The present investigation addresses whether the midgut (MG) of Atlantic cod ( Gadus morhua L.) is an infection route for Vibrio ( Listonella) anguillarum serotype 02 β and if Carnobacterium maltaromaticum, a probiotic bacterium, can out-compete the pathogen and modulate the autochthonous MG microbiota. This was investigated by using an ex vivo method the intestinal sac, utilized previously in studies on Atlantic cod and Atlantic salmon ( Salmo salar L.). Exposure of the MG to V. ( L.) anguillarum did not reveal any cell damage indicating that the MG does not appear to be an infection route for V. ( L.) anguillarum in healthy Atlantic cod. This finding together with previous observations on Atlantic cod and Atlantic salmon indicate that intestine as an infection route might vary between these two species. When the MG was exposed to C. maltaromaticum, no cell damage or cellular disruptions were observed. As budding from the apices of microvilli was observed in all treatments exposed to bacteria, we suggest that budding might be involved in the primary barrier against bacterial infection. However, to clarify this hypothesis, further studies are needed. Exposure of the MG to the probiotics and pathogenic bacteria indicated that C. maltaromaticum, to some extent, is able to out-compete V. ( L.) anguillarum but the topic merits further investigation. Analysis of the MG microbiota after sterile saline solution and bacterial exposure indicates that bacteria related to Staphylococcus sciuri belong to the autochthonous gut microbiota in Atlantic cod. Intensive fish production worldwide has increased the risk of infectious diseases. However, before any infection can be established, pathogens must penetrate the primary barrier. In fish, the three major routes of infection are the skin, gills and gastrointestinal (GI) tract. The GI tract is essentially a muscular tube lined by a mucous membrane of columnar epithelial cells that exhibit a regional variation in structure and function. In the last two decades, our understanding of the endocytosis and translocation of bacteria across this mucosa, and the sorts of cell damage caused by pathogenic bacteria, has increased. Electron microscopy has made a valuable contribution to this knowledge. In the fish-farming industry, severe economic losses are caused by furunculosis (agent, Aeromonas salmonicida spp. salmonicida) and vibriosis [agent, Vibrio ( Listonella) anguillarum]. This article provides an overview of the GI tract of fish from an electron microscopical perspective focusing on cellular damage (specific attack on tight junctions and desmosomes) caused by pathogenic bacteria, and interactions between the ‘good’ intestinal bacteria [e.g. lactic acid bacteria (LAB)] and pathogens. Using different in vitro methods, several studies have demonstrated that co-incubation of Atlantic salmon ( Salmo salar L.) foregut (proximal intestine) with LAB and pathogens can have beneficial effects, the cell damage caused by the pathogens being prevented, to some extent, by the LAB. However, there is uncertainty over whether or not similar effects are observed in other species such as Atlantic cod ( Gadus morhua L.). When discussing cellular damage in the GI tract of fish caused by pathogenic bacteria, several important questions arise including: (1) Do different pathogenic bacteria use different mechanisms to infect the gut? (2) Does the gradual development of the GI tract from larva to adult affect infection? (3) Are there different infection patterns between different fish species? Background. Recently we reported on cerebral metabolic changes suggesting ischemia in piglets during nitroprusside-induced low-pressure CPB. We here investigated whether a mean arterial pressure (MAP) of 40-45 mmHg could provoke similar changes by a NO-independent intervention. Methods. Piglets underwent 60 minutes normothermic followed by 90 minutes hypothermic CPB. The LP-group (n=8) had MAP of 40-45 mmHg by phentolamine while the HP-group (n=8) had MAP of 60-80 mmHg by norepinephrine. Cerebral glucose, lactate, pyruvate and glycerol were determined. In the last two animals of each group, cerebral tissue was examined by electron microscopy. Results. Cerebral lactate was higher in the LP-group than the HP-group during normothermic CPB. Compared with baseline, cerebral glucose of the LP-group decreased whereas lactate/pyruvate-ratio, lactate and glycerol-concentrations increased during normothermic CPB. In the HP-group these parameters remained unchanged. Electron microscopy showed 31.2% and 8.3% altered mitochondria in the cortical micrographs taken from the LP- and the HP-group, respectively (p<0.001). Conclusion. Atlantic salmon ( Salmo salar L.) were fed fishmeal protein for 46 days, and 500 g kg−1 of fishmeal protein substituted with meal from Northern krill ( Meganyctiphanes norvegica). No differences were observed in weight gain, length gain, feed conversion or specific growth rate between the groups that could be attributed to dietary manipulation. The adherent microbiota in the hindgut of the two rearing groups were further investigated. By substituting fishmeal with krillmeal, the total viable counts of aerobic and facultative aerobic bacteria colonizing the hindgut of Atlantic salmon increased from 8.5 × 104 to 2.2 × 106. Furthermore, dietary krillmeal affected the adherent hindgut microbiota. The Gram-positive bacteria Carnobacteria piscicola, Microbacterium oxydans, Microbacterium luteolum and Staphylococcus equorum spp. linens and the Gram-negatives Psychrobacter spp. and Psychrobacter glacincola were not isolated from hindgut of fish fed the krill diet. On the other hand, Pseudomonas fulgida, Pseudomonas reactans and Stenotrophomonas maltophila were not isolated from the control group fed fishmeal. Acinetobacter lwoffi, which is not normally found in the fish gut, was isolated from both feeding groups. Transmission electron microscopy showed bacteria-like profiles between the hindgut microvilli in both feeding groups indicating autochthonous microbiota. The primary aim of the present study was to evaluate the population level of adherent (autochthonous) aerobic and facultative anaerobic bacteria in the hindgut of healthy Arctic charr ( Salvelinus alpinus L.) fed dextrin or inulin. This was assessed by the dilution plate technique, and visualized using both transmission and scanning electron microscopy. A population level of 4.8 × 105 adherent bacteria per gram wet mass was found in the hindgut of fish fed a casein-based diet supplemented with 15% dextrin. However, substituting dextrin with 15% inulin reduced the bacterial population level in the hindgut (3.56 × 104). A total of 217 bacterial isolates were identified by key phenotypical and biochemical characteristics. In addition, 22 strains were also identified by partial sequencing of the 16S rRNA gene. The composition of bacteria colonizing the hindgut of Arctic charr fed dextrin was dominated by the genera Staphylococcus, Pseudomonas, Micrococcus, Psychrobacter glacincola and Streptococcus. However, bacteria colonizing the hindgut of fish fed inulin were dominated by Gram-positive bacteria of the genera Staphylococcus, Streptococcus, Carnobacterium and Bacillus. While Carnobacterium divergens-like strains were isolated from charr fed dextrin, Carnobacterium maltaromicus-like strains were isolated from the hindgut of fish fed inulin. Electron microscopical analysis of hindgut regions confirmed traditional culture-based microbial analysis as fewer bacterial cells were observed between microvilli and associated with the surfaces of enterocytes of fish fed inulin rather than dextrin. The article focuses on the electron microscopic study of endocytosis of indigenous bacteria and cell damage caused by Vibrio anguillarum in the foregut and hindgut of spotted wolffish Anarhichas minor. The authors suggested two mechanisms involved in bacterial translocation of indigenous gut bacteria. The difference in bacterial translocation between indigenous gut bacteria and pathogens has not been elucidated. The results reached a hypothesis that paracellular bacteria translocation is a secondary effect of detachment and loss of enterocytes and increased intracellular gap when the fry is exposed to V. anguillarum.

In fish, bacterial pathogens can enter the host by one or more of three different routes: (a) skin, (b) gills and (c) gastrointestinal tract. Bacteria can cross the gastrointestinal lining in three different ways. In undamaged tissue, bacteria can translocate by transcellular or paracellular routes. Alternatively, bacteria can damage the intestinal lining with extracellular enzymes or toxins before entering. Using an in vitro (Ussing chamber) model, this paper describes intestinal cell damage in Atlantic salmon (Salmo salarL.) caused by the fish pathogenAeromonas salmonicidassp.salmonicida, the causative agent of furunculosis. The in vitro method clearly demonstrated substantial detachment of enterocytes from anterior region of the intestine (foregut) upon exposure to the pathogen. In the hindgut (posterior part of the intestine), little detachment was observed but cellular damage involved microvilli, desmosomes and tight junctions. Rainbow trout ( Oncorhynchus mykiss Walbaum) were fed purified diets containing fish oil for six weeks and then soybean lecithin or soybean oil for 25 days. The gastrointestinal tract segments, stomach, midgut and hindgut were then sampled for lipid and fatty acid composition and electron microscopy. Membrane lipid class composition was fairly similar in all three segments of trout fed fish oil. Hindgut contained slightly more phosphatidylserine