The formation of environmental literacy in the education system through education for sustainable development (EfSD) experiences many obstacles. The aim of the research is to analyze the impact of interdisciplinary science learning for interdisciplinary oriented education for sustainable development to improve interdisciplinary thinking and environmental literacy of prospective teachers. The experimental method with a quasi-experimental pretest posttest group design was used in this study. The data collection technique used is an environmental literacy questionnaire and an interdisciplinary thinking test. Data analysis technique used independent t test. The results obtained were that the science interdisciplinary learning effective in improving interdisciplinary thinking performance and environmental literacy. The performance of students' interdisciplinary thinking affects environmental literacy of students. The university forms student teacher candidates who have the ability to think interdisciplinary so that they can manage science learning in an interdisciplinary and environmentally literate manner. The peatland conservation model is generally developed based on the community. A new education-based conservation model was developed through temporary activities through seminars, workshops or non-formal learning. The aim of this research is to develop and validation a peatland conservation model through interdisciplinary science learning that connects universities, schools and communities in peatland conservation activities. The development of a peatland conservation model went through third stages following interdisciplinary research patterns. Validation was determined by content and empirical validation. The peatland conservation model consisted of three interrelated dimensions in peatland conservation, namely universities, schools and communities. The model was equipped with implementation tools in the field in the form of science learning tools. The form of a peatland conservation model book, teaching practice materials, an environmental literacy questionnaire, and an interdisciplinary thinking performance rubric were developed in this study. The validated aspects included theory, assumptions, and conceptual model representations with the Aiken score was 0.88 as a valid category and the ICC value of 0.457 showed that the reliability was not good. Empirical validation showed that the dimensions of the university, school and community constructs had internal consistency reliability and validity. The relation between dimension that universities had an influence on schools and society, and schools have an influence on society. The peatland conservation model was equipped with practical teaching learning tools that directly connected universities and schools. However, this model was also suitable for non-educational natural sciences courses or pure natural sciences to form interdisciplinary thinking skills and environmental literacy. Neglected tropical diseases are those caused by infectious agents or parasites and are considered endemic in low-income populations. These diseases also have unacceptable indicators and low investment in research, drug production, and control. Tropical diseases such as leishmaniasis are some of the main causes of morbidity and mortality around the globe. Electrochemical immunosensors are promising tools for diagnostics against these diseases. One such benefit is the possibility of assisting diagnosis in isolated regions, where laboratory infrastructure is lacking. In this work, different peptides were investigated to detect antibodies against Leishmania in human and canine serum samples. The peptides evaluated (395-KKG and 395-G) have the same recognition site but differ on their solid-binding domains, which ensure affinity to spontaneously bind to either graphene oxide (GO) or graphene quantum dots (GQD). Cyclic voltammetry and differential pulse voltammetry were employed to investigate the electrochemical behavior of each assembly step and the role of each solid-binding domain coupled to its anchoring material. The graphene affinity peptide (395-G) showed better reproducibility and selectivity when coupled to GQD. Under the optimized set of experimental conditions, negative and positive human serum samples responses were distinguished based on a cut-off value of 82.5% at a 95% confidence level. The immunosensor showed selective behavior to antibodies against Mycobacterium leprae and Mycobacterium tuberculosis, which are similar antibodies and potentially sources of false positive tests. Caspases are crucial mediators of programmed cell death (apoptosis). Apoptosis can occur in spermatozoa during spermatogenesis or epididymal transit, as well as in ejaculated spermatozoa. A high proportion of apoptotic sperm would be a poor indicator of the freezability of a raw seminal sample. Alpaca spermatozoa are notoriously difficult to freeze successfully. Therefore, the objectives of this study were to study caspase activation during incubation (37°C) of fresh alpaca spermatozoa, as well as before and after cryopreservation, to gain some insight into the