Brassica rapa line K185 was identified with broad-spectrum extreme resistance to turnip mosaic virus (TuMV). The resistance was effective against TuMV isolates UK 1, CZE 1, GBR 6, POL 1 and CDN 1 (representing major pathotypes 1, 3 and 4 and major resistance-breaking isolates) following mechanical inoculation. F1 plants from a cross between K185 and the rapid-cycling B. rapa ssp. trilocularis line R-o-18 (uniformly susceptible to all the above TuMV isolates), were resistant following challenge with TuMV isolates CDN 1 and GBR 6, indicating the involvement of dominant gene(s). F2 plants derived from a single F1, CDN 1- and GBR 6-resistant plant segregated for resistance to TuMV isolate GBR 6. The segregation ratio of resistant to susceptible plants was consistent with at least two loci controlling resistance and with both loci having dominant alleles for resistance. Other sources of broad-spectrum resistance to TuMV have been shown to involve the recessive allele retr01. The broad-spectrum resistance in K185 clearly involves different alleles to retr01 and provides the opportunity to introgress an alternative form of TuMV resistance into commercial B. rapa lines and thereby reduce the selection pressure for retr01 resistance breaking. Background: Populations living in fragmented habitats may suffer from loss of genetic variation and reduced between-patch dispersal, which are processes that can result in genetic differentiation. This occurs frequently in species with reduced mobility, whereas genetic differentiation is less common among mobile species such as migratory birds. The high dispersal capacity in the latter species usually allows for gene flow even in fragmented landscapes. However, strongly philopatric behaviour can reinforce relative isolation and the degree of genetic differentiation. The Southern Dunlin (Calidris alpina schinzii) is a philopatric, long-distance migratory shorebird and shows reduced dispersal between isolated breeding patches. The endangered population of the Southern Dunlin breeding at the Baltic Sea has suffered from habitat deterioration and fragmentation of coastal meadows. We sampled DNA across the entire population and used 12 polymorphic microsatellite loci to examine whether the environmental changes have resulted in genetic structuring and loss of variation. Results: We found a pattern of isolation-by-distance across the whole Baltic population and genetic differentiation between local populations, even within the southern Baltic. Observed heterozygosity was lower than expected throughout the range and internal relatedness values were positive indicating inbreeding. Conclusions: Our results provide long-term, empirical evidence for the theoretically expected links between habitat fragmentation, population subdivision, and gene flow. They also demonstrate a rare case of genetic differentiation between populations of a long-distance migratory species. The Baltic Southern Dunlin differs from many related shorebird species that show near panmixia, reflecting its philopatric life history and the reduced connectivity of its breeding patches. The results have important implications as they suggest that reduced connectivity of breeding habitats can threaten even long-distance migrants if they show strong philopatry during breeding. Males of many species theoretically face a fitness tradeoff between mating and parental effort, but quantification of this is rare. We estimated the magnitude of the mating opportunity cost paid by incubating male Temminck's stints ( Calidris temminckii), taking advantage of uniparental care provided by both sexes in this species. 'Incubating males' provide all care for an early clutch, limiting subsequent mating possibilities. 'Non-incubating' males include males that failed to obtain, lost to predation, or actively avoided incubating clutches. These males were free to pursue extrapair copulations and to mate with females laying later clutches, which females usually incubate themselves. Male incubation classes did not differ in measures of quality, and many individuals changed classes between years, suggesting the use of conditional reproductive tactics. However, specialist non-incubators may also exist. Using microsatellites to assign parentage, we compare males' total fertilizations and the subset 'free of care' fertilizations between incubation classes. Incubators were more likely to gain at least one fertilization per season and averaged one more per season than non-incubators. However, successful non-incubators were more likely to gain 'free of care' fertilizations, averaging two more than successful incubators. The relative success of male incubation classes also changed with local sex ratios. With higher female proportions, non-incubators gained disproportionately more offspring, suggesting that the use of tactics should be partly determined by the availability of potentially incubating females. Aim We study the population differentiation and phylogeography of the Temminck's Stint ( Calidris temminckii). Specifically, we seek signs of past and present population size changes and dispersal events and evaluate management and conservation unit status of the populations. We also study the possibility of introgression as the origin of two mitochondrial DNA (mtDNA) lineages found and estimate the divergence time of the lineages. Location Northern Eurasia. Methods We analysed 583 bp of mtDNA control region domains I and II and 11 microsatellite loci from 13 localities throughout the breeding range. In addition, we used mitochondrial