Myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML) are severe myeloid disorders associated with significant morbidity and mortality. Because of patient and disease factors, many older adults are treated as outpatients with less-intensive therapy. Optimal supportive care strategies to minimize bleeding and infectious complications in this patient population have not been systematically evaluated. We conducted a survey of Canadian hematologists to explore current practice in the use of tranexamic acid (TXA) and prophylactic antimicrobials in patients with MDS/AML treated with less-intensive therapy, and to evaluate equipoise for future trials. Survey items were generated through a combination of literature review and discussion with content experts. The survey was disseminated to 304 potential respondents with a response rate of 52%. Prophylactic platelet transfusions were used by 95%, while prophylactic TXA was used by 57%; the most frequent reason for not using TXA was uncertainty about benefit or harm. Use of prophylactic antimicrobials varied by chemotherapy regimen. If antimicrobial prophylaxis was used, the most frequently prescribed antibacterials were fluroquinolones (90%) and trimethoprim/sulfamethoxazole (21%); the most commonly used antifungals were fluconazole (66%) and voriconazole (36%). The most common reason for not using prophylactic antimicrobials was insufficient evidence of benefit. Most respondents agreed that clinical trials are needed to define the use of TXA and prophylactic antimicrobials in this patient population. Among survey respondents, there was variation in the use of supportive care strategies to address bleeding and infection risk in older adults with MDS/AML. Simple Summary: CPX-351 has been approved for the treatment of acute myeloid leukemia. Previous studies have indicated that dose adjustment is not warranted for patients with mild to moderate renal impairment. However, the effect of severe renal impairment on the use of CPX-351 has not been established. In this study, we evaluated CPX-351 in patients with normal renal function and in patients with moderate and severe renal impairment. We compared how CPX-351 moves through the body, as well as the safety of the drug, in these different patient groups. The results suggest that CPX-351 dose adjustment is not required for patients with hematologic malignancies with moderate or severe renal impairment. This open-label phase 1 study (clinicaltrials.gov, NCT03555955) assessed CPX-351 pharmacokinetics (PK) and safety in patients with hematologic malignancies with normal or impaired renal function. Patients were enrolled into three cohorts based on their creatinine clearance (CrCl): ≥90 mL/min (Cohort 1, normal renal function, n = 7), 30 to <59 mL/min (Cohort 2, moderate renal impairment, n = 8), or <30 mL/min (Cohort 3, severe renal impairment, n = 6). Patients received intravenous CPX-351 for initial induction; blood and urine samples were collected for PK analysis. The primary objective was to assess the PK parameters for cytarabine, daunorubicin, and their respective metabolites, arabinosyluracil (Ara-U) and daunorubicinol. Renal impairment did not significantly impact the cytarabine, daunorubicin, or daunorubicinol exposure, but it caused a slight increase in the Ara-U exposure. The CPX-351 side effect profile was similar in patients with impaired renal function compared to those with normal renal function. All the patients reported ≥1 treatment-emergent adverse event (TEAE), most commonly febrile neutropenia and nausea (57% each) and hyperglycemia (43%); no patients discontinued treatment due to TEAEs. Tremendous progress has been made in variety development and host plant resistance to mitigate the impact of Fusarium head blight (FHB) since the disease manifested in the southeastern United States in the early 2000s. Much of this improvement was made possible through the establishment of and recurring support from the US Wheat & Barley Scab Initiative (USWBSI). Since its inception in 1997, the USWBSI has enabled land‐grant institutions to make advances in reducing the annual threat of devastating FHB epidemics. A coordinated field phenotyping effort for annual germplasm screening has become a staple tool for selection in public and private soft red winter wheat (SRWW) breeding programmes. Dedicated efforts of many SRWW breeders to identify and utilize resistance genes from both native and exotic sources provided a strong foundation for improvement. In recent years, implementation of genomics‐enabled breeding has further accelerated genetic gains in FHB resistance. This article reflects on the improvement of FHB resistance in southern SRWW and contextualizes the monumental progress made by collaborative, persistent, and good old‐fashioned cultivar development. Rye (Secale cereale L.) grain production in Kentucky is insufficient to meet the needs of distillers and bakers, in part because there is a knowledge gap about rye management that discourages farmers from choosing this crop. We conducted an economic study to develop recommendations for profitable rye grain production. The aim of this study was to determine the influence of two different nitrogen (N) rates (35 lb N acre−1 and 70 lb N acre−1) on yield and profitability of winter rye grain production. Experiments were conducted in 2020–2021 season at three Kentucky locations: Lexington, Princeton, and Adairville. Twenty‐four rye entries were planted in a split plot design experiment and the two N rates (35 lb N acre−1 and 70 lb N acre−1) were assigned to main plots. There was no significant difference in mean yield between 35 and 70 lb N acre−1. This indicates that less investment in N fertilizer will not adversely affect grain yield level, will enhance profitability of production, and will benefit distillers due to the higher alcohol yield associated with higher starch and lower protein levels. The presence or absence of awns—whether wheat heads are 'bearded' or 'smooth' – is the most visible phenotype distinguishing wheat cultivars. Previous studies suggest that awns may improve yields in heat or water-stressed environments, but the exact contribution of awns to yield