Background: The liver has a solid inbuilt antioxidant defense system to regulate oxidative stress. However, exposure to an excessive level of ROS causes liver injury. This study examined the cytoprotective effect of neoxanthin, a xanthophyll antioxidant molecule isolated from Solanum trilobatum in stress-induced HepG2 cells. Methods and results: The cytotoxic effect of H2O2 and cytoprotective potential of β-carotene, lutein, and neoxanthin was analyzed by WST-1 assay. The intracellular ROS level and mitochondrial membrane potential (MMP) were measured using DCFH-DA (2′, 7′-dichlorofluorescin diacetate) and JC-10 MMP assay. The expression of anti-oxidant and apoptotic markers was measured by western blot analysis. Neoxanthin pretreatment exhibited better protection than β-carotene and lutein against cell death caused by H2O2. It significantly arrested H2O2-mediated elevation of intracellular ROS levels and protected MMP. The intracellular antioxidant enzymes HO-1 and SOD-2 were upregulated by neoxanthin pretreatment. Neoxanthin also activated the protein expression of redox-sensitive transactivation factors, Nrf2 and NF-kB. The cytoprotective effect of neoxanthin was associated with increased expression of the anti-apoptotic protein, Bcl-2 and decreased pro-apoptotic protein Bax. Conclusions: For the first time, our results demonstrate that neoxanthin offers adequate protection against stress-mediated cytotoxicity in hepatocytes by activating the intracellular antioxidant defense system and blocking apoptosis. SUMMARY: Neoxanthin (Neo), which is only bound to the peripheral antenna proteins of photosystem (PS) II, is a conserved carotenoid in all green plants. It has been demonstrated that Neo plays an important role in photoprotection and its deficiency fails to impact LHCII stability in vitro and indoor plant growth in vivo. Whether Neo is involved in maintaining the PSII complex structure or adaptive mechanisms for the everchanging environment has not yet been elucidated. In this study, the role of Neo in maintaining the structure and function of the PSII‐LHCII supercomplexes was studied using Neo deficient Arabidopsis mutants. Our results show that Neo deficiency had little effect on the electron transport capacity and the plant fitness, but the PSII‐LHCII supercomplexes were significantly impacted by the lack of Neo. In the absence of Neo, the M‐type LHCII trimer cannot effectively associate with the C2S2‐type PSII‐LHCII supercomplexes even in moderate light conditions. Interestingly, Neo deficiency also leads to decreased PSII protein phosphorylation but rapid transition from state 1 to state 2. We suggest that Neo might enforce the interactions between LHCII and the minor antennas and that the absence of Neo makes M‐type LHCII disassociate from the PSII complex, leading to the disassembly of the PSII‐LHCII C2S2M2 supercomplexes, which results in alterations in the phosphorylation patterns of the thylakoid photosynthetic proteins and the kinetics of state transition. Significance Statement: Although Neoxanthin (Neo), existing in all green plants, is dispensable for plant growth and development in controlled conditions, it might play unidentified roles in adaptive mechanisms for the everchanging environment. Here we show that the interaction pattern of PSII‐LHCII supercomplexes and the state transition kinetics are significantly impacted in three Neo deficient Arabidopsis mutants, highlighting the important role of Neo in stabilizing PSII high‐order structures for maintaining high light harvesting and energy transfer efficiency under everchanging conditions. In this study, two carotenoid antioxidants lutein and neoxanthin were investigated for their ability to prevent oxidatively induced DNA damage. A Chlamydomonas reinhardtii alga was used as the natural source of lutein and neoxanthin. Analysis using HPLC with Diode Array Detector (DAD) showed that lutein and neoxanthin were the most abundant carotenoids in the extract of C. reinhardtii. An analysis using GC–MS/MS was performed to measure the damage induced by Fenton reaction to DNA and to evaluate the ability of lutein, neoxanthin and C. reinhardtii extract to inhibit such damage. The results showed that lutein and neoxanthin have the ability to reduce oxidatively induced DNA base damage to less than half of its magnitude. Extract of C. reinhardtii inhibited about 76% of the damage in comparison to control sample. Neoxanthin in lower concentrations than those of lutein proved to be better inhibitor of oxidatively induced DNA damage. [ABSTRACT FROM AUTHOR]

