Purpose: To assess the complications, satisfaction, and quality of life of patients rehabilitated with delayed and immediate loading of single crowns. Materials and Methods: An electronic search was conducted in PubMed/MEDLINE, Scopus, Web of Science, Cochrane Library, and Embase databases up to February 2023. Only prospective studies and randomized and nonrandomized clinical trials comparing immediate and delayed loading were included. For the quantitative analysis, dichotomous and consistent variables were evaluated with a 95% confidence interval. Results: A total of 20 studies were evaluated. No statistically significant difference was observed between protocols: satisfaction (I²: 0%; P = .42), quality of life (I²: 0 %; P = .05), biologic complications (I²: 9%; P = .17), mechanical complications (I²: 58%; P = .84), and survival rate (I²: 0%; P = .38). The subgroup analysis showed significant differences only for marginal bone loss (MBL) when immediate implants were placed in the mandible (I²: 15%; P = .01) and posterior zone (I²: 0%; P = .001). Conclusions: Complications and patient-centered outcomes for immediate single-implant crowns were comparable to delayed loading. Scientific evidence showed no significant difference between loading protocols for survival rates. Note that several factors could interfere with the complication events, implant failures, and MBL. The subgroup analysis showed that only immediate implants placed in the posterior mandible had a higher and statistically significant mean MBL. [ABSTRACT FROM AUTHOR]

Purpose: The purpose of this study was to investigate the mechanical properties of acrylic resins at different aging times for denture bases manufactured using the conventional method, microwave processing, milling, and 3D printing. Materials and Methods: A total of 160 rectangular samples (64 × 10 × 3.3 ± 0.03 mm) were prepared, divided among the four main resin groups, and subdivided into four analysis times (T0, T1, T2, and T3), resulting in 10 samples per subgroup. The samples were stored in distilled water at 37° ± 2°C for 24 hours (T0), then subjected to thermocycling at temperatures of 5° ± 1°C and 55° ± 1°C in different numbers of cycles: 5,000 (T1); 10,000 (T2); and 20,000 (T3). The mechanical properties evaluated were surface microhardness, flexural strength, and modulus of elasticity. Statistical differences between resin groups and aging time were evaluated using two-way analysis of variance (P < .05). Results: The 3D-printed resin showed the significantly lowest values of microhardness, flexural strength, and modulus of elasticity compared to other resins (P < .001). Conclusions: The CAD/CAM--milled denture resin showed mechanical properties similar to those of traditional resins (conventional and microwave-processed). The 3D-printing resin did not show adequate mechanical properties for long-term clinical use. Despite this, new studies are developing better properties of this resin for long-term use. This study aimed to evaluate the effects of adding chromium-methionine (Cr-meth) chelate in extruded diets containing reduced protein content on growth performance, carcass composition, nutrient retention, and hematobiochemical responses in Nile tilapia juveniles. The fish were fed with two control diets (no Cr-meth): positive control (311 g kg−1 of crude protein, estimated 258 g kg−1 of digestible protein) and negative control (255 g kg−1 of crude protein, estimated 207 g kg−1 of digestible protein). Another five experimental diets (mean 251 g kg−1 of crude protein, mean estimated 204 g kg−1 of digestible protein) were supplemented with 0.2, 0.4, 0.6, 0.8, and 1.0 mg Cr-meth kg diet−1. Juveniles (34.31 g) were organized in a randomized design (seven treatments in triplicate) in 21 tanks (250 L) and fed until apparent satiety. The experiment lasted 9 weeks. Fish fed 0.8 and 1.0 mg Cr-meth kg diet−1 showed zootechnical variables similar to the positive control. The reduced protein diets showed lower costs, and supplementation with 0.91 mg kg−1 of chromium matched the profitability observed in the positive control diet. Carcass gross energy and energy retention, plasma cholesterol (total and LDL), and triglycerides values were higher in fish fed 0.6 mg Cr-meth kg diet−1 than in the positive control. Treatments with dietary Cr-meth (mainly between 0.4 and 0.8 mg kg−1) decreased carcass Cr, selenium, and copper retention and increased carcass nitrogen retention and muscle glycogen levels compared to the positive control. This treatment (positive control) also had lower values for total leukocytes and monocytes, respectively, than fish fed 0.2 and 0.4 mg Cr-meth kg diet−1 and for eosinophils and lymphocytes than fish fed 0.8 mg Cr-meth kg diet−1. In