Global warming is a major challenge to the sustainable and humane production of food because of the increased risk of livestock to heat stress. Here, the example of the prolactin receptor (PRLR) gene is used to demonstrate how gene editing can increase the resistance of cattle to heat stress by the introduction of mutations conferring thermotolerance. Several cattle populations in South and Central America possess natural mutations in PRLR that result in affected animals having short hair and being thermotolerant. CRISPR/Cas9 technology was used to introduce variants of PRLR in two thermosensitive breeds of cattle – Angus and Jersey. Gene‐edited animals exhibited superior ability to regulate vaginal temperature (heifers) and rectal temperature (bulls) compared to animals that were not gene‐edited. Moreover, gene‐edited animals exhibited superior growth characteristics and had larger scrotal circumference. There was no evidence for deleterious effects of the mutation on carcass characteristics or male reproductive function. Although the beneficial effect of biostimulation on reproduction has been reported, the influence of selectivity and social factors on the response to biostimulation has not received sufficient research attention in both Bos indicus and Bos indicus influenced cattle. Furthermore, 'green and cheap' strategies to improve cattle reproduction are currently in demand while Bos indicus influenced cattle with inferior reproductive performance, and farmers with economic limitations are common in tropical zones. Hence, to assess the reproductive response of crossbred taurus × indicus cows to biostimulation by pre‐pubertal (PPM) or pubertal (PM) teasers males, two trials of 2 years each were conducted. Trial 1 n = 187 cows (Year 1:85 cows exposed to PPM and Year 2:102 cows exposed to PM). Trial 2 n = 196 cows (Year 1:101 cows exposed to PPM and Year 2:95 cows exposed to PM). The effect of exposing cows to PPM and PM on the intervals calving to first service (ICFS), calving to conception (ICC) and economic cost of days open (ECDO) was analysed using Kruskal–Wallis ANOVA and the effect of exposing cows to PPM and to PM on reproductive status at 90 days (RS90) and proportion of cows requiring hormonal protocols (PRH) was compared using χ2 analysis. Both ICFS and ICC were shorter (p <.0001) for PM‐exposed females (96.12 ± 4.1 and 110.93 ± 2.9 days; respectively) compared with those PPM‐exposed (134.41 ± 3.3 and 135.64 ± 2.4 days; respectively). With RS90, more (p <.0001) PM‐exposed cows (50.7%) were pregnant compared with PPM‐exposed cows (16.1%). The PRH was greater (p <.0001) in PPM‐exposed cows (79.0%) compared with PM‐exposed (27.9%). The ECDO was less (p <.0001) in PM‐exposed cows (US$ 142.9 ± 3.8) compared with PPM‐exposed (US$ 176.3 ± 2.9). In conclusion, cows exposed to PM had shorter ICFS and ICC compared with cows exposed to PPM. More cows exposed to PM were pregnant after 90 days, and PRH was less than cows exposed to PPM. Background: Massive blood transfusion (MBT) following older adult trauma poses unique challenges. Despite extensive evidence on optimal resuscitative strategies in the younger adult patients, there is limited research in the older adult population. Methods: We used the Trauma Quality Improvement Program (TQIP) database from 2013 to 2017 to identify all patients over 65 years old who received a MBT. We stratified our population into six fresh‐frozen plasma:packed red blood cell (FFP:pRBC) ratio cohorts (1:1, 1:2, 1:3, 1:4, 1:5, 1:6+). Our primary outcomes were 24‐h and 30‐day mortality. We constructed multivariable regression models with 1:1 group as the baseline and adjusted for confounders to estimate the independent effect of blood ratios on mortality. Results: A total of 3134 patients met our inclusion criteria (median age 73 ± 7.6 years, 65% male). On risk‐adjusted multivariable analysis, 1:1 FFP:pRBC ratio was independently associated with lowest 24‐h mortality (1:2 odds ratio [OR] 1.60, 95% confidence interval [CI] 1.25–2.06, p < 0.001) and 30‐day mortality (1:2 OR 1.44, 95% CI 1.15–1.80, p = 0.002). Conclusions: Compared to all other ratios, the 1:1 FFP:pRBC ratio had the lowest 24‐h and 30‐day mortality following older adult trauma consistent with findings in the younger adult population. During lockdowns associated with the COVID-19 pandemic, individuals have experienced poor sleep quality and sleep regularity, changes in lifestyle behaviours, and heightened depression and anxiety. However, the inter-relationship and relative strength of those behaviours on mental health outcomes is still unknown. We collected data between 12 May and 15 June 2020 from 1048 South African adults (age: 32.76 ± 14.43 years; n = 767 female; n = 473 students) using an online questionnaire. Using