Parkinsonism is a clinical syndrome characterized by akinesia/bradykinesia, muscle rigidity, resting tremor, and postural instability. Within the group of parkinsonisms is Parkinson's disease, also known as neurodegenerative parkinsonian syndrome. The group of atypical parkinsonisms was established due to the existence of sporadic parkinsonisms that do not share the exact etiology of Parkinson's disease. Additionally, parkinsonisms that arise from causes other than neurodegeneration have been classified as secondary parkinsonisms. With this in mind, given the diversity of etiologies that can trigger parkinsonism, it is crucial to understand the symptomatology and its relationship with the basal ganglia (including damage to the nigrostriatal pathway, neuroinflammation, and neuronal damage). Simple Summary: In male rats, the major pelvic ganglion is the principal component of the pelvic plexus and plays a crucial role in regulating various physiological functions such as urination, defecation, erection, ejaculation, and glandular secretion. This ganglion is considered mixed, as it receives both sympathetic and parasympathetic innervation through the hypogastric and pelvic nerves, respectively. Homologous structures with similar functions are present in other species, including cats, dogs, and pigs; however, differences exist in nomenclature, anatomical complexity, and functionality. Although anatomical, histological, and immunohistochemical studies have been conducted on these structures across various species, the major pelvic ganglion of the rat has been the most extensively studied due to its ease of identification and manipulation. This review provides a comprehensive analysis of the pelvic plexus and its regulation across various mammalian species, including rats, cats, dogs, and pigs. The pelvic and hypogastric nerves play crucial roles in regulating pelvic functions such as micturition, defecation, and erection. The anatomical organization of these nerves varies, forming either well-defined ganglia or complex plexuses. Despite these variations, the neurons within these structures are consistently regulated by key neurotransmitters, norepinephrine and acetylcholine. These neurons also possess receptors for testosterone and prolactin, particularly in rats, indicating the significant role of these hormones in neuronal function and development. Moreover, neuropeptides such as vasoactive intestinal peptide (VIP), substance P, neuropeptide Y (NPY), somatostatin (SOM), galanin (GAL), and calcitonin gene-related peptide (CGRP) are co-released with neurotransmitters to modulate pelvic functions. This review highlights the complex interplay between neurotransmitters, neuropeptides, and hormones in regulating pelvic physiology and emphasizes the importance of hormonal regulation in maintaining the functionality and health of the pelvic plexus across different species. Parkinson's disease is currently a global public health challenge due to the rapid growth of aging populations. To understand its pathophysiology is necessary to study the functional correlation between the basal ganglia (BG) and the cerebellum, which are involved in motor control. Herein, we explored multiunit electrical activity (MUA) in the cerebellum of rats with induced Parkinsonism as a result of lesions following bilateral placement of electrodes and passing of current in the ventrolateral striatum (VLS). In one control group, the electrodes descended without electrical current, and another group was left intact in VLS. MUA was recorded in Sim B and Crus II lobes, and in the dentate nucleus (DN) during the execution of exploration behaviors (horizontal and vertical) and grooming. The lesioned and sham groups showed a decrease in MUA amplitude in the Crus II lobe compared to the intact group in all recorded behaviors. However, Sim B and DN did not express differences. Both electrical and physical insults to the VLS induced Parkinsonism, which results in less MUA in Crus II during the execution of motor behaviors. Autism is a complex alteration in children's developing nervous system that manifests in behavioral patterns that do not match those of typical subjects. Moreover, starting at puberty, these children may encounter problems regarding social interaction related to sexual encounters. As studies are scarce, we used the valproate model of autism in the zebrafish to contribute to the knowledge related to sexual behavior in this disorder. Young zebrafish were reproduced, embryos collected, and organized in groups of control or treated with valproic acid, as the autism model. Sexual behavior was recorded in fish from these embryos as they became sexually active. The results show that the zebrafish's sexual behavior is organized into appetitive, preconsummatory, and consummatory behaviors, as in other vertebrates. In the autism model, the patterns of sexual behavior are present but with significant modifications. The behavior of males was the most affected, while in females, the most affected parameter was oviposition. These results show that, in the autism model of zebrafish, sexual behavior is not suppressed, but it seems that critical changes occur in the neuroendocrine system that reduces reproductive success. Furthermore, the enriched environment was beneficial in maintaining the consummatory behaviors of females and males with autism-like behaviors. [ABSTRACT FROM AUTHOR]