than stomach and midgut, while midgut contained more phosphatidylcholine and less lysophospatidylcholine/sphingomyelin. Feeding soybean products appeared to marginally decrease free cholesterol. The fatty acid compositions of the main lipid classes showed significant regional differences. In control fish, stomach had higher levels of arachidonic acid (20:4n-6) and n-6 polyunsaturated fatty acids than midgut and hindgut, and lower content of docosahexaenoic acid (22:6n-3) and n-3 polyunsaturated fatty acids. Midgut phosphatidylethanolamine also had higher levels of saturated fatty acids and less n-3 polyunsaturated fatty acids than the other tissues. Feeding soybean products decreased the n-3/n-6 ratio mainly due to increases in linoleic (18:2n-6) and 20:4n-6 and decreases in 22:6n-3 and eicosapentaenoic acid (20:5n-3). Phosphatidylcholine and to a lesser extent phosphatidylethanolamine adapted more readily to dietary changes by major increases in 18:2n-6 and C20−22 n-6 polyunsaturated fatty acids. The composition of phosphatidyl-serine and -inositol appeared to be under more strict metabolic control. Linoleic acid hardly increased at all while the increase in other n-6 polyunsaturated fatty acids was less pronounced than for the other lipid classes. Regardless of lipid class, stomach resisted dietary changes more strongly than midgut and hindgut. Increases in n-6 polyunsaturated fatty acids were minor as were the loss of n-3 polyunsaturated fatty acids. The dead-end product 20:2n-6 accumulated to a higher degree in hindgut phosphatidyl-ethanolamine and -coline compared to midgut and stomach, suggesting that the activity of Δ6 desaturation is higher in the anterior part of the intestine where most of the lipid is absorbed. Feeding soybean oil caused massive accumulation of free lipid droplets in midgut enterocytes while little lipid droplets were observed in trout fed fish oil or soybean lecithin. Since both soybean products influenced intestinal composition to the same degree, altered fatty acid profiles in membranes is not responsible for the observed lipid accumulation. This supports previous observations in Arctic charr ( Salvelinus alpinus L.), suggesting that fish may require exogenous phospholipids in order to sustain a sufficient rate of lipoprotein synthesis. The aim of this study was to provide data on the morphology of the gastrointestinal tract of Adélie penguins (Pygoscelis adeliae). It was found to consist of a long oesophagus, a two-chambered stomach, a small intestine measuring only 5.2×body length, two rudimentary caeca and a short colon, typical of carnivorous birds. The stomach comprised a glandular proventriculus and a muscular gizzard that frequently contained grit. An acidic pH was recorded in both chambers. Ultrastructural studies of the small intestinal mucosal membrane revealed epithelial cells with elongated, irregular microvilli and high affinity for toluidine blue, absorptive intestinal epithelial cells and goblet cells. Numerous large lymphocyte-like cells were observed close to the brush border of the epithelium, and empty spaces on the epithelial surface reflected normal cell loss in the small intestine. Groups of Atlantic salmon ( Salmo salar L.) in feeding (guts filled with faeces) or fasted (three days of diet deprivation) states were subjected to 15 minutes of acute stress. Blood samples and intestinal tissue were collected and prepared for chemical and ultrastructural analyses at intervals post stress until 53 h of recovery. Subjecting fish to acute stress led to significant alterations of the ultrastructure of the enterocytes lining the gastrointestinal tract (GI tract). The most notable effect was substantial damage to the intercellular junctional complexes in midgut regions. These effects appeared within the first hour after stress, were maintained for at least 12 h and were more pronounced in fed than fasted fish. In contrast, hindgut was influenced less by stress and damage was rarely observed. Stress also influenced fish intestinal microbiota. Adherent bacteria decreased in both midgut and hindgut of stressed fish, and this was accompanied by a significant increase in the bacterial contents of faeces. It is suggested that this was due to the sloughing off of mucus eliminating existing microflora and allowing remaining bacteria (also pathogenic) in the gut lumen to colonize the surface of the enterocytes. Although blood haematocrit and plasma cortisol increased following stress, the response appeared to be greater in fasted fish. There were also significant differences in carbohydrate metabolism. While liver glycogen stores were depleted in fasted fish following the mobilization of glucose into plasma, liver glycogen was never depleted in fed fish. As a consequence, plasma glucose levels remained high for more than 12 h of