The assessment of skin changes in the context of possible child abuse is an important task in forensic medicine. This requires knowledge of pigmentation variants and pigmentation disorders such as congenital dermal melanocytosis, which includes Mongolian spots. Particularly in the case of atypical localization and dark pigmented skin, the differentiation from hematomas can be challenging. A case of two Nigerian siblings with extensive and atypically localized Mongolian spots is reported. The 1.5-year-old girl showed Mongolian spots on her back and the right side of her trunk. The 8-year-old boy showed Mongolian spots only on the back of his thighs. The authors are not aware of any case in which so called Mongolian spots were present exclusively on the back of the thighs and this case is all the more noteworthy as the back of the thigh is a common localization of blunt force trauma. This article compares the mitochondrial changes in the right ventricle (RV) and left ventricle (LV) in response to chronic pressure overload. The study used rat models with two stages of disease (compensated and decompensated) and conducted transcriptomic and proteomic analyses to identify differentially expressed mitochondrial genes and proteins. The results showed that mitochondrial dysfunction contributes to disease progression in both right and left heart failure. While there were ventricle-specific differences in mitochondrial gene and protein expression, the overall changes in gene expression were similar between the RV and LV. [Extracted from the article]

We investigate barrier-crossing processes corresponding to collective hydrogen-bond rearrangements in liquid water using Markov state modeling techniques. The analysis is based on trajectories from classical molecular dynamics simulations and accounts for the full dynamics of relative angular and separation coordinates of water clusters and requires no predefined hydrogen bond criterium. We account for the complete 12-dimensional conformational subspace of three water molecules and distinguish five well-separated slow dynamic processes with relaxation times in the picosecond range, followed by a quasi-continuum spectrum of faster modes. By analysis of the Markov eigenstates, these processes are shown to correspond to different collective interchanges of hydrogen-bond donors and acceptors. Using a projection onto hydrogen-bond states, we also analyze the switching of one hydrogen bond between two acceptor water molecules and derive the complete transition network. The most probable pathway corresponds to a direct switch without an intermediate, in agreement with previous studies. However, a considerable fraction of paths proceeds along alternative routes that involve different intermediate states with short-lived alternative hydrogen bonds or weakly bound states. Background: Obstructive sleep apnea (OSA) imposes vascular and metabolic risks through chronic intermittent hypoxia (CIH) and impairs skeletal muscle performance. As studies addressing limb muscles are rare, the reasons for the lower exercise capacity are unknown. We hypothesize that CIH-related morphological alterations in neuromuscular junctions (NMJ) and mitochondrial integrity might be the cause of functional disorders in skeletal muscles. Methods: Mice were kept under 6 weeks of CIH (alternating 7% and 21% O2 fractions every 30 s, 8 h/day, 5 days/week) compared to normoxia (NOX). Analyses included neuromuscular junctions (NMJ) postsynaptic morphology and integrity, fiber cross-sectional area (CSA) and composition (ATPase), mitochondrial ultrastructure (transmission-electron-microscopy), and relevant transcripts (RT-qPCR). Besides wildtype (WT), we included inducible nitric oxide synthase knockout mice (iNOS−/−) to evaluate whether iNOS is protective or risk-mediating. Results: In WT soleus muscle, CIH vs. NOX reduced NMJ size (− 37.0%, p < 0.001) and length (− 25.0%, p < 0.05) together with fiber CSA of type IIa fibers (− 14%, p < 0.05) and increased centronucleated fiber fraction (p < 0.001). Moreover, CIH vs. NOX increased the fraction of damaged mitochondria (1.8-fold, p < 0.001). Compared to WT, iNOS−/− similarly decreased NMJ area and length with NOX (− 55%, p < 0.001 and − 33%, p < 0.05, respectively) or with CIH (− 37%, p < 0.05 and − 29%, p < 0.05), however, prompted no fiber atrophy. Moreover, increased fractions of damaged (2.1-fold, p < 0.001) or swollen (> 6-fold, p < 0.001) mitochondria were observed with iNOS−/− vs. WT under NOX and similarly under CIH. Both, CIH- and iNOS−/− massively upregulated suppressor-of-cytokine-signaling-3 (SOCS3) > 10-fold without changes in IL6 mRNA expression. Furthermore, inflammatory markers like CD68 (macrophages) and IL1β were significantly lower in CIH vs. NOX. None of these morphological alterations with CIH- or iNOS−/− were detected in the gastrocnemius muscle. Notably, iNOS expression was undetectable in WT muscle, unlike the liver, where it was massively decreased with CIH. Conclusion: CIH leads to NMJ and mitochondrial damage associated with fiber atrophy/centronucleation selectively in slow-twitch muscle of WT. This effect is largely mimicked by iNOS−/− at NOX (except for atrophy). Both conditions involve massive SOCS3 upregulation likely through denervation without Il6 upregulation but accompanied by a decrease of macrophage density especially next to denervated endplates. In the absence of muscular iNOS expression in WT, this damage may arise from extramuscular, e.g., motoneuronal iNOS deficiency (through CIH or knockout) awaiting functional evaluation. An updated set of goals and objectives for the Mercury Gamma and Neutron Spectrometer (MGNS) are presented based on the most recent findings of the MESSENGER mission. The updated design of MGNS with the new CeBr3 crystal for detection of gamma-ray along with its benefits for the detection of 40K and K/Th ratio are