Simple Summary: Maternal behavior involves active and passive responses associated with the willingness to nurse and protect the young. In some species, its expression is very selective toward individuals that are recognized as their own and may be long-lasting, whereas in other species the expression is not as selective or may be short-lasting. Brain processes of acceptance, social recognition, inhibition of rejection/fear, and increase in care motivation mediate its expression. The neurocircuitry of maternal behavior is activated upon exposure to the right natural stimuli, such as those that occur during pregnancy, parturition, and lactation. However, even virgin females and males can respond with maternal behaviors if they develop sensitization to the offspring via cohabitation or cross-sensitization via mating. Herein, we discuss behavioral expression in different species, the natural triggering stimuli, and the putative neurocircuitries of acceptance, social recognition, motivation, and rejection during maternal behavior. Among the different species of mammals, the expression of maternal behavior varies considerably, although the end points of nurturance and protection are the same. Females may display passive or active responses of acceptance, recognition, rejection/fear, or motivation to care for the offspring. Each type of response may indicate different levels of neural activation. Different natural stimuli can trigger the expression of maternal and paternal behavior in both pregnant or virgin females and males, such as hormone priming during pregnancy, vagino-cervical stimulation during parturition, mating, exposure to pups, previous experience, or environmental enrichment. Herein, we discuss how the olfactory pathways and the interconnections of the medial preoptic area (mPOA) with structures such as nucleus accumbens, ventral tegmental area, amygdala, and bed nucleus of stria terminalis mediate maternal behavior. We also discuss how the triggering stimuli activate oxytocin, vasopressin, dopamine, galanin, and opioids in neurocircuitries that mediate acceptance, recognition, maternal motivation, and rejection/fear. Brain mechanisms of sexual attraction toward reproductive partners develop from a systematic interrelationship between biology (nature) and learning (nurture). However, the causes of attraction toward non-reproductive partners are poorly understood. Here, we explored the role of Pavlovian learning under dopaminergic agonism on the development of sexual preference and brain activation for young male rats. During conditioning, adult sexually naïve males received either Saline (Saline-Paired) or the D2-receptor agonist quinpirole (QNP-Paired) and cohabited in contingency, or out of contingency (QNP-Unpaired) during 24 h with an almond-scented prepubertal juvenile male (PD25). Conditioning occurred every 4 days for three trials. Social and sexual responses were assessed four days after the last conditioning trial in a drug-free test, and males chose freely between a scented young male (PD37) and a novel receptive female. Four days later, males were exposed to the conditioned odor only and brain Fos-IR and serum testosterone were analyzed. Saline-Paired and QNP-Unpaired males displayed more non-contact erections (NCEs) and genital investigations for females, whereas QNP-Paired males expressed more NCEs and genital investigations for young males. In the QNP-Paired group, exposure to the young male-paired odor evoked more Fos-IR in limbic, hypothalamic and cortical areas, but no differences in serum testosterone were observed. Cohabitation with juvenile males during enhanced D2 agonism results in atypical appetitive sexual responses and a higher pattern of brain response for the young male-paired odor, with no changes in serum testosterone. Purpose of Review: The aim of this review is to provide current evidence on the biological and psychological mechanisms that underlie sexual partner preferences (SPP) in humans and animals.Recent Findings: SPP depend mainly on prenatal (adaptive) organization of the brain, but can be drastically modified via learning under enhanced dopaminergic (DA) and oxytocinergic (OT) activity.Summary: SPP can be categorized as in those directed towards partners who display indicators of biological fitness (IBF) or towards partners who do not show those indicators. The IBF function as unconditioned stimuli that presumably activate prenatally organized brain areas that mediate the salience of those stimuli. However, we discuss some evidence indicating that SPP not directed towards IBF (i.e., paraphilias) might be consequence of a learning process that occurs under enhanced DA or OT activity, resulting in new powerful learning with additional brain areas involved. Because of its primary role in drug-seeking, consumption and addictive behaviour, there is a growing interest in identifying the neural circuits and molecular mechanisms underlying the formation, maintenance and retrieval of drug-related memories. Human studies, which focused on neuronal systems that store and control drug-conditioned memories, have found cerebellar activations during the retrieval of drug-associated cue memory. However, at the pre-clinical level, almost no attention has been paid to a possible role of the cerebellum in drug-related memories. In the present study, we ought to fill this gap by aiming to investigate the pattern of neuronal activation (as revealed by cFos expression) in