PD-1/PD-L1 blockade therapies have displayed extraordinary clinical efficacy for melanoma, renal, bladder and lung cancer; however, only a minority of colorectal cancer (CRC) patients benefit from these treatments. The efficacy of PD-1/PD-L1 blockade in CRC is limited by the complexities of tumor microenvironment. PD-1/PD-L1 blockade immunotherapy is based on T cell-centered view of tumor immunity. However, the onset and maintenance of T cell responses and the development of long-lasting memory T cells depend on innate immune responses. Acknowledging the pivotal role of innate immunity in anti-tumor immune response, this review encapsulates the employment of combinational therapies those involve PD-1/PD-L1 blockade alongside the activation of innate immunity and explores the underlying cellular mechanisms, aiming to harnessing innate immune responses to induce long-lasting tumor control for CRC patients who received PD-1/PD-L1 blockade therapy. Background: As a prominent part of complementary and alternative medicine, Chinese Medicine (CM) has proved its strengths in treating a diverse range of acute and chronic medical conditions and is at present recognized in 196 countries and territories worldwide. In 2012, Australia regulated the CM profession under the National Regulation and Accreditation Scheme (NRAS) by legislation and reports quarterly demographic information about individual CM practitioners so to ensure public interest, although research examining the change of CM workforce in Australia has been scarce. Objective: This study aims to investigate the construction of the CM workforce in Australia and more importantly, evaluated its development in the last decade to capture the trajectory and trend in the present period and future potential changes. Methods: Data were sourced from the Australian Health Practitioner Regulation Agency (AHPRA) annual reports and the Chinese Medicine Board of Australia (CMBA) registration statistics from 2012 to 2023. A descriptive analysis was conducted with demographic variables, including profession, age, and gender, and chi-square tests and linear regression modeling were carried out to assess the variations between regions and across years. Results: The population of CM practitioners in 2022/2023 stagnated with slight decrease to 4,823, in contrast to the increase rate of 2.9% in the whole health care community. The number of young CM registrants (<35 y) shrank by 37.5% from 691 in 2012 to 432 in 2023. In comparison with other health care professions, CM comprises the smallest proportion of the population aged younger than 25 (0.2%) and the largest proportion aged older than 65 years (16.2%), advancing into an aging era. Conclusions: This study indicates a worrying potential decline in CM workforce in Australia, which is likely to be further exacerbated by the lack of new graduates and rise of median age among practitioners. Meanwhile, continued advancement in Western medicine technology and standards requires substantial efforts to increase both a better understanding of CM and demonstration of its efficacy. Furthermore, greater effort is needed to recruit and educate new young CM practitioners in Australia and to broaden the international training pipeline for a sustainable development of CM practice. Summary: TMEM56, a gene coding a transmembrane protein, is abundantly expressed in erythroid cells. Despite this, its role in erythropoiesis has not been well characterized. In this study, we sought to clarify the function of TMEM56 in erythroid development, focusing specifically on its involvement in haem biosynthesis and cell cycle progression. To do this, we used CD34+ haematopoietic stem cells derived from umbilical cord blood and differentiated them into erythroid cells in an ex vivo model. Our results indicate that the loss of TMEM56 disrupts haem biosynthesis and impairs erythroid differentiation. Furthermore, deletion of Tmem56 in the erythroid lineage in murine models using erythropoietin receptor (EpoR)‐Cre revealed defects in erythroid progenitors within the bone marrow under both normal conditions and during haemolytic anaemia. These observations underscore the regulatory role of TMEM56 in maintaining erythroid lineage homeostasis. Taken together, our results unveil a previously unrecognized function of TMEM56 in erythroid differentiation and suggest its potential as an unfounded target for therapeutic strategies in the treatment of erythropoietic disorders. Background: The increasing incidence of endometrial cancer (EC) has highlighted the need for improved early detection methods. This study aimed to develop and validate a novel DNA methylation classifier, EMPap, for EC detection using cervical scrapings. Methods: EMPap incorporated the methylation status of BHLHE22 and CDO1, along with age and body mass index (BMI), into a logistic regression model to calculate the endometrial cancer methylation (EM) score for identifying EC in cervical scrapings. We enrolled 1297 patients with highly suspected EC, including 196 confirmed EC cases, and assessed the EMPap performance in detecting EC. Results: EMPap demonstrated robust diagnostic accuracy, with an area under the curve of 0.93, sensitivity of 90.3%, and specificity of 89.3%. It effectively detected EC across various disease stages, grades, and histological subtypes, and consistently performed well across patient demographics and symptoms. EMPap correctly identified 87.5% of the type II ECs and 53.8% of premalignant lesions. Notably, compared with transvaginal ultrasonography (TVS) in patients with postmenopausal bleeding, EMPap exhibited superior sensitivity (100% vs. 82.0%) and specificity (85.2% vs. 38.5%). In asymptomatic postmenopausal women, EMPap maintained high sensitivity (89.5%) and negative predictive value (NPV) (98.3%). Conclusions: This study demonstrated the potential of EMPap as an effective tool for EC detection. Despite the limited sample size, EMPap showed promise for identifying type II EC and detecting over 50% of premalignant lesions. With the recent developments of composite polymer materials, the usage of synthetic