cytochrome c oxidase subunit I ( COI), a barcoding gene, to search for signs of introgression. Results More population differentiation was found from microsatellites than from mtDNA, although differentiation was weak in both markers. Signs of past population growth were observed, in addition to more recent decline in some areas. Both control region and COI sequences revealed two maternal lineages coexisting in Fennoscandia and in north-west Siberia. No signs of introgression were detected. Lineage divergence time was estimated to have occurred during the glacial periods of Pleistocene. Main conclusions Slight differences in mtDNA and microsatellite differentiation and diversity may reflect different features - such as the mutation rate and effective population size - of the markers used, or female-biased dispersal pattern and high male site-fidelity of the species. The coexistence of the two mitochondrial lineages is most likely a consequence of post-glacial mixing of two refugial Pleistocene populations. Based on genetic information alone, global conservation concerns are not imminent. In some animal populations, immigrants have lower survival than philopatric individuals. Costs of dispersal or low phenotypic quality of dispersers may explain the pattern. However, apparent adult survival estimates, which describe real survival combined with site fidelity cannot be separated from permanent emigration. Thus, heterogeneity in breeding dispersal propensities of immigrants and philopatrics can bias fitness correlates of dispersal. Differences in breeding dispersal propensities may be caused by different strategies in response to environmental cues inducing dispersal, such as reproductive success. In such cases, the reported differences between immigrants and philopatric individuals may not reflect true variation in survival. We studied whether dispersal status specific apparent adult survival is associated with reproductive success in a Temminck's stint Calidris temminckii population. We analysed two long term capture-recapture datasets characterised by low and high nest predation levels. Philopatric individuals had higher apparent adult survival than immigrants in both datasets and the difference was highlighted during the high nest predation period. By contrasting return rates between successful and unsuccessful breeders as a proxy for dispersal, we found that unsuccessful immigrants breeding for the first time dispersed more likely than successful immigrants, but such a pattern was not found among philopatric individuals. Our results support the hypothesis that immigrant and philopatric individuals have different breeding dispersal strategies following reproductive failure and that their apparent adult survival differences are at least partly explained by different breeding dispersal propensities. Our results also suggest that the recent decline of the study population reflects a multiple response to increased nest predation through decreased local recruitment and increased emigration. Temminck's stint breeds in Eurasian arctic tundra and subarctic and temperate boreal zones in a range extending from Fennoscandia to easternmost Siberia. In contrast to the favourable global conservation status of the species, it has been classified as vulnerable in Finland and near threatened in Sweden. A fragment of the control region of mtDNA was sequenced from 127 individuals from breeding areas in Fennoscandia in the west (three populations) and in the eastern end of the range. The mtDNA variability and structuring were among the lowest values reported for waders (FST -0.02616). The mtDNA sequences revealed seven haplotypes, of which four were present in single individuals. The most common haplotype occurred in 81% of all individuals and in all birds in the Siberian sample. There was evidence of two maternal lineages. The most common lineage occurred in 95% of the individuals and was the only one present in the Siberian sample. The lineages coexisted in all three Fennoscandian populations, indicating a secondary contact of two previously isolated populations. The mtDNA variation and the mitochondrial nucleotide and haplotype diversities indicated panmixis of the populations. However, a higher degree of population differentiation was detected in microsatellite allele frequencies (125 birds, six loci) in Fennoscandia between the Bothnian Bay population and the two inland populations (Lapland and southern Norway). The difference may be caused by the female-biased dispersal pattern of the species. We measured nesting success of the Temminck's Stint Calidris temminckii along the Finnish Bothnian Bay coast during 19 breeding seasons (1983–2001) and conducted a population census (1999–2002). We found 105 pairs, showing a marked decline from the previous survey (170 pairs 1987–95). Of the 424 ‘known-fate’ nests, 47% hatched. Depredation caused 79.9% of the nest losses. Nesting failures increased from 1983–91 to 1992–2001 owing to a rise in nest predation. The proportion of failed nests that failed because of predation rose from 48.9 to 87.7%. When only depredated nests were considered as losses, Mayfield nest survival probability over the incubation period dropped from 69 to 31% (461 nests). This pattern emerged both in man-made and in natural habitats. Survival probability was independent of habitat type (natural vs. man-made). In an experiment involving videotaping of dummy nests, Common Gull Larus canus and Ruddy Turnstone Arenaria interpres were found to be the most important egg predators. [ABSTRACT FROM AUTHOR]