differences remains unclear. Here we leverage historical phenotypic, genotypic, and climate data for wheat (Triticum aestivum) to estimate the yield effects of awns under different environmental conditions over a 12-year period in the southeastern USA. Lines were classified as awned or awnless based on sequence data, and observed heading dates were used to associate grain fill periods of each line in each environment with climatic data and grain yield. In most environments, awn suppression was associated with higher yields, but awns were associated with better performance in heat-stressed environments more common at southern locations. Wheat breeders in environments where awns are only beneficial in some years may consider selection for awned lines to reduce year-to-year yield variability, and with an eye towards future climates. Focus on local food production and supply chains has heightened in recent years, as evidenced and amplified by the COVID-19 pandemic. This study aimed to assess the suitability of soft red winter (SRW) wheat breeding lines for local artisan bakers interested in locally sourced, strong gluten wheat for bread. Seventy-six genotyped SRW wheat breeding lines were milled into whole wheat flour and baked into small loaves. Bread aroma, flavor, and texture were evaluated by a sensory panel, and bread quality traits, including sedimentation volume, dough extensibility, and loaf volume, were measured to estimate heritability. SE-HPLC was performed on white flour, and breeding lines were characterized for different protein fraction ratios. Heritability of loaf volume was moderately high (h2 = 0.68), while heritability of sedimentation volume, a much easier trait to measure, was slightly lower (h2 = 0.55). Certain protein fraction ratios strongly related to loaf volume had high heritability (h2 = 0.7). Even though only a moderate heritability estimate of dough extensibility was found in our study, high positive correlations were found between this parameter and sedimentation volume (r = 0.6) and loaf volume (r = 0.53). This low-input and highly repeatable parameter could be useful to estimate dough functionality characteristics. Flavor and texture heritability estimates ranged from 0.16 to 0.37, and the heritability estimate of aroma was not significantly different from zero. However, the sensorial characteristics were significantly correlated with each other, suggesting that we might be able to select indirectly for aroma by selecting for flavor or texture characteristics. From a genome-wide association study (GWAS), we identified six SNPs (single nucleotide polymorphisms) associated with loaf volume that could be useful in breeding for this trait. Cereal rye (Secale cereale) is a multi‐purpose crop, harvested for grain (for food, feed, and other products) as well as used as a forage or cover crop. The functionality of rye across these spheres is due to its abiotic and biotic stress tolerance and unique phytochemical composition. In this review, we focus on the current state and prospects of breeding rye for quality traits, where we define quality traits as grain, feed, or forage quality. Rye grain for human consumption has numerous healthful compounds, notably high dietary fiber content in grain and unique flavors that contribute to the signature properties of rye baked products and distilled spirits. As a forage, rye is useful in marginal environments and has high quality, including high crude protein, and is thus valuable for livestock production. While yield remains a major focus of breeding efforts, due to recent improvements in rye genomic and phenotypic resources, there are now opportunities for increased attention on breeding high‐quality ryes for multiple end uses. Through genetic improvement of quality traits, cereal rye has the potential to expand and serve multiple markets as a climate‐resilient crop. Reducing generation time is critical to achieving the goals of genetic gain in important crops like wheat (Triticum aestivum). Speed breeding (SB) has been shown to considerably reduce generation times in crop plants. Unlike spring wheat cultivars, winter wheat varieties require typically 6–9 weeks of cold treatment, called vernalization, for flowering which extends the generation time for the development of improved winter wheat cultivars. Here, we optimized the SB method using a set of 48 diverse soft red winter wheat (SRWW) cultivars by testing vernalization duration, light and temperature requirements, and the viability of seeds harvested after different durations post‐anthesis under extended daylight conditions. We have found that using a 22‐h setting (22 h day/2 h night, 25°C/22°C) in high‐density 50‐cell trays results in rapid generation advancement. We used genotypic data for a panel of soft red winter wheat varieties from the regional programs to determine the impact of photoperiod and vernalization alleles on the efficiency of the SB approach. Using a set of 48 SRWW cultivars and germplasm from Maryland and four other public breeding programs, we establish that this protocol can allow for the advancement of four generations per year in controlled conditions for winter wheat varieties, experimental lines, or emerging cultivars. Our work shows the potential to reduce generation time by ∼30 days per generation faster than what had been reported in the SB strategies for winter wheat, thus allowing for a quicker turnaround time from original cross to genetically stable experimental genotypes that can be tested in field settings. Kentucky's wheat (Triticum aestivum L.) crop is planted into a rotation in which corn (Zea mays L.) is followed by wheat, which is followed by double‐crop soybean [Glycine max (L.) Merr.]. The causal agent of Fusarium head blight (FHB), Fusarium graminearum Schwabe, overwinters in corn stubble; thus, the Kentucky wheat crop is always at risk for FHB. The preeminent goal of the University of Kentucky wheat breeding program is releasing FHB‐resistant winter wheat cultivars. 