Background: Some insects can degrade both natural and synthetic plastic polymers, their host and gut microbes play crucial roles in this process. However, there is still a scientific gap in understanding how the insect adapted to the polystyrene (PS) diet from natural feed. In this study, we analyzed diet consumption, gut microbiota responses, and metabolic pathways of Tenebrio molitor larvae exposed to PS and corn straw (CS). Results: T. molitor larvae were incubated under controlled conditions (25 ± 1 °C, 75 ± 5% humidity) for 30 days by using PS foam with weight-, number-, and size-average molecular weight (Mw, Mn, and Mz) of 120.0, 73.2, and 150.7 kDa as a diet, respectively. The larvae exhibited lower PS consumption (32.5%) than CS (52.0%), and these diets had no adverse effects on their survival. The gut microbiota structures, metabolic pathways, and enzymatic profiles of PS- and CS-fed larvae showed similar responses. The gut microbiota of larvae analysis indicated Serratia sp., Staphylococcus sp., and Rhodococcus sp. were associated with both PS and CS diets. Metatranscriptomic analysis revealed that xenobiotics, aromatic compounds, and fatty acid degradation pathways were enriched in PS- and CS-fed groups; laccase-like multicopper oxidases, cytochrome P450, monooxygenase, superoxidase, and dehydrogenase were involved in lignin and PS degradation. Furthermore, the upregulated gene lac640 in both PS- and CS-fed groups was overexpressed in E. coli and exhibited PS and lignin degradation ability. Conclusions: The high similarity of gut microbiomes adapted to biodegradation of PS and CS indicated the plastics-degrading ability of the T. molitor larvae originated through an ancient mechanism that degrades the natural lignocellulose. Phytoplasmas are a threat to cassava production, but have not been found from cassava in Japan. Here, we present the discovery of phytoplasma in asymptomatic plants propagated in Japan. A 16S rRNA gene sequence analysis showed that the phytoplasma was related to ‘Candidatus Phytoplasma pruni’. Analyses based on 16S rRNA, secY, and ribosomal protein gene sequences revealed that the phytoplasma was closely related to phytoplasmas found in cassava in Latin America. Infected plants remained asymptomatic during cultivation for more than 11 months. This is the first molecular evidence of phytoplasma infection in cassava in Japan. [ABSTRACT FROM AUTHOR]

Deregulated expression of an Arabidopsis H/Ca antiporter (sCAX1) in agricultural crops increases total calcium (Ca) but may result in yield losses due to Ca deficiency-like symptoms. Here we demonstrate that co-expression of a maize calreticulin (CRT, a Ca binding protein located at endoplasmic reticulum) in sCAX1-expressing tobacco and tomato plants mitigated these adverse effects while maintaining enhanced Ca content. Co-expression of CRT and sCAX1 could alleviate the hypersensitivity to ion imbalance in tobacco plants. Furthermore, blossom-end rot (BER) in tomato may be linked to changes in CAX activity and enhanced CRT expression mitigated BER in sCAX1 expressing lines. These findings suggest that co-expressing Ca transporters and binding proteins at different intracellular compartments can alter the content and distribution of Ca within the plant matrix. Bottle gourd ( Lagenaria siceraria Standl.) has been commonly used as a source of rootstock for watermelon. To improve its performance as a rootstock without adverse effects on the scion, the bottle gourd was genetically engineered using a modified Arabidopsis Ca2+/H+ exchanger sCAX2B. This transporter provides enhanced Ca2+ substrate specificity and decreased Mn2+ transport capability. Our previous work demonstrated that s CAX2B bottle gourds were more robust and nutrient dense than controls. Here, several cucurbit crops were test-grafted onto the transgenic bottle gourd to determine its effect on the scions. The grafted watermelons and melons onto the transgenic rootstocks appeared to show more robust growth than the controls 35 days after greenhouse transplanting. Watermelon fruits with the watermelon/transgenic bottle gourd (scion/rootstock) combination demonstrated higher osmotic pressure and more soluble solids than controls. These results suggest that s CAX2B expression in the bottle gourd rootstock facilitates improved watermelon quality through the translocation of nutrients and/or water toward enhancing the biomass of scion. Cation levels within the cytosol are coordinated by a network of transporters. Here, we examine the functional roles of calcium exchanger 1 (CAX1), a vacuolar H+/Ca2+ transporter, and the closely related transporter CAX3. We demonstrate that like CAX1, CAX3 is also