Background: Human papillomavirus (HPV) is a crucial prognostic factor in oropharyngeal cancer (OPC). p16 is a surrogate marker for diagnosing HPV+ OPC, however it is not direct evidence of HPV existence. Objective: The purpose of our study was to evaluate an HPV DNA test—Cobas HPV assay—in diagnosing HPV+ OPC through neck lymph node aspiration. Methods: Patients with suspected neck mass who received fine needle aspiration (FNA) or core needle biopsy (CNB) at the National Taiwan University Hospital between January 2018 and December 2022 were reviewed. Besides routine cytology and pathology study, needle rinse fluid was collected for the Cobas HPV assay to detect high-risk HPV. Results: We analyzed 137 patients with suspected lymph nodes, 32 (23.4%) of whom were HPV+ OPC patients and 105 (76.6%) of whom had non-HPV-related disease. FNA was performed in 31 patients and CNB was performed in 106 patients, according to the size and necrosis status of the lymph nodes. For diagnosing HPV+ OPC, CNB combined with p16 immunohistochemistry staining showed sensitivity of 93.3%, specificity of 97.8%, positive predictive value (PPV) of 87.5%, negative predictive value (NPV) of 98.9%, and accuracy of 97.2%. On the other hand, for the needle rinse Roche Cobas HPV assay, the test showed sensitivity of 96.9%, specificity of 100%, PPV of 100%, NPV of 99.1%, and accuracy of 99.3%. Compared with p16 IHC staining, the Cobas HPV test showed better PPV with statistical significance (p = 0.04). Conclusion: The Cobas HPV assay is a US FDA-approved, highly automated, and readily used technique to directly detect the presence of high-risk HPV. To investigate the feasibility and efficacy of combining ultrasound-guided sharp needle technique with percutaneous transluminal angioplasty (PTA) for treating outflow stenosis or dysfunction in arteriovenous fistula (AVF) among hemodialysis patients. From October 2021 to March 2023, patients with occluded or malfunctional fistula veins not amenable to regularly angioplasty were retrospectively enrolled in the study. They underwent ultrasound-guided sharp needle intervention followed by PTA. Data on the location and length between the two veins, technical success, clinical outcomes, and complications were collected. Patency rates post-angioplasty were calculated through Kaplan-Meier analysis. A total of 23 patients were included. The mean length of the reconstructed extraluminal segment was 3.18 cm. The sharp needle opening was performed on the basilic vein (60.9%), brachial vein (26.1%), or upper arm cephalic vein (13%) to create outflow channels. Postoperatively, all cases presented with mild subcutaneous hematomas around the tunneling site and minor diffuse bleeding. The immediate patency rate for the internal fistulas was 100%, with 3-month, 6-month, and 12-month patency rates at 91.3%, 78.3%, and 43.5%, respectively. Sharp needle technology merged with PTA presents an effective and secure minimally invasive method for reconstructing the outflow tract, offering a new solution for recanalizing high-pressure or occluded fistulas. Tuberculosis (TB) was a public health problem in the past and remained a threat worldwide, including in Taiwan. We aimed to estimate annual percentage changes (APC) in TB mortality and the age-period-cohort effects on TB mortality. Taiwan TB mortality data from 1978 to 2022 was used to estimate the APC through the Joinpoint regression analysis and age-period-cohort models. Crude and age-standardized mortality rates (ASMR) reduced from 23.46 to 3.02 and 41.70 to 1.59 per 100,000 population in males, while 8.84 to 1.09 and 14.58 to 0.45, respectively, in females. During the COVID-19 pandemic, the crude rates still significantly declined in males from 2020 to 2022 but not in females. APC in ASMR significantly decreased in males and females (-7.17% and -7.64%), while after adjusted age-period-cohort effects, the estimated APC were -8.83% and -9.77%. Age-period-cohort effects were observed, with the highest peaks in the youngest (aged 0–4) and the period 1978–1982 in both sexes and peaks in the male cohort 1898–1902 and female cohort 1903– 1907. TB mortality in Taiwan has declined in all ages, sexes, periods, and cohorts. However, the COVID-19 pandemic stopped TB mortality from decreasing. [ABSTRACT FROM AUTHOR]

Conflicting data exists regarding the baseline determinants of virological nonsuppression outcomes in treatment-experienced people living with human immunodeficiency virus (PWH) switching to antiretroviral treatment (ART) with bictegravir/emtricitabine/tenofovir alafenamide fumarate (BIC/FTC/TAF) or dolutegravir/lamivudine (DTG/3TC) in Asia. This retrospective observational study, conducted at a designated HIV-care hospital from October 2019 to January 2023, aimed to address this gap. We assessed the odds of virological nonsuppression (VNS) at weeks 48 using logistic regression. A total of 988 patients were included, 35 patients (3.5%) with VNS at week 48. Pre-existing primary resistance-associated mutations (RAM) to nucleoside reverse transcriptase inhibitor (NRTI) and non-nucleoside reverse transcriptase inhibitor (NNRTI) were identified in 11.0% (51/465) and 14.4% (67/465), respectively. The identified risk factor was a record of virological failure ≥2 times (AOR 5.32, 95% CI 2.04–13.85), while an HIV viral load <50 copies/mL within the past three months before switch (AOR: 0.27, 95% CI 0.11–0.72) was identified as a protective factor. No cases of acquired drug resistance-associated mutations were detected at week 48. Additionally, DTG/3TC was noninferior to BIC/FTC/TAF in achieving or maintaining HIV RNA levels of <50 copies/mL, within a -10% noninferiority margin in the per-protocol analysis (responder proportion: 98.2% vs. 95.0%, respectively; adjusted treatment difference [95% CI], 3.2% [0.7% to 5.3%]). In conclusion, DTG/3TC and BIC/FTC/TAF demonstrated good effectiveness in a real-world cohort, but frequent virological failure before the switch might impact the benefits of these regimens in the short term of follow-up. The Crepidinae are the largest subtribe of the Cichorieae (Asteraceae). Debate remains over the circumscription and phylogeny of this subtribe, mainly due to its complex morphology and the poor phylogenetic signal provided by traditional Sanger sequencing markers. In this study, a well‐resolved phylogeny of the subtribe Crepidinae, consisting of seven highly supported clades, was obtained for the first time using nuclear data with a phylogenomics approach (Hyb‐Seq). Using this phylogeny along with other evidence, we propose a new taxonomic framework for the Crepidinae with seven lines and 29 genera, which merges subtribe Chondrillinae with the Crepidinae. We also describe a new monotypic genus, Qineryangia, that is characterized by broad involucres, loose imbricate phyllaries with wavy margins, and thick pappus bristles. [ABSTRACT FROM AUTHOR]