'Pembroke 2021' (Reg. no. CV‐1198, PI 701381) is an early‐maturing, semi‐dwarf, soft red winter wheat cultivar developed and released in 2020 by the Kentucky Agricultural Experiment Station for FHB resistance, high yield and test weight potential, and lodging resistance. Pembroke 2021, tested as KY 07C‐1145‐94‐12‐5, was developed from the cross 'IL99‐15867'/B990081//KY97C‐0554‐04‐05. The pedigree of the KY parent is VA94‐54‐549/Roane//Kristy. Pembroke 2021 was named for the Pembroke silt loam soil series prevalent in the main wheat‐producing area of Kentucky. Pembroke 2021 was selected from F4:5 head rows using a modified bulk breeding method; selected rows carried the resistance allele at the following FHB resistance quantitative trait loci: 1B Jamestown, 1A Neuse, 4A Neuse, and 3B Massey. Pembroke 2021 was tested in Kentucky in multi‐location replicated yield trials, the Uniform Eastern Soft Red Winter Wheat Nursery, the Mason‐Dixon nursery, the Uniform Northern Winter Wheat Scab Nursery, and the Kentucky Wheat Variety Trial. Breeding wheat (Triticum aestivum L.) has resulted in small gains in improved nutrient acquisition and use as numerous traits are involved. In this study, we evaluated the impact of breeding on P-acquisition and identified both plant and soil variables that could be used to inform the selection of germplasm with increased P acquisition efficiency. We previously screened a historic panel of winter wheat cultivars for root system architecture and root tip organic acid content when grown in P-deficient solution/agar and used these characteristics together with breeding history to develop a predicted P extraction potential (PEP). We tested the validity of the PEP classification by growing cultivars under sufficient and insufficient soil P conditions. Old, wild-type cultivars had the greatest P utilization efficiency (PUtE) when grown under insufficient P, likely a result of the chemical potential of wild-type (with respect to Rht-B1) cultivars (greater organic acid production) rather than root system size. Wild-type plants had differences in rhizosphere microbial community structure, rhizosphere bicarbonate-extractable P, and bulk soil Fe and Al, indicating the utilization of typically less available P pools. Wheat is a globally important crop and one of the “big three” US field crops. But unlike the other two (maize and soybean), in the United States its development is commercially unattractive, and so its breeding takes place primarily in public universities. Troublingly, the incentive structures within these universities may be hindering genetic improvement just as climate change is complicating breeding efforts. “Business as usual” in the US public wheat-breeding infrastructure may not sustain productivity increases. To address this concern, we held a multidisciplinary conference in which researchers from 12 US (public) universities and one European university shared the current state of knowledge in their disciplines, aired concerns, and proposed initiatives that could facilitate maintaining genetic improvement of wheat in the face of climate change. We discovered that climate-change- oriented breeding efforts are currently considered too risky and/or costly for most university wheat breeders to undertake, leading to a relative lack of breeding efforts that focus on abiotic stressors such as drought and heat. We hypothesize that this risk/cost burden can be reduced through the development of appropriate germplasm, relevant screening mechanisms, consistent germplasm characterization, and innovative models predicting the performance of germplasm under projected future climate conditions. However, doing so will require coordinated, longer-term, inter-regional efforts to generate phenotype data, and the modification of incentive structures to consistently reward such efforts. Acute myeloid leukemia (AML) is a hematological malignancy that predominantly affects the elderly. Prognosis declines with age. For those who cannot tolerate intensive chemotherapy, historically established treatment options have been hypomethylating agents (HMAs), low dose cytarabine (LDAC), and best supportive care (BSC). As the standard of care evolves for those unfit for intensive chemotherapy, there is a need to understand established treatment pathways, clinical outcomes and healthcare resource utilization in Canada. The CURRENT study was a retrospective chart review of AML patients not eligible for intensive chemotherapy who initiated first-line treatment between 1 January 2015 and 31 December 2018. Data were collected from 170 Canadian patients treated at six hematology centers, of whom 118 received systemic therapy and 52 received BSC as first-line treatment. Median overall survival was 8.58 months and varied from 2.96 months for BSC to 13.31 months for HMAs. Over 80% of patients had at least one outpatient visit, and 67% of patients receiving systemic therapy and 71% of those receiving BSC had at least one admission to hospital, during their first line of therapy. A total of 96 (81.4%) patients receiving first line systemic therapy and 39 (75.0%) of those receiving first line BSC had at least one red blood cell or platelet transfusion. Acute myeloid leukemia (AML) is a hematologic malignancy that most frequently develops in older adults. Overall, AML is associated with a high mortality although advancements in genetic risk stratification and new treatments are leading to improvements in outcomes for some subgroups. In this review, we discuss an individualized approach to intensive therapy with a focus on the role of recently approved novel therapies as well as the selection of post-remission therapies for patients in first remission. We discuss the management of patients with relapsed and refractory AML, including the role of targeted treatment and allogeneic stem cell transplant. Next, we review non-intensive treatment for older and unfit AML patients including the use of azacitidine and venetoclax. Objectives: This retrospective chart review examined real‐world healthcare resource utilization (HRU) in patients with AML ineligible for intensive therapy who received first‐line systemic therapy or best supportive care (BSC). Methods: Data were collected anonymously on patients with AML who initiated first‐line hypomethylating agents (HMA), low‐dose cytarabine (LDAC), other systemic therapy, or BSC. HRU endpoints