Background: Studies on haemosporidian diversity, including origin of human malaria parasites, malaria's zoonotic dynamic, and regional biodiversity patterns, have used target gene approaches. However, current methods have a trade-off between scalability and data quality. Here, a long-read Next-Generation Sequencing protocol using PacBio HiFi is presented. The data processing is supported by a pipeline that uses machine-learning for analysing the reads. Methods: A set of primers was designed to target approximately 6 kb, almost the entire length of the haemosporidian mitochondrial genome. Amplicons from different samples were multiplexed in an SMRTbell® library preparation. A pipeline (HmtG-PacBio Pipeline) to process the reads is also provided; it integrates multiple sequence alignments, a machine-learning algorithm that uses modified variational autoencoders, and a clustering method to identify the mitochondrial haplotypes/species in a sample. Although 192 specimens could be studied simultaneously, a pilot experiment with 15 specimens is presented, including in silico experiments where multiple data combinations were tested. Results: The primers amplified various haemosporidian parasite genomes and yielded high-quality mt genome sequences. This new protocol allowed the detection and characterization of mixed infections and co-infections in the samples. The machine-learning approach converged into reproducible haplotypes with a low error rate, averaging 0.2% per read (minimum of 0.03% and maximum of 0.46%). The minimum recommended coverage per haplotype is 30X based on the detected error rates. The pipeline facilitates inspecting the data, including a local blast against a file of provided mitochondrial sequences that the researcher can customize. Conclusions: This is not a diagnostic approach but a high-throughput method to study haemosporidian sequence assemblages and perform genotyping by targeting the mitochondrial genome. Accordingly, the methodology allowed for examining specimens with multiple infections and co-infections of different haemosporidian parasites. The pipeline enables data quality assessment and comparison of the haplotypes obtained to those from previous studies. Human influences on natural environments are now ubiquitous but manifest in multiple and unique ways depending on local environments and communities. Attempts to control, or mediate, local pests to residences or to agriculture can impart important negative consequences on systems. Secondary exposure to anticoagulant rodenticides (ARs) can cause numerous adverse effects on wild carnivores including death. Few studies have quantified AR prevalence, investigated their pathway of exposure, or associations with specific location types in the northeastern U.S. We hypothesized that ARs would be found in the mesocarnivore community throughout Pennsylvania and have the greatest detection rate in highly urbanized or agricultural landscapes. From 2019 through early 2022, we collected carcasses to obtain liver samples (n = 265) from three species of carnivores: bobcats (Lynx rufus (Schreber, 1777)), fishers (Pekania pennanti (Erxleben, 1777)), and river otters (Lontra canadensis (Schreber, 1777)). We used generalized linear models to test for differences in AR detection rates among species and spatial scales including the six Pennsylvania Game Commission regions and 23 wildlife management units. Summary: The long‐term outcomes of adults with Fanconi anaemia (FA) have improved with advances in haematopoietic stem cell transplantation (HSCT) and more detailed follow‐up and screening guidelines. The phenotype of those who survive to adulthood may differ from the typical presentation of FA. We collected retrospective clinical data on adults with FA who received their care at the Cincinnati Children's Hospital Medical Center. In our final cohort of 52 patients, there were 29 females and 23 males, with median (range) age of 21 (18–37) years. Overall, 42 patients (81%) were alive at last follow‐up. In all, 36 adults (69%) had undergone HSCT, including eight who had developed myelodysplasia or acute myeloid leukaemia. Eight (15%) developed squamous cell carcinoma. Endocrine complications were common, including hypothyroidism (42%), diabetes (10%), low body mass index (31%) and low bone mineral density (51%). The majority of adults with FA were employed (52%) or full‐time students (13%). A significant subset of patients with FA are surviving into adulthood without requiring HSCT. Endocrine abnormalities and the development of solid tumours complicate adulthood. With improved survival outcomes following HSCT and more aggressive malignancy screening protocols, ongoing longitudinal analysis will be important to further characterise this cohort and the phenotype of untransplanted