recovery. In fed fish, plasma lactate was also higher than in fasted salmon, and the clearance rate appeared slower. Macrophage dysfunction is a likely mechanism underlying common diabetic complications such as increased susceptibility to infection, accelerated atherosclerosis, and disturbed wound healing. There are no available studies on the function of tissue macrophages in diabetes in humans. We have therefore studied peritoneal macrophages from diabetic type 2-like db/db mice. We found that the release of tumor necrosis factor-alpha and interleukin-1beta from lipopolysaccharide plus interferon-gamma-stimulated macrophages and vascular endothelial growth factor from both stimulated and nonstimulated macrophages was significantly reduced in diabetic animals compared with nondiabetic controls. Nitric oxide production from the stimulated db/db macrophages was significantly higher than that in the db/+ cultures, whereas there was no difference in their ability to generate reactive oxygen species. When studied both at light and electron microscopic levels, macrophages in diabetic animals had an altered morphological appearance compared with those of normal controls. We conclude that the function and morphology of the macrophages are disturbed in db/db mice and that this disturbance is related to the mechanisms underlying common inflammatory and degenerative manifestations in diabetes. The concentrations of catecholamines in the heart chambers of elasmobranchs were measured by the fluorimetric method of Bertler et al. (1958). Noradrenaline (NA) can be detected in all the chambers, but the sinus venosus is by far the richest in NA. This can either be due to the presence of storage sites for this amine in the sinus wall, or to a transport of amine to the sinus venosus from the anterior chromaffin bodies. The sinus wall contains large numbers of 'granule containing cells' and axon-like processes, both with numerous dense-core vesicles of about 1800 Å diameter. The dense-core vesicles contain a uranophilic matrix indicating the presence of protein, phospholipids and/or nucleic acid. The reactions failed to demonstrate amine, which may be due to a loss of amine by diffusion, to a relatively low intravesicular amine concentration, or, to the absence of amines in these granule-containing cells and processes. Heavy accumulations of granule-containing processes occur in the subendothelial area. The endothelium contains fenestrae and pores through which granule-containing fibres protrude into the venous cavity. Granule-containing cells are innervated by presumed cholinergic nerve endings. It is suggested that the granule-containing cells and fibres belong to a neurosecretory system with a cholinergic input, releasing the contents of the dense-core vesicles into the blood stream at the level of the venous cavity. The membrane systems of the cardiac muscle cell of Munida tenuimana G. O. Sars are described. The sarcolemma invaginates at the Z level, forming tubules. Narrow tubules branch off in a longitudinal direction from these transverse and radially arranged tubules, forming a narrow transverse collar at the H level where dyadic and triadic junctions are formed with the sarcoplasmic reticulum. Atrial specific granules first appear in the human auricle by about 7 weeks of fetal development. They are rapidly formed between the 7 and 8 week stages. Granules are fairly evenly spread at the onset of their appearance. As they increase in number they soon aggregate in the juxtanuclear and subsarcolemmal positions. The electron dense matrix of granules of young cardiomyoblasts is condensed and/or synthesized within the Golgi cisternae. Large and electron dense cisternae are sometimes observed in close connection with mature specific granules. Bristle coated vesicles develop by a 'budding off' process from the lateral margins of the Golgi cisternae. A fusion between bristle coated vesicles and specific granules is indicated. The myofibrillogenesis in the human embryonic heart is described. The synthesis of thin filaments, which are the first to appear, takes place in close proximity to smooth surfaced SR tubules. Z-band material is closely related to the thin filaments and appears first as irregularly distributed patches in the filamenteous mass. Further cellular differentiation includes an organization of the thin filaments/Z-band material. The synthesis of thick filaments, which follows that of the thin filaments, takes place in ribosome rich areas of the cell. They are rapidly incorporated into the strings of organized thin filaments/Z-band material. The periodic binding sites on both kinds of filaments are believed to play an important role in the precise ordering of the filaments. The formation of myofilaments in the adult hypertrophied human heart is also described. Primary 9+0 