Background: Folk biology commonly contains knowledge of many more taxa than those of immediate economic importance. Species with little or no practical use are, however, often overlooked by ethnobiological research. An example are a few Myxomycetes taxa which played an important role in the folk biology and beliefs of pre-industrial Sweden and adjacent Nordic countries, Denmark, Norway and Finland. Such organisms are not of less interest for the understanding of human-biota relationships; local knowledge about the entire biota in a given environment should therefore be studied to comprehend the full range of folk biology. Methodology: This qualitative study analyses and reviews historical data available in archives and published ethnographic collections as well as scattered and fragmentary notes in the literature using a historical ethnobiological approach. Results: Peasants in the Nordic countries recognized three taxa of myxomycetes. Scrambled egg slime, Fuligo septica (L.) F.H.Wigg., in particular attracted interest and is known by many local folk names. This slime had no practical value or use, yet it was well known in folk biology and associated with a supernatural malevolent being which in the shape of a hare or cat stole milk or butter on behalf of a witch. The belief in evil origins of slime became the cause of violent actions such as whipping and burning of the organism. Two other taxa, Lycogala epidendron (L.) Fr. and Mucilago crustacea F.H.Wigg., have also been observed in folk biology, but data on how they were perceived and treated is sparse. Conclusions: The sudden appearance of strange shapes and clear colours of myxomycetes in damp weather created both fear and curiosity; these odd organisms required explanations, interpretations and actions. Our example of the economically irrelevant myxomycetes in Sweden and nearby Nordic countries indicates that interpretations in pre-industrial societies of natural phenomena and various organisms, connections with beliefs and perceptions about the surrounding environment and the possible consequent actions should be studied alongside economic plants and animals in ethnobiological research, for a deeper understanding about folk biology and the multilayered and multidimensional relationships between humans and biota. Babesia spp. and Theileria spp. are tick-borne apicomplexan protozoa that can cause disease in animals and humans. Deer are considered reservoirs for a wide variety of Piroplasmida species, including some potentially zoonotic. This study aimed to investigate the occurrence and genetic diversity of piroplasmids in wild deer sampled in four Brazilian states (São Paulo, Mato Grosso do Sul, Paraná and Goiás). For this purpose, extracted DNA samples from 181 deer buffy coat samples (138 Blastocerus dichotomus, 26 Subulo gouazoubira, 4 Mazama jucunda, 3 Mazama rufa and 10 Ozotocerus bezoarticus) were subjected to a nested PCR (nPCR) assay based on the 18S rRNA gene in order to perform a screening for piroplasmids and characterized based on the near-complete 18S rRNA, hsp70 and cox-3 genes. As a result, 75.14% (136/181) samples were positive for piroplasmids. Of these, 108 (79.41%), 101 (74.26%) and 67 (49.26%) were positive to near complete 18S rRNA, hsp70 and cox-3 genes, respectively. Phylogenetic analyses based on three molecular markers showed similar topology to each other. All sequences obtained in the present study were positioned into the Theileria sensu stricto clade, forming a distinct clade, albeit close to T. cervi. Most sequences grouped together into a large clade divided into subclades, which were often related to deer genus/species, showing that Theileria lineages seemed to show specificity according to deer genus/species. Two 18S rRNA sequences (one obtained from S. gouazoubira and another from M. jucunda) were positioned into a different clade, apart from other sequences detected in this study, indicating that different species of Theileria occur in deer from Brazil. Two subclusters were observed in the phylogenetic analysis based on the hsp70 gene: the first containing only sequences detected in marsh deer and the second grouping sequences detected in brocket deer (Mazama spp. and S. gouazoubira). The latter was also divided into smaller clades that grouped Theileria genotypes according to deer species (M. jucunda, M. rufa and S. gouazoubira). This study provides the first molecular evidence of Theileria infection in M. jucunda, as well as co-infection by distinct Theileria (sub)species/genotypes in the same deer was evidenced. Canine visceral leishmaniasis (CVL) is a parasitic disease caused by the protozoan Leishmania infantum. Neurological infection occurs due to the parasite's ability to cross the blood–brain barrier. It is known that dogs can remain infected with a subclinical infection for life, potentially acting as reservoirs for L. infantum when bitten by sandflies. In this context, the objective of this study was to evaluate the occurrence of Leishmania spp. in the brains of dogs from the metropolitan region of Rio Grande do Sul, Brazil, without a history of neurological disease but residing in an endemic area for L. infantum. A total of 200 samples, from 2022 to 2023, were evaluated using conventional Polymerase Chain Reaction (PCR) with the primers Leishmini-F GGKAGGGGCGTTCTGC and Leishmini-R STATWTTACACCAACCCC, aiming to amplify a product of 120 base pairs for Leishmania spp. To identify the species, a multiplex PCR was used, differentiating L. braziliensis (127 bp), L. amazonensis (100 bp), and L. infantum (60 bp), with the molecular target being the conserved region of the kinetoplast DNA (kDNA) minicircle, specific to Leishmania spp. Of the 200 samples evaluated, 26.5% (53/200) tested positive in the conventional PCR reaction for Leishmania spp., with the PCR multiplex the only species detected was Leishmania infantum. The average age of the positive animals was 5.08 years, with 47.2% being females and 52.8% being males; among these, mixed-breed dogs were the most predominant, representing 43.4% of the total. Clinical signs varied: hepatomegaly in two dogs, pronounced neutrophilic hepatitis in one, splenomegaly in one with lymphoid hyperplasia, and glomerulonephritis and nephritis in two animals. Mild anemia and thrombocytopenia were found in eight, with pale mucous membranes in three, and diffuse alveolar edema in one case. Notable pathological findings included suspected distemper in one animal and lymphoplasmacytic meningitis in another. Histopathological findings revealed alveolar edema and acute renal failure. A third dog exhibited bilateral hydrocephalus and diffuse edema in the brain. Additional changes, such as mild inflammatory infiltrate and slight vacuolar degeneration, were observed in 11.3% of the analyzed brains. There was no clinical suspicion of leishmaniasis in any of the studied cases. Therefore, the detection of L. infantum DNA in the brains of dogs suggests that animals with subclinical infection may play a crucial role in the spread of leishmaniasis, and infection by Leishmania spp. should be considered as a differential diagnosis for neurological disease in endemic areas for the protozoan. Yeast feed additives present a natural approach for mitigating ruminal greenhouse gases (GHG) in an environmentally sustainable manner. This study aimed to isolate yeast strains from ruminal fluids capable of reducing GHG from Aflatoxin (AFB1) contaminated diets. Two isolates of Pichia manchuria (FFNLYFC1 and FFNLYFC2) were isolated and identified from the ruminal contents of dairy Zaraibi goats. An in vitro gas production assay was conducted to evaluate the impact of the yeast supplementations on a basal diet contaminated with AFB1 or not. The treatments were control (-AFB1; basal diet without supplements), control with AFB1 contamination (+ AFB1; basal diet containing 20 ppb AFB1), and yeast-supplemented diets (basal diet supplemented with Saccharomyces cerevisiae, and three treatments of P. manchuria [FFNLYFC1, FFNLYFC2, and their mixture at 1:1 ratio (Mix)]. High biological components were detected in abundance of both FFNLYFC1, FFNLYFC2 filtrates (e.g., diisooctyl phthalate). The Mix and FFNLYFC2 of P. manchuria reduced (P < 0.05) methane by 23.5 and 20.8%, respectively, while only Mix inhibited carbon dioxide by 44% compared to the + AFB1 diet. All yeast diets improved (P < 0.05) ammonia concentration, total protozoal and Entodinium spp. counts compared to + AFB1 diet. The Mix exhibited higher (P < 0.05) values of ruminal degraded cellulose, total short-chain fatty acids, acetate and propionate compared to the individual isolates diets. The results suggest synergistic interactions among P. manshurica isolates, leading to enhanced ruminal fermentation and reduced GHG emissions while alleviating the adverse effects of AFB1. The present experiment was conducted to evaluate the nutritional value of red Alternanthera sessilis for fattening lambs when they were replaced with alfalfa forage. Forty growing lambs with an average weight of 21.12 kg and an age of 5 months were randomly assigned to four experimental treatments. Growth performance, nutrient digestibility, rumen fermentation parameters, protozoa population, blood parameters, and composition of carcass components were evaluated. The results of this experiment showed that the use of Alternanthera sessilis in the diet significantly reduced feed intake, increased the average daily weight gain, and improved the