Ungulate herbivory might induce different effects on the diversity and growth of trees and shrubs. The density, distribution, and the species of ungulates as well as plant communities' composition and other factors determine whether ungulate herbivory promotes or limits plants' diversity and growth. The impacts of ungulates on woody plants are commonly surveyed with exclosure-control approaches. In practice, such surveys frequently only cover short periods of time, addressing immediate management needs. Long-term surveys, documenting lasting effects of ungulate herbivory, are highly needed, but still rare. However, the general transferability of outcomes of long-term surveys might be limited due to different disturbing factors. This study addresses two basic aspects of long-term monitoring in mountainous forests, based on a unique 30-year data set: (1) Possible long-term effects of herbivores on forest vegetation (e.g., species/structural diversity of woody plants) and (2) potential differences between short-term and long-term surveys in terms of height growth patterns. In our study, diversity of woody plant species showed great variability with no significant impact of ungulate herbivory. The presence of ungulates had a significantly negative effect on the vertical structural diversity and growth of trees. Due to the slower growth on control plots, it took trees longer to reach a 160 cm height-threshold with their terminal shoots than on exclosure plots. Our long-term control-exclosure data set indicated that long-term survey data indeed might differ from growth patterns represented by short-term surveys. This can be induced by several factors, like site-specific growth patterns of trees, occurrences of natural abiotic disturbances that influence the functional life of exclosures, and others. Purpose The aim of this study was to study the impact of obesity on the secretory activity of the murine PVAT. Methods We exploited proteomics to profile the secretome of peri-aortic and canonical AT depots in wild-type (wt) and obese (ob/ob) mice. In parallel, fat tissues were processed for biochemical and histological analysis and mechanistical experiments were performed in vitro on primary neuronal cultures. Results Proteomics on ATs conditioned media from wt mice revealed that each fat depot displays a unique secretory profile. The enrichment of neuronal cell-adhesion molecules detected in PVAT secretomes reflected a higher content of intra-parenchymal sympathetic projections compared to non-perivascular ATs. A significant decrease of the same neuronal proteins in PVAT conditioned media from ob/ob mice was found to be associated with a substantial reduction of the perivascular sympathetic innervation. Intriguingly, a similar decrease of sympathetic markers was detected in the epicardial AT from obese patients. Mechanistically, the conditioned media from ob/ob AT explants was found to exert a deleterious effect on the axons of primary sympathetic neurons in vitro, indicating that this neuropathy is due to local alterations of the PVAT secretome that detrimentally impact on the embedded sympathetic neurites. Among proteins significantly down-regulated in the secretomes of ob/ob PVAT samples, neuronal growth regulator 1 (Negr1) was found to promote axonal elongation and branching on sympathetic neurons in vitro. Administration of recombinant Negr1 also partially restored the neurotrophic effect of ob/ob AT secretomes on sympathetic axons both in vitro and in vivo. Conclusions Obesity-related alterations in the secretome of PVAT severely affect the homeostasis of the perivascular environment, leading to a loss of perivascular sympathetic innervation. Recent task-switching work in which paper-and-pencil administered single-task lists were compared with task-alternation lists has demonstrated large increases in task-switch costs with concurrent articulatory suppression (AS), implicating a crucial role for verbalization during switching (Baddeley, Chincotta, & Adlam, 2001; Emerson & Miyake, 2003). Experiment 1 replicated this result, using computerized assessment, albeit with much smaller effect sizes than in the original reports. In Experiment 2, AS interference was reduced when a sequential cue (spatial location) that indicated the current position in the sequence of task alternations was given. Finally, in Experiment 3, switch trials and no-switch trials were compared within a block of alternating runs of two tasks. Again, AS interference was obtained mainly when the endogenous sequencing demand was high, and it was comparable for no-switch and switch trials. These results suggest that verbalization may be critical