Résumé: À ce jour, seules quelques études épidémiologiques ont tenu compte de l'exposition prénatale et de la petite enfance à des mélanges de polluants chimiques pour évaluer les associations avec les perturbations et les troubles du neurodéveloppement de l'enfant. Les substances chimiques au sein d'un mélange peuvent avoir des effets additifs, synergiques ou antagonistes. Des études récentes de cohortes mère-enfant ont appliqué des modèles d'analyse multi-polluants pour étudier les effets des mélanges. Elles ont examiné l'impact sur le neurodéveloppement (cognitif, comportemental) du mélange, identifié les substances chimiques de ce mélange qui jouent un rôle clé dans la neurotoxicité (métaux, bisphénols, certains métabolites de phtalates, etc.) et montré des effets interactifs. D'autres domaines de l'exposome (nutriments, mode de vie, facteurs sociaux) peuvent également être impliqués dans l'association avec les effets neurodéveloppementaux. Les études toxicologiques ont montré que les polluants chimiques partagent des modes et des mécanismes d'action communs, le stress oxydant étant souvent décrit comme l'un des événements clés conduisant à des perturbations cellulaires et tissulaires à différents niveaux du cerveau pendant les périodes critiques de vulnérabilité. Les modifications épigénétiques associées à certains polluants (phtalates, bisphénols, pesticides, particules fines, etc.) altèrent l'expression et la régulation de gènes importants pour le neurodéveloppement et contribuent au risque de troubles neurodéveloppementaux. Les progrès dans la connaissance des marqueurs épigénétiques devraient favoriser une approche préventive. To date, only a few epidemiological studies have considered prenatal and early childhood exposure to mixtures of chemical pollutants to assess their associations with neurodevelopmental disabilities in children. Chemicals in a mixture may have additive, synergistic or antagonistic effects. Some recent birth cohort studies investigating chemicals and neurodevelopmental effects have applied multipollutant models to the investigation of mixtures. They have examined the overall impact of mixtures, identified the chemicals from mixtures that play a key role in neurotoxicity (metals, bisphenols, phthalates, etc.) and investigated their interactive effects. Other domains of the exposome (nutrients, lifestyle, and social factors) may be also implicated in the association with neurodevelopmental effects. Toxicological studies have shown that chemical pollutants share common modes and mechanisms of action, with oxidative stress often described as one of the key events leading to cellular and tissue disruptions at different levels of the brain during critical periods of vulnerability. Epigenetic modifications associated with a number of pollutants (phthalates, bisphenols, pesticides, fine particles, etc.) alter the expression and regulation of genes important for neurodevelopment and contribute to the risk of neurodevelopmental disorders. Résumé: L'exposition aux pesticides, qu'elle soit professionnelle ou environnementale, pose un problème de santé publique. De nombreuses études épidémiologiques montrent que l'exposition professionnelle aux pesticides est corrélée positivement à un risque accru de développer certaines pathologies. Les études épidémiologiques réalisées chez les consommateurs d'aliments issus de l'agriculture biologique, dont les teneurs en pesticides sont sensiblement réduites, montrent une relation inverse avec l'incidence des maladies métaboliques et les taux de certains cancers. Les études expérimentales apportent des arguments quant à la plausibilité du lien pesticide-santé. Les pesticides sont des composés biologiquement actifs capables individuellement d'exercer des effets spécifiques sur différentes cibles ayant un rôle primordial dans le fonctionnement cellulaire et par extension dans les grandes fonctions physiologiques de l'organisme. Le consommateur est exposé à de faibles doses de pesticides mais à un mélange de ces composés. Dans un mélange, les pesticides peuvent interagir entre eux et induire des effets synergiques. La base des interactions entre pesticides est un phénomène complexe impliquant un réseau de cibles et de mécanismes à différents niveaux, à l'échelle de la cellule et de l'organisme. Ainsi, la prédiction des effets cocktails est aujourd'hui difficile à réaliser, compte tenu de la diversité des effets des pesticides, des niveaux d'interactions et des capacités d'adaptation et de réponses biologiques de l'organisme. Dans une étude publiée récemment nous montrons qu'un cocktail de six pesticides présents dans l'alimentation