included hospitalizations, outpatient consultations, transfusions, and supportive care. Results: Of 1762 patients included, 46% received HMA, 11% received LDAC, 17% received other systemic therapy, 26% received BSC; median treatment durations were 118, 35, 33, and 57 days, respectively. Most patients were hospitalized, most commonly for treatment administration, transfusion, or infection (HMA 82%, LDAC 93%, other systemic therapy 83%, BSC 83%). A median number of hospitalizations were 2–6 across systemic groups and two for BSC, with median durations of 8–18 days. Transfusion rates and outpatient consultations were highest for HMA (80% and 79%) versus LDAC (57% and 53%), other systemic therapy (57% and 63%), and BSC (71% and 66%). Antivirals/antibiotics and antifungals were used more frequently than growth factors (72–92%, 34–63%, and 7–27%, respectively). Objectives: Acute myeloid leukaemia (AML) is a disease of older adults, who are vulnerable to socio‐economic factors. We determined AML incidence in older adults and the impact of socio‐economic factors on outcomes. Methods: We included 3024 AML patients (1996–2016) identified from a population‐based registry. Results: AML incidence in patients ≥60 years increased from 11.01 (2001–2005) to 12.76 (2011–2016) per 100 000 population. Among 879 patients ≥60 years in recent eras (2010–2016), rural residents (<100 000 population) were less likely to be assessed by a leukaemia specialist (39% rural, 47% urban, p =.032); no difference was seen for lower (43%, quintile 1–3) vs. higher (47%, quintile 4–5) incomes (p =.235). Similar numbers received induction chemotherapy between residence (16% rural, 18% urban, p =.578) and incomes (17% lower, 17% high, p = 1.0). Differences between incomes were seen for hypomethylating agent treatment (14% low, 20% high, p =.041); this was not seen for residence (13% rural, 18% urban, p =.092). Among non‐adverse karyotype patients ≥70 years, 2‐year overall survival was worse for rural (5% rural, 12% urban, p =.006) and lower income (6% low, 15% high, p =.017) patients. Conclusions: AML incidence in older adults is increasing, and outcomes are worse for older rural and low‐income residents; these patients face treatment barriers. [ABSTRACT FROM AUTHOR]

Acute myeloid leukemia (AML) predominantly affects the elderly, and prognosis declines with age. Induction chemotherapy plus consolidation therapy is standard of care for fit patients; options for unfit patients include hypomethylating agents (HMA), low-dose cytarabine (LDAC), targeted therapies, and best supportive care (BSC). This retrospective chart review evaluated clinical outcomes in unfit patients with AML who initiated first-line treatment or BSC 01/01/2015–12/31/2018. Overall survival (OS), progression-free survival (PFS), time-to-treatment failure (TTF), and response rates were assessed. Of 1762 patients, 1310 received systemic therapies: 809 HMA, 199 LDAC, and 302 other therapies; 452 received BSC. Median OS was 9.9, 7.9, 5.4, and 2.5 months for HMA, LDAC, other, and BSC, respectively. Median PFS was 7.5, 5.3, 4.1, and 2.1 months for HMA, LDAC, other, and BSC, respectively; median TTF was 4.9, 2.1, 2.2, and 2.1 months, respectively. There has been an explosion of knowledge about the role of metabolism and the mitochondria in acute myeloid leukemia (AML). We have also recently seen several waves of novel therapies change the treatment landscape for AML, such as the selective B-cell lymphoma 2 (BCL-2) inhibitor venetoclax. In this new context, we review the rapidly advancing literature on the role of metabolism and the mitochondria in AML pathogenesis, and how these are interwoven with the mechanisms of action for novel therapeutics in AML. We also review the role of oxidative phosphorylation (OxPhos) in maintaining leukemia stem cells (LSCs), how recurrent genomic alterations in AML alter downstream metabolism, and focus on how the BCL-2 pathway and the mitochondria are inextricably linked in AML. Thus, we provide an overview of the mitochondria and metabolism in the context of our new therapeutic world for AML and outline how targeting these vulnerabilities may produce novel therapeutic strategies. Previous results from our laboratory have shown that optical sorter–based indirect selection reduced deoxynivalenol (DON) accumulation in soft red winter wheat (SRWW). In this paper we evaluate the efficacy of optical sorter–augmented genomic selection (OSA‐GS) for lowering DON accumulation at three selection intensities across 2 yr. In total, 758 SRWW breeding lines were genotyped and then phenotyped in an inoculated Fusarium head blight (FHB) nursery. Accumulation of DON was measured on all breeding lines. The proportion of Fusarium‐damaged kernels estimated using an optical sorter (FDKos) was measured on 120 lines; these data were used to train a genomic prediction model. Genomic estimated breeding values (GEBVs) for FDKos were computed for all lines without actual FDKos data. The top 20, 30, and 40% of lines without actual FDKos data were selected based on FDKos GEBVs. The same was done using actual measured DON values. Both strategies lowered DON, but traditional direct phenotypic selection based on actual DON values outperformed OSA‐GS. In other words, phenotypic selection was necessary to achieve the greatest reductions in DON. However, using previously published cost estimates for the price of an optical sorter, DON analysis, and genotyping, we determined that OSA‐GS required less financial investment than phenotypic selection based on measured DON. Taken together, our findings indicate that OSA‐GS is a cost‐effective method for lowering DON accumulation and support the usefulness of an optical sorter as a tool for FHB resistance breeding in SRWW. Organic acids are exuded by plant roots into the rhizosphere for a variety of reasons, including nutrient acquisition. The most frequently reported organic acids are those involved in the Krebs cycle and other important biochemical pathways that are responsible for the production of important plant metabolites, including oxalic, malic, and citric acid. Many studies report increased citric and malic acid exudation when plants are