cilia have been found in the embryonic and the adult human heart. Proximally the cilia show the typical (+0 filament arrangement. Rearrangement of the filaments occur in their distal regions. The cilia are usually found in deep invaginations of the cell membrane, but can also be found in a superficial position. The ultrastructure of myocardial cells was studied in four patients with left ventricular myocardial hypertrophy due to aortic and/or mitral valvular disease. Signs of cellular lysis and synthesis were present in juxtanuclear, interfibrillar, and subsarcolemmal areas. Subsarcolemmal areas contained a granular matrix with foci of polyribosomes, mitochondria, well preserved membranes, a proliferating sarcoplasmic reticulum, and thick and thin filaments and Z-band formations at different stages of development. Z-band substance and thin filaments appeared in the cytoplasm in close proximity to free polyribosomes and endoplasmic membranes and were seemingly not associated with the sarcolemma. These findings were interpreted as evidence for a proteosynthetic activity leading to the formation of new myofibrils and sarcomeres during the chronic stage of hypertrophy. Comparative studies showed that this process has much in common with the genesis of contractile cardiac units in the mammalian embryo. Anomalous Z-band accumulations were studied in normal myocardial cells of various vertebrates and in cells of the overloaded human heart. The origin of cardiac myofibrils in cells from the atrial wall in human embryos was studied. Z-band substance appears throughout the cytoplasm as irregular electron dense patches in a network of thin filaments. The thin and thick filaments are synthesized as separate units in the sarcoplasm and are later aggregated into myofibrils. Complexes of Z substance and thin filaments occur numerously at different stages of myofibrillar organisation. A comparative study of the pigeon ventricular myocardial cell has been performed by transmission electron microscopy (TEM) and by scanning electron microscopy (SEM). Three-dimensional access to the cell interior was obtained by cryo-fracturing paraffin-embedded tissue immersed in liquid nitrogen. The TEM studies revealed parallelly arranged myofibrils separated by rows of mitochondria. The sarcoplasmic reticulum is represented by a well-developed network of tubules which, at the Z- and H-band level of the sarcomere, expands to form belt-like cisternae. The cisternae at the Z-band level lie in close proximity to both myofilaments and mitochondria. Transverse tubules are absent and thus only peripheral couplings are present. SEM observations of the fractured tissue revealed the spatial relationship between the different cell organelles, the most important of these being the parallel myofibrils and the mitochondria. The conspicuous ridges transversing the myofibril at the Z-band level consist mainly of expanded Z-bands, but overlying SR-tubules also contribute to these ridges. Traces of the SR can sometimes be seen covering the myofibrils. The close proximity between the SR and the mitochondria was also confirmed in the SEM. Preparation and examination of SEM prepared tissue in the TEM confirmed that no essential damage or reorganization of cell organelles had taken place during the SEM procedure. The ultrastructure of atrial and ventricular myocardial cells from Acipenser stellatus is described. The cells of the atrium are more loosely connected than those of the ventricle. Cell contact is by simple intercalated discs and by desmosomes. The cells are flattened, with peripheral myofibrils and a central region of mitochondria and the nucleus. The sarcoplasmic reticulum consists of subsarcolemmal tubules, that frequently extend towards the central mitochondria. Dyads are small and positioned at any sarcomeric level. Following subcutaneous injections of isoproterenol hydrochloride (ISO), atrial cells present a large number of partly degranulated or completely clear 'specific granules' enclosed by an intact membrane. Such profiles were never encountered in normal controls and might suggest ISO-induced release of a secretory product. Permeability of perigranular membrane was tested using the extracellular macromolecular tracer horseradish peroxidase (HRP). Reaction product was entirely absent within granules of atrial cells in which the sarcolemma was made permeable to HRP molecules by the ISO injections. This seemed to be the case even in heavily labelled cells in which the peroxidase had penetrated the mitochondrial membranes. In atrial cells impermeable to the tracer, the specific granules closely apposed to the sarcolemma were always HRP-negative. The transverse tubule (T-tubule) system in papillary muscles of the sand rat and the mouse were studied with the aid of a diffusion tracer (horseradish peroxidase). The