induit, chez la souris exposée pendant un an à des doses non toxiques, des perturbations métaboliques (diabète et surpoids) uniquement chez les mâles. Chez les femelles exposées, on observe un impact sur le statut oxydoréducteur hépatique et sur le microbiote. Ainsi, les pesticides à des doses considérées comme non toxiques peuvent avoir un impact sur la physiologie de l'organisme lors d'une exposition chronique de ces composés en mélange. Occupational or environmental exposure to pesticides is a public health concern. Numerous epidemiologic studies show that occupational exposure to these substances is positively correlated with an increased risk of developing certain diseases. Epidemiologic studies among consumers of organic food products (which have greatly reduced pesticide levels) show an inverse relation with the rates of some cancers and metabolic diseases. Experimental studies provide arguments of a plausible link between pesticides and health. Pesticides are biologically active compounds that can individually exert specific effects on different targets that play a major role in cellular function and by extension in the physiological functions of the body. Consumers are exposed to low doses of mixtures of pesticides. When combined, they can interact with one another and induce synergistic effects. Their interactions are complex phenomena involving a network of targets and mechanisms at both the cell and organ levels. It is thus difficult to predict the effects of any given pesticide cocktail, given the diversity of their effects and interaction levels and of the organism's adaptability and biological responses. In a recent study, we showed that a cocktail of six pesticides, present in the diet of mice exposed for one year at non-toxic doses, induced metabolic disorders (diabetes and overweight) only in males. In exposed females, there was an impact on hepatic redox status and microbiota activity. Thus, chronic exposure to a combination of pesticides at doses considered non-toxic can affect human physiology. [ABSTRACT FROM AUTHOR]

The cleavage of seven RNA model 2-hydroxypropyl aryl phosphates (1) catalyzed by a dinuclear Zn(II) complex of bis[1,4-N1,N1′(1,5,9-triazacyclododecanyl)]butane (4) was studied in methanol and ethanol at 25 °C under pH controlled conditions. The results are compared with what was reported earlier for the dinuclear Zn(II) complex of the lower homologue bis[1,3-N1,N1′(1,5,9-triazacyclododecanyl)]propane (3). In methanol, the higher homologue exhibits saturation binding with substrates having poor aryloxy leaving groups. With good leaving groups there is an observed linear dependence of kobs versus complex concentration without saturation binding over the catalyst concentration range investigated. In ethanol, strong saturation binding between the active form of the catalyst ((RO–):Zn(II)2:4) and all substrates is observed, the results observed in both solvents being similar to what was reported for the lower (RO–):Zn(II)2:3 homologue. Energetics calculations are presented for the (RO–):Zn(II)2:4-catalyzed cleavage of each substrate in both solvents to assess the catalytic efficiency via the ΔΔG‡ for catalyst binding a transition state comprising [RO–:1]‡ or its kinetic equivalent. Opérant dans des conditions de pH contrôlé, à 25 C, en solution dans le méthanol et l’éthanol, on a étudié la réaction de clivage de sept phosphates d’aryle et de 2-hydroxypropyle (1), des modèles de l’ARN, catalysé par un complexe dinucléaire du Zn(II) et le bis[1,4-N1,N1′(1,5,9-triazacyclododécanyl)]butane (4). On compare les résultats avec ceux rapportés antérieurement pour le complexe dinucléaire du Zn(II) l’homologue inférieur bis[1,3-N1,N1′(1,5,9-triazacyclodécanyl)]propane (3). Dans le méthanol, l’homologue supérieur on observe qu’il y a saturation du site de fixation avec les substrats pour lesquels l’habilité à partir des groupes aryloxy est faible. Avec de bons groupes partant, pour toute la plage de concentration de catalyseur utilisée dans ce travail, on observe une dépendance linéaire de kobs en fonction de la concentration du complexe. Dans l’éthanol, on observe une forte saturation des sites de fixation entre tous les substrats et la forme active du catalyseur [(RO–):Zn(II)2:4]; dans les deux solvants, les résultats sont similaires à ceux rapportés pour l’homologue inférieur [(RO–):Zn(II)2:3]. Afin d’évaluer l’efficacité catalytique par le biais du ΔΔG‡ de la fixation par le catalyseur d’un état d’activation comportant [RO–:1]‡ ou son équivalent cinétique, on a effectué des calculs d’énergie pour le clivage de chacun des substrats, catalysés par [(RO–):Zn(II)2:4].