feed conversion ratio compared to the control treatment (P < 0.05). The digestibility of dry matter and protein was significantly increased (P < 0.05), while the digestibility of neutral detergent fiber, acid detergent fiber, and organic matter showed a numerical increase. Diets containing different levels of the Alternanthera sessilis plant did not affect pH, but the increase in the amount of this plant in the diet led to an increase ammonia nitrogen concentration and rumen protozoa population (P < 0.05). The addition of Alternanthera sessilis to the diet significantly reduced the concentration of blood glucose and cholesterol and increased the concentration of blood urea nitrogen (P < 0.05). Except for the weight of the thigh and neck, the effect of experimental treatments on other carcass components was not significant. Overall, the results of this experiment showed that using the Alternanthera sessilis plant in the rations of fattened lambs as a substitute for alfalfa forage not only had no negative effect on the studied parameters but also improved them in some cases. The problem most often encountered in fish farming is disease which can cause a decrease in fish production levels. Parasites cause one of the infectious diseases found in fish farming. The presence of parasites can cause a decrease in the quality of the growth of fish production. The purpose of this study was to identify the prevalence of ectoparasites on Nile tilapia in Sidoarjo, Indonesia. Ninety-six samples from two ponds were examined under the microscope to find out the presence of ectoparasite that exists in the outside body part of the fish. Samples were collected from skin, fins, tail, and gills using native methods and observed under a microscope in the Division of Parasitology Laboratory, Faculty of Veterinary Medicine, Universitas Airlangga. The results of this study showed 86 samples positively infected by ectoparasite and 10 showed negative. The ectoparasites found in this study were Trichodina sp., Chilodonella sp., Ichthyophthirius multifillis, Gyrodactylus sp., and Dactylogyrus sp. The highest prevalence is Trichodina sp. and Dactylogyrus sp. The number of fish infested by ectoparasite is 89.56%, this is classified in the usual category. The highest prevalence occurs in infestations of Trichodina sp. i.e.72.91% which were contained in the 70 samples. While the lowest prevalence was in Gyrodactylus sp. by 8.32% in 8 fish samples. Alternatives to conventional chemical treatments for parasitic diseases in fish are needed. Microalgal‐sourced fatty acid ethyl esters (FAEEs) have shown an antiparasitic effect against Gyrodactylus turnbulli infection in guppies. Here, we tested a range of commercial FAEEs of various carbon chain lengths and unsaturation levels against two fish parasites. Guppies and barramundi infected with G. turnbulli and Trichodina sp., respectively, were used. The most effective FAEE, after excluding those toxic to fish, was ethyl laurate (12:0). For both parasites, the LD50 was 18.75 μM within 250 min of incubation. Ethyl eicosapentaenoate (20:5n3) was the next most effective FAEE against G. turnbulli, and dihomo‐γ‐linolenic acid ethyl ester (20:3n6) and ethyl α‐linolenate (18:3n3) were the next most effective against Trichodina sp. In addition, FAEEs prepared from the microalga Phaeodactylum tricornutum residue, after fucoxanthin extraction, were examined against Trichodina sp. infection in barramundi for the first time. LD85 and LD100 was achieved at 2.5 and 5 μL mL−1 of the FAEE preparation, respectively. In vivo, immersion of infected barramundi in 1.25 μL mL−1 of this preparation for 24 h reduced infection prevalence from 100% to 53% and was non‐toxic to fish. [ABSTRACT FROM AUTHOR]

Medical parasitology is the science that studies human parasites, diseases caused by them, and measures to combat these diseases. Medical parasitology is an important part of the science of parasitology (which includes zooparasitology and phytoparasitology) and studies the causes, transmission, pathogenesis, clinic, epidemiology, immunology and treatment of parasitic diseases in humans, as well as ways of prevention and control of these diseases. Traditionally, parasitic diseases include diseases caused by protozoa (protozoa), diseases caused by helminths (helminthoses) and diseases caused by arthropods. One of the most important tasks facing parasitology is the development and implementation of scientific-based measures against diseases caused by parasites in order to protect people's health, protect agricultural animals and plants from various diseases. This study aimed to carry out a molecular screening for the presence of Giardia, Cryptosporidium, and/or Entamoeba in the feces of pet and stray/feral cats in Jordan. G. duodenalis was found in 27.9% (95% CI, 23.2–32.9) of the 348 sampled cats overall; E. histolytica was found in only 0.6% (95% CI, 0.1–2.1) of the cats, while none of the sampled cats had Cryptosporidium infections. The infection rate of G. duodenalis among indoor cats (32.3%) did not differ significantly from that among outdoor cats (24.1%). There were significantly more infections (p = 0.0004) geographically in the cold semiarid areas (67%) than in the cold desert areas (24%). Multilocus sequence typing analysis of amplicons based on the bg, tpi, and gdh genes revealed that the majority of G. duodenalis infections were zoonotic assemblage B (65.9%; 64 of 97 positive samples); followed by feline-specific assemblage F (18.5%, 18/97); cattle-specific assemblage E (5.2%, 5/97); and then assemblage C that was shared with canids (1.0%; 1/97). Within Giardia isolates, a substitution mutation (A/G) was found at position 297 of the complete protein coding sequence (cds) of tpi-assemblage B, which may represent a new spreading mutation within this gene among the cat population in Jordan. The results of the present study suggest that close human-cat interactions could play a role in zoonotic transmission of Giardia, but further research is needed to determine the possible contribution of cats to the transmission of other protozoa to humans. Intestinal parasitic infections (IPIs) remain a significant contributor to morbidity and mortality globally, particularly in developing countries such as Ethiopia. Periodic assessments of IPI prevalence are essential prerequisite for effective control measures. Therefore, this cross‐sectional study is aimed at determining the prevalence of gastrointestinal parasitic infections and associated risk factors among schoolchildren at Wonji Shoa Secondary School, East Shoa Zone, Adama district, Oromia region, Ethiopia, between January and June 2022. A simple random stratified sampling technique was employed to select participants. Sociodemographic and risk factor data were collected using a structured questionnaire. Stool samples were examined to identify parasites. Data were analyzed using SPSS version 20. Descriptive statistics, chi‐square tests, and logistic regression were conducted to assess associations between variables and then the strength of the association. A p value < 0.05 was considered statistically significant. Of the 403 selected students, 330 completed the study that makes 81.89% response success. The overall IPI prevalence was 16.66% (55/330), with a higher prevalence among males (60%, 33/55) than females (40%, 22/55). Five parasite species were identified: two protozoa (Entamoeba histolytica and Giardia lamblia) with a combined prevalence of 9.70% (32/330) and three helminths (Ascaris lumbricoides, Hymenolepis nana, and Taenia spp.) with a combined prevalence of 6.97% (23/330). Cysts were detected in 62.5% of E. histolytica cases (15/24), and eggs were detected in 76.92% of A. lumbricoides cases (10/13). The study revealed a substantial IPI prevalence (16.66%) among the students. This finding underscores the need for effective prevention and control strategies. The predominance of parasitic infections among males is notable requiring further investigation of the factors. The identification of multiple parasite species indicates a complex epidemiological scenario. Recently a number of broad-range stool parasite PCR assays have been developed. However, there is ongoing disagreement regarding their diagnostic performance, as various studies have produced contradictory results. In this study, we compared the diagnostic accuracy of the Seegene Allplex GI-Parasite and Allplex GI-Helminth assays (SA) with the conventional methods used at the travel clinic of the Institute of Tropical Medicine (ITM) including microscopy, antigen testing, and molecular detection in order to provide insights into the strengths and limitations of this diagnostic tool which may be crucial to select the most appropriate diagnostic tools for the suspected pathogen. A total of 97 native stool samples from 95 patients with suspected gastrointestinal illness were analyzed, including 26 from a frozen collection and 71 prospectively collected samples. The diagnostic performance of SA was notably superior to the conventional workflow in detecting Dientamoeba fragilis (sensitivity 100% vs. 47.4%) and Blastocystis hominis (sensitivity 95% vs. 77.5%). SA had a comparable performance with the conventional workflow in detecting pathogenic protozoa (sensitivity 90% vs. 95%). In contrast, SA had a much lower diagnostic performance in detecting helminths (59.1%) compared to the conventional workflow (100%). We conclude that the Seegene Allplex GI-Parasite assay may be