide EBNA2 binding in EBV-1 and EBV-2 transformed human B cells to identify shared and unique EBNA2 interactions with the human genome, revealing thousands of type-specific EBNA2 ChIP-seq peaks. Computational predictions based on hTF motifs and subsequent ChIP-seq experiments revealed that both type 1 and 2 EBNA2 co-occupy the genome with SPI1 and AP-1 (BATF and JUNB) hTFs. However, type 1 EBNA2 showed preferential co-occupancy with EBF1, and type 2 EBNA2 preferred RBPJ. These differences in hTF co-occupancy revealed possible mechanisms underlying type-specific gene expression of known EBNA2 human target genes: MYC (shared), CXCR7 (type 1 specific), and CD21 (type 2 specific). Both type 1 and 2 EBNA2 binding events were enriched at systemic lupus erythematosus (SLE) and multiple sclerosis (MS) risk loci, while primary biliary cholangitis (PBC) risk loci were specifically enriched for type 2 peaks. Conclusions: This study reveals extensive type-specific EBNA2 interactions with the human genome, possible differences in EBNA2 interaction partners, and a possible new role for type 2 EBNA2 in autoimmune disorders. Objectives Agriculture entered the discussions about safety climates late, despite being one of the most hazardous industries. We recently developed a safety climate scale in Bordeaux vineyards, for which we provided good evidence of reliability and construct validity (Grimbuhler and Viel 2019). In this study, we aimed to establish the external validity of this safety climate scale with the help of an independent national sample of vineyard professionals. Population and methods We approached vineyard managers and operators during compulsory training and certification procedures for pesticide-related activities. Trainees giving informed consent for participation in the study were asked to complete a safety climate questionnaire at the start and end of a training session. In total, 406 vineyard managers or operators completed the questionnaire at the start of the study, 37 of whom declined to complete the questionnaire at the end of the training session, leaving 369 subjects available for pretraining/post-training comparisons. Statistical comparisons were based on t -tests and mixed models for repeated measures. Results A mean safety climate score of 82.91 (SD: 9.06) was obtained in the initial survey in the Bordeaux region, whereas the safety climate score was estimated at 83.78 (SD 10.39) in this nationwide survey (P = 0.23). A significant increase was observed after the training course, for the safety climate score (7.5%, P < 10–15) and for each of its 7 dimensions (P < 10–4 or less), in both univariate and multivariate analyses. However, the magnitude of these increases varied with dimension, ranging from 2.4% for rules and best practices to 15.5% for communication and feedback. Conclusions External validity was demonstrated by transferability and sensitivity to intervention. Flow cytometry is a relevant tool to meet the requirements of academic and industrial research projects aimed at estimating the features of a bacterial population (e.g., quantity, viability, activity). One of the remaining challenges is now the safe assessment of bacterial viability while minimizing the risks inherent to existing protocols. In our core facility at the Paris‐Saclay University, we have addressed this issue with two objectives: measuring bacterial viability in biological samples and preventing bacterial contamination and chemical exposure of the staff and cytometers used on the platform. Here, we report the development of a protocol achieving these two objectives, including a viability labeling step before bacteria fixation, which removes the risk of biological exposure, and the decrease of the use of reagents such as propidium iodide (PI), which are dangerous for health (CMR: carcinogenic, mutagenic, and reprotoxic). For this purpose, we looked for a non‐CMR viability dye that can irreversibly label dead bacteria before fixation procedures and maintain intense fluorescence after further staining. We decided to test on the bacteria, eFluor Fixable Viability dyes, which are usually used on eukaryotic cells. Since the bacteria had size and granularity characteristics very similar to those associated with flow cytometry background signals, a step of bacterial DNA labeling with SYTO or DRAQ5 was necessarily added to differentiate them from the background. Three marker combinations (viability‐DNA) were tested on LSR Fortessa and validated on pure bacterial populations (Gram+, Gram−) and polybacterial cultures. Any of the three methods can be used and adapted to the needs of each project and allow users to adapt the combination according to the configuration of their cytometer. Having been tested on six bacterial populations, validated on two cytometers, and repeated at least two times in each evaluated