for endogenous maintenance and updating of a sequential plan, rather than exclusively for the actual switching process. The Sverdrup Basin of the Canadian Arctic Islands contains a sedimentary record, with only short breaks, from Early Carboniferous to Late Cretaceous time and can be used to document the nature of sediments delivered from northern Canada and Greenland. Sm-Nd isotopic analysis of 72 sedimentary rock samples from the Sverdrup Basin, coupled with trace element characterization, shows that for most of Carboniferous to Late Cretaceous time, the sediment supply in the northern part of North America was dominated by a single broad provenance; 56 of the 72 samples lie squarely within the Nd isotopic evolution of a clastic sedimentary cover delivered to the region following 450--350 Ma Caledonian and Franklinian mountain building in Greenland and the Canadian Arctic Islands. Cratonic Shield sources in Greenland and Canada are hardly evident in the record, and significant contributions to the sediment budget from any source other than the post-mid-Paleozoic orogenic cover occurred only during four relatively short periods. First, during Carboniferous time, pre--Late Ordovician rocks of the Franklinian orogen contributed to alluvial clastic rocks in small rift basins in northern Ellesmere Island. Second, during Early Cretaceous time, Shield basement contributed to more widespread deltaic deposits in central and southern Ellesmere Island. Third, minor volcanic contributions to much of the basin occurred during Late Triassic-- materials from Caledonian and Franklinian mountains dominated the provenance of the continental and continent-margin sedimentary system for at least 370 m.yr., a period of time extending far beyond the existence of the mountains themselves. This dominance was achieved by recycling of widespread Middle and Upper Devonian strata into Mesozoic units in the Canadian Arctic and Cordillera. In two experiments, young and older adults solved arithmetic chain tasks with single-digit operands, with or without a concurrent memory load of three or six digits. Variables in the arithmetic tasks had to be replaced by digits from the screen or from the memory set. A task-irrelevant concurrent load impaired neither speed nor accuracy of arithmetic in younger adults. In Experiment 2, this was also true for older adults. A large decrease in arithmetic performance was observed, however, when variables in the arithmetic task had to be substituted by digits from the memory list. Older adults had specific problems with this condition in Experiment 1, where the substitution involved two successive steps, but not in Experiment 2, where the substitution from memory could be done in a single step. The results are difficult to reconcile with models assuming a common resource for storage and processing. Rather, they are compatible with the hypothesis that a concurrent memory load interferes with a processing task only during the points of access to working memory. Further, even though access to working memory was found to be the critical source of concurrent-load interference, it was found to be insensitive to the effects of adult aging. The stability, pH-dependence and kinetic properties of the Mn and FDP-activated NAD-dependent lactic acid dehydrogenases from Lactobacillus casei ssp. casei (ATCC 393) and L. curvatus (DSM) 20010) were studied after the enzymes were purified to homogeneity by affinity chromatography. Both enzymes are virtually unidirectional, catalysing efficiently only the reduction of pyruvate. They are similar with respect to the effector requirement and pH-optimum. They differ, however, in their electrophoretic mobility, heat stability, pH-dependence of the Mn requirement and several kinetic properties. It is suggested that most of these differences are caused by differences of the negative charges in the vicinity of the FDP-binding site or the site responsible for the interaction of the subunits of the enzymatically active oligomeres. Forty-four Parkinson patients (19 patients of the rigid-akinetic type, 13, of the rigid-akinetic-tremor type, and 12, of the tremor type) were included in a study in order to analyse correlations of the expression of the motor symptoms tremor, rigidity, akinesia, with other clinical parameters, computertomographic aspect of brain atrophy and psychometrically assessed cognitive parameters. Rigidity and akinesia are significantly positively correlated with the severity of motor dysability, stage of the disease, and brain atrophy, as is akinesia with a history of pharmacotoxic psychosis. Tremor is significantly negatively correlated with motor dysability, stage