useful for protozoa screening in low-endemic industrialized countries. The aim of this research is to identify, determine prevalence, and degree infection of protozoa gastrointestinal tract in dairy cow at KUTT Suka Makmur, Pasuruan district. The research was conducted from July until September 2023 by taking 100 dairy cow faeces sample from 4 villages of KUTT Suka Makmur. The samples were examined by sedimentation and floating methods. Species are identified by looking at morphology of protozoa gastrointestinal. There were of 85 (85%) samples positively infected with protozoan. There were 3 species that infect the digestive tract, those are Eimeria sp., Balantidium sp., and Blastocystis sp. The highest infection of protozoa in this research was Balantidium sp. (54%) followed by Eimeria sp. (47%). In this study, we found that 54 samples (54%) infected by one kind of protozoa and 31 samples (31%) infected by mixed protozoa. The result of Chi Square showed no significant difference in the age on the prevalence of digestive tract protozoa. The genus Naegleria is a taxonomic subfamily consisting of 47 free-living amoebae. The genus can be found in warm aqueous or soil habitats worldwide. The species Naegleria fowleri is probably the best-known species of this genus. As a facultative parasite, the protist is not dependent on hosts to complete its life cycle. However, it can infect humans by entering the nose during water contact, such as swimming, and travel along the olfactory nerve to the brain. There it causes a purulent meningitis (primary amoebic meningoencephalitis or PAME). Symptoms are severe and death usually occurs within the first week. PAME is a frightening infectious disease for which there is neither a proven cure nor a vaccine. In order to contain the disease and give patients any chance to survival, action must be taken quickly. A rapid diagnosis is therefore crucial. PAME is diagnosed by the detection of amoebae in the liquor and later in the cerebrospinal fluid. For this purpose, CSF samples are cultured and stained and finally examined microscopically. Acanthamoeba, a widely distributed free-living amoeba with 20 genotypes identified through rRNA gene sequencing, exhibits varying degrees of pathogenicity influenced by its genotype. This study focuses on assessing the prevalence of Acanthamoeba species in the surface waters of Ilam, located in western Iran, utilizing morphological analysis and sequencing of the 18S rRNA gene through the PCR method. A total of 50 water samples were collected from various regions within Ilam city, situated in the southwest of Iran. To isolate Acanthamoeba parasites from the samples, a culture method was used, and all utilized culture media were scrutinized through microscopic and molecular techniques. The parasite’s genotype was determined by sequencing a 500-bp fragment of the 18S rRNA gene. Using microscopic and molecular methods, 19 and 16 water samples tested positive, respectively. The 18S rRNA sequences revealed that the isolates belonged to the T4, T2, and T11 genotypes. This study emphasizes the presence and inclination for close contact with highly pathogenic genotypes of Acanthamoeba in the surface waters of Ilam City. [ABSTRACT FROM AUTHOR]

Francisella tularensis is the pathogen of tularemia, a zoonotic disease that have a broad range of hosts. Its epidemiology is related to aquatic environments, particularly in the subspecies holarctica. In this review, we explore the role of water and mosquitoes in the epidemiology of Francisella in Europe. F. tularensis epidemiology has been linked to natural waters, where its persistence has been associated with biofilm and amebas. In Sweden and Finland, the European countries where most human cases have been reported, mosquito bites are a main route of transmission. F. tularensis is present in other European countries, but to date positive mosquitoes have not been found. Biofilm and amebas are potential sources of Francisella for mosquito larvae, however, mosquito vector capacity has not been demonstrated experimentally, with the need to be studied using local species to uncover a potential transmission adaptation. Transstadial, for persistence through life stages, and mechanical transmission, suggesting contaminated media as a source for infection, have been studied experimentally for mosquitoes, but their natural occurrence needs to be evaluated. It is important to clear up the role of different local mosquito species in the epidemiology of F. tularensis and their importance in all areas where tularemia is present. [ABSTRACT FROM AUTHOR]

Background: The encystation of Acanthamoeba castellanii has important ecological and medical significance. Blocking encystation is the key to preventing transmission and curing infections caused by A. castellanii. The formation of autophagosomes is one of the most important changes that occur during the encystation of Acanthamoeba. Our previous studies have shown that the heat shock protein 20 of A. castellanii (Ac-HSP20) is involved in its encystation. This study aimed to determine the role and mechanism of Ac-HSP20 in regulating autophagy involved in the encystation of A. castellanii. Methods: Immunofluorescence assay, western blotting and transmission electron microscopy were used to analyze the dynamic changes in autophagy during the initiation and continuation of encystation. The knockdown of Ac-HSP20 was performed to clarify its regulation of encystation and autophagy and to elucidate the molecular mechanism by which Ac-HSP20 participates in autophagy to promote cyst maturation. Results: The encystation rates and autophagosomes were significantly decreased by treatment with the autophagy inhibitor 3-MA. The autophagy marker LC3B and autophagic lysosomes increased with the induced duration of encystation and reached the maximum at 48 h. The encystation rate, LC3B expression and autophagosomes decreased when Ac-HSP20 was knocked down by siRNA transfection. In addition, the expression levels of Ac-HSP20 and LC3B increased and the expressions of p-AKT and p-mTOR decreased after 48 h of encystation without knockdown. However, the expressions of p-AKT and p-mTOR increased while the expression of LC3B decreased under the knockdown of Ac-HSP20. Furthermore, the protein expression of LC3B increased when the PI3K/AKT/mTOR signaling pathway was inhibited but decreased when the pathway was activated. Conclusions: The results demonstrated that autophagy is positively correlated with the encystation of A. castellanii, and Ac-HSP20 regulates autophagy to maintain the homeostasis of A. castellanii by inhibiting the PI3K /AKT /mTOR signaling pathway, thus promoting the maturation and stability of encystation. Ciliate protozoa are an integral part of the rumen microbial community involved in a variety of metabolic processes. These processes are thought to be in part the outcome of interactions with their associated prokaryotic community. For example, methane production is enhanced through interspecies hydrogen transfer between protozoa and archaea. We hypothesize that ciliate protozoa are host to a stable prokaryotic community dictated by specific functions they carry. Here, we modify the microbial community by varying the forage‐to‐concentrate ratios and show that, despite major changes in the prokaryotic community, several taxa remain stably associated with ciliate protozoa. By quantifying genes belonging to various known reduction pathways in the rumen, we find that the bacterial community associated with protozoa is enriched in genes belonging to hydrogen utilization pathways and that these genes correspond to the same taxonomic affiliations seen enriched in protozoa. Our results show that ciliate protozoa in the rumen may serve as a hub for various hydrogenotrophic functions and a better understanding of the processes driven by different protozoa may unveil the potential role of ciliates in shaping rumen metabolism. Irrigated and rain-fed rice fields are unique agroecosystems and anthropogenic wetlands whose main feature is seasonal flooding. Flooded soils are characterized by spatiotemporal shifts and oscillation of the oxygen status and redox potential, sustaining varieties of microbial metabolisms, where bacteria and methanogenic archaea play principal roles and thus have been the major research targets. In this review, we focus on the diversity and ecology of protists—often overlooked biological entities—in wetland rice field soils. Protists with different ecological functions, i.e., phagotrophs, phototrophs, saprotrophs, and parasites, inhabit a rice field soil with a community- and individual-level adaptation to the wide range of oxygen tensions and redox potential. Other agricultural managements like fertilization and char application also influence the protist community. They link to the material cycling in rice soil and affect the activities and community composition of the microorganisms involved in the biogeochemical cycles. Rice roots are the hot spot for protists, which control the rhizospheric bacterial community and could increase the plant productivity through enhancing nutrient release and altering bacterial activities. This review highlights the essential roles of protists in a wetland rice field soil and needs for further research to fill the gaps in knowledge regarding the diversity and functions of the protists in this unique agroecosystem. Background: Malaria, transmitted by Plasmodium parasites and anopheline