conclusion, we recommend supplementation with 0.8 mg Cr-meth kg diet−1 for diets with low protein content, as it produces a dietary protein-sparing effect, reducing costs, improving immune defense, muscle glycogen, and nitrogen retention without compromising growth performance, physiology, and fish metabolism. The shrimp nursery is an important tool to improve the shrimp zootechnical and health performances. However, diseases and high mortality rates are the main limitations of the nursery system. Therefore, this work aimed to evaluate the influence of stocking density in the synbiotic nursery system, through growth, microbial composition, and water quality. The experiment lasted 30 days and used Penaeus vannamei postlarvae (PL10, 0.003 ± 0.001 g), fed with commercial feed (40–45% crude protein) and treatments were completely randomized with 2000, 4000, and 6000 PL‧m−3, with three replicates. Before and after stocking the postlarvae, in the experimental units (10 m3), were added a mixture of rice bran, brown sugar, commercial probiotic, and sterilized water (Synbiotic). The bottom water of tanks was drained twice a week, totaling a total 5% water exchange during the total experiment and alkalinity was corrected with calcium hydroxide (Ca(OH)2) when the alkalinity values were < 100 mg CaCO3‧L−1. Weekly, the total ammonia nitrogen, nitrite nitrogen, nitrate nitrogen, and total alkalinity were monitored and microbiological analyses were conducted at the beginning and end. After the trial (30 days), the influence of the different stocking densities on the final weight, survival, feed conversion ratio, weekly growth rate, and productivity, with higher values for 2000 PL‧m−3. Also observed. Vibrio spp., Lactobacillus spp., total heterotrophic bacteria, and yeast count values increased by the end of the period in all shrimp stocking densities, however, with only significant differences in yeast count. In relation to ammonia, nitrite, and nitrate, no significant differences were observed in the influence of the different stocking densities. Based on the data obtained, we recommend using a density of 2000 PL‧m−3 in the nursery phase, with better zootechnical performance, water quality, and microbial composition. [ABSTRACT FROM AUTHOR]

Standardized monitoring strategies are often used to study spatial and temporal ecological patterns and trends. Such approaches are applied for many study taxa, including bats (Mammalia, Chiroptera). However, local characteristics of individual field sites, including species assemblages, terrain, climatic factors, and presence or lack of landscape features, may affect the efficacy of these standardized surveys. In this paper, we completed mist-netting surveys for bats in two widely separated field sites, Calakmul Biosphere Reserve (CBR), a Mexican lowland tropical forest, and Krka National Park (KNP), a Mediterranean dry scrub forest in Croatia. Standardized surveys were conducted along predefined transects for six hours. We also completed targeted surveys in KNP that focused on the key bat activity period (the first two to three hours after sunset), with nets being deployed at sites of known or assumed value to bats (independent of predefined transects). We analyzed how survey success differed in standardized surveys between CBR and KNP and between standardized and targeted surveys in KNP. Survey success was measured through three parameters: capture rate = the number of individual bats captured per net hour, inventory rate = the number of unique bat species recorded per net hour, and inventory efficacy = the percentage of known species assemblage recorded per net hour across all surveys. Results for all three parameters indicate that standardized surveys in CBR were vastly more effective than those in KNP (e.g., mist-netting in CBR detected 69.8% of the species assemblage, compared to just 8.3% in KNP), and it was only by employing targeted mist-netting in KNP that meaningful capture rates could be achieved. This study contributes further evidence to discussions around how and when standardized survey methods should be employed, and the alternative approaches that can be taken in ecosystems where generally effective methods underperform. Introduction: The sufCDSUB gene cluster, encoding the sole iron-sulfur (Fe-S) cluster assembly system in S. mutans , was recently shown to be up-regulated in response to oxidative stressors and Fe limitation. Methods: In this study, luciferase reporter fusion assays, electrophoretic gel mobility shift assays (EMSA) and in vitro transcription assays (IVT) were used to dissect the cis- and trans- acting factors that regulate the expression of sufCDSUB. Results and discussion: Results showed deletion of perR , for the only Fur-family