materials in interior furnishing has rapidly increased. Polymeric foams such as polyurethane (PU) and polyethylene (PE) are widely applied as upholstered furniture and wall insulations. Nevertheless, the occurrence of fire in a confined environment surrounded by combustible polymeric materials can lead to disastrous consequences. This is because the toxic emission of polymers, specifically carbon monoxide (CO), is responsible for most deaths and injuries in building fires. Thus, it is essential to gain an in-depth understanding of material flammability and gaseous product emissions, as well as the correlation against various radiant heat fluxes, temperatures and ventilation conditions. In this study, cone calorimeter with Fourier Transform Infrared Spectroscopy (FT-IR) was performed on PU, PE and Ethylene–vinyl acetate (EVA) foam to (i) characterise the critical combustion performances; and (ii) obtain real-time emission profiles of asphyxiant gases such as hydrogen cyanide (HCN), formaldehyde (HCHO), and acrolein, under flaming conditions. Furthermore, a fire dynamics simulator (FDS) model was constructed to study the burning characteristics of the selected materials numerically. The reinforcement of drug-seeking motivation following drug withdrawal is recognized as a significant factor contributing to relapse. The ventral pallidum (VP) plays a crucial role in encoding and translating motivational aspects of reward. However, current research lacks a clear understanding of how the VP mediates drug-seeking motivation and the feedback modulation between the VP and the nucleus accumbens (NAc) following drug withdrawal. Therefore, utilizing a rat model of cocaine self-administration, we investigated the circuitry mechanisms underlying drug-seeking behavior post-drug withdrawal involving the NAc-VP pathway. Initially, we observed a significant enhancement in drug-seeking behavior 14 days after cocaine withdrawal. Subsequently, we identified the feedback mechanism through which the NAc-VP regulates this behavior. Immunofluorescence results indicated an increase in c-Fos expression levels in the ventral pallidum ventromedial (VPvm) and ventrolateral ventral pallidum (VPvl) following drug withdrawal. Calcium fiber photometry further elucidated that during the expression of high motivational drug-seeking behavior, there was a specific enhancement in VPvm neuronal activity, and retrograde tracing techniques suggested a weakened transmission function in the NAc-VPm pathway. Additionally, chemical genetic techniques demonstrated that inhibiting the activity of the NAc-VP pathway could increase the motivational level of drug-seeking behavior. These findings indicate that the reduced inhibitory function of the NAc-VP pathway following prolonged cocaine withdrawal forms the basis for heightened reactivity in VPvm neurons, thus regulating the expression of high motivational behavior triggered by drug-related cues. Our study results suggest that maintaining normal NAc-VP pathway functionality may decrease drug-seeking motivation post long-term drug withdrawal, offering new insights for interventions targeting relapse. Rutin is a flavonoid compound with potential for treating Alzheimer's disease, preventing brain damage, mitigating cerebral ischemia–reperfusion injury, and exhibiting anti-glioblastoma activity. However, its efficacy is limited by its low solubility, poor bioavailability, and limited permeability across the blood–brain barrier (BBB). To enhance the bioavailability and brain-targeting ability of Rutin, transferrin-modified Rutin liposome (Tf-Rutin-Lip) was developed using liposomes as a delivery system. Rutin liposomes were prepared using the thin-film dispersion method, and the preparation conditions were optimized using the response surface methodology. Then, transferrin (Tf) was incorporated into the liposomes through covalent modification, yielding Tf-Rutin liposomes. The toxicity of these liposomes on bEnd.3 cells, as well as their impact on the tight junctions of these cells, was rigorously evaluated. Additionally, in vitro and in vivo experiments were conducted to validate the brain-targeting efficacy of the Tf-Rutin liposomes. A susceptible detection method was developed to characterize the pharmacokinetics of Tf-Rutin-Lip further. The optimized conditions for the preparation of Tf-Rutin-Lip were determined as follows: a lipid-to-cholesterol ratio of 4.63:1, a drug-to-lipid ratio of 1:45.84, a preparation temperature of 42.7 °C, a hydration volume of 20 mL, a sonication time of 10 min, a surfactant concentration of 80 mg/mL, a DSPE-MPEG-2000 concentration of 5%, and a DSPE-PEG2000-COOH to DSPE-MPEG-2000 molar ratio of 10%. The liposomes did not affect the cell activity of bEnd.3 cells at 24 h and did not disrupt the tight junction of the blood–brain barrier. Tf-modified liposomes were taken up by bEnd.3 cells, which, in turn, passed through the BBB, thus improving liposomal brain targeting. Furthermore, the results of pharmacokinetic experiments showed that the Cmax, AUC0-∞, AUC0-t, MRT0-∞, and t1/2 of Tf-Rutin-Lip increased 1.99-fold, 2.77-fold, 2.58-fold, 1.26-fold, and 1.19-fold compared to those of free Rutin solution, respectively. The dosing of fluconazole for young infants remains empirical because of the limited pharmacokinetic (PK) data. We aimed to establish a population PK model and assess the systematic exposure-response of commonly used regimens of fluconazole in Chinese infants. We included infants with a postnatal age of less than 120 days and received intravenous fluconazole. Both scheduled and scavenged plasma samples were collected, and fluconzaole concentration was determined by a validated ultra-performance liquid chromatography-tandem mass spectrometry assay. Population PK analysis was conducted using Phoenix NLME, and then Monte Carlo simulation was conducted to predict the probability of target attainment (PTA) of empirically used regimens of both prophylactic and therapeutic purposes. Based on 304 plasma samples from 183 young infants, fluconazole concentration data was best described by a one-compartment model