The assessment of two diamine derivatives, TMD and IPDA, as corrosion inhibitors for aluminium in hydrochloric acid (HCl) at concentrations of 0.2 M, 0.3 M, and 0.4 M. (4S)—2,2,4-trimethyl hexane-1,6-diamine (TMD) and (1R,3R)-3-(amino methyl)-3,5,5-trimethyl cyclohexane-1-amine (IPDA) are verified for their ability to inhibit corrosion of Al in acidic conditions. The evaluation is conducted using electrochemical impedance spectroscopy (EIS), potentiodynamics polarization (PDP), and gravimetric methods. The inhibitors' effectiveness depends on their concentration (ranging from 20 to 50 mM) and molecular structure. Because TMD has more anchoring functional groups, it had the best inhibitory efficacy, reaching a maximum of 98.3% at 50 mM concentration. It is looked into how temperature (313–333 K) affects corrosion behaviour. As per Langmuir's adsorption isotherm, the temperature probably influences the ability of inhibitors to adsorb on the surface of aluminium, pointing to a monolayer adsorption mechanism. Calculated and described are the thermodynamics activation factors for the dissolution process of Al in both inhibited along uninhibited solutions. The stability of the inhibitor-Al contact and the reaction kinetics are revealed by these characteristics. The creation of a protective covering on the Al surfaces was validated by surface investigation techniques such as atomic force microscopy, scanning electron microscopy, and energy-dispersive X-ray (EDX) analysis, demonstrating the inhibitors' efficacy in preventing corrosion. The inhibitors' molecular chemical makeup as well as the degree of inhibitory efficacy is correlated in theoretical investigations employing molecular dynamics simulations and density functional theory. In this paper, an unexplored solid solution (1-x)Bi(Cu1/2Ti1/2)O3-(x)PbTiO3 has been synthesized and characterized for its crystal structure, dielectric and ferroelectric properties aimed at a selected composition, i.e., 0.30 Bi(Cu1/2Ti1/2)O3-0.70PbTiO3. The paper unveils the different synthesis approaches to achieve pure-phase in Pb-based perovskite-structured materials that are prone to produce impurities during synthesis. The Rietveld refinement of the XRD pattern confirms a tetragonal crystal structure (P4mm) of the composition. The ferroelectric properties of the as-prepared composition are also discussed using P-E hysteresis and PUND characteristics. Exploring the functionalities of materials requires a profound understanding of the crystal structure. In this paper, room temperature crystal structures of a new solid solution (1 − x)Ba(Cu1/3Nb2/3)O3-(x)PbTiO3 have been investigated in the entire compositional span and different crystallographic phases and phase coexistence regions have been discovered. The confirmation of the symmetry of these crystal structures has been done with the help of Rietveld analysis of the high-resolution XRD data. Despite both the end components, Ba(Cu1/3Nb2/3)O3 and PbTiO3, having tetragonal (P4mm) symmetry, new cubic and monoclinic phases have been discovered for the intermediate compositions with multiple phase boundaries. The composition region 0.05 ≤ x ≤ 0.55 exhibits a cubic crystal structure and increasing PbTiO3 concentration to 0.62 results in a unique coexistence of two tetragonal phases with different tetragonalities. This transformation is mediated by coexisting cubic and tetragonal phases, for 0.59 and 0.60. The crystal structure of the solid solution later transforms into coexisting monoclinic and tetragonal phases for a wider compositional span i.e., 0.65 ≤ x ≤ 0.85. This composition region is very fascinating, as two phenomenologically different monoclinic structures have been observed in it. Finally, a tetragonal phase at x = 1 is achieved through the mediation of two coexisting tetragonal phases, for the region 0.85 < x ≤ 0.975. The evolution of different crystallographic structures and the coexisting phases are critically comprehended using the variations in the lattice parameters and unit cell volume. The presence of multiple phase boundaries spread across a wide range of compositions makes this solid solution very intriguing and a viable choice for exploring different properties with compositional tuning. The studies permitted ‘Seasonal incidence of pod pest complex of pigeonpea [Cajanus cajan (L.) Millsp.]’ were conducted during Kharif 2020-2021. The experiment was carried out at the Agricultural Research Farm, Institute of Agricultural Sciences, Banaras Hindu University, Varanasi. Seasonal incidence of pigeonpea against pod pest complex revealed that the maximum incidence of Melanagromyza obtusa (Malloch), Clavigralla gibbosa (Spinola) and Helicoverpa armigera (Hubner) was 5.33 maggots/plant, 7.53 bugs/plant, 5.20 larvae/plant respectively in the 9th SMW (1st week of March). Correlation of the pod pest complex and weather parameters revealed that the incidence of M. obtusa were showed the significant negative correlation with evening relative humidity (r= -0.669*), while the significant positive correlation with maximum temperature (r= 0.614*) and sunshine (0.779*), the incidence of C. gibbosa showed non-significant negative correlation with the evening relative humidity (r= -0.401), while the significance positive correlation with sunshine (r= 0.782*) and the incidence of H. armigera showed significantly negative correlation with evening relative humidity (r= -0.591**) and significant positive correlation with maximum temperature (r= 0.544**) and sunshine (r= 0.612*). The present study was conducted to evaluate the comparative efficacy and economics of different insecticides against Melanagromyza obtusa, Clavigralla gibbosa and Helicoverpa armigera infesting pigeonpea variety Bahar during Kharif 2020-21. Among all the treatments, Thiamethoxam 25 WG @ 75g a.i./ ha provided better control of M. obtusa and C. gibbosa in terms of higher percent reduction in population over control, lower pod and grain damage and higher grain yield. Spinosad 45 SC @ 73g a.i./ ha was found to be most effective against H. armigera with minimum mean larval population (1.14 larvae/ plant), pod damage (5.20%) and grain damage (1.72%). All the treatments were statistically significant in increasing grain yield (i.e. 986.89 kg/ha to 1039.49 kg/ha) over untreated control (803.02 kg/ha). Theileriosis is an economically important haemoprotozoan disease of cattle caused by Theileria spp. It causes production losses and mortality in dairy industry. A total of 135 cattle suspected of theileriosis were screened based on clinical signs and blood smear examination (Giemsa's stain). Out of them, 64 animals were diagnosed with theileriosis. Twenty-four cattle out of them were selected and divided into four equal groups, viz., B, C, D, and E, each comprised of six animals for four different treatment protocols to evaluate their therapeutic efficacy, while group A of six healthy cattle served as control. The most common clinical findings of theileriosis infected cattle (n = 24) in descending order were anorexia (100%), dullness and depression, decreased milk production, weakness, salivation, enlarged pre-scapular lymph nodes, pale conjunctival mucous membrane, presence of ticks over the body, emaciation, nasal discharge, cessation of rumination, lacrimation, diarrhea and sternal recumbency (8.33%). The mean rectal temperature, heart rate, and respiratory rate were 104.5 ± 0.33° F, 79.5 ± 2.85/min and 42.83 ± 2.67/min, respectively. The haematological findings revealed decreased values of haemoglobin, packed cell volume, total erythrocytes count (TEC), lymphocytes, and mean corpuscular hemoglobin concentration (MCHC), and increased values of neutrophils, eosinophils, Mean corpuscular volume (MCV), and mean corpuscular hemoglobin (MCH) in theileriosis infected cattle as compared to healthy cattle. Biochemical findings revealed decreased values of total protein and albumin while increased total bilirubin, Aspartate aminotransferase (AST), Alanine aminotransferase (ALT), and Gamma-glutamyl transferase (GGT) in theileriosis-infected cattle. These altered haemato-biochemical profiles were found to be normalized by the 14th day in different treatment groups. The therapeutic efficacy of Buparvaquone 2 dose (Group B) and Buparvaquone plus Oxytetracycline LA (Group D) was better (100.00%) than the Arteether (Group C, 66.66%) and Arteether plus Oxytetracycline LA (Group E, 83.33%) in theileriosis infected cattle, hence may be advocated to the field veterinarians for effective treatment of theileriosis in cattle. New diagnostic tests for the etiology of childhood pneumonia are needed. We evaluated the antibody-in-lymphocyte supernatant (ALS) assay to detect immunoglobulin (Ig) G secretion from ex vivo peripheral blood mononuclear cell (PBMC) culture, as a potential diagnostic test for pneumococcal pneumonia. We enrolled 348 children with pneumonia admitted to Patan Hospital, Kathmandu, Nepal between December 2015 and September 2016. PBMCs sampled