discussed. MGNS will then be capable of measuring the elemental composition of shallow subsurface in order to empirically evaluate the fittest model on the origin of Mercury, as well as the presence of possible water ice deposits on the permanently shadowed polar craters on the planet. The recently discovered colliding-wind binary (CWB) Apep has been shown to emit luminously from radio to X-rays, with the emission driven by a binary composed of two Wolf–Rayet (WR) stars of one carbon-sequence (WC8) and one nitrogen-sequence (WN4–6b). Mid-infrared imaging revealed a giant spiral dust plume that is reminiscent of a pinwheel nebula but with additional features that suggest Apep is a unique system. We have conducted observations with the Australian Long Baseline Array to resolve Apep's radio emission on milliarcsecond scales, allowing us to relate the geometry of the wind-collision region to that of the spiral plume. The observed radio emission shows a bow-shaped structure, confirming its origin as a wind-collision region. The shape and orientation of this region is consistent with being originated by the two stars and with being likely dominated by the stronger wind of the WN4–6b star. This shape allowed us to provide a rough estimation of the opening angle of ∼150○ assuming ideal conditions. The orientation and opening angle of the emission also confirms it as the basis for the spiral dust plume. We also provide estimations for the two stars in the system to milliarcsecond precision. The observed radio emission, one order of magnitude brighter and more luminous than any other known non-thermal radio-emitting CWB, confirms it is produced by an extremely powerful wind collision. Such a powerful wind-collision region is consistent with Apep being a binary composed of two WR stars, so far the first unambiguously confirmed system of its kind. [ABSTRACT FROM AUTHOR]

We report the detection of a bright fast radio burst, FRB 191108, with Apertif on the Westerbork Synthesis Radio Telescope. The interferometer allows us to localize the FRB to a narrow 5 arcsec × 7 arcmin ellipse by employing both multibeam information within the Apertif phased-array feed beam pattern, and across different tied-array beams. The resulting sightline passes close to Local Group galaxy M33, with an impact parameter of only 18 kpc with respect to the core. It also traverses the much larger circumgalactic medium (CGM) of M31, the Andromeda Galaxy. We find that the shared plasma of the Local Group galaxies could contribute ∼10 per cent of its dispersion measure of 588 pc cm−3. FRB 191108 has a Faraday rotation measure (RM) of +474  |$\pm \, 3$|  rad m−2, which is too large to be explained by either the Milky Way or the intergalactic medium. Based on the more moderate RMs of other extragalactic sources that traverse the halo of M33, we conclude that the dense magnetized plasma resides in the host galaxy. The FRB exhibits frequency structure on two scales, one that is consistent with quenched Galactic scintillation and broader spectral structure with Δ ν ≈ 40 MHz. If the latter is due to scattering in the shared M33/M31 CGM, our results constrain the Local Group plasma environment. Context. Flares in radio-loud active galactic nuclei are thought to be associated with the injection of fresh plasma into the compact jet base. Such flares are usually strongest and appear earlier at shorter radio wavelengths. Hence, very long baseline interferometry (VLBI) at millimeter(mm)-wavelengths is the best-suited technique for studying the earliest structural changes of compact jets associated with emission flares. Aims. We study the morphological changes of the parsec-scale jet in the nearby (z = 0.049) γ-ray bright radio galaxy 3C 111 following a flare that developed into a major radio outburst in 2007. Methods. We analyse three successive observations of 3C 111 at 86 GHz with the Global mm-VLBI Array (GMVA) between 2007 and 2008 which yield a very high angular resolution of ∼45 μas. In addition, we make use of single-dish radio flux density measurements from the F-GAMMA and POLAMI programmes, archival single-dish and VLBI data. Results. We resolve the flare into multiple plasma components with a distinct morphology resembling a bend in an otherwise remarkably straight jet. The flare-associated features move with apparent velocities of ∼4.0c to ∼4.5c and can be traced also at lower frequencies in later epochs. Near the base of the jet, we find two bright features with high brightness temperatures up to ∼1011 K, which we associate with the core and a stationary feature in the jet. Conclusions. The flare led to multiple new jet components indicative of a dynamic modulation during the ejection. We interpret the bend-like feature as a direct result of the outburst which makes it possible to trace the transverse structure of the jet. In this scenario, the components follow different paths in the jet stream consistent with expectations for a spine-sheath structure, which is not seen during intermediate levels of activity. The possibility of coordinated multiwavelength observations during a future bright radio flare in 3C 111 makes this source an excellent target for probing the radio-γ-ray connection. Due to high migration inflows to Europe, forensic age assessment of living persons has clearly gained importance. If there is a legal justification for X-ray examinations without a medical indication, the Study Group on Forensic Age Diagnostics (AGFAD) recommends the combination of a physical examination with anamnesis, an X-ray examination of the hand and a dental examination with evaluation of an orthopantomogram for age assessments of adolescents and young adults. If the development of the hand skeleton has been completed, an additional CT examination of the clavicles is to be performed. To demonstrate the outcome of forensic age assessments according to AGFAD recommendations