mechanisms behind the vulnerability of alpaca spermatozoa. Eleven sperm samples were incubated for 4 h at 37°C (Study 1), and 23 samples were frozen using an automated system (Study 2). Caspase‐3/7 activation was assessed at 0,1,2,3, and 4 h in samples incubated at 37°C (Study 1); and before/after cryopreservation (Study 2) using CellEvent™ Caspase 3/7 Green Detection Reagent and flow cytometry. The proportions of alpaca spermatozoa with caspase‐3/7 activated increased (p < 0.05) after 3–4 h of incubation at 37°C; however, caspase activation was similar before and after cryopreservation (36.2 ± 11.2% vs. 36.6 ± 33.7%, p > 0.05). The high standard deviation found after freezing could be explained by the existence of two subpopulations: one subpopulation where caspase‐3/7 activation decreased during cryopreservation (from 36.6 ± 9.1% to 1.5 ± 2.2%), and the other subpopulation where caspase‐3/7 activation increased after cryopreservation (from 37.7 ± 13.0% to 64.3 ± 16.7%). In conclusion, after 3–4 h of incubation, caspase‐3/7 activation increased in fresh alpaca sperm, whereas cryopreservation affects alpaca sperm samples in different ways. [ABSTRACT FROM AUTHOR]

Eighty‐five sperm samples were cryopreserved and SYBR14/PI, MitoTracker Deep Red FM, FITC‐PSA/PI and chlortetracycline were used for imaging flow cytometry evaluation of sperm viability, mitochondrial membrane potential (MMP), acrosome integrity and sperm capacitation, respectively. Sperm motility was also registered. Sperm motility (46.1 ± 7.7 vs. 24.1% ± 6.5%), sperm viability (49.8 ± 11.5 vs. 32.3% ± 9.6%) and high MMP (49.8% ± 12.4% vs. 34.9% ± 9.9%) decreased significantly (p <.05) during cryopreservation process, in contrast to acrosome‐reacted in viable spermatozoa (1.0% ± 1.6% vs. 1.0% ± 1.0%) and sperm capacitation (10.0 ± 9.8 vs. 8.2% ± 12.4%) that were similar (p >.05) before and after cryopreservation. Positive correlations were found between sperm motility versus high MMP (r =.63), sperm motility versus sperm viability (r =.67) and sperm viability versus high MMP (r =.88). In conclusion, cryopreservation of alpaca spermatozoa is related to a decrease in sperm motility, sperm viability and high MMP, meanwhile acrosome integrity and sperm capacitation are not affected. The development of immunosensors to detect antibodies or antigens has stood out in the face of traditional methods for diagnosing emerging diseases such as the one caused by the SARS-CoV-2 virus. The present study reports the construction of a simplified electrochemical immunosensor using a graphene-binding peptide applied as a recognition site to detect SARS-CoV-2 antibodies. A screen-printed electrode was used for sensor preparation by adding a solution of peptide and reduced graphene oxide (rGO). The peptide-rGO suspension was characterized by scanning electron microscopy (SEM), Raman spectroscopy, and Fourier transform infrared spectroscopy (FT-IR). The electrochemical characterization (electrochemical impedance spectroscopy—EIS, cyclic voltammetry—CV and differential pulse voltammetry—DPV) was performed on the modified electrode. The immunosensor response is based on the decrease in the faradaic signal of an electrochemical probe resulting from immunocomplex formation. Using the best set of experimental conditions, the analytic curve obtained showed a good linear regression (r2 = 0.913) and a limit of detection (LOD) of 0.77 μg mL−1 for antibody detection. The CV and EIS results proved the efficiency of device assembly. The high selectivity of the platform, which can be attributed to the peptide, was demonstrated by the decrease in the current percentage for samples with antibody against the SARS-CoV-2 S protein and the increase in the other antibodies tested. Additionally, the DPV measurements showed a clearly distinguishable response in assays against human serum samples, with sera with a response above 95% being considered negative, whereas responses below this value were considered positive. The diagnostic platform developed with specific peptides is promising and has the potential for application in the diagnosis of other infections that lead to high antibody titers. The emergence of antibiotic-resistant bacteria in the intestinal microbiota of broiler chickens, and the possibility of antibiotic residues in meat and meat products are seen as a threat to animal and human health. The search for alternatives to conventional drugs, including natural compounds, is an interesting approach to prevent the adverse effects of antibiotics. Therefore, the aim of the present study was to evaluate whether the addition of tannin (as a quebracho extract) in the diet of broiler chickens could be used to replace in-feed antimicrobials and coccidiostatic drugs, to exert positive effects on animal health and performance. The use of tannin in the diet of broiler chicks as an additive (at 0.5% for 1–10-day-old birds; and 1.0% for 11–42-day-old broiler chickens) enhanced their bodyweight, weight gain and daily weight gain on Day 42 of life, compared with the untreated control group (P < 0.05). However, these findings were not observed when tannin was added