Strong coupling between a single quantum emitter and an optical cavity (at rate Ω) accesses fundamental quantum optics and provides an essential building block for photonic quantum technologies. However, the minimum mode volume of conventional dielectric cavities restricts their operation to cryogenic temperature for strong coupling. Here we harness surface self-assembly to make deterministic strong coupling at room temperature using CdSe/CdS quantum dots (QDs) in nanoparticle-on-mirror (NPoM) plasmonic nanocavities. We achieve a fabrication yield of ~70% for single QD strong coupling by optimizing their size and nano-assembly. A clear and reliable Rabi splitting is observed both in the scattering of each nanocavity and their photoluminescence, which are however not equal. Integrating these quantum elements with electrical pumping allows demonstration of strong coupling in their electroluminescence. This advance provides a straightforward way to achieve practical quantum devices at room temperature, and opens up exploration of their nonlinear, electrical, and quantum correlation properties. Due to the severity and high prevalence of cancer, as well as its complex pathological condition, new strategies for cancer treatment and diagnostics are required. As such, it is important to design a toolbox that integrates multiple functions on a single smart platform. Theranostic hydrogels offer an innovative and personalized method to tackle cancer while also considering patient comfort, thereby facilitating future implementation and translation to the clinic. In terms of theranostic systems used in cancer therapy, nanoparticles are widely used as diagnostic and therapeutic tools. Nanoparticles can achieve systemic circulation, evade host defenses, and deliver drugs and signaling agents at the targeted site, to diagnose and treat the disease at a cellular and molecular level. In this context, hydrogel microneedles have a high potential for multifunctional operation in medical devices, while avoiding the complications associated with the systemic delivery of therapeutics. Compared with oral administration and subcutaneous injection, microneedles offer advantages such as better patient compliance, faster onset of action, and improved permeability and efficacy. In addition, they comprise highly biocompatible polymers with excellent degradability and tunable properties. Nanoparticles and microneedles thus offer the possibility to expand the theranostic potential through combined synergistic use of their respective features. We review herein recent advances concerning processing methods and material requirements within the realm of hydrogel microneedles as theranostic platforms, various approaches toward cancer therapy, and the incorporation of nanoparticles for added functionality. Electron tomography has become a commonly used tool to investigate the three-dimensional (3D) structure of nanomaterials, including colloidal nanoparticle assemblies. However, electron microscopy is typically done under high-vacuum conditions, requiring sample preparation for assemblies obtained by wet colloid chemistry methods. This involves solvent evaporation and deposition on a solid support, which consistently alters the nanoparticle organization. Here, we suggest using electron tomography to study nanoparticle assemblies in their original colloidal liquid environment. To address the challenges related to electron tomography in liquid, we devise a method that combines fast data acquisition in a commercial liquid-cell with a dedicated alignment and reconstruction workflow. We present the advantages of this methodology in accurately characterizing two different systems. 3D reconstructions of assemblies comprising polystyrene-capped Au nanoparticles encapsulated in polymeric shells reveal less compact and more distorted configurations for experiments performed in a liquid medium compared to their dried counterparts. A similar expansion can be observed in quantitative analysis of the surface-to-surface distances of self-assembled Au nanorods in water rather than in a vacuum, in agreement with bulk measurements. This study, therefore, emphasizes the importance of developing high-resolution characterization tools that preserve the native environment of colloidal nanostructures. Drying force-induced deformation complicates the characterization of the 3D structure of colloidal assemblies. During chemical growth, the handedness of nanocrystals is usually tuned by selecting the corresponding enantiomer of chiral molecules involved in asymmetric growth, often known as chiral inducers. We report that, even using the same chiral inducer enantiomer, the handedness of chiral gold nanocrystals can be reversed by using Au nanorod seeds with either single crystalline or pentatwinned structure. This effect holds for chiral growth induced both by amino acids and by chiral micelles. 4D printing is the 3D printing of objects that change chemically or physically in response to an external stimulus over time. Photothermally responsive shape memory materials are attractive for their ability to undergo remote activation. While photothermal methods using gold nanorods (AuNRs) are used for shape recovery, 3D patterning of these materials into objects with complex geometries using degradable materials is not addressed. Here, the fabrication of 3D printed shape memory bioplastics with photo‐activated shape recovery is reported. Protein‐based nanocomposites based on bovine serum albumin (BSA), poly (ethylene glycol) diacrylate (PEGDA), and AuNRs are developed for vat photopolymerization. These 3D printed bioplastics are mechanically deformed under high loads, and the proteins served as mechano‐active elements that unfolded in an energy‐dissipating mechanism that prevented fracture of the thermoset. The bioplastic object maintained its metastable shape‐programmed state under ambient conditions. Subsequently, up to 99% shape recovery is achieved within 1 min of irradiation with near‐infrared (NIR) light. Mechanical characterization and small angle X‐ray scattering (SAXS) analysis suggest that the proteins mechanically unfold during the shape programming step and may refold during shape recovery. The tumor microenvironment (TME) is a dynamic pseudoorgan that shapes the development and progression of cancers. It is a complex ecosystem shaped by interactions between tumor and stromal cells. Although the traditional focus has been on the paracrine communication mediated by protein messengers, recent attention has turned to the metabolic secretome in tumors. Metabolic enzymes, together with exchanged substrates and products, have emerged as potential biomarkers and therapeutic targets. However, traditional techniques for profiling secreted metabolites in complex cellular contexts are limited. Surface-enhanced Raman scattering (SERS) has emerged as a promising alternative due to its nontargeted nature and simplicity of operation. Although SERS has demonstrated its potential for detecting metabolites in biological settings, its application in deciphering metabolic interactions within multicellular systems like the TME remains underexplored. In this study, we introduce a SERS-based strategy to investigate the secreted purine metabolites of tumor cells lacking methylthioadenosine phosphorylase (MTAP), a common genetic event associated with poor prognosis in various cancers. Our SERS analysis reveals that MTAP-deficient cancer cells selectively produce methylthioadenosine (MTA), which is taken up and metabolized by fibroblasts. Fibroblasts exposed to MTA exhibit: i) molecular reprogramming compatible with cancer aggressiveness, ii) a significant production of purine derivatives that could be readily recycled by cancer cells, and iii) the capacity to secrete purine derivatives that induce macrophage polarization. Our study supports the potential of SERS for cancer metabolism research and reveals an unprecedented paracrine crosstalk that explains TME reprogramming in MTAP-deleted cancers. Micro-and nanorobots have the potential to perform non-invasive drug delivery, sensing, and surgery in living organisms, with the aid of diverse medical imaging techniques. To perform such actions, microrobots require high spatiotemporal resolution tracking with real-time closed-loop feedback. To that end, photoacoustic imaging has appeared as a promising technique for imaging microrobots in deep tissue with higher molecular specificity and contrast. Here, we present different strategies to track magnetically-driven micromotors with improved contrast and specificity using dedicated contrast agents (Au nanorods and nanostars). Furthermore, we discuss the possibility of improving the light absorption properties of the employed nanomaterials considering possible light scattering and coupling to the underlying metal-oxide layers on the micromotor's surface. For that, 2D COMSOL simulation and experimental results were correlated, confirming that an increased spacing between the Au-nanostructures and the increase of thickness of the underlying oxide layer lead to enhanced light absorption and preservation of the characteristic absorption peak. These characteristics are important when visualizing the micromotors in a complex in vivo environment, to distinguish them from the light absorption properties of the surrounding natural chromophores. Chiral plasmonic nanoparticles (and their assemblies) interact with biomolecules in a variety of different ways, resulting in distinct optical signatures when probed by circular dichroism spectroscopy. These systems show promise for biosensing applications and offer several advantages over achiral plasmonic systems. Arguably the most notable advantage is that chiral nanoparticles can differentiate between molecular enantiomers and can, therefore, act as sensors for enantiomeric purity. Furthermore, chiral nanoparticles can couple more effectively to chiral biomolecules in biological systems if they have a matching handedness, improving their effectiveness as biomedical agents. In this article, we review the different types of interactions that occur between chiral plasmonic nanoparticle systems and biomolecules, and discuss how circular dichroism spectroscopy can probe these interactions and inform how to optimize systems for biosensing and biomedical applications. Chiral metal nanoparticles (NPs) offer a powerful means of inducing and harnessing optical activity. However, due to the incomplete knowledge of the underlying growth mechanisms, there is still limited control over the achievable morphological detail and, consequently, over the resulting optical activity. Therefore, theoretical modeling is needed to guide experimental development toward optimizing the plasmonic chiroptical response. Toward filling this gap, herein an extensive parametric analysis is presented, via computer‐aided‐design (CAD) models and full‐wave electrodynamic simulations, which aims at systematically analyzing the influence of structural changes on the plasmonic circular dichroism (CD) spectra of rod‐shaped gold NPs comprising helical indentations on achiral nanorod cores. From this analysis, interesting patterns in the plasmon‐mediated resonant behavior are identified and cause–effect relationships are drawn that may serve as a go‐to recipe for the understanding and fabrication of these NPs and their applications, such as spectroscopic (bio)detection including CD spectral shifts and surface‐enhanced Raman optical activity. Anti-Stokes photoluminescence (PL) is light emission at a higher photon energy than the excitation, with applications in optical cooling, bioimaging, lasing, and quantum optics. Here, we show how plasmonic nano-cavities activate anti-Stokes PL in WSe2 monolayers through resonant excitation of a dark exciton at room temperature. The optical near-fields of the plasmonic cavities excite the out-of-plane transition dipole of the dark exciton, leading to light emission from the bright exciton at higher energy. Through statistical measurements on hundreds of plasmonic cavities, we show that coupling to the dark exciton leads to a near hundred-fold enhancement of the upconverted PL intensity. This is further corroborated by experiments in which the laser excitation wavelength is tuned across the dark exciton. We show that a precise nanoparticle geometry is key for a consistent enhancement, with decahedral nanoparticle shapes providing an efficient PL upconversion. Finally, we demonstrate a selective and reversible switching of the upconverted PL via electrochemical gating. Our work introduces the dark exciton as an excitation channel for anti-Stokes PL in WSe2 and paves the way for large-area substrates providing nanoscale optical cooling, anti-Stokes lasing, and radiative engineering of excitons. Hyperthermia, as the process of heating a malignant site above 42 °C to trigger cell death, has emerged as an effective and selective cancer therapy strategy. Various modalities of hyperthermia have been proposed, among which magnetic and photothermal hyperthermia are known to benefit from the use of nanomaterials. In this context, we introduce herein a hybrid colloidal nanostructure comprising plasmonic gold nanorods (AuNRs) covered by a silica shell, onto which iron oxide nanoparticles (IONPs) are subsequently grown. The resulting hybrid nanostructures are responsive to both external magnetic fields and near-infrared irradiation. As a result, they can be applied for the targeted magnetic separation of selected cell populations – upon targeting by antibody functionalization – as well as for photothermal heating. Through this combined functionality, the therapeutic effect of photothermal heating can be enhanced. Bottom‐up assembly of nanoparticle‐on‐mirror (NPoM) nanocavities enables precise inter‐metal gap control down to ≈ 0.4 nm for confining light to sub‐nanometer scales, thereby opening opportunities for developing innovative nanophotonic devices. However limited understanding, prediction, and optimization of light coupling and the difficulty of controlling nanoparticle facet shapes restricts the use of such building blocks. Here, an ultraprecise symmetry‐breaking plasmonic nanocavity based on gold nanodecahedra is presented, to form the nanodecahedron‐on‐mirror (NDoM) which shows highly consistent cavity modes and fields. By characterizing > 20 000 individual NDoMs, the variability of light in/output coupling is thoroughly explored and a set of robust higher‐order plasmonic whispering gallery modes uniquely localized at the edges of the triangular facet in contact with the metallic substrate is found. Assisted by quasinormal mode simulations, systematic elaboration of NDoMs is proposed to give nanocavities with near hundred‐fold enhanced radiative efficiencies. A new methodology is presented to count the number of atoms in multimetallic nanocrystals by combining energy dispersive X‐ray spectroscopy (EDX) and high angle annular dark field scanning transmission electron microscopy (HAADF STEM). For this purpose, the existence of a linear relationship between the incoherent HAADF STEM and EDX images is exploited. Next to the number of atoms for each element in the atomic columns, the method also allows quantification of the error in the obtained number of atoms, which is of importance given the noisy nature of the acquired EDX signals. Using experimental images of an Au@Ag core–shell nanorod, it is demonstrated that 3D structural information can be extracted at the atomic scale. Furthermore, simulated data of an Au@Pt core–shell nanorod show the prospect to characterize heterogeneous nanostructures with adjacent atomic numbers. [ABSTRACT FROM AUTHOR]

Controlled assembly of gold nanoparticles (AuNPs) into clusters is a promising avenue for the development of sensitive bioimaging and diagnostic tools based on surface‐enhanced Raman scattering (SERS). However, several challenges, such as biocompatibility or colloidal and structural stability in biological environments, remain before AuNPs can be used as a tool for in vivo bioimaging. A versatile strategy for the preparation of colloidally stable and biocompatible AuNP clusters (AuNPCs) is introduced with high SERS signals, which are used as SERS contrast bioimaging agents (SERS tags). By tuning the ligand ratio of Raman reporter molecules to stabilizing polymer on the surface of each AuNP, aggregation can be carefully controlled. The resulting AuNPCs exhibit redshifted surface plasmon resonances in the near‐infrared (NIR) region, as well as distinct electromagnetic hotspots that give rise to SERS analytical enhancement factors above 104, compared to non‐clustered spherical AuNPs. Thanks to the protective polymer shell, high levels of cellular uptake with low cytotoxicity are observed, allowing 3D SERS mapping of cells with sufficiently high spatial resolution to detect AuNPCs within intracellular organelles. Thin films sustaining plasmonic circular dichroism (PCD) have acquired high scientific relevance and a great potential for applications. While most efforts in PCD thin film structures focus on lithographically fabricated static metasurfaces, the bottom‐up fabrication of active chiral plasmonic films constitutes an alternative approach. Herein, the preparation of PCD thin films by melting and freezing a mixture of liquid crystal (LC), a chiral dopant, and gold nanoparticles (Au NPs), serving as helical matrix, symmetry breaking inducer, and plasmonic component, respectively is reported. UV–vis and circular dichroism spectroscopies, as well as theoretical modeling are used to disclose the interactions among thin film components, toward maximizing the PCD dissymmetry factor (g‐factor). Variation of substrate temperature affords reversible off/on switching of the chiroptical response. The soft nature of LC matrix enables patterning of the films via a thermal nanoimprinting method, using a polydimethylsiloxane mold for transfer‐printing onto a flexible substrate, leading to stretchable PCD films. The PCD wavelengths can be readily tuned by varying the geometry of the Au NPs. Macroporous materials with controlled pore sizes are of high scientific and technological interest, due to their low specific weight, as well as unique acoustic, thermal, or optical properties. Solid foams made of titania, silica, or silicon, as representative materials, have been previously obtained with several hundred nanometer pore sizes, by using sacrificial templates such as spherical emulsion droplets or colloidal particles. Macroporous structures in particular are excellent candidates as photonic materials with applications in structural coloration and photonic bandgap devices. However, whereas using spherical building blocks as templates may provide tight control over pore shape and size, it results in materials with an often unfavorable local topology. Templating dry‐foam or compressed‐emulsion structures appear as attractive alternatives, but have not been demonstrated so far for submicron pore sizes. Herein, the use of soft, flexible microgel colloids decorated with silica nanoparticles as templates of macroporous foams is reported. These purposely synthesized core–shell colloids are assembled at ultra‐high effective volume fractions by centrifuging and thermal swelling, thereby resulting in uniform disordered materials with facetted pores, mimicking dry foams. With the advent of flexible electronics, the old fashioned and conventional solid‐state technology will be replaced by conductive inks combined with low‐cost printing techniques. Graphene is an ideal candidate to produce conductive inks, due to its excellent conductivity and zero bandgap. The possibility to chemically modify graphene with active molecules opens up the field of responsive conductive inks. Herein, a bioresponsive, electroactive, and inkjet‐printable graphene ink is presented. The ink is based on graphene chemically modified with selected enzymes and an electrochemical mediator, to transduce the products of the enzymatic reaction into an electron flow, proportional to the analyte concentration. A water‐based formulation is engineered to be respectful with the enzymatic activity while matching the stringent requirements of inkjet printing. The efficient electrochemical performance of the ink, as well as a proof‐of‐concept application in biosensing, is demonstrated. The versatility of the system is demonstrated by modifying graphene with various oxidoreductases, obtaining inks with selectivity toward glucose, lactate, methanol, and ethanol. Plasmonic metal‐organic frameworks are composite nanoparticles comprising plasmonic metal nanoparticles (NPs) embedded within a metal‐organic framework (MOF) matrix. As a result, not only the functionalities of the individual components are retained, but synergistic effects additionally provide improved chemical and physical properties. Recent progress in plasmonic MOFs has demonstrated the potential for nanofabrication and various nanotechnology applications. Synthetic challenges toward plasmonic MOFs have been recently addressed, resulting in new opportunities toward practical applications, such as surface‐enhanced Raman scattering, therapy, and catalysis. The impact of key parameters (thermodynamic vs. kinetic) on the synthetic pathways of plasmonic MOFs is reviewed, while providing insight into related progress toward structure‐derived applications. Lattice plasmons, i.e., diffractively coupled localized surface plasmon resonances, occur in long‐range ordered plasmonic nanostructures such as 1D and 2D periodic lattices. Such far‐field coupled resonances can be employed for ultrasensitive surface‐enhanced Raman spectroscopy (SERS), provided they are spectrally matched to the excitation wavelength. The spectral positions of lattice plasmon modes critically depend on the lattice period and uniformity, owing to their pronounced sensitivity to structural disorder. We report the fabrication of superlattices by templated self‐assembly of gold nanoparticles on a flexible support, with tunable lattice‐plasmon resonances by means of macroscopic strain. We demonstrate that the highest SERS performance is achieved by matching the lattice plasmon mode to the excitation wavelength, by post‐assembly fine‐tuning of long‐range structural parameters. Both asymmetric and symmetric lattice deformations can be used to adapt a single lattice structure to both red‐shifted and blue‐shifted excitation lines, as exemplified by lattice expansion and contraction, respectively. This proof‐of‐principle study represents a basis for alternative designs of adaptive functional nanostructures with mechanically tunable lattice resonances using strain as a macroscopic control parameter. Bimetallic nanoparticles display unique physical and chemical properties, including improved chemical stability, enhanced optical properties, or higher catalytic activity. Here, a synthetic methodology is described to obtain bimetallic heterostructures and alloyed plasmonic nanocrystals through the irradiation of colloidal Au@Ag core–shell nanorods (Au@Ag NRs) with femtosecond laser pulses. Depending on the energy deposited on the Au@Ag NRs, different morphologies and degrees of alloying are obtained, such as hot‐dog‐like and rice‐like (partially alloyed) NRs, as well as fully alloyed nanospheres. By using advanced electron microscopy techniques and energy‐dispersive X‐ray spectroscopy (EDX) tomography, both the morphology and the elemental distribution of the irradiated nanoparticles can be disclosed, and correlated to detailed investigations of their optical properties using electromagnetic simulations. The wide variety of bimetallic species provided by the proposed approach is a clear indication of the potential of combining synthetic colloidal methods with fs‐pulsed laser irradiation for the fabrication of unique multielemental nanoparticles. From a geometrical perspective, a chiral object does not have mirror planes or inversion symmetry. It exhibits the same physical properties as its mirror image (enantiomer), except for the chiroptical activity, which is often the opposite. Recent advancements have identified particularly interesting implications of chirality on the optical properties of metal nanoparticles, which are intimately related to localized surface plasmon resonance phenomena. Although such resonances are usually independent of the circular polarization of light, specific strategies have been applied to induce chirality, both in assemblies and at the single-particle level. In this tutorial review, we discuss the origin of plasmonic chirality, as well as theoretical models that have been proposed to explain it. We then summarise recent developments in the synthesis of discrete nanoparticles with plasmonic chirality by means of wet-chemistry methods. We conclude with a discussion of promising applications for discrete chiral nanoparticles. We expect this tutorial review to be of interest to researchers from a wide variety of disciplines where chiral plasmonics can be exploited at the nanoparticle level, such as chemical sensing, photocatalysis, photodynamic or photothermal therapies, etc. [ABSTRACT FROM AUTHOR]

Raman spectroscopy is becoming increasingly popular as an in vitro bioimaging technique, when coupled with plasmonic substrates such as gold nanoparticles (AuNPs). Plasmonic AuNPs not only display excellent biocompatibility but can also induce the surface‐enhanced Raman scattering (SERS) effect, which can be exploited for cell labeling, as an interesting alternative to fluorescence‐based techniques. SERS bioimaging requires the use of so‐called SERS tags or SERS‐encoded AuNPs. A remaining difficulty toward the general implementation of this method is the difficulty to correlate the SERS signal (spectral intensity) with the number of SERS tags. Therefore, a general correlation method, suitable for arbitrary AuNP morphologies and Raman‐active molecules (Raman reporters or RaRs), should largely improve the quantitative character of SERS as an imaging technique. We propose a protocol, with an associated app (SERSTEM), which enables the user to determine the average SERS intensity per nanoparticle from transmission electron microscopy (TEM) and SERS data. As a proof of concept, we demonstrated the method for Au nanostars and nanorods, carrying four different RaRs, and implemented the SERSTEM app, which is publicly available from an open‐source platform. Self-assembly of cellulose nanocrystals (CNCs) doped with anisotropic gold nanorods (AuNRs) was studied by small-angle neutron scattering. Correlation distances and structured domains were analysed to determine the influence of CNC and AuNR concentration on structuring. The transfer of the nematic structure of CNCs to AuNRs is explained in terms of an entropy-driven evolution from an isotropic to a cholesteric phase, with small nematic domains already present in the "isotropic" phase in equilibrium with the chiral nematic phase. 3D printing strategies have acquired great relevance toward the design of 3D scaffolds with precise macroporous structures, for supported mammalian cell growth. Despite advances in 3D model designs, there is still a shortage of detection tools to precisely monitor in situ cell behavior in 3D, thereby allowing a better understanding of the progression of diseases or to test the efficacy of drugs in a more realistic microenvironment. Even if the number of available inks has exponentially increased, they do not necessarily offer the required functionalities to be used as internal sensors. Herein the potential of surface‐enhanced Raman scattering (SERS) spectroscopy for the detection of biorelevant analytes within a plasmonic hydrogel‐based, 3D‐printed scaffold is demonstrated. Such SERS‐active scaffolds allow for the 3D detection of model molecules, such as 4‐mercaptobenzoic acid. Flexibility in the choice of plasmonic nanoparticles is demonstrated through the use of gold nanoparticles with different morphologies, gold nanorods showing the best balance between SERS enhancement and scaffold transparency. Detection of the biomarker adenosine is also demonstrated as a proof‐of‐concept toward the use of these plasmonic scaffolds for SERS sensing of cell‐secreted molecules over extended periods of time. Perovskite nanocrystals (NCs) have revolutionized optoelectronic devices because of their versatile optical properties. However, controlling and extending these functionalities often requires a light‐management strategy involving additional processing steps. Herein, we introduce a simple approach to shape perovskite nanocrystals (NC) into photonic architectures that provide light management by directly shaping the active material. Pre‐patterned polydimethylsiloxane (PDMS) templates are used for the template‐induced self‐assembly of 10 nm CsPbBr3 perovskite NC colloids into large area (1 cm2) 2D photonic crystals with tunable lattice spacing, ranging from 400 nm up to several microns. The photonic crystal arrangement facilitates efficient light coupling to the nanocrystal layer, thereby increasing the electric field intensity within the perovskite film. As a result, CsPbBr3 2D photonic crystals show amplified spontaneous emission (ASE) under lower optical excitation fluences in the near‐IR, in contrast to equivalent flat NC films prepared using the same colloidal ink. A detailed 3D investigation of nanoparticles at a local scale is of great importance to connect their structure and composition to their properties. Electron tomography has therefore become an important tool for the 3D characterization of nanomaterials. 3D investigations typically comprise multiple steps, including acquisition, reconstruction, and analysis/quantification. Usually, the latter two steps are performed offline, at a dedicated workstation. This sequential workflow prevents on‐the‐fly control of experimental parameters to improve the quality of the 3D reconstruction, to select a relevant nanoparticle for further characterization, or to steer an in situ tomography experiment. Here, an efficient approach to overcome these limitations is presented, based on the real‐time reconstruction of arbitrary 2D reconstructed slices through a 3D object. Colloidal anisotropic gold nanocrystals play a central role in the field of plasmonics owing to their tunable optical activity across a wide spectral range. However, achieving sufficient optical quality for practical implementation requires advanced synthetic protocols yielding gold nanocrystals with the desired morphology and plasmonic properties. This Minireview focuses on a fundamental step during the growth of anisotropic nanocrystals, namely symmetry breaking. In connection with thermodynamic and kinetic control of nanocrystal growth, we discuss the complex interplay between the role of seed morphology and that of surfactants, shape‐directing additives and reducing agents. We revisit some iconic syntheses of anisotropic gold nanoparticles, including nanorods, nanotriangles, and nanobipyramids. Finally, we analyze the use of co‐surfactants as an emerging strategy to disconnect the symmetry breaking event from the anisotropic growth process, overcoming current limitations in the synthesis of anisotropic gold nanocrystals. The composition and intercellular interactions of tumor cells in the tissues dictate the biochemical and metabolic properties of the tumor microenvironment. The metabolic rewiring has a profound impact on the properties of the microenvironment, to an extent that monitoring such perturbations could harbor diagnostic and therapeutic relevance. A growing interest in these phenomena has inspired the development of novel technologies with sufficient sensitivity and resolution to monitor metabolic alterations in the tumor microenvironment. In this context, surface‐enhanced Raman scattering (SERS) can be used for the label‐free detection and imaging of diverse molecules of interest among extracellular components. Herein, the application of nanostructured plasmonic substrates comprising Au nanoparticles, self‐assembled as ordered superlattices, to the precise SERS detection of selected tumor metabolites, is