mosquitoes, continues to be a leading cause of global disease and death. This retrospective investigation from 2018 to 2023 examines the epidemiological attributes of malaria in Saravan, southeastern Iran. It seeks to evaluate the prevalence, transmission causes, local population impact, and health system effects. Materials and Methods: Blood samples from suspected malaria cases in Saravan health centers were collected for this analysis. Each positive case was detailed with demographic data in a questionnaire. The SPSS 26 statistical program scrutinized data with t-tests comparing the variables. Results: The study indicated fluctuating malaria cases peaking in 2023, with an annual parasite incidence. (API) of 17.27. Plasmodium vivax was the predominant species (P < 0.001), with the majority of cases in individuals over 15, notably males. A significant number of cases were reported in September (20.7%). Conclusion: The findings emphasize the persistent malaria challenges in Saravan, accentuating the urgent need to strengthen prevention and control strategies. Reducing disease burden demands focused approaches, including improving prevention and treatment programs, enhancing surveillance systems, developing health infrastructures, and implementing localized therapies, especially considering recent climatic and rainfall patterns. The socio-economic importance of parasitic diseases, above all malaria and intestinal helminthosis, is so great that this pathology creates an obstacle for the economic development of many countries of the world. Cases of introduction of tropical parasitic diseases in non-endemic countries seriously harm people's health. Considering the global importance of parasitic pathology, most countries of the world have set the problem of fighting parasitic diseases as a goal to be solved. Protozoan diseases and helminthoses, which are the subject of parasitology, are an important part of infectious pathology. According to the WHO, more than 4.5 billion people worldwide suffer from parasitic diseases. About one million parasitic patients are registered in Russia every year. Their actual number exceeds 20 million people. WHO shows that diseases caused by helminths are the third most important infectious and parasitic diseases in the world, and diseases caused by plasmodials take the fourth place (1.4 billion and 600 million cases, respectively). All this requires medical workers to be familiar with the modern principles of diagnosis, treatment and prevention of human parasitic diseases and makes it necessary to teach them. Intestinal parasites, including helminths and protozoa, account for a significant portion of the global health burden. The gastrointestinal (GI) tract not only serves as the stage for these parasitic infections but also as the residence for millions of microbes. As the intricacies of the GI microbial milieu continue to unfold, it is becoming increasingly apparent that the interactions between host, parasite, and resident microbes help dictate parasite survival and, ultimately, disease outcomes. Across both clinical and experimental models, intestinal parasites have been shown to impact microbial composition and diversity. Reciprocally, microbes can directly influence parasitic survival, colonization and expulsion. The gut microbiota can also indirectly impact parasites through the influence and manipulation of the host. Studying this host–parasite–microbiota axis may help bring about novel therapeutic strategies for intestinal parasitic infection as well as conditions such as inflammatory bowel disease (IBD). In this review, we explore the relationship between intestinal parasites, with a particular focus on common protozoa and helminths, and the gut microbiota, and how these interactions can influence the host defence and intestinal immune response. Parasitism by protozoa and monogenean flatworms in freshwater fish from Romania was studied by collecting and examining samples from two major river systems there: 183 fish from 17 species from the Olt River and its tributaries; and 155 fish from 16 species from the Mureș River and its tributary, Târnava Mare. The average rates of parasitism in the samples from the two rivers and their tributaries were as follows: Ichthyiophthirius multifiliis (2%), Trichodina spp. (21%), Apiosoma spp. (18%), Mixobolus spp. (8%), Dactylogyrus spp. (9%), and Gyrodactylus spp. (10%). The number of parasite species varied from one river to another. I. multifiliis was found in only 3 fish species, Trichodina spp. in 13 species, Glosatella spp. in 6 species, and Mixobollus spp., Dactylogyrus spp., and Gyrodactylus spp. in 7 different species each. The highest number of parasite species (six) were identified in the European chub (Squalius cephalus) and schneider (Alburnoides bipunctatus), which seem more susceptible to parasitic infections. A systematic study of the interaction of hydrobiological, hydrochemical, hydrological, and hydrophysical processes that occur in the ecosystem of the Vistula Lagoon (VL) of the Baltic Sea has been carried out using mathematical modeling and taking into account the processes in the active layer of bottom sediments (BSs). The features of the dynamics of nitrogen and phosphorus compounds, the turnover time, and the components of the balance of compounds of biogenic elements in the VL ecosystem, as well as the degree of their temporal variability, have been analyzed. It is shown that the variability of nitrogen and phosphorus compounds in the aquatic environment increases in spring and autumn periods. This is due to an increase in river flow into the lagoon and an increase in the level of external biogenic load on the water area of the VL. Model calculations of matter fluxes allow us to state that the inputs of dissolved organic and suspended matter into the lagoon from external sources and the removal to the Baltic Sea are important processes that significantly affect their content in the VL ecosystem. It has been established that under strong winds the process of sedimentation of suspensions can be largely blocked, and the intensity of BS resuspension can increase significantly. An analysis of the main qualitative and quantitative features of the nature of the temporal dynamics of the DON and DOP turnover times, as well as the mineral compounds of nitrogen and phosphorus, indicates a close conjugation of the processes of transformation of dissolved organic and inorganic substances in the water of the VL. A quantitative assessment of the processes of consumption and excretion of compounds of biogenic elements by different groups of aquatic organisms has been performed, and the important role of heterotrophic bacterioplankton and protozoa in the processes of transformation of compounds of nutrients in the VL ecosystem has been shown. Background Relapsing babesiosis often occurs in highly immunocompromised patients and has been attributed to the acquisition of resistance against drugs commonly used for treatment such as atovaquone, azithromycin, and clindamycin. Tafenoquine, which is approved for malaria prophylaxis and presumptive antirelapse treatment of Plasmodium vivax malaria, has shown activity against Babesia microti in several animal models of acute infection and in a single human case of relapsing babesiosis. Here, we report 5 cases of relapsing babesiosis treated with tafenoquine, including the previous case, and begin to define the conditions for optimal use of tafenoquine in relapsing babesiosis. Methods A definitive diagnosis of babesiosis was made by microscopic examination of Giemsa-stained thin blood smears or a real-time polymerase chain reaction (PCR) that targets the parasite 18S rRNA gene. Clearance of B. microti infection was ascertained by use of blood smear and real-time PCR. Results Tafenoquine was initiated with a loading dose of 600 mg. A weekly maintenance dose consisted of 200 mg or 300 mg; the lower dose was associated with a delayed clearance of B. microti. In 2 cases, all antimicrobial agents but tafenoquine were discontinued prior to clearance of infection. In 2 other cases, clearance was achieved while tafenoquine was administered along with other antimicrobial agents. In 3 of these 4 cases, tafenoquine was used in combination with atovaquone-proguanil. Other agents included atovaquone, azithromycin, and/or clindamycin. In 1 case, tafenoquine was administered alone and failed to prevent relapse. The relationship between the protozoan communities and environmental variables was studied in the Nile River to evaluate their potential as water quality indicators. Protozoans were sampled monthly at six sampling sites in the Nile's Damietta Branch across a spatial gradient of environmental conditions during a 1-year cycle (February 2016–January 2017). The Protozoa community was comprised of 54 species belonging to six main heterotrophic Protozoa phyla. The abundance (average, 1089 ± 576.18 individuals L−1) and biomass (average, 86.60 ± 106.13 μg L−1) were comparable between sites. Ciliates comprised the majority of protozoan species richness (30 species), abundance (79.72%), and biomass (82.90%). Cluster analysis resulted in the distribution of protozoan species into three groups, with the most dominant species being the omnivorous ciliate Paradileptus elephantinus. Aluminium, fluoride, and turbidity negatively affected abundance and biomass, while dissolved oxygen and potassium positively impacted biomass. Of the dominant species recorded over the study area, the