with first-order elimination. Gestational Age (GA), postnatal age (PNA), and body weight (BW) were included in the final model as CL = 0.02*(GA/214)2.77*(PNA/13)0.24*exp(nCL); V = 1.56*(BW/1435)0.90*exp(nV). Model validation revealed the final model had qualified stability and acceptable predictive properties. Monte Carlo simulation indicated that under the same minimum inhibitory concentration (MIC) value and administration regimen, PTA decreased with GA and PNA. The commonly used prophylactic regimens can meet the clinical need, while higher doses might be needed for treatment of invasive candidiasis. This population PK model of fluconazole discriminated the impact of GA and PNA on CL and BW on V. Dosing adjustment was needed according to the GA and PNA of infants to achieve targeted exposures. Objective: To observe the anti-inflammatory and glycometabolic effects of glutathione (GSH) on macrophages in collagen induced arthritis (CIA) mice. Methods: 1 CIA model establishment and groups: A total of 14 female DBA/1J mice were randomly divided into: CIA+PBS group and CIA+GSH group. The mice were sacrificed on the 50th day, collecting serum and isolating bone marrow derived macrophages (BMDM), which were marked as BMDM1. 2 Trained immunity model establishment and groups: BMDM were isolated from normal DBA/1J mice, and were pretreated with histone H3K27 demethylases inhibitor (GSKJ1) or PBS for 2 h. Then, serum from CIA model mice in vivo was incubated for 24 h, and the samples were grouped as follows: (CIA+GSH)+PBS group, (CIA+GSH)+GSKJ1 group, (CIA+PBS)+PBS group, (CIA+PBS)+GSKJ1 group. Lipopolysaccharide (LPS) was adopted to stimulated cells on the 6th day, which were marked as BMDM2. 3 RNA sequencing was used to detect differentially expressed genes (DEGs) and their function in BMDM1 and BMDM2. q-PCR was adopted to estimate the mRNA levels of PFK and Idh3g. The culture supernatants were used to measure the protein levels of TNF-α and IL-6 by ELISA. Results: 1 Compared with CIA+PBS group, the mice in CIA+GSH group showed lighter of joint swelling (P<0.05), less arthritis index (P<0.05), HE staining suggested less inflammatory cell infiltration, Safranin O-fast green staining showed more chondrocytes, TRAP staining indicated reduced osteoclasts. 2 In BMDM1, GO analysis showed that DEGs were mainly involved in glutathione derivative metabolic process, IL-6 production, inflammatory response, innate immune response, regulation of primary metabolic process and glycolipid binding, compared with CIA+ GSH and CIA+PBS group. In CIA+GSH group, the mRNA level of PFK was significantly decreased (P<0.05), while Idh3g was also significantly up-regulated (P<0.05), and the expression of TNF-α, IL-6 were both reduced (P<0.05) compared with CIA+PBS group. 3 In BMDM2, GO analysis showed that DEGs were also involved in inflammatory response, activation of innate immune response, regulation of tumor necrosis factor production, positive regulation of IL-6 production, regulation of glycolytic process and 1, 3- β-Dglucan binding between CIA+GSH and CIA+PBS group. Furthermore, in (CIA+GSH)+PBS group, PFK was decreased (P<0.05), Idh3g was up-regulated (P<0.05), and IL-6 was also significantly down-regulated (P<0.05) compared with (CIA+PBS)+PBS group. However, there was no significant difference in the expression of Idh3g and PFK, moreover, TNF-α and IL-6 were significantly upregulated compared with (CIA+GSH)+GSKJ1 group and (CIA+GSH)+PBS group. Conclusion: GSH can regulate glycometabolism and inflammatory response of macrophages via demethylation of histone H3K27, and it can also alleviate CIA in mice. [ABSTRACT FROM AUTHOR]

The performance and stability of organic metal halide perovskite (OMHP) optoelectronic devices have been associated with ion migration. Understanding of nanoscale resolved organic cation migration mechanism would facilitate structure engineering and commercialization of OMHP. Here, we report a three‐dimensional approach for in situ nanoscale infrared imaging of organic ion migration behavior in OMHPs, enabling to distinguish migrations along grain boundary and in crystal lattice. Our results reveal that organic cation migration along OMHP film surface and grain boundaries (GBs) occurs at lower biases than in crystal lattice. We visualize the transition of organic cation migration channels from GBs to volume upon increasing electric field. The temporal resolved results demonstrate the fast MA+ migration kinetics at GBs, which is comparable to diffusivity of halide ions. Our findings help understand the role of organic cations in the performance of OMHP devices, and the proposed approach holds broad applicability for revealing migration mechanisms of organic ions in OMHPs based optoelectronic devices. Many major diseases of the retina often show symptoms of lesions in the fundus of the eye. The extraction of blood vessels from retinal fundus images is essential to assist doctors. Some of the existing methods do not fully extract the detailed features of retinal images or lose some information, making it difficult to accurately segment capillaries located at the edges of the images. In this paper, we propose a multi-scale retinal vessel segmentation network (SCIE_Net) based on skip connection information enhancement. Firstly, the network processes retinal images at multiple scales to achieve network capture of features at different scales. Secondly, the feature aggregation module is proposed to aggregate the rich information of the shallow network. Further, the skip connection information enhancement module is proposed to take into account the detailed features of the shallow layer and the advanced features of the deeper network to avoid the problem of incomplete information interaction between the layers of the network. Finally, SCIE_Net achieves better vessel segmentation performance and results on the publicly available retinal image standard datasets DRIVE, CHASE_DB1, and STARE. [ABSTRACT FROM AUTHOR]

