The semislug Megaustenia siamensis, commonly found in Thailand, is notable for its exceptional capacity to produce biological adhesives, enabling it to adhere to tree leaves even during heavy rainfall. In this study, we generated the first reference genome for M. siamensis using a combination of three sequencing technologies: Illumina's short-read, Pac-Bio's HIFI long-read, and Hi-C. The assembled genome size was 2593 billion base pairs (bp), containing 34,882 protein-coding genes. Our analysis revealed positive selection in pathways associated with the ubiquitin–proteasome system. Furthermore, RNA sequencing of foot and mantle tissues unveiled the primary constituents of the adhesive, including lectin-like proteins (C-lectin, H-lectin, and C1q) and matrilin-like proteins (VWA and EGF). Additionally, antimicrobial peptides were identified. The comprehensive M. siamensis genome and tissue-specific transcriptomic data provided here offer valuable resources for understanding its biology and exploring potential medical applications. The mechanism of bioluminescence in the luminous land snails remains largely unknown. Here, we analyzed basic biochemical properties of the luminous land snail, Quantula weinkauffiana. The luminescence activity was extracted from the light organ located near the mouth using a neutral buffer containing detergent. The reaction of the crude buffer extract was triggered by the addition of only hydrogen peroxide (H2O2). These results are inconsistent with the single precedent report on the bioluminescence in the Quantula striata by Shimomura and Haneda in 1986, in which the luminescence of the buffer extract (without detergent) from the light organ was induced by the coaddition of three indispensable components H2O2, ferrous ion, and 2‐mercaptoethanol. Based on the present findings, we suggested that an insoluble photoprotein is involved in the bioluminescence of the luminous land snails and the luminescence reaction is simply triggered by H2O2. Land snails were collected for the project ‘Conserving Myanmar’s Karst Biodiversity’ from the limestone karsts in Mon, Kayin, and Shan states and in the regions of Tanintharyi and Mandalay between 2015 and 2017, through cooperation with Fauna and Flora International (FFI) and the Forestry Department of Myanmar. Here, we report on a portion of the collection, and list 17 species from seven genera of the Hypselostomatidae microsnails. Three new species from two genera are described as Bensonella taiyaiorum Tongkerd & Panha, sp. nov., B. lophiodera Tongkerd & Panha, sp. nov., and Gyliotrachela aunglini Tongkerd & Panha, sp. nov. All new species are known only from the type locality in Shan State (Bensonella) and Kayin State (Gyliotrachela). A new combination of Acinolaemus dayanum and three newly recorded species, namely A. cryptidentatus, B. anguloobtusa and G. hungerfordiana are discussed. The low morphological variability of the widely distributed G. hungerfordiana is discussed, and two species are proposed for formal synonymisation. The mysterious world of the bioluminescent molluscs in terrestrial ecosystems is mesmerizing, but Quantula striata was previously the only terrestrial mollusc known to be luminescent. Here, we document the new discovery of bioluminescence in four land snails, namely Phuphania crossei, P. globosa, P. carinata, and P. costata. Our observations establish clearly that these four species of Phuphania produce a continuous greenish light from the light-emitting cells located within the mantle and the foot, and that its bright luminescence is intracellular and is not due to any luminous secretion. Although both Quantula and Phuphania can produce a green light, the luminescence patterns are different. The luminescence displayed by Quantula is rhythmical blinking or flashing, while Phuphania glows continuously. The Euconulidae is a globally distributed land snail family but there is no record of this family from Thailand. In this study, we describe a new genus, Siamoconus gen. nov., based on comparative studies of the shell characteristics, external morphology of animals, radula, genital and spermatophore structures, and molecular phylogeny. We performed phylogenetic analyses of two mitochondrial gene fragment (cytochrome c oxidase I, COI; and 16S rRNA) and one nuclear (28S rRNA) gene fragment to clarify the relationships with other euconulid genera and its position in the superfamily Trochomorphoidea. We also analysed shell morphometrics, male genitalia and mantle pigmentation