This study investigates the impact of bunch microclimate on the evolution of some relevant carotenoids in Nebbiolo grapes. Four bunch-zone microclimates, defined by different vineyard aspect and vine vigor, were characterized by radiation and temperature indices. Berry samples were collected from green phase up to harvest, during two consecutive seasons and carotenoid determination was assessed by High-Performance Liquid Chromatography (HPLC). High carotenoid concentrations were highlighted in Nebbiolo. Lutein and neoxanthin contents (μg berry−1) varied similarly in both seasons achieving a concentration peak after veraison especially in the cooler plots while a variety effect on the lutein seasonal trend was presumed. Conversely, β-carotene content remained generally constant during ripening, with the exception of the south plots showing dissimilar evolution between the seasons. Furthermore, higher temperature in the less vigorous and south facing vineyards led to lower amounts of carotenoids, both during ripening and at harvest. Bunch zone temperature and light condition may affect both synthesis and degradation of grape carotenoids determining their amount and profile at harvest. These findings add further knowledge about the influence of climate changes on grape aroma precursors, and are useful to adapt cultural strategies and preserve grape quality consequently. Nitrogen limitation is considered a good strategy for enhancement of algal lipid production while conversely N repletion has been shown to result in biomass rich in proteins. In this study, the influence of long-term N limitation on Euglena gracilis fatty acid (FA), protein, chlorophyll a, and carotenoid concentrations was studied in N limited cultures. Biomass composition was analyzed from three-time points from N starved late stationary phase cultures, exposed to three different initial N concentrations in the growth medium. Total lipid content increased under N limitation in ageing cultures, but the low N content and prolonged cultivation time resulted in the formation of a high proportion of saturated FAs. Furthermore, growth as well as the production of proteins, chlorophyll a and carotenoids were enhanced in higher N concentrations and metabolism of these cellular components stayed stable during the stationary growth phase. Our findings showed that a higher N availability and a shorter cultivation time is a good strategy for efficient E. gracilis biomass production, regardless of whether the produced biomass is intended for maximal recovery of polyunsaturated FAs, proteins, or photosynthetic pigments. Oncidium 'Gower Ramsey' ( Onc. GR) is a popular cut flower, but its colour is limited to bright yellow. The β-ring carotene hydroxylase ( BCH2) gene is involved in carotenoid biogenesis for pigment formation. However, the role of BCH2 in Onc. GR is poorly understood. Here, we investigated the functions of three BCH2 genes, BCH-A2, BCH-B2 and BCH-C2 isolated from Onc. GR, to analyse their roles in flower colour. RT- PCR expression profiling suggested that BCH2 was mainly expressed in flowers. The expression of BCH-B2 remained constant while that of BCH-A2 gradually decreased during flower development. Using Agrobacterium tumefaciens to introduce BCH2 RNA interference ( RNAi), we created transgenic Oncidium plants with down-regulated BCH expression. In the transgenic plants, flower colour changed from the bright yellow of the wild type to light and white-yellow. BCH-A2 and BCH-B2 expression levels were significantly reduced in the transgenic flower lips, which make up the major portion of the Oncidium flower. Sectional magnification of the flower lip showed that the amount of pigmentation in the papillate cells of the adaxial epidermis was proportional to the intensity of yellow colouration. HPLC analyses of the