Textures in molybdenum‐rhenium (Mo‐Re) alloys are inevitable during thermal fabrication. [110] and [100] are common orientations in Mo‐Re alloys and effect mechanical responses. However, orientation dependence of mechanical responses in Mo‐Re alloys is not quite clear yet. To clarity this problem, micro‐pillar compression tests are conducted in grains with orientation [100] and [110] separately. Orientation‐dependent compressive properties are found in Mo‐14Re and Mo‐42Re (wt.%), but are not found in Mo and Mo‐5Re, which may be attributed to activated multi‐slip planes as increased Re. Solid solution effect of Re not only relies on orientations, but also on Re contents. Softening effect occurs in both [100] and [110] Mo‐5Re. while, strong strengthening effect happens in [110] Mo‐14Re and Mo‐42Re. Our research clarifies that Mo‐Re alloys with [110] orientation/texture could be preferred to obtain good strengthening effect. [ABSTRACT FROM AUTHOR]

This study investigates the effects of breeds, feeding methods, and parts on the volatile flavor of yak meat. Gas chromatography–ion mobility spectrometry (GC-IMS) and multivariate analysis were used to analyze the volatile organic components (VOCs) in yak meat from various sources. A total of 71 volatile compounds were identified, 53 of which were annotated based on the GC-IMS database. These include 20 alcohols, 16 ketones, 10 aldehydes, four alkenes, one ester, one acid, and one furan. Using VOC fingerprinting and multivariate analysis, yak meats from different sources were distinctly categorized. Breed had the most significant impact on yak meat VOCs, followed by feeding method and then part. Six volatiles with a variable importance in projection value greater than one were identified as potential markers for distinguishing yak meat. This study offers insights into the flavor profile of yak meat from different sources and demonstrates the efficacy of GC-IMS and multivariate analysis in characterizing and discriminating meats. Plantarflexors provide propulsion during walking and receive input from both corticospinal and corticoreticulospinal tracts, which exhibit some frequency-specificity that allows potential differentiation of each tract's descending drive. Given that stroke may differentially affect each tract and impair the function of plantarflexors during walking; here, we examined this frequency-specificity and its relation to walking-specific measures during post-stroke walking. Fourteen individuals with chronic stroke walked on an instrumented treadmill at self-selected and fast walking speed (SSWS and FWS, respectively) while surface electromyography (sEMG) from soleus (SOL), lateral gastrocnemius (LG), and medial gastrocnemius (MG) and ground reaction forces (GRF) were collected. We calculated the intermuscular coherences (IMC; alpha, beta, and low-gamma bands between SOL–LG, SOL–MG, LG–MG) and propulsive impulse using sEMG and GRF, respectively. We examined the interlimb and intralimb IMC comparisons and their relationships with propulsive impulse and walking speed. Interlimb IMC comparisons revealed that beta LG–MG (SSWS) and low-gamma SOL–LG (FWS) IMCs were degraded on the paretic side. Intralimb IMC comparisons revealed that only alpha IMCs (both speeds) exhibited a statistically significant difference to random coherence. Further, alpha LG–MG IMC was positively correlated with propulsive impulse in the paretic limb (SSWS). Alpha and beta/low-gamma bands may have a differential functional role, which may be related to the frequency-specificity of the underlying descending drives. The persistence of alpha band in plantarflexors and its strong positive relationship with propulsive impulse suggests relative alteration of corticoreticulospinal tract after stroke. Cavity optomechanical systems have received widespread attentions because they provide a novel platform for metrology, sensing, hybrid systems and quantum information processing. Their nonlinear dynamics has rich physics and plays an important role in the application scenarios. Previous works devoted to this subject have usually focused on the self-induced oscillation and chaos, whereas other parts of the rich nonlinear-dynamics picture are almost uncharted waters. In this study, we fill this gap and report the first experimental observation of limit-torus attractor, whose dynamics exhibits a torus-like trajectory in phase space. Moreover, we investigate the sharp decrease of oscillating amplitude along the up scanning transmission spectrum, referred to as catastrophe point, for the first time. The location of catastrophe point is independent of the pump power and the coupling distance. Our findings enrich the nonlinear dynamics in optomechanical systems, and open up new ways towards exploiting these systems as versatile building blocks in various applications including communication, quantum information processing, sensing and metrology. Constructing highly efficient electrocatalysts is vital to enhance oxygen evolution reaction (OER) performance at industrially relevant current densities. Herein, three-phase coupled Ni3S2/r-NiS/h-NiS composites are grown in situ on Ni foam (NNSN/NF) via a one-step solvothermal approach. The as-prepared composites need overpotentials of only 377 mV, 451 mV and 476 mV at 1000 mA cm−2 for the OER in alkaline freshwater, simulated seawater and seawater, respectively. In addition, the optimized catalyst exhibited long-term durability at 300 mA cm−2. Our work clarifies designing and preparing cost-effective Ni-based sulfide electrocatalysts for the OER in alkaline freshwater/simulated seawater/seawater under industrially relevant current densities. Background: A dosimetric evaluation is still lacking in terms of clinical target volume (CTV) omission in stage III patients treated with 4D-CT Intensity-Modulated Radiation Therapy (IMRT). Methods: 49 stage III NSCLC patients received 4D-CT IMRT were reviewed. Target volumes and organs at risk (OARs) were re-delineated. Four IMRT plans were conducted retrospectively to deliver different prescribed dose (74 Gy–60 Gy), and with or without CTV implementation. Dose and volume histogram (DVH) parameters were collected and compared. Results: In the PTV-g 60 Gy plan (PTV-g refers to the PTV generated