at 0.5%. Moreover, broiler chickens fed with an extract containing tannins showed higher erythrocyte counts, as well as haemoglobin and haematocrit concentrations than did those in the control group, whereas counts of total leukocytes and lymphocytes were lower (P < 0.05). The use of a diet with tannins did not influence meat quality compared with a diet containing zinc bacitracin and salinomycin, which altered some parameters associated with meat colour. Moreover, the diet with tannins reduced (P < 0.05) the number of oocysts of Eimeria sp. (36-day-old broilers) and the total faecal bacterial counts (42-day-old broilers) compared with the control group. Finally, intestinal histopathology showed that the addition of tannins minimised the degree of lesions (Degree 1) compared with the control group (Degree 3). On the basis of these results, we concluded that the use of quebracho extract containing tannins can be a suitable approach to improve the performance of broiler chickens, replacing in-feed antibiotics and coccidiostatic drugs. The use of tannins did not change meat quality; however, it caused positive effects on the immune system, and exerted potent bactericidal and coccidiostatic properties, reinforcing its use as a replacement for conventional drugs. Quebracho extract containing tannins when used in broiler chicken diets is capable of replacing in-feed drugs used as growth promoters (antimicrobials and coccidiostatics), because both experimental groups performed similarly. Tannins in the diet showed antimicrobial and anticoccidial effects, with results similar to those seen in animals that received conventional chemical drugs. Tannin caused an anti-inflammatory effect against coccidiosis, and, consequently, it reduced intestinal lesions caused by this parasite and its usual exacerbated inflammatory response. Contents The objective of this study was to detect changes in intracellular reactive oxygen species (superoxide anion and hydrogen peroxide) production and lipid peroxidation during cryopreservation of alpaca spermatozoa. Twelve alpaca semen samples were conventionally cryopreserved. Intracellular superoxide anion and hydrogen peroxide were evaluated by fluorescence microscopy using dihydroethidium ( DHE)/ YO- PRO-1 and dichlorofluorescein diacetate (H2 DCFDA)/propidium iodide ( PI), respectively. Evaluations were performed during cooling curve at (1) 25°C, (2) 15°C, (3) 5°C/0 min, (4) 5°C/15 min, (5) 5°C/30 min and (6) after freezing/thawing. Evaluation of lipid peroxidation by measuring malondialdehyde ( MDA) was performed at 25°C, 5°C/30 min and after thawing. Maximum percentages of total spermatozoa producing superoxide anion and hydrogen peroxide were found at 5°C/30 min (62.8 ± 6.3% and 30.5 ± 5.6%, respectively), and these results were higher ( p < .05) than initial (25°C: 10.8 ± 3.8% and 6.8 ± 0.7%, respectively) and after thawing (29.8 ± 9.5% and 7.5 ± 1.8%, respectively) values. However, considering only viable spermatozoa, production of superoxide anion and hydrogen peroxide during overall stabilization at 5°C (>76% and >91%, respectively) and after thawing (74.9 ± 5.0% and 78.9 ± 2.2%, respectively) was higher ( p < .05) than initial values at 25°C (38.7 ± 3.1% and 53.6 ± 2.0%, respectively). Lipid peroxidation at 25°C, 5°C/30 min, and post-thawing were 346.5 ± 99.8, 401.1 ± 64.8 and 527.7 ± 142.8 ng/ ml MDA, respectively. These results showed that high percentage of viable alpaca spermatozoa produces intracellular reactive species oxygen ( ROS) during the cryopreservation process of alpaca semen. Contents Although computer-assisted systems for sperm morphometry and morphological analysis are important tools in the study of male fertility, their use in extensive systems in alpacas is limited by factors such as the expense of equipment and the high altitudes of the Andean region. The objectives of this study were to evaluate alpaca sperm head morphometry using a nonautomated digital method and determine the frequency of sperm abnormalities based on strict criteria for sperm morphology in fertile male alpacas. Ejaculates ( n = 15) from seven alpacas were collected, and sperm smears stained with modified Papanicolaou were processed. For morphometric analysis, 3,000 sperm (200 cells/sample) images were captured at 400× magnification and Quick Photo MICRO 3.0 software was used for manual measurement of basic (sperm head length, width, perimeter and area) and derived variables (ellipticity, shape factor, elongation and regularity). For morphology assessment, smears were observed at 1000× magnification according to WHO and strict criteria. Average morphometric parameters were length 5.48 μm, width 2.99 μm, perimeter 13.62 μm, area 12.43 μm2, ellipticity 1.86, shape factor 1.20, elongation 0.29 and regularity 1.05. Significant between-individual and within-individual differences were found in morphometric parameters. Based on morphometric study, sperm heads were classified as elliptical or normal (49%), long (18%), short (2%), pyriform (12%), round (9%), large (6%) and small (4%). Morphological analysis found no additional sperm head defects in 49% of normal