Honey has been recognized as a reliable indicator of environmental quality because of honeybees' intense foraging activity, which brings them into contact with many persistent organic pollutants around the hive. In this study, four types of honey (meadow, acacia, chestnut, and honey in comb) collected at three different locations were analyzed for Co, Cr, Cu, Fe, Mn, Pb, and Zn levels. The highest levels of Fe and Cu in chestnut honey, Co and Zn in meadow honey, and Pb in honey in comb were observed in Varaždin County. The lowest levels of Pb in meadow honey and Co in comb honey were found from apiaries in Sisak-Moslavina County. Significant differences in the mean concentrations of Cr, Cu, Mn, and Fe were observed among the four honey types. Conversely, no significant differences in Co, Pb, and Zn levels were found. Most of the significant differences between the elements are related to chestnut honey. While sampling location (Fe) and type of honey (Pb), or both (Cr and Zn), significantly influenced the concentrations of some elements, these factors were found to be irrelevant for Mn, Co, and Cu. Glyphosate is the most used herbicide in agriculture. Its major metabolite is AMPA (aminomethylphosphonic acid), but N-acetyl-AMPA and N-acetylglyphosate are also metabolites of interest. For risk assessment, a general residue definition was proposed as the sum of glyphosate, AMPA, N-acetyl-glyphosate and N-acetyl-AMPA, expressed as glyphosate. A confirmatory method for glyphosate in fat, liver and kidneys, as well as a confirmatory method for AMPA and N-acetyl-glyphosate in all matrices, are still missing. In this paper, we present a method for the quantitative determination of glyphosate residues and its metabolites AMPA, N-acetyl-AMPA and N-acetyl-glyphosate by liquid chromatography–mass spectrometry (LC-MS/MS) in adipose tissue, liver, eggs, milk and honey without derivatization. Different chromatographic columns were tested, with the Hypercarb column providing the best results. The analytes were eluted with mobile phases of acidified water with 1.2% formic acid and 0.5% formic acid in acetonitrile. Sample purification procedures were also optimized by varying the solvent extraction mixtures (water, methanol and mixture ψ (methanol, water) = 1:1, each with the addition of 1% formic acid (v/v)), using different sorbents in solid phase extraction (SPE) (polymeric cationic (PCX) and anionic (PAX)) and using dispersive solid phase extraction (dSPE) (C18 and PSA) by modifying the extraction procedures. Finally, the analytes were extracted from the samples with 1% formic acid in water (v/v). Milk and adipose tissue were purified by the addition of dichloromethane, while liver and egg samples were purified by SPE with a mixed cation exchange sorbent and ultrafiltration with cut-off filters. The proposed analytical procedures were validated according to SANTE/11312/2021 guidelines: linearity, limits of quantification, precision and accuracy were determined for all matrices. The limits of quantification (LOQs) ranged from 0.025 to 0.2 mg kg−1. Precision, expressed as relative standard deviation, was <20%, while accuracy, expressed as analytical recovery, ranged from 70% to 120%. During method validation, the measurement uncertainty was estimated to be <50% for all analytes. Good validation parameters according to the SANTE document were achieved for all analytes. Therefore, the method can be considered reliable and sensitive enough for routine monitoring of polar pesticides. The application of the accredited method in routine analysis will provide data that are useful for the re-evaluation of risk assessment studies in foods of animal origin. [ABSTRACT FROM AUTHOR]

This study was conducted to evaluate withdrawal time of levamisole in eggs after oral administration in laying hens at different doses. Sampling of eggs was conducted for 37 days after the end of treatment, and levamisole concentrations were measured by liquid chromatography-tandem mass spectrometry validated according to the Commission Decision 2002/657/EC. Estimated validation parameters were as follows: decision limit, 0.54 μg/kg; detection capability, 0.56 μg/kg; limit of detection, 0.04 μg/kg; limit of quantification, 0.15 μg/kg; accuracy (recovery), between 92.9 and 102.3%; precision (relative standard deviation), ≤4.62%; and within-laboratory precision (relative standard deviation), ≤5.19%. Levamisole residue levels were significantly higher in egg yolks than in egg whites. The highest levels of levamisole were detected on day 2 posttreatment in groups receiving 50 mg/kg of body weight (556.2 μg/kg in egg yolks and 166.5 μg/kg in egg whites). Significant elimination occurred within 5 days after the cessation of treatment in all groups, with an elimination half-life of 1.3 days. Levamisole was still detectable on day 30 after the end of treatment in egg whites (0.06 μg/kg) and on day 37 in egg yolks (0.06 μg/kg). The longest withdrawal time for levamisole in eggs (14.9 days) was determined in a group treated with 25 mg of levamisole per kg of body weight for two consecutive days. According to the results, oral treatment of laying hens with levamisole may result in noncompliant egg samples even 14 days after treatment. [ABSTRACT FROM AUTHOR]

Honey is an important food supplement in the human diet because of its nutritional quality. Therefore, honey should be safe for human consumption. The purpose of this research was to evaluate physicochemical and microbiological properties of 64 honey samples. The physicochemical analysis (water content, hydroxymethylfurfural, electrical conductivity and diastase activity) showed that 95.31% of samples meet Croatian and European standards. Microbiological quality of tested samples was considered good with no pathogenic bacteria detected. In 34.38% of the samples, the yeast and mold count exceeded the limit established by legislation. The presence of yeasts was noted in 17 samples varying from 18 to 1300 CFU/g, whereas mold count ranged from 18 to 182 CFU/g. The study revealed that out of six genera of molds recovered, Cladosporium was the most frequent, followed by Alternaria, Penicillium, Mucor, Aureobasidium, and Stachybotrys. Considering that the majority of molds identified in this study are commonly found in bee hive environment and in digestive tract of honey bees, it can be concluded that the mold contamination of honey in this study derives mainly from primary sources. The presence of Stachybotrys sp. is an indicator of contamination that comes from secondary sources. Practical applications: Due to continuous expansion of the world honey market and increasing awareness of consumers towards healthy food, the importance of quality control of honey has also grown. In respond to these demands, this study presents physicochemical and microbiological properties of different types of Croatian honey, with special emphasis laid on the identification of the present molds in incompatible samples. The results of this study provide useful data that Croatian honey has good level of quality and contribute to better understanding of the genera of molds present in the honey from this region.