with regard to migrants of doubtful minority declaration, this study analyzes the expert reports carried out at the Institute of Legal Medicine, Münster, from 2009 to 2018. A total of 597 age assessments were performed during the study period. A total of 227 age assessments were issued on behalf of youth welfare offices in the legal area of social law, 282 in family law proceedings, 76 in criminal proceedings, and 12 age assessments in immigration law proceedings. In 211 out of 597 cases, the stated age was compatible with the findings of the age assessment. In the remaining 386 cases, the average difference between the stated age and the minimum age was 1.9 years. The average difference between stated age and most probable age was 5.1 years. Of the 521 age assessments carried out outside criminal proceedings, 197 unaccompanied minors with questionable age minority (37.8%) have reached the age of majority beyond doubt. A total of 388 unaccompanied minors (74.5%) have most probably reached the age of majority. Forensic age assessments with the AGFAD methodology make an important contribution to legal certainty, the welfare of the child, and the fair distribution of resources. [ABSTRACT FROM AUTHOR]

The number of older patients admitted to emergency departments (ED) increases continuously. The Identification of Seniors at Risk (ISAR) score is currently recommended to screen patients in German ED, but its appropriateness is being criticized. ISAR scores and clinical characteristics from 98 emergency admissions (EA), 80 from acute geriatrics (AG) and 89 from a geriatric rehabilitation (GR) unit were compared retrospectively. No significant differences were found between groups, being the ISAR score positive in 87.7% of EA, 94.9% of AG and 94.4% of GR cases. None of positively identified geriatric patients in the EA was transferred to the geriatric ward of competence. EA patients showed significantly higher number of functional impairments (p = 0.001) and higher BI score (p < 0.0001) compared to AG and GR groups. A higher ISAR score threshold and additional functional information might be needed to better select patients in need of prompt treatment by a geriatric team. [ABSTRACT FROM AUTHOR]

A self-consistent two-dimensional particle model coupled to the external circuit equations was developed in an asymmetrical configuration for the self-bias voltage calculation and the reactor design study. An intermediate modeling was performed in one and two symmetrical geometries. The one-dimensional model is used to optimize the computing time which is reduced by a factor of 10 by using some optimization techniques. It is also used to validate the charged particle and basic data choices. We have shown that the consideration of only two charged particle species (electron and H[sub 3][sup +] positive ion) is sufficient in the present hydrogen radio-frequency discharge modeling. Computational results (i.e., power density and self-bias voltage) are in good agreement with experimental results. A strong gradient of the plasma parameters (such as electric field, potential, charged particle densities and energies) was observed in the periphery of the driven electrode. Furthermore, the present two-dimensional asymmetric model shows that the interelectrode distance increase (from 1.7 up to 3.7 cm) can lead to reducing the plasma heterogeneity due to the geometrical electric field. © 2001 American Institute of Physics. Aims. In the framework of the multi-wavelength and very long baseline interferometry (VLBI) monitoring program TANAMI (Tracking Active Nuclei with Austral Milliarcsecond Interferometry), we study the evolution of the parsec-scale radio emission in radio galaxies in the southern hemisphere and their relationship to the γ-ray properties of the sources. Our study investigates systematically, for the first time, the relationship between the two energy regimes in radio galaxies. In this first paper, we focus on Fermi-LAT-detected sources. Methods. The TANAMI program monitors a large sample of radio-loud AGN at 8.4 GHz and 22.3 GHz with the Australian long baseline array (LBA) and associated telescopes in Antarctica, Chile, New Zealand and South Africa. We performed a kinematic analysis for five γ-ray detected radio galaxies using multi-epoch 8.4 GHz VLBI images, deriving limits on intrinsic jet parameters such as speed and viewing angle. We analyzed 103 months of Fermi-LAT data in order to study possible connections between the γ-ray properties and the pc-scale jets of Fermi-LAT-detected radio galaxies, both in terms of variability and average properties. We discuss the individual source results and draw preliminary conclusions on sample properties including published VLBI results from the MOJAVE (Monitoring Of Jets in Active galactic nuclei with VLBA Experiments) survey, with a total of fifteen sources. Results. We find that the first γ-ray detection of Pictor A might be associated with the passage of a new VLBI component through the radio core, which appears to be a defining feature of high-energy emitting Fanaroff-Riley type II radio galaxies. We detect subluminal parsec-scale jet motions in the peculiar AGN PKS 0521−36, and we confirm the presence of fast γ-ray variability in the source down to timescales of six hours, which is not accompanied by variations in the VLBI jet. We robustly confirm the presence of significant superluminal motion, up to βapp ∼ 3, in the jet of the TeV radio galaxy PKS 0625−35. Our VLBI results constrain the jet viewing angle to be θ <  53°, allowing for the possibility of a closely aligned jet. Finally, by analyzing the first pc-scale multi-epoch images of the prototypical compact symmetric object (CSO) PKS 1718−649, we place an upper limit on the separation speed between the two mini-lobes. This in turn allows us to derive a lower limit on the age of the source. Conclusions. We can draw some preliminary conclusions on the relationship between pc-scale jets and γ-ray emission in radio galaxies, based on Fermi-LAT-detected sources with available multi-epoch VLBI measurements. We find that the VLBI core flux density correlates with the γ-ray flux, as seen in blazars. On the other hand, the γ-ray luminosity does not show any dependence on the core brightness temperature and core dominance, which are two common indicators of jet Doppler boosting. This seems to indicate that γ-ray emission in radio galaxies is not driven by orientation-dependent effects, as in blazars, in accordance with the unified model of jetted AGN. [ABSTRACT FROM AUTHOR]