Background: The effects of heat acclimation (HA) on the hypothalamus after exertional heatstroke (EHS) and the specific mechanism have not been fully elucidated, and this study aimed to address these questions. Methods: In the present study, rats were randomly assigned to the control, EHS, HA, or HA + EHS groups (n = 9). Hematoxylin and eosin (H&E) staining was used to examine pathology. Tandem mass tag (TMT)-based proteomic analysis was utilized to explore the impact of HA on the protein expression profile of the hypothalamus after EHS. Bioinformatics analysis was used to predict the functions of the differentially expressed proteins. The differential proteins were validated by western blotting. An enzyme-linked immunosorbent assay was used to measure the expression levels of inflammatory cytokines in the serum. Results: The H&E staining (n = 5) results revealed that there were less structural changes in hypothalamus in the HA + EHS group compared with the EHS group. Proteomic analysis (n = 4) revealed that proinflammatory proteins such as argininosuccinate synthetase (ASS1), high mobility group protein B2 (HMGB2) and vimentin were evidently downregulated in the HA + EHS group. The levels of interleukin (IL)-1β, IL-1, and IL-8 were decreased in the serum samples (n = 3) from HA + EHS rats. Conclusions: HA may alleviate hypothalamic damage caused by heat attack by inhibiting inflammatory activities, and ASS1, HMGB2 and vimentin could be candidate factors involved in the exact mechanism. Several pulsar timing array (PTA) collaborations have recently reported the evidence for a stochastic gravitational-wave background (SGWB), which can unveil the formation of primordial seeds of inhomogeneities in the early universe. With the SGWB parameters inferred from PTAs data, we can make a prediction of the seeds for early galaxy formation from the domain walls in the axion-like particles (ALPs) field distribution. This also naturally provides a solution to the observation of high redshifts by the James Webb Space Telescope. Background: The association of body mass index (BMI) with survival outcomes in patients with advanced non-small cell lung cancer (NSCLC) treated with first-line chemotherapy, immunotherapy, or chemoimmunotherapy is controversial. We aimed to investigate these associations, including associations in male and female patients specifically, in a multicenter cohort study. Methods: We retrospectively analyzed data from seven cohorts comprising 7021 advanced non-small cell lung cancer patients who received chemotherapy (three cohorts), immunotherapy (two cohorts), and chemoimmunotherapy (two cohorts) from five data sources, including a de-identified nationwide (US-based) NSCLC clinico-genomic database and two randomized, double-blind, phase 3 clinical trials. BMI was categorized as underweight, normal weight, overweight, or obese. Underweight patients were excluded because of their small proportion. The primary endpoints were the associations between BMI and progression-free survival (PFS) and overall survival (OS) stratified by treatment type and sex, which were assessed using Kaplan–Meier methods and adjusted Cox modeling. Meta-analyses were performed to combine the adjusted hazard ratios. Results: In the pooled analysis, obesity was significantly associated with improved OS in patients receiving chemotherapy (hazard ratios [HR] = 0.84, 95% confidence interval (CI) 0.76–0.93), but there was no association with PFS (HR = 0.91, 95% CI 0.82–1.02). The association of BMI with OS for patients receiving chemotherapy differed by sex, with an inverse association in men (HR = 0.74, 95% CI 0.64–0.84), but no association observed in women (HR = 0.96, 95% CI 0.81–1.13, Pinteraction = 0.018). No impact of BMI on OS or PFS was detected in patients receiving immunotherapy or chemoimmunotherapy. Obese patients had the lowest level of tumor mutational burden, similar level of programmed death-ligand 1 expression and ESTIMATE scores. Conclusions: Obesity may be associated with an increased overall survival among male patients treated with chemotherapy, whereas not associated with the outcomes in patients treated with immunotherapy or chemoimmunotherapy. The success of platinum-based chemotherapeutic drugs for clinical cancer treatments has inspired tremendous research efforts on developing new metallic anticancer agents with improved cytotoxic activity and reduced side effects. 2,2′;6′,2″-Terpyridine and its 4′-substituted derivatives have showed great potential as ligand compartments for designing new anticancer drug candidates involving base metals. In this work, we synthesized a series of cobalt and iron coordination compounds based on 4′-pyridyl-2,2′;6′,2″-terpyridine, including homoleptic complexes, a dinuclear bridged complex and 1- and 2-dimensional coordination polymers/networks. The polymorphism of two homoleptic CoII and FeII complexes has been described along with the structural characterization of a CoII coordination polymer and dinuclear FeIII complex by X-ray crystallography. These compounds were tested preliminarily as precatalysts for the regioselective hydrosilylation of styrene. Their cytotoxic activities against two human breast cancer cell lines (MCF-7 and MDA-MB 468) and a normal breast epithelial cell line (MCF-10A) were investigated in order to observe the best-performing drug candidates. Objective To investigate the dynamic developmental trajectories of early suicidal ideation among adolescents, and the influence of factors such as gender, left-behind status, only-child status, boarding status, and social support on the trajectories of suicidal ideation development. Methods A cluster sampling method was employed to conduct three follow-up surveys among 989 7th-grade students in a certain urban middle school in Hunan Province from March to December 2023. The adolescent suicidal ideation scale (PANSI) and adolescent social support scale (CASSS) were used for questionnaire surveys. A latent growth mixture model (LGMM) was utilized to analyze and explore different categories of early suicidal ideation among adolescents. Results Four potential categories of developmental trajectories of early suicidal ideation among adolescents were identified, labeled as C1 group "high-risk-stable" (7.2%), C2 group "medium-risk-worsening" (8.8%), C3 group "medium-risk-stable" (11.1%) and C4 group "low-risk-stable" (72.9%). Being female, left-behind, and only-child were identified as risk factors for the developmental trajectories of early suicidal ideation (all P < 0.05), while social support was identified as a protective factor for these trajectories. Background/Objectives: Brain tumor patients confront numerous challenges arising from diagnosis and treatment, and these impact the patient's physical, mental, and social functions at all levels. Primary informal caregivers assume a pivotal role in home-based patient care. Of particular importance are the coping strategies employed by family caregivers, as they can influence both their own health and the overall quality of home care. This study aimed to explore the associations among family function, caregiving competence, and coping strategies among primary informal caregivers. Methods: This study adopted a cross-sectional correlational design and convenience sampling to survey the primary informal caregivers of 111 brain tumor patients. The study instruments included the Family Assessment Device General Function, Caregiving Competence Scale, and Revised Ways of Coping Checklist. Results: The findings of this study revealed a significant positive correlation between the family function of primary informal caregivers and their employment of emotion-focused coping (r = 0.209, p < 0.05). Furthermore, caregiving competence exhibited a positive association with problem-focused coping (r = 0.242, p < 0.05) and emerged as a significant predictor of problem-focused coping (β = 0.182, p < 0.05). However, neither family function (r = 0.059, p < 0.05) nor caregiving competence (r = 0.031, p < 0.05) demonstrated significant associations with total coping strategies. Conclusions: The findings of this study affirmed that enhancing the caregiving competence of primary