Introduction Lung cancer in never-smoker (LCINS) patients accounts for 20% of lung cancer cases, and its biology remains poorly understood, particularly in genetically admixed populations. We elucidated the molecular profile of driver genes in Brazilian LCINS. Methods The mutational and gene fusion status of 119 lung adenocarcinomas from self-reported never-smoker patients, was assessed using targeted sequencing (NGS), nCounter, and immunohistochemistry. A panel of 46 ancestry-informative markers determined patients' genetic ancestry. Results The most frequently mutated gene was EGFR (49.6%), followed by TP53 (39.5%), ALK (12.6%), ERBB2 (7.6%), KRAS (5.9%), PIK3CA (1.7%), and less than 1% alterations in RET , NTRK1 , MET ∆ex14, PDGFRA , and BRAF. Except for TP53 and PIK3CA , all other alterations were mutually exclusive. Genetic ancestry analysis revealed a predominance of European (71.1%), and a higher African ancestry was associated with TP53 mutations. Conclusion Brazilian LCINS exhibited a similar molecular profile to other populations, except the increased ALK and TP53 alterations. Soybean is the main oilseed cultivated worldwide. Even though Brazil is the world's largest producer and exporter of soybean, its production is severely limited by biotic factors. Soil borne diseases are the most damaging biotic stressors since they significantly reduce yield and are challenging to manage. In this context, the present study aimed to evaluate the potential of a bacterial strain (Ag109) as a biocontrol agent for different soil pathogens (nematodes and fungi) of soybean. In addition, the genome of Ag109 was wholly sequenced and genes related to secondary metabolite production and plant growth promotion were mined. Ag109 showed nematode control in soybean and controlled 69 and 45% of the populations of Meloidogyne javanica and Pratylenchus brachyurus, respectively. Regarding antifungal activity, these strains showed activity against Macrophomia phaseolina, Rhizoctonia solani, and Sclerotinia sclerotiorum. For S. sclerotiorum, this strain increased the number of healthy plants and root dry mass compared to the control (with inoculation). Based on the average nucleotide identity and digital DNA-DNA hybridization, this strain was identified as Bacillus velezensis. Diverse clusters of specific genes related to secondary metabolite biosynthesis and root growth promotion were identified, highlighting the potential of this strain to be used as a multifunctional microbial inoculant that acts as a biological control agent while promoting plant growth in soybean. Our previous studies have shown that 'Palmer' mangoes immersed in solutions containing 2.5% sorbitol and stored under a controlled atmosphere (CA) at 8 °C for 30 days had fewer symptoms of a chilling injury. However, there is no information regarding the effectiveness of sorbitol treatment in other atmospheres and/or in combination with lower temperatures. Thus, the objective of this study was to assess the impact of dipping 'Palmer' mangoes in 0.1% and 2.5% (w/v) sorbitol solutions and storing the fruit under a CA without atmosphere modification (21 kPa O2 + 0.03 kPa CO2) at 8 °C/95% relative humidity (RH) or with 5 kPa O2 + 5 kPa CO2 at 4 °C/95% RH for 28 days. The fruits were evaluated periodically for chilling injuries, quality, and oxidative metabolism. A chilling injury (CI) was correlated with increased fresh weight loss (FWL) and changes in the color of the epicarp (Lpeel, h°peel, and Cpeel) and mesocarp (L*pulp). Lipid peroxidation (LPpulp and LPpeel) and the hydrogen peroxide content (H2O2peel and H2O2pulp) were associated with the development of a CI, particularly after being transferred to ambient. The treatment with 2.5% sorbitol was more effective in minimizing the chilling injury symptoms and did not compromise the fruit quality, especially when it was stored at 4 °C in association with a CA containing 5 kPa O2 + 5 kPa CO2. This treatment reduced lipid peroxidation and increased the activities of the superoxide dismutase (SOD) and ascorbate peroxidase (APX) enzymes in the epicarp and mesocarp, providing greater cold tolerance. The use of 2.5% sorbitol has been identified as the most efficacious approach for mitigating the adverse impacts of chilling injuries, preserving the fruit quality, and enhancing oxidative metabolism, even at lower temperatures. Aegla is a genus of freshwater anomuran Crustacea known for having epimorphic postembryonic development. The newly-hatched juveniles' morphology have been described for only 11 of the 93 known species. These descriptions are important tools for identifying juveniles and useful characteristics for phylogenetic analyses. This study aims to describe and illustrate the external morphology of the first juvenile stage of Aegla lata. Ovigerous females were collected, fed, and monitored periodically in the laboratory until the eggs hatched. The newly-hatched juveniles were fixed in an alcohol series and preserved in a 70% alcohol and glycerine solution (2:1 ratio) for subsequent dissection and analysis under light microscopy. The appendages were illustrated using a camera lucida and then described. Scanning electron microscopy was used to analyse some specimens, allowing the detailed observation of the setal morphology, some cephalothoracic appendages, and lineae aeglicae. Pappose setae were also observed in the protopod and dactylus of cheliped. The newly-hatched juvenile of A. lata has a set of the following characteristics: basal segment of antennule with serrate setae; distal segment of antennule with pappose setae; merus of second maxilliped with simple setae; and merus of third maxilliped without simple setae. Simple Summary: Neoadjuvant therapy is a crucial part of breast cancer treatment, but identifying the patients who will benefit the most from it remains challenging. This summary discusses a study of breast cancer treatment, specifically focusing on neoadjuvant endocrine therapy (NET) for postmenopausal, hormone-receptor positive, HER2-negative breast cancer patients. The study aimed to determine whether Magee equations (MEs) could predict how well patients respond to NET. Among the 75 female participants, ME3 emerged as a promising predictor with an AUC of 0.734. This means that ME3 could effectively identify patients likely to respond well to NET. In the analysis, factors such as clinical staging and molecular subtype also showed significant associations with treatment response. Specifically, patients with ME3 values less than 20 were more likely to have a positive response to treatment. The study suggests that using ME3 as a predictive tool could help identify breast cancer patients who are likely to respond positively to NET. This approach offers a cost-effective alternative to the use of Oncotype DX, a commonly used but expensive tool for treatment decision-making. However, larger prospective studies are needed to further validate these findings, which could ultimately improve the selection of the most suitable candidates for NET. Background: Breast cancer (BC) remains a significant health care challenge, and treatment approaches continue to evolve. Among these, neoadjuvant endocrine therapy (NET) has gained prominence, particularly for postmenopausal, hormone-receptor positive, HER2-negative (HR+/HER2−) BC patients. Despite this, a significant gap exists in identifying patients who stand to benefit from NET. The objective of this study was to assess whether Magee equations (MEs) could serve as predictors of response to NET. Methods: This retrospective study included adult patients with invasive BC who underwent NET followed by curative surgery. Assessment of sociodemographic, clinical, and tumor-related variables was conducted. The ME1, ME2, ME3, and ME mean were analyzed to explore their predictive role for NET response. Receiver operating characteristic (ROC) curves were employed, along with the determination of optimal cutoff points. Logistic regression models were utilized to identify the most significant predictors of pathological response. Results: Among the 75 female participants, the mean age was 69.4 years, with the majority being postmenopausal (n = 72, 96%) and having an ECOG-PS of 0/1 (n = 63, 84%). Most patients were classified as luminal A (n = 41, 54.7%). ME3 emerged as a promising predictor, boasting an AUC of 0.734, with sensitivity of 90.62% and specificity of 57.50% when the threshold was ≤ 19.97. In univariate analysis, clinical staging (p = 0.002), molecular subtype (p = 0.001), and ME3 (continuous = 0.001, original 3-tier: p = 0.013, new 2-tier: <0.001) categories exhibited significant associations with pathological response. In the multivariate model, clinical staging and new 2-tier ME3 (<20 vs. ≥20) were included as significant variables. Conclusions: Patients with ME3 < 20 have a higher likelihood of presenting a pathological response, offering a cost-effective alternative tool to Oncotype DX. The morphologies of only 10 of the 93 described species of Aegla Leach, 1820 are known. We describe and illustrate the external morphology of newly-hatched, first-stage juveniles of Aegla jacutinga Marçal & Teixeira in Marçal, Páez, Souza-Shibatta, Sofia & Teixeira, 2020 using light microscopy and scanning electron microscopy (SEM) comparing them with the same stage in other congeners and providing diagnostic characters. Aegla is morphologically a conservative genus, which creates many problems in the identification of its many cryptic species. It thus becomes necessary to refine the taxonomic characters used in the taxonomy of the group by providing a new sets of morphological characters to be analyzed. [ABSTRACT FROM AUTHOR]

NTRK1, 2, and 3 fusions are important therapeutic targets for NSCLC patients, but their prevalence in South American admixed populations needs to be better explored. NTRK fusion detection in small biopsies is a challenge, and distinct methodologies are used, such as RNA-based next-generation sequencing (NGS), immunohistochemistry, and RNA-based nCounter. This study aimed to evaluate the frequency and concordance of positive samples for NTRK fusions using a custom nCounter assay in a real-world scenario of a single institution in Brazil. Out of 147 NSCLC patients, 12 (8.2%) cases depicted pan-NTRK positivity by IHC. Due to the absence of biological material, RNA-based NGS and/or nCounter could be performed in six of the 12 IHC-positive cases (50%). We found one case exhibiting an NTRK1 fusion and another an NTRK3 gene fusion by both RNA-based NGS and nCounter techniques. Both NTRK fusions were detected in patients diagnosed with lung adenocarcinoma, with no history of tobacco consumption. Moreover, no concomitant EGFR, KRAS, and ALK gene alterations were detected in NTRK-positive patients. The concordance rate between IHC and RNA-based NGS was 33.4%, and between immunohistochemistry and nCounter was 40%. Our findings indicate that NTRK fusions in Brazilian NSCLC patients are relatively rare (1.3%), and RNA-based nCounter methodology is a suitable approach for NRTK fusion identification in small biopsies. Purpose: Medulloblastomas are the most common primary malignant brain tumors in children. They are divided into molecular subgroups: WNT-activated, SHH-Activated, TP53 mutant or wild type, and non-WNT/non-SHH (Groups 3 and 4). WNT-activated medulloblastomas are usually caused by mutations in the CTNNB1 gene (85%-90%), and most remaining cases of CTNNB1 wild type are thought to be caused by germline mutations in APC. So far, the frequencies of CTNNB1 have been reported mainly in North American and European populations. The aim of this study was to report the frequency of CTNNB1 mutations in WNT-activated medulloblastomas in a Latin-Iberian population and correlate with their clinicopathological characteristics. Methods: A total of 266 medulloblastomas from seven different institutions from Brazil (n=211), Portugal (n=38), and Argentina (n=17) were evaluated. Following RNA and DNA isolation from formalin-fixed, paraffin-embedded (FFPE) tumor tissues, the molecular classification and CTNNB1 mutation analysis were performed by nCounter and Sanger sequencing, respectively. Results: WNT-activated medulloblastomas accounted for 15% (40/266) of the series. We observed that 73% of WNT-activated medulloblastomas harbored CTNNB1 mutations. CTNNB1 wild-type cases (27%) were more prevalent in female individuals and suggested to be associated with a worse outcome. Among the CTNNB1 wild-type cases, the available analysis of family history revealed two cases with familiar adenomatous polyposis, harboring APC germline variants. Conclusion: We observed a lower incidence of CTNNB1 mutations in WNT-activated medulloblastomas in our Latin-Iberian cohort compared to frequencies previously described in other populations. Considering that CTNNB1 wild-type cases may exhibit APC germline mutations, our study suggests a higher incidence (~30%) of hereditary WNT-activated medulloblastomas in the Latin-Iberian population. In this work a dilute‐and‐shoot procedure using digital image colorimetry (DIC) was evaluated for the determination of aluminum in antiperspirants. Derivatization was performed by means of the reaction between Al(III) and Alizarin Red S (ARS) reagent. Digital images were acquired using a table scanner and the average values for red, green and blue channels (RGB) were obtained. Optimal conditions were set at pH=4.5, ARS concentrations 10‐fold higher than Al(III) and using the G channel. The method presented linearity in the 2.0–10.0 mg L−1 range, the limit of detection (LOD) was 1.1 mg L−1 and precision was better than 2.55 %. Accuracy was checked by the analysis of spiked samples at 2.0 and 4.0 mg L−1 Al(III), and good recoveries (82.6–117.5 %) were obtained. Eight samples of roll‐on antiperspirants were analyzed by the proposed colorimetric method and the well‐stablished flame atomic absorption spectrometry (FAAS) comparative method. The obtained results were not statistically different at 95 % confidence level. An analytical greenness assessment was performed for both methods and the procedure based on digital image colorimetry was proven to be a greener approach due to miniaturization, low sample consumption, least amount of waste and high sample throughput. Foodborne diseases are a major challenge in the global food industry, especially those caused by multidrug-resistant (MDR) bacteria. Bacteria capable of biofilm formation, in addition to MDR strains, reduce the treatment efficacy, posing a significant threat to bacterial control. Bacteriophages, which are viruses that infect and kill bacteria, are considered a promising alternative in combating MDR bacteria, both in human medicine and animal production. Phage cocktails, comprising multiple phages, are commonly employed to broaden the host range and prevent or delay the development of phage resistance. There are numerous techniques and protocols available to evaluate the lytic activity of bacteriophages, with the most commonly used methods being Spot Test Assays, Efficiency of Plating (EOP), and infection assays in liquid culture. However, there is currently no standardization for which analyses should be employed and the possible differences among them in order to precisely determine the host range of phages and the composition of a cocktail. A preliminary selection using the Spot Test Assay resulted in four phages for subsequent evaluation against a panel of 36 Salmonella isolates of numerous serovars. Comparing EOP and infection assays in liquid culture revealed that EOP could underestimate the lytic activity of phages, directly influencing phage cocktail development. Moreover, the phage cocktail containing the four selected phages was able to control or remove biofilms formed by 66% (23/35) of the isolates, including those exhibiting low susceptibility to phages, according to EOP. Phages were characterized genomically, revealing the absence of genes associated with antibiotic resistance, virulence factors, or integrases. According to confocal laser scanning microscopy analysis, the biofilm maturation of one Salmonella isolate, which exhibited high susceptibility to phages in liquid culture and 96-well plates biofilm viability assays but had low values for EOP, was found to be inhibited and controlled by the phage cocktail. These observations indicate that phages could control and remove Salmonella biofilms throughout their growth and maturation process, despite their low EOP values. Moreover, using infection assays in liquid culture enables a more precise study of phage interactions for cocktail design timelessly and effortlessly. Hence, integrating strategies and techniques to comprehensively assess the host range and lytic activity of bacteriophages under different conditions can demonstrate more accurately the antibacterial potential of phage cocktails. The aim of this study was to determine the activity of Brevibacillus brevis strain LABIM17 against Xanthomonas euvesicatoria pv. euvesicatoria and the control of bacterial spot on tomato. The antimicrobial activity of the strain LABIM17 against X. euvesicatoria pv. euvesicatoria strain 75-3 was evaluated by the agar diffusion method, the pour plate. The minimum inhibitory concentration (MIC) for the growth of the strain 75-3 was 500 µg mL−1 of the LABIM17 extract, the lyophilized cells free supernatant (CFS). Tomato plants sprayed with LABIM17 suspension at the concentrations of 3.4 × 107 and 1.7 × 107 CFU mL−1 and inoculated three days later with strain 75-3 showed reduction in the severity of the disease. Further, the plants that received these applications of strain LABIM17 presented reduction of 80 and 86%, respectively, in bacterial spot severity, the same level of disease reduction provided by the commercial chemical inducer acibenzolar-S-methyl. To our knowledge, this is the first report of the antimicrobial activity of B. brevis strain LABIM17 against X. euvesicatoria pv. euvesicatoria. The characterization of this antimicrobial activity may bring new insights on the mechanisms of control of bacterial spot in tomato by this promising biocontrol agent. [ABSTRACT FROM AUTHOR]

The COVID-19 pandemic has increased the incidence of depression and other mental disorders in the general population, influenced by various individual and contextual factors. Physical activity (PA) interventions offer a promising approach to mitigating the negative mental health effects of the pandemic. This study aims to analyze the association between PA and depressive symptoms. A total of 785 individuals aged 37.4 ± 13.2 years (72.5% female) were evaluated at two different time points: the first between 2018 and 2019, and the second during the COVID-19 pandemic in 2020. Depressive symptoms, demographic, and socioeconomic data were assessed using the Beck Depression Inventory to estimate depressive symptoms. Frequency analysis and binary and multinomial regression were employed for data analysis. The prevalence of mild depressive symptoms increased from 23.1% before the pandemic to 35.1% during the pandemic. Our findings reveal that practicing PA before the pandemic was a protective factor (OR: 0.19; 95% CI: 0.13, 0.30; p < 0.001) against mild depressive symptoms. Additionally, individuals who continued to practice PA during the pandemic had a lower chance of presenting mild (OR: 0.21; 95% CI: 0.15, 0.30) and moderate/severe (OR: 0.15; 95% CI: 0.08, 0.27) symptoms. Furthermore, our study shows that PA, which was already a protective factor before the pandemic, remained protective during the pandemic, even for those with the highest levels of depression. Choroid plexus tumors (CPTs) are rare intracranial neoplasms, representing <1% of all brain tumors, yet they represent 20% of first-year pediatric brain tumors. Although these tumors have been linked to TP53 germline mutations in the context of Li-Fraumeni syndrome, their somatic driver alterations remain poorly understood. In this study, we report two cases of lateral ventricle tumors: 3-yr-old male diagnosed with an atypical choroid plexus papilloma (aCPP), and a 6-mo-old female diagnosed with a choroid plexus carcinoma (CPC). We performed whole-exome sequencing of paired blood and tumor tissue in both patients, categorized somatic variants, and determined copy-number alterations. Our analysis revealed a tier II variant (Association for Molecular Pathology [AMP] criteria) in BRD1, a H3 and TP53 acetylation agent, in the aCPP. In addition, we detected copy-number gains on Chromosomes 12, 18, and 20 and copy-number losses on Chromosomes 13q and 22q (BRD1 locus) in this tumor. The CPC tumor had only a pathogenic germline TP53 variant, based on American College of Medical Genetics (ACMG) criteria, with a clinical and familiar history of Li-Fraumeni syndrome. The CPC patient presented loss of heterozygosity (LoH) of TP53 loci and hyperdiploid genome. Both tumors were microsatellite-stable. This is the first study performing whole-exome sequencing in Brazilian choroid plexus tumors, and in line with the literature, we corroborate the absence of recurrent somatic mutations in these tumors. Background: Physical exercise programs are typically composed of activities directed to the development of different physical abilities, usually stimulated in the same session. Objective: The aim of the study was to determine the effect of one session of aerobic exercise at high intensity to 1% and 10% gradient on the height (HJump) and kinematics of the depth jump (PExc - eccentric, concentric phase PCon, and contact time -- CT). Methods: Twenty-five moderately trained men (VO2Max 53.2 ± 4.3 mL.kg-1.min-1) attended five visits in the laboratory. Familiarity with the procedures in depth jump, VO2Max measures and their velocity associated (VVO2Max), and time to exhaustion performance (TLim) were performed at two initial visits. Results: On the three subsequent visits, the volunteers were subjected to three maximum depth jumps before and 10 min after the following conditions: (1) running intervals at high intensity of 10% gradient (R10%), (2) at 1% gradient (R1%), and control condition (CON). The order of conditions was determined randomly. A running condition did not induce significant changes from HJump (R1% 1.1% vs 1.0% R10%) when expressed as percentage difference from the CON condition. The PExc, PCon, and CT also did not change after running sessions (P> 0.05). None of the intervals running strategies were able to generate significant change in height and kinematics of the vertical depth jump. Conclusion: The prescription of the running at VVO2Max in 1% or 10% gradient does not seem to lead to concurrent effect, is suggested to ensure the concomitant development of maximal aerobic power and explosive strength. The present study demonstrates the biocontrol potential of a plant growth-promoting bacterial strain using three different approaches: (i) an in vitro evaluation of antagonistic activity against important phytopathogenic fungi; (ii) an evaluation under greenhouse conditions with strawberry plants to assess the control of gray mold; and (iii) an in silico whole genome sequence mining to assign genetic features such as gene clusters or isolated genes to the strain activity. The in vitro assay showed that the B.BV10 strain presented antagonistic activity, inhibiting the mycelial growth in all the phytopathogenic fungi evaluated. The application of the Bacillus velezensis strain B.BV10 under greenhouse conditions reduced the presence of Botrytis cinerea and increased the mean fruit biomass. The genome of B.BV10 was estimated at 3,917,533 bp, with a GC content of 46.6% and 4088 coding DNA sequences, and was identified as B. velezensis. Biosynthetic gene clusters related to the synthesis of the molecules with antifungal activity were found in its genome. Genes related to the regulation/formation of biofilms, motility, and the important properties for the rhizospheric colonization were also found in the genome. The current study offers a comprehensive understanding of the genomic architecture and control activity of phytopathogenic fungi by the B. velezensis strain B.BV10 that may substantiate the industrialization of this strain in the future. The use of grasses as ground cover in tropical climate guarantees soil protection against erosion processes, however, its capacity for nutrients mineralization is poorly understood. In this sense, the aim of the study was to evaluate the decomposition of vegetable residues from grasses and the mineralization of macronutrients in tropical conditions. The study was conducted in a randomized block design with four replications, in a 4 × 9 factorial scheme with four cultural residues: sorghum, millet, black oats, and fallow; and nine evaluation times: 0, 14, 28, 42, 56, 70, 84, 98, and 112 days after setting up the study (DAA), in the field. Vegetable residues were disposed on the soil surface for remaining dry matter (RDM), half-life (t1/2) and mineralization rates. Lower decomposition rates were observed in sorghum and millet residues, with RDMs of 40% and 31% when compared to oats and fallow (17% RDM). Thus, the waste mineralization rates decreased in the following order: fallow = oats > millet > sorghum. The mineralization of macronutrients was considered high and decreased in the following order: K > P > N > C > Mg > S > Ca, corresponding to 96, 88, 88, 80, 76, and 74%. The fallow, millet, oat, and sorghum residues released 84, 17, 225, and 17 kg t−1 of N, P, K, and S, with different mineralization rates. The development of bio-based products has increased in recent years, and species of the Bacillus genus have been widely used for product development due to their elevated production of antimicrobial molecules and resistance to extreme environmental conditions through endospore formation. In this context, the antifungal potential of Bacillus velezensis CMRP 4489 was investigated using in silico predictions of secondary metabolites in its genome and in vitro tests against the following phytopathogenic fungi: Sclerotinia sclerotiorum, Macrophomina phaseolina, and Botrytis cinerea. The in-silico predictions indicated that CMRP 4489 possesses several Biosynthetic Gene Clusters (BGCs) capable of producing molecules with antifungal properties and other non-identified BGCs. The in vitro assay results evidenced strong antifungal activity, inhibiting more than 60% of the tested fungi, and the isolate's molecules were stable under diverse physicochemical conditions. The in vitro assay evidenced significant antifungal activity, deformation of the hyphal structure in SS, biofilm formation capacity, and swarming motility. In the colonization assay, we observed attachment, colonization, and net-shaped biofilm formation, with the strain transitioning from the seeds to nearby structures. Therefore, CMRP 4489 showed to be a potential biocontrol agent against various diseases with agronomic importance and can be used under adverse environmental conditions. [ABSTRACT FROM AUTHOR]

BACKGROUND: Treatment with low temperatures can be used to quarantine mangoes against fruit‐fly infestation. However, mangoes can develop chilling injury (CI) when stored at temperatures below 13 °C. We demonstrated that the immersion in polyol solutions can alleviate CI symptoms in 'Palmer' mangoes stored at 8 °C. These suggest that polyols can be used to reduce CI in mangoes during quarantine at low temperatures. Thus, we investigated the efficacy of applying 0.1% (v/v) glycerol, propylene glycol, or sorbitol to 'Palmer' mangoes subjected to cold treatment (1.0 °C) for 28 days. Mangoes were then ripened at 23 °C for 7 days. RESULTS: Among these polyols, sorbitol was the most effective in alleviating CI for up to 14 days of cold treatment. Mangoes treated with sorbitol showed lower levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2), and reduced polyphenol oxidase (PPO) activity. These fruit also had elevated levels of ascorbate (AsA), especially in the epicarp, and increased superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) activity. Shoot organogenesis is critical for the shortening of long breeding cycles and circumvent the barrier of cloning mature Eucalyptus cloeziana trees. It enables large-scale production of plants from transformed tissues. This study evaluates the effect of α-naphthaleneacetic acid (NAA), thidiazuron (TDZ) and benzylaminopurine (BAP) on the organogenesis of E.cloeziana from hypocotyls and cotyledonary leaves. In the induction stage, hypocotyls and cotyledonary leaves were established in a Murashige and Skoog (MS) culture medium supplemented with NAA or TDZ. Callus tissues were cultivated in a MS culture medium containing only BAP or different concentrations of BAP/NAA in the differentiation stage. Adventitious buds were multiplied in vitro and elongated in a WPM culture medium supplemented with 0.89 µM BAP and 0.05 µM NAA. Cotyledonary leaves exhibited the best in vitro regeneration. The induction of adventitious buds occurred only in calluses induced from TDZ. In the differentiation stage, 4.4 µM BAP treatment promoted an increase of adventitious bud regeneration. Micro-cuttings from regenerated shoots were acclimatized and rooted ex vitro in mini-incubators. The results confirm the establishment of an efficient protocol for the in vitro regeneration of E. cloeziana by indirect organogenesis, providing new insights regarding cloning of this species. Soybean and maize are some of the main drivers of Brazilian agribusiness. However, biotic and abiotic factors are of great concern, causing huge grain yield and quality losses. Phosphorus (P) deficiency is important among the abiotic factors because most Brazilian soils have a highly P-fixing nature. Thus, large amounts of phosphate fertilizers are regularly applied to overcome the rapid precipitation of P. Searching for alternatives to improve the use of P by crops is essential to reduce the demand for P input. The use of multifunctional rhizobacteria can be considered one of these alternatives. In this sense, the objective of the present work was to select and validate bacterial strains with triple action (plant growth promoter, phosphate solubilizer, and biocontrol agent) in maize and soybean, aiming to develop a multifunctional microbial inoculant for Brazilian agriculture. Bacterial strains with high indole acetic acid (IAA) production, phosphate solubilization, and antifungal activity against soil pathogenic fungi (Rhizoctonia solani, Macrophomina phaseolina, and Fusarium solani) were selected from the maize rhizosphere. Then, they were evaluated as growth promoters in maize under greenhouse conditions. Based on this study, strain 03 (Ag75) was selected due to its high potential for increasing biomass (root and shoot) and shoot P content in maize. This strain was identified through genomic sequencing as Bacillus velezensis. In field experiments, the inoculation of this bacterium increased maize and soybean yields by 17.8 and 26.5%, respectively, compared to the control (25 kg P2O5). In addition, the inoculation results did not differ from the control with 84 kg P2O5, indicating that it is possible to reduce the application of phosphate in these crops. Thus, the Ag75 strain has great potential for developing a multifunctional microbial inoculant that combines the ability to solubilize phosphate, promote plant growth, and be a biocontrol agent for several phytopathogenic fungi. Low Field Time-Domain Nuclear Magnetic Resonance (TD-NMR) relaxometry was used to determine moisture, fat, and defatted dry matter contents in "requeijão cremoso" (RC) processed cheese directly in commercial packaged (plastic cups or tubes with approximately 200 g). Forty-five samples of commercial RC types (traditional, light, lactose-free, vegan, and fiber) were analyzed using longitudinal (T1) and transverse (T2) relaxation measurements in a wide bore Halbach magnet (0.23 T) with a 100 mm probe. The T1 and T2 analyses were performed using CWFP-T1 (Continuous Wave Free Precession) and CPMG (Carr-Purcell-Meiboom-Gill) single shot pulses. The scores of the principal component analysis (PCA) of CWFP-T1 and CPMG signals did not show clustering related to the RC types. Optimization by variable selection was carried out with ordered predictors selection (OPS), providing simpler and predictive partial least squares (PLS) calibration models. The best results were obtained with CWFP-T1 data, with root-mean-square errors of prediction (RMSEP) of 1.38, 4.71, 3.28, and 3.00% for defatted dry mass, fat in the dry and wet matter, and moisture, respectively. Increasing phosphorus (P) use efficiency in agricultural systems is urgent and essential to significantly reduce the global demand for this nutrient. Applying phosphate-solubilizing and plant growth-promoting bacteria in the rhizosphere represents a strategy worthy of attention. In this context, the present work aimed to select and validate bacterial strains capable of solubilizing phosphorous and promoting maize growth, aiming to develop a microbial inoculant to be used in Brazilian agriculture. Bacterial strains from the maize rhizosphere were evaluated based on their ability to solubilize phosphate and produce indole acetic acid. Based on these characteristics, 24 strains were selected to be further evaluated under laboratory, greenhouse, and field conditions. Among the selected strains, four (I04, I12, I13, and I17) showed a high potential to increase maize root growth and shoot P content. Strains I13 (Ag87) and I17 (Ag94) were identified by genomic sequencing as Bacillus megaterium and Lysinibacillus sp., respectively. These strains presented superior yield increments relative to the control treatment with 30% P. In addition, combining Ag87 and Ag94 resulted in even higher yield gains, indicating a synergistic effect that could be harnessed in a commercial inoculant for Brazilian agriculture. Micropropagation technique has been recommended for plant tissue rejuvenation/reinvigoration and the improvement of clonal plants production commercially. Concerning the limiting factors on in vitro cultivation of Eucalyptus grandis × E. urophylla hybrid clone, the effects of light intensity on in vitro introduction and multiplication phases were assessed. The experimental tissue was collected from ministumps cultivated in a semi-hydroponic system. The results of light intensity on in vitro introduction and multiplication were evaluated with four treatments (fluorescent lamp/40 µmol m−2 s−1, red/blue LEDs 20 µmol m−2 s−1, red/blue LEDs 40 µmol m−2 s−1, and red/blue LEDs 80 µmol m−2 s−1). The explants' morphological, anatomical, and genetic characteristics were evaluated at 30 d in the in vitro introduction, and 12 subcultures for in vitro multiplication. Based on the results, the treatments F/L 40 µmol m−2 s−1 and R/B 80 µmol m−2 s−1 are the most suitable during in vitro introduction and multiplication of Eucalyptus grandis × E. urophylla. It provides a higher percentage of responsive explants, shoots per explant, vigor, photosynthetic pigment content, adaxial epidermis thickness, abaxial epidermis, mesophyll, palisade parenchyma, spongy parenchyma, and vascular tissues. Furthermore, it was found that the micropropagated plants are identical clones of the Eucalyptus grandis × E. urophylla selected tree; that is, there was no somaclonal variation during the twelve subcultures on in vitro multiplication. Purpose: Medulloblastoma is the most frequent pediatric malignant brain tumor, and is divided into four main subgroups: WNT, SHH, group 3, and group 4. MYCN amplification is an important medulloblastoma prognostic biomarker. We aimed to molecular classify and predict MYCN amplification in a single assay. Methods: It was included 209 medulloblastomas from 205 patients (Brazil, Argentina, and Portugal), divided into training (n = 50) and validation (n = 159) sets. A nCounter assay was carried out using a custom panel for molecular classification, with additional genes, including MYCN. nSolver 4.0 software and the R environment were used for profiling and MYCN mRNA analysis. MYCN amplification by FISH was performed in 64 cases. Results: The 205 medulloblastomas were classified in SHH (44.9%), WNT (15.6%), group 3 (18.1%) and group 4 (21.4%). In the training set, MYCN amplification was detected in three SHH medulloblastomas by FISH, which showed significantly higher MYCN mRNA counts than non-FISH amplified cases, and a cutoff for MYCN amplification was established ( X ¯ + 4σ = 11,124.3). Applying this threshold value in the validation set, we identified MYCN mRNA counts above the cutoff in three cases, which were FISH validated. Conclusion: We successfully stratified medulloblastoma molecular subgroups and predicted MYCN amplification using a single nCounter assay without the requirement of additional biological tissue, costs, or bench time. BACKGROUND: Sunscald is a physiological disorder that occurs in many horticultural products when exposed to excessive solar radiation and high temperatures. Traditionally, sunscald is controlled using physical barriers that reflect radiation, however this practice is not always efficient. A possible alternative would be the use of chemical barriers, such as mycosporine‐like amino acids (MAAs), which protect aquatic organisms against ultraviolet (UV) radiation. Thus, this study aimed to develop a lipid‐based emulsion containing MAAs for using in the preharvest of horticultural products. RESULTS: Emulsions were developed using 10% (w/v) of corn oil (CO) and soybean oil (SO), carnauba wax (CW), and beeswax (BW) as lipid bases (LBs). The emulsion containing CW and ammonium hydroxide was the most stable, resembling commercial wax. Therefore, this formulation was used as the basis for the incorporation of the commercial product Helioguard™ 365, a source of MAA, in concentrations of 0%, 1%, 2%, and 4% (v/v). The MAA incorporation resulted in little modifications in the stability of the emulsion, providing an increase in the absorbance with peaks in the UV‐B ranging from 280 to 300 nm. The spatial and seasonal distributions of benthic marine organisms are generally linked to the physical and chemical features of the environment. The abundance of Libinia ferreirae Brito Capello, 1871 was analysed in relation to the environmental factors: water temperature (surface and bottom), water salinity (bottom), and substratum (texture and organic matter of the sediment). Samples were collected monthly, from January 1998 to December 1999, in the Ubatumirim (UBM), Ubatuba (UBA) and Mar Virado (MV) Bays, on the northern coast of São Paulo State, Brazil. We caught a total of 213 spider crabs (Ubatumirim = 39; Ubatuba = 53 and Mar Virado = 121). The abundance of L. ferreirae was highest during the spring (October to December) in the three bays. We assume that this result is associated with the entrance of the South Atlantic Central Water during this season, whose features favoured primary productivity and, consequently, displayed a higher food availability for other trophic levels. The lowest number of specimens found during the autumn might be caused by the migration of the population to other sites. The high abundance of L. ferreirae in MV is probably associated with sediment composition, i. e., fine grains which benefit the species' habit of burrowing in the sediment. Hence, the combination of a favourable sediment homogeneity and the highest food availability seems to be effective modulators of the abundance and distribution of L. ferreira in the bays. Freshwater crustaceans of the genus Aegla are endemic to southern South America. In Brazil, the greatest diversity of aeglid species occurs in river basins in the Rio Grande do Sul state. Nevertheless, recent studies have shown that the potential for discovering new species of Aegla in the Paraná state is huge. In recent years, for example, seven new species have been described for the state of Paraná. Furthermore, there is a need to solve problems related to cryptic species and refine taxonomic information about the group. Here we expand our current knowledge of the freshwater biodiversity of Aegla in the region by describing three new species. Aegla nanopedis sp. nov., Aegla abrupta sp. nov., and Aegla santosi sp. nov. were discriminated based on morphological and molecular evidence. Particular morphological characters of the carapace, chelipeds, epimeron and uropods distinguished the new taxa. Molecular results also support the separation of the new species from the closely related species A. jacutinga, A. castro and A. lata. The new species have limited distribution and are categorised as endangered (Aegla nanopedis sp. nov.) and vulnerable (Aegla abrupta sp. nov. and Aegla santosi sp. nov.) according to International Union for Conservation of Nature Red List Categories and Criteria. We characterized the natural diet and feeding dynamics of Aegla castro Schmitt, 1942 (Aeglidae), an endemic freshwater crab. Research was undertaken on the trophic spectrum of the species by analyzing the stomach contents of individuals collected in a stream in southeastern Brazil, and evaluating the diet through the index of relative importance (IRI). Seasonal and circadian variations in diet composition were analyzed, and the feeding dynamics investigated through repletion index (RI) analysis. We analyzed a total of 240 stomachs and identified 20 main food categories of the diet, which were studied throughout the seasons. Aegla castro maintained a constant RI throughout the year, with a peak of food activity between midnight (00) and 0600. The species was characterized as an omnivorous generalist and opportunist. The high overlay in the diet composition of adults of both sexes and juveniles indicates that they share the same food resources, even though they seem to exploit them in different quantities and frequencies. Future research should focus on the species of Aegla whose diets have not yet been described, and use available data to inform trophic network analyses and models. [ABSTRACT FROM AUTHOR]

Aegla castro, is an endemic anomuran crab from Brazil found in headwater streams of the Atlantic Forest on the Paranapanema River basin. This study aims to evaluate the gonadal maturity of A. castro based on macroscopic and microscopic analyses. Crabs were collected in Mauá da Serra (23º57' S; 51º06ʹ W), Paraná, Brazil in the winter and spring of 2015, and summer, autumn, and spring of 2016. Individuals had their gonads analysed and categorized into stages of development. Stage1, ovaries are macroscopically undetectable through abdominal transparency, while ovaries of stages 2 and 3 could be detected through abdominal transparency and have characteristic colour and length in relation to the abdominal somites. Ovaries have pre-vitellogenic and vitellogenic oocytes in all stages of development, with germ cells and structural epithelium characteristic of these gonadal maturation phases. Spermatogenesis in males begins in spring, with the production of spermatozoa in autumn and spermiation in winter. Aegla castro has seasonal reproduction. The macroscopic stages of gonadal development correspond to the histological gonadal maturation, indicating that the macroscopic observation of the gonads of live animals is feasible in studies of the reproductive cycle of aeglids. The main objective of this study was to evaluate Bacillus velezensis strain CMRP 4490 regarding its ability to inhibit soil-borne plant pathogens and to increase plant growth. The study included evaluation of in vitro antifungal control, sequencing the bacterial genome, mining genes responsible for the synthesis of secondary metabolites, root colonization ability, and greenhouse studies for the assessment of plant growth–promoting ability. The strain was obtained from soil samples in the north of Paraná in Brazil and was classified as a B. velezensis , which is considered a promising biological control agent. In vitro assay showed that B. velezensis CMRP 4490 presented antagonistic activity against Sclerotinia sclerotiorum , Macrophomina phaseolina , Botrytis cinerea , and Rhizoctonia solani with a mycelial growth inhibition of approximately 60%, without any significant difference among them. To well understand this strain and to validate its effect on growth-promoting rhizobacteria, it was decided to explore its genetic content through genome sequencing, in vitro , and greenhouse studies. The genome of CMRP 4490 was estimated at 3,996,396 bp with a GC content of 46.4% and presents 4,042 coding DNA sequences. Biosynthetic gene clusters related to the synthesis of molecules with antifungal activity were found in the genome. Genes linked to the regulation/formation of biofilms, motility, and important properties for rhizospheric colonization were also found in the genome. Application of CMRP 4490 as a coating film on soybean increased from 55.5 to 64% on germination rates when compared to the control; no differences were observed among treatments for the maize germination. We investigated the population dynamics of the spider crab Libinia ferreirae, focusing on the frequency distribution of individuals in size classes, sex ratio and the action of environmental variables (temperature, salinity, texture and organic matter content in the sediment) on reproduction and recruitment. Monthly collections were made in the Ubatuba region from January 1998 to December 2000. A total of 222 individuals were collected, including 123 juveniles (males and females), 43 adult males, 25 non-ovigerous adult females and 31 ovigerous females. Unlike most adult brachyurans, there was no significant size difference between sexes, and sexual dimorphism seems to be a varying characteristic for this crab genus. The reproductive period and recruitment were continuous with peaks that could be related to water mass dynamics and higher food availability in the Ubatuba region. This study was motivated by the data about a Rimapenaeus constrictus population from Ubatuba Bay, obtained in the 1990s, and by the establishment of a marine protected area (MPA) in the same region in 2008. We obtained comparable data about the distribution and abundance of this species collected 20 years apart, to evaluate the effectiveness of the MPA, for its preservation and possible changes in the populational profile. Biotic and abiotic data were obtained monthly, in September 1995–August 1996 (period 1 = 1ºP), and in September 2016–August 2017 (period 2 = 2ºP), from five transects within Ubatuba Bay. In total, 710 and 2,362 individuals were caught in the 1oP and 2oP, respectively. Despite this increase in abundance, aspects of the spatial distribution did not change substantially. The high abundance recorded at 5 m of depth was probably due to the creation of the MPA and to the accumulation of detritus at this depth. The high abundance seen in the transect sheltered from waves may be related to sediment heterogeneity, which was higher in this transect, and to the fact that this area is naturally protected from fisheries. In the 1ºP, the highest abundances were recorded in spring and summer, whereas in the 2ºP, they occurred in autumn and winter. This change may be explained by the entrance of the South Atlantic Central Water (SACW), which drives shrimps towards shallower areas. During the 2ºP, a fishing ban occurred in autumn (March 1st until May 31st), which is also when the SACW retreats and water temperature and salinity increase. These two factors combined may have been responsible for the increased shrimp abundance. In conclusion, both conservation practices (MPA and fishing ban) along with the existence of an area naturally protected from fisheries seem to have positively affected the population of R. constrictus, leading to an increase in abundance over the 20‐year interval. Soil fertilization with dehydrated sewage sludge (DSS) accelerates the recovery process of degraded areas by improving nutrient concentration, and favors the development of trophic webs with pioneer plants such as Acacia auriculiformis A. Cunn. ex Beth (Fabales: Fabaceae), phytophagous Hemiptera, predators, and protocooperanting ants. This study aimed to evaluate the development and production of A. auriculiformis litter with or without dehydrated sewage sludge application and the ecological indices of sucking insects (Hemiptera), their predators and protocooperating ants, as bioindicators, in a degraded area for 24 months. Complete randomization was applied for two treatments (with or without application of dehydrated sewage sludge) in 24 replications (one repetition = one plant). We evaluated the number of leaves/branch and branches/plant, percentage of soil cover (litter), ecological indices of phytophagous Hemiptera, their predators, and protocooperating ants. The plants of A. auriculiformis, that were applied with dehydrated sewage sludge, had superior development when compared to plants where DSS were not applied. The highest abundance and richness of phytophagous Hemiptera species and Sternorrhyncha predators occurred on A. auriculiformis plants that were applied with dehydrated sewage sludge. The increase in richness of species of protocooperanting ants that established mutualistic relationships positively influenced the phytophagous Hemiptera. The use of A. auriculiformis, with application of dehydrated sewage sludge, can increase recovery of degraded areas due to its higher soil cover (e.g., litter) and results in higher ecological indices of phytophagous Hemiptera and their predators. Nutrients from dehydrated sewage sludge play an essential role in the development of many plants such as Terminalia argentea, in the recovery of degraded areas. The aims were to assess the abundance, diversity and species richness of phytophagous, pollinators and predators arthropods, as well as the percentage of defoliation of T. argentea trees, fertilized (or not) with dehydrated sewage sludge in a degraded area. The abundance, diversity and species richness of phytophagous Coleoptera and total predators (predator insects + protocooperating ants + spiders); abundance and species richness of Diptera, pollinator insects, spiders, and predators (predator insects + spiders) were higher on trees fertilized with dehydrated sewage sludge. The abundance of phytophagous Coleoptera declined with the presence of phytophagous Hemiptera and protocooperating ants; population of phytophagous Orthoptera declined in response to phytophagous Coleoptera and total predators; the numbers of the leafminer Lyriomyza sp. directly increased with the numbers of spiders. The ecological indices of phytophagous, pollinators, and predator arthopods increased on Terminalia argentea trees fertilized with dehydrated sewage sludge; such a better ecological indices in fertilized than in unfertilized trees, show it more suitable for the recovery of degraded areas. Summary: Due to the importance of banana consumption in developing countries and producing zinc‐rich foods, the objective of this study was to evaluate the feasibility of banana bunch sprays and bunch stalk feeding with Zn aiming to biofortify this fruit. Zn sprays were carried out with a dose of 80 g ha−1 using ZnCl2 and ZnCl2 combined with a chelating agent in two banana cultivars ('Maravilha' and 'FHIA 18'). Bunch stalk feeding with Zn was carried out by fixing plastic bags on the stem end containing solutions of ZnSO4 (0, 0.5, 1.0 and 2.0 g L−1) in two banana cultivars ('Williams' and 'Prata Anã'). Both strategies resulted in a maximum fruit Zn content of 0.14 mg 100 g−1. Although it was possible to increase the Zn content in banana fruit from 'FHIA 18' and 'Prata Anã' cultivars, both strategies did not result in biofortified banana fruit with a Zn level which would fulfil the Zn dietary reference intake (DRI) of 2 mg dia−1 for children. Banana bunch sprays with Zn did not affect the fruit quality, but bunch stalk feeding with Zn resulted in fruits with lower ratio, pH and ascorbic acid content. [ABSTRACT FROM AUTHOR]

Computer Aided Design of Chemical Process is now a well established eld in the design of new process as well as in the optimization, revamp and retrot of existing ones. The use of powerful process simulators available today allows the process engineer to simulate even an entire process, but the majority of process simulators have only classical unit operations. So, if the process has a non-classical unit operation it needs to be simulated using a suitable computer language and further to be linked to the simulator. In this paper we addressed the problem of including a new unit operation in a process simulator and how to use the virtual plant to optimize and to evaluate the environmental impact of a chemical process. We used the free chemical process simulator COCO to simulate two styrene process production plant. The firrst one uses as a reactor a conventional PFR that is available in the simulator. The second plant uses a membrane reactor that was simulated using the software Scilab that was embedded in COCO simulator by using the CAPE-OPEN protocol. Then, we used both virtual plants to develop meta-models of the processes by using experimental design and surface responses. These empirical models were used after to optimize the plants and the results shown that it is possible to increase the styrene productivity up to 27.32 kmol/h using a PFR reactor and up to 30.56 kmol/h using a membrane reactor. Finally, we calculated the Potential of Environmental Impact (PEI) for each process using the WAR algorithm and we shown that both processes have PEI very similar. Therefore, the route that uses membrane reactor has an advantage over the route that uses PFR reactor since it allows to obtain higher styrene productivities. [ABSTRACT FROM AUTHOR]

Aegla lata Bond-Buckup & Buckup, 1994 is a critically endangered species, extinct in its type-locality. Its currently known geographical distribution is restricted to small streams of the lower Tibagi River Basin (Paraná state, southern Brazil), where it is vulnerable to the environmental degradation that takes place in these watersheds. Scientific knowledge on the species is limited to the original description and a few occurrence records. No information about its biology is available. Aiming to fill this gap, we investigated the relative growth, sexual maturity, sexual dimorphism, and the reproductive and recruitment periods of A. lata. Monthly sampling occurred from May 2014 through June 2015 in the Bule stream in the southern limit of Mata dos Godoy State Park, Londrina, Paraná state, Brazil (23°27ʹ20.86″S, 51°16ʹ32.44″W). The analysis of allometric growth was used to differentiate juveniles and adults. According to the development stage of the chelae, two sequential groups of adult males were recognized (morphotypes I and II). The estimated average cephalothorax length (CL) at the onset of morphometric maturity of males and females are similar, suggesting puberty molt takes place at nearly the same size in both sexes. Males and females are heterochelous, with a clear predominance of the left chela as the most developed of the pair. Both chelae are larger in adult males than in adult females of similar CL. The reproductive period is markedly seasonal, extending from the austral autumn to winter (May to August), as well as those recorded in other species of Aegla from similar latitudes. Summary: Alcohol‐free beer with isotonic properties is getting more popular and its production can be carried out by different production strategies; however, interrupted fermentation is still a challenge. Therefore, the objective of this study was to develop a low‐alcohol isotonic beer (<0.5% v/v) by interrupted fermentation. Moreover, the major objective is to compare the developed product to commercial beverages (sports drinks, 'Pilsen' regular beer, alcohol‐free beers and low‐alcohol isotonic beer). The beverages were evaluated based on pH, alcohol content (% v/v), total titratable acidity (mEq L−1), osmolality (mOsmol kg−1), bitterness International Bitterness Units, colour European Brewery Convention, total phenolic compounds (mg L−1 gallic acid), reducing and total sugars (%) and Na and K contents (mg L−1). The developed low‐alcohol isotonic beer presented characteristics similar to sports drinks, with the advantage of being richer in phenolic compounds and suitable osmolality. Despite salts were added in its formulation, the grades attributed to all beers employed in the sensory evaluation, as well as the purchase intention did not present significant differences.