Context. Few active galactic nuclei (AGN) reveal double-sided jet systems. However, these systems are crucial to understand basic physical properties of extragalactic jets. Aims. We address the questions whether jets in AGN are symmetric in nature, how well they are collimated on small scales, and how they evolve with time. Methods. We monitored the sub-parsec scale morphology of NGC 1052 with the Very Long Baseline Array at 43 GHz from 2005 to 2009. Results. A detailed study of 29 epochs show a remarkable asymmetry between both jets. A kinematic analysis of the outflows reveals higher apparent velocities for the eastern (approaching) jet as compared to the western (receding) jet, i.e., βej = 0.529 ± 0.038 and βwj = 0.343 ± 0.037, respectively. Contradictory to previous studies, we find higher flux densities for the western jet as compared to the eastern. The distribution of brightness temperature and jet width features well-collimated jets up to 1 mas distance to the dynamic center and a nearly conical outflow further outward. By combining flux density ratios and velocities of the jet flows, we were unable to find a combination of intrinsic velocities and inclination angles of the jets that is consistent for all four years of observation; this contradicts findings for symmetrically evolving jets. Spectral index maps between quasi-simultaneous 22 GHz and 43 GHz observations support the existence of an optically thick absorber covering the innermost ≃1.6 mas around the 43 GHz central feature and an optically thin jet emission with a spectral index of ≤−1. Conclusions. Our results fit into a picture in which we expect larger internal energy and/or magnetic flux in the western jet and higher kinetic energy in the eastern jet. Previous observations at lower frequencies have found slower velocities of the moving jet features as compared to this work. Considering the different velocities in different areas, we suggest a spine-sheath structure with a faster inner layer and slower outer layer. [ABSTRACT FROM AUTHOR]

The energetic feedback that is generated by radio jets in active galactic nuclei (AGNs) has been suggested to be able to produce fast outflows of atomic hydrogen (H I) gas, which can be studied in absorption at high spatial resolution. We have used the Very Large Array (VLA) and a global very long baseline interferometry (VLBI) array to locate and study in detail the H I outflow discovered with the Westerbork Synthesis Radio Telescope (WSRT) in the restarted radio galaxy 3C 236. Based on the VLA data, we confirm a blueshifted wing of the H I with a width of ~1000 km s−1. This H I outflow is partially recovered by the VLBI observation. In particular, we detect four clouds with masses of 0.28 − 1.5 × 104M⊙ with VLBI that do not follow the regular rotation of most of the H I. Three of these clouds are located, in projection, against the nuclear region on scales of ≲40 pc, while the fourth is cospatial to the southeast lobe at a projected distance of ~270 pc. Their velocities are between 150 and 640 km s−1 blueshifted with respect to the velocity of the disk-related H I. These findings suggest that the outflow is at least partly formed by clouds, as predicted by some numerical simulations, and that it originates already in the inner (few tens of pc) region of the radio galaxy. Our results indicate that the entire outflow might consist of many clouds, possibly with comparable properties as those clearly detected, but distributed at larger radii from the nucleus where the lower brightness of the lobe does not allow us to detect them. However, we cannot rule out a diffuse component of the outflow. Because 3C 236 is a low-excitation radio galaxy, it is less likely that the optical AGN is able to produce strong radiative winds. The improvement of bahiagrass, Paspalum notatum Flüggé, has been inhibited by reduced levels of genetic diversity in sexual tetraploid germplasm. A few experimental sexual tetraploid genotypes (ESTGs) have been generated by chromosome doubling, but these plants typically exhibit low vigor. The objectives of this work were to generate and characterize the ploidy level, mode of reproduction, and fertility of a novel 308 individual sexual synthetic tetraploid population (SSTP) developed by intercrossing 29 sexual F1 hybrids originated by hybridizing several naturally occurring apomictic tetraploids from diverse origin with a few ESTGs. Ploidy levels were determined using flow cytometry, and reproductive modes were evaluated by a molecular assay with aposporylinked markers and embryo sac observations. The tetraploid level and the sexual mode of reproduction remained stable after two cycles of recombination during the generation of the SSTP. Fertility was evaluated based on seed set under self- and open pollination during 3 yr. The SSTP exhibited in average 30.2 and 15.2% seed set under open and self-pollination, respectively, showing a predominantly cross-pollination behavior with variable levels of self-fertility. There were no differences in terms of fertility between the SSTP and the ESTG. The novel tetraploid population behaves as sexual and cross-pollinated, and it is expected to allow a more efficient genetic improvement under the proposed breeding