Volatile amines are usually problematic compounds in sampling, sample pretreatment and gas chromatographic analysis due to their chemical characteristics (polarity, basicity and reactivity). Headspace solid-phase microextraction (SPME) Arrow sampling of aqueous samples were proven to be complicated since moisture in the headspace was also sorbed into the SPME sorbent and resulted in distorted or split peaks for the volatile amines. This was the case especially with old used sorbents not so much with the new ones. Volume of the water sample, sampling conditions, quality of the SPME sorbent and desorption conditions greatly influenced the concentration of water in the headspace and in the sorbent phase. This, in turn, affected the length of the water film in the column which determined the degree of peak splitting and distance between the split amine peaks (water film trapped part of the amine molecules). Addition of the salt to the sample solution and additional drying of the SPME sorbent after the sampling were shown to effectively decrease the amount of water in the headspace and in the sorbent phase. This combined effect of salt addition and drying step resulted in much better peak shapes and intensities for the amines. Biogenic volatile organic compounds (BVOCs) emitted by terrestrial vegetation participate in a diversity of natural processes. These compounds impact both shortrange processes, such as on plant protection and communication, and long-range processes, for example by participating in aerosol particle formation and growth. The biodiversity of plant species around the Earth, the vast assortment of emitted BVOCs, and their trace atmospheric concentrations contribute to the substantial remaining uncertainties about the effects of these compounds on atmospheric chemistry and physics, and call for the development of novel collection devices that can offer portability with improved selectivity and capacity. In this study, a novel solid-phase microextraction (SPME) Arrow sampling system was used for the static and dynamic collection of BVOCs from a boreal forest, and samples were subsequently analyzed on site by gas chromatography-mass spectrometry (GC-MS). This system offers higher sampling capacity and improved robustness when compared to traditional equilibrium-based SPME techniques, such as SPME fibers. Field measurements were performed in summer 2017 at the Station for Measuring Ecosystem-Atmosphere Relations (SMEAR II) in Hyytiälä, Finland. Complementary laboratory tests were also performed to compare the SPME-based techniques under controlled experimental conditions and to evaluate the effect of temperature and relative humidity on their extraction performance. The most abundant monoterpenes and aldehydes were successfully collected. A significant improvement on sampling capacity was observed with the new SPME Arrow system over SPME fibers, with collected amounts being approximately 2× higher for monoterpenes and 7-8× higher for aldehydes. BVOC species exhibited different affinities for the type of sorbent materials used (polydimethylsiloxane (PDMS)-carbon wide range (WR) vs. PDMS-divinylbenzene (DVB)). Higher extraction efficiencies were obtained with dynamic collection prior to equilibrium regime, but this benefit during the field measurements was small, probably due to the natural agitation provided by the wind. An increase in temperature and relative humidity caused a decrease in the amounts of analytes extracted under controlled experimental conditions, even though the effect was more significant for PDMS-carbon WR than for PDMS-DVB. Bioaerosols are ubiquitous in the atmosphere and constitute ∼ 30 % of atmospheric aerosol particle mass in sizes > 1 μm. Bioaerosol components, such as bacteria, fungi and pollen, may affect the climate by acting as could-active particles, thus having an effect on cloud and precipitation formation processes. In this study, size-segregated aerosol samples (< 1.0, 1-2.5, 2.5-10 and > 10 μm) were collected in boreal forest (Hyytiälä, Finland) during one year and analyzed for free amino acids (FAAs), DNA concentration and microorganism (bacteria, Pseudomonas and fungi). Measurements were performed using tandem mass spectrometry, spectrophotometry and qPCR, respectively. Meteorological parameters and statistical analysis were used to study their atmospheric implication for results. Distinct annual patterns of bioaerosol components were observed, late spring and autumn being seasons of dominant occurrence. Elevated abundances of FAAs and bacteria were observed during the local pollen season, whereas fungi were observed at highest level during autumn. Meteorological parameters, such as air and soil temperature, radiation and rainfall were observed to possess close relationship with bioaerosol abundances on an annual scale. [ABSTRACT FROM AUTHOR]