amoebozoa Centropyxis aculeata was associated with runoff variables, while the bacterivorous ciliates Colpidium colpoda, Glaucoma scintillans, and Vorticella convallaria were related to the abundance of heterotrophic bacteria, phytoplankton biomass, and total organic carbon. Total dissolved salts, PO4, NH3, NO2, dissolved oxygen, and total organic carbon were the strongest causative factors for protozoa distribution. The α-Mesosaprobic environment at site VI confirmed a high load of agricultural runoffs compared to other sites. Introduction: Ongoing global changes, including natural land conversion for agriculture and urbanization, modify the dynamics of human–primate contacts, resulting in increased zoonotic risks. Although Asia shelters high primate diversity and experiences rapid expansion of human–primate contact zones, there remains little documentation regarding zoonotic surveillance in the primates of this region. Methods: Using the PRISMA guidelines, we conducted a systematic review to compile an inventory of zoonotic pathogens detected in wild Asian primates, while highlighting the coverage of primate species, countries, and pathogen groups surveyed, as well as the diagnostic methods used across the studies. Moreover, we compared the species richness of pathogens harbored by primates across diverse types of habitats classified according to their degree of anthropization (i.e., urban vs. rural vs. forest habitats). Results and discussion: Searches of Scopus, PubMed, and the Global Mammal Parasite Database yielded 152 articles on 39 primate species. We inventoried 183 pathogens, including 63 helminthic gastrointestinal parasites, two blood-borne parasites, 42 protozoa, 45 viruses, 30 bacteria, and one fungus. Considering each study as a sample, species accumulation curves revealed no significant differences in specific richness between habitat types for any of the pathogen groups analyzed. This is likely due to the insufficient sampling effort (i.e., a limited number of studies), which prevents drawing conclusive findings. This systematic review identified several publication biases, particularly the uneven representation of host species and pathogen groups studied, as well as a lack of use of generic diagnostic methods. Addressing these gaps necessitates a multidisciplinary strategy framed in a One Health approach, which may facilitate a broader inventory of pathogens and ultimately limit the risk of cross-species transmission at the human–primate interface. Strengthening the zoonotic surveillance in primates of this region could be realized notably through the application of more comprehensive diagnostic techniques such as broad-spectrum analyses without a priori selection. The data obtained from this study is important to literate the farm owners, farmers and other stakeholders about health, hygiene and the impact of this on animals as well as on themselves. Background: Ruminants and their products play an important role in providing food for many people. They are able to convert plants that cannot be digested by humans into usable protein for humans. New animal feed has been commonly developed for long years. To improve food production from ruminants, it is important to explore new feed sources. New or less exploited types of feed can replace or supplement traditional feeds in ruminant diets. Sesbania is a multipurpose plant that is cultivated for consumption as fodder and green manure. As a fast- growing plant, Sesbania has been mentioned as a food source to reduce feed costs. The food value of some of its species is equal to alfalfa and several other valuable plants. Due to proper digestibility, low fiber, and relatively high protein, it is possible to replace it with hay for livestock, especially small ruminants. In addition, as a tropical legume, Sesbania can easily withstand temperatures above 35°C, which are limiting for most legume plants, and grow well under salinity stress conditions if there is sufficient moisture in the soil. Therefore, this research aimed to investigate the nutritional value of Sesbania sp. L. for ruminants. Methods: The present research was carried out in the laboratories and teaching-research station of animal husbandry at the Agricultural Sciences and Natural Resources University of Khuzestan. The experimental treatments included a control diet (without the Sesbania plant), 25, 50, 75, and 100% replacements of the Sesbania plant with alfalfa. The chemical compositions of the Sesbania plant, alfalfa hay, treatments, and other experimental samples, including crude protein, dry matter, ash, acid detergent fiber, and total tannin, were measured by standard methods. The neutral detergent fiber was quantified with the usual method without using the alpha-amylase enzyme and sodium sulfite by removing ash. The amount of gas produced by the samples was recorded at 2, 4, 6, 8, 10, 12, 24, 48, 72, and 96 hours after incubation, and gas production parameters, digestibility of organic matter, their metabolizable energy, ammonia nitrogen concentration, pH, and protozoa population of culture medium were estimated in the treatments. In the gas production experiment, rumen fluid was taken from the rumen of three male sheep fed with a forage-based diet at the maintenance level and before morning feeding using a stomach tube. Ruminal fluid was filtered with a four-layer cheesecloth, mixed, and transferred to the laboratory in a flask containing hot water at 39 °C. To count protozoa at the end of 24 hours, a sample was prepared from the gas culture medium and mixed with 10 ml of 10% formaldehyde, and protozoa were counted using a hemocytometer slide. The resulting data were analyzed with a completely randomized design. In the present experiment, the chemical composition, digestion, and fermentation of the Sesbania plant were also compared with alfalfa hay. Results: Compared to the control, Sesbania-containing diets replaced with alfalfa had significantly the lowest potential and gas production rate at all levels (P < 0.05). The comparison of the Sesbania plant and alfalfa hay showed that the potential and gas production rate of alfalfa were higher than that of the Sesbania plant. In the experimental diets, the partitioning factor, microbial biomass production, and microbial biomass production efficiency were higher at all levels of Sesbania replaced with alfalfa than the control diet, and the levels of 75% and 100% of Sesbania replaced with alfalfa had a significantly higher partitioning factor than the control and other treatments (P < 0.05). However, the truly degradable organic matter was not affected by the experimental treatments. The results of the comparison of the Sesbania plant and alfalfa hay showed that the partitioning factor, microbial biomass production, and microbial biomass production efficiency of the Sesbania plant were significantly higher than alfalfa (P < 0.05). The results of fermentation parameters after 24 hours of incubation showed that the pH of the rumen culture medium was not affected by the experimental treatments, but the ammonia nitrogen concentration was affected by the experimental treatments (P < 0.05), and the control treatment contained the highest ammonia nitrogen concentration. In the comparison of the Sesbania plant with alfalfa hay, the pH of the rumen culture medium was not affected by the experimental treatments, and the ammonia nitrogen concentration of the Sesbania plant was significantly lower than that of alfalfa hay (P < 0.05). The helotrich, entodinomorph, and cellulolytic protozoa population, and the total rumen protozoa population were the highest in the control treatment (29.33) and the lowest in the 100% replacement of the Sesbania plant with alfalfa hay (9.66). The results of the comparison of the Sesbania plant with alfalfa hay showed an increase in the population of Helotrich, Entodinomorph, and Cellulolytic protozoa and the total population of rumen protozoa in alfalfa hay, with a decrease in the population of the Sesbania plant. The results of in vitro digestibility indicated the highest digestibility of dry matter, NDF, and, ADF in the control treatment. The comparison of the Sesbania plant with alfalfa hay revealed that the digestibility of dry matter, NDF, and ADF of alfalfa was higher than that of the Sesbania plant. Conclusion: In general, the results of the present experiment demonstrate that the chemical composition and digestibility of the Sesbania plant are similar to alfalfa hay. Background: Bone defects, resulting from substantial bone loss that exceeds the natural self-healing capacity, pose significant challenges to current therapeutic approaches due to various limitations. In the quest for alternative therapeutic strategies, bone tissue engineering has emerged as a promising avenue. Notably, excretory proteins from Toxoplasma gondii (TgEP), recognized for their immunogenicity and broad spectrum of biological activities secreted or excreted during the parasite's lifecycle, have been identified as potential facilitators of osteogenic differentiation in human bone marrow mesenchymal stem cells (hBMSCs). Building on our previous findings that TgEP can enhance osteogenic differentiation, this study investigated the molecular mechanisms underlying this effect and assessed its therapeutic potential in vivo. Methods: We determined the optimum concentration of TgEP through cell cytotoxicity and cell proliferation assays. Subsequently, hBMSCs were treated with the appropriate