carotenoid composition of the transgenic flowers suggested major reductions in neoxanthin and violaxanthin. In conclusion, BCH2 expression regulated the accumulation of yellow pigments in the Oncidium flower, and the down-regulation of BCH-A2 and BCH-B2 changed the flower colour from bright yellow to light and white-yellow. A novel super-complex of photosystem I (PSI)-light-harvesting complex I (LHCI) was isolated from a siphonaceous marine green alga, Bryopsis corticulans. The super-complex contained 9-10 Lhca antennas as external LHCI bound to the core complex. The super-complex was further disintegrated into PSI core and LHCI sub-complexes, and analysis of the pigment compositions by high-performance liquid chromatography revealed unique characteristics of the B. corticulans PSI in that one PSI core contained around 14 α-carotenes and 1-2 ε-carotenes. This is in sharp contrast to the PSI core from higher plants and most cyanobacteria where only β-carotenes were present, and is the first report for an α-carotene-type PSI core complex among photosynthetic eukaryotes, suggesting a structural flexibility of the PSI core. Lhca antennas from B. corticulans contained seven kinds of carotenoids (siphonaxanthin, all- trans neoxanthin, 9′- cis neoxanthin, violaxanthin, siphonein, ε-carotene, and α-carotene) and showed a high carotenoid:chlorophyll ratio of around 7.5:13. PSI-LHCI super-complex and PSI core showed fluorescence emission peaks at 716 and 718 nm at 77 K, respectively; whereas two Lhca oligomers had fluorescence peaks at 681 and 684 nm, respectively. By comparison with spinach PSI preparations, it was found that B. corticulans PSI had less red chlorophylls, most of them are present in the core complex but not in the outer light-harvesting systems. These characteristics may contribute to the fine tuning of the energy transfer network, and to acclimate to the ever-changing light conditions under which the unique green alga inhabits. The trimeric light-harvesting complexes II (LHCII) of plants and green algae are pigment-protein complexes involved in light harvesting and photoprotection. Different conformational states have been proposed to be responsible for their different functions. At present, detergent-solubilized LHCII is used as a model for the 'light-harvesting conformation', whereas the 'quenched conformation' is mimicked by LHCII aggregates. However, none of these conditions seem to perfectly reproduce the properties of LHCII in vivo. In addition, several monomeric LHC complexes are not fully stable in detergent. There is thus a need to find conditions that allow analyzing LHCs in vitro in stable and, hopefully, more native-like conformations. Here, we report a study of LHCII, the major antenna complex of plants, in complex with amphipols. We have trapped trimeric LHCII and monomeric Lhcb1 with either polyanionic or non-ionic amphipols and studied the effect of these polymers on the properties of the complexes. We show that, as compared to detergent solutions, amphipols have a stabilizing effect on LHCII. We also show that the average fluorescence lifetime of LHCII trapped in an anionic amphipol is ~30 % shorter than in α-dodecylmaltoside, due to the presence of a conformation with 230-ps lifetime that is not present in detergent solutions. A pot experiment was conducted to compare the responses of a non-transgenic tobacco plant (WT) and plants with genetically prolonged life-span (SAG) to risk elements of As, Cd and Zn. Plants were grown in control soil and in soil with higher levels of risk elements. The pigment contents were established by HPLC and chlorophyll fluorescence parameters were measured from slow kinetics after a 15 min dark period with the PAM fluorometer. Top ( i.e. young) leaves of both WT and SAG plants were more sensitive to photoinhibition caused by these risk elements but plants showed acclimation to such elements in the bottom leaves. Plants differed in the participation of individual pigments of xanthophyll cycle: increased levels of risk elements seem to stimulate especially first (violaxanthin to antheraxanthin) and second (anhtheraxanthin to zeaxanthin) steps of the cycle in WT plants. BACKGROUND Wild