approaches. Background: Peripheral blood dyscrasias in older patients are repeatedly seen in geriatric clinical practice; however, there is substantial lack of data about the epidemiology, possible causes and treatment options in this patient group. Proton pump inhibitors (PPI) are extensively used in older patients and associated with leukopenia. The primary objective of this study was the assessment of encoded cytopenia prevalence in a geriatric patient cohort and the secondary objective was the assessment of putative causes and the analysis of PPI administration in patients with cytopenia.Methods: Retrospective evaluation of patients admitted to the geriatric department of a German urban hospital between 2010 and 2012. Electronic patient data were screened for encoded diagnosis of cytopenia according to the International Classification of Diseases (ICD) 10. Inclusion criteria were ICD code D69.0–9 and/or D70.0–7, age ≥60 years and exclusion criteria were no ICD code D69.0–9 and/or D70.0–7 and age <60 years. Out of 9328 screened inpatients 54 patients remained for analysis. Study parameters included hemoglobin (Hb), red blood cell count (RBC), leucocytes, platelets, mean cell volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), red cell distribution width (RDW), presence of leukopenia (<4000/µl), presence of thrombocytopenia (<140,000/µl) and presence of anemia according to the World Health Organization (WHO). Substitution of blood products, medication with PPI and potential causes for dyscrasias were evaluated based on electronic patient records.Results: The mean age was 78.3 ± 6.5 years (27 females, 27 males), anemia was seen in 78%, leukopenia was encoded in13% and thrombocytopenia in 44.4%. In most of the patients no substitution of blood products was documented. In most of the patients (20.4%) cytopenia was attributed to either heparin-induced thrombocytopenia (HIT) or hemato-oncologic (20.4%) diseases, followed by drug association in 18.5%. In 70.8% of the study patients PPIs were administered but the indication for PPI administration remained unclear in 20.4%.Conclusion: The results encourage accurate assessment of blood dyscrasias and appropriate documentation as well as indication check for PPI treatment in geriatric inpatients. One central goal of conservation biology is to conserve the genetic diversity of species in order to protect their adaptive potential. The main objective of this study was to identify management units (MUs) for the threatened noble crayfish ( Astacus astacus) in Western Europe by utilizing sequence and microsatellite analysis to determine populations in need of focused conservation programs. With the analysis of noble crayfish from 31 sampling sites from Belgium, France, The Netherlands and Germany, and further comparison of this data with a European-wide dataset, we propose four distinct MUs: the French Meuse (MU 1), the French Rhine (MU 2), the Belgian Scheldt and Meuse (MU 3) as well as populations from the French Seine (MU 4). This knowledge enables advanced A. astacus conservation management practises in these catchments by distinguishing between outbreeding and inbreeding populations and by preserving the maximum genetic diversity. When required, a high genetic diversity can be conserved by strengthen existing populations via stocking with populations that either bear the most common haplotype or population-specific private haplotypes in order to maintain recent and regional adaptions. Above all, stocking with populations that exhibit haplotypes from outside Western Europe should be avoided in these catchments. Evaluation of the degree of ossification of the medial clavicular epiphysis plays a crucial role in determining with an adequate degree of probability whether legally relevant age boundaries after the age of 17 have been crossed. In view of the need to avoid unnecessary radiation exposure, establishing non-X-ray methods for investigating the clavicle has long been a key objective in forensic age assessment research. Based on magnetic resonance imaging examinations in a large sample of healthy subjects, the current study for the first time presents statistical measures which allow inferences to be drawn for forensic age assessment in both sexes. We undertook a prospective study of a reference sample of 334 female and 335 male German volunteers aged from 12 to 24 using a 3-T MRI scanner. A 3D FFE (fast field echo) T1-weighted sequence with fat saturation was acquired. To stage ossification of the medial clavicular epiphysis, we used the clavicular ossification staging systems described by Schmeling et al. and Kellinghaus et al. Ossification stage IIIc offers a means in both sexes of demonstrating that the age of 18 has been attained prior to complete ossification of the epiphyseal plate. In both sexes, if a subject has reached ossification stage IV, it can be stated that he or she has attained the age of 21. Background: Our understanding of the molecular mechanisms implementing pubertal maturation of the testis in vertebrates is incomplete. This topic is relevant in Atlantic salmon aquaculture, since precocious male puberty negatively impacts animal welfare and growth. We hypothesize that certain miRNAs modulate mRNAs relevant for the initiation of puberty. To explore which miRNAs regulate mRNAs during initiation of puberty in salmon, we performed an integrated transcriptome analysis (miRNA and mRNA-seq) of salmon testis at three stages of development: an immature, long-term quiescent stage, a prepubertal stage just before, and a pubertal stage just after the onset of single cell proliferation activity in the testis. Results: Differentially expressed miRNAs clustered into 5 distinct expression profiles related to the immature, prepubertal and pubertal salmon testis. Potential mRNA targets of these miRNAs were predicted with miRmap and filtered for mRNAs displaying negatively