Background: Cardiovascular diseases (CVDs) are the leading cause of death worldwide, and the prevalence of CVDs increases markedly with age. Due to the high energetic demand, the heart is highly sensitive to mitochondrial dysfunction. The complexity of the cardiac mitochondrial proteome hinders the development of effective strategies that target mitochondrial dysfunction in CVDs. Mammalian mitochondria are composed of over 1000 proteins, most of which can undergo post-translational modifications (PTMs). Top-down proteomics is a powerful technique for characterizing and quantifying proteoform sequence variations and PTMs. However, there are still knowledge gaps in the study of age-related mitochondrial proteoform changes using this technique. In this study, we used top-down proteomics to identify intact mitochondrial proteoforms in young and old hearts and determined changes in protein abundance and PTMs in cardiac aging. Methods: Intact mitochondria were isolated from the hearts of young (4-month-old) and old (24-25-month-old) mice. The mitochondria were lysed, and mitochondrial lysates were subjected to denaturation, reduction, and alkylation. For quantitative top-down analysis, there were 12 runs in total arising from 3 biological replicates in two conditions, with technical duplicates for each sample. The collected top-down datasets were deconvoluted and quantified, and then the proteoforms were identified. Results: From a total of 12 LC-MS/MS runs, we identified 134 unique mitochondrial proteins in the different sub-mitochondrial compartments (OMM, IMS, IMM, matrix). 823 unique proteoforms in different mass ranges were identified. Compared to cardiac mitochondria of young mice, 7 proteoforms exhibited increased abundance and 13 proteoforms exhibited decreased abundance in cardiac mitochondria of old mice. Our analysis also detected PTMs of mitochondrial proteoforms, including N-terminal acetylation, lysine succinylation, lysine acetylation, oxidation, and phosphorylation. Conclusion: By combining mitochondrial protein enrichment using mitochondrial fractionation with quantitative top-down analysis using ultrahigh-pressure liquid chromatography (UPLC)-MS and label-free quantitation, we successfully identified and quantified intact proteoforms in the complex mitochondrial proteome. We present the results of an interdisciplinary study conducted at the Pedernales-5 site, situated in the Salar de Pedernales basin (26°S; 3356 masl), dated from 10,510−10,749 to 11,201−11,612 cal. BP. Unlike other Late Pleistocene and Holocene sites in the Andean Puna, where vicuñas (Vicugna vicugna) were the primary prey, at Pedernales-5, the zooarcheological assemblage consists almost exclusively of flamingo (Phoenicopteridae) and bird remains. Through the analysis of lithic and bone artifacts, archeobotanical remains, pigment composition, and paleoenvironmental data, we propose that the emphasis on flamingo exploitation cannot be solely explained by alimentary consumption but also served cultural and symbolic purposes related to the acquisition of feathers, hides, and bones. The unique archeological context of Pedernales-5 offers a distinctive perspective on human dynamics in the Andean highlands during the Early Holocene. [ABSTRACT FROM AUTHOR]

Salinity is a major constraint limiting the yield of tomatoes. However, grafting strategies may help to overcome the salt toxicity of this important horticultural species if appropriate rootstocks are identified. The present study aimed to test a new rootstock, JUPAFORT1, obtained by crossing the glycophyte Solanum lycopersicum (cv. Poncho Negro) with the halophyte wild-related species Solanum chilense to improve the salinity tolerance of the Chilean tomato landrace Old Limachino Tomato (OLT). Intact OLT plants were exposed to 0, 80, or 160 mM of NaCl for 21 days at the vegetative stage and compared with self-grafted (L/L) and Limachino plants grafted on JUPAFORT1 rootstock (L/R) under a completely randomized design. JUPAFORT1 increased OLT scion vigor in the absence of salt but did not significantly increase fresh weight under stress conditions. However, JUPAFORT1 confers to the scion an anisohydric behavior contrasting with the isohydric behavior of L and L/L plants as indicated by measurements of stomatal conductance; L/R plants were able to maintain their metabolic status despite a slight decrease in the leaf's relative water content. JUPAFORT1 rootstock also enabled the maintenance of photosynthetic pigment concentrations in the scion in contrast to L and L/L plants, which exhibited a decrease in photosynthetic pigments under stress conditions. L/R plants encountered oxidative stress at the highest stress intensity (160 mM of NaCl) only, while L and L/L plants suffered from oxidative damage at a lower dose (80 mM of NaCl). L/R plants behaved as includer plants and did not sequester Na+ in the root system, in contrast to L and L/L, which behaved as excluder plants retaining Na+ in the root system to avoid its translocation to the shoots. The expression of genes coding for ion transporters (HKT1.1, HKT1.2, LKT1, SKOR, SOS2, and SOS3) in the root system was not modified by salinity in L/R. In contrast, their expression varied in response to salinity in L and L/L. Overall, L/R plants exhibited higher physiological stability than L/L or L plants in response to an increasing NaCl dose and