localized to the tonoplast. We show that CAX1 is predominately expressed in leaves, while CAX3 is highly expressed in roots. Previously, using a yeast assay, we demonstrated that an N-terminal truncation of CAX1 functions as an H+/Ca2+ transporter. Here, we use the same yeast assay to show that full-length CAX1 and full-length CAX3 can partially, but not fully, suppress the Ca2+ hypersensitive yeast phenotype and coexpression of full-length CAX1 and CAX3 conferred phenotypes not produced when either transporter was expressed individually. In planta, CAX3 null alleles were modestly sensitive to exogenous Ca2+ and also displayed a 22% reduction in vacuolar H+-ATPase activity, caxl/cax3 double mutants displayed a severe reduction in growth, including leaf tip and flower necrosis and pronounced sensitivity to exogenous Ca2+ and other ions. These growth defects were partially suppressed by addition of exogenous Mg2+. The double mutant displayed a 42% decrease in vacuolar H+/Ca2+ transport, and a 47% decrease in H+-ATPase activity. While the ionome of cax1 and cax3 lines were modestly perturbed, the cax1/cax3 lines displayed increased PO3-4, Mn2+, and Zn2+ and decreased Ca2+ and Mg2+ in shoot tissue. Abstract: The Arabidopsis Ca2+/H+ antiporters cation exchanger (CAX) 1 and 2 utilise an electrochemical gradient to transport Ca2+ into the vacuole to help mediate Ca2+ homeostasis. Previous whole plant studies indicate that activity of Ca2+/H+ antiporters is regulated by pH. However, the pH regulation of individual Ca2+/H+ antiporters has not been examined. To determine whether CAX1 and CAX2 activity is affected by pH, Ca2+/H+ antiport activity was measured in vacuolar membrane vesicles isolated from yeast heterologously expressing either transporter. Ca2+ transport by CAX1 and CAX2 was regulated by cytosolic pH and each transporter had a distinct cytosolic pH profile. Screening of CAX1/CAX2 chimeras identified an amino acid domain within CAX2 that altered the pH-dependent Ca2+ transport profile so that it was almost identical to the pH profile of CAX1. Results from mutagenesis of a specific His residue within this domain suggests a role for this residue in pH regulation. [Copyright &y& Elsevier]

The Ca2+-dependent protein phosphatase calcineurin is an important regulator of ion transporters from many organisms, including theSaccharomyces cerevisiaevacuolar Ca2+/H+ exchanger Vcx1p. In yeast and plants, cation/H+ exchangers are important in shaping cytosolic Ca2+ levels involved in signal transduction and providing tolerance to potentially toxic concentrations of cations such as Ca2+, Mn2+ and Cd2+. Previous genetic evidence suggested Vcx1p is negatively regulated by calcineurin. By utilizing direct transport measurements into vacuolar membrane vesicles, we demonstrate that Vcx1p is a low-affinity Ca2+ transporter and may also function in Cd2+ transport, but cannot transport Mn2+. Furthermore, direct Ca2+ transport by Vcx1p is calcineurin sensitive. Using a yeast growth assay, a mutant allele ofVCX1(VCX1-S204A/L208P), termedVCX1-M1, was previously found to confer strong Mn2+ tolerance. Here we demonstrate that this Mn2+ tolerance is independent of the Ca2+/Mn2+-ATPase Pmr1p and results from Mn2+-specific vacuolar transport activity of Vcx1-M1p. This Mn2+ transport by Vcx1-M1p is calcineurin dependent, although the localization of Vcx1-M1p to the vacuole appears to be calcineurin independent. Additionally, we demonstrate that mutation of L208P alone is enough to confer calcineurin-dependent Mn2+ tolerance. Ca2+ levels in plants are controlled in part by H+/Ca2+ exchangers. Structure/function analysis of the Arabidopsis H+/cation exchanger, CAX1, revealed that a nine amino acid region (87–95) is involved in CAX1-mediated Ca2+ specificity. CAX3 is 77% identical (93% similar) to CAX1, and when expressed in yeast, localizes to the vacuole but does not suppress yeast mutants defective in vacuolar Ca2+ transport. Transgenic tobacco plants expressing CAX3 containing the 9 amino acid Ca2+ domain (Cad) from CAX1 (CAX3-9) displayed altered stress sensitivities similar to CAX1-expressing plants, whereas CAX3-9-expressing plants did not have any altered stress sensitivities. A single leucine-to-isoleucine change at position 87 (CAX3-I) within the Cad of CAX3 allows this protein to weakly transport Ca2+ in yeast (less than 10% of CAX1). Site-directed mutagenesis of the leucine in the CAX3 Cad demonstrated that no amino acid change tested could confer more activity than CAX3-I. Transport studies in yeast demonstrated that the first three amino acids of the CAX1 Cad could confer twice the Ca2+ transport capability compared to CAX3-I. The entire Cad