from the internal gross tumor volume), only 5 of 49 patients had the isodose ≥ 50 Gy line covering at least 95% of the PTV-c (PTV-c refers to the PTV generated from the internal CTV) volume. When the prescribed dose was elevated to 74 Gy to the PTV-g, 33 of 49 patients could have the isodose ≥ 50 Gy line covering at least 95% of the PTV-c volume. In terms of OARs protection, the SIB-IMRT plan showed the lowest value of V5, V20, and mean dose of lung, had the lowest V55 of esophagus, and the lowest estimated radiation doses to immune cells (EDIC). The V20, V30, and mean dose of heart was lower in the simultaneous integrated boost (SIB) IMRT (SIB-IMRT) plan than that of the PTV-c 60 Gy plan. Conclusions: CTV omission was not suitable for stage III patients when the prescribed dose to PTV-g was 60 Gy in the era of 4D-CT IMRT. CTV omission plus high dose to PTV-g (74 Gy for example) warranted further exploration. The SIB-IMRT plan had the best protection to normal tissue including lymphocytes, and might be the optimal choice. [ABSTRACT FROM AUTHOR]

Glioblastoma organoids (GBOs) serve as a powerful and reliable tool to study glioblastoma stem cells (GSCs) and glioblastoma (GBM). GBOs can be derived from different materials using different methods. To identify the predominant generation methods and the most applications of GBOs, we searched four databases (PubMed, Embase, Web of Science, and Wiley Online Laboratory) from August 2021 to August 2023. After screening, 42 out of 295 articles were included and analyzed. GBOs in these articles were generated using only one material, such as tumor tissues, tumor cells, and gene-edited multifunctional stem cells, or simultaneously using two materials, such as tumor cells and normal organoids. Methodologically, direct cultivation of GBM cells or tissues was the most commonly used method to generate GBOs. Embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) were the frequently used multifunctional stem cells to generate GBOs by simultaneously silencing P53, NF1, and PTEN using CRISPR/Cas9. In terms of applications, GBOs generated by direct cultivation of GBM tissue had the most applications, including molecular mechanisms, therapy, and culture technique. Some critical civil infrastructure projects, such as government offices, command buildings, airport stations, and transportation routes, are highly susceptible to missile attacks during conflicts. To investigate the influence of impact velocity ( v 0) and impact location on the dynamic responses of RC slabs and projectiles, finite element models were established to simulate the perforation of the 300 mm-diameter projectile into the 100 mm-thick RC slab. The results indicate that both v 0 and impact location have effects on the relative relationship between kinetic energy and internal energy of the RC slab. Stresses exceeding 48 MPa ( f c) and maximum displacement of the RC slab are primarily concentrated around the bullet hole, extending to a distance of approximately 450 mm (1.5 d) from the impact center. After perforation, the relationship between velocity loss ratio and v 0 , as well as the relationship between kinetic energy loss ratio and v 0 , conforms to the power function distribution. Considering the damage to the RC slab, the Chen model, Peng model, and Konyaew model of the residual velocity prove more suitable for this working condition, especially the Chen model. The study of oxygen evolution reaction (OER) catalysts is crucial for the development of hydrogen energy technology. However, industrial water splitting requires high current densities, and monometallic catalysts have poor stability at high current densities, so the development of multimetallic high-entropy catalyst has become a focus of attention. Herein, we report an amorphous high-entropy FeCoNiZnOOH electrocatalyst that exhibits exceptional stability during the OER under alkaline conditions for over 150 h at 1 A cm−2, an industrial current density condition. And after the OER, the catalyst performance was further improved to achieve a current density of 100 mA cm−2 with only 191 mV overpotential. This phenomenon is attributed to the leaching of Zn, increasing the lattice oxygen content. Meanwhile, the catalyst nanoparticles unfold to form nanoflower-like catalysts, which increases the ECSA of the catalyst. Precise DNA replication is fundamental for genetic inheritance. In eukaryotes, replication initiates at multiple origins that are first "licensed" and subsequently "fired" to activate DNA synthesis. Despite the success in identifying origins with specific DNA motifs in Saccharomyces cerevisiae, no consensus sequence or sequences with a predictive value of replication origins have been recognized in metazoan genomes. Rather, epigenetic rules and chromatin structures are believed to play important roles in governing the selection and activation of replication origins. We propose that replication initiation is facilitated by a group of sequence‐specific "replication pioneer factors," which function to increase chromatin accessibility and foster a chromatin environment that is conducive to the loading of the prereplication complex. Nowadays, with the approaching carbon neutrality deadlines and the implementation of zero-net carbon emission policies, the research and development of flame retardants are increasingly influenced by stringent regulations and laws. In this context, natural inorganic materials have garnered significant attention as promising flame retardants to enhance the fire resistance of polymer composites. These materials offer unique advantages, such as being environmentally friendly, cost-effective, and producing no carbon emissions during preparation. Consequently, in recent years, there has been a rapid increase in research on natural mineral flame retardants. This review systematically introduces the research progress on natural minerals as flame retardants in epoxy resin, highlighting their ability to provide exceptional fire resistance. Additionally, we categorize the various chemical modification approaches for natural minerals and explore different various