from participants were incubated for 48 h before harvesting of cell culture supernatant (ALS). We used a fluorescence-based multiplexed immunoassay to measure the concentration of IgG in ALS against five conserved pneumococcal protein antigens. Of children with pneumonia, 68 had a confirmed etiological diagnosis: 12 children had pneumococcal pneumonia (defined as blood or pleural fluid culture-confirmed; or plasma CRP concentration ≥60 mg/l and nasopharyngeal carriage of serotype 1 pneumococci), and 56 children had non-pneumococcal pneumonia. Children with non-pneumococcal pneumonia had either a bacterial pathogen isolated from blood (six children); or C-reactive protein <60 mg/l, absence of radiographic consolidation and detection of a pathogenic virus by multiplex PCR (respiratory syncytial virus, influenza viruses, or parainfluenza viruses; 23 children). Concentrations of ALS IgG to all five pneumococcal proteins were significantly higher in children with pneumococcal pneumonia than in children with non-pneumococcal pneumonia. The concentration of IgG in ALS to the best-performing antigen discriminated between children with pneumococcal and non-pneumococcal pneumonia with a sensitivity of 1.0 (95% CI 0.73–1.0), specificity of 0.66 (95% CI 0.52–0.78) and area under the receiver-operating characteristic curve (AUROCC) 0.85 (95% CI 0.75–0.94). Children with pneumococcal pneumonia were older than children with non-pneumococcal pneumonia (median 5.6 and 2.0 years, respectively, p < 0.001). When the analysis was limited to children ≥2 years of age, assay of IgG ALS to pneumococcal proteins was unable to discriminate between children with pneumococcal pneumonia and non-pneumococcal pneumonia (AUROCC 0.67, 95% CI 0.47–0.88). This method detected spontaneous secretion of IgG to pneumococcal protein antigens from cultured PBMCs. However, when stratified by age group, assay of IgG in ALS to pneumococcal proteins showed limited utility as a test to discriminate between pneumococcal and non-pneumococcal pneumonia in children. Background. Diabetic patients are more susceptible to urinary tract infection compared to nondiabetic patients, Escherichia coli being the most common uropathogen causing UTI. Unreasonable and incorrect antibiotic prescription for UTI in these patients may induce the development of antibiotic-resistant urinary pathogens resulting in delayed recovery and longer hospitalization. In addition to these, biofilm forming capacity of the pathogen may worsen the problem. The main aim of this cross-sectional study (conducted from March to September 2015) is to detect the biofilm forming capacity of UTI causing micro-organisms and compare the antibiotic resistance pattern of Escherichia coli, the most common cause of UTI, which will help the physician in choosing the best antibiotic. Method. Total of 1,099 clean-catch mid stream urine (CCMSU) was processed by standard microbiological technique; 182 were from the diabetic group and 917 nondiabetic. Following identification, all isolates were subjected to antibiotic susceptibility testing using modified Kirby-Bauer disc diffusion method. In-vitro biofilm forming capacity of the isolates were detected by Microtitre plate method. The data were analyzed using SPSS software 16. Result. Urinary tract infection was found to be significantly higher in diabetic patients (42.9%) compared to nondiabetic patients (17.4%) with Escherichia coli as the most common uropathogen in both diabetic and nondiabetic groups. Similarly, UTI was more common in elderly population (29.5%). Imipenem, nitrofurantoin and amikacin were found to be the most effective drug for uropathogenic E. coli in both diabetic and nondiabetic patients, whereas amoxicillin, ciprofloxacin, and cotrimoxazole were least effective. Of the total bacterial isolates, 43.3% showed positive results for in-vitro biofilm production by the Microtitre plate method. A significantly higher resistance rate was observed among biofilm producing E. coli for quinolones, cotrimoxazole, and third generation cephalosporin ceftriaxone. Most of the biofilm producers (79.5%) were found to be MDR (p-value 0.015). Conclusion. Elderly populations with diabetes are at a higher risk of UTI. Higher biofilm production and resistance to in-use antimicrobial agents in this study render its inefficacy for empirical treatment and point out the importance of biofilm screening to ensure the effective management of infection. Background: Salmonella serovars Typhi (S. Typhi) and