concentration of TgEP. We assessed osteogenic protein markers, including alkaline phosphatase (ALP), Runx2, and Osx, as well as components of the BMP/Smad signaling pathway using quantitative real-time PCR (qRT-PCR), siRNA interference of hBMSCs, Western blot analysis, and other methods. Furthermore, we created a bone defect model in Sprague-Dawley (SD) male rats and filled the defect areas with the GelMa hydrogel, with or without TgEP. Microcomputed tomography (micro-CT) was employed to analyze the bone parameters of defect sites. H&E, Masson and immunohistochemical staining were used to assess the repair conditions of the defect area. Results: Our results indicate that TgEP promotes the expression of key osteogenic markers, including ALP, Runx2, and Osx, as well as the activation of Smad1, BMP2, and phosphorylated Smad1/5—crucial elements of the BMP/Smad signaling pathway. Furthermore, in vivo experiments using a bone defect model in rats demonstrated that TgEP markedly promoted bone defect repair. Conclusion: Our results provide compelling evidence that TgEP facilitates hBMSC osteogenic differentiation through the BMP/Smad signaling pathway, highlighting its potential as a therapeutic approach for bone tissue engineering for bone defect healing. Malaria, a mosquito-borne infectious disease caused by the parasite Plasmodium, has been predominantly associated with humans. However, recent studies have uncovered a potential reservoir for Plasmodium spp., within livestock populations. Malaria, caused by the parasitic protozoan Plasmodium spp., is a deadly disease that affects millions of people worldwide. Although human is the primary host for the malaria parasite, it has been discovered that certain animals can also serve as reservoirs. One such potential reservoir is livestock populations, as they have been found to carry Plasmodium spp. parasites. This article will discuss the implications of livestock being potential reservoirs for malaria and the importance of understanding this relationship for effective disease control. Livestock populations have long been overlooked as potential reservoirs for malaria transmission, but recent advancements in diagnostic techniques and surveillance studies have sparked interest in exploring this field. Background: Sarcocystis includes a global group of apicomplexan parasites with two-host life cycle frequently circulating in wildlife and domestic hosts, including humans. Two of the most important wild terrestrial carnivores acting as definitive hosts are the red fox and raccoon dog, due to their wide distribution in Europe and usage of wild and farmed animals as prey. This study was conducted to determine the prevalence of Sarcocystis in hunted red foxes and raccoon dogs from nine regions of the Czech Republic and to identify isolated sporocysts by molecular techniques. Methods: Approximately 5 g of the contents of large intestine from 200 animals (197 red foxes and three raccoon dogs) were examined by flotation centrifugation coprological method. Only samples of 50 red foxes and one raccoon dog positive to Sarcocystis spp. were used for the nested PCR (nPCR) method to amplify a fragment or partial sequence on the cox1 gene. Ten species-specific primer pairs for detection of Sarcocystis spp. using farm animals as intermediate hosts were utilized. Results: In total, 38.1% of the red foxes and 66.7% of the raccoon dogs were positive to Sarcocystis by light microscopy. The molecular characterization resulted in the identification of five species in the red fox: S. arieticanis, S. capracanis, S. cruzi, S. miescheriana, and S. tenella, while the PCR was negative for the sole raccoon dog. The highest intraspecific variation was found for S. miescheriana, while S. tenella was the most prevalent. Co-infections occurred in the large intestine of the red fox. No zoonotic species were found in our samples. Conclusion: This is the first study where the potential role of the red fox and raccoon dogs as spreaders of Sarcocystis to farm animals in the Czech Republic is shown. This narrative review was aimed at collecting updated knowledge on the risk factors, illnesses caused, and measures for the prevention of protozoan infections transmitted by food and drinking water. Reports screened dated from 2019 to the present and regarded global prevalence in food handlers, occurrence in food and drinking water, impact on human health, and recently reported outbreaks and cases of severe infections attributable to the dietary route. Cryptosporidium spp., Cyclospora cayetanensis, Entamoeba histolytica, and Cystoisospora belli were the protozoans most frequently involved in recently reported waterborne and foodborne outbreaks and cases. Blastocystis hominis was reported to be the most widespread intestinal protozoan in humans, and two case reports indicated its pathogenic potential. Dientamoeba fragilis, Endolimax nana, and Pentatrichomonas hominis are also frequent but still require further investigation on their ability to cause illness. A progressive improvement in surveillance of protozoan infections and infection sources took place in developed countries where the implementation of reporting systems and the application of molecular diagnostic methods led to an enhanced capacity to identify epidemiological links and improve the prevention of foodborne and waterborne protozoan infections. Introduction: Eimeria spp. are intracellular protozoan parasites of the phylum Apicomplexa causing economic losses to various wild and domestic animals. An eimerian species infecting Columba livia domestica was identified in this study. Methods: A total of 15 faecal samples were examined by floatation technique, a prevalence rate of 60% was reported. Eimerian oocysts were sporulated in 2.5% potassium dichromate solution then identified using morphological and molecular (DNA amplification of the 18S rRNA and ITS-1 genes) diagnostic techniques. Results: Sporulated oocysts were identified as Eimeria labbeana-like, after morphometry with typical bi-layered wall with spherical to subspherical oocysts morphology. A polar granule is present, but no micropyle or oocyst residuum. Sporocysts are elongated ovoidal with stieda body. Sporocyst residuum with many granules and sporozoites with refractile bodies and nucleus. Both 18S rRNA and ITS-1 sequences have been deposited in GenBank database. DNA sequences from the partial 18S rRNA generated from the oocysts were found to be related to eimerian and isosporan parasites found in domestic pigeons. For the first time, ITS-1 sequences for E. labbeana-like were provided. Conclusion: The necessity of using molecular techniques to describe pigeon intestinal coccidian parasites in conjunction with traditional morphology-based tools was emphasized in this work in order to understand the biology of such parasites. Reactive oxygen species (ROS) contain at least one oxygen atom and one or more unpaired electrons and include singlet oxygen, superoxide anion radical, hydroxyl radical, hydroperoxyl radical, and free nitrogen radicals. Intracellular ROS can be formed as a consequence of several factors, including ultra-violet (UV) radiation, electron leakage during aerobic respiration, inflammatory responses mediated by macrophages, and other external stimuli or stress. The enhanced production of ROS is termed oxidative stress and this leads to cellular damage, such as protein carbonylation, lipid peroxidation, deoxyribonucleic acid (DNA) damage, and base modifications. This damage may manifest in various pathological states, including ageing, cancer, neurological diseases, and metabolic disorders like diabetes. On the other hand, the optimum levels of ROS have been implicated in the regulation of many important physiological processes. For example, the ROS generated in the mitochondria (mitochondrial ROS or mt-ROS), as a byproduct of the electron transport chain (ETC), participate in a plethora of physiological functions, which include ageing, cell growth, cell proliferation, and immune response and regulation. In this current review, we will focus on the mechanisms by which mt-ROS regulate different pathways of host immune responses in the context of infection by bacteria, protozoan parasites, viruses, and fungi. We will also discuss how these pathogens, in turn, modulate mt-ROS to evade host immunity. The human oral cavity is normally colonized by microorganisms including bacteria, fungi, archaea, viruses and protozoa. The aim of this study was to determine the frequency of Candida spp., in de oral cavity in a group of medical students from the north of Mexico. Oral sample were obtained from 240 healthy students. The specimens were analyzed by traditional microbiology cultures and DNA sequencing. Candida spp., grew in Sabouraud dextrose agar from 57 samples and subsequently were isolated and phenotyped. The definitive identification to the species level was done by sequence analysis. The yeasts were identified as follow: 28 Clavispora lusitaniae, 20 Candida albicans, 5 Pichia kudriavzevii and 4 Candida parapsilosis. Our findings revealed that 23.75% of the healthy population has a potential pathogen in their mouth. Surprisingly, C. albicans is not the predominant yeast; instead other non-Candida species are the colonizers of the oral cavity as normal microbiota. C. lusitaniae is considered an emerging opportunistic pathogen in immunosuppressive patients. Prokaryotes play a fundamental role in global ocean biogeochemical cycles. However, how the