did not require additional energy investment to trigger an adaptative response to salinity. This suggests that the constitutive salinity tolerance of the halophyte S. chilense was maintained in the interspecific rootstock. JUPAFORT1 issued from S. lycopersicum x S. chilense may thus improve salt-stress resilience in OLT tomatoes. MicroRNAs (miRNAs) exert their gene regulatory effects on numerous biological processes based on their selection of target transcripts. Current experimental methods available to identify miRNA targets are laborious and require millions of cells. Here we have overcome these limitations by fusing the miRNA effector protein Argonaute2 to the RNA editing domain of ADAR2, allowing the detection of miRNA targets transcriptome-wide in single cells. miRNAs guide the fusion protein to their natural target transcripts, causing them to undergo A>I editing, which can be detected by sensitive single-cell RNA sequencing. We show that agoTRIBE identifies functional miRNA targets, which are supported by evolutionary sequence conservation. In one application of the method we study microRNA interactions in single cells and identify substantial differential targeting across the cell cycle. AgoTRIBE also provides transcriptome-wide measurements of RNA abundance and allows the deconvolution of miRNA targeting in complex tissues at the single-cell level. Objectives: Investigating if the proportion of fixed assets over total assets is positively associated with the asymmetric cost behavior of public and private hospitals in Brazil. Methods: In order to test the sticky cost phenomenon in a different sector of companies and industries, we used panel data regression to investigate the asymmetric cost behavior in Brazilian hospitals, analyzing the hospital cost behavior regarding the variation in revenues and verifying whether the proportion of fixed assets over total assets is positively associated with the asymmetric cost behavior. As a result, this research took the findings obtained by the models applied to data from the 101 hospitals comprising the sample, spread over the 2010–2019 period. The research was divided into four sections. The first section tested asymmetry for fixed assets over total assets for hospitals in general. The second section divided the sample into public and private hospitals. The third section analyzed the sample of conglomerates against a single hospital. Finally, the fourth section tested the asymmetry of the hospitals in the sample measured by the number of beds. Results: The evidence documented here partially confirms the results of literature on the existence of asymmetric cost behavior regarding variations in revenue. The H1 hypothesis that the proportion of fixed assets over total assets is positively associated with the asymmetric cost behavior was confirmed, especially for private and small hospitals regarding fixed assets. Disease severity and drought due to climate change present significant challenges to orchard productivity. This study examines the effects of spring inoculation with Pseudomonas syringae pv. syringae (Pss) on sweet cherry plants, cvs. Bing and Santina with varying defense responses, assessing plant growth, physiological variables (water potential, gas exchange, and plant hydraulic conductance), and the levels of abscisic acid (ABA) and salicylic acid (SA) under two summer irrigation levels. Pss inoculation elicited a more pronounced response in 'Santina' compared to 'Bing' at 14 days post-inoculation (dpi), and those plants inoculated with Pss exhibited a slower leaf growth and reduced transpiration compared to control plants during 60 dpi. During differential irrigations, leaf area was reduced 14% and 44% in Pss inoculated plants of 'Bing' and 'Santina' respectively, under well-watered (WW) conditions, without changes in plant water status or gas exchange. Conversely, water-deficit (WD) conditions led to gas exchange limitations and a 43% decrease in plant biomass compared to that under WW conditions, with no differences between inoculation treatments. ABA levels were lower under WW than under WD at 90 dpi, while SA levels were significantly higher in Pss-inoculated plants under WW conditions. These findings underscore the influence on plant growth during summer in sweet cherry cultivars that showed a differential response to Pss inoculations and how the relationship between ABA and SA changes in plant drought level responses. Epitranscriptomics is a field that delves into post-transcriptional changes. Among these modifications, the conversion of adenosine to inosine, traduced as guanosine (A>I(G)), is one of the known RNA-editing mechanisms, catalyzed by ADARs. This type of RNA editing is the most common type of editing in mammals and contributes to biological diversity. Disruption in the A>I(G) RNA-editing balance has been linked to diseases, including several types of cancer. Drug resistance in patients with cancer represents a significant public health concern, contributing to increased mortality rates resulting from therapy non-responsiveness and disease progression, representing the greatest challenge for researchers in this field. The A>I(G) RNA editing is involved in several mechanisms over the immunotherapy and genotoxic drug response and drug resistance. This review investigates the relationship between ADAR1 and specific A>I(G) RNA-edited