correlated expression patterns. In summary, this analysis revealed miRNAs previously known to be regulated in immature vertebrate testis (miR-101, miR-137, miR-92b, miR-18a, miR-20a), but also miRNAs first reported here as regulated in the testis (miR-new289, miR-30c, miR-724, miR-26b, miR-new271, miR- 217, miR-216a, miR-135a, miR-new194 and the novel predicted n268). By KEGG enrichment analysis, progesterone signaling and cell cycle pathway genes were found regulated by these differentially expressed miRNAs. During the transition into puberty we found differential expression of miRNAs previously associated (let7a/b/c), or newly associated (miR-15c, miR-2184, miR-145 and the novel predicted n7a and b) with this stage. KEGG enrichment analysis revealed that mRNAs of the Wnt, Hedgehog and Apelin signaling pathways were potential regulated targets during the transition into puberty. Likewise, several regulated miRNAs in the pubertal stage had earlier been associated (miR-20a, miR-25, miR-181a, miR-202, let7c/d/a, miR-125b, miR-222a/b, miR-190a) or have now been found connected (miR-2188, miR-144, miR-731, miR-8157 and the novel n2) to the initiation of puberty. Conclusions: This study has - for the first time - linked testis maturation to specific miRNAs and their inversely correlated expressed targets in Atlantic salmon. The study indicates a broad functional conservation of already known miRNAs and associated pathways involved in the transition into puberty in vertebrates. This work presents HVOF as an alternative means to produce dense Babbitt coatings by thermal spray. A radial injection setup and low fuel flow rates were used to minimize heat transfer to the low melting point alloy. In-flight particle diagnostic systems were used to correlate spray parameters with the changes in particle velocity and thermal radiation intensity. The use of particles with larger diameters resulted in higher deposition efficiencies. It was shown that HVOF Babbitt coatings combine a dense structure and a fine distribution of intermetallic phases when compared to more traditional babbitting techniques. [ABSTRACT FROM AUTHOR]

Most current cartilage testing devices require the preparation of excised samples and therefore do not allow intra-operative application for diagnostic purposes. The gold standard during open or arthroscopic surgery is still the subjective perception of manual palpation. This work presents a new diagnostic method of ultrasound palpation (USP) to acquire applied stress and strain data during manual palpation of articular cartilage. With the proposed method, we obtain cartilage thickness and stiffness. Moreover, repeated palpations allow the quantification of relaxation effects. USP measurements on elastomer phantoms demonstrated very good repeatability for both, stage-guided (97.2%) and handheld (96.0%) applications. The USP measurements were compared with conventional indentation experiments and revealed very good agreement on elastomer phantoms ( $$r = 0.98$$ ) and good agreement on porcine cartilage samples ( $$r = 0.76$$ ). Artificially degenerated cartilage samples showed reduced stiffness, weak capacity to relax after palpation and an increase of stiffness of approximately 50% with each single palpation. Intact cartilage was measured by USP directly at the patella (in situ) and after excision and removal of the subchondral bone (ex situ), leading to stiffness values of $$12.1\pm 5.5$$ and $$8.5\pm 5.9\,\hbox {MPa}$$ ( $$p<0.05$$ ), respectively. The results demonstrate the potential of the USP system for cartilage testing, its sensitivity to degenerative changes and as a method for quantifying relaxation processes by means of repeated palpations. Furthermore, the differences in the results of in-situ and ex-situ measurements are of general interest, since such comparison has not been reported previously. Context. 67P/Churyumov-Gerasimenko (67P/C-G) is a short-period Jupiter family comet with an orbital period of 6.55 yr. Being the target comet of ESA's Rosetta mission, 67P/C-G has become one of the most intensively studied minor bodies of the solar system. The Rosetta Orbiter and the Philae Lander have brought us unique information about the structure and activity of the comet nucleus, as well as its activity along the orbit, composition of gas, and dust particles emitted into the coma. However, as Rosetta stayed in very close proximity to the cometary nucleus (less than 500 km with a few short excursions reaching up to 1500 km), it could not see the global picture of a coma at the scales reachable by telescopic observations (10³-105 km). Aims. In this work we aim to connect in-situ observations made by Rosetta with the morphological evolution of the coma structures monitored by the ground-based observations. In particular, we concentrate on causal relationships between the coma morphology and evolution observed with the Nordic Optical Telescope (NOT) in the Canary Islands, and the seasonal changes of the insolation and the activity of the comet observed by the Rosetta instruments. Methods. Comet 67P/C-G was monitored with the NOT in imaging mode in two colors. Imaging optical observations were performed roughly on a weekly basis, which provides good coverage of short- and long-term variability. With the three dimensional modeling of the coma produced by active regions on the southern hemisphere, we aim to qualify the observed morphology by connecting it to the activity observed by Rosetta. Results. During our monitoring program, we detected major changes in the coma morphology of comet 67P/C-G. These were long-term and long-lasting changes. They do not represent any sudden outburst or short transient event, but are connected to seasonal changes of the surface insolation and the emergence of new active regions on the irregular shaped comet nucleus. We have also found significant deviations in morphological