Paratyphi A (S. Paratyphi A), the causative agents of enteric fever, have been routinely isolated organisms from the blood of febrile patients in the Kathmandu Valley since the early 1990s. Susceptibility against commonly used antimicrobials for treating enteric fever has gradually changed throughout South Asia since this time, posing serious treatment challenges. Here, we aimed to longitudinally describe trends in the isolation of Salmonella enterica and assess changes in their antimicrobial susceptibility in Kathmandu over a 23-year period. Methods: We conducted a retrospective analysis of standardised microbiological data from April 1992 to December 2014 at a single healthcare facility in Kathmandu, examining time trends of Salmonella-associated bacteraemia and the corresponding antimicrobial susceptibility profiles of the isolated organisms. Results: Over 23 years there were 30,353 positive blood cultures. Salmonella enterica accounted for 65.4% (19,857/30,353) of all the bacteria positive blood cultures. S. Typhi and S. Paratyphi A were the dominant serovars, constituting 68.5% (13,592/19,857) and 30.5% (6,057/19,857) of all isolated Salmonellae. We observed (i) a peak in the number of Salmonella-positive cultures in 2002, a year of heavy rainfall and flooding in the Kathmandu Valley, followed by a decline toward pre-flood baseline by 2014, (ii) an increase in the proportion of S. Paratyphi in all Salmonella-positive cultures between 1992 and 2014, (iii) a decrease in the prevalence of MDR for both S. Typhi and S. Paratyphi, and (iv) a recent increase in fluoroquinolone non-susceptibility in both S. Typhi and S. Paratyphi isolates. Conclusions: Our work describes significant changes in the epidemiology of Salmonella enterica in the Kathmandu Valley during the last quarter of a century. We highlight the need to examine current treatment protocols for enteric fever and suggest a change from fluoroquinolone monotherapy to combination therapies of macrolides or cephalosporins along with older first-line antimicrobials that have regained their efficacy. The GeneXpert MTB/RIF assay (Xpert) is a novel automated diagnostic tool for tuberculosis but its optimal placement in the healthcare system has not been determined. The objective of this study was to determine the possibility of additional case detection for pulmonary tuberculosis (PTB) by offering Xpert to smear-negative patients in a low-HIV burden setting with no Mycobacterium tuberculosis (M.tb.) culture facilities. Patients routinely presenting with symptoms suggestive of PTB with negative smears were offered single Xpert test on a fee-paying basis. Data were retrospectively reviewed to determine case detection in patients tested from February to December 2013. Symptoms associated with a positive test were analysed to determine if refinement of clinical criteria would reduce unnecessary testing. 258 smear-negative patients were included and M.tb. was detected in 55 (21.32%, n=55/258). Using standard clinical assessment for selection, testing 5 patients detected one case of smear-negative PTB. These results demonstrate that fee-paying Xpert service in low-income setting can increase TB case confirmation substantially and further systematic studies of health economic implications should be conducted to determine optimal implementation models to increase access to Xpert in low- and middle-income countries. Background: In many rural areas at risk for enteric fever, there are few data on Salmonella enterica serotypes Typhi (S. Typhi) and Paratyphi (S. Paratyphi) incidence, due to limited laboratory capacity for microbiologic culture. Here, we describe an approach that permits recovery of the causative agents of enteric fever in such settings. This approach involves the use of an electricity-free incubator based upon use of phase-change materials. We compared this against conventional blood culture for detection of typhoidal Salmonella. Methodology/Principal Findings: Three hundred and four patients with undifferentiated fever attending the outpatient and emergency departments of a public hospital in the Kathmandu Valley of Nepal were recruited. Conventional blood culture was compared against an electricity-free culture approach. Blood from 66 (21.7%) patients tested positive for a Gram-negative bacterium by at least one of the two methods. Sixty-five (21.4%) patients tested blood culture positive for S. Typhi (30; 9.9%) or S. Paratyphi A (35; 11.5%). From the 65 individuals with culture-confirmed