vegetables have traditionally been consumed as part of the Mediterranean diet, being valuable sources of nutrients and bioactive compounds. The objective of this work was to analyse the carotenoid content of the edible young shoots of four species ( Asparagus acutifolius L., Humulus lupulus L., Bryonia dioica Jacq. and Tamus communis L.) as part of a wider study on the characterisation of the nutritional composition of wild edible plants commonly consumed in Spain. Samples were gathered from two locations in Central Spain for two consecutive years. RESULTS Lutein, β-carotene, neoxanthin and violaxanthin were quantified by high-performance liquid chromatography with a photodiode array detector. Median concentration ranges (mg g−1 edible wet weight) were: β-carotene, 33.9-66.9; lutein, 54.4-191.3; neoxanthin, 51.7-173.7; and violaxanthin, 20.8-89.3. The highest carotenoid content was that of B. dioica (590.1 mg g−1) and the lowest was found in A. acutifolius (175.8 mg g−1). The biosynthesis pathway to diadinoxanthin and fucoxanthin was elucidated in Phaeodactylum tricornutum by a combined approach involving metabolite analysis identification of gene function. For the initial steps leading to β-carotene, putative genes were selected from the genomic database and the function of several of them identified by genetic pathway complementation in Escherichia coli. They included genes encoding a phytoene synthase, a phytoene desaturase, a ζ-carotene desaturase, and a lycopene β-cyclase. Intermediates of the pathway beyond β-carotene, present in trace amounts, were separated by TLC and identified as violaxanthin and neoxanthin in the enriched fraction. Neoxanthin is a branching point for the synthesis of both diadinoxanthin and fucoxanthin and the mechanisms for their formation were proposed. A single isomerization of one of the allenic double bounds in neoxanthin yields diadinoxanhin. Two reactions, hydroxylation at C8 in combination with a keto-enol tautomerization and acetylation of the 3′-HO group results in the formation of fucoxanthin. [ABSTRACT FROM AUTHOR]

Ulam or traditional vegetables in Malaysia comprise more than 120 species representing various families ranging from groundcovers, shrubs to trees. The leaves, shoots, flowers, fruits, roots and rhizomes of the vegetables are eaten fresh as salad or cooked and are consumed to add variety and flavor to the diet, as well as for their health benefits. Ulam species are rich in carbohydrate, protein, mineral and vitamin. This study established that ulam species differ greatly with respect to types and concentrations of carotenoids in leaves. A total of 10 species were evaluated for quantitative and qualitative carotenoid composition through spectrophotometry and HPLC analysis. The main carotenoids identified in these selected ulam were lutein, neoxanthin, violaxanthin, zeaxanthin and ß-carotene. The ratio of these carotenoids varies between species. [ABSTRACT FROM AUTHOR]

Epoxyxanthophylls (epoxide-containing xanthophylls), a group of carotenoids, are ubiquitously distributed in edible plants. Among them, neoxanthin in green leafy vegetables and fucoxanthin in brown algae have been reported to exhibit an antiproliferative effect on several human cancer cells in vitro. However, there is little information about the intestinal absorption and metabolic fate of dietary epoxyxanthophylls in humans. To estimate the intestinal absorption of neoxanthin and fucoxanthin in humans, we evaluated the plasma epoxyxanthophyll concentrations before and after 1-week dietary interventions with spinach (Spinacia oleracea) and wakame (Undaria pinnatifida). The epoxyxanthophylls and their metabolites in the plasma extracts were determined by HPLC after partial purification and concentration with solid-phase extraction cartridges. Even after 1 week of spinach intake (3·0 mg neoxanthin/d), the plasma concentrations of neoxanthin and its metabolites (neochrome stereoisomers) remained very low (about 1 nmol/l), whereas those of β-carotene and lutein were markedly increased. Similarly, the plasma concentration of fucoxanthinol, a gastrointestinal metabolite of fucoxanthin, was < 1 nmol/l after 1 week of wakame intake (6·1 mg fucoxanthin/d). These results indicated that the plasma response to dietary epoxyxanthophylls was very low in humans even after 1-week intake of epoxyxanthophyll-rich diets. [ABSTRACT FROM PUBLISHER]