adults with FA. The purpose of this study was to understand Polynesian American (PA) values, preferences, and beliefs about psychotherapy in light of their culture. An Interpretative Phenomenological Analysis (IPA) was conducted to collect and analyze culturally relevant preferences and expectations of psychotherapy with Polynesian Americans. The study consisted of 13 in-depth interviews with individuals of Pacific Islander descent who are currently living in the United States. The results of the analysis showed three culturally informed themes shared by study participants that informed this sample's expectations and preferences of psychotherapy: ʻOhana (family), Lōkahi (harmony) and Aloha (warmth, compassion, love). These values provide unique insights to therapy adaptations that should be emphasized when working with Polynesian American clients, such as using a family centered approach to therapy that takes into account the collective needs of a client's entire family, participating in therapist self-disclosure and the sharing of personal backgrounds, looking at clients challenges through a holistic lens, and demonstrating genuine warmth in the client-counsellor relationship. Special education (SPED) teacher preparation programs are intended to help preservice teachers (PSTs) learn and adopt effective teaching practices to serve students with special needs. To help SPED-PSTs learn needed skills, SPED mentor teachers (MTs) must employ high-leverage practices (HLPs) such as performance feedback, conferencing, and modeling. Although most teacher preparation programs utilize modeling in preparing PSTs, little is known about how SPED-MTs use this mentoring tool. An interpretative phenomenological analysis was conducted to explore how SPED-MTs engaged PSTs in modeling practices during a practicum experience. Data from semi-structured interviews were analyzed to describe SPED-PST and SPED-MT attitudes, experiences, and use of modeling over the course of a special education practicum, focusing on MTs' modeling strategies and PSTs' subsequent behaviors. The relative impacts of simple and cognitive modeling on the PST-MT relationship were compared. SPED-MTs' modeling foci were compared to recommended HLPs. This qualitative case study describes three adult siblings' experiences and their perceptions of support connected with the time before and after their father's suicide. At the time of the suicide, participants were ages 1, 5, and 8 years old. We considered commonalities and disparities among the three survivors' perceptions. We also considered how their reported experiences compared to extant literature on child survivors of parent suicide. Our findings suggest that, although the siblings experienced the same traumatic event, each had unique perceptions of the parent's suicide. Immediately prior to closing each interview, to deescalate from the intense topic of suicide, participants offered their impressions of potentially therapeutic children's books and how bibliotherapy may or may not support surviving children. Participants' perceptions of selected children's picture books offer insights about opening communication and addressing challenges specific to a parent's suicide. Implications for teachers, parents, and school-based mental health professionals are provided. We conclude that postvention must consider and monitor each child's perceptions and provide individualized interventions that encourage open communication and support adaptive coping to navigate the intense grief associated with a parent's suicide. Highlights: Following a parent's suicide, children's memories are highly influenced by what adults say and do following the suicide. Even though siblings share a similar trauma, their narratives may indicate highly individualized memories and responses. Highly mobile predators can show strong numerical responses to pulsed resources, sometimes resulting in irruptions where large numbers of young invade landscapes at a continental scale. High production of young in irruption years may have a strong influence on the population dynamics unless immature survival is reduced compared to non-irruption years. This could occur if subordinate individuals (mainly immatures) are forced into suboptimal habitats due to density-dependent effects in irruption years. To test whether irruptive individuals had lower survival than non-irruptive ones, we combined necropsy results (N = 365) with telemetry (N = 185) from more than 20 years to record timing and causes of mortality in snowy owls (Bubo scandiacus), which irrupt into eastern North America during winter following high breeding output caused by lemming peaks in the Arctic. Mortality was more than four times higher in irruption years than non-irruption years, but only for immatures, and