changes from the prediction models based on previous apparitions of 67P/C-G, like the time delay of the morphology changes and the reduced activity in the northern hemisphere. According to our modeling of coma structures and geometry of observations, the changes are clearly connected with the activity in the southern hemisphere observed by the Rosetta spacecraft. In the first part of this article, we extend the formal upscaling of a diffusion–precipitation model through a two-scale asymptotic expansion in a level set framework to three dimensions. We obtain upscaled partial differential equations, more precisely, a non-linear diffusion equation with effective coefficients coupled to a level set equation. As a first step, we consider a parametrization of the underlying pore geometry by a single parameter, e.g. by a generalized “radius” or the porosity. Then, the level set equation transforms to an ordinary differential equation for the parameter. For such an idealized setting, the degeneration of the diffusion tensor with respect to porosity is illustrated with numerical simulations. The second part and main objective of this article is the analytical investigation of the resulting coupled partial differential equation–ordinary differential equation model. In the case of non-degenerating coefficients, local-in-time existence of at least one strong solution is shown by applying Schauder's fixed point theorem. Additionally, non-negativity, uniqueness, and global existence or existence up to possible closure of some pores, i.e. up to the limit of degenerating coefficients, is guaranteed. Thermal requirements of larval weatherfish Misgurnus fossilis were investigated in terms of growth, survival and aerobic performance. Growth and survival of M. fossilis larvae acclimated to five temperatures (11, 15, 19, 23 and 27° C) were measured over 25 days. In the upper temperature treatments (19, 23 and 27° C), survival of larvae was stable throughout the entire rearing period (>75%), whereas 11 and 15° C resulted in severe declines in survival (to <10%). Growth of larvae (expressed as dry mass and total length) was highest at 19 and 23° C, but significantly decreased at 27° C. Routine metabolic rate of 3 days post-hatch larvae was estimated as oxygen consumption rate ( Ṁ O2) during acute exposure (30 min to 1 h) to seven temperatures (11, 15, 19, 23, 27, 31 and 35° C). Larval oxygen uptake increased with each consecutive temperature step from 11 to 27° C, until a plateau was reached at temperatures >27° C. All larvae of the 35° C regime, however, died within the Ṁ O2 measurement period. M. fossilis larvae show greater than expected tolerance of high temperatures. On the other hand, low temperatures that are within the range of likely habitat conditions are critical because they might lead to high mortality rates when larvae are exposed over periods >10 days. These findings help to improve rearing conditions and to identify suitable waters for stocking and thus support the management of re-introduction activities for endangered M. fossilis. To improve the accuracy of forensic age estimation where there is no legal basis for carrying out x-ray examinations, it would be useful to establish non-x-ray imaging techniques. The objective of this study was to provide reference data for the magnetic resonance imaging-based evaluation of the ossification stage of the distal radius. Furthermore, we tested a new criterion of the maturity of the distal radial epiphysis for determining whether an individual has completed the age of 18. We investigated 668 MRI scans of the distal radial epiphysis from 333 female and 335 male subjects ranging in age from 12 to 24. To determine the ossification stage, we used the clavicular ossification staging systems described by Schmeling et al. and Kellinghaus et al. Ossification stage IV as described by Schmeling et al. was divided into two sub-stages, IVa and IVb, depending on whether or not it was possible to identify a triple-banded meta-epiphyseal zone of calcification. All study subjects were able to be assigned to an ossification stage without ambiguity. We present statistics relating to the distribution of ossification stages divided by sex. The age of the youngest female subject assessed as ossification stage IVb was 16.8, the age of the youngest male subject 18.6. The youngest age at which female subjects were assessed as ossification stage V was 22.3; for male subjects it was 23.1. Further independent studies should be carried out to determine whether ossification stage IVb can indeed be used to reliably determine whether a male subject has completed the age of 18. Evaluation of the ossification of the medial clavicular epiphysis plays a key role in forensic age estimation, particularly in determining whether the age of 18 has been attained. A key research objective in the forensic age estimation field at present is to establish non-X-ray methods for investigating the clavicle. This paper looks at the use of magnetic resonance imaging for evaluating the developmental state of the medial clavicular epiphysis. Clavicle specimens obtained from autopsies of 125 female and 270 male subjects aged from 10 to 30 were examined using a 3-T magnetic resonance scanner. One FFE-3D-T1 gradient echo sequence and one 2D-T2 turbo spin echo sequence were acquired. In each case, two investigators undertook a consensual determination of the ossification stage of the medial clavicular epiphysis using recognised classification systems. To determine intra-observer and inter-observer agreement, 80 clavicle specimens were subjected to repeat evaluation. We present statistics relating to the ossification stages. The inclusion of established sub-stages of clavicular ossification offers an additional option for determining whether a subject has attained the age of 18 which is applicable in both sexes. For both sexes, the minimum