P‐deficient. Because plant breeding is typically carried out under heavily fertilized conditions, it is possible that wheat (Triticum aestivum L.) breeders have unintentionally selected for varieties with decreased nutrient acquisition capability from soil organic matter and soil colloids. In this study, we evaluated a diverse panel of 10 winter wheat cultivars, which included wild‐type (tall) and semidwarf varieties, for organic acid content. Plants were grown in solution culture with and without P for 24 d. Thirty 1‐cm root tips were bulked per sample and the extracts were analyzed by high‐performance liquid chromatography for citric, malic, and oxalic acid content. Under P deficiency, total organic acid content differed between wild‐type and semidwarf varieties, with wild‐type cultivars containing more organic acids in the root tips. This difference in total production was driven by oxalic acid, whereas citric and malic acid concentrations were conserved. Unlike citric and malic acid, oxalic acid is not necessary for major metabolic processes and therefore may be expendable. Breeding for semidwarf wheat corresponds with unintentional changes to root system function that could impact nutrient acquisition and other rhizosphere processes. Use of genetic resistance is one of the most important strategies to manage the devastating disease Fusarium head blight (FHB) in wheat. Numerous quantitative trait loci (QTL) having varying effects on reducing FHB and the mycotoxin deoxynivalenol (DON) accumulation have been reported from Asian, European, or distant sources such as wild relatives of wheat (Triticum aestivum L.). However, coming from nonadapted backgrounds, the incorporation of such QTL into regional breeding programs has often resulted in the simultaneous transfer of other undesirable traits. Therefore, it is important to identify, characterize, and deploy sources of genetic resistance that do not suffer from poor adaptability and/or linkage drag. In the present work, QTL associated with FHB resistance in a high‐yielding, moderately resistant soft red winter wheat cultivar 'Jamestown' were mapped and validated. The QTL mapping was done using a recombinant inbred line (RIL) population of Pioneer '25R47' × Jamestown having 186 individuals. Phenotyping over 2 yr at three locations, and genotyping using the 90K single nucleotide polymorphism (SNP) platform identified two new QTL, named QFHB.vt‐1B.1 and QFHB.vt‐1B.2, on the chromosome 1B long arm. The QTL contributed to FHB incidence, FHB severity, Fusarium‐damaged kernels, and DON content. Independent mapping of these QTL using two additional RIL populations of FG95195 × Jamestown (170 RILs) and Jamestown × LA97113UC‐124 (77 RILs) validated their stability and effectiveness in different genetic backgrounds. Kompetitive allele specific polymerase chain reaction (KASP) assays were developed using linked SNPs for marker‐assisted selection of the QTL. Previous results from our lab have shown that using an optical sorter to identify Fusarium head blight (FHB) resistant breeding lines was effective at reducing the toxin deoxynivalenol (DON) and FHB-associated kernel damage. In this paper we quantified the proportion of desirable genotypes at FHB resistance QTL in lines from three selection cycles of optical sorting. Breeding lines were genotyped at loci on chromosomes 3BS, 2DL, and 5A using the following DNA markers: TaHRC, CFD233, and GWM304. TaHRC is a KASP marker for Fhb1 , a major FHB resistance QTL on chromosome 3BS. CFD233 is an SSR marker for Qfhs.nau-2DL on chromosome 2DL. GWM304 is an SSR marker for Qfhs.ifa-5A on chromosome 5A. Sorter selection was effective at identifying lines that had the resistant genotype at TaHRC; in other words, the sorter was able to identify lines with resistance alleles at Fhb1. The sorter was less effective at selecting for the resistant genotype at CFD233 and GWM304. However, the proportion of lines with resistant genotypes at GWM304 did increase with additional sorter selection, just not to the degree that was observed for the Fhb1 -associated marker. The proportion of lines with resistant alleles at CFD233 did not show a consistent trend. In addition to increasing the proportion of lines with Fhb1 and Qfhs.ifa-5A each selection cycle, optical sorter-based mass selection enhanced FHB resistance in different marker genotype combinations evaluated in this study. For example, there were net reductions in DON and kernel damage after two cycles of sorter selection in 15X110601S07002, a line with Fhb1 , with Qfhs.nau-2DL, and with Qfhs.ifa-5A; final C3 DON levels were 63% of the resistant check (KY02C-3005-25). Kernel damage was also reduced in 15X110601A08221 a line without Fhb1 , without Qfhs.nau-2DL, and without Qfhs.ifa-5A. Our findings suggest the increased resistance observed in different marker genotype combinations was conferred by QTL other than Fhb1 , QFhs.nau-2DL, and Qfhs.ifa-5, and validate our previous results that the optical sorter is effective at selecting FHB-resistant breeding material. usarium graminearum, the major causal agent of Fusarium head blight (FHB) of wheat (Triticum aestivum) in the U.S., can produce mycotoxins, such as deoxynivalenol (DON), during infection. Contamination of wheat grain with DON is a major concern for wheat producers and millers, and the U.S. Food and Drug Administration (FDA) has set advisory levels for DON in finished wheat products for human and animal consumption. Practices utilized to manage FHB and DON contamination include planting wheat cultivars with moderate resistance to FHB and applying efficacious fungicides at the beginning of anthesis. Under severe epidemics, DON contamination can exceed FDA advisory levels despite implementation of these measures. Additionally, fungicide efficacy can be limited when anthesis is not uniform among plants in the field, which can occur when planting is delayed or if there is non-uniform seedling establishment. The objectives of this study were to evaluate the effect of (1) in-furrow phosphorus application at planting and seeding rate on heading and anthesis uniformity, FHB symptomology, DON contamination, grain yield, yield components, and test weight; and (2) harvesting at different grain moisture concentrations on FHB symptomology, DON contamination, grain yield and test weight. Field trials were established in Princeton, Kentucky, from 2017 to 2019, to evaluate in-furrow phosphorus application at planting (0 kg P2O5 ha-1 and 47 kg P2O5 ha-1); seeding rate (377 live seeds m-2 and 603 live seeds m-2); and grain moisture at harvest (20 to 22% and 13 to 15%). In-furrow phosphorus increased grain yield and spikes m-2, but had no effect on heading and anthesis uniformity or DON contamination. The 603 live seeds m-2 seeding rate decreased the number of days to Zadoks 60 for the November planted wheat, and decreased FHB incidence, but did not decrease DON contamination. Harvesting at 20 to 22% grain moisture decreased Fusarium damaged kernel ratings and percent kernel infection but increased DON contamination in the harvested grain. Although in-furrow phosphorus, seeding rate, and harvesting 20 to 22% grain moisture did not decrease DON contamination, there is potential for these treatments to alleviate negative effects of late planted wheat grown in stressful environments. Fusarium head blight (FHB) is a devastating disease in cereals around the world. Because it is quantitatively inherited and technically difficult to reproduce, breeding to increase resistance in wheat germplasm is difficult and slow. Genomic selection (GS) is a form of marker-assisted selection (MAS) that simultaneously estimates all locus, haplotype, or marker effects across the entire genome to calculate genomic estimated breeding values (GEBVs). Since its inception, there have been many studies that demonstrate the utility of GS approaches to breeding for disease resistance in crops. In this study, the Uniform Northern (NUS) and Uniform Southern (SUS) soft red winter wheat scab nurseries (a total 452 lines) were evaluated as possible training populations (TP) to predict FHB traits in breeding lines of the UK (University of Kentucky) wheat breeding program. DON was best predicted by the SUS; Fusarium damaged kernels (FDK), FHB rating, and two indices, DSK index and DK index were best predicted by NUS. The highest prediction accuracies were obtained when the NUS and SUS were combined, reaching up to 0.5 for almost all traits except FHB rating. Highest prediction accuracies were obtained with bigger TP sizes (300–400) and there were not significant effects of TP optimization method for all traits, although at small TP size, the PEVmean algorithm worked better than other methods. To select for lines with tolerance to DON accumulation, a primary breeding target for many breeders, we compared selection based on DON BLUES with selection based on DON GEBVs, DSK GEBVs, and DK GEBVs. At selection intensities (SI) of 30–40%, DSK index showed the best performance with a 4–6% increase over direct selection for DON. Our results confirm the usefulness of regional nurseries as a source of lines to predict GEBVs for local breeding programs, and shows that an index that includes DON, together with FDK and FHB rating could be an excellent choice to identify lines with low DON content and an overall improved FHB resistance. Cardiovascular (CV) disease is a common comorbidity in acute leukemia (AL) patients and can be worsened by the use of anthracyclines. Interruptions and underutilization of CV medications during AL treatments may negatively impact the CV health of these patients. A 30-question electronic survey was distributed to Canadian hematologists who treat adults with AL to determine the frequency, timing and rationale for interruptions in statins, antiplatelets and angiotensin antagonists in patients undergoing intensive chemotherapy. Strategies for mitigating anthracycline cardiotoxicity, methods of establishing baseline CV risk and utilization of clinical pharmacists were also assessed. Results indicate that it is common for AL patients undergoing intensive chemotherapy to require CV medication interruptions. This highlights the need for collaboration between hematology and cardiology healthcare teams and utilization of multidisciplinary healthcare professionals to improve CV care during AL. Fusarium head blight (FHB) is a destructive disease in wheat (Triticum aestivum) and other small grains. Morphological traits can act as barrier between pathogen and plant and provide a passive form of resistance to disease. The objectives were: (1) to phenotype 250 soft red winter wheat (SRWW) breeding lines and cultivars for morphological traits in 2015–2016; (2) to estimate their correlations with disease resistance; and (3) to identify QTL for morphological traits associated with FHB resistance through genome-wide association study (GWAS). Morphological traits measured were anther extrusion, plant height, peduncle length, spike length (SL), spikelet number (SN), spike density (SD), spike inclination (SI). Agronomic traits included heading date and yield. Disease traits were: FHB-rating, incidence, severity, FHB-index, Fusarium damaged kernels and deoxynivalenol (DON) concentration. There were significant (p < 0.01) differences among genotypes for all morphological traits measured. Moderate heritabilities (0.41–0.66) for spike traits were estimated. Morphological and disease traits were generally negatively correlated; e.g. there was a significant (p < 0.01) negative correlation between spike inclination and DON. GWAS identified 29 significant (p < 0.001) SNPs associated with morphological traits, ranging from − 3.54 to 9.58% of the trait mean. Potential SNPs for yield were located on chromosomes 1B, 3B and 6A. Despite the small effects identified for the SNPs, several morphological traits should be considered for phenotypic and/or genomic selection in FHB resistance breeding programs. Weather and climate forecasts can help agricultural producers improve management choices in anticipation of uncertain growing conditions. Current literature conjectures that the extent to which forecasts are useful depends on their accuracy, that is, the probability with which a forecasted event, such as precipitation, is projected to occur. Too little accuracy can potentially render forecasts effectively useless, even if they convey