different regions of the prefrontal cortex and cerebellum of mice trained to develop conditioned preference for an olfactory stimulus (CS+) paired with cocaine. Our results indicate that CS+ preference was directly associated with cFos expression in cells at the apical region of the granule cell layer of the cerebellar vermis; this relationship being more prominent in some specific lobules. Conversely, cFos+ immunostaining in other cerebellar regions seems to be unrelated to CS+ preference but to other aspects of the conditioning procedure. At the prefrontal cortex, cFos expression seemed to be related to cocaine administration rather than to its ability to establish conditioned preference. The present results suggest that as it has been observed in some clinical studies, the cerebellum might be an important and largely overlooked part of the neural circuits involved in generating, maintaining and/or retrieving drug memories. The sexual behavior of male rats constitutes a natural model to study learning of motor skills at the level of the central nervous system. We previously showed that sexual behavior increases Fos expression in granule cells at lobules 6 to 9 of the vermis cerebellum. Herein, we obtained multiunit recordings of lobules 6a and 7 during the training period of naive subjects, and during consecutive ejaculations of expert males. Recordings from both lobules and the inferior olive showed that the maximum amplitude of mount, intromission, and ejaculation signals were similar, but sexual behavior during training tests produced a decrease in the amplitude for mount and intromission signals. The fastigial nucleus showed an inverse mirror-like response. Thus, the cerebellum is involved in the neural basis of sexual behavior and the learning of appropriate behavioral displays during copulation, with a wiring that involves the cerebellar cortex, inferior olive, and fastigial nucleus. Background: The prostate is a sexual gland that produces important substances for the potency of sperm to fertilize eggs within the female reproductive tract, and is under complex endocrine control. Taking advantage of the peculiar behavioral pattern of copulating male rats, we developed experimental paradigms to determine the influence of sexual behavior on the level of serum testosterone, prostate androgen receptors, and mRNA for androgen receptors in male rats displaying up to four consecutive ejaculations. Methods: The effect of four consecutive ejaculations was investigated by determining levels of (i) testosterone in serum by solid phase RIA, (ii) androgen receptors at the ventral prostate with Western Blots, and (iii) androgen receptors-mRNA with RT-PCR. Data were analyzed with a one-way ANOVA followed by a post hoc application of Dunnett's test if required. Results: The constant execution of sexual behavior did not produce any change in the weight of the ventral prostate. Serum testosterone increased after the second ejaculation, and remained elevated even after four ejaculations. The androgen receptor at the ventral prostate was higher after the first to third ejaculations, but returned suddenly to baseline levels after the fourth ejaculation. The level of mRNA increased after the first ejaculation, continued to increase after the second, and reached the highest peak after the third ejaculation; however, it returned suddenly to baseline levels after the fourth ejaculation. Conclusion: Four consecutive ejaculations by sexually experienced male rats had important effects on the physiological responses of the ventral prostate. Fast responses were induced as a result of sexual behavior that involved an increase and decrease in androgen receptors after one and four ejaculations, respectively. However, a progressive response was observed in the elevation of mRNA for androgen receptors, which also showed a fast decrease after four ejaculations. All of these changes with the prostate gland occurred in the presence of a sustained elevation of testosterone in the serum that started after two ejaculations. A consideration of these fast-induced changes suggests that the nerve supply plays a key role in prostate physiology during the sexual behavior of male rats. [ABSTRACT FROM AUTHOR]

Background: The prostate is a key gland in the sexual physiology of male mammals. Its sensitivity to steroid hormones is widely known, but its response to prolactin is still poorly known. Previous studies have shown a correlation between sexual behaviour, prolactin release and prostate physiology. Thus, here we used the sexual behaviour of male rats as a model for studying this correlation. Hence, we developed experimental paradigms to determine the influence of prolactin on sexual behaviour and prostate organization of male rats. Methods: In addition to sexual behaviour recordings, we developed the ELISA procedure to quantify the serum level of prolactin, and the hematoxilin-eosin technique for analysis of the histological organization of the prostate. Also, different experimental manipulations were carried out; they included pituitary grafts, and haloperidol and ovine prolactin treatments. Data were analyzed with a One way ANOVA followed by post hoc Dunnet test if required. Results: Data showed that male prolactin has a basal level with two peaks at the light-dark-light transitions. Consecutive ejaculations increased serum prolactin after the first ejaculation, which reached the highest level after the