ages for ossification stages 4 and 5 allow conclusions to be drawn about a subject's age at a point in time lying several years in the past. Magnetic resonance imaging is a valid investigatory procedure for determining the ossification stage of the medial clavicular epiphysis. INSL3 (insulin-like peptide 3) is a relaxin peptide family member expressed by Leydig cells in the vertebrate testis. In mammals, INSL3 mediates testicular descent during embryogenesis but information on its function in adults is limited. In fish, the testes remain in the body cavity, although the insl3 gene is still expressed, suggesting yet undiscovered, evolutionary older functions. Anti-Müllerian hormone (Amh), in addition to inhibiting spermatogonial differentiation and androgen release, inhibits the Fsh (follicle-stimulating hormone)-induced increase in insl3 transcript levels in zebrafish testis. Therefore, the two growth factors might have antagonistic effects. We examine human INSL3 (hINSL3) effects on zebrafish germ cell proliferation/differentiation and androgen release by using a testis tissue culture system. hINSL3 increases the proliferation of type A undifferentiated (A) but not of type A differentiating (A) spermatogonia, while reducing the proliferation of Sertoli cells associated with proliferating A. Since the area occupied by A decreases and that of A increases, we conclude that hINSL3 recruits A into differentiation; this is supported by the hINSL3-induced down-regulation of nanos2 transcript levels, a marker of single A spermatogonia in zebrafish and other vertebrates. Pulse-chase experiments with a mitosis marker also indicate that hINSL3 promotes spermatogonial differentiation. However, hINSL3 does not modulate basal or Fsh-stimulated androgen release or growth factor transcript levels, including those of amh. Thus, hINSL3 seems to recruit A spermatogonia into differentiation, potentially mediating an Fsh effect on spermatogenesis. [ABSTRACT FROM AUTHOR]

Cardiac hypertrophy is a mechanism to compensate for increased cardiac work load, that is, after myocardial infarction or upon pressure overload. However, in the long run cardiac hypertrophy is a prevailing risk factor for the development of heart failure. During pathological remodelling processes leading to heart failure, decompensated hypertrophy, death of cardiomyocytes by apoptosis or necroptosis and fibrosis as well as a progressive dysfunction of cardiomyocytes are apparent. Interestingly, the induction of hypertrophy, cell death or fibrosis is mediated by similar signalling pathways. Therefore, tiny changes in the signalling cascade are able to switch physiological cardiac remodelling to the development of heart failure. In the present review, we will describe examples of these molecular switches that change compensated hypertrophy to the development of heart failure and will focus on the importance of the signalling cascades of the TGFβ superfamily in this process. In this context, potential therapeutic targets for pharmacological interventions that could attenuate the progression of heart failure will be discussed. [ABSTRACT FROM AUTHOR]

Context: Most deaths in the United States occur among older persons who have 1 or more disabling conditions. As a result, many deaths are preceded by an extended period during which family members provide care to their disabled relative.Objective: To better understand the effect of bereavement on family caregivers by examining predeath vs postdeath changes in self-reported and objective health outcomes among elderly persons providing varying levels of care prior to their spouse's death.Design and Setting: Prospective, population-based cohort study conducted in 4 US communities between 1993 and 1998.Participants: One hundred twenty-nine individuals aged 66 to 96 years whose spouse died during an average 4-year follow-up. Individuals were classified as noncaregivers (n = 40), caregivers who reported no strain (n = 37), or strained caregivers (n = 52).Main Outcome Measures: Changes in depression symptoms (assessed by the 10-item Center for Epidemiological Studies-Depression [CES-D] scale), antidepressant medication use, 6 health risk behaviors, and weight among the 3 groups of participants.Results: Controlling for age, sex, race, education, prevalent cardiovascular disease at baseline, and interval between predeath and postdeath assessments, CES-D scores remained high but did not change among strained caregivers (9.44 vs 9.19; P =.76), while these scores increased for both noncaregivers (4.74 vs 8.25; F(1,116) = 14.33; P<.001) and nonstrained caregivers (4.94 vs 7.13; F(1,116) = 4.35; P =.04). Noncaregivers were significantly more likely to be using nontricyclic antidepressant medications following the death than the nonstrained caregiver group (odds ratio [OR], 12.85; 95% confidence interval [CI], 1.02-162.13; P =.05). The strained caregiver group experienced significant improvement in health risk behaviors following the death of their spouse (1.47 vs 0.66 behaviors; F(1,118) = 20.23; P<.001), while the noncaregiver and nonstrained caregiver groups showed little change (0.27 vs 0.27 [P =.99] and 0.46 vs 0.27 [P =.39] behaviors, respectively). Noncaregivers experienced significant weight loss following the death (149.1 vs 145.3 lb [67.1 vs 65.4 kg]; F(1,101) = 8.12; P =.005), while the strained and nonstrained caregiving groups did not show significant weight change (156.2 vs 155.2 lb [70.3 vs 69.8 kg] [P =.41] and 156.2 vs 154.0 lb [70.3 vs 69.3 kg] [P =.12], respectively).Conclusions: These data indicate that the impact of losing one's spouse among older persons varies as a function of the caregiving experiences that precede the death. Among individuals who are already strained prior to the death of their spouse, the death itself does not increase their level of distress. Instead, they show reductions in health risk behaviors.