structural equation modelling, we investigated how insomnia symptoms, sleep regularity, exercise intensity/frequency and sitting/screen-use (sedentary screen-use) interacted to predict depressive and anxiety-related symptoms before and during lockdown. We also controlled for the effects of sex and student status. Irrespective of lockdown, (a) more severe symptoms of insomnia and greater sedentary screen-use predicted greater symptoms of depression and anxiety and (b) the effects of sedentary screen-use on mental health outcomes were mediated by insomnia. The effects of physical activity on mental health outcomes, however, were only significant during lockdown. Low physical activity predicted greater insomnia symptom severity, which in turn predicted increased depressive and anxiety-related symptoms. Overall, relationships between the study variables and mental health outcomes were amplified during lockdown. Objective: The impact of food insecurity on mental health has not yet been examined in graduate students, a population widely considered at elevated risk for financial strain and negative mental health outcomes. This study aimed to derive initial prevalence estimates of food insecurity in a sample of current graduate students at a large state university and to elucidate the relationship between food insecurity and depression, anxiety and stress in this sample.Design: Cross-sectional online survey including the US Household Food Security Survey Module: Six-Item Short Form and the Depression, Anxiety, and Stress Scales (DASS-21).Setting: University in the northeastern region of the USA.Participants: Two hundred sixty-three graduate students.Results: In the present sample, 59·7 % of participants reported high/marginal food security, 18·5 % reported low food security and 21·8 % reported very low food security. Graduate students with very low food security reported significantly greater depression (η2 = 0·09), anxiety (η2 = 0·10) and stress (η2 = 0·10), compared with those with low food security and high food security (all P's < 0·001).Conclusions: Food insecurity occurred in nearly half of the graduate students surveyed, and very low food security was associated with elevated levels of depression, anxiety and stress. The association of musculoskeletal pain, respiratory distress, gastrointestinal discomfort, and genitourinary pain to sleep quality in 89 persons with and without dementia was examined in the current exploratory, cross-sectional, observational quantitative design. Sources of pain were recorded, with 37.1% of the sample reporting urinary retention, 30.3% reporting genitourinary discomfort, and 53.9% reporting gastrointestinal discomfort. Mild scores of musculoskeletal pain and respiratory discomfort were elicited in >75% of the sample. An actigraph was used to measure sleep variables. Findings from this research illustrated dementia, gender, pillow use, respiratory distress, and urinary retention were statistically significantly associated with nighttime sleep quality. Results suggest comprehensive assessments of pain and sleep for older adults with and without dementia are needed. Developing and testing interventions that address factors contributing to sleep quality may improve outcomes. The objective of this study was to assess aspects of the social behavior of a mixed-breed herd of beef cows as a potential source for stress and economic losses. Angus (AN; N = 10), Brahman (BR; N = 10), and Senepol (SE; N = 10) cows were assigned to two groups (N = 15 each containing equal breed numbers) on separate pastures. Agonistic interactions (win/loss) during feeding were recorded daily for 45 days. Dominance values were estimated as the proportion of individuals dominated to total herdmates. From this, individuals were placed into social categories based upon linear ranking as follows: dominants (D), intermediate (I), and subordinates (S). Breed influenced (P < 0.01) social category, with SE cows being dominants (P < 0.05) over AN and BR cows. Interactions between AN and BR cows were less (P < 0.0005) than interactions between AN and SE (53 vs 140, respectively). Within breeds, BR (152) and SE (182) cows had more (P < 0.0005) agonistic interactions than AN (107) cows. Although apparently influenced by breed, agonistic interactions occurred more frequently (P < 0.005) between social categories than within social categories (814 vs 310, respectively). Dominant cows were involved in more agonistic interactions with cows from different social categories than were intermediate and subordinate cows (P < 0.0005). However, intermediate (100) and subordinate (157) cows generated more (P < 0.0005) agonistic interactions within their own social category than dominant cows (53). It was concluded that, in mixed-breed herds, breed influences both social organization and agonistic interactions which could be considered as potential sources of stress and economic losses. The development of shared memories, beliefs, and norms is a fundamental characteristic of human communities. These emergent outcomes are thought to occur owing to a dynamic system of information sharing and memory updating, which fundamentally depends on communication. Here we report results on the formation of collective memories in laboratory-created communities. We manipulated conversational network structure in a series of real-time, computer-mediated interactions in fourteen 10-member communities. The results show that mnemonic convergence, measured as the degree of overlap among community members' memories, is influenced by both individual-level information-processing phenomena and by the conversational social network structure created during conversational recall. By studying laboratory-created social networks, we show how large-scale social phenomena (i.e., collective memory) can emerge out of microlevel local dynamics (i.e.,mnemonic reinforcement and suppression effects). The social-interactionist approach proposed herein points to optimal strategies for spreading information in social networks and provides a framework for measuring and forging collective memories in communities of individuals. Foamy virus (FV) vectors are promising for hematopoietic stem cell (HSC) gene therapy but preclinical data on the clonal composition of FV vector-transduced human repopulating cells is needed. Human CD34+ human cord blood cells were transduced with an FV vector encoding a methylguanine methyltransferase (MGMT)P140K transgene, transplanted into immunodeficient NOD/SCID IL2Rγnull mice, and selected in vivo for gene-modified cells. The retroviral insertion site profile of repopulating clones was examined using modified genomic sequencing PCR. We observed polyclonal repopulation with no evidence of clonal dominance even with the use of a strong internal spleen focus forming virus promoter known to be genotoxic. Our data supports the use of FV vectors with MGMTP140K for HSC gene therapy but also suggests additional safety features should be developed and evaluated. [ABSTRACT FROM AUTHOR]

This article is the presentation of the main phobia data from the Epidemiologic Catchment Area (ECA) program, with a sample size of n = 18.571. Work on this article was initiated in 1981 at the beginning of the ECA study, but publication has been delayed a decade. Phobias are determined from information from the Diagnostic Interview Schedule (DIS), classified according to DSM III. Phobias are found to be the most common psychiatric disorder in the community, more common than major depression or alcohol abuse or dependence in the month prior to interview. The one month prevalence is between 4.0 and 11.1%, with the estimated prevalence in the United States being 6.2%. There were nine community surveys of the prevalence of phobia that pre-dated the ECA studies, which found a wide range of prevalence rates from 1.2% to 26.1%. By far the strongest risk factor associated with phobias is the presence of another psychiatric disorder. Prevalence rates of simple phobia and agoraphobia are found in the ECA studies to be significantly higher in women; social phobia, which is less prevalent, has no significant sex difference. The prevalence rates are higher in younger age groups, and in those with low socioeconomic status (SES). The onset of phobias occurs primarily in the childhood or teenage years, and they tend to be chronic conditions. Less than a quarter of phobics receive treatment. [ABSTRACT FROM AUTHOR]

Inclusion of Bos indicus genetics improves production traits of cattle maintained in hot climates. Limited information exists detailing pregnan-cy-specific events as influenced by variable amounts of Bos indicus genetics. Three experiments were complet-ed to examine the effect of Bos taurus and Bos indicus genotypes on fetal size and plasma pregnancy-associ-ated glycoprotein (PAG) concentrations. In all experi-ments, cows were bred by AI after synchronization of ovulation. Fetal measurements were completed by transrectal ultrasonography and plasma PAG concentra-tions were quantified from plasma harvested the day of each fetal measurement. In Exp. 1, fetal size and plasma PAG concentrations were measured at d 53 of preg-nancy in cows composed of various fractions of Angus and Brahman (n = 9 to 21 cows/group). Fetus size was greater in cows containing >80% Angus genetics com-pared with cows containing <80% Angus influence (3.40 ± 0.28 vs. 2.86 ± 0.28 cm crown-rump length; P < 0.01). Plasma PAG concentrations were reduced (P < 0.01) in cows containing >80% Angus genetics when