of Siamoconus gen. nov., confirming the status of three new species with keeled shells (S. boreas sp. nov., S. coleus sp. nov. and S. geotrochoides sp. nov.) and one new species with a rounded shell (S. destitutus sp. nov.). These new species are restricted to limestone areas in northern and north-eastern Thailand. We also re-examined the status of other genera in the superfamily Trochomorphoidea based on shell features, body pigmentation, radula, genital anatomy, spermatophore morphology, and a phylogenetic analysis of all available trochomorphoidean DNA sequences. Our analysis suggests that the family Geotrochidae, previously synonymised with the Trochomorphidae, should be resurrected to represent the genus Geotrochus from Borneo, and this family is retrieved as the sister clade of the Dyakiidae. Salween River basin’s karst ecosystems in southeastern Myanmar remain largely unexplored and are likely to harbour a high terrestrial snail diversity that are often associated with high levels of snail endemism. Here, an outstanding group of new karst-associated terrestrial snails, Burmochlamys gen. nov., are discovered. A study of the comparative morphological and anatomical data reveals that the reproductive tract and radula of this new genus are closely related to the helicarionid genus Sophina Benson, 1859 but shell morphology (shape, size, and sculpture) and mantle extensions are distinct from the latter genus. Burmochlamys gen. nov. now consists of four known nominal species, B. cassidula comb. nov., B. cauisa comb. nov., B. perpaula comb. nov., and B. poongee comb. nov., and five new species; B. albida sp. nov., B. fasciola sp. nov., B. moulmeinica sp. nov., B. versicolor sp. nov., and B. whitteni sp. nov. The highlight is that the members of the new genus show site-specific endemism, being restricted to karstic habitat islands of the Salween River basin. In addition, the discovery supports that the unique and complex structure of Salween River basin’s karst ecosystems are habitats in which the terrestrial malacofauna have speciated and become endemic. Knowledge of Thai semislugs remains scarce, especially the dancing semislug genus Cryptosemelus. Prior to the present study, only a single species has been recognized with little available information. To address this knowledge gap, we surveyed for semislugs in western and southern Thailand, which yielded three species belonging to the genus Cryptosemelus. The little-known type species C. gracilis is redescribed herein, including a comparison with the type specimens. Two additional species, C. betarmon sp. nov. and C. tigrinus sp. nov., are described as new to science. All three species are characterized by differences in their genital anatomy, especially with respect to anatomical details of the penis, epiphallus, and spermatophore. An enigmatic land snail species restricted to the karst ecosystem in southern Cambodia and formerly placed in the family Ariophantidae is now reclassified into the family Dyakiidae. Its current systematic position is based on morpho-anatomical characters combined with a molecular phylogeny based on two partial mitochondrial markers (COI and 16S rDNA) and a nuclear gene fragment (28S rDNA). Our results indicate that Khmerquantula gen. nov. is a member of the Dyakiidae with very strong support from both morphological and molecular phylogenetic analyses. The diagnostic characters of this new genus and new species, Khmerquantula leipo are a dextral shell with a well-polished surface, animal with only three dorsal lobes, and reproductive organ without penial verge and amatorial organ papilla. Members of the terrestrial snail family Dyakiidae from Southeast Asia show a distinct geographical distribution pattern and possess different degrees of complexity in their amatorial organ gland. This study is the first molecular phylogeny of ten of the 12 genera in this family, performed to provide insights into the origin of Dyakiidae and the evolution of their shells and amatorial organ gland structure. A new genus and new species, Pseudoquantula lenticularis Jirapatrasilp & Panha gen. & sp. nov. , was uncovered based on its distinct morphological characters and molecular divergence. All other genera were retrieved as monophyletic except for Dyakia. Mainland Southeast Asia was inferred to be the ancestral range of the Dyakiidae, and the lineages then dispersed to and diversified in Borneo. Cladistic analysis showed that all 14 morphological characters used in this study were homoplastic. These