informal caregivers of brain tumor patients can facilitate the adoption of problem-focused coping strategies. Aim: This study utilized latent profile analysis to investigate care needs subgroups among older adults with urinary incontinence. Methods: The "Elderly Urinary Incontinence Care Needs Inventory" surveyed 510 participants in two Guangzhou City hospitals from July 2022 to June 2023. Latent profile analysis created a classification model, and variance and correlation analysis assessed influencing factors. Results: A total of 510 older adults with urinary incontinence participated. The standardized total care needs score was 78.77 ± 5.03, with variations across dimensions: social participation needs scored (71.16 ± 10.32), daily life care needs (78.80 ± 5.51), medical care needs (77.33 ± 12.17), psychological comfort needs (76.97 ± 6.51) and health education needs scored highest (82.67 ± 6.77). Three distinct profiles emerged: "medium," "high SPN‐PCN" and "high DLCN‐MCN‐HEN". The majority belonged to the "high SPN‐PCN" profile. Significant correlations were found with age, education, leaks and frequency of micturitions. Conclusion: Research findings showed the existence of three distinct categories, with a notable majority of participants belonging to the "high SPN‐PCN" group. The significance of having these classes identified lies in the move away from a one‐size‐fits‐all approach to a more nuanced understanding of care needs. Customized nursing interventions can be devised based on specific factors, such as age, education level, urinary incontinence‐related symptoms and potential category. For instance, for the "high SPN‐PCN" group, our nursing strategy can encompass heightened psychological support and expanded opportunities for social engagement.Furthermore, in the training and education of healthcare professionals, recognizing and meeting the needs of each potential category of older adults might require more attention. To investigate the role of miR-223-3p in the modulatory effect of paeonol (Pae) on high glucose (HG)-induced endothelial cell apoptosis. HG (25 mmol/L) was used to induce cellular damage and apoptosis in the mouse cardiac microvascular endothelial cells (MCMECs). Various concentration of Pae was tested and 60 μmol/L Pae was selected for the subsequent studies. MCMECs were transfected with exogenous miR-223-3p mimics or anti-miR-223-3p inhibitors. Cell viability was assessed by MTT assay and apoptosis was quantified by flow cytometry. The expression of miR-223-3p and NLRP3 mRNA was measured using real-time quantitative RT-PCR, and protein level of NLRP3 and apoptosis-related proteins was detected by immunoblotting. Pae significantly attenuated HG-induced apoptosis of MCMECs in a concentration-dependent manner. In addition, Pae (60 µmol/L) significantly reversed HG-induced down-regulation of miR-223-3p and up-regulation of NLRP3. Pae (60 µmol/L) also significantly blocked HG-induced up-regulation of Bax and Caspase-3 as well as down-regulation of Bcl-2. Moreover, exogenous miR-223-3p mimics not only significantly attenuated HG-induced apoptosis, but also significantly suppressed NRLP-3 and pro-apoptotic proteins in the MCMECs. In contrast, transfection of exogenous miR-223-3p inhibitors into the MCMECs resulted in not only significantly increased apoptosis of the cells, but also significant suppression of NLRP3 and pro-apoptotic proteins in the cells. Pae attenuated HG-induced apoptosis of MCMECs in a concentration-dependent manner. Fragile X syndrome (FXS) is the most common form of inherited intellectual disability and a monogenic cause of autism spectrum disorders. Deficiencies in the fragile X messenger ribonucleoprotein, encoded by the FMR1 gene, lead to various anatomical and pathophysiological abnormalities and behavioral deficits, such as spine dysmorphogenesis and learning and memory impairments. Synaptic cell adhesion molecules (CAMs) play crucial roles in synapse formation and neural signal transmission by promoting the formation of new synaptic contacts, accurately organizing presynaptic and postsynaptic protein complexes, and ensuring the accuracy of signal transmission. Recent studies have implicated synaptic CAMs such as the immunoglobulin superfamily, N-cadherin, leucine-rich repeat proteins, and neuroligin-1 in the pathogenesis of FXS and found that they contribute to defects in dendritic spines and synaptic plasticity in FXS animal models. This review systematically summarizes the biological associations between nine representative synaptic CAMs and FMRP, as well as the functional consequences of the interaction, to provide new insights into the mechanisms of abnormal synaptic development in FXS. Background The bla B, bla GOB and bla CME genes are thought to confer β-lactam resistance to Elizabethkingia anophelis , based on experiments conducted primarily on Escherichia coli. Objectives To determine the individual contributions of β-lactamase genes to increased MICs in E. anophelis and to assess their impact on the in vivo efficacy of carbapenem therapy. Methods Scarless gene deletion of one or more β-lactamase gene(s) was performed in three clinical E. anophelis isolates. MICs were determined by broth microdilution. Hydrolytic activity and expressions of β-lactamase genes were measured by an enzymatic assay and quantitative RT–PCR, respectively. In vivo efficacy was determined using Galleria mellonella and murine thigh infection models. Results The presence of bla B resulted in >16-fold increases, while bla GOB caused 4–16-fold increases of carbapenem MICs. Hydrolysis of carbapenems was highest in lysates of bla B-positive strains, possibly due to the constitutionally higher expression of bla B. Imipenem was ineffective against bla B-positive isolates in vivo in terms of improvement of the survival of wax moth larvae and reduction of murine bacterial load. The deletion of bla B restored the efficacy of imipenem. The bla B gene was also responsible for a >4-fold increase of ampicillin/sulbactam and piperacillin/tazobactam MICs. The presence of bla CME, but not bla B or bla GOB, increased the MICs of ceftazidime and cefepime by 8–16- and 4–8-fold, respectively. Conclusions The constitutionally and highly expressed bla B gene in E. anophelis was responsible for increased MICs of carbapenems and led to their poor in vivo efficacy. bla CME increased the MICs of ceftazidime and cefepime. Extracellular ATP (eATP) signaling through the P2X7 receptor pathway is widely believed to trigger NLRP3 inflammasome assembly in microglia, potentially contributing to depression. However, the cellular stress responses of microglia to both eATP and stress itself remain largely unexplored. Mitochondria-associated membranes (MAMs) is a platform facilitating calcium transport between the endoplasmic reticulum (ER) and mitochondria, regulating ER stress responses and mitochondrial homeostasis. This study aims to investigate how MAMs influence microglial reaction and their involvement in the development of depression-like symptoms in response to chronic social defeat stress (CSDS). CSDS induced ER stress, MAMs' modifications, mitochondrial damage, and the formation of the IP3R3-GRP75-VDAC1 complex at the ER-mitochondria interface in hippocampal microglia, all concomitant with depression-like behaviors. Additionally, exposing microglia to eATP to mimic CSDS conditions resulted in analogous outcomes. Furthermore, knocking down GRP75 in BV2 cells impeded ER-mitochondria contact, calcium transfer, ER stress, mitochondrial damage, mitochondrial