sites, focusing particularly on breast cancer, and the impact of these sites on DNA damage repair and the immune response over anti-cancer therapy. We address the underlying mechanisms, bioinformatics, and in vitro strategies for the identification and validation of A>I(G) RNA-edited sites. We gathered databases related to A>I(G) RNA editing and cancer and discussed the potential clinical and research implications of understanding A>I(G) RNA-editing patterns. Understanding the intricate role of ADAR1-mediated A>I(G) RNA editing in breast cancer holds significant promise for the development of personalized treatment approaches tailored to individual patients' A>I(G) RNA-editing profiles. Antimicrobial resistance has been stated to be a global health problem. In Chile, the use of antibiotics should be declared by medical prescription, but it is unknown what happens to the drugs once the treatment ends. Among the possibilities for their disposal are the trash or the drain; regardless of which scenario arises, antibiotics could accumulate in the environment, stimulating the emergence of antimicrobial resistance mechanisms and their transfer between microorganisms. Unfortunately, sometimes wastewater ends up in bodies of water, due to the dragging of elements by rain, or by the presence of illegal water discharges. In this work, shotgun metagenomics was used to elucidate the functional and microbial composition of biohazard elements in the bay of Puerto Varas City, Chile. As expected, a high diversity of microorganisms was found, including bacterial elements described as human or animal pathogens. Also, a diverse repertory of antimicrobial resistant genes (ARGs) was detected, which confers mainly resistance to macrolides, beta-lactams, and tetracyclines, consistent with the families of antibiotics most used in Chile. Similar ARGs were identified in DNA mobile elements. In addition, we tested the antimicrobial susceptibility in 14 bacterial strains isolated from Llanquihue Lake. This is the first report of the presence of genomic elements that could constitute a health problem, considering the importance of the interconnection between environmental, animal, and human health, a concept known as One Health. Simple Summary: Sortilin is a protein with tumour-suppressor activity. Among other functions, sortilin moves around other proteins in the cell and also out of it by leading them to small vesicles termed exosomes. The aim of this study was to establish the degree of abnormal expression of the gene in non-small cell lung cancer. We show here that the alternative forms of the gene are affected both qualitatively and quantitatively. In addition, the ratio of their expression changes. We demonstrate that expression changes of the SORT1 form are due to DNA methylation of its regulatory region. As sortilin is involved in trafficking EGFR, a known target of current drugs, it is possible that these alterations, beyond their involvement in cancer development, may be predictive of the response to these therapies. Sortilin is an important regulator with potential tumour-suppressor function by limiting EGFR signalling. In this study, we undertook a comprehensive expression analysis of sortilin transcript variants and the DNA methylation status of their corresponding promoters in human non-small cell carcinomas (NSCLCs). RNA/DNA was extracted from 81 NSCLC samples and paired normal tissue. mRNA expression was measured by qPCR and DNA methylation determined by pyrosequencing. BigDye-terminator sequencing was used to confirm exon-8 alternative splicing. Results demonstrated that both SORT1A and SORT1B variants were downregulated in lung tumours. The SORT1A/SORT1B expression ratio was higher in tumours compared to normal tissue. SORT1B promoter hypermethylation was detected in lung tumours compared to normal lung (median difference 14%, Mann–Whitney test p = 10−6). Interestingly, SORT1B is hypermethylated in white blood cells, but a small and very consistent drop in methylation (6%, p = 10−15) was observed in the lung cancer cases compared to control subjects. We demonstrate that the SORT1B exon-8 splice variation, reported in sequence databases, is also a feature of SORT1A. The significantly altered quantitative and qualitative characteristics of sortilin mRNA in NSCLC indicate a significant involvement in tumour pathogenesis and may have significant impact for its utility as a predictive marker in lung cancer management. Introduction: Although stentrievers (SRs) have been a mainstay of mechanical thrombectomy (MT), and current guidelines recommend the use of SRs in the treatment of large vessel occlusion stroke (LVO), there is a paucity of studies in the literature comparing SRs directly against each other in terms of mechanical and functional properties. Timely access to endovascular therapy and the ability to restore intracranial flow in a safe, efficient, and efficacious manner have been critical to the success of MT. This study aimed to investigate the impact of contemporary SR characteristics, including model, brand, size, and length, on the first-pass effect (FPE) in patients with acute ischemic stroke. Methods: Consecutive patients with M1 occlusion treated with a single SR+BGC were recruited from the ROSSETTI registry. The primary outcome was the FPE that was defined as modified (mFPE) or true (tFPE) for the achievement of modified