Alkylamines are important precursors in secondary aerosol formation in the boreal forest atmosphere. To better understand the behavior and sources of two alkylamines, dimethylamine (DMA) and diethylamine (DEA), we estimated the magnitudes of soil-atmosphere fluxes of DMA and DEA using a gradient-diffusion approximation based on measured concentrations in soil solution and in the canopy air space. The ambient air concentration of DMA used in this study was a sum of DMA and ethylamine. To compute the amine fluxes, we first estimated the soil air space concentration from the measured soil solution amine concentration using soil physical (temperature, soil water content) and chemical (pH) state variables. Then, we used the resistance analogy to account for gas transport mechanisms in the soil, soil boundary layer, and canopy air space. The resulting flux estimates revealed that the boreal forest soil with a typical long-term mean pH?5.3 is a possible source of DMA (170±?51nmolm-2day-1) and a sink of DEA (-1.2±1.2?nmolm-2day-1). We also investigated the potential role of fungi as a reservoir for alkylamines in boreal forest soil. We found high DMA and DEA concentrations both in fungal hyphae collected from field humus samples and in fungal pure cultures. The highest DMA and DEA concentrations were found in fungal strains belonging to decay and ectomycorrhizal fungal groups, indicating that boreal forest soil and, in particular, fungal biomass may be important reservoirs for these alkylamines. Emerging organic contaminants in wastewater are usually analyzed by targeted approaches, and especially estrogens have been the focus of environmental research due to their high hormonal activity. The selection of specific target compounds means, however, that most of the sample components, including transformation products and potential new contaminants, are neglected. In this study, the fate of steroidal compounds in wastewater treatment processes was evaluated by a nontargeted approach based on comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry. The potential of the nontargeted approach to generate comprehensive information about sample constituents was demonstrated with use of statistical tools. Transformation pathways of the tentatively identified compounds with steroidal four-ring structure were proposed. The purification efficiency of the wastewater treatment plants was studied, and the distribution of the compounds of interest in the suspended solids, effluent water, and sludge was measured. The results showed that, owing to strong adsorption of hydrophobic compounds onto the solid matter, the steroids were mostly bound to the suspended solids of the effluent water and the sewage sludge at the end of the treatment process. The most abundant steroid class was androstanes in the aqueous phase and cholestanes in the solid phase. 17β-estradiol was the most abundant estrogen in the aqueous phase, but it was only detected in the influent samples indicating efficient removal during the treatment process. Volatile organic compounds (VOCs) play a key role in atmospheric chemistry and physics. They participate in photochemical reactions in the atmosphere, which have direct implications on climate through, e.g. aerosol particle formation. Forests are important sources of VOCs, and the limited resources and infrastructures often found in many remote environments call for the development of portable devices. In this research, the potential of needle trap microextraction and portable gas chromatography-mass spectrometry for the study of VOCs at forest site was evaluated. Measurements were performed in summer and autumn 2014 at the Station for Measuring Ecosystem-Atmosphere Relations (SMEAR II) in Hyytiälä, Finland. During the first part of the campaign (summer) the applicability of the developed method was tested for the determination of monoterpenes, pinonaldehyde, aldehydes, amines and anthropogenic compounds. The temporal variation of aerosol precursors was determined, and evaluated against temperature and aerosol number concentration data. The most abundant monoterpenes, pinonaldehyde and aldehydes were successfully measured, their relative amounts being lower during days when particle number concentration was higher. Ethylbenzene, p- and m-xylene were also found when wind direction was from cities with substantial anthropogenic activity. An accumulation of VOCs in