Abstract: The location of the neoxanthin binding site in CP26 and CP29 was investigated by site-directed mutagenesis. The crystallographic structure of LHCII shows that the binding of neoxanthin to the N1 site is stabilised by an H bond with a tyrosine in the lumenal loop. This residue is conserved in CP26 and CP29. Mutation of this tyrosine into phenylalanine induced specific loss of neoxanthin without affecting violaxanthin binding. In contrast to previous proposals, it is thus concluded that also in these minor antenna complexes neoxanthin is accommodated in the N1 site. A novel abscisic acid (ABA)-deficient mutant, aba4, was identified in a screen for paclobutrazol-resistant germination. Compared with wild-type, the mutant showed reduced endogenous ABA levels in both dehydrated rosettes and seeds. Carotenoid composition analysis demonstrated that the defective locus affects neoxanthin synthesis. The ABA4 gene was identified by map-based cloning, and found to be a unique gene in the Arabidopsis genome. The predicted protein has four putative helical transmembrane domains and shows significant similarity to predicted proteins from tomato, rice and cyanobacteria. Constitutive expression of the ABA4 gene in Arabidopsis transgenic plants led to increased accumulation of trans-neoxanthin, indicating that the ABA4 protein has a direct role in neoxanthin synthesis. aba4 mutant phenotypes were mild compared with previously identified ABA-deficient mutants that exhibit vegetative tissue phenotypes. Indeed, ABA levels in seeds of aba4 mutants were higher than those of aba1 mutants. As aba1 mutants are also affected in a unique gene, this suggests that ABA can be produced in the aba4 mutant by an alternative pathway using violaxanthin as a substrate. It appears, therefore, that in Arabidopsis both violaxanthin and neoxanthin are in vivo substrates for 9- cis-epoxycarotenoid dioxygenases. Furthermore, significantly reduced levels of ABA were synthesized in the aba4 mutant on dehydration, demonstrating that ABA biosynthesis in response to stress must occur mainly via neoxanthin isomer precursors. Photosynthetic antenna systems are mainly involved in the absorption of light energy required for photosynthesis. The typical green plants arrange chlorophylls a and b and carotenoids, including lutein and 9′- cis neoxanthin, in their antenna systems; such antenna systems have prospered on earth. Therefore, these antenna systems should be highly evolved and should adapt to the photoenvironments in which plants grow. However, little information is available on the diversity and evolution of antenna systems in green plants as a whole. To approach this, the present study focused on the antenna systems in the Prasinophyceae, an assemblage of early diverging lineages of green plants and analyzed their photosynthetic pigments in detail. In the present study, various novel blue–green light-absorbing siphonaxanthin series were detected in the early diverging species of the Prasinophyceae and the distribution of these carotenoids was revealed. Additionally, to clarify the evolution of antenna systems in the Ulvophyceae, a highly developed green algal group that specializes in inhabiting various aquatic environments, members of the Cladophorales belonging to this class were selected and their carotenoid compositions were determined to compare them with the molecular phylogenetic tree constructed on the basis of the 18S rRNA gene sequences of the Cladophorales. In this review, these data will be summarized and the remarkable variation of photosynthetic pigments will be presented. Neoxanthin, a major carotenoid in green leafy vegetables, was reported to exhibit potent antiproliferative effect via apoptosis induction on human prostate cancer cells. However, the metabolic fate of dietary neoxanthin in mammals remains unknown. In the present study, we investigated the gastrointestinal metabolism of neoxanthin in mice and the in vitro digestion of spinach, and estimated the