presented. The potential of this technology is first demonstrated through the analysis of kynurenine, a secreted immunomodulatory derivative of the tumor metabolism and the related molecules tryptophan and purine derivatives. SERS facilitates the unambiguous identification of trace metabolites and allows the multiplex detection of their characteristic fingerprints under different conditions. Finally, the effective plasmonic SERS substrate is combined with a hydrogel‐based three‐dimensional cancer model, which recreates the tumor microenvironment, for the real‐time imaging of metabolite alterations and cytotoxic effects on tumor cells. The design and development of contrast agents (CAs) for magnetic resonance imaging (MRI) in clinical analysis is expected to improve the image spatial resolution and to increase the detection sensitivity, especially regarding neurological disorders and cancer disease. In particular, advanced CAs for T1-weighted images are investigated to achieve the sensitive detection of early-stage primary tumors or brain metastases. In this study, we present a strategy toward diagnostic T1 CAs for MRI, based on polymer-modified MnO nanoparticles (NPs). Two different nanosystems were synthesized, consisting of (1) colloidal MnO nanocrystals wrapped by a multidentate amphiphilic polymer and (2) MnO nanocrystals embedded within poly-(lactic-co-glycolic acid) (PLGA) NPs. These nanosystems were compared in terms of their MRI contrast power and biological safety. The latter system combines the excellent biocompatibility of PLGA with the unique magnetic properties of MnO NPs and allows sustained contrast enhancement over time. Longitudinal relaxivities of both MnO composite nanomaterials proved to be higher than those of commercial Gd-based CAs and Teslascan, both in phosphate-buffered saline and in plasma, also exhibiting low cytotoxicity. Methods to image complex 3D cell cultures are limited by issues such as fluorophore photobleaching and decomposition, poor excitation light penetration, and lack of complementary techniques to verify the 3D structure. Although it remains insufficiently demonstrated, surface‐enhanced Raman scattering (SERS) imaging is a promising tool for the characterization of biological complex systems. To this aim, a controllable 3D cell culture model which spans nearly 1 cm2 in surface footprint is designed. This structure is composed of fibroblasts containing SERS‐encoded nanoparticles (i.e., SERS tags), arranged in an alternating layered structure. This "sandwich" type structure allows monitoring of the SERS signals in the z‐axis and with mm dimensions in the xy‐axis. Taking advantage of correlative microscopy techniques such as electron microscopy, it is possible to corroborate nanoparticle positioning and distances in z‐depths of up to 150 µm. Once introduced into the human body, nanoparticles often interact with blood proteins, which in turn undergo structural changes upon adsorption. Although protein corona formation is a widely studied phenomenon, the structure of proteins adsorbed on nanoparticles is far less understood. We propose a model to describe the interaction between human serum albumin (HSA) and nanoparticles (NPs) with arbitrary coatings. Our model takes into account the competition between protonated and unprotonated polymer ends and the curvature of the NPs. To this end, we explored the effects of surface ligands (citrate, PEG-OMe, PEG-NH2, PEG-COOH, and glycan) on gold nanoparticles (AuNPs) and the pH of the medium on structural changes in the most abundant protein in blood plasma (HSA), as well as the impact of such changes on cytotoxicity and cellular uptake. We observed a counterintuitive effect on the ζ-potential upon binding of negatively charged HSA, while circular dichroism spectroscopy at various pH values showed an unexpected pattern in the reduction of α-helix content, as a function of surface chemistry and curvature. Our model qualitatively reproduces the decrease in α-helix content, thereby offering a rationale based on particle curvature. The simulations quantitatively reproduce the charge inversion measured experimentally through the ζ-potential of the AuNPs in the presence of HSA. Finally, we found that AuNPs with adsorbed HSA display lower toxicity and slower cell uptake rates, compared to functionalized systems in the absence of protein. Our study allows examining and explaining the conformational dynamics of blood proteins triggered by NPs and corona formation, thereby opening new avenues toward designing safer NPs for drug delivery and nanomedical applications.