condition, we consider this method reliable in the context of these conditions. The reliability of the results obtained in flow cytometry was successfully validated by applying this protocol to confocal microscopy, permeabilization, and also to follow cultures over time. Background: Anthropometric measurements play a crucial role in medico-legal practices. Actually, several scanning technologies are employed in post-mortem investigations for forensic anthropological measurements. This study aims to evaluate the precision, inter-rater reliability, and accuracy of a handheld scanner in measuring various body parts. Methods: Three independent raters measured seven longitudinal distances using an iPad Pro equipped with a LiDAR sensor and specific software. These measurements were statistically compared to manual measurements conducted by an operator using a laser level and a meterstick (considered the gold standard). Results: The Friedman test revealed minimal intra-rater variability in digital measurements. Inter-rater variability analysis yielded an ICC = 1, signifying high agreement among the three independent raters. Additionally, the accuracy of digital measurements displayed errors below 1.5%. Conclusions: Preliminary findings demonstrate that the pairing of LiDAR technology with the Polycam app (ver. 3.2.11) and subsequent digital measurements with the MeshLab software (ver. 2022.02) exhibits high precision, inter-rater agreement, and accuracy. Handheld scanners show potential in forensic anthropology due to their simplicity, affordability, and portability. Background: Reports of dual carriers of pathogenic BRCA1 variants in trans are extremely rare, and so far, most individuals have been associated with a Fanconi Anemia-like phenotype. Methods: We identified two families with a BRCA1 in-frame exon 20 duplication (Ex20dup). In one male individual, the variant was in trans with the BRCA1 frameshift variant c.2475delC p.(Asp825Glufs*21). We performed splicing analysis and used a transcription activation domain (TAD) assay to assess the functional impact of Ex20dup. We collected pedigrees and mapped the breakpoints of the duplication by long- and short-read genome sequencing. In addition, we performed a mitomycin C (MMC) assay from the dual carrier using cultured lymphoblastoid cells. Results: Genome sequencing and RNA analysis revealed the BRCA1 exon 20 duplication to be in tandem. The duplication was expressed without skipping any one of the two exon 20 copies, resulting in a lack of wild-type transcripts from this allele. TAD assay indicated that the Ex20dup variant has a functional level similar to the well-known moderate penetrant pathogenic BRCA1 variant c.5096G > A p.(Arg1699Gln). MMC assay of the dual carrier indicated a slightly impaired chromosomal repair ability. Conclusions: This is the first reported case where two BRCA1 variants with demonstrated functional impact are identified in trans in a male patient with an apparently normal clinical phenotype and no BRCA1-associated cancer. The results pinpoint a minimum necessary BRCA1 protein activity to avoid a Fanconi Anemia-like phenotype in compound heterozygous status and yet still predispose carriers to hormone-related cancers. These findings urge caution when counseling families regarding potential Fanconi Anemia risk. Inflammation is a natural protective reaction of the body against endogenous and exogenous damage, such as tissue injuries, trauma, and infections. Thus, when the response is adequate, inflammation becomes a defense mechanism to repair damaged tissue, whereas when the response is inadequate and persistent, the increase in inflammatory cells, cytosines, and chymosins impair tissue regeneration and promote a response harmful to the organism. One example is chronic tissue inflammation, in which a simple lesion can progress to ulcers and even necrosis. In this situation, the anti-inflammatory medications available in therapy are not always effective. For this reason, the search for new treatments, developed from medicinal plants, has increased. In this direction, the plants Agave sisalana (sisal) and Punica granatum (pomegranate) are rich in saponins, which are secondary metabolites known for their therapeutic properties, including anti-inflammatory effects. Although Brazil is the world's leading sisal producer, approximately 95% of the leaves are discarded after fiber extraction. Similarly, pomegranate peel waste is abundant in Brazil. To address the need for safe and effective anti-inflammatory treatments, this study aimed to create a topical mucoadhesive gel containing a combination of sisal (RS) and pomegranate residue (PR) extracts. In vitro experiments examined isolated and combined extracts, as well as the resulting formulation, focusing on (1) a phytochemical analysis (total saponin content); (2) cytotoxicity (MTT