occurred disproportionately in early winter for immatures, but not adults. Mortality was also higher in eastern North America, where owl abundance fluctuates considerably between years, compared to core winter regions of the Arctic and Prairies where populations are more stable. Most mortality was not due to starvation, but rather associated with human activity, especially vehicle collisions. We conclude that immature snowy owls that irrupt into eastern North America are limited by density-dependent factors, such as increased competition forcing individuals to occupy risky human-altered habitats. For highly mobile, irruptive animals, resource pulses may have a limited impact on population dynamics due to low subsequent survival of breeding output during the nonbreeding season. Annually, 13% of special educators leave teaching, twice that of general educators (Wong et al., 2017). In June 2017, the U.S. Department of Education and Office of Postsecondary Education announced that 46 states were drastically short of special education teachers (Robinson et al., 2019). This study explores how university special education (SPED) teacher preparation program descriptions entice students to enroll in university SPED program. Background: Unwanted intrusive thoughts of intentionally harming one's infant (intrusive harm thoughts) are common distressing experiences among postpartum mothers and fathers. Aim: To understand infant crying as a stimulus for intrusive harm thoughts and associated emotional responses in prepartum and postpartum mothers and fathers in response to infant cry. Method: Following completion of self-report measures of negative mood and anger, prepartum (n = 48) and postpartum (n = 44) samples of mother and father pairs completed 10 minutes of listening to audio-recorded infant crying. Post-test questionnaires assessed harm thoughts, negative emotions, urges to comfort and flee, and thoughts of shaking as a soothing or coping strategy. Results: One quarter of prepartum and 44% of postpartum parents reported intrusive infant-related harm thoughts following crying. Mothers and fathers did not differ in the likelihood of reporting harm thoughts, nor in the number of thoughts reported. Women reported more internalizing emotions compared with men. Hostile emotions were stronger among postpartum parents, and parents reporting harm thoughts. All parents reported strong urges to comfort the infant. Urges to flee were stronger among parents who reported harm thoughts. The likelihood of using infant shaking as a soothing or coping strategy was minimally endorsed, albeit more strongly by fathers and parents who also reported harm thoughts. Conclusions: In response to crying, harm thoughts are common and are associated with hostile emotions, urges to flee, and increased thoughts of using infant shaking. Highly efficient transmetalation between gold phosphites and iodopalladium species is presented. In addition to successful transfer of cyclic and acyclic phosphites, studies involving P‐chiral substrates revealed that an initial protodeauration as well as the target transmetalation were both stereospecific, and that the overall process occurred with retention of configuration at phosphorus. Building on the mechanistic work, a catalytic approach to the synthesis of chiral P‐arylated nucleoside derivatives was developed. Obesity is a growing epidemic, yet few patients with obesity receive a clinical diagnosis of obesity or appropriate counseling. We examined the socioeconomic and demographic factors associated with the accurate diagnosis of obesity during ambulatory care visits. We used data from the National Hospital Ambulatory and National Ambulatory Medical Care Surveys (NHAMCS and NAMCS) to determine if a patient with obesity had been clinically diagnosed with obesity during the visit by either of the following: (1) a diagnosis listed in the patient's record; or (2) the provider's answer to the question 'despite the diagnoses listed, does this patient have obesity?' We used multivariate models to examine the association between the accurate diagnosis of obesity and socioeconomic and demographic factors. We examined 885,291,770 weighted office visits involving individuals 5 years of age and older between 2006 and 2010. Providers were less likely to diagnose obesity at office visits involving children (5-12 years) with obesity (23.4 %) than at visits for adolescents (13-21 years; 39.7 %), young adults (22-34 years; 45.4 %), adults (35-64 years; 43.9 %) or elderly adults (≥65 years; 39.6 %; P < 0.001 for all). Individuals with obesity residing in more highly educated areas were more likely to be diagnosed than those living in less highly educated areas (44.2 vs. 40.9 %; AOR 1.4; 95 % CI 1.2-1.6). Males with obesity were less likely to be