antiproliferative effect of neoxanthin metabolites on PC-3 human prostate cancer cells. Two hours after the oral administration to mice of purified neoxanthin, unchanged neoxanthin and stereoisomers of neochrome (8'-R/S) were detected in the plasma, liver, and small intestinal contents. To estimate the effect of intragastric acidity on the conversion of dietary neoxanthin into neochrome (epoxide-furanoid rearrangement), spinach was digested in vitro by incubating it with a pepsin-HCl solution at pH 2.0 or 3.0 (gastric phase) followed by a pancreatin-bile salt solution (intestinal phase). Spinach neoxanthin was largely converted into (R/S)-neochrome during the digestion when the gastric phase was set at pH 2.0, whereas the rearrangement was observed to a lesser extent at pH 3.0. (R/S)-neochrome dose-dependently inhibited the proliferation of PC-3 cells as well as neoxanthin at concentrations < or = 20 micromol/L. Although neoxanthin induced evident apoptotic cell death, (R/S)-neochrome inhibited the cell proliferation without obvious apoptosis induction. These results indicate that dietary neoxanthin is partially converted into (R/S)-neochrome by intragastric acidity before intestinal absorption and that (R/S)-neochrome exhibits an antiproliferative effect on PC-3 cells by the induction of cytostasis. High irradiance (HI) effects on xanthophyll cycle pigments (XCP) and activity of violaxanthin de-epoxidase (VDE) in terms of de-epoxidation index (DEI) were studied in soybean calli. The calli from the hypocotyl segments of 5-d seedlings were induced on a solid (1.1 % agar) MS medium (pH 5.8) supplemented with 4.52 μM 2,4-dichloro-phenoxyacetic acid, 2.32 μM kinetin, and 3 % sucrose. After a 30 d cultivation, the green calli were irradiated for 24 h with “white light” (HI, 1 300 μmol m−2 s−1) and VDE was isolated from the photosystem 2 (PS2) particles. In the control (0 h irradiation) callus, the reaction of PS2 particles with VDE in the presence or absence of Tween 20 resulted in the decrease of VIO content and the increase of ZEA content. In the 24 h HI-callus, the reaction of PS2 particles in the absence of VDE led to the decrease of VIO and ANT contents and increase of ZEA content. In the control, DEIs in the presence of VDE with or without 0.1 % Tween 20 (1.04 and 1.06, respectively) were significantly higher than the DEI (0.76) in the absence of VDE. In the HI-callus, DEIs in the presence of VDE with or without 0.1 % Tween 20 (0.98 and 0.96, respectively) were similar to that (1.03) in the absence of VDE. [ABSTRACT FROM AUTHOR]

The photosynthetic pigment composition of Mesostigma viride Lauterborn, a primitive green alga, was determined. This alga contained chl a and b , lycopene, lutein, siphonaxanthin, γ-carotene, β-carotene, antheraxanthin, violaxanthin, neoxanthin, and two novel carotenoid fatty acid esters, siphonaxanthin C12:0 ester and siphonaxanthin C14:0 ester. The esters were saturated, whereas all previously identified siphonaxanthin and loroxanthin esters have been mono-unsaturated (trans -Δ2). Neoxanthin was the all-trans form. This is the first such case detected in the chloroplasts of green plants. The 9′-cis form of neoxanthin is believed to be universally present in the chloroplasts of green plants (Streptophyta and Chlorophyta) and is a precursor of abscisic acid. However, the 9′-cis form was not found in M. viride. Based on these results, we discuss the phylogenetic implications and early evolution of the antenna pigment system in green plants. Foliage of Scots pine ( Pinus sylvestris L.) and pedunculate oak ( Quercus robur L.) was collected in a mixed pine/oak forest at canopy positions differing in radiation environment. In both species, chlorophyll (Chl) a/b ratios were higher in foliage of canopy positions exposed to higher irradiance as compared to more shaded crown layers. Throughout the growing season, pine needles exhibited significantly lower Chl a/b ratios than oak leaves acclimated to a similar photon availability. Hence, pine needles showed shade-type pigment characteristics relative to foliage of oak. At a given radiation environment, pine needles tended to contain more neoxanthin and lutein per unit of Chl than oak