thrombolysis in cerebral infarction (mTICI) grades 2b-3 or 3 after a single device pass, respectively. We compared patients who achieved mFPE with those who achieved tFPE according to SR characteristics. Results: We included 610 patients (52.3% female and 47.7% male, mean age 75.1 ± 13.62 years). mFPE was achieved in 357 patients (58.5%), whereas tFPE was achieved in 264 (43.3%). There was no significant association between SR characteristics and mFPE or tFPE. Specifically, the SR size did not show a statistically significant relationship with improvement in FPE. Similarly, the length of the SR did not yield significant differences in the mFPE and tFPE, even when the data were grouped. Conclusions: Our data indicate that contemporary SR-mediated thrombectomy characteristics, including model, brand, size, and length, do not significantly affect the FPE. [ABSTRACT FROM AUTHOR]

This editorial explores the use of rootstocks in crops and fruit trees as a means to address the challenges posed by climate change and abiotic stress in agriculture. By grafting plants onto tolerant rootstocks, growers can enhance crop resilience and reduce the need for resources like water and fertilizers. The selection of suitable rootstocks is a complex process that involves considering various root-related traits. The editorial also delves into the physiological and molecular aspects of tolerance to different types of abiotic stresses associated with the use of rootstocks. The document discusses the identification of significant QTLs related to tolerance to iron chlorosis in Prunus plants, with the goal of developing tools and biotechnological technologies to improve the performance of plants grafted on stress-tolerant rootstocks. Tropical forest management has a potential role in forest conservation if it is sustainable. This study of a forest under management in Bolivian Amazonia strongly suggests that the management project is not sustainable and that no potential changes in management would be likely to make it so. In a 216.41 ha harvested area, 278 commercial trees from 10 families, 15 genera, and 15 species were measured. The density of commercial species with diameter at breast height (DBH) ≥ 50 cm was 1.28 trees ha−1, and the harvestable commercial volume was 12.40 m3 ha−1. Due to market restrictions, the actual amounts harvested were much lower: 96 trees were harvested with commercial boles totaling 2.7 m3 ha−1. Of the total impact on biomass and carbon (above- and belowground), the logs removed from the area represented only 13.4%, while 86.6% was from losses in the forest as follows: 14.5% from the stumps, crowns, and roots of harvested trees (DBH ≥ 50 cm) plus 72.1% from the trees (DBH ≥ 10 cm) in the forest lost to roads, log landings, and skid tracks and the gap openings caused by felling the harvested trees. The estimated expenses exceeded the gross revenue of the management company (USD 519.15 ha−1), a fact confirmed by the company's subsequent bankruptcy. The project's low harvest intensity reduces the environmental impact per hectare but increases the impact per cubic meter of wood harvested because producing a given volume of wood requires disturbing a larger area and because more kilometers of access roads and skid tracks have to be installed to extract a given volume of wood. Because many costs are fixed regardless of harvest intensity, small harvest volume can render such projects financially unfeasible, increasing the likelihood that they will be abandoned and not provide long-term "sustainable" forest protection. However, this does not mean that higher harvest intensity results in sustainability, as other constraints apply to high-intensity projects. Simple Summary: In this study, it is described that embryos produced in vitro release nanoparticles charged with fragments of embryonic DNA. These nanoparticles, alongside their DNA content, can be recovered from the liquid medium where the embryos develop in vitro and used to assess the genetics of embryos. The analysis of gene markers is a common practice in clinics for selecting human embryos without genomic errors. In animals, it can help in selecting embryos with high genetic value. So far, this has been achieved using embryonic biopsy to collect several cells; however, this is invasive due to the manipulation of the embryo. In this sense, the secreted nanoparticles containing DNA could stand as samples for genetic analysis, without disturbing the embryo. The main concern is whether these particles are mainly released by low-quality embryos that will eventually die. This work aimed to evaluate whether the rate of cell death in the embryo affects the release of particles containing DNA. It was confirmed that viable and good-quality embryos secrete particles with DNA in the same way as low-quality embryos; therefore, they represent a suitable alternative for embryonic genetic diagnosis, avoiding the invasive nature and technical complexity of a biopsy. Pre-implantation embryos release extracellular vesicles containing different molecules, including DNA. The presence of embryonic DNA in E-EVs released into the culture medium during in vitro embryo production could be useful for genetic diagnosis. However, the vesicles containing DNA might be derived from embryos suffering from apoptosis, i.e., embryos of bad quality. This work intended to confirm that embryos