of the disease, and history of pharmacotoxic psychosis. Akinesia is correlated with visuomotor dysfunction (tested with Bender Gestalt Test) and rigidity with the depression score (Zung scale). Four patients meeting the clinical criteria of the rigid spine syndrome are presented; they are one girl with a positive family history and three boys. Clinical and histological findings are discussed in relation to the 14 cases of rigid spine syndrome reported in the literature. The delineations of the syndrome from other benign myopathies with early contractures are discussed suggesting that the rigid spine syndrome probably does not represent a single nosological entity. Rationale Myofilament proteins are responsible for cardiac contraction. The myofilament subproteome, however, has not been comprehensively analysed thus far. Methods Cardiomyocytes were isolated from rodent hearts and stimulated with endothelin-1 and isoproterenol, potent inducers of myofilament protein phosphorylation. Subsequently, cardiomyocytes were 'skinned-tm) and the myofilament subproteome analysed using a high mass accuracy ion trap tandem mass spectrometer (LTQ Orbitrap XL) equipped with electron transfer dissociation. Results As expected, a small number of myofilament proteins constituted the majority of the total protein mass, with several known phosphorylation sites being confirmed by electron transfer dissociation. More than 600 additional proteins were identified in the cardiac myofilament subproteome, including kinases and phosphatase subunits. The proteomic comparison of myofilaments from control and treated cardiomyocytes suggested that isoproterenol treatment altered the subcellular localisation of protein phosphatase 2A regulatory subunit B56α. Immunoblot analysis of myocyte fractions confirmed that β-adrenergic stimulation by isoproterenol decreased the B56α content of the myofilament fraction in the absence of significant changes in the myosin phosphatase target subunit isoforms 1 and 2 (MYPT1 and MYPT2). Furthermore, immunolabelling and confocal microscopy revealed the spatial redistribution of these proteins, with a loss of B56α from Z-disc and M-band regions but increased association of MYPT1/2 with A-band regions of the sarcomere, following β-adrenergic stimulation. Conclusion We present the first comprehensive proteomic dataset of skinned cardiomyocytes and demonstrate the potential of proteomics to unravel dynamic changes in protein composition that may contribute to the neurohormonal regulation of myofilament contraction. Purpose: With regards to biomarker discovery, proteomics has overpromised, but underdelivered. The cardiovascular field is no exception. The plasma proteome is the most complex proteome of the human body. An alternative strategy is the application of proteomics to diseased tissues, where the potential biomarkers are less dilute.Methods and Results: We have applied our proteomics method for cardiac extracellular proteins to biomarker discovery in septic hearts. C57Bl6 mice were injected with LPS, their cardiac proteome was subfractionated and extracellular proteins were analysed by liquid chromatography tandem mass spectrometry. Among the most differentially expressed proteins was long pentraxin 3 (PTX3), an acute phase protein, secreted by various cells and tissues, that is known to be associated with increased mortality in septic patients. Interestingly, PTX3 accumulated in the cardiac extracellular matrix as an octamer due to disulphide-bond formation. An octameric and tetrameric moiety of PTX3 was also detectable in plasma. Over a time course of 11 days, the redox-state of PTX3 was quantified in septic patients (n=31). On admission to the intensive care unit, there was no difference in the redox state of PTX3 between survivors and non-survivors. From day 2 onwards, however, the conversion of octameric to monomeric PTX3 was consistently associated with a greater survival after 28 days of follow-up. For example, on day 2 post admission, octameric PTX3 was undetectable in survivors, but still constituted more than half of total PTX3 in non-survivors (p<0.001). Although monomeric PTX3 was inversely associated with cardiac damage markers such as NT-proBNP, high sensitive troponin I and T, octamer- and monomer-PTX3 levels predicted the 28-day survival better than a model containing NT-proBNP only.Conclusion: Sepsis is associated with significant mortality. In comparison to the conventional measurements of total PTX3, the assessment of the redox-sensitive oligomerization of PTX3 was more dynamic and could be a superior predictor of inflammation resolution and disease outcome in septic patients.