natural mineral-based flame retardants. Furthermore, we elucidate the flame retardant mechanisms behind both natural and modified mineral flame retardant systems. Beyond summarizing and concluding the current state of research, we also project future research directions and identify challenges in the development of natural mineral flame retardants. Two series of vanillin derivatives containing 1,3,4-oxadiazole and 1,3-thiazolidin-4-one scaffolds were prepared and evaluated for their antifungal activity. The results revealed that compounds 6j (29.73 μg/ml) and 7a (38.15 μg/ml) displayed excellent inhibitory activity against the spore of Fusarium solani. The inhibitory activity of compound 7d (10.53 μg/ml) against the spore of Alternaria solani was more than 42-fold that of vanillin. Compound 7a (37.54 μg/ml) showed better antifungal activity against the spore of B. cinerea than positive controls. Simple Summary: This study was the first attempt to use heat-killed Lactobacillus acidophilus as a feed additive to replace antibiotics. The results of the evaluation showed that it could improve the performance of piglets, increase intestinal microbial diversity and thus ensure the intestinal health of piglets. First, a feed-additive sample of heat-killed Lactobacillus acidophilus IFFI 6005 was prepared by culture. Second, 96 (initial BW = 14.38 ± 0.67 kg, weaning age of 40 days) healthy piglets were selected and randomized into four treatment groups. Each treatment group consisted of three replicates (n = 8). Pigs were fed a basal diet (NC), basal diet plus antibiotics (PC), basal diet plus Lactobacillus acidophilus IFFI 6005 at 600 g/t (LA, 1.0 × 1010 cfu/g), and basal diet plus heat-killed Lactobacillus acidophilus IFFI 6005 at 600 g/t (HKLA), respectively; the trial lasted for 30 days. The results showed that the ratios of feed to gain (F:G) and diarrhea rate of both the HKLA and PC groups were significantly lower compared with the NC and LA groups (p < 0.05); however, there was no significant difference between the HKLA and PC group (p > 0.05). In addition, the average daily weight gain (ADG) of the HKLA group was significantly higher (p < 0.05) than that of the other three groups in terms of growth performance. Finally, 16S rRNA sequencing and metabolome analysis based on fecal samples further elaborated that the addition of heat-killed Lactobacillus acidophilus IFFI 6005 to the feed improved the intestinal microbial diversity and abundance (p < 0.05) and reduced the abundance of pathogenic bacteria (p < 0.05), but it did not affect the abundance of Lactobacillus (p > 0.05). Through the comparison of microbial abundance and metabolite content between the two groups (NC_vs_HKLA), the largest differences were found in six microorganisms and 10 metabolites in the intestine (p < 0.05). These differential metabolites were involved in the digestion, absorption and utilization of protein and starch, as well as in oxidative stress. Deep learning is a powerful branch of machine learning, which presents a promising new approach for diagnose diseases. However, the deep learning for detecting anterior cruciate ligament still limits to the evaluation of whether there are injuries. Plant Growth-Promoting Rhizobacteria (PGPR) are gaining increasing attention, but their interactions with indigenous rhizosphere microbiomes remain unclear. To address this issue, we isolated a strain of Priestia aryabhattai with a growth-promoting effect. Under greenhouse conditions, its growth-promoting effect on alfalfa was evaluated, and amplicon sequencing was used to analyze changes in the rhizosphere microbial community to explore the growth promotion mechanism. Our study shows that inoculation with Priestia aryabhattai increases the α-diversity index of the alfalfa rhizosphere microbiome and enhances the abundance of beneficial bacterial genera. This is likely because inoculation with Priestia aryabhattai increased the abundance of carbon-sequestering genera, particularly Gemmatimonas, thereby improving the soil environment. The increased abundance of beneficial bacteria stimulates root development in alfalfa and enhances nutrient uptake, particularly phosphorus, which in turn boosts photosynthesis and promotes alfalfa growth. In summary, Priestia aryabhattai improves soil environment and promotes alfalfa growth by enhancing the carbon sequestration capacity of the rhizosphere microbial community. Developing optical anti-counterfeiting technology with dynamic color changes offers new hope for improving the replication difficulty of anti-counterfeiting labels and promoting the updating and iteration of high-level security technology and information encryption, but remains a huge challenge. Herein, we develop a ground-breaking multi-wavelength excitation-dependent anti-counterfeiting technology based on security inks containing three luminescent materials that respond to 254 nm, 365 nm, and 980 nm excitation, respectively. A blue carbon dots/calcium carbonate (B-CDs/CaCO3) composite and a green carbon dots/silicon dioxide (G-CDs/SiO2) composite are prepared via in situ loading processes. The former exhibits blue fluorescence (FL) emission light and green room-temperature phosphorescence (RTP) when turning on/off 365 nm excitation, and the latter emits green FL under the same conditions. Under 254 nm excitation, excellent red emission is realized in the YVO4:Eu3+ (R-Eu) phosphor. Blue and green upconversion luminescence (UCL) are obtained in YVO4:Tm3+/Yb3+ and YVO4:Er3+/Yb3+ phosphors upon 980 nm excitation. When integrating these luminescent materials into invisible security inks, the printed security patterns exhibit triple-mode polychromatic information and spatial dynamic color-gradient characteristics by varying the excitation conditions (wavelength, intensity, illumination angle, and exposure time of excitation light). The unique optical properties enable the resulting security labels to show a high-level anti-counterfeiting strength and demonstrate application value in next-generation anti-counterfeiting. This study first analyzed the differences in physiological and