release DNA that is useful for genotyping by evaluating the effect of embryonic apoptosis on DNA content in E-EVs. Bovine embryos were produced by parthenogenesis and in vitro fertilization (IVF). On Day 5, morulae were transferred to individual cultures in an EV-depleted SOF medium. On Day 7, embryos were used to evaluate cellular apoptosis, and each culture medium was collected to evaluate E-EV concentration, characterization, and DNA quantification. While no effect of the origin of the embryo on the apoptotic rate was found, arrested morulae had a higher apoptotic rate. E-EVs containing DNA were identified in all samples, and the concentration of those vesicles was not affected by the origin or quality of the embryos. However, the concentration of DNA was higher in EVs released by the arrested parthenogenetic embryos. There was a correlation between the concentration of E-EVs, the concentration of DNA-positive E-EVs, and the concentration of DNA. There was no negative effect of apoptotic rate on DNA-positive E-EVs and DNA concentration; however, embryos of the best quality with a low apoptotic rate still released EVs containing DNA. This study confirms that the presence of DNA in E-EVs is independent of embryo quality. Therefore, E-EVs could be used in liquid biopsy for noninvasive genetic diagnosis. [ABSTRACT FROM AUTHOR]

Dysregulated A>I(G) RNA editing, which is mainly catalyzed by ADAR1 and is a type of post-transcriptional modification, has been linked to cancer. A low response to therapy in breast cancer (BC) is a significant contributor to mortality. However, it remains unclear if there is an association between A>I(G) RNA-edited sites and sensitivity to genotoxic drugs. To address this issue, we employed a stringent bioinformatics approach to identify differentially RNA-edited sites (DESs) associated with low or high sensitivity (FDR 0.1, log2 fold change 2.5) according to the IC50 of PARP inhibitors, anthracyclines, and alkylating agents using WGS/RNA-seq data in BC cell lines. We then validated these findings in patients with basal subtype BC. These DESs are mainly located in non-coding regions, but a lesser proportion in coding regions showed predicted deleterious consequences. Notably, some of these DESs are previously reported as oncogenic variants, and in genes related to DNA damage repair, drug metabolism, gene regulation, the cell cycle, and immune response. In patients with BC, we uncovered DESs predominantly in immune response genes, and a subset with a significant association (log-rank test p < 0.05) between RNA editing level in LSR, SMPDL3B, HTRA4, and LL22NC03-80A10.6 genes, and progression-free survival. Our findings provide a landscape of RNA-edited sites that may be involved in drug response mechanisms, highlighting the value of A>I(G) RNA editing in clinical outcomes for BC. Mycorrhizal symbioses (mycorrhizas) of Ericaceae, including ericoid mycorrhiza (ErM), have been mainly studied in the Northern Hemisphere, although the highest diversity of ericaceous plants is located in the Southern Hemisphere, where several regions remain largely unexplored. One of them is South America, which harbors a remarkably high diversity of Ericaceae (691 species and 33 genera) in a wide range of environmental conditions, and a specific mycorrhizal type called cavendishioid. In this review, we compile all available information on mycorrhizas of Ericaceae in South America. We report data on the mycorrhizal type and fungal diversity in 17 and 11 ericaceous genera, respectively. We show that South American Ericaceae exhibit a high diversity of habitats and life forms and that some species from typical ErM subfamilies may also host arbuscular mycorrhiza. Also, a possible geographical pattern in South American ErM fungal communities is suggested, with Sebacinales being the dominant mycorrhizal partners of the Andean clade species from tropical mountains, while archetypal ErM fungi are common partners in southern South America species. The gathered information challenges some common assumptions about ErM and suggests that focusing on understudied regions would improve our understanding of the evolution of mycorrhizal associations in this intriguing family. The distribution of copper, iron, and sulfur during the oxidation of La Caridad copper concentrate particles under simulated flash smelting conditions was studied in a laboratory reactor. Six wet-sieved size fractions and the unsieved copper concentrate were oxidized at 1123 K and 40% and 70% O2 by volume in the process gas during the experiments. Samples of partially oxidized particles were collected at 0.2, 0.8, and 0.9 m from the point of entry and analyzed in a QEMSCAN® unit to determine the elemental composition within the population of particles. The distribution of the major elements during oxidation was strongly dependent upon the size and chemical composition of the initial particles. Overall, the copper content tended to increase and sulfur content decreased along the reactor length within all sizes. In contrast, the distribution of iron did not follow a general trend, as it was found to increase, decrease, or remain unchanged depending on the particle size. This finding may represent a key feature to further investigate the reaction path followed by particles during flash smelting, especially those associated with particle fragmentation. In general, the