some form of information. In this study, we collect farmer-based data through a questionnaire and a framed field experiment to test for the existence of an accuracy threshold for forecasts, below which forecasts do not induce any behavioral changes. We do this in the context of a very specific management choice—the timing and amount of nitrogen that Kentucky farmers apply to their wheat in early spring—in response to randomly generated 6-to-10-day forecasts of rainfall conditions. We find that forecasts provide economically significant value to decision-makers only when they depart dramatically from what is normally expected. These results have implications that extend beyond the nitrogen-application decision for winter wheat: if this type of behavior is widespread, at current accuracy levels, other types of forecasts may be of little value to decision-makers and therefore go unheeded. Fusarium head blight (FHB) is an important disease of wheat (Triticum aestivum L.) that has caused billions of dollars in losses in recent decades. Although a massive breeding effort has been undertaken on multiple continents, there are no wheat cultivars with immunity to the disease. Resistance is conditioned by multiple loci and is further complicated by the role of the environment in expression of the disease phenotype. The objectives of our study were (i) to evaluate the phenotypic response to FHB in a large, diverse soft red winter wheat mapping panel; and (ii) to identify promising quantitative trait loci (QTL) associated with FHB resistance based on a genome‐wide association study (GWAS). We evaluated the mapping panel in 2014–2015 and 2015–2016 in an irrigated, inoculated scab nursery near Lexington, KY. Traits evaluated were heading date, plant height, FHB rating, severity, incidence, index, Fusarium‐damaged kernels (FDK), and deoxynivalenol (DON). There were significant (p < 0.05) differences among genotypes for all traits measured. The GWAS (based on 2‐yr entry means) identified 16 significant (p < 0.001) single nucleotide polymorphisms (SNPs) associated with disease traits on multiple chromosomes. Single nucleotide polymorphism association ranged from −2.14 to 4.01% of the mean of a given trait. We detected SNPs associated with FDK and DON on chromosomes 4A, 5B and 6B, and these SNPs decreased DON levels by 1.5, 2.1 and 3.2 mg kg−1, respectively. The majority of studies evaluating genomic selection (GS) for plant breeding have used single‐trait, single‐site models that ignore genotype × environment interaction (GEI) effects. However, such studies do not accurately reflect the complexities of many applied breeding programs, and previous papers have found that models that incorporate GEI effects and multiple traits can increase the accuracy of genomic estimated breeding values (GEBVs). This study's goal was to test GS methods for prediction in scenarios that simulate early‐generation yield testing by correcting for field spatial variation, and fitting multienvironment and multitrait models on data for 14 traits of varying heritability evaluated in unbalanced designs across four environments. Corrections for spatial variation increased across‐environment trait heritability by 25%, on average, but had little effect on model predictive ability. Results between all models were generally equivalent when predicting the performance of newly introduced genotypes. However, models incorporating GEI information and multiple traits increased prediction accuracy by up to 9.6% for low‐heritability traits when phenotypic data were sparsely collected across environments. The results suggest that GS models using multiple traits and incorporating GEI effects may best be suited to predicting line performance in new environments when phenotypic data have already been collected across a subset of the total testing environments. Grain yield is a trait of paramount importance in the breeding of all cereals. In wheat (Triticum aestivum L.), yield has steadily increased since the Green Revolution, though the current rate of increase is not forecasted to keep pace with demand due to growing world population and increasing affluence. While several genome-wide association studies (GWAS) on yield and related component traits have been performed in wheat, the previous lack of a reference genome has made comparisons between studies difficult. In this study, a GWAS for yield and yield-related traits was carried out on a population of 322 soft red winter wheat lines across a total of four rain-fed environments in the state of Virginia using single-nucleotide polymorphism (SNP) marker data generated by a genotyping-by-sequencing (GBS) protocol. Two separate mixed linear models were used to identify significant marker-trait associations (MTAs). The first was a single-locus model utilizing a leave-one-chromosome-out approach to estimating kinship. The second was a sub-setting kinship estimation multi-locus method (FarmCPU). The single-locus model identified nine significant MTAs for various yield-related traits, while the FarmCPU model identified 74 significant MTAs. The availability of the wheat reference genome allowed for the description of MTAs in terms of both genetic and physical positions, and enabled more extensive post-GWAS characterization of significant MTAs. The results indicate a number of promising candidate genes contributing to grain yield, including an ortholog of the rice aberrant panicle organization (APO1) protein and a gibberellin oxidase protein (GA2ox-A1) affecting the trait grains per square meter, an ortholog of the Arabidopsis thaliana mother of flowering time and terminal flowering 1 (MFT) gene affecting the trait seeds per square meter, and a B2 heat stress response protein affecting the trait seeds per head. Breeding for resilience to climate change is a daunting prospect. Crop and climate models tell us that global wheat yields are likely to decline as the climate warms, causing a significant risk to global food security. High temperatures are known to affect crop development yet breeding for tolerance to heat stress is difficult to achieve in field environments. We conducted an active warming study over two years to quantify the