abundance and metabolic activity of ecologically distinct subgroups (i.e., high nucleic acid (HNA) and low nucleic acid (LNA) cells), and their regulating factors, change in response to changing marine environmental conditions remains poorly understood. Here, we delved into the time-evolving dynamic responses of the HNA and LNA prokaryotic subgroups to declining resource availability and selective grazing by protozoa by conducting a 73-day incubation experiment in a large-volume (117,000 L) macrocosm that facilitates community-level exploration. We found that the metabolic activity of the HNA subgroup was higher than that of the LNA subgroup when the macrocosm was resource replete but that the HNA subgroup declined more rapidly than the LNA subgroup as the resources became increasingly scarce, leading to a steadily increasing contribution of LNA cells to prokaryotic activity. Meanwhile, as resources in the macrocosm became limited, protozoan grazing preference shifted from the HNA to the LNA subgroup and the contributions of the LNA subgroup to the carbon flow within the macrocosm increased. The findings highlight the resilience of LNA cells in resource-limited environments, illuminate the critical role of selective grazing by protozoa in balancing distinct prokaryotic subgroups under changing resource conditions, and demonstrate the complex and adaptive interactions between protozoa and prokaryotes across diverse environmental contexts. Soil-transmitted helminths (STHs) are a group of parasites that are globally distributed and are the most prevalent neglected disease (NTD) in Latin America and the Caribbean (LAC); their presence is associated with health and development problems. In Argentina, their distribution is heterogenous, and there are highly endemic areas in the north of the country. The World Health Organization (WHO) recommends the mass deworming of children as a first-line strategy for the prevention and control of STHs and recently also encourage the development of more sensitive diagnostic tests. The aim of this study was to determine the prevalence of STHs in Tartagal (Salta, Argentina) after four years of deworming interruption with albendazole and ivermectin. A total of 437 fecal samples were analyzed using standard coprological techniques, a subset of which were selected to molecularly typify protozoan parasites; 257 blood samples were analyzed for the presence of specific antibodies to the STH Strongyloides stercoralis. The most prevalent protozoan species were G. intestinalis (19.6–49.2%) and B. hominis (19.1–38.5%). Molecular characterization allowed us to evidence possible zoonotic or human-to-human transmission pathways for Giardia intestinalis or Blastocystis spp., while serology for S. stercoralis proved to be a useful screening tool for monitoring this parasite after treatment. In general, a decrease in the prevalence of STHs was observed in the area, from 60% to 2.9–20% for hookworms and from 51% to 1–9.3% for S. stercoralis four years after treatment, demonstrating the effectiveness and duration of anthelmintic treatment with these two drugs. Background and Objective: Despite the significant burden of Plasmodium falciparum (Pf) malaria and the licensure of two vaccines for use in infants and young children that are partially effective in preventing clinical malaria caused by Pf, a highly effective vaccine against Pf infection is still lacking. Live attenuated vaccines using Pf sporozoites as the immunogen (PfSPZ Vaccines) hold promise for addressing this gap. Here we review the safety and efficacy of two of the most promising PfSPZ approaches: PfSPZ Vaccine (radiation attenuated PfSPZ) and PfSPZ-CVac (chemo-attenuated PfSPZ). Methods: We conducted a systematic review and meta-analysis by searching PubMed, EMBASE, SCOPUS, CENTRAL, and WOS until 22nd December 2021. We included randomized controlled trials (RCTs) of these two vaccine approaches that measured protection against parasitaemia following controlled human malaria infection (CHMI) in malaria-naive and malaria-exposed adults or following exposure to naturally transmitted Pf malaria in African adults and children (primary outcome) and that also measured the incidence of solicited and unsolicited adverse events as indicators of safety and tolerability after vaccination (secondary outcome). We included randomized controlled trials (RCTs) that measured the detected parasitaemia after vaccination (primary outcome) and the incidence of various solicited and unsolicited adverse events (secondary outcome). The quality of the included RCTs using the Cochrane ROB 1 tool and the quality of evidence using the GRADE system were evaluated. We pooled dichotomous data using the risk ratio (RR) for development of parasitemia in vaccinees relative to controls as a measure of vaccine efficacy (VE), including the corresponding confidence interval (CI). This study was registered with PROSPERO (CRD42022308057). Results: We included 19 RCTs. Pooled RR favoured PfSPZ Vaccine (RR: 0.65 with 95% CI [0.53, 0.79], P = 0.0001) and PfSPZ-table (RR: 0.42 with 95% CI [0.27, 0.67], P = 0.0002) for preventing parasitaemia, relative to normal saline placebo. Pooled RR showed no difference between PfSPZ Vaccine and the control in the occurrence of any solicited adverse event (RR: 1.00 with 95% CI [0.82, 1.23], P = 0.98), any local solicited adverse events (RR: 0.73 with 95% CI [0.49, 1.08], P = 0.11), any systemic solicited adverse events (RR: 0.94 with 95% CI [0.75, 1.17], P = 0.58), and any unsolicited adverse event (RR: 0.93 with 95% CI [0.78, 1.10], P = 0.37). Conclusion: PfSPZ and PfSPZ-CVacs showed comparable efficacy. Therefore, they can introduce a promising strategy for malaria prophylaxis, but more large-scale field trials are required to sustain efficacy and yield clinically applicable findings. Mosquitoes are hematophagous carriers for hundreds of pathogenic viruses that are the etiological agents of human diseases. Malaria, Zika fever, chikungunya and dengue fever, etc., are among the diseases that mosquitoes are involved in transmitting. In nature, mosquito-borne viruses maintain a life cycle between mosquitoes and vertebrates. Malaria, transmitted by Anopheles mosquitoes, remains a major cause of morbidity and mortality worldwide, particularly in sub-Saharan Africa. Understanding mosquito ecology is crucial for malaria control efforts. Zika fever, associated with severe neurological complications, has emerged as a global concern, primarily transmitted by Aedes mosquitoes. Urbanization facilitates the proliferation of Aedes populations, exacerbating viral transmission. Chikungunya, spread by Aedes mosquitoes, has expanded beyond endemic regions due to international travel, causing fever, rash, and joint pain. Dengue fever, caused by dengue viruses transmitted by Aedes mosquitoes, witnesses a global resurgence, with climate factors influencing disease transmission dynamics. Effective mosquito control strategies are essential for mitigating the impact of these vector-borne diseases on global health. The inland bearded dragon (Pogona vitticeps) is a lizard species commonly kept as a pet worldwide. Endoparasites are among the most important pathogens affecting bearded dragons. The aim of this study was to evaluate the endoparasites in captive P. vitticeps in Italy. Faecal samples from 30 P. vitticeps were analysed by fresh faecal smears, flotation tests, the Mini-FLOTAC technique, and a rapid immunoassay to detect Cryptosporidium spp. To search for microsporidia, PCR and sequencing were performed on the faecal samples. Data were statistically analysed. The overall positivity rate for endoparasites was 83.3% (25/30). The identified endoparasites were oxyurids (17/30, 56.7%), Isosospora amphiboluri (13/30, 43.3%), Encephalitozoon pogonae (4/18, 22.22%), and Cryptosporidium sp. (1/30, 3.33%). The positivity for protozoa was significantly higher in juveniles compared to adults. Moreover, the frequency of clinical signs was significantly higher in the positive animals. The results obtained here emphasize the importance of regular veterinary examinations of captive P. vitticeps, aimed at the diagnosis, treatment, and control of endoparasites. Background: Intestinal parasitic infections (IP) are a major source of morbidity in people living with Human immunodeficiency virus (HIV), particularly in resource-limited settings, mostly as a result of high viral load. Hence, this study aimed to investigate the magnitude of intestinal parasitic infections and its determinants among patients with HIV/AIDS attending public health facilities in East and West Gojam Zones in Ethiopia. Methods: Institution-based cross-sectional study was conducted on 327 people living with HIV visiting public health facilities from December 2022 to May 2023. A simple random sampling technique was used to recruit participants. Face-to-face interviews were used to collect socio-demographics and determinants. The fresh stool was collected from each patient, transported, and tested in accordance with laboratory standard operating procedures of wet mount, formol-ether concentration technique, and modified acid-fast staining. Data were entered and analyzed in the statistical package for Social Science (SPSS) version 20. A 95% CI with p-value < 0.05 was considered statistically significant. Results: The overall prevalence of IP in patients with HIV/AIDS was 19.3% (63/327). Hookworm was the most identified parasite 33.3% (21/63) followed by E.histolytica 