larger the particle size was, the larger the change in the content of the major elements within the particle population. Based on the experimental results, particles within a size fraction of <45 µm tended to follow a reaction path consisting of rapid melting followed by the collision and coalescence of reacting droplets during flight. In contrast, particles within the fraction of 45–53 µm tended to react individually. The oxidation behavior of the unsieved concentrate particles showed a combination of both reaction paths. The negative impacts of climate change on native forest ecosystems have created challenging conditions for the sustainability of natural forest regeneration. These challenges arise primarily from abiotic stresses that affect the early stages of forest tree development. While there is extensive evidence on the diversity of juvenile microbial symbioses in agricultural and fruit crops, there is a notable lack of reports on native forest plants. This review aims to summarize the critical studies conducted on the diversity of juvenile plant–microbe interactions in forest plants and to highlight the main benefits of beneficial microorganisms in overcoming environmental stresses such as drought, high and low temperatures, metal(loid) toxicity, nutrient deficiency, and salinity. The reviewed studies have consistently demonstrated the positive effects of juvenile plant–microbiota interactions and have highlighted the potential beneficial attributes to improve plantlet development. In addition, this review discusses the beneficial attributes of managing juvenile plant–microbiota symbiosis in the context of native forest restoration, including its impact on plant responses to phytopathogens, promotion of nutrient uptake, facilitation of seedling adaptation, resource exchange through shared hyphal networks, stimulation of native soil microbial communities, and modulation of gene and protein expression to enhance adaptation to adverse environmental conditions. One of the causal agents of bacterial canker is Pseudomonas amygdali pv. morsprunorum—Pam (formerly Pseudomonas syringae pv. morsprunorum). Recently detected in Chile, Pam is known to cause lesions in the aerial parts of the plant, followed by more severe symptoms such as cankers and gummosis in the later stages of the disease. This study presents the design of PCR and LAMP detection methods for the specific and sensitive identification of Pseudomonas amygdali pv. morsprunorum (Pam) from cherry trees. Twelve Pseudomonas isolates were collected, sequenced, and later characterized by Multi-locus Sequence Analysis (MLSA) and Average Nucleotide Identity by blast (ANIb). Three of them (11116B2, S1 Pam, and S2 Pam) were identified as Pseudomonas amygdali pv. morsprunorum and were used to find specific genes through RAST server, by comparing their genome with that of other Pseudomonas, including isolates from other Pam strains. The effector gene HopAU1 was selected for the design of primers to be used for both techniques, evaluating sensitivity and specificity, and the ability to detect Pam directly from plant tissues. While the PCR detection limit was 100 pg of purified bacterial DNA per reaction, the LAMP assays were able to detect up to 1 fg of purified DNA per reaction. Similar results were observed using plant tissues, LAMP being more sensitive than PCR, including when using DNA extracted from infected plant tissues. Alternative lengthening of telomeres (ALT) is a telomere maintenance mechanism activated in ~10–15% of cancers, characterized by telomeric damage. Telomeric damage-induced long non-coding RNAs (dilncRNAs) are transcribed at dysfunctional telomeres and contribute to telomeric DNA damage response (DDR) activation and repair. Here we observed that telomeric dilncRNAs are preferentially elevated in ALT cells. Inhibition of C-rich (teloC) dilncRNAs with antisense oligonucleotides leads to DNA replication stress responses, increased genomic instability, and apoptosis induction selectively in ALT cells. Cell death is dependent on DNA replication and is increased by DNA replication stress. Mechanistically, teloC dilncRNA inhibition reduces RAD51 and 53BP1 recruitment to telomeres, boosts the engagement of BIR machinery, and increases C-circles and telomeric sister chromatid exchanges, without increasing telomeric non-S phase synthesis. These results indicate that teloC dilncRNA is necessary for a coordinated recruitment of DDR factors to ALT telomeres and it is essential for ALT cancer cells survival. ALT cells use an alternative lengthening mechanism of telomeres and bear telomeric DNA damage with increased levels of damage-induced long non-coding RNA. Faunal remains from archeological sites worldwide, especially feces and regurgitated pellets, are usually subjected to paleoparasitological examination, allowing the identification of past animal parasites. In this study, we analyzed 10 samples of South American camelid dung, which played an important role for the ancient human groups in the South‐Central Andes, dated between 341 and 1635 calCE, from the Iluga Túmulos site (Pampa del Tamarugal, Tarapacá region, Atacama Desert, Chile). Microscopy examination revealed parasite remains in seven samples, in which oocysts of Eimeria macusaniensis and eggs of Lamanema chavezi/Nematodirus lamae, Trichuris sp., Moniezia sp., capillariids, strongylids, and