compared with their contemporaries (6.0 ± 1.5 ng/mL vs. 9.4 ± 1.5 ng/mL). In Exp. 2, fetal measurements and plasma PAG concentrations were determined at d 35 and 62 of pregnancy in Angus and Brangus cows. Breed did not affect fetus size at d 35, but Angus cows contained larger fetuses than Brangus cows at d 62 [3.0 ± 0.03 vs. 2.8 ± 0.03 cm crown-nose length (CNL; P > 0.01)]. Plasma PAG concentrations were not different between breed at d 35 and 62 (P >0.1). In Exp. 3, fetal measurements and plasma samples were collected at d 33/34, 40/41, 47/48, and 54/55 post-AI in Angus and Brangus cows. Fetus size was not different (P > 0.05) between genotypes on d 33/34,40/41, and 47/48. Angus fetuses were larger than Brangus fetuses at d 54/55 (2.1 ± 0.03 vs. 1.9 ± 0.03 cm CNL; P = 0.001 ). Plasma PAG concentrations were less in Angus than Brangus cows at each time point (average 4.9 ± 0.9 vs. 8.2 ± 0.9 ng/ mL; P = 0.005). In conclusion, these studies determined that the Bos taurus x Bos indicus genotype impacts fetal size and rate of fetal development by 7 wk of gestation. Plasma PAG concentrations were increased in cattle with Bos indicus genetics in 2 of 3 studies, suggesting that genotype is one of several determinants of PAG production and secretion in cattle. The aim of this study was to evaluate potential sampling strategies for detection of infected flocks that could be applied during an outbreak of low pathogenicity notifiable avian influenza (LPNAI) initiated in duck holdings, following initial detection. A simulation model of avian influenza virus transmission and spread within and between holdings, respectively, was used to predict the impact on the size and duration of an outbreak of (i) changing the tracing window within which premises that might be the source of infection or that may have been infected by the index premises were sampled and (ii) changing the number of birds sampled in the flock being tested. It has shown that there is potential benefit in increasing the tracing window in terms of reducing the likelihood of a large outbreak. The objectives of this study were to determine the fraction of additive genetic variance explained by the SNP from the Illumina Bovine3K chip; to compare the ranking of animals evaluated with genomic-polygenic, genomic, and polygenic models; and to assess trends in predicted values from these 3 models for residual feed intake (RFI), daily feed intake (DFI), feed conversion ratio (FCR), and postweaning BW gain (PWG) in a multibreed Angus-Brahman cattle population under subtropical conditions. Data consisted of phenotypes and genotypes from 620 bulls, steers, and heifers ranging from 100% Angus to 100% Brahman. Phenotypes were collected in a GrowSafe automated feeding facility (GrowSafe Systems, Ltd., Airdrie, Alberta, Canada) from 2006 to 2010. Variance components were estimated using single-trait genomic-polygenic mixed models with option VCE (Markov chain Monte Carlo) of the program GS3. Fixed effects were contemporary group (year-pen), age of dam, sex of calf, age of calf, Brahman fraction of calf, and heterozygosity of calf. Random effects were additive SNP, animal polygenic, and residual effects. Genomic predictions were computed using a model without polygenic effects and polygenic predictions with a model that excluded additive SNP effects. Heritabilities were 0.20 for RFI, 0.31 for DFI, 0.21 for FCR, and 0.36 for PWG. The fraction of the additive genetic variance explained by SNP in the Illumina 3Kchip was 15% for RFI, 11% for DFI, 25% for FCR, and 15% for PWG. These fractions will likely differ in other multibreed populations. Rank correlations between genomic-polygenic and polygenic predictions were high (0.95 to 0.99; P < 0.0001), whereas those between genomic-polygenic and genomic predictions were low (0.65 to 0.74; P < 0.0001). Genomic-polygenic, genomic, and polygenic predictions for all traits tended to decrease as Brahman fraction increased, indicating that calves with greater Brahman fraction were more efficient but grew more slowly than calves with greater Angus fraction. Predicted SNP values were small for all traits, and those above and below 0.2 SNP SD were in multiple chromosomes, supporting the contention that quantitative traits are determined by large numbers of alleles with small effects located throughout the genome. The article explains the role of lymphatic fluid in screening Mycobacterium avium subspecies paratumberculosis (MAP) in cattle. The study was performed on 58 cows, where lymph fluid was extracted from their udders to identify traces of MAP through nested polymerase chain reaction. Results revealed that MAP was identified in 43.1 percent of lymph samples, where it was discovered in 66.7 percent cows with clinical Johne's disease.