enteric fever, 55 (84.6%) were identified by the conventional blood culture and 60 (92.3%) were identified by the experimental method. Median time-to-positivity was 2 days for both procedures. The experimental approach was falsely positive due to probable skin contaminants in 2 of 239 individuals (0.8%). The percentages of positive and negative agreement for diagnosis of enteric fever were 90.9% (95% CI: 80.0%–97.0%) and 96.0% (92.7%–98.1%), respectively. After initial incubation, Salmonella isolates could be readily recovered from blood culture bottles maintained at room temperature for six months. Conclusions/Significance: A simple culture approach based upon a phase-change incubator can be used to isolate agents of enteric fever. This approach could be used as a surveillance tool to assess incidence and drug resistance of the etiologic agents of enteric fever in settings without reliable local access to electricity or local diagnostic microbiology laboratories. Author Summary: Every year, 20 million people worldwide suffer from typhoid, a bacterial infection spread by contaminated food and water, and over 200,000 die from the infection. However, few data are available on the prevalence and antimicrobial resistance profiles of the causative agents of typhoid, especially in settings without reliable access to laboratories and electricity. Here, we describe an approach that permits recovery of the causative agents of typhoid that requires no electricity, laboratory infrastructure, or specialized laboratory personnel at the site of patient contact. This approach involves the use of an electricity-free incubator, consisting of an insulated container and reusable packets filled with a chemical that upon warming in hot water or direct sun, maintains 38°C for 24 hours. We used blood culture bottles with a color indicator that signals growth of bacteria and an antibiotic that selects for Gram-negative bacteria. We validated this approach in a clinical study among individuals with fever presenting to a hospital in urban Nepal, demonstrating that the approach performed comparably to conventional blood culture for isolating the bacteria causing typhoid. Background: Multi-drug resistance (MDR) and extensive-drug resistance (XDR) associated with extended-spectrum beta-lactamases (ESBLs) and carbapenemases in Gram-negative bacteria are global public health concerns. Data on circulating antimicrobial resistance (AMR) genes in Gram-negative bacteria and their correlation with MDR and ESBL phenotypes from Nepal is scarce. Methods: A retrospective study was performed investigating the distribution of ESBL and carbapenemase genes and their potential association with ESBL and MDR phenotypes in E. coli, Klebsiella spp., Enterobacter spp. and Acinetobacter spp. isolated in a major tertiary hospital in Kathmandu, Nepal, between 2012 and 2018. Results: During this period, the hospital isolated 719 E. coli, 532 Klebsiella spp., 520 Enterobacter spp. and 382 Acinetobacter spp.; 1955/2153 (90.1%) of isolates were MDR and half (1080/2153) were ESBL producers. Upon PCR amplification, blaTEM (1281/1771; 72%), blaCTXM-1 (930/1771; 53%) and blaCTXM-8 (419/1771; 24%) were the most prevalent ESBL genes in the enteric bacilli. BlaOXA and blaOXA-51 were the most common blaOXA family genes in the enteric bacilli (918/1771; 25%) and Acinetobacter spp. (218/382; 57%) respectively. Sixteen percent (342/2153) of all isolates and 20% (357/1771) of enteric bacilli harboured blaNDM-1 and blaKPC carbapenemase genes respectively. Of enteric bacilli, Enterobacter spp. was the most frequently positive for blaKPC gene (201/337; 60%). The presence of each blaCTX-M and blaOXA were significantly associated with non-susceptibility to third generation cephalosporins (OR 14.7, p < 0.001 and OR 2.3, p < 0.05, respectively).The presence of each blaTEM, blaCTXM and blaOXA family genes were significantly associated with ESBL positivity (OR 2.96, p < 0.001; OR 14.2, p < 0.001 and OR 1.3, p < 0.05 respectively) and being MDR (OR 1.96, p < 0.001; OR 5.9, p < 0.001 and OR 2.3, p < 0.001 respectively). Conclusions: This study documents an alarming level of AMR with high prevalence of MDR ESBL- and carbapenemase-positive ESKAPE microorganisms in our clinical setting. These data suggest a scenario where the clinical management of infected patients is increasingly difficult and requires the use of last-resort antimicrobials, which in turn is likely to intensify the magnitude of global AMR crisis.