sperm obtained by morphometry, although a 4% incidence of neck/mid-piece defects and a 16% incidence of principal-piece defects were found. We conclude that sperm head morphometry assessment in fertile alpacas using a nonautomated digital method is feasible, and that defects in sperm heads constitute the main morphological alteration (>50% of the sperm population), based on WHO and strict criteria. The objective was to evaluate the effect of different cryoprotectant agents in the cryopreservation of Peruvian Paso horse semen. Twenty semen samples were collected from five Peruvian Paso horse stallions. Each sample was divided into 12 parts to form the groups: dimethylacetamide ( DMA), dimethyl sulfoxide ( DMSO), ethylene glycol ( EG) and glycerol ( GLY), at 3%, 4% and 5%. Samples were frozen using a rate-controlled freezer. Sperm parameters evaluated were motility and viability/acrosomal status. After thawing, progressive motility in DMA group was higher ( p < .05) than in DMSO, EG and GLY groups. Similarly, viable acrosome-intact spermatozoa were higher ( p < .05) using DMA in comparison with DMSO. No differences were found when comparing concentrations for any of the cryoprotectant agents. In conclusion, DMA seems to be a good cryoprotectant agent for the cryopreservation of Peruvian Paso horse stallion semen. [ABSTRACT FROM AUTHOR]

Aim: To evaluate two extenders and two cryoprotectant agents (CPA) for alpaca semen cryopreservation. Methods: Semen samples were obtained from four adult alpacas ( Lamapacos) and frozen using extender I (TRIS, citrate, egg yolk and glucose) or extender II (skim milk, egg yolk and fructose), each containing either glycerol (G) or ethylene glycol (EG) as CPA. Consequently, four groups were formed: 1) extender I-G; 2) extender I-EG; 3) extender II-G; and 4) extender II-EG. Semen was diluted in a two-step process: for cooling to 5 °C (extenders without CPA), and for freezing (extenders with CPA). Viability and acrosome integrity were assessed using trypan blue and Giemsa stains. Results: When compared, the motility after thawing was higher ( P < 0.05) in groups II-EG (20.0 %± 6.7 %) and II-G (15.3 %± 4.1 %) than that in groups I-G (4.0 %± 1.1 %) and I-EG (1.0 %± 1.4 %). Viable spermatozoa with intact acrosomes in groups II-EG (18.7 %± 2.9 %) and II-G (12.7 %± 5.9 %) were higher than that in groups I-G (5.7 %± 1.5 %) and I-EG (4.0 %±1.0 %). Conclusion: The skim milk- and egg yolk-based extenders containing ethylene glycol or glycerol to freeze alpaca semen seems to promote the survival of more sperm cells with intact acrosomes than the other extenders. Spermatogonial stem cell (SSC) is known for its self‐renewal capacity. We have studied the in vitro proliferation of isolated SSC from adult alpaca (Vicugna pacos) testes. A total of 107 samples were evaluated of which 31 were evaluated at baseline, 36 were cultivated in DMEM and 40 in STEMPRO media. Half of the cultivated samples was analysed after 14 days, and the rest after 21 days. Round cell subpopulations were identified with FITC‐DBA by flow cytometry: strongly positive DBA (sDBA+) as SSC, weakly positive DBA (wDBA+) as in early differentiation and negative DBA as differentiated. At the beginning, 4.16% of the cells were SSC, 37.61% wDBA+ while 54.12% were DBA−. After 14 days, 42.28% of SSC, 44.68% wDBA+ and 11.07% DBA− were found in DMEM while 47.09% of SSC, 32.57% wDBA+ and 18.48% DBA− in STEMPRO. After 21 days 38.66% were SSC, 52.78% wDBA and 7.65% DBA− in DMEM and on STEMPRO media 47.92% SSC, 44.20% wDBA+, 4.93% DBA−. There is a significant difference between the number of initial and SSC cultivated, as well as between DBA− (p < 0.05), while there is no significant difference between the wDBA+ (p > 0.05). Our results suggest that both culture media are appropriate for the in vitro proliferation of alpacas SSC. [ABSTRACT FROM AUTHOR]

The identification system of spermatogonial stem cell (SSC) was established in alpaca using the molecular expression as well as the reactivity pattern to Dolichos biflorus agglutinin (DBA) by flow cytometry. Twenty‐four testicles with their epididymis were recovered from adult alpacas at the slaughterhouse of Huancavelica—Perú. Samples were transported to the Laboratory of Reproductive Physiology at Universidad Nacional Mayor de San Marcos. Testes were selected for our study when the progressive motility of epididymal spermatozoa (ESPM) was above 30%. Isolation of SSC was performed with two enzymatic digestions. Finally, sperm viability was evaluated by means of the trypan blue vital stain in spermatogonial round cells. Samples with more than 80% viability were selected. Isolated cells cultured for 2 days were used for identifying the presence of SSCs by the expression of integrin β1 (116 bp) and PLZF (206 bp) genes. Spermatogonia were classified according to the DBA reactivity. Spermatogonia with a strong positive to DBA (sDBA+) were classified as SSC (Mean ± SEM=4.44 ± 0.68%). Spermatogonia in early differentiation stages stained weakly positive with DBA (wDBA+) (Mean ± SEM=37.44 ± 3.07%) and differentiated round cells as DBA negative (Mean ± SEM=54.12 ± 3.18%).