The objective of this study was to compare performance and aspects of adaptability attributes of cattle from a Florida Angus bloodline (local source from a mostly closed herd for over 50 yr) to cattle that are representative of modern Angus bloodlines (outside source) in US subtropical conditions. Embryos from both sources were transferred to Brahman-crossbred cows in South Florida, and calves (n = 82) were born in 3 yr. Before weaning, summer tympanic temperatures were recorded hourly for 3 d in each year. Heifers were placed with fertile bulls until diagnosed pregnant. Traits relative to sexual maturation of bulls were recorded at 1- or 2-mo intervals until approximately 17 mo of age. Calves from outside sources had greater hip height at weaning than calves from the local source (P < 0.001; 108.8 ± 0.62 and 104.7 ± 0.68 cm, respectively). Local-source calves (n = 37) had greater (P = 0.03) exit velocity (2.7 ± 0.3 m/s) than outside-source (n = 45) calves (2.0 ± 0.29 m/s), which may be indicative of more nervous or temperamental disposition. However, no source differences were detected for other assessments of disposition (chute or pen score, P > 0.8). Few source differences for minimum, maximum, or range of daily tympanic (inner ear) temperatures were detected. At 17 mo of age, outside-source heifers were heavier (P = 0.05) and had greater (P < 0.001) hip height than Angus heifers from the local source. Heifers from the outside source were younger (P < 0.001) at the time of their first conception (454 ± 17.5 d) than heifers from the local source (550 ± 16.9 d). Outside-source heifers also had greater (P < 0.02) pregnancy and calving rates (0.7 ± 0.119 and 0.62 ± 0.125, respectively) from exposure to bulls within a year from weaning than the heifers from the local source (0.29 ± 0.089 and 0.19 ± 0.077, respectively). Bulls from the outside source were heavier (P = 0.05) at 320 d of age than local-source bulls. From 14 through 17 mo of age, outside-source bulls had greater (P ⩽ 0.05) scrotal circumference and tended (P ⩽ 0.15) to be heavier than local-source bulls. There appeared to be no performance or adaptation advantages for the local-source Angus through 17 mo of age. The large source difference for age at first conception in heifers merits additional attention and comparison with cow lifetime production performance for the 2 sources. [ABSTRACT FROM AUTHOR]

Mineral oils have been historically favoured over plant oils for insect pest control in horticultural crops because of their greater efficacy. Recently the increased pressure for environmentally sustainable pest management strategies has renewed interest in the use of plant oils and also in the reasons for differences in efficacy between plant and mineral oils. Efficacy of canola and mineral oils were compared for two modes of action: asphyxia in control of Saissetia oleae on olives and as an oviposition deterrent in control of Phyllocnistis citrella on lemons. On olives both canola and mineral oil treatments significantly reduced the number of black scale in comparison to the control but mineral oil reduced the number of black scale significantly more than canola oil. When oils were applied to lemons as a preventative spray, concentrations of canola oil above 0.5% significantly reduced the number of P. citrella mines per leaf compared to the control and there were no significant differences between any concentration above 0.5% canola oil and 0.5% mineral oil. Canola oil applied at a concentration of 0.5% was significantly less effective than mineral oil applied at the same concentration. Efficacy of canola oil was found to be lower than that of mineral oil in all experiments, but the higher efficacy of mineral oil was more pronounced in suffocation of S. oleae than in deterrence of P. citrella oviposition. Our results indicate that even though canola oil has very different molecular structures than mineral oils the resulting physical characteristics of canola oil, primarily high boiling point and viscosity, may contribute to their lower efficacy against arthropod pests. However, low phytotoxicity of canola oil indicates that the chemical structure of molecules contained in canola oil had much more influence on processes on the plant surface than the physical characteristics of the oil. The article discusses research on the accuracy of diagnostic methods that are commonly used for identifying Anaplasma marginale infection among dairy cattle that are lactating in Puerto Rico. It references a study by Pedro Melendez and colleagues, published in the 2010 issue of the "Journal of the Veterinary Diagnostic Investigation." The researchers used the Bayesian methods to examine the cattle if they are suffering from the disease called Bovine anaplasmosis. They found that among the cases, only five cows were tested positive to the infection.