the snow cover was observed in the autumn campaign. Results demonstrated the successful applicability of needle trap microextraction and portable gas chromatography-mass spectrometry for the rapid in situ determination of organic gaseous compounds in the atmosphere. Atmospheric aerosol particles of primary or secondary, biogenic or anthropogenic origin are highly complex samples of changing composition in time and space. To assess their effects on climate or human health, the size-dependent chemical composition of these ubiquitous atmospheric constituents must be known. The development of novel analytical methods has enabled more detailed characterization of the organic composition of aerosols. This review gives an overview of the methods used in the chemical characterization of atmospheric aerosol particles, with a focus on mass-spectrometry techniques for organic compounds, either alone or in combination with chromatographic separation. Off-line, on-site, and on-line methods are covered, and the advantages and limitations of the different methods are discussed. The main emphasis is on methods used for detailed characterization of the composition of the organic compounds in aerosol particles. We address and summarize the current state of analytical methods used in aerosol research and discuss the importance of developing novel sampling strategies and analytical instrumentation. RATIONALE: On-line chemical characterization methods of atmospheric aerosols are essential to increase our understanding of physicochemical processes in the atmosphere, and to study biosphere-atmosphere interactions. Several techniques, including aerosol mass spectrometry, are nowadays available, but they all suffer from some disadvantages. In this research, desorption atmospheric pressure photoionization high-resolution (Orbitrap) mass spectrometry (DAPPIHRMS) is introduced as a complementary technique for the fast analysis of aerosol chemical composition without the need for sample preparation. METHODS: Atmospheric aerosols from city air were collected on a filter, desorbed in a DAPPI source with a hot stream of toluene and nitrogen, and ionized using a vacuum ultraviolet lamp at atmospheric pressure. To study the applicability of the technique for ambient aerosol analysis, several samples were collected onto filters and analyzed, with the focus being on selected organic acids. To compare the DAPPI-HRMS data with results obtained by an established method, each filter sample was divided into two equal parts, and the second half of the filter was extracted and analyzed by liquid chromatography/mass spectrometry (LC/MS). RESULTS: The DAPPI results agreed with the measured aerosol particle number. In addition to the targeted acids, the LC/MS and DAPPI-HRMS methods were found to detect different compounds, thus providing complementary information about the aerosol samples. CONCLUSIONS: DAPPI-HRMS showed several important oxidation products of terpenes, and numerous compounds were tentatively identified. Thanks to the soft ionization, high mass resolution, fast analysis, simplicity and on-line applicability, the proposed methodology has high potential in the field of atmospheric research. A complete methodology based on LC-anisole-toluene dopant-assisted atmospheric pressure photoionization-IT-MS was developed for the determination of aldehydes in atmospheric aerosol particles. For the derivatization, ultrasound was used to accelerate the reaction between the target analytes and 2,4-dinitrophenylhydrazine. The developed methodology was validated for three different samples, gas phase, ultrafine (Dp = 30 ± 4 nm; where Dp stands for particle diameter) and all-sized particles, collected on Teflon filters. The method quantitation limits ranged from 5 to 227 pg. The accuracy and the potential matrix effects were evaluated using standard addition methodology. Recoveries ranged between 91.7 and 109.9%, and the repeatability and the reproducibility of the method developed between 0.5 and 8.0% and between 2.9 and 11.1%, respectively. The results obtained by the developed methodology compared to those provided by the previously validated method revealed no statistical differences. The method developed was applied to the determination of aldehydes in 16 atmospheric aerosol samples (30 nm and all-sized samples) collected at the Station for Measuring Forest Ecosystem-Atmosphere Relations II during spring 2011. Comprehensive two dimensional gas chromatography-time-of-flight mass spectrometry (GC×GC-TOF-MS) was used for screening and semiquantitation of semivolatile organic compounds in aerosol particles. As the