diagnosed than females with obesity (36.1 vs. 45.8 %; AOR 0.7; 95 % CI 0.6-0.8). After controlling for socioeconomic-status we did not find a consistent difference in the diagnosis of obesity by race. The diagnosis of obesity was made at less than half of all office visits involving patients with obesity. Recent evidence suggesting that adolescent suicidality has increased over the past decade has emphasized the importance of assessing contributory psychosocial risk factors. This present study has examined the association of family conflict and family cohesion with adolescent suicidality. Data was collected from a multisite sample of 195 adolescent primary care patients using the Family Environment Survey (FES) and the Scale for Suicidal Ideation (BSI). Results indicated that family cohesion was significantly and inversely associated with suicidality ( r = −.3904, p < .001) and that family conflict was significantly and positively associated with suicidality ( r = .3899, p < .001). All (100 %) of the adolescents with BSI scores indicative of elevated risk for suicidal behavior reported above-median levels of family conflict. Reports of suicidality were particularly elevated among adolescents who reported high family conflict and low family cohesion. The ability to synthesize and salvage purines is crucial for colonization by a variety of human bacterial pathogens. Helicobacter pylori colonizes the gastric epithelium of humans, yet its specific purine requirements are poorly understood, and the transport mechanisms underlying purine uptake remain unknown. Using a fully defined synthetic growth medium, we determined that H. pylori 26695 possesses a complete salvage pathway that allows for growth on any biological purine nucleobase or nucleoside with the exception of xanthosine. Doubling times in this medium varied between 7 and 14 hours depending on the purine source, with hypoxanthine, inosine and adenosine representing the purines utilized most efficiently for growth. The ability to grow on adenine or adenosine was studied using enzyme assays, revealing deamination of adenosine but not adenine by H. pylori 26695 cell lysates. Using mutant analysis we show that a strain lacking the gene encoding a NupC homolog (HP1180) was growth-retarded in a defined medium supplemented with certain purines. This strain was attenuated for uptake of radiolabeled adenosine, guanosine, and inosine, showing a role for this transporter in uptake of purine nucleosides. Deletion of the GMP biosynthesis gene guaA had no discernible effect on mouse stomach colonization, in contrast to findings in numerous bacterial pathogens. In this study we define a more comprehensive model for purine acquisition and salvage in H. pylori that includes purine uptake by a NupC homolog and catabolism of adenosine via adenosine deaminase. Although prior research has examined the consequences of childhood maltreatment, little research has focused specifically on the associations of childhood emotional abuse and cruel physical punishment with symptoms of generalized anxiety disorder and panic disorder during adolescence. This study was conducted to investigate these associations. Childhood emotional abuse, cruel physical punishment, and anxiety disorder symptoms were assessed in a multisite sample of 194 adolescent primary care patients. Results indicated that both childhood emotional abuse and cruel physical punishment were significantly correlated with elevated levels of generalized anxiety disorder and panic disorder symptoms. Findings were consistent with the hypothesis that childhood emotional abuse and cruel physical punishment may be correlated with elevated risk for adolescent anxiety symptoms. [ABSTRACT FROM AUTHOR]

Teachers have a central role in developing new learning models in schools. This paper describes a study that explored teachers' confidence and competence in using Information and Communication Technologies (ICT) as they participated in an ICT development project conducted by an Australian education system in 12 primary schools. The project aimed to develop ICT integrated teaching practices by providing in-class equipment and teacher professional development in ICT use, curriculum development and teaching strategies. The study used qualitative data from teacher records of Professional Development and Action Learning to identify three of four stages proposed to describe teachers' ICT learning. Teachers moved from gaining basic ICT skills, to conducting ICT-focused lessons and eventually appropriate ICT integration. Teachers who progressed to the third stage were able to exploit additional learning opportunities and begin to make fundamental changes to their pedagogy, but needed more time to reach the final stage of challenging existing pedagogical structures.