unidentified nematodes were found. Although some of these species pose a potential health risk to camelids, most are not considered to be of major zoonotic importance, which could explain the absence of these parasites on human coprolites from this site analyzed previously. This first paleoparasitological study on camelid fecal remains from northern Chile sheds light on the occurrence of gastrointestinal parasites and its relationship with Andean ancient human populations and the environment. Background: Achieving and maintaining a low-risk profile is associated with favorable outcome in pulmonary arterial hypertension (PAH). The effects of treatment on risk profile are variable among patients. Objective: To Identify variables that might predict the response to treatment with phosphodiesterase-5 inhibitors (PDE-5i) in PAH. Methods: We carried out a cohort analysis of the Spanish PAH registry in 830 patients diagnosed with PAH that started PDE5i treatment and had > 1 year follow-up. 644 patients started PDE-5i either in mono- or add-on therapy and 186 started combined treatment with PDE-5i and endothelin receptor antagonist (ERA). Responders were considered when at 1 year they: (1) were alive; (2) did not present clinical worsening; and (3) improved European Society of Cardiology/European Respiratory Society (ESC/ERS) risk score or remained in low-risk. Univariate and multivariate logistic regression models were used to analyze variables associated with a favorable response. Results: Two hundred and ten patients (33%) starting PDE-5i alone were classified as responders, irrespective of whether it was mono- or add-on therapy. In addition to known predictors of PAH outcome (low-risk at baseline, younger age), male sex and diagnosis of portopulmonary hypertension (PoPH) or HIV-PAH were independent predictors of favorable response to PDE-5i. Diffusing capacity for carbon monoxide (DLco) ≤ 40% of predicted was associated with an unfavorable response. When PDE-5i were used in upfront combination, 58% of patients were responders. In this group, diagnosis of idiopathic PAH (IPAH) was an independent predictor of favorable response, whereas connective tissue disease-PAH was associated with an unfavorable response. Conclusion: Male sex and diagnosis of PoPH or HIV-PAH are predictors of favorable effect of PDE-5i on risk profile when used as mono- or add-on therapy. Patients with IPAH respond more favorably to PDE-5i when used in upfront combination. These results identify patient profiles that may respond favorably to PDE-5i in monotherapy and those who might benefit from alternative treatment strategies. [ABSTRACT FROM AUTHOR]

En el presente artículo se realiza el análisis filológico del volumen 3169 de la Biblioteca Nacional de España en el cual se encuentran foliados importantes manuscritos incunables de la cultura andina. En primer lugar, se revisan algunos de los fenómenos textuales que se repiten en los márgenes a lo largo de todo el volumen, proponiendo hipótesis inter-pretativas sobre su aparición y uso. Luego, se reflexiona acerca de las fechas y autoría de los manuscritos, distinguiendo aquellas reales de las que se ha propuesto a partir de datos contextuales con distinto grado de fiabilidad. Finalmente, se discute una hipótesis sobre el proyecto textual del volumen en cuestión. The present article performs a philological analysis of volume 3169 of the National Library of Spain, wherein numbered folios of incunabula of Andean culture can be found. First, some of the textual phenomena that recur throughout volume 3169 are examined and interpretive hypotheses about their appearance and use are proposed. Second, the dates and authorship of the manuscripts are discussed, distinguishing between those actually known and those that have been deduced from contextual data with varying degrees of trustworthiness. This article reports the first case of Rhizobium rhizogenes causing crown gall disease on blueberry and cherry plants in Chile. Crown gall is a significant disease that affects fruit trees and can cause substantial losses in orchards and nurseries. Previous reports in Chile have identified other species of Agrobacterium and Rhizobium causing crown gall, but R. rhizogenes had only been associated with hairy root disease. The researchers isolated and tested twelve bacteria from gall samples, and only two isolates of R. rhizogenes induced tumors in tomato, kalanchoe, blueberry, and cherry plants. Genomic analysis confirmed the identity of the isolates as R. rhizogenes, and further analysis revealed the presence of a Ti plasmid, which is associated with tumor induction. This finding expands our understanding of the pathogenicity of R. rhizogenes and its impact on fruit trees in Chile. [Extracted from the article]