• The SLICK1 mutation in bovine PRLR confers cattle with a short hair coat and increased ability for thermoregulation. • Consequences for other physiological functions are not well understood. • It was found that animals with the mutation have slight alterations in hepatic gene expression. • Thus, inheritance of the SLICK1 allele can exert functional changes beyond those related to hair growth. • The mutation acts as a dominant allele because there were few differences in body temperature and liver gene expression between animals with one or two copies of the gene. The SLICK1 mutation in bovine PRLR (c.1382del; rs517047387) is a deletion mutation resulting in a protein with a truncated intracellular domain. Cattle carrying at least one allele have a phenotype characterized by a short hair coat (slick phenotype) and increased resistance to heat stress. Given the pleiotropic nature of prolactin, the mutation may affect other physiological characteristics. The liver is one organ that could potentially be affected because of the expression of PRLR. The mutation is a dominant allele, and heterozygous animals have a similar hair coat to that of animals homozygous for the mutation. Present objectives were to determine whether inheritance of the SLICK1 mutation affects liver gene expression and if animals homozygous for the SLICK1 allele differ from heterozygotes in liver gene expression and regulation of body temperature during heat stress. In one experiment, rectal and ruminal temperatures were less for Holstein heifers that were heterozygous for the SLICK1 allele compared with wildtype heifers. There were 71 differentially expressed genes in liver, with 13 upregulated and 58 downregulated in SLICK1 heterozygotes. Among the ontologies characteristic of differentially expressed genes were those related to immune function and fatty acid and amino acid metabolism. In a prospective cohort study conducted with adult Senepol cattle, body temperature and hepatic gene expression were compared between animals heterozygous or homozygous for the SLICK1 mutation. There were no differences in ruminal temperatures between genotypes, rectal temperature was higher in animals homozygous for the SLICK1 mutation, and there was only one gene in liver that was differentially expressed. It was concluded that inheritance of the SLICK1 allele can exert functional changes beyond those related to hair growth although changes in liver gene expression were not extensive. Paratuberculosis (Johne’s disease), caused by Mycobacterium avium subsp. paratuberculosis, is an important disease for bovines, although its genetic basis is poorly understood. In this study, three candidate genes were typed to study the associations between single nucleotide polymorphisms (SNPs) and paratuberculosis susceptibility (measured in a 1 or 0 form) at the haplotype level. A significant risk haplotype, constructed by a variant allele (C) at the first SNP and a common allele (A) at the second SNP, within the CARD15 gene was detected to trigger genetic effects on paratuberculosis infection in an overdominace manner. Marginally significant haplotypes were identified for the other two genes. The results obtained will provide scientific guidance about the selection and prediction of resistance types in bovines. Paratuberculosis is a chronic, infectious disease of ruminants that entails a serious concern for the cattle industry. One of the main issues relates to the efficiency of diagnosis of subclinically infected animals. The objective of this field study was to analyse the association among results of a serum enzyme-linked immunosorbent assays (ELISA), faecal culture and nested PCR tests on milk, blood and faeces for Mycobacterium avium subsp. paratuberculosis detection in dairy cows. Faeces, blood and milk samples were collected from 328 lactating dairy cows in four known infected herds. Results were analysed to determine associations and levels of agreement between pairs of tests. A total of 61 animals (18.6%) tested positive when all the tests were interpreted in parallel. The agreement between results in different pairs of tests was poor, slight and fair in two, five and three of the 10 possible combinations respectively. Faecal culture and faecal polymerase chain reaction (PCR) resulted in the highest kappa coefficient (0.39; fair agreement), with the lowest agreement being for ELISA and blood PCR (−0.036; poor agreement). Fisher’s exact test resulted in statistically significant associations ( P ≤ 0.05) between the following test pairs: ELISA : faecal culture; ELISA : faecal PCR; milk PCR : faecal PCR, blood PCR : faecal PCR and faecal culture : faecal PCR. Enzyme-linked immunosorbent assays showed the highest complementary sensitivity values for all the possible two-test combinations, followed by faecal PCR. Cow and calf genetic and environmental factors were evaluated for their association with ELISA scores for paratuberculosis in a multibreed population of beef cattle. The ELISA scores are a measure of the presence or absence of antibodies against Mycobacterium avium subsp. paratuberculosis in bovine serum. The linear mixed-model analysis used 352 ELISA scores from 238 cows: 51 Angus (A); 34 Brahman (B); 41 (¾ A¼ B); 45 (½ A½ B); 34 (¼A¾ B); and 33 Brangus (5/8 A 3/8 B). Cows were assumed to be unrelated. Year affected (P < 0.001) ELISA scores, but age of cow did not, which was expected to be significant because of the chronic progressive nature of this disease. Important regressions on fixed effects associated with cows were 1) a positive estimate of cow B breed effect (0.59 ± 0.24; P < 0.017), indicating an upward trend of ELISA scores