superoxide production, and NLRP3 inflammasome aggregation induced by eATP. In addition, reduced GRP75 expression in microglia of Cx3cr1CreER/+Hspa9f/+ mice lead to reduce depressive behaviors, decreased NLRP3 inflammasome aggregation, and fewer ER-mitochondria contacts in hippocampal microglia during CSDS. Here, we show the role of MAMs, particularly the formation of a tripartite complex involving IP3R3, GRP75, and VDAC1 within MAMs, in facilitating communication between the ER and mitochondria in microglia, thereby contributing to the development of depression-like phenotypes in male mice. Malfunctioning of cellular organelles, such as the endoplasmic reticulum (ER) and mitochondria, can have a role in stress-induced pathophysiological changes. Background: Infantile liver failure syndrome type 1 (ILFS1, OMIM #615,438), caused by leucyl-tRNA synthase 1 (LARS1, OMIM *151,350) deficiency, is a rare autosomal-recessive disorder. The clinical manifestations, molecular-genetic features, and prognosis of LARS1 disease remain largely elusive. Methods: Three new instances of ILFS1 with confirmed variants in LARS1, encoding LARS1, were identified. Disease characteristics were summarized together with those of 33 reported cases. Kaplan-Meier analysis was performed to assess prognostic factors in ILFS1 patients. Results: The 3 new ILFS1 patients harbored 6 novel variants in LARS1. Among the 36 known patients, 12 died or underwent liver transplantation. The main clinical features of ILFS1 were intrauterine growth restriction (31/32 patients in whom this finding was specifically described), failure to thrive (30/31), hypoalbuminemia (32/32), microcytic anemia (32/33), acute liver failure (24/34), neurodevelopmental delay (25/30), seizures (22/29), and muscular hypotonia (13/27). No significant correlations were observed between genotype and either presence of liver failure or clinical severity of disease. Kaplan-Meier analysis indicated that age of onset < 3mo (p = 0.0015, hazard ratio = 12.29, 95% confidence interval [CI] = 3.74–40.3), like liver failure (p = 0.0343, hazard ratio = 6.57, 95% CI = 1.96-22.0), conferred poor prognosis. Background: Osteoarthritis (OA) is a major cause of human disability. Despite receiving treatment, patients with the middle and late stage of OA have poor survival outcomes. Therefore, within the framework of predictive, preventive, and personalized medicine (PPPM/3PM), early personalized diagnosis of OA is particularly prominent. PPPM aims to accurately identify disease by integrating multiple omic techniques; however, the efficiency of currently available methods and biomarkers in predicting and diagnosing OA should be improved. Disulfidptosis, a novel programmed cell death mechanism and appeared in particular metabolic status, plays a mysterious characteristic in the occurrence and development of OA, which warrants further investigation. Methods: In this study, we integrated three public datasets from the Gene Expression Omnibus (GEO) database, including 26 OA samples and 20 normal samples. Via a series of bioinformatic analysis and machine learning, we identified the diagnostic biomarkers and several subtypes of OA. Moreover, the expression of these biomarkers were verified in our in-house cohort and the single cell dataset. Results: Three significant regulators of disulfidptosis (NCKAP1, OXSM, and SLC3A2) were identified through differential expression analysis and machine learning. And a nomogram constructed based on these three regulators exhibited ideal efficiency in predicting early- and late-stage OA. Furthermore, based on the expression of three regulators, we identified two disulfidptosis-related subtypes of OA with different infiltration of immune cells and personalized expression level of immune checkpoints. Notably, the expression of the three regulators was demonstrated in a single-cell RNA profile and verified in the synovial tissue in our in-house cohort including 6 OA patients and 6 normal people. Finally, an efficient disulfidptosis-mediated diagnostic model was constructed for OA, with the AUC value of 97.6923% in the training set and 93.3333% and 100% in two validation sets. Background: The activation of G protein-coupled receptors (GPCR) signaling by external stimuli has been implicated in inducing cardiac stress and stress responses. GPR22 is an orphan GPCR expressed in brains and hearts, while its expression level is associated with cardiovascular damage in diabetes. Previous studies have suggested a protective role of GPR22 in mechanical cardiac stress, as loss of its expression increases susceptibility to heart failure post-ventricular pressure overload. However, the involvement and underlying signaling of GPR22 in cardiac stress response to ischemic stress remains unexplored. Methods: In this study, we used cultured cells and a transgenic mouse model with cardiomyocyte-specific GPR22 overexpression to investigate the impact of ischemic stress on GPR22 expression and to elucidate its role in myocardial ischemic injury. Acute myocardial infarction (AMI) was induced by left coronary artery ligation in eight-week-old male GPR22 transgenic mice, followed by histopathological and biochemical examination four weeks post-AMI induction. Results: GPR22 expression in H9C2 and RL-14 cells, two cardiomyocyte cell lines, was decreased by cobalt chloride (CoCl2) treatment. Similarly, reduced expression of myocardial GPR22 was observed in mice with AMI. Histopathological examinations revealed a protective effect of GPR22 overexpression in attenuating myocardial infarction in mice with AMI. Furthermore, myocardial levels of Bcl-2 and activation of PI3K-Akt signaling were downregulated by ischemic stress and upregulated by GPR22 overexpression. Conversely, the expression levels of caspase-3 and phosphorylated ERK1/2 in the infarcted myocardium were downregulated with GPR22 overexpression. Conclusion: Myocardial ischemic stress downregulates cardiac expression of GPR22, whereas overexpression of GPR22 in cardiomyocytes upregulates Akt signaling, downregulates ERK activation, and mitigates ischemia-induced myocardial injury. Purpose: To explore care requirements of older adults with urinary incontinence (UI) and contributing factors. Method: This cross-sectional study used the Older Adults Urinary Incontinence Care Needs Inventory to survey participants with UI in three large-scale tertiary hospitals located in Guangzhou City, China, from January 2023 to November 2023. Statistical analyses, including analysis of variance, t tests, correlation analyses, and linear regression models, were conducted to assess factors influencing participants' care needs. Results: A total of 530 older adults with UI participated in the survey and mean standardized score for overall care needs was 78.65 (SD = 5.01), with mean scores for each dimension ranging from 70.88 (SD = 10.55) for social participation needs to 82.45 (SD = 7.11) for health education needs. Factors that were found to influence incontinence care needs in older adults included age, literacy level, number of leaks, and type of disease (F = 37.07, adjusted R2 = 0.290, p < 0.001). Objective: This study explores subjective and objective driving style similarity to identify how similarity can be used to develop driver-compatible vehicle automation. Background: Similarity in the ways that interaction partners perform tasks can be measured subjectively, through questionnaires, or objectively by characterizing each agent's actions. Although subjective measures have advantages in prediction, objective