results disagree with the previous amatorial organ transformation series, in which neither Pseudoplecta nor Quantula was ancestral to the other genera. Sophina is a poorly known and neglected genus due to the inaccessibility of the Salween Basin, Southern Myanmar. Taxonomic status, distribution, and phylogeny are being revised based on an integrative analysis of genitalia, radula, and molecular data. Morphological variation in shells and genitalia, together with a phylogenetic tree from concatenated data of mitochondrial and nuclear genes, revealed the existence of ten species/subspecies. Penial morphology and genetic divergence were generally consistent and useful in delimiting species, while shell characters showed little overall taxonomic utility in some species. Taxonomic placement of the previous subspecies "bensoni" shows clear distinction in both genitalia and molecular evidence, and is currently recognized as a distinct species. The nominal species "S. schistostelis" and "S. calias" possess similar genitalia and shell morphology, and molecular evidence suggested that they are sister taxa representing geographically isolated populations. Four new species are additionally described herein as S. furfuracea n. sp., S. pisinna n. sp., S. salweenica n. sp., and S. tonywhitteni n. sp. based on both morphology and molecular evidence. Phylogenetic analyses supported monophyly of Sophina, and further a split into two principal clades. These two clades showed little difference in genitalia, but more clear differences in the umbilical area and allopatric distribution in upper and lower reaches of the Gyaing River. High genetic divergence was revealed and this coincided with remarkable degree of endemism and localization with a pattern of one outcrop for one lineage. These data highlight the importance of the Salween Basin's karst ecosystems as an evolutionary and endemic biodiversity hotspot, and indicate that a focus on conservation and management in this area is urgently required. Members of the land snail genus Sarika Godwin-Austen, 1907 are superficially similar and difficult to differentiate by their shell morphology so that their species limits are still unclear. In order to resolve the taxonomy of this group, a phylogenetic reconstruction of Sarika is presented, based on morphological and anatomical characters, as well as on partial sequences of the mitochondrial cytochrome c oxidase subunit I (COI) gene. In total, 23 species of Sarika are recognised in Thailand, and nine species are new to science, namely S. caligina Pholyotha & Panha, sp. nov., S. gratesi Pholyotha & Panha, sp. nov., S. inferospira Pholyotha & Panha, sp. nov., S. lactospira Pholyotha & Panha, sp. nov., S. megalogyne Pholyotha & Panha, sp. nov., S. melanospira Pholyotha & Panha, sp. nov., S. pellosa Pholyotha & Panha, sp. nov., S. solemi Pholyotha & Panha, sp. nov., and S. subheptagyra Pholyotha & Panha, sp. nov. Results from genital examination and COI analyses confirm the monophyly of Sarika and its species. The intra- and inter-specific genetic distances of Sarika were 0-3.7% and 4.6-12.0%, respectively. Colour images of the living adults, shell, and genitalia along with SEM images of the spermatophore and radula are given. The Tonle Sap Lake in Cambodia is a crucial freshwater biodiversity hotspot and supports one of the world's largest inland fisheries. Within the Tonle Sap basin, freshwater molluscs provide vital ecosystem services and are among the fauna targetted for commercial harvesting. Despite their importance, freshwater molluscs of the Tonle Sap basin remain poorly studied. The historical literature was reviewed and at least 153 species of freshwater molluscs have been previously recorded from throughout Cambodia, including 33 from the Tonle Sap basin. Surveys of the Tonle Sap Lake and surrounding watershed were also conducted and found 31 species, 15 bivalves (five families) and 16 gastropods (eight families), in the Tonle Sap basin, including three new records for Cambodia (Scaphula minuta, Novaculina siamensis, Wattebledia siamensis), the presence of globally invasive Pomacea maculata and potential pest species like Limnoperna fortunei. This study represents the most comprehensive documentation of freshwater molluscs of the Tonle Sap basin, and voucher specimens deposited at the Inland Fisheries Research and Development Institute, Cambodia, represent the first known reference collection of freshwater molluscs in the country. In order to combat the combined anthropogenic pressures, including invasive species, climate change and dams along the Mekong River, a multi-pronged approach is urgently required to study the biodiversity, ecology, ecosystem functioning of freshwater molluscs and other aquatic fauna in the Tonle Sap basin. Prior to this study, few collections and records were made of the land snails in Cambodia and the historical taxa had never been reviewed. Herein a report on the land snail diversity based on specimens collected recently from karstic and non-karstic areas in southern Cambodia is provided. This checklist presents 36 species of land snails (two Neritimorpha, six Caenogastropoda, and 28 Heterobranchia). Illustrations and brief taxonomic notes/remarks are provided for every species. We also described Georrisa carinata Sutcharit & Jirapatrasilp, sp. nov. based on some distinct shell morphological characters. Since the first descriptions during the colonial period in the nineteenth century, some land snail species (e.g., Trichochloritis norodomiana, Durgella russeola, Anceyoconcha siamensis obesula comb. nov., Anceyoconcha chaudoensis comb. nov., and Succinea tenuis) have not been reported subsequently. This probably reflects a lack of knowledge concerning land snail biodiversity in this country. To our knowledge, this is the first comprehensive survey of land snails in southern Cambodia. The land area of Laos is composed of a large variety of undisturbed habitats, such as high mountainous areas, huge limestone karsts and the lower Mekong Basin. Therefore, Laos is expected to have a high species diversity, especially for the land snails. However, with respect to research on malacology, Laos is probably the least well-researched area for land snail diversity in Indochina (including Laos) over the past few centuries. The handful of species lists have never been systematically revised from the colonial period to the present, so these classifications are outdated. Herein we present the first comprehensive annotated checklist with an up-to-date systematic framework of the land snail fauna in Laos based on both field investigations and literature surveys. This annotated checklist is collectively composed of 231 nominal species (62 'prosobranch' and 169 heterobranches), of which 221 nominal species are illustrated. The type specimens of 143 species from several museum collections and/or 144 species of newly collected specimens are illustrated. There are 58 species recorded as new to the malacofauna of the country, and two new replacement names are proposed as Hemiplecta lanxangnica Inkhavilay and Panha, nomen novum (Ariophantidae) and Chloritis khammouanensis Inkhavilay and Panha, nomen novum (Camaenidae). Four recently described species of the genus Amphidromus from Laos, "thakhekensis", "richgoldbergi", "attapeuensis" and "phuonglinhae" are synonymized with previously described species. In addition, thirteen nominal species are listed as uncertain records that may or may not occur in Laos. The taxonomic status of the limacoid genus Macrochlamys Gray, 1847 in Thailand has long been plagued by much ambiguity due to the inability to distinguish members from each other, and a few other genera, by shell morphology alone. The taxonomy of the Thai species is evaluated resulting in recognition of seven species, including five new species. The new species, Macrochlamys aurantia, M. coleus and M. tanymentula from Kanchanaburi Province (West Thailand), and M. caverna and M. lemma from Lopburi Province (Central Thailand), are distinguished and described based on shell, animal colouration, radula and genitalia characters. The status of Macrochlamys brunnea Möllendorff, 1902b is at present unclear. Although described from Bangkok, no material identifiable as this species was found in recent extensive surveys and it is provisionally retained in the genus without anatomical evaluation. Macrochlamys kelantanensis Möllendorff, 1902a is re-described with additional morphological and anatomical information. The five new species appear to have a restricted distribution in Western and Central Thailand, whereas M. kelantanensis is more wide-ranging, from Thailand to Malaysia. Detailed descriptions and illustrations of the shell morphology, external morphology, genital anatomy and radulae are provided. The results show that despite the shells being morphologically similar, aspects of classical morphology based taxonomy for Macrochlamys species, especially the genitalia morphology, remain useful and reliable for species delimitation.