toward 100% B cows; 2) a negative estimate for weight change from before calving (late November) to the date of the blood sample in May (-0.0062 ± 0.0019 score/kg; P < 0.002), indicating that poorer maintenance of cow weights was associated with higher ELISA scores; and 3) a positive estimate for days in lactation of cow on the date of the blood sample (0.0086 ± 000034 score/d; P < 0.021), indicating the production of larger amounts of antibodies against Mycobacterium avium subsp. paratuberculosis as lactation progressed. Relevant regressions on fixed effects associated with calves were 1) calf birth weight (-0.022 ± 0.010 score! kg; P < 0.035), and 2) calf gain from birth to the date of the cow blood sample (-0.0092 ± 0.0027 score/kg; P < 0.001). These estimates indicate that cows that produced lighter calves at birth and/or calves with slower preweaning growth tended to have greater ELISA scores. Although the sensitivity (percentage of infected animals detected) of ELISA was only 50%, these results suggest that subclinical paratuberculosis may be negatively affecting cows and their offspring. Little reliable information exists about the genetic events that control the onset and timing of seed-fill in soybean cotyledons. To identify the genes involved in this process, cDNA libraries were prepared from mRNAs isolated from seeds at 7 and 21 days after flowering (DAF), which represent times just before and after the initiation of seed-fill. For the soybean variety Resnik, which was used for this study, seed-fill and the establishment of an endoreduplicative cell cycle occurred 12–14 DAF. Suppression subtractive hybridization was then applied to identify sequences that were differentially expressed at each of these two developmental stages. False positives in the libraries were reduced by using mirror orientation selection (MOS). The libraries of differentially expressed genes that resulted were analysed and the nucleotide sequences obtained were compared with those in existing databases. Several genes from each library were chosen and their expression profile during seed development was analysed by quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) using RNA preparations originating from different seed developmental stages. The microstructure of the Sn-Ag-Cu solder is examined by optical microscopy and scanning electron microscopy (SEM) for various compositions near the ternary eutectic for different cooling rates from the solder melt. Focus is on the size and orientation of Sn grains as indicated by cross-polarized, light optical microscopy, and pole figures from x-ray diffraction. We fred that both composition and cooling rate have strong influences on Sn grain size, with Sn grain size increasing an order of magnitude as Cu concentration increases from 0% to 1.1%. Cyclic growth twinning, with twinning angles near 60°, is observed in Sn-Ag-Cu alloys near the composition Sn-3.9Ag-0.6Cu. [ABSTRACT FROM AUTHOR]

Background: To establish that magnetic resonance imaging (MRI) can provide an objective measurement of the biophysical state of the cervix and to measure the response of the cervix to prostaglandins in the first trimester of pregnancy.Methods: A comparative study, with each patient serving as her own control, was carried out on 10 primigravid women with 49--84 days amenorrhoea undergoing surgical termination of pregnancy. Each woman had two MRI scans of the uterus and cervix. The first scan was performed prior to administration of prostaglandins and the second scan 2.5--3 h following an 800 microg dose of misoprostol given vaginally. Cervical change was assessed by measurement of cervical length, transverse diameter of the cervix, diameter of the internal os and cervical stromal signal intensity.Results: A significant difference in cervical length (P = 0.012), transverse diameter (P = 0.001) and diameter of the internal os (P = 0.008) was detected following the administration of misoprostol. In five women a significant change in one or both components of the cervical stromal signal was detectable and in five women no change could be demonstrated.Conclusion: MRI is capable of detecting changes in the physical parameters of the cervix following administration of prostaglandin. However, the changes detected in the stromal signal intensity are less consistent. The efficacy of pest control of three different oil formulations in multiple low-concentration spray programmes on sweet orange and pummelo were compared with unsprayed and normal farmer-treatments over three years. Phytotoxicity of sprays was assessed in terms of fruit and leafdrop, fruityield and external fruitquality. Trees sprayed with any type of oil had lower pest numbers than unsprayed trees. On sweet orange, the heaviest oil was better than the normal farm practice at controlling chaff scale on fruit and red mite and whitefly on leaves. On pummelo it provided the best control of red mite on leaves. All three oils were as effective as the normal farm practice in control of rust mite on sweet orange fruit and leaves and red mite, rust mite and red scale on pummelo fruit. On sweet orange trees there was no evidence of phytotoxicity, and the external quality of fruit generally improved over time. On pummelo trees, oil sprays were unable to improve the external fruitquality. The number of pummelo per tree was reduced in three of the oil spray schedules in 1995 and one in 1996, but the total weight of fruit harvested per tree was unaffected. [ABSTRACT FROM AUTHOR]