second, and started to decrease after the third ejaculation. These normal levels of prolactin did not induce any change at the prostate tissue. However, treatments for constant elevations of serum prolactin decreased sexual potency and increased the weight of the gland, the alveoli area and the epithelial cell height. Treatments for transient elevation of serum prolactin did not affect the sexual behaviour of males, but triggered these significant effects mainly at the ventral prostate. Conclusion: The prostate is a sexual gland that responds to prolactin. Mating-induced prolactin release is required during sexual encounters to activate the epithelial cells in the gland. Here we saw a precise mechanism controlling the release of prolactin during ejaculations that avoid the detrimental effects produced by constant levels. However, we showed that minor elevations of prolactin which do not affect the sexual behaviour of males, produced significant changes at the prostate epithelium that could account for triggering the development of hyperplasia or cancer. Thus, it is suggested that minute elevations of serum prolactin in healthy subjects are at the etiology of prostate abnormal growth. [ABSTRACT FROM AUTHOR]

Background: Messenger RNA (mRNA) comprises three major parts: a 5'-UTR (UnTranslated Region), a coding region, and a 3'-UTR. The 3'-UTR contains signal sequences involved in polyadenylation, degradation and localization/stabilization processes. Some sequences in the 3'-UTR are involved in the localization of mRNAs in (e.g.) neurons, epithelial cells, oocytes and early embryos, but such localization has been most thoroughly studied in neurons. Neuronal polarity is maintained by the microtubules (MTs) found along both dendrites and axon and is partially influenced by sub-cellular mRNA localization. A widely studied mRNA is that for Tau protein, which is located in the axon hillock and growth cone; its localization depends on the wellcharacterized cis-acting signal (U-rich region) in the 3'-UTR. Methods: We compared the cis-acting signal of Tau with mRNAs in the axonal regions of neurons using the ClustalW program for alignment of sequences and the Mfold program for analysis of secondary structures. Results: We found that at least 3 out of 12 mRNA analyzed (GRP75, cofilin and synuclein) have a sequence similar to the cis-acting signal of Tau in the 3'-UTR. This could indicate that these messengers are localized specifically in the axon. The Mfold program showed that these mRNAs have a similar "bubble" structure in the putative sequence signal. Conclusion: Hence, we suggest that a U-rich sequence in the 3'-UTR region of the mRNA could act as a signal for its localization in the axon in neuronal cells. Sequences homologous to the DTE sequence of BC1 mRNA could direct the messenger to the dendrites. Messengers with homologues of both types of sequence, e.g. β-actin, might be located in both dendrites and axon. [ABSTRACT FROM AUTHOR]

Abstract The scrotal nerves in the rat were studied electrophysiologically to describe their topographic organization in the skin and in the dorsal roots, and to determine if the electrical properties of these afferents are under the influence of testosterone. There are 2 afferent nerve branches innervating this area, the proximal scrotal branch supplying the proximal half of the scrotum (PSb), and the distal scrotal branch supplying the distal half (DSb). Results showed that afferent axons from PSb enter the spinal cord through the L5 and L6 dorsal roots, and axons from DSb enter the cord through the L6 and S1 dorsal roots. Mechanical stimulation of the scrotal skin exposed 3 dermatome areas, the L5, L6, and S1, with L6 being the longest dermatome with intermingled terminals from PSb and DSb. Decreased levels of testosterone after castration increased the threshold to trigger the activity of scrotal afferents, and the exogenous administration of the hormone reverted it. This effect was similar in the dorsal penile nerve. The decrease in androgen levels produced a reduction in the skin sensory field to mechanical stimuli that was restored after the administration of the hormone. As the scrotal nerves are highly stimulated during copulation in parallel with the dorsal penile nerve, it is suggested that the sensory field of the scrotal skin could be closely related to the modifications in sexual behavior after testosterone manipulation. European rabbits ( Oryctolagus cuniculus) of both sexes possess a variety of skin glands important in chemical communication. These include sizable inguinal glands, the strong-smelling secretion from which accumulates in conspicuous pouches of skin on either side of the genital opening. To test the hypothesis that this secretion advertises sexual receptivity in females, inguinal gland secretions from nine intact and six ovariectomized females were presented on dummy rabbits or in a simultaneous choice paradigm, and the responses of nine stud males to these secretions were recorded. Although the males clearly perceived the stimuli, no differences in responses to secretions from the intact and ovariectomized does were observed, and no differences were found in the size, weight, or secretory activity of the does' glands. It is concluded that inguinal gland secretion plays little role in communicating sexual receptivity in female rabbits and that ovarian steroids play little part in regulating inguinal gland activity.