biochemical indicators, muscle histological characteristics, and muscle fiber types. A total of 248 mRNAs, 25 miRNAs, and 432 lncRNAs were identified as differentially expressed by whole transcriptome sequencing. Key genes that may influence meat production traits include MTMR14, PPP1R3A, PYGM, PGAM2, MYH1, and MYH7. Simple Summary: Following weaning, piglets exhibit an immature immune system, reduced capacity to digest and absorb nutrients, and encounter diverse stressors that significantly compromise their overall health. The supplementation of probiotics in animal feed has proven to be a beneficial strategy for enhancing the welfare of livestock and poultry. The findings of this research demonstrate that supplementing the diet with 5 × 108 CFU/kg of Clostridium butyricum enhances fecal microflora diversity and richness through the modulation of specific metabolites such as L-aspartic acid, 5-hydroxyindole-3-acetic acid, stearic acid, and adipic acid. This mechanism contributes to a reduction in the incidence of diarrhea in weaned piglets triggered by E. coli K88. This study aimed to evaluate the effects of Clostridium butyricum (C. butyricum) on the prevention of the diarrhea rates and growth performances of weaned piglets induced by Escherichia coli K88 (E. coli K88). Twenty-four weaned piglets (6.92 ± 0.11 kg) were randomly assigned to one of three treatment groups for a period of 21 days. Each group consisted of eight pigs, with each pig being housed in an individual pen. Group I received the control diet along with normal saline, Group II received the control diet along with E. coli K88, and Group III received the control diet supplemented with 5 × 108 CFU/kg of C. butyricum and E. coli K88. We examined alterations in rectal microbiota and metabolites, analyzed the incidence of diarrhea, and investigated the interactions between microbiota and metabolites through the application of Illumina MiSeq sequencing and liquid chromatography–mass spectrometry. The results showed that, from days 14 to 21, the diarrhea incidence in Group III decreased significantly by 83.29% compared to Group II (p < 0.05). Over the entire experimental duration, the average daily feed intake of Group III decreased significantly by 11.13% compared to Group I (p < 0.05), while the diarrhea incidence in Group III decreased by 71.46% compared to Group II (p < 0.05). The predominant microbial flora in the rectum consisted of Firmicutes (57.32%), Bacteroidetes (41.03%), and Proteobacteria (0.66%). Administering E. coli K88 orally can elevate the relative abundance of Megasphaera (p < 0.05). Conversely, the supplementation of C. butyricum in the diet reduced the relative abundance of Megasphaera (p < 0.05), while increasing the relative abundance of unclassified_f_Lachnospiraceae (p < 0.05). Rectal metabolomics analysis revealed that supplementing C. butyricum in the feed significantly altered the amino acids and fatty acids of the piglets infected with E. coli K88 (p < 0.05). The correlation analysis showed that the occurrence of diarrhea was inversely related to adipic acid (p < 0.05) and positively associated with (5-hydroxyindol-3-YL) acetic acid and L-aspartic acid (p < 0.05). Alloying elements segregating to grain boundaries (GBs) is vital in determining structural stability and mechanical properties of alloys, especially in high‐entropy alloys (HEAs) that consist of multiple elements. However, some details remain unclear, such as the process of varied atoms (Cr and Co) cosegregation to GBs in FeNiCrCo alloy. Therefore, the competition and cooperation between Fe, Ni, Cr, and Co atoms segregating to GBs of FeNiCrCo alloy were investigated via molecular dynamic (MD) simulation. Five [110] symmetric tilt GBs of FeNiCrCo were applied to study the relation between the tendency of atomic segregation especially Cr and Co and GB characters including GB energy and GB excess free volume (GB excess volume). Background: The expression of CYP19A1 has implications for the prognosis of female bladder cancer. However, this study aimed to explore the association between single nucleotide polymorphisms (SNPs) in CYP19A1 and bladder cancer risk, as no prior research has addressed this association. Research design and methods: We selected and genotyped five CYP19A1 SNPs (rs4646, rs6493487, rs1062033, rs17601876, and rs3751599) in 217 patients and 550 controls using the Agena MassARRAY system. Logistic regression analysis was employed to calculate the odds ratio (OR) and 95% confidence intervals (CIs). Bioinformatics predicted SNP functions and CYP19A1 involving pathways. Results: Our study revealed a significant association between bladder cancer risk and four SNPs (rs4646 (AC vs. CC: OR = 1.71, FDR-p = 0.005), rs6493487 (G vs. A: OR = 0.68, FDR-p = 0.011), rs1062033 (G vs. C: OR = 0.36, FDR-p < 0.001), and rs17601876 (GA vs. GG: OR = 1.66, FDR-p = 0.008)) in CYP19A1. The three SNPs (rs4646, rs1062033, and rs17601876) were significantly correlated with CYP19A1 expression levels in normal whole blood (p < 0.05). Moreover, CYP19A1 was found to primarily participate in the steroid hormone biosynthesis and metabolic pathways. The environment is an important factor affecting the composition and abundance of metabolites in O. sinensis, which indirectly determines its edible function and medicinal potential. This study integrated metabolomics and redundancy analysis (RDA) to analyze the metabolite profile characteristics and key environmental factors influencing O. sinensis in various production areas. A total of 700 differentially accumulated metabolites (DAMs) were identified, primarily comprising lipids, organic acids, and organoheterocyclic compounds. Results from hierarchical cluster analysis and KEGG indicated distinct accumulation patterns of these DAMs in O. sinensis from different regions, with enrichment in pathways such as tryptophan metabolism and glycerophospholipid metabolism. Environmental factors like annual mean precipitation, pH, temperature, and altitude were found to significantly influence metabolite composition, particularly lipids, organic acids, and nucleosides. NLRP6 