Background: The aim of this study was to determine the patterns and determinants of service use in severely mentally ill persons drawn from the National Institute of Mental Health Epidemiological Catchment Area (ECA) program, a community-based epidemiologic survey. This information provides a baseline against which to track ongoing changes in the US mental health service system.Methods: Severe mental illness (SMI) was defined according to US Senate Appropriations Committee guidelines. Comparisons were made with persons who had a mental disorder that did not meet these criteria (non-SMI). Sociodemographic factors, and 1-year volume and intensity of mental or addictive services use were determined. Differences between those who used services and those who did not were examined using logistic regression.Results: Persons with SMI differed from persons with non-SMI in most sociodemographic characteristics. A higher proportion of persons with SMI used ambulatory services, but the mean number of visits per person did not differ from the non-SMI population. Persons with SMI comprised the bulk of hospital inpatients admitted during a 1-year period. Several significant sociodemographic determinants of service use were found, with different patterns for general medical and specialty service use, pointing out potential barriers to care.Conclusions: As health care reform measures continue to be debated, attention to the service needs of the severely mentally ill is of crucial importance. Pre-managed care (pre-1993) baseline service use benchmarks will be essential to assess the impact of managed care on access to care, particularly for the severely mentally ill. Periodic collection of epidemiologic data on prevalence and service use would thus greatly facilitate service planning and addressing barriers to receiving mental health services in this population. The Madin-Darby canine kidney (MDCK) cell line expresses many characteristics of the renal collecting duct. The MDCK-C7 subclone forms a high-resistance, hormone-responsive model of the principal cells, which are found in distal sections of the renal tubule. The electrophysiological technique of short-circuit current measurement was used to examine the response to antidiuretic hormone (ADH) in the MDCK-C7 clone. Three discrete electrogenic ion transport phenomena can be distinguished temporally and by the use of inhibitors and effectors. Initially the cells exhibit anion secretion through the cystic fibrosis transmembrane conductance regulator (CFTR). The presence of CFTR was confirmed by immunoprecipitation followed by Western blotting. The CFTR-mediated anion secretion is transient and is followed, in time, by a verapamil- and Ba2+-sensitive anion secretion or cation absorption and, finally, by Na+ reabsorption via epithelial Na+ channels (ENaC). In contrast to other studies of MDCK cells, we see no indication that the presence of CFTR functionally inhibits ENaC. The characterization of the various ion transport phenomena substantiates this cell line as a model renal epithelium that can be used to study the hormonal and metabolic regulation of ion transport. The efficacy of petroleum spray oil against citrus psylla was tested in Guangzhou, China. As the oil concentration increased from 0.25% to 1.0% there was a linear decrease in the number of psylla of each stage present on foliar shoots after 8 days. When psylla infested shoots were sprayed with equivalent oil concentrations, survival was significantly different between oil treatments. Psylla survival was also stage-dependent, with 1st- 2nd instars being the most susceptible and eggs being the most tolerant to oil. Oil efficacy was compared to an organophosphate pesticide and an insect growth regulator, and all provided similarly effective controlofpsylla nymphs. Rates of sodium absorption in the presence of bacterial fatty acids and bile salts are unknown along the length of the colon. Such information may guide resection of colon with a view to leaving colon most optimal for ion absorption. Absorption of sodium and permeability of the colonic mucosa to51Cr EDTA was measured in the proximal (PC) and distal colon (DC) instilled with NaCl (120 mM), n-butyrate (40 mM) and chenodeoxycholic acid (1 or 2 mM) in varying combinations. Sodium absorption in the PC was 123.6±19 (nmoles/min/cm2) (n=7) with saline alone and was doubled (P<.001) when 40 mM n-butyrate replaced chloride anions. Sodium absorption in the DC with saline alone was 106.9±18 without significant alteration by addition of n-butyrate. CDC (1 mM) diminished sodium absorption in the PC and DC. Diminished sodium absorption induced by bile salts was significantly reversed by n-butyrate in the PC but not DC. Permeability of51Cr EDTA was greatest with 2 mM CDC in the distal colon. Mucosal function—sodium absorption with SCFAs, permeability changes, and protection by bacterial fatty acids from sodium losses due to bile salts—was superior in the PC compared with DC. Our results suggest that it may be preferable to preserve the proximal rather than distal colon in operations of the colon to secure optimal absorption of sodium. [ABSTRACT FROM AUTHOR]

A comparison has been made of costs within in-patient units in a large Scottish psychiatric hospital. Cost differences were found between short- stay, rehabilitation and long-stay psychosis units and beds for the elderly with mental disorder. These differences reflected staffing levels, particularly of nurses. The costs were greater than average for psychiatric hospitals and approximated to geriatric units-a finding in keeping with the high occupancy of beds by the elderly. Existing long-stay elderly patients require to be considered separately in distinguishing elderly in-patient costs without total psychogeriatric care offered by society. [ABSTRACT FROM AUTHOR]