T-tubule system in the sand rat showed a typical mammalian pattern with sarcolemmal tubules invaginating at the Z-band level of the sarcomere. These tubules follow a transverse direction in the cell with frequent longitudinal side-branches which connect tubules at different Z-band levels. In the mouse myocardium, the T-tubules also start as lateral invaginations from the sarcolemma at the Z-band levels. In the cell interior, however, the tubules ramify and brake up into a complicated system of spirally running tubules. The papillary muscle of the heart of adult white mice is investigated. Intrafibrillarly located leptomeric fibrils, frequently encountered in the Z-band region of the myofibrils. The leptomeric fibrils are always running in a transverse direction and often in close proximity to the transverse tubules (which are also located at this level). There seems to be a close connection between the dense striae of the leptofibrils and the Z-bands of ordinary myofibrils. The leptomeric fibrils are spindle-shaped and have a length varying between 0.6 and 1.2 μm. The banding periodicity of the fibrils is approximately 0.16 μm. The membrane systems of cardiac muscle cells of the euphausiacean Meganychtiphanes norvegica are described. Transverse tubules are found both at the Z-band level (T-tubules) and at the H-band level (T-tubules). Within the sarcomere narrow longitudinal tubules branch off from the T-tubules. At the H-band level these tubules expand forming flattened cisternae in dyadic and triadic couplings with the sarcoplasmic reticulum (SR). Adjacent myofibrils are separated by a well developed SR. The membrane systems of the cardiac muscle cell of the amphipod Tmetonyx cicada (O. Fabricius) are described. The sarcolemma invaginates and forms a transverse network of tubules at the level of the Z band. Narrow longitudinal tubules branch from the network and connect to another transverse network of tubules at the H band level, where dyadic and triadic junctions are formed with the sarcoplasmic reticulum. Adjacent myofibrils are normally separated by a well developed double layer of the sarcoplasmic reticulum. To determine the protective effect during ischaemia and reperfusion of removing oxygen radicals two groups of isolated Langendorff perfused rat hearts were arrested with cardioplegic solution at 4°C and kept ischaemic at 15°C for 210 min before being reperfused for 60 min at 37°C. To remove oxygen radicals superoxide dismutase and catalase were added to the cardioplegic solution and to the buffer during the first 30 min of reperfusion in one group, the other group serving as control. At the end of reperfusion the first derivative of left ventricular developed pressure (dP/dt), coronary flow, high energy phosphate concentrations, and ultrastructure were determined. The ultrastructure was examined using a stereological method based on point counting and the results presented as volume fractions (Vv). DP/dt after 60 min of reperfusion was 61.6(5.6)% (mean(SEM)) of the initial values in the control group and 77.6(3.4)% in the superoxide dismutase and catalase supplemented group (p<0.05). In the supplemented group coronary flow was significantly higher than in the control group but only in the first part of reperfusion. The concentrations of adenosine triphosphate and creatine phosphate in the control group were 9.9(1.0) and 19.6(1.8) μmol·g−1 dry weight respectively; corresponding values in the supplemented group were 14.4(2.1) and 29.4(3.6) μmol·g−1 dry weight. The morphometric examination of the ultrastructure showed no significant difference in interstitial fluid accumulation evaluated by Vv(myocyte/myocardium) measurements and there was no difference in mitochondrial alteration between the two groups. There was, however, a significant reduction in the volume fraction of cellular oedema (Vv(cell oedema/myocyte)) in the supplemented group. This may indicate a protective effect at the cell membrane level. The use of enzymes capable of removing oxygen radicals during and after prolonged hypothermic cardioplegia may partly prevent the loss of cellular integrity and contractile performance. To investigate the cardiac effects of enzymatically generated oxygen radicals isolated Langendorff rat heart preparations were perfused with hypoxanthine (0.96 mmol-litre−1) plus xanthine oxidase (0.025 U-ml−1). Oxygen radicals produced an immediate increase in coronary flow. After 10 min a pronounced reduction in contractile performance, as well as in concentrations of high energy phosphates, was seen. Electron microscopical examination showed damage with cellular oedema as a prominent finding. These effects were all effectively reversed by the specific enzymes, superoxide dismutase and catalase, proving that they were due to oxygen radicals. [ABSTRACT FROM PUBLISHER]