leaves. The differences in pigment composition between foliage of pine and oak can be explained by a higher ratio of outer antennae Chl to core complex Chl in needles of P. sylvestris which enhances the efficiency of photon capture under limiting irradiance. The shade-type pigment composition of pine relative to oak foliage could have been due to a reduced mesophyll internal photon exposure of chloroplasts in needles of Scots pine, resulting from their xeromorphic anatomy. Two cultivars of Capsicum annuum L. were acclimated for 5 d at sub-optimal temperature (14 °C) and irradiance of 250 µmol m−2 s−1. This cold-hardening resulted in some reduction in the extent of photoinhibition during an 8 h exposure to high irradiance at 4 °C. Obvious differences were observed between non-hardened leaves (NHL) and cold-hardened leaves (CHL) in the recovery under low irradiance at room temperature. The CHL of both cultivars recovered faster than NHL, especially during the initial fast phase of recovery. Compared with NHL, the total content of carotenoids (Cars), based on chlorophyll, Chl ( a+ b), and the proportions of xanthophyll cycle pigments referred to total Cars increased in CHL, mainly due to an increase of violaxanthin (V) + antheraxanthin (A) + zeaxanthin (Z) content per mol Chl ( a+ b). Faster development and a higher non-photochemical quenching (NPQ) of Chl fluorescence, related to a stronger deepoxidation of the larger xanthophyll cycle pool in NHL, could act as a major defence mechanism to reduce the formation of reactive oxygen species during severe chilling. This is suggested by higher content of Z or Z+A in photoinhibition as well as by its rapid decline during the initial fast phase of recovery. Recent research has revealed that abscisic acid (ABA), synthesised in response to water stress, is an apo-carotenoid. Two potential carotenoid precursors, 9′-cis-neoxanthin and 9-cis-violaxanthin, have been identified in light-grown and etiolated leaves, and in roots of a variety of species. Experiments utilizing etiolated Phaseolus vulgaris leaves and deuterium oxide strongly suggest that 9′-cis-neoxanthin, synthesised from all-trans-violaxanthin, is the immediate pre-cleavage precursor of ABA. The cleavage of 9′-cis-neoxanthin, performed by an inducible and specific dioxygenase, is likely to be the rate-limiting step in ABA biosynthesis. Any apocarotenoids formed as by-products of cleavage are probably rapidly degraded by lipoxygenase or related enzymes. The distribution and geometric form (cis or trans) of neoxanthin, one of the major carotenoids of oxygenic phototrophs, were systematically examined. The 9′-cis form of neoxanthin, but not the all-trans form, was found in chlo-roplasts of seed plants, ferns, mosses and green algae, all of which contain chlorophylls a and b. In contrast, neoxanthin was not found in other algal classes, such as Hetero-kontophyta, Rhodophyta and oxygenic phototrophic pro-karyotes. Consequently, with regard to phylogeny, the appearance of neoxanthin appears to be associated with the appearance of chlorophyll b. In non-photosynthetic organs, such as petals and fruits, the presence of neoxanthin was classified into four types; those having only the 9′-cis form, those with only the all-trans form, those with both forms, and those without either form. Thus only 9′-cis neoxanthin is found in chloroplasts, and the all-trans neoxanthin is found only in non-photosynthetic organs. Because the absorption spectra of both forms are almost identical, their functions in photosynthesis might be similar. 9′-Cis neoxanthin is not involved in the xanthophyll-cycle, whereas it is a suitable substrate for abscisic acid synthesis. The article presents information on a study regarding antiproliferative effect of neoxanthin and fucoxanthin on cultured cells. Neoxanthin and fucoxanthin are one of the major xanthophylls in the chloroplast of higher plants and brown algae, respectively. The present study indicated that fucoxanthin was more effective in reducing the viability of human colorectal adeno carcinoma human colon cancer cells as well as PC-3 human prostate cancer cells than the viability of the other cancer and normal cell lines tested.