Photoactive nanoparticles are used to reduce microbial colonization and self-clean surfaces of materials such as glass and ceramics. To test whether such an approach is feasible for wood surfaces, we treated radiata pine samples with TiO2 (rutile and anatase) or ZnO nanoparticles and then coated the samples with different finishes. Coated samples and uncoated controls were exposed outdoors for six months. After exposure, fungi colonizing wood surfaces were identified using molecular techniques and microscopy, and colour changes in the wood samples were also measured. Treatment of uncoated surfaces with nanoparticles reduced the discolouration of wood during weathering but had little effect on colonization of wood by black mould fungi. In contrast, pretreatment of samples with titanium dioxide nanoparticles increased the number and diversity of fungi including basidiomycetes colonizing coated samples, whereas zinc oxide nanoparticles had the opposite effect. Zinc oxide nanoparticles, however, were less effective than rutile titanium dioxide nanoparticles at reducing the discolouration of coated samples exposed to natural weathering. We conclude that none of the photoactive nanoparticles on their own are able to reduce microbial colonization and discoloration of samples. The European Union aims for its existing building stock to be highly energy-efficient and decarbonized by 2050 through long-term renovation strategies so that all residential buildings are nearly zero-energy buildings. The objective of this work is to determine the optimal energy renovation solution for rural residential buildings located in cold climate zones of Spain to achieve nearly zero-energy buildings. For this purpose, the energy, environmental and economic impacts of 48 energy renovation proposals in three different climate zones are assessed, taking La Rioja as a case study. Considering these impacts, the optimal solution is a solution that improves the thermal envelope, applying the life cycle cost analysis, and that uses renewable energy sources to meet thermal needs and a portion of the electrical energy needs. Under the optimal solution, overall savings of up to EUR 2.4 can be achieved for each euro invested, resulting in reductions in non-renewable primary energy consumption by up to 97%, total primary energy consumption by up to 81% and CO2 emissions by up to 97%. Chilling injury is a physiological disorder caused by cold storage in peaches and nectarines. The main symptom of chilling injury is mealiness/wooliness, described as a lack of juice in fruit flesh. In this work, we studied two nectarine varieties (Andes Nec-2 and Andes Nec-3) with contrasting susceptibility to mealiness after cold storage. A non-targeted metabolomic analysis was conducted by GC-MS to understand if changes in metabolite abundance are associated with nectarine mealiness induced by cold storage. Multivariate analyses indicated that in unripe nectarines, cold storage promoted a higher accumulation of amino acids in both varieties. Interestingly, for ripe nectarines, cold storage induced an accumulation of fewer amino acids in both varieties and showed an increased abundance of sugars and organic acids. A pathway reconstruction of primary metabolism revealed that in ripe nectarines, cold storage disrupted metabolite abundance in sugar metabolism and the TCA cycle, leading to a differential accumulation of amino acids, organic acids, and sugars in mealy and juicy nectarines. Dravet syndrome (DS) is an epileptic encephalopathy caused by mutations in the Scn1a gene encoding the α1 subunit of the Nav1.1 sodium channel, which is associated with recurrent and generalized seizures, even leading to death. In experimental models of DS, histological alterations have been found in the brain; however, the retina is a projection of the brain and there are no studies that analyze the possible histological changes that may occur in the disease. This study analyzes the retinal histological changes in glial cells (microglia and astrocytes), retinal ganglion cells (RGCs) and GABAergic amacrine cells in an experimental model of DS (Syn-Cre/Scn1aWT/A1783V) compared to a control group at postnatal day (PND) 25. Retinal whole-mounts were labeled with anti-GFAP, anti-Iba-1, anti-Brn3a and anti-GAD65/67. Signs of microglial and astroglial activation, and the number of Brn3a+ and GAD65+67+ cells were quantified. We found retinal activation of astroglial and microglial cells but not death of RGCs and GABAergic amacrine cells. These changes are similar to those found at the level of the hippocampus in the same experimental model in PND25, indicating a relationship between brain and retinal changes in DS. Total lymphoid irradiation (TLI) is an alternative treatment for chronic lung allograft dysfunction (CLAD). However, data regarding its efficacy and tolerance are scarce. This study included patients with CLAD treated with TLI at our center between 2011 and 2018. Clinical characteristics before and after TLI and related complications were analyzed. Forty patients with CLAD (twenty‐nine bronchiolitis obliterans syndrome [BOS], nine restrictive allograft syndrome [RAS], and two mixed) were included. Significant attenuation of the forced expiratory volume in 1‐sec (FEV1) decline slope was observed in all phenotypes, in both the BOS and RAS. The median FEV1 12, 6, and 3 months pre‐TLI were as follows: 1980 (IQR 1720‐2560), 1665 (IQR 1300‐2340) and 1300 (IQR 1040‐1740) ml (p <.001), while the median FEV1 at 3, 6, and 12 months post‐TLI was 1110 (IQR 810–1440), 1130 (IQR 860–1470), and 1115 (IQR 865–1490) ml (p =.769). No dropouts due to radiation toxicity were observed. The mean survival according to the Karnofsky Performance Status Scale (KPS) >70 or ≤70 at baseline was 1837 (IQR 259–2522) versus 298 (IQR 128–554) days (p <.0001), respectively. In conclusion, TLI may stop FEV1 decline in both BOS and RAS. Moreover, a good KPS score may be an important prognostic factor. [ABSTRACT FROM AUTHOR]