measures are more useful when operationalizing interventions based on these measures. Showing how objective and subjective similarity are related is therefore prudent for aligning future machine performance with human preferences. Methods: A driving simulator study was conducted with stop-and-go scenarios. Participants experienced conservative, moderate, and aggressive automated driving styles and rated the similarity between their own driving style and that of the automation. Objective similarity between the manual and automated driving speed profiles was calculated using three distance measures: dynamic time warping, Euclidean distance, and time alignment measure. Linear mixed effects models were used to examine how different components of the stopping profile and the three objective similarity measures predicted subjective similarity. Results: Objective similarity using Euclidean distance best predicted subjective similarity. However, this was only observed for participants' approach to the intersection and not their departure. Conclusion: Developing driving styles that drivers perceive to be similar to their own is an important step toward driver-compatible automation. In this paper, the contact behavior of silicone rubbers under spherical and conical indenters is investigated by simulation and experiment. The accuracy and applicability of seven hyperelastic constitutive models are discussed for the contact behavior of silicone rubbers. The stress-strain curves obtained by uniaxial tension/compression experiments are used to fit parameters of hyperelastic constitutive models. Indentation experiments are carried out to compare with simulation results. The fitting accuracy of constitutive models under different strains is analyzed. It is found that the constitutive parameters fitted by the uniaxial compression experiments are the best for simulating the contact behavior of silicone rubbers. [ABSTRACT FROM AUTHOR]

Introduction: Chronic physical disease (CPD) makes life filled with many negative events in adolescents, but not all adolescents experiencing negative life events proceed to develop emotional distress, only those with low emotional distress tolerance (EDT). A valid and reliable scale to measure EDT in CPD adolescents is important for caring for their emotional distress. Therefore, the purpose of this study is to translate the 15‐item English version Distress Tolerance Scale (DTS) into a Chinese version and then validate the scale for measuring EDT of adolescents with CPD. Methods: The 15‐item English version DTS was translated into a Chinese version using the translation guidelines for cross‐cultural research. Two cohorts of adolescents with CPD were recruited from four hospitals in southern Taiwan, with the first cohort including 124 adolescents with CPD employed to conduct exploratory factor analysis, corrected item‐total correlation and reliability testing, while the second cohort, consisting of 238 adolescents with CPD, was utilized to examine confirmatory factor analysis and concurrent validity. Results: The two‐factor nine‐item Chinese version DTS for Adolescents with CPD (C‐DTS‐A) was developed. Lower scores of the C‐DTS‐A were significantly associated with higher diabetes distress, poorer self‐management, and worse glycaemic control; their correlation coefficients sequentially were −.40,.17 and −.23. Cronbach's α and the test–retest reliability of the two‐factor C‐DTS‐A ranged from.81 to.87 and from.79 to.89, respectively. Background: Patients with type 2 diabetes are at high risk for developing multiple chronic complications. However, there is a lack of studies of the cumulative number of diabetic complications in China. Methods: A retrospective cohort study was performed from 2009 to 2021. Type 2 diabetes patients who were first diagnosed after the age of 35 years between January 1, 2009, and December 31, 2017, were included. Five states were defined according to the number of chronic complications: no (S0), one (S1), two (S2), three (S3), and four or more complications (S4). A multi-state Markov model was constructed to estimate transition probability, transition intensity, mean sojourn time, and the possible factors for each state. Results: The study included 32653 type 2 diabetes patients (mean age, 59.59 years; 15929 (48.8%) male), and mean follow-up time of 7.75 years. In all, 4375 transitions were observed. The 12-year transition probability of from state S0 to S1 was the lowest at 16.4%, while that from S2 to S3 was the highest, at 45.6%. Higher fasting blood glucose, lower high-density lipoprotein cholesterol, higher total cholesterol, and an unhealthy diet were associated with higher risk of progression from S0 to S1. Being female, less than 60 years old, weekly physical activity, and vegetarian diet decreased this risk. Being female and less than 60 years old reduced the likelihood of transition from S1 to S2, whereas lower high-density lipoprotein cholesterol increased this likelihood. Conclusions: Following the occurrence of two complications in type 2 diabetes patients, the risk for accumulating a third complication within a short time is significantly increased. It is important to take advantage of the stable window period when patients have fewer than two complications, strengthen the monitoring of blood glucose and blood lipids, and encourage patients to maintain good living habits to prevent further deterioration. Recently, more and more evidence shows that lipid metabolism disorder has been observed in tumor, which impacts tumor cell proliferation, survival, invasion, metastasis, and response to the tumor microenvironment (TME) and tumor treatment. However, hitherto there has not been sufficient research to demonstrate the role of lipid metabolism in pancreatic cancer. This study contrives to get an insight into the relationship between the characteristics of lipid metabolism and pancreatic cancer. We collected samples of patients with pancreatic cancer from the Gene Expression Omnibus (GEO), the Therapeutically Applicable Research to Generate Effective Treatments (TARGET), and the International Cancer Genome Consortium (ICGC) databases. Firstly, we implemented univariate regression analysis to get prognosis-related lipid metabolism genes screened and a construction of protein–protein interaction (PPI) network ensued. Then, contingent on our screening results, we explored the molecular subtypes mediated by lipid metabolism-related genes and the correlated TME cell infiltration. Additionally, we studied the disparately expressed genes among disparate lipid metabolism subtypes and established a scoring model of lipid metabolism-related characteristics using the least absolute shrinkage and selection operator (LASSO) regression analysis. At last, we explored the relationship between the scoring model and disease prognosis, tumor stage, tumor microenvironment, and immunotherapy. Two subtypes, C1 and C2, were identified, and lipid metabolism-related genes were studied. The result indicated that the patients with subtype C2 have a significantly lower survival rate than that of the patients with subtype C1, and we found difference in abundance of different immune-infiltrating cells. The gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses revealed the association of these differentially expressed genes with functions and pathways related to lipid metabolism. Finally, we established a scoring model of lipid metabolism-related characteristics based on the disparately expressed genes. The results show that our scoring model have a substantial effect on forecasting the prognosis of patients with pancreatic cancer. Using the model, the relationship between disease prognosis, molecular subtypes, TME cell infiltration characteristics, and immunotherapy in pancreatic cancer patients could be explored. The Internet of Vehicles (IoV) autonomous driving technology based on deep learning has achieved great success. However, under the tunnel environment, the computer vision‐based IoV may fail due to low illumination. In order to handle this issue, this paper deploys an image enhancement module at the terminal of the IoV to alleviate the low illumination influence. The enhanced images can be submitted through IoT to the cloud server for further processing. The core algorithm of image enhancement is implemented by a dynamic graph embedded transformer network based on federated learning which can fully utilize the data resources of multiple devices in IoV and improve the generalization. Extensive comparative experiments are conducted on the publicly available dataset and the self‐built dataset which is collected under the tunnel environment. Compared with other deep models, all results confirm that the proposed graph embedded Transformer model can effectively enhance the detail information of the low‐light image, which can greatly improve the following tasks in IoV. Objectives: This study is to investigate the effect of group music therapy on depression and anxiety in acute psychiatric in-patients. Methods: Two groups of experiments were pretest-posttest designed. The research subjects were 122 inpatients in the acute psychiatric ward of a medical center. The experimental group received 50 minutes of a group music therapy intervention every Wednesday and Friday for a total of 8 times over 4 weeks. The control group received routine care. Both groups were administered the Beck Depression Inventory (BDI-II) and Beck Anxiety Inventory (BAI) pretest and posttest. Results: For the experimental group, the music therapy intervention showed a significant reduction in both depression (p <.001) and anxiety (p <.001). Implications for practice: The results of this study provide an empirical basis for the use of group music therapy as an intervention to reduce depression and anxiety in in-patients with mental illness. It is suggested that mental health care professionals may find music therapy helpful in routine group therapy programs to reduce depression and anxiety symptoms in acute psychiatric in-patients. To avoid carbonate precipitation for CO2 electrolysis, developing CO2 conversion in an acid electrolyte is viewed as an ultimately challenging technology. The treatment of brain tumors is greatly limited due to the presence of the blood tumor barrier (BTB). Our previous research has shown that the essential oil of chuanxiong promoted temozolomide entry into glioma cells and enhanced temozolomide-induced anticancer efficiency in vivo. Phthalides account for the largest proportion of essential oil and, therefore, this study was conducted to investigate the effects of six phthalides on the transmembrane transport of compatible compounds across the BTB and the mechanism. The results showed that different concentrations of senkyunolide H (SH), senkyunolide A (SA), ligustilide (LIG), butylidenephthalide (BP), n-butylphthalide (NBP) and neocnidilide (NOL) significantly increase the permeability from A (apical side) to B (basolateral side) (Papp(AB)) of fluorescein isothiocyanate 4000 (FITC4000), while NBP, NOL and SA could also significantly increase Papp(B→A). Western blot analysis indicated that these phthalides down-regulated expressions of claudin-5, occludin and ZO-1 to increase the permeability of BTB. LIG, SA, SH, NBP and NOL significantly increased Papp(AB) and the efflux ratio of the P-gp substrate rhodamine 123 attribute to down-regulate the expressions of P-gp. The expression of caveolin-1 was also down-regulated by the six phthalides. Objective To explore the mediating role of self-esteem in the relationship between childhood adverse experiences (ACEs) and depressive symptoms of junior high school students, and the regulatory effect of social support. Methods A total of 1 308 students from 3 junior middle schools in Xiangxi were selected by stratified random cluster sampling from February to April 2023. A questionnaire survey was conducted with the use of Childhood adverse experience questionnaire, Adolescent social support scale, PHQ-9 Depression symptom screening scale, and Rosenberg self-esteem scale. The data were analyzed by correlation analysis, intermediary analysis, and moderated intermediary model test. Results (1) ACEs positively predicted depressive symptoms of junior high school students (β=0.801, t=7.246, P<0.05), while self-esteem and social support negatively predicted depressive symptoms of junior high school students (β=-0.406, t=-14.276, P < 0.05; β =-0.027, t=-2.781, P < 0.05). (2) Self-esteem played a mediating role between ACEs and depressive symptoms (effect=0.468, P < 0.05), and the mediating effect accounted for 31.33% of the total effect. (3) Social support directly regulated the relationship between ACEs and depressive symptoms (β=-0.017, t=-3.043, P<0.01), and social support played a regulatory role in the latter half of the mediating pathway of "ACEs self-esteem and depressive symptoms" (β=-0.004, t=-2.887, P<0.01). Conclusion There is a mediating effect between ACEs and depressive symptoms of junior high school students, and self-esteem is the mediating variable of the relationship between them. Social support buffers the influence of ACEs on the risk of depressive symptoms of junior high school students and strengthens the relationship between self-esteem and depressive symptoms. Therefore, giving more care and social support to junior high school students with ACEs and improving their self-esteem will help prevent and reduce the occurrence of depressive symptoms. This study proposes a novel framework that couples the general likelihood uncertainty estimation (GLUE) method with a deterministic forecasting approach to conduct a new uncertainty analysis approach for assessing the energy production of operational wind turbines installed in the Jhongtun wind farm at Penghu (an island in the middle of Taiwan Strait). The 10-year measured data of wind speeds and energy output collected on these wind turbines is divided into two 5-year data sets for the present analysis framework of execution and validation to demonstrate the predictability of the GLUE method. The present study considers 15 scenario testing cases with various time periods, i.e., twelve months, the strong-wind (October-March) regime, the weak-wind (April-September) regime, and one year, for the framework to investigate the applicability of the GLUE method on long-term wind energy forecasting. In the execution framework, the 5-year measured data is used by the GLUE method to access the uncertainties involved in the deterministic approach (i.e., the shape and scale parameters of the Weibull wind speed distribution (WWSD), the performance curve, and the capacity factor) with two confidence intervals of 50% and 90%. The framework is then validated by the measured capacity factors in the last 5-year data and compared with the results of the uncertainty analysis approach by the Monte Carlo (MC) approach to discover the applicability of the new uncertainty analysis approach. From the simulated results, it is found that the proposed uncertainty analysis approach provides predictions of confidence intervals that match the measured data better than the MC-based uncertainty analysis approach. Specifically, the proposed approach can match the measured capacity factors in all the simulated scenarios. Conversely, the MC-based approach is found to create narrow confidence intervals that cannot completely capture the measured capacity factors, particularly for the strong-wind, weak-wind, and one-year scenarios. Metabolic dysfunction-associated fatty liver disease (MAFLD) has reached epidemic proportions worldwide and is the most frequent cause of chronic liver disease in developed countries. Within the spectrum of liver disease in MAFLD, steatohepatitis is a progressive form of liver disease and hepatocyte ballooning (HB) is a cardinal pathological feature of steatohepatitis. The accurate and reproducible diagnosis of HB is therefore critical for the early detection and treatment of steatohepatitis. Currently, a diagnosis of HB relies on pathological examination by expert pathologists, which may be a time-consuming and subjective process. Hence, there has been interest in developing automated methods for diagnosing HB. This narrative review briefly discusses the development of artificial intelligence (AI) technology for diagnosing fatty liver disease pathology over the last 30 years and provides an overview of the current research status of AI algorithms for the identification of HB, including published articles on traditional machine learning algorithms and deep learning algorithms. This narrative review also provides a summary of object detection algorithms, including the principles, historical developments, and applications in the medical image analysis. The potential benefits of object detection algorithms for HB diagnosis (specifically those combined with a transformer architecture) are discussed, along with the future directions of object detection algorithms in HB diagnosis and the potential applications of generative AI on transformer architecture in this field. In conclusion, object detection algorithms have huge potential for the identification of HB and could make the diagnosis of MAFLD more accurate and efficient in the near future. Transitional metal oxides (TMOs) with ultra‐high specific surface areas (SSAs), large pore volume, and tailored exposed facets appeal to significant interests in heterogeneous catalysis. Nevertheless, synthesizing the metal oxides with all the above features is challenging. Herein, the so‐called seeds/NaCl‐mediated growth method is successfully developed based on a bottom‐up route. First, the (Brunauer‐Emmett‐Teller) BET SSAs of TMOs prepared with this method are significantly higher, where the BET SSAs of CeO2, SnO2, Nb2O5, Fe3O4, Mn3O4, Mg(OH)2, and ZrO2 reached 187, 275, 518, 212, 147, 186, and 332 m2 g−1, respectively. Second, these TMOs exhibit unique mesoporous structures, generated mainly by the aggregation of rod‐like or other aspherical primary nanoparticles. More importantly, no environmental‐unfriendly organic surfactants or expensive metal alkoxides are involved in this method. Introduction: Osteoarthritis (OA) is a common degenerative disease of joints, which can appear in almost any joint of the body. Therefore, the widespread occurrence of this disease has a huge impact on the lives of patients around the world. As an important part of metabolism, lipid metabolism is closely related to the occurrence and development of osteoarthritis. Method: We screened UGCG and KLF4 based on weighted co-expression network analysis (WGCNA) and SVM-REF analysis. The data from Gene Expression Omnibus (GEO) and single-cell data verified the expression of these two genes. We analyzed KLF4-related genes and established a diagnosis model of OA related to lipid metabolism through the least absolute shrinkage and selection operator (LASSO) analysis. RT-PCR was used to verify the expression of KLF4 in osteoarthritis. Results: Ten important lipid metabolism related genes (LMRGs) in OA were obtained. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that they are involve in the formation of immune microenvironment in osteoarthritis. CIBERSORT analysis revealed that there were significant differences in the immune microenvironment between osteoarthritis patients and normal controls. RT-PCR results showed that the expression of KLF4 in OA samples was lower than that in normal samples. The diagnostic model can be used to diagnose OA patients well. Conclusions: Overall, we demonstrated the potential relationship between the abnormal lipid metabolism and the pathological process of OA. Finally, we identified KLF4 as our significant LMRG and constructed a KLF4-related scoring model to accurately diagnose OA. Objective: To investigate the feasibility and efficacy of combining ultrasound-guided sharp needle technique with percutaneous transluminal angioplasty (PTA) for treating outflow stenosis or dysfunction in arteriovenous fistula (AVF) among hemodialysis patients. Methods: From October 2021 to March 2023, patients with occluded or malfunctional fistula veins not amenable to regularly angioplasty were retrospectively enrolled in the study. They underwent ultrasound-guided sharp needle intervention followed by PTA. Data on the location and length between the two veins, technical success, clinical outcomes, and complications were collected. Patency rates post-angioplasty were calculated through Kaplan-Meier analysis. Results: A total of 23 patients were included. The mean length of the reconstructed extraluminal segment was 3.18cm. The sharp needle opening was performed on the basilic vein (60.9%), brachial vein (26.1%), or upper arm cephalic vein (13%) to create outflow channels. Postoperatively, all cases presented with mild subcutaneous hematomas around the tunneling site and minor diffuse bleeding. The immediate patency rate for the internal fistulas was 100%, with 3-month, 6-month, and 12-month patency rates at 91.3%, 78.3%, and 43.5%, respectively. Conclusion: Sharp needle technology merged with PTA presents an effective and secure minimally invasive method for reconstructing the outflow tract, offering a new solution for recanalizing high-pressure or occluded fistulas. Chilli pepper is one of the most important and widely cultivated pepper types in China. It has high nutritional and economic value. Capsicum frutescens L. is one of the chilli pepper types in production. In October 2022, a new disease showing blight of the entire chilli pepper plant was observed in Nanchang City, Jiangxi Province. The fungal isolates consistently obtained from the diseased samples were identified as Diaporthe caryae C. M. Tian & Qin Yang based on morphological characteristics, pathogenicity test and phylogenetic analysis of four genes, including internal transcribed spacer region (rDNA‐ITS), translation elongation factor 1‐alpha gene (TEF1), beta‐tubulin gene (TUB2), and calmodulin (CAL). Although other Diaporthe species have been reported as pathogens of peppers previously, to our knowledge, this is the first report of D. caryae causing stem blight of pepper in the world. D. caryae was reported to be able to infect various fruit trees such as kiwifruit, peach, pear, and pecan, resulting in kiwifruit rot, peach constriction canker, pear shoot canker, and pecan dieback, respectively. While the worldwide distribution of geogenic arsenic (As)-affected groundwater is highly overlapped with the areas with abundant groundwater, utilization of As-contained groundwater is an inevitable compromise in those areas where surface water is not enough for irrigation. Since the occurrence of As in groundwater is often accompanied by high iron (Fe) contents, the facilitation of As and Fe precipitation without adding additional oxidizers and adsorbents is considered an environmental-friendly approach to removing As in groundwater. In the present study, the oxidation/filtration dual-process with sprinkling height of 25 cm and 120 kg filter media efficiently increased the dissolved oxygen (DO) concentration (0.36–1.52 mg/L) and oxidation–reduction potential (ORP) (24–63 mV), which facilitated the formation of Fe oxides and As co-precipitation. The correlation of As removal efficiencies with their respective flow rates indicated that a decrease in groundwater Fe and an increase of Fe in sands and gravels filters as the flow rate increased evidenced the rapid oxidation of Fe to form the Fe hydroxides. In a 40-hour continuous aeration/filtration operation, As and Fe concentrations in groundwater were reduced by 79.5% and 64.88% within 40 hrs, respectively. Background: Family members often undertake caregiving responsibilities over long periods of time, which could lead to caregiving burden. A theory‐based and culturally sensitive learned resourcefulness program may help family caregivers mitigate stress by learning and using self‐help strategies. Objectives: This study's aim is to use rigorous methods to investigate the effects of a learned resourcefulness program called SOURCE (Chen et al., Geriatric Nursing, 2021, 45, 1129) to reduce caregiver burden and improve quality of life (QOL) for family caregivers. SOURCE is an acronym for the six self‐help behaviors at the core of this theory‐based learned resourcefulness program, developed by the Principal Investigator (PI) in collaboration with other researchers (Chen et al., Geriatric Nursing, 2021, 45, 1129). These behaviors are: seeking problem‐solving strategies, organizing daily actions, using self‐regulation, reframing positive situations, changing negative self‐thinking, and exploring new thinking and skills. Methods: We used a quasi‐experimental design with repeated measures for this study. The potential subjects were family caregivers whose family member was receiving home care. PI recruited 94 family caregivers (60 years old or older) who were caring for disabled and/or ill family members who also were 60 years old or older and living at home at the time of the study. The PI recruited these caregivers through the home care services department of a hospital and two community healthcare centers in Taiwan. The experimental group (n = 46) received the four‐week in‐person SOURCE while their family member continued to receive home care services, whereas the control group (n = 48) did not receive the SOURCE program while their family member received home care services. We collected data from the participants using the Caregiver Burden Scale, EuroQol‐5 Dimension, and Rosenbaum's Self‐Control Scale at baseline (T0, Week 1), after the intervention (T1, Week 5), and at follow‐up (T2, Week 9). We analyzed the data using paired‐sample t‐tests and used the generalized estimating equation method to compare paired data between the baseline and follow‐up. Results: Of the 94 recruited family caregivers, 90.4% (n = 85) completed the study. We found no significant differences between the experimental and control groups in terms of demographic, caregiver burden, and QOL variables at baseline. Compared to the control group, the experimental group had significantly improved caregiver burden scores (indicating a reduction in caregiver burden) at the end of the intervention and at follow‐up. The experimental group had significantly improved QOL scores at follow‐up only. The experimental group also showed high levels of adherence to and satisfaction with the intervention. The paired‐sample t‐test results show significantly improved learned resourcefulness between T0 and T1 and between T0 and T2 (t = −5.84, p < 0.001; t = −6.77, p < 0.001) for the experimental group. Conclusion: The SOURCE program helped family caregivers develop and use self‐help behaviors during their daily caregiving tasks. The family caregivers reported satisfaction with the program as well as reduced caregiver burden and improved QOL after the program ended. Implications for practice: The SOURCE program can be implemented to improve family caregivers' caregiver burden and QOL. Community health nurses can improve self‐help abilities of family caregivers by helping them to acquire abilities and skills that allow them to effectively handle stress and reduce their caregiver burden. Background: Corneal fluorescein staining is a key biomarker in evaluating dry eye disease. However, subjective scales of corneal fluorescein staining are lacking in consistency and increase the difficulties of an accurate diagnosis for clinicians. This study aimed to propose an automatic machine learning-based method for corneal fluorescein staining evaluation by utilizing prior information about the spatial connection and distribution of the staining region. Methods: We proposed an end-to-end automatic machine learning-based classification model that consists of staining region identification, feature signature construction, and machine learning-based classification, which fully scrutinizes the multiscale topological features together with conventional texture and morphological features. The proposed model was evaluated using retrospective data from Beijing Tongren Hospital. Two masked ophthalmologists scored images independently using the Sjögren's International Collaborative Clinical Alliance Ocular Staining Score scale. Results: A total of 382 images were enrolled in the study. A signature with six topological features, two textural features, and two morphological features was constructed after feature extraction and selection. Support vector machines showed the best classification performance (accuracy: 82.67%, area under the curve: 96.59%) with the designed signature. Meanwhile, topological features contributed more to the classification, compared with other features. According to the distribution and correlation with features and scores, topological features performed better than others. Conclusions: An automatic machine learning-based method was advanced for corneal fluorescein staining evaluation. The topological features in presenting the spatial connectivity and distribution of staining regions are essential for an efficient corneal fluorescein staining evaluation. Elimination of cancer stem cells (CSCs) and reinvigoration of antitumor immunity remain unmet challenges for cancer therapy. Tumor-associated macrophages (TAMs) constitute the prominant population of immune cells in tumor tissues, contributing to the formation of CSC niches and a suppressive immune microenvironment. Here, we report that high expression of inhibitor of differentiation 1 (ID1) in TAMs correlates with poor outcome in patients with colorectal cancer (CRC). ID1 expressing macrophages maintain cancer stemness and impede CD8+ T cell infiltration. Mechanistically, ID1 interacts with STAT1 to induce its cytoplasmic distribution and inhibits STAT1-mediated SerpinB2 and CCL4 transcription, two secretory factors responsible for cancer stemness inhibition and CD8+ T cell recruitment. Reducing ID1 expression ameliorates CRC progression and enhances tumor sensitivity to immunotherapy and chemotherapy. Collectively, our study highlights the pivotal role of ID1 in controlling the protumor phenotype of TAMs and paves the way for therapeutic targeting of ID1 in CRC.