plays a crucial role in maintaining intestinal homeostasis by regulating the interaction between the intestinal mucosa and the microbiota. However, the impact of NLRP6 deficiency on intestinal damage following hematopoietic stem cell transplantation (HSCT) remains poorly understood. In this study, we established a syngeneic HSCT mouse model using C57BL/6 mice as donors and NLRP6-/- or C57BL/6 mice as recipients. Our findings revealed that NLRP6 deficiency had minimal influence on peripheral blood cell counts and splenic immune cell proportions in transplanted mice. However, it exacerbated pathological changes in the small intestine on day 14 post-transplantation, accompanied by increased proportions of macrophages, dendritic cells, and neutrophils. Furthermore, the NLRP6 deficiency resulted in elevated expression of MPO and CD11b, while reducing the levels mature caspase-1 and mature IL-1β in the intestine. Moreover, the NLRP6 deficiency disturbed the expression of apoptosis-related molecules and decreased the tight junction protein occludin. Notably, recipient mice with NLRP6 deficiency exhibited lower mRNA expression levels of antimicrobial genes, such as Reg3γ and Pla2g2a. The short-term increase in inflammatory cell infiltration caused by NLRP6 deficiency was associated with intestinal damage, increased apoptosis, reduced expression of antimicrobial peptides, and impaired intestinal repair. Background: Logsplitter Injury is a type of high-energy ankle fracture dislocation. The mechanism of injury has not been described in detail. A detailed understanding of the radiological features and pathological changes can further guide treatment. Methods: Between April 2009 and December 2018, a retrospective analysis was conducted on 62 patients with Logsplitter injury. The study analysed the characteristics of fibular injury, tibial injury, syndesmosis injury, medial injury and lateral ligament injury on preoperative X-ray and CT scans. The incidence of the different injury types was summarised. The correlation between Logsplitter injuries and the mechanisms causing them were analysed using the Lauge-Hansen classification of ankle fractures. Results: The study provides data on the types of fractures observed. Of the total fractures, 98.4% were open fractures. The fibula injuries were classified as no fracture (1.6%), transverse or short oblique fractures (61.3%), butterfly fragments (25.8%), and comminuted fractures (11.3%). The tibial injuries included compression of lateral articular surfaces (38.7%) and posterior compressions (6.5%). Medial injuries, including medial malleolar fractures, accounted for 87.1%, and deltoid ligament rupture accounted for 12.9%. The study found that injuries to the syndesmosis consisted of simple ligament ruptures (11.3%), Tillaux fractures (8.1%), Volkmann fractures (43.5%), and Tillaux and Volkmann fractures (37.1%). In 12.9% of cases, there was a complete rupture of the lateral collateral ligament. Based on the Lauge-Hansen classification, 87.1% of injuries were pronation-abduction injuries, while 8.1% were pronation and external rotation injuries, and 1.6% were supination external rotation injuries. Furthermore, 3.2% of cases could not be classified. Conclusion: The pathoanatomic characteristics of Logsplitter injury are diverse, with some cases accompanied by collateral ligament injury. It is important to note that these evaluations are objective and based on current results. The most common injury mechanism is vertical violence combined with abduction, although in some cases, it may be a vertical combined external-rotation injury. Level of evidence: (4) case series. Due to the inherent intermittency, variability, and randomness, photovoltaic (PV) power generation faces significant challenges in energy grid integration. To address these challenges, current research mainly focuses on developing more efficient energy management systems and prediction technologies. Through optimizing scheduling and integration in PV power generation, the stability and reliability of the power grid can be further improved. In this study, a new prediction model is introduced that combines the strengths of convolutional neural networks (CNNs), long short-term memory (LSTM) networks, and attention mechanisms, so we call this algorithm CNN-LSTM-Attention (CLA). In addition, the Crested Porcupine Optimizer (CPO) algorithm is utilized to solve the short-term prediction problem in photovoltaic power generation. This model is abbreviated as CPO-CLA. This is the first time that the CPO algorithm has been introduced into the LSTM algorithm for parameter optimization. To effectively capture univariate and multivariate time series patterns, multiple relevant and target variables prediction patterns (MRTPPs) are employed in the CPO-CLA model. The results show that the CPO-CLA model is superior to traditional methods and recent popular models in terms of prediction accuracy and stability, especially in the 13 h timestep. The integration of attention mechanisms enables the model to adaptively focus on the most relevant historical data for future power prediction. The CPO algorithm further optimizes the LSTM network parameters, which ensures the robust generalization ability of the model. The research results are of great significance for energy generation scheduling and establishing trust in the energy market. This paper proposes a computational method for the reliability of 2-separable networks. Based on graph theory and probability theory, this method simplifies the calculation process by constructing a network equivalent model and designing corresponding algorithms to achieve the efficient evaluation of reliability. Considering independent random failures of edges with equal probability q, this method can accurately calculate the reliability of 2-separable networks, and its effectiveness and accuracy are verified through examples. In addition, to demonstrate the generality of our method, we have also applied it to other 2-separable networks with fractal structures and proposed linear algorithms for calculating their all-terminal reliability.