transcriptional regulator in S. mutans , resulted in >5-fold increases in luciferase activity under the control of the sufCDSUB promoter (P<0.01), as compared to the parent strain, UA159 when the reporter strains were grown in medium with no supplemental iron. Site-directed mutagenesis of a PerR-box in the promoter region led to elevation of the reporter activity by >1.6-fold (P <0.01). In an EMSA, recombinant PerR (rPerR) was shown to bind to the cognate sufCDSUB promoter leading to mobility retardation. On the other hand, the reporter activity was increased by >84-fold (P<0.001) in response to the addition of cysteine at 4 mM to the culture medium. Deletion of cysR , for a LysR-type of transcriptional regulator, led to reduction of the reporter activity by >11.6-fold (P <0.001). Addition of recombinant CysR (rCysR) to an EMSA caused mobility shift of the sufCDSUB promoter probe, indicative of rCysR-promoter interaction, and rCysR was shown to enhance sufC transcription under the direction of sufCDSUB promoter in vitro. These results suggest that multiple factors are involved in the regulation of sufCDSUB expression in response to environmental cues, including cysteine and Fe availability, consistent with the important role of sufCDSUB in S. mutans physiology. Although the benefits of incorporating algal oil (AO) in plant‐based aquafeeds have been established for most commercial fish species, knowledge on the effects of such incorporation in shrimp feeds is scarce. The present study assessed how AO inclusion at different levels in a plant‐based shrimp feed could promote growth and fatty acid (FA) deposition in the muscle. Pacific white shrimp, Litopenaeus vannamei, were fed a reference diet with marine animal ingredients (F0) or a marine‐free plant‐based diet with vegetable oil (VO) alone (P0), or in combination with 1% (P1) or 2% (P2) AO. After 51 days of culture, zootechnical and nutritional performance of shrimp fed with AO supplemented diets was significantly improved. The sensory attributes of shrimp fed P1 and P2 were slightly improved relative to P0 while the eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) content was three to four times higher, thereby increasing the nutritional value of shrimp. Therefore, plant‐based feeds can sustain an efficient production of L. vannamei with controlled FA profiles, provided they are supplemented with AO. In addition, the use of AO eliminates the reliance of shrimp feeds on the limited supplies of marine ingredients while maintaining the forage fish dependency ratio for fish oil (FO) of zero and allows for the use of more flexible formulae based on different meals of nonmarine origin. The rs12979860 polymorphism, related to the IFNL4 gene, is suggested as a factor that impacts fibrosis progression in hepatitis C virus (HCV) infection and exhibits a wide distribution pattern across global populations. In this retrospective cross-sectional study, we aimed to investigate the frequency of this variant in an Amazonian population from Brazil, as well as its association with liver fibrosis development and its staging in HCV carriers. Our results show a significant association of the TT genotype in the sample of patients with HCV (OR = 2.291; 95% CI = 1.088–4.826; p = 0.033) and the greater frequency of the T allele (62.1%), which is similar to the those of African populational groups. Populational genetics analysis showed significant differences in allele frequencies on global levels. The frequency of the C allele in the study population (37.8%) was like that of the African population (39.7%), and differed from all other populations, which ranged from 62.5% to 92.9%. These findings suggest that rs12979860 plays a role in susceptibility to hepatitis C. Additionally, they allow us to propose that the response to hepatitis C infection in this group may resemble that of the African population. [ABSTRACT FROM AUTHOR]

An emerging hypothesis linking arsenic toxicity involves altered epigenetic mechanisms, such as DNA methylation. In this study, we examined the relationship between parents' arsenic exposure and DNA methylation in tissues obtained from 28 infants with spina bifida from Bangladesh. We analyzed arsenic in parents' toenails using inductively coupled plasma mass spectrometry (ICP-MS). DNA methylation was measured in infants' dural tissue, buccal swabs, and whole blood using the Illumina Infinium MethylationEPIC BeadChip. We performed epigenome-wide association analyses (EWAS) and tested differentially methylated regions (DMRs). In EWAS, DNA methylation at cg24039697 in dural tissue was positively associated (β = 0.59, p = 7.6 × 10−9) with father's toenail arsenic concentrations, adjusting for covariates. We did not identify any CpG