Copper (Cu) dry etching is demonstrated using a narrow-gap hydrogen plasma generated at 13.3 kPa (100 Torr) for applications in the Cu wiring technology of integrated circuits. A localized hydrogen plasma is generated around the apex of a fine pipe electrode. The Cu etching can be observed only when the process gas contains hydrogen, and the etching rates decrease with decreased hydrogen concentration. The plasma heating effect owing to plasma localization is negligible for the Cu etching because no etching occurs in the presence of pure N2 plasma whose volume is almost equal to that of the pure H2 plasma. Furthermore, the influences of physical sputtering and vacuum ultraviolet irradiation on the Cu etching are confirmed to be insignificant by exposing the samples to rare-gas plasma. The maximum Cu etching rate of 500 nm/min can be achieved at a stage temperature of 0 °C. However, the Cu etching rate has no obvious dependence on the stage temperature in a range from -20 to 330 °C. In contrast, the etching rates for Si and SiO2 at a stage temperature of 0 °C are 100 lm/min and 50 nm/min, respectively. The Cu etching rate is 10 times higher than that of SiO2, which implies that this etching technique has potential applications for Cu wiring on an SiO2 layer. The Cu surface etched by the hydrogen plasma is roughened and exhibits many round pits and bumps, which seems to be owing to excessive incorporation of the diffused hydrogen in the Cu bulk. Cellular proteins do not work in isolation. Instead, they often function as part of large macromolecular complexes, which are transported and concentrated into specific cellular compartments and function in a highly crowded environment. A central theme of modern cell biology is to understand how such macromolecular complexes are assembled efficiently and find their destinations faithfully. In this Opinion article, we will focus on HEAT repeats, flexible arrays of amphiphilic helices found in many eukaryotic proteins, such as karyopherins and condensins, and discuss how these uniquely designed helical repeats might underlie dynamic protein-protein interactions and support cellular functions in crowded environments. We will make bold speculations on functional similarities between the action of HEAT repeats and intrinsically disordered regions (IDRs) in macromolecular phase separation. Potential contributions of HEAT-HEAT interactions, as well as cooperation between HEATs and IDRs, to mesoscale organelle assembly will be discussed. [ABSTRACT FROM AUTHOR]

Elongation of seminal and lateral roots of rice seedlings was markedly inhibited by high ammonium levels in growth medium. However, high exogenous nitrate concentrations had little inhibitory effect on root growth. The objective of this study was to elucidate the relationship between inhibition of rice root growth induced by high ammonium conditions and ammonium assimilation in the seedlings. Activity of glutamine synthetase (GS) was kept at a low level in the seminal roots of the seedlings grown under high nitrate levels. In contrast, high ammonium levels significantly enhanced the GS activity in the roots, so that Gln abundantly accumulated in the shoots. These results indicate that ammonium assimilation may be activated in the seminal roots under high ammonium conditions. Application of methionine sulfoximine (MSO), an inhibitor of GS, relieved the repression of the seminal root elongation induced by high ammonium concentrations. However, the elongation of lateral roots remained inhibited even under the same condition. Furthermore, MSO drastically increased ammonium level and remarkably decreased Gln level in the shoots grown under high ammonium conditions. These results show that, for rice seedlings, an assimilatory product of ammonium, and not ammonium itself, may serve as an endogenous indicator of the nitrogen status involved in the inhibition of seminal root elongation induced by high levels of exogenous ammonium. Abstract  In order to study the effect of repression of 14-3-3 genes on actual activity of the nitrate reductase (NR) in Nicotiana benthamiana leaves, Nb14-3-3a gene was silenced by virus-induced gene silencing (VIGS) method using potato virus X (PVX). Expression of Nb14-3-3a as well as Nb14-3-3b genes was altogether repressed in the leaves of PVX-14-3a-infected plants. Furthermore, two-dimensional gel electrophoresis and immunoblot analysis with anti-14-3-3 antiserum suggested that the expressions of Nb14-3-3a and Nb14-3-3b proteins are accordingly repressed in PVX-14-3a-infected plants. It is well known that binding of 14-3-3 proteins to phosphorylated NR leads to substantial decrease in NR activity of leaves under darkness. Therefore, we studied the changes in NR activity in response to light/dark transitions in the leaves of PVX-14-3a-infected plants. NR activation state was kept at a high level under darkness in PVX-14-3a-infected plants, but not in PVX-green fluorescent protein (GFP)-infected and control plants. This result suggests that Nb14-3-3a and/or Nb14-3-3b proteins are indeed involved in the inactivation of NR activity under darkness in N. benthamiana. [ABSTRACT FROM AUTHOR]

We have investigated the electromagnetic and thermal properties of a REBaCuO ring bulk with an inhomogeneous critical current density, Jc, profile during pulsed field magnetization (PFM) using a numerical simulation and compared those to a bulk with a homogeneous Jc profile. A notch was introduced in the bulk periphery, which was assumed as a crack existing in the actual bulk material. A sudden flux penetration (flux jump) took place through the notch area and as a result, a large temperature rise also took place around this notch. Consequently, the final trapped field profile was simulated to be a ‘C-shaped profile’, which qualitatively reproduced our previous experimental results. The size and position dependences of the notch on the flux penetration behaviour were also simulated, in which a larger and outer notch promotes the flux jump phenomenon easily. On the other hand, in the homogeneous model, under the same conditions, no flux jump phenomenon was observed. These results suggest that the imperfection in the bulk can be a possible starting point of the flux jump. The electromagnetic and thermal hoop stresses were also simulated in the ring bulk during PFM, in which the electromagnetic stress and the thermal stress were both observed to be lower than the fracture strength of the bulk material. The author discusses a study which demonstrated the role of condensin II in the formation of chromosome territories during interphase, by Bauer and colleagues, published within the issue. He commends the study authors for showing how interphase chromosomes could be organized, and describes the chromosome organization and dynamics of Drosophila melanogaster. He adds that the study reinforced the role of condensin II in chromosome organization.