assay); and (3) a pharmacological assessment of anti-inflammatory activity (phagocytosis, macrophage spreading, and membrane stability). The results revealed saponin concentrations in grams per 100 g of dry extract as follows: SR—29.91 ± 0.33, PR—15.83 ± 0.93, association (A)—22.99 ± 0.01, base gel (G1)—0.00 ± 0.00, and association gel (G2)—0.52 ± 0.05. In MTT tests for isolated extracts, cytotoxicity values (µg/mL) were 3757.00 for SR and 2064.91 for PR. Conversely, A and G2 exhibited no cytotoxicity, with increased cell viability over time. All three anti-inflammatory tests confirmed the presence of this activity in SR, PR, and A. Notably, G2 demonstrated an anti-inflammatory effect comparable to dexamethasone. In conclusion, the gel containing SR and PR (i.e., A) holds promise as a novel herbal anti-inflammatory treatment. Objective: While the incidence and type of blood malignancies are well documented amid primary Sjögren's syndrome patients (pSS), data focusing on solid neoplasms are more conflicting. We aimed to describe clinical, pathological, and immunological characteristics of pSS patients with cancers, along with the chronological interplay between the two conditions. Methods: Outcomes concerning both pSS and cancer were retrospectively collected from Montpellier University Hospital (tertiary center) between 2019 and 2020. pSS characteristics were compared to a control group of pSS patients without cancer. Results: A total of 165 patients with pSS were included: 55 patients with cancer (52 female, mean age 58.4 ± 10.4 years at pSS diagnosis; mean follow-up 10.5 ± 10.1 years, 12 patients had multiple cancers) and 110 controls without cancer. Characteristics of pSS patients with cancers were different from controls mostly for lymphoma prognosis factors. Among the 70 cancers, we recorded 55 solid neoplasms (whom 27 breast cancers and 8 lung cancers, and 82% of adenocarcinomas), with no evidence of disease at the end of follow-up in 85% of them. Among the 15 recorded blood malignancies, ten were lymphomas with an excellent prognosis. Regarding chronological interplay between cancer and pSS, most cancers (43%) were diagnosed close (± 5 years) to pSS diagnosis. Breast cancers were diagnosed before or close to pSS diagnosis (mean delay − 1.8 ± 13.0 years), at an early stage, with only two relapses (no cancer-related death), while lung cancers were diagnosed late after. Conclusions: The tight chronological interplay between breast cancer and pSS and the intriguing pathological and immunological pattern of pSS in these patients suggest a hypothesis of immune control of cancer. (1) Background: MLH1 hypermethylation is an epigenetic alteration in the tumorigenesis of colorectal cancer (CRC) and endometrial cancer (EC), causing gene silencing, and, as a consequence, microsatellite instability. Commonly, MLH1 hypermethylation is considered a somatic and sporadic event in cancer, and its detection is recognized as a useful tool to distinguish sporadic from inherited conditions (such as, Lynch syndrome (LS)). However, MLH1 hypermethylation has been described in rare cases of CRC and EC in LS patients. (2) Methods: A total of 61 cancers (31 CRCs, 27 ECs, 2 ovarian cancers, and 1 stomach cancer) from 56 patients referred to cancer genetic counselling were selected for loss of MLH1 protein expression and microsatellite instability. All cases were investigated for MLH1 promoter methylation and MLH1/PMS2 germline variants. (3) Results: Somatic MLH1 promoter hypermethylation was identified in 16.7% of CRC and in 40% of EC carriers of MLH1 germline pathogenic variants. In two families, primary and secondary MLH1 epimutations were demonstrated. (4) Conclusions: MLH1 hypermethylation should not be exclusively considered as a sporadic cancer mechanism, as a non-negligible number of LS-related cancers are MLH1 hypermethylated. Summary: Background and Objectives: The histological PRO score (I–III) helps to assess the malignant potential of actinic keratoses (AK) by grading the dermal‐epidermal junction (DEJ) undulation. Line‐field confocal optical coherence tomography (LC‐OCT) provides non‐invasive real‐time PRO score quantification. From LC‐OCT imaging data, training of an artificial intelligence (AI), using Convolutional Neural Networks (CNNs) for automated PRO score quantification of AK in vivo may be achieved. Patients and Methods: CNNs were trained to segment LC‐OCT images of healthy skin and AK. PRO score models were developed in accordance with the histopathological gold standard and trained on a subset of 237 LC‐OCT AK images and tested on 76 images, comparing AI‐computed PRO score to the imaging experts' visual consensus. Results: Significant agreement was found in 57/76 (75%) cases. AI‐automated grading correlated best with the visual score for PRO II (84.8%) vs. PRO III (69.2%) vs. PRO I (66.6%). Misinterpretation occurred in 25% of the cases mostly due to shadowing of the DEJ and disruptive features such as hair follicles. Conclusions: The findings suggest that CNNs are helpful for automated PRO score quantification in LC‐OCT images. On 5–6 July 2017, an unstable atmospheric condition caused an unusual concentration of rainfall above the Northern part of Kyushu Island, triggering a set of hydro-meteorological hazards. Within the affected area, the mountainous subwatershed of the Akatani River was significantly impacted by numerous landslides combined with debris flow and floods. National and local agencies deployed a plan of reconstruction to restore the floodplain and protect inhabitants. Regarding the hydrosystem in the Akatani watershed, this reconstruction project mainly focuses on the restoration of damaged protection systems and the construction of new infrastructures. Thus, this paper aims to explain the restoration plan of the Akatani River in terms of the strategic Japanese River System Sabo and then as a model of a national-scale spatial plan. It draws on (i) a literature review based on the historical evolution of Japanese protection systems and the River Sabo System; (ii) field surveys in 2019, 2022 and 2023, in conjunction with (iii) interviews with local, regional, and national officials; and (iv) a Geographical Information System analysis of previously and newly built protection systems through aerial photograph interpretation and geospatial data. Sabo works implemented in the Akatani watershed illustrate the engineering vision of Japanese river management. They also constitute a comprehensive system and include a downstream–upstream logic which echoes that of the River System Sabo. In addition, the disaster of July 2017 and the government's response emphasize the continuous adaptation and improvement of the Japanese disaster management system, which mitigates severe disasters. Cataract, the opacification of the lens, is the leading cause of blindness worldwide. Although effective, cataract surgery is costly and can lead to complications. Toward identifying alternate treatments, it is imperative to develop organoid models relevant for lens studies and drug screening. Here, we demonstrate that by culturing mouse lens epithelial cells under defined three-dimensional (3D) culture conditions, it is possible to generate organoids that display optical properties and recapitulate many aspects of lens organization and biology. These organoids can be rapidly produced in large amounts. High-throughput RNA sequencing (RNA-seq) on specific organoid regions isolated via laser capture microdissection (LCM) and immunofluorescence assays demonstrate that these lens organoids display a spatiotemporal expression of key lens genes, e.g., Jag1, Pax6, Prox1, Hsf4 and Cryab. Further, these lens organoids are amenable to the induction of opacities. Finally, the knockdown of a cataract-linked RNA-binding protein encoding gene, Celf1, induces opacities in these organoids, indicating their use in rapidly screening for genes that are functionally relevant to lens biology and cataract. In sum, this lens organoid model represents a compelling new tool to advance the understanding of lens biology and pathology and can find future use in the rapid screening of compounds aimed at preventing and/or treating cataracts. Simple Summary: Urolithiasis is the formation of salt crystals in the kidney and can be caused by several factors, one of which is the use of unbalanced diets for feeding animals. This study aims to evaluate the effectiveness of different additives in preventing urolithiasis in lambs fed a high-concentrate diet. Thirty-two noncastrated male lambs were divided into four treatments, three including additives in the diet and one group without additives (CON), and fed for 56 days in a feedlot. The additives used were ammonium chloride (ACL), calcium chloride (CCL), and benzoic acid (BZA), and each was added to a diet composed of 6% Cynodon ssp. hay and 94% concentrate. The use of additives in the diet did not affect the performance of the lambs. Regarding urine pH, inclusion of ACL and CCL in the diet acidified the urine, but none of the additives prevented urolithiasis. However, the formation of these crystals did not show obstruction of the urinary tract or affect the lambs' health. This study aimed to evaluate the effectiveness of different additives in preventing urolithiasis in lambs fed a diet rich in concentrate and their impact on performance and blood and urinary parameters. Thirty-two noncastrated male lambs, crossbred Dorper × Santa Inês, with initial body weights (BWs) of 23 ± 0.1 kg and ages of 50 ± 5 days, were kept in individual pens and fed a diet composed of 6% Cynodon ssp. hay and 94% concentrate and subjected to four treatments: CON without inclusion of additives, addition of ACL 5 g/kg of dry matter (DM), addition of CCL 6.3 g/kg of DM, and addition of BZA 5 g/kg of DM. There was no effect of treatment or interaction with time on blood parameters (p > 0.050), and performance characteristics, morphometry of ruminal papillae, and scores of cecum and rumen lesions were not affected by the addition of additives to the diet (p > 0.050). Greater urinary acidification was observed in animals from CCL and ACL treatments (p = 0.033). Calcium chloride acidified the urinary pH and can be used instead of ammonium chloride at a concentration of 0.63% based on DM, when this is the objective. [ABSTRACT FROM AUTHOR]