volatility was a prerequisite parameter for the analysis, some compounds were transformed via derivatization such as silylation into more volatile ones. The identification of the analytes was made by comparing the GC retention indices and the TOF mass spectra with the NIST and the Golm metabolome database reference libraries. The data treatment was simplified by exploiting an additional classification of the identified compounds, namely the main functional group or specific element present in the molecule leading to different groups of compounds. This methodology was applied to identify compounds in 30 ± 4 nm, 50 ± 5 nm and total suspended particles (TSP) collected during spring and autumn of 2009 and summer of 2010 at the Station for Measuring Forest Ecosystem Atmosphere Relations (SMEAR II) at Hyytiälä (Finland). The number of identified compounds was higher than 400, which were the most relevant compounds present in the samples, in terms of concentrations. The analysis of aerosol particles of different sizes, collected simultaneously, revealed that the number of compounds increased with the particle size whereas the normalized response factor decreased in most of the cases, aldehydes being an exception. Interactions between Intralipid dispersion and local anesthetics (bupivacaine, prilocaine, and lidocaine) were investigated. The amount of bupivacaine (the most cardiotoxic analyte of the local anesthetics studied) entrapped in Intralipid in the presence of plasma was studied using an off-line filtration and solid phase extraction method combined with capillary zone electrophoresis for quantification of free unbound bupivacaine. To confirm interactions between the analytes and Intralipid at lower concentrations, direct injection mass spectrometry was used. The use of immobilized Intralipid chromatography-atmospheric pressure ionization–ion trap mass spectrometry in the study of interactions between drugs and Intralipid dispersion is demonstrated. Finally, interactions between Intralipid dispersion and local anesthetics were investigated by electrokinetic capillary chromatography. The electrophoretic mobility of the Intralipid dispersed phase was calculated using the iterative procedure and a homologous series of alkyl phenyl benzoates (C1–C6), and the retention factors for the analytes were determined. Oxidation of β-caryophyllene with ozone followed by reduction was carried out in dichloromethane. Reaction led to the formation of β-caryophyllene aldehyde (11%, purity 90.8%) and β-nocaryophyllone aldehyde (79%, purity 99%). Compounds were purified and separated by liquid–liquid extraction and flash chromatography. Identifications were achieved by electron-impact and electrospray ionization mass spectrometry, and precise structures were confirmed by one and two-dimensional nuclear magnetic resonance spectroscopy. The prepared compounds were used in the analysis of ambient aerosol samples collected during spring 2003 at Hyytiälä, Finland. β-Nocaryophyllone aldehyde was positively identified and quantified in ambient aerosol samples for the first time. The amount of quantified β-nocaryophyllone aldehyde of aerosol origin was 17.4 ± 1.0 ng m−3 of sampled air. Comparison with previous findings led to the conclusion that oxidation products of β-caryophyllene participate in biogenic aerosol formation over boreal forest during late spring. The use of biochar (BC) has been suggested for remediation of contaminated soils. This study aims to investigate the role of microorganisms in sorption of PAH to BC-amended soils. Fungi, especially the wood and litter-degrading fungi, have shown the ability for humification and to degrade recalcitrant molecules, and are thus suitable model organisms. Haplic Arenosol with high organic matter content was chosen to highlight the importance of soil organic matter (SOM) in PAH sorption, possibly to form non-extractable residue. Basidiomycetous fungi Agrocybe praecox and Phanerochaete velutina grown on pine bark were inoculated in organic matter (OM)-rich Haplic Arenosol and OM-poor sandy loam with either BC or chemically activated BC (ABC) and 14C-labelled pyrene for 60 days. Fungi did not mineralize pyrene, but increased sorption up to 47-56% in BC-amended Haplic Arenosol in comparison with controls (13-25%) without a fungus irrespective of the presence of an adsorbent. In OM-poor sandy loam, only 9-12% of pyrene was sorbed to amended soil in the presence of fungus and adsorbent. The results suggest that BC and fungal amendment increased sorption of pyrene, especially to Haplic Arenosol more than by either BC or fungi alone. [ABSTRACT FROM AUTHOR]