17% (11/63) and G.lamblia 14.3% (9/63). Parasitic infections were significantly higher among viral load > 1000cps/ml (p = 0.035), WHO stage 4 (p = 0.002), CD4 < 200 cell/mm3 (p = 0.001), and bare foot walking (p = 0.001). Conclusion: IP infections are moderately high among patients with HIV/AIDS in the study area. The proportion of parasites was greatly affected by high viral load, WHO stage 4, CD4 < 200 cell/mm3, and being barefoot; this gives valuable insight to health professionals, health planners and community health workers. As a result, viral load monitoring, and WHO stage controlling were periodically assessed in patients with HIV/AIDS. Inflammatory bowel disease (IBD) is a multifactorial disease involving the interaction of the gut microbiota, genes, host immunity, and environmental factors. Dysbiosis in IBD is associated with pathobiont proliferation, so targeted antibiotic therapy is a rational strategy. When restoring the microbiota with probiotics, it is necessary to take into account the mutual influence of co-cultivated microorganisms, as the microbiota is a dynamic community of species that mediates homeostasis and physiological processes in the intestine. The aim of our study was to investigate the recovery efficacy of two potential probiotic bacteria, L. johnsonii and E. faecalis, in Muc2−/− mice with impaired mucosal layer. Two approaches were used to determine the efficacy of probiotic supplementation in mice with dysbiosis caused by mucin-2 deficiency: bacterial seeding on selective media and real-time PCR analysis. The recovery time and the type of probiotic bacteria relocated affected only the number of E. faecalis. A significant positive correlation was found between colony-forming unit (CFU) and the amount of E. faecalis DNA in the group that was replanted with probiotic E. faecalis. As for L. johnsonii, it could be restored to its original level even without any additional bacteria supplementation after two weeks. Interestingly, the treatment of mice with L. johnsonii caused a decrease in the amount of E. faecalis. Furthermore, either L. johnsonii or E. faecalis treatment eliminated protozoan overgrowth caused by antibiotic administration. [ABSTRACT FROM AUTHOR]

Simple Summary: This research conducted in Serbia aimed to identify intestinal parasites in dogs that could potentially infect humans. Total prevalence of intestinal endoparasites was 62.6%. Various endoparasites such as Cystoisospora spp., Sarcocystis spp., Neospora caninum/Hammondia spp., Giardia intestinalis, Toxocara canis, Toxascaris leonina, Ancylostomatidae, Trichuris vulpis, Capillaria spp., Alaria alata and Taeniidae were found. Factors like age, outdoor living, attitude and diet were linked to higher infection rates. This study emphasizes the importance of educating dog owners, conducting routine parasitological tests on their pets and regular deworming strategies. Dogs are the most popular pets worldwide. Close contact between dogs and people increases the risk of transmission of various zoonotic parasitic infections. Given the importance of veterinary medicine in preserving the One Health concept, the aim of this research was to identify intestinal parasites that may have zoonotic potential and to evaluate risk factors (individual and environmental). The research was conducted in Serbia in 2022 and 2023 on 382 owned dogs, using qualitative methods of coprological examination with a concentration on parasitic elements. The overall prevalence of intestinal parasites was 62.6%, with the following detected: protozoa: Cystoisospora spp. (9.2%), Sarcocystis spp. (4.5%), Neospora caninum/Hammondia spp. (3.7%), Giardia intestinalis (11.8%); nematoda: Toxocara canis (11.5%), Toxascaris leonina (4.2%), family Ancylostomatidae (38.0%), Trichuris vulpis (21.5%), Capillaria spp. (10.5%); trematoda: Alaria alata (1.6%) and cestodes from the Taeniidae family (1.3%). Factors like age, size and coat length, as well as the way of living, attitude and diet were linked to a significantly higher (p < 0.05) prevalence of intestinal parasites. Based on the results of coprological diagnostics, this research indicates the importance of educating dog owners, conducting routine parasitological tests on their pets and regular deworming strategies. Background: Rodents are recognized as the hosts of many vector-borne bacteria and protozoan parasites and play an important role in their transmission and maintenance. Intensive studies have focused on their infections in vectors, especially in ticks, however, vector-borne bacterial and protozoan infections in rodents are poorly understood although human cases presenting with fever may due to their infection have been found. Methods: From May to October 2019, 192 wild rodents were trapped in wild environment of Guangxi Province, and the spleen samples were collected to reveal the presence of vector-borne bacterial and protozoan infections in them. The microorganisms in rodents were identified by detecting their DNA using (semi-)nested PCR. All the PCR products of the expected size were subjected to sequencing, and then analyzed by BLASTn. Furthermore, all the recovered sequences were subjected to nucleotide identity and phylogenetic analyses. Results: As a result, 192 rodents representing seven species were captured, and Bandicota indica were the dominant species, followed by Rattus andamanensis. Based on the (semi-)nested PCR, our results suggested that Anaplasma bovis, Anaplasma capra, Anaplasma ovis, Anaplasma phagocytophilum, "Candidatus Neoehrlichia mikurensis", "Candidatus E. hainanensis", "Candidatus E. zunyiensis", three uncultured Ehrlichia spp., Bartonella coopersplainsensis, Bartonella tribocorum, Bartonella rattimassiliensis, Bartonella silvatica, two uncultured Bartonella spp., Babesia microti and diverse Hepatozoon were identified in six rodent species. More importantly, six species (including two Anaplasma, two Bartonella, "Ca. N. mikurensis" and Bab. microti) are zoonotic pathogens except Anaplasma bovis and Anaplasma ovis with zoonotic potential. Furthermore, dual infection was observed between different microorganisms, and the most common type of co-infection is between "Ca. N. mikurensis" and other microorganisms. Additionally, potential novel Bartonella species and Hepatozoon species demonstrated the presence of more diverse rodent-associated Bartonella and Hepatozoon. Conclusions: The results in this work indicated great genetic diversity of vector-borne infections in wild rodents, and highlighted the potential risk of human pathogens transmitted from rodents to humans through vectors. In China, little information is known about the infection of vector-borne pathogens in rodents although the rodent species are very diverse. In this study, seven wild rodent species were collected to identify the genetic diversity of vector-borne bacteria and protozoan. Interestingly, diverse vector-borne microorganisms, including four Anaplasma, five Ehrlichia, "Candidatus Neoehrlichia mikurensis", six Bartonella, Babesia microti and diverse Hepatozoon were identified in six rodent species. Furthermore, two potential novel Bartonella species were detected. All these data suggested great genetic diversity of vector-borne pathogens infections in rodents. Background: Ascetosporea (Endomyxa, Rhizaria) is a group of unicellular parasites infecting aquatic invertebrates. They are increasingly being recognized as widespread and important in marine environments, causing large annual losses in invertebrate aquaculture. Despite their importance, little molecular data of Ascetosporea exist, with only two genome assemblies published to date. Accordingly, the evolutionary origin of these parasites is unclear, including their phylogenetic position and the genomic adaptations that accompanied the transition from a free-living lifestyle to parasitism. Here, we sequenced and assembled three new ascetosporean genomes, as well as the genome of a closely related amphizoic species, to investigate the phylogeny, origin, and genomic adaptations to parasitism in Ascetosporea. Results: Using a phylogenomic approach, we confirm the monophyly of Ascetosporea and show that Paramyxida group with Mikrocytida, with Haplosporida being sister to both groups. We report that the genomes of these parasites are relatively small (12–36 Mb) and gene-sparse (~ 2300–5200 genes), while containing surprisingly high amounts of non-coding sequence (~ 70–90% of the genomes). Performing gene-tree aware ancestral reconstruction of gene families, we demonstrate extensive gene losses at the origin of parasitism in Ascetosporea, primarily of metabolic functions, and little gene gain except on terminal branches. Finally, we highlight some functional gene classes that have undergone expansions during evolution of the group. Conclusions: We present important new genomic information from a lineage of enigmatic but important parasites of invertebrates and illuminate some of the genomic innovations accompanying the evolutionary transition to parasitism in this lineage. Under adverse environmental conditions, microorganisms are able to enter a state of cellular dormancy which consists of cell cycle arrest and interruption of multiplication. This process ensures their perpetuation in the infected host organism and enables the spread of disease. Throughout biological evolution, dormancy allowed microorganisms to persist in a harsh niche until favorable conditions for their reactivation were re‐established.