Antioxidants have been proposed as a treatment for diseases of the central nervous system. However, few studies actually studied their effects in the brain. To test central actions of antioxidants, we used the lithium-pilocarpine (Li-Pilo) model of status epilepticus (SE) in the rat in which seizures are accompanied by significant oxidative stress. We used in vivo microdialysis to determine isoprostane levels during SE in real time and brain homogenates for other measures of oxidative stress. Six different antioxidants were tested in acute and preventive experiments (vitamin C, vitamin E, ebselen, resveratrol, n-tert-butyl-α-phenylnitrone and coenzyme Q10). None of the antioxidants had an effect when given acutely during SE. In contrast, when antioxidants were given for 3 days prior to seizure induction, vitamins C and E reduced isoprostane formation by 58% and 65%, respectively. Pretreatment with the other antioxidants was ineffective. In brain homogenates prepared after 90 min of seizures, SE decreased the ratio of reduced vs. oxidized glutathione (GSH/ GSSG ratio) from 60.8 to 7.50 and caused a twofold increase of 8-hydroxy-2'-deoxyguanosine (8-OHdG) levels and protein carbonyls. Pretreatment with vitamin C or vitamin E mitigated these effects and increased the GSH/GSSG ratio to 23.9 and 28.3, respectively. Again, the other antioxidants were not effective. We conclude that preventive treatment with vitamin C or vitamin E ameliorates seizure-induced oxidative damage in the brain. Several well-studied antioxidants were inactive, possibly due to limited brain permeability or a lack of chain-breaking antioxidant activity in hydrophilic compounds. [ABSTRACT FROM AUTHOR]

Two binuclear heteroleptic CuI complexes, namely Cu−NIR1 and Cu−NIR2, bearing rigid chelating diphosphines and π‐conjugated 2,5‐di(pyridin‐2‐yl)thiazolo[5,4‐d]thiazole as the bis‐bidentate ligand are presented. The proposed dinuclearization strategy yields a large bathochromic shift of the emission when compared to the mononuclear counterparts (M1–M2) and enables shifting luminescence into the near‐infrared (NIR) region in both solution and solid state, showing emission maximum at ca. 750 and 712 nm, respectively. The radiative process is assigned to an excited state with triplet metal‐to‐ligand charge transfer (3MLCT) character as demonstrated by in‐depth photophysical and computational investigation. Noteworthy, X‐ray analysis of the binuclear complexes unravels two interligand π–π‐stacking interactions yielding a doubly locked structure that disfavours flattening of the tetrahedral coordination around the CuI centre in the excited state and maintain enhanced NIR luminescence. No such interaction is present in M1–M2. These findings prompt the successful use of Cu−NIR1 and Cu−NIR2 in NIR light‐emitting electrochemical cells (LECs), which display electroluminescence maximum up to 756 nm and peak external quantum efficiency (EQE) of 0.43 %. Their suitability for the fabrication of white‐emitting LECs is also demonstrated. Simple Summary: Excessive intake of certain minerals can modify the requirement of others. For example, Mo and S are the main antagonists of Cu, in which high dietary levels of these minerals increase Cu requirements in ruminants. Thus, if Cu levels in the body are low, higher amounts of Mo may become a toxic element for animals. In this paper, we investigated the effects of organic and inorganic sources of Cu and S and their interaction with Mo on the performance, carcass traits, and blood concentration of these minerals, and we also investigated the serum ceruloplasmin activity in lambs. We found that molybdenum and inorganic and organic sources of Cu and S did not improve performance, nor did they have an impact on carcass traits. However, a clear negative effect of Mo and S was detected on Cu bioavailability and metabolism, either due to serum Cu concentrations or through ceruloplasmin activity. However, it was not possible to identify a pattern in the variables studied. This study aimed to evaluate the effects of molybdenum (Mo) and organic and inorganic sources of copper (Cu) and sulfur (S) on the performance, carcass traits, and blood concentration of these minerals in