of CAX3 (87–95) inserted into CAX1 abolishes CAX1-mediated Ca2+ transport. However, single, double, or triple amino acid replacements within the native CAX1 Cad did not block CAX1 mediated Ca2+ transport. Together these findings suggest that other domains within CAX1 and CAX3 influence Ca2+ transport. Xanthomonas campestris pv. campestris induces two types of symptoms, namely, black rot and blight. Black rot symptoms are V-shaped lesions and black veins on the leaf, and blight symptoms are sudden collapse of interveinal tissues following the lack of veinal necrosis at early stages of infection. These two symptoms can occur simultaneously. However, the tendency to induce either symptom type is strain-dependent. Six strains were evaluated for their rate and pattern of spread in misted seedbeds by using strain-specific monoclonal antibodies and miniplate enrichment/ELISA. Data on pathogen incidence was defined as the presence of the pathogen in or on plants rather than visual symptoms. The results indicated that blight-inducing strains spread to more seedlings than black rot-inducing strains. The high incidences of blight-inducing strains in experimental plots were associated with non-randomness of spatial pattern of pathogen spread, indicating that high incidence is primarily due to the spread from adjacent plants by leaf contact and water splash. Most ELISA-positive seedlings were symptomless, indicating that the sensitivity of the system used in this study was adequate for detection of latent or epiphytic spread. [ABSTRACT FROM AUTHOR]

Betulinic acid (BA) is a major constituent of Zizyphus seeds that have been long used as therapeutic agents for sleep-related issues in Asia. BA is a pentacyclic triterpenoid. It also possesses various anti-cancer and anti-inflammatory effects. Current commercially available sleep aids typically use GABAergic regulation, for which many studies are being actively conducted. However, few studies have focused on acetylcholine receptors that regulate wakefulness. In this study, we utilized BA as an antagonist of α3β4 nicotinic acetylcholine receptors (α3β4 nAChRs) known to regulate rapid-eye-movement (REM) sleep and wakefulness. Effects of BA on α3β4 nAChRs were concentration-dependent, reversible, voltage-independent, and non-competitive. Site-directed mutagenesis and molecular-docking studies confirmed the binding of BA at the molecular level and showed that the α3 subunit L257 and the β4 subunit I263 residues affected BA binding. These data demonstrate that BA can bind to a binding site different from the site for the receptor's ligand, acetylcholine (ACh). This suggests that BA may be an effective antagonist that is unaffected by large amounts of ACh released during wakefulness and REM sleep. Sainfoin, Onobrychis viciifolia Scop., is an introduced forage legume grown on a limited basis in the Rocky Mountain region of the northern United States and southern Canada (Heath et al., 1973). The major attributes of sainfoin are its non-bloating characteristic as green forage and the high digestible nutrient content. It is drought tolerant, resistant to the alfalfa weevil, winter hardy, and produces good early spring growth tolerant to frost. If commercial Rhizobium inoculant is used, it requires no nitrogen fertilizer and has a low requirement for phosphorous. It has extremely good forage palatability. Therefore, it has the potential to provide a sustainable pasture system without required chemical inputs. The objective of our study was to determine if the combination of meadow bromegrass and our nematode-resistant sainfoin line would in crease the sustainability of sainfoin in the presence of the northern root-knot nematode (NRKN). Under environmentally controlled conditions, initial population density of five NRKN (Meloidogyne hapla Chitwood) eggs/cm3 soil resulted in 100% of sainfoin (Onobrychis viciifolia Scop.) roots with galls after70 days. One hundred percent of remaining sainfoin plants were galled in two other con trolled studies after 6 months, and 86% were galled in a field study after 2 years. Resistance to NRKN in an experimental sainfoin line was expressed in lower plant mortality, lower root galling, and lower egg production, compared to the susceptible cultivar Remont. The NRKN also appeared to be parasitic on roots of 'Regar' bromegrass (Bromus riparius Rehm.). Although root and shoot biomass were both reduced, there was no reproduction on roots and no bromegrass plants were killed. Under relatively high nematode soil populations, intercropping sainfoin with bromegrass provided no protection for sainfoin against the NRKN in either environmentally controlled or field plot studies.