effects of heat stress on genetic variation of soft red winter (SRW) wheat (Triticum aestivum L.). Forty SRW cultivars and breeding lines were chosen based on marker genotypes at photoperiod sensitivity and reduced height loci. These genotypes were planted in a randomized complete block design replicated twice across two environments, ambient and artificially warmed. Average heading date occurred 5 days earlier in the warmed environment than in the ambient environment over both years (p ≤ 0.05). On average, grain yield was significantly reduced in the warmed environment by 211.41 kg/ha (p ≤ 0.05) or 4.84%, though we identified 13 genotypes with increased yield in response to warming in both years. Of these genotypes, eight had significantly increased N uptake while six showed significantly increased N utilization efficiency under warming. Under warming, genotypes with wild‐type alleles at the Rht‐D1 locus display significantly greater yields (p ≤ 0.01) and biomass (p ≤ 0.001) than genotypes with reduced height alleles. Of the 13 genotypes with higher (p ≤ 0.01) yields under warming, nine have the wild‐type allele at the Rht‐D1 locus in addition to being photoperiod insensitive. The next steps will be to validate these findings in other populations and to develop an efficient breeding/phenotyping scheme that will lead to more resilient cultivars. Increasing global temperatures will reduce wheat yield and productivity. To identify genetic variation for resilience to warming, we screened a set of diverse wheat genotypes in a warmed and a control environment. On average, grain yield was significantly (p < 0.05) reduced in the warmed environment though we identified genotypes which had higher yields in response to warming in both years. Genomic selection (GS) uses training population (TP) data to estimate the value of lines in a selection population. In breeding, the TP and selection population are often grown in different environments, which can cause low prediction accuracy when the correlation of genetic effects between the environments is low. Subsets of TP data may be more predictive than using all TP data. Our objectives were (i) to evaluate the effect of using subsets of TP data on GS accuracy between environments, and (ii) to assess the accuracy of models incorporating marker X environment interaction (MEI). Two wheat (Triticum aestivum L.) populations were phenotyped for 11 traits in independent environments and genotyped with single-nucleotide polymorphism markers. Within each population- trait combination, environments were clustered. Data from one cluster were used as the TP to predict the value of the same lines in the other cluster(s) of environments. Models were built using all TP data or subsets of markers selected for their effect and stability. The GS accuracy using all TP data was >0.25 for 9 of 11 traits. The between-environment accuracy was generally greatest using a subset of stable and significant markers; accuracy increased up to 48% relative to using all TP data. We also assessed accuracy using each population as the TP and the other as the selection population. Using subsets of TP data or the MEI models did not improve accuracy between populations. Using optimized subsets of markers within a population can improve GS accuracy by reducing noise in the prediction data set. [ABSTRACT FROM AUTHOR]

Summary: Treatment of Burkitt lymphoma (BL) with intensive, multi‐agent chemotherapy with aggressive central nervous system (CNS) prophylaxis results in high cure rates, although no regimen is standard of care. We examined population‐based survival outcomes of adults with BL treated with a modified combination of cyclophosphamide, vincristine, doxorubicin, prednisone and systemic high‐dose methotrexate (MTX) (CODOX‐M) with IVAC (ifosfamide, mesna, etoposide, cytarabine and intrathecal MTX) (CODOX‐M/IVAC) ± rituximab over a 15‐year period in British Columbia. For the 81 patients identified (including 8 with CNS involvement and 18 with human immunodeficiency virus‐associated BL), 5‐year progression‐free survival (PFS) and overall survival (OS) were 75% [95% confidence interval (CI): 63–83%] and 77% (95% CI: 66–85%), respectively, with no treatment‐related deaths. Those who completed the regimen per protocol (n = 38) had significantly improved 5‐year PFS 86% (P = 0·04) and OS 92% (P = 0·008), as did those under 60 years with 5‐year PFS 82% (P = 0·005) and OS 86% (P = 0·002), which remained significant in multivariate analysis [PFS: hazard ratio (HR) 3·36, P = 0·018; OS HR 4·03, P = 0·012]. Incorporation of high‐dose systemic methotrexate also significantly affected multivariate survival outcomes (OS HR 0·28, P = 0·025). Stem cell transplant in first remission had no effect on OS or PFS. This large, real‐world analysis of BL patients treated with CODOX‐M/IVAC ± rituximab demonstrates excellent survival outcomes comparable to clinical trials. Global temperature increases will affect Fusarium head blight (FHB) levels in wheat (Triticum aestivum L.). A pressing question is whether current sources of resistance will be effective in a warmer environment. We evaluated phenotypic response to disease in 238 soft winter wheat breeding lines and cultivars grown in 2015-2016 and 2016-2017 under control and warmed (+3 °C) conditions. Warming was achieved with heating cables buried 3 cm in the rhizosphere. We measured heading date, plant height, yield, FHB rating, Fusarium damaged kernels (FDK), deoxynivalenol (DON), leaf blotch rating, powdery mildew rating and leaf rust rating. There were significant (p < 0.01) differences among genotypes for all traits measured. Genome-wide association study (GWAS) identified 19 and 10 significant SNPs in the control and warmed treatments, respectively. FDK and DON levels were often significantly (p < 0.05) higher in warmed than in control when we contrasted alleles at important quantitative trait locus (QTL) such as Fhb1, Rht-B1 and D1 and all vernalization and photoperiod loci. Increased rhizosphere temperature resulted in a significantly (p < 0.01) earlier heading date (~3.5 days) both years of the study. Rank correlation between warmed and control treatments was moderate (r = 0.56).