lambs. Forty male non-castrated crossbred Dorper x Santa Inês lambs (20 ± 1.2 kg of body weight and 90 ± 2 d of age) were randomly allocated into one of the ten following treatments: (T0) control, basal diet; (T1) Mo; (T2) inorganic Cu and inorganic S; (T3) inorganic Cu and organic S; (T4) organic Cu and inorganic S; (T5) organic Cu and organic S; (T6) Mo plus inorganic Cu and inorganic S; (T7) Mo plus inorganic Cu and organic S; (T8) Mo plus organic Cu and inorganic S; and (T9) Mo plus organic Cu and organic S. Regardless of the source, Mo, Cu, and S were added at levels of 10 mg, 10 mg, and 2000mg/kg DM, respectively. The mineral supplements (Mo, Cu, and S) were added into the total mixed ration (TMR) by mixing them apart with the mineral and vitamin premix and then put into the TMR. The animals were kept in individual pens and received a total mixed ration for 84 days. Body weight and blood sampling was performed every 28 days. All animals were slaughtered after 84 days, and carcass traits were evaluated. Although organic sources of Cu and S added to Mo supplementation had increased the ADG throughout the study, this effect did not reflect in the heavier final BW outcomes for this treatment. In addition, no effect of these treatments was observed on the carcass traits. The serum Cu concentration was higher for the T0 group compared to the other groups; otherwise, Mo reduced the serum Cu concentration compared to the other groups. Considering the interaction among the minerals and their sources at 84 d of study, organic sources of Cu and S treatment and Mo associated with inorganic sources of Cu plus organic S treatment had an increased serum Cu concentration compared to other groups. Regardless of time, organic sources of Cu and S increased serum S concentration. At 84 days after enrollment, serum Mo concentration was lower for the control group compared to the other groups. Further, Mo supplementation increased its blood concentration compared to the control group throughout the study. The control group had the highest ceruloplasmin activity compared to the other groups; otherwise, at 84 d of the study, either Mo or inorganic S supplementation reduced ceruloplasmin activity. Serum ceruloplasmin activity was higher when Cu supplementation, regardless of source, was associated with organic S. However, at d 84 of the study, inorganic Cu associated to organic S supplements increased serum ceruloplasmin activity. In this current study, it was not possible to identify a pattern in the variables studied, however, further studies are needed to confirm that organic sources of Cu and S interacted alone without a defined pattern. The present study investigates the langauge and text of the Fiore, a work transmitted by a manuscript conserved in Montpellier, at the Bibliothèque interuniversitaire, Section Médecine, H 438. From a linguistic point of view, the analysis will focus on non-Florentine language traces, as indicated by Castellani, revealing a more recent layer of Eastern and Western Tuscan, as well as some 14th-century Florentine features. As regards the establishment of the text, the study will give a classification of the scribal errors identified in the manuscript, with special attention to the ones, which can be considered as evidence of complex and multilayered scribal activity. The novel amphiphilic polyacrylate grafted with cholesterol moieties, PAAbCH, previously synthesized, was deeply characterized and investigated in the lab and on a pre-industrial scale. Solid-state NMR analysis confirmed the polymer structure, and several water-based pharmaceutical and cosmetic products were developed. In particular, stable oil/water emulsions with vegetable oils, squalene, and ceramides were prepared, as well as hydrophilic medicated films loaded with diclofenac, providing a prolonged drug release. PAAbCH also formed polyelectrolyte hydrogel complexes with chitosan, both at the macro- and nano-scale. The results demonstrate that this polymer has promising potential as an innovative excipient, acting as a solubility enhancer, viscosity enhancer, and emulsifying agent with an easy scale-up transfer process. Quantitative biomarkers of facial skin ageing were studied from one hundred healthy Caucasian female volunteers, aged 20–70 years, using in vivo 3D Line-field Confocal Optical Coherence Tomography (LC-OCT) imaging coupled with Artificial Intelligence (AI)-based quantification algorithms. Layer metrics, i.e. stratum corneum thickness (SC), viable epidermal thickness and Dermal–Epidermal Junction (DEJ) undulation, as well as cellular metrics were measured for the temple, cheekbone and mandible. For all three investigated facial areas, minimal age-related variations were observed in the thickness of the SC and viable epidermis layers. A flatter and more homogeneous epidermis (decrease in the standard deviation of the number of layers means), a less dense cellular network with fewer cells per layer (decrease in cell surface density), and larger and more heterogeneous nuclei within each layer (increase in nuclei volume and their standard deviation) were found with significant variations with age. The higher atypia scores further reflected the heterogeneity of nuclei throughout the viable epidermis. The 3D visualisation of fine structures in the skin at the micrometric resolution and the 1200 µm × 500 µm field of view achieved with LC-OCT imaging enabled to compute relevant quantitative biomarkers for a better understanding of skin biology and the ageing process in vivo. The warm-hot intergalactic medium (WHIM) contains a significant portion of the 'missing baryons'. Its detection in emission remains a challenge. Integral field spectrometers like X-IFU on board of the Athena satellite will secure WHIM detection in absorption and emission and, for the first time, allow us to investigate its physical properties. In our research, we use the CAMELS simulations to model the surface brightness maps of the OVII and OVIII ion lines and compute summary statistics like photon counts and 2-point correlation functions to infer the properties of the WHIM. Our findings confirm that detectable WHIM emission is primarily associated with galaxy haloes, and the properties of the WHIM show minimal evolution from z ∼ 0.5 to the present time. By exploring a wide range of parameters within the CAMELS suite, we investigate the sensitivity of WHIM properties to cosmology and energy feedback mechanisms influenced by active galactic nuclei and stellar activity. This approach allows us to separate the cosmological aspects from the baryonic processes and place constraints on the latter. Additionally, we provide forecasts for WHIM observations using a spectrometer similar to X-IFU. We anticipate detecting 1–3 WHIM emission lines per pixel and mapping the WHIM emission profile around haloes up to a few tens of arcminutes, surpassing the typical size of a WHIM emitter. Overall, our work demonstrates the potential of emission studies to probe the densest phase of the WHIM, shedding light on its physical properties and offering insights into the cosmological and baryonic processes at play. The current study evaluated the success of implementing a blended + flipped structure in a Calculus for Life and Management Sciences course. By reimagining two of the three weekly instructional hours as online asynchronous lessons, we redesigned a 200-person section into 30-person sections, with minimal additional instructor resources. Findings indicated that students viewed the course positively, with perceptions positively correlating with course outcomes. Further, students reported more positive beliefs relating to their ability to accomplish course learning outcomes, comfort with math, and perceptions of in-class readiness compared to those in the traditional course format. The Alcohol Use Disorders Identification Test (AUDIT) and its short form, the AUDIT-C, the main clinical instruments used to identify unhealthy drinking behaviors, are influenced by memory bias and under-reporting. In recent years, phosphatidylethanol (PEth) in blood has emerged as a marker of unhealthy alcohol use. This systematic review aims to investigate the molecular characteristics of PEth and summarize the last ten years of published literature and its use compared to structured questionnaires. A systematic search was performed, adhering to PRISMA guidelines, through "MeSH" and "free-text" protocols in the databases PubMed, SCOPUS, and Web of Science. The inclusion criteria were as follows: PEth was used for detecting unhealthy alcohol consumption in the general population and quantified in blood through liquid chromatography coupled to mass spectrometry, with full texts in the English language. Quality assessment was performed using the JBI critical appraisal checklist. Twelve papers were included (0.79% of total retrieved records), comprising nine cross-sectional studies and three cohort studies. All studies stratified alcohol exposure and quantified PEth 16:0/18:1 through liquid chromatography coupled to mass spectrometry (LC-MS) in liquid blood or dried blood spots (DBS) with lower limits of quantitation (LLOQ) ranging from 1.7 ng/mL to 20 ng/mL. A correlation between blood PEth level and the amount of alcohol ingested in the previous two weeks was generally observed. PEth interpretative cut-offs varied greatly among the included records, ranging from 4.2 ng/mL to 250 ng/mL, with sensitivity and specificity in the ranges of 58–100% and 64–100%, respectively. Although the biomarker seems promising, further research elucidating the variability in PEth formation and degradation, as well as the molecular mechanisms behind that variability, are necessary.