sites related to father's arsenic exposure in the other tissues, or any CpG sites related to mother's arsenic exposure. Gene ontology analysis identified many biological pathways of interest, including the Wnt signaling pathways. We identified several DMRs across the tissues related to arsenic exposure that included probes mapping to genes that have previously been identified in studies of neural tube defects. This study emphasizes the potential impact of arsenic exposure in fathers, often understudied in epidemiological studies, on DNA methylation in a unique neurological tissue specific to spina bifida. The cortical epigenetic clock was developed in brain tissue as a biomarker of brain aging. As one way to identify mechanisms underlying aging, we conducted a GWAS of cortical age. We leveraged postmortem cortex tissue and genotyping array data from 694 participants of the Rush Memory and Aging Project and Religious Orders Study (ROSMAP; 11000,000 SNPs), and meta-analysed ROSMAP with 522 participants of Brains for Dementia Research (5,000,000 overlapping SNPs). We confirmed results using eQTL (cortical bulk and single nucleus gene expression), cortical protein levels (ROSMAP), and phenome-wide association studies (clinical/neuropathologic phenotypes, ROSMAP). In the meta-analysis, the strongest association was rs4244620 (p = 1.29 × 10−7), which also exhibited FDR-significant cis-eQTL effects for CD46 in bulk and single nucleus (microglia, astrocyte, oligodendrocyte, neuron) cortical gene expression. Additionally, rs4244620 was nominally associated with lower cognition, faster slopes of cognitive decline, and greater Parkinsonian signs (n ~ 1700 ROSMAP with SNP/phenotypic data; all p ≤ 0.04). In ROSMAP alone, the top SNP was rs4721030 (p = 8.64 × 10−8) annotated to TMEM106B and THSD7A. Further, in ROSMAP (n = 849), TMEM106B and THSD7A protein levels in cortex were related to many phenotypes, including greater AD pathology and lower cognition (all p ≤ 0.0007). Overall, we identified converging evidence of CD46 and possibly TMEM106B/THSD7A for potential roles in cortical epigenetic clock age. [ABSTRACT FROM AUTHOR]

Machado-Joseph disease (MJD) is an autosomal dominant spinocerebellar ataxia (SCA) caused by a polyglutamine expansion in the ataxin-3 protein, which initiates a cascade of pathogenic events, including transcriptional dysregulation. Genotype-phenotype correlations in MJD are incomplete, suggesting an influence of additional factors, such as epigenetic modifications, underlying the MJD pathogenesis. DNA methylation is known to impact the pathophysiology of neurodegenerative disorders through gene expression regulation and increased methylation has been reported for other SCAs. In this work we aimed to analyse global methylation in MJD carriers. Global 5-mC levels were quantified in blood samples of 33 MJD mutation carriers (patients and preclinical subjects) and 33 healthy controls, matched by age, sex, and smoking status. For a subset of 16 MJD subjects, a pilot follow-up analysis with two time points was also conducted. No differences were found in median global 5-mC levels between MJD mutation carriers and controls and no correlations between methylation levels and clinical or genetic variables were detected. Also, no alterations in global 5-mC levels were observed over time. Our findings do not support an increase in global blood methylation levels associated with MJD. [ABSTRACT FROM AUTHOR]

Arsenic and its inorganic compounds affect numerous organs and systemic functions, such as the nervous and hematopoietic systems, liver, kidneys, and skin. Despite a large number of studies on arsenic toxicity, rare reports have investigated the leukopenia incidence in workers exposed to arsenic. In workplaces, the main source of workers' exposure is the contaminated air by the inorganic arsenic in mines, arsenic or copper smelter industries, and chemical factories. Erythropoiesis inhibition is one of the arsenic effects and it is related to regulatory factor GATA-1. This factor is necessary for the normal differentiation of early erythroid progenitors. JAK-STAT is an important intracellular signal transduction pathway responsible for the mediating normal functions of several cytokines related to cell proliferation and hematopoietic systems development and regulation. Arsenic inactivates JAK-STAT by inhibiting JAK tyrosine kinase and using the IFNγ pathway. The intravascular hemolysis starts after the absorption phase when arsenic binds to the globin of hemoglobin in erythrocytes and is transported into the body, which increases the oxidation of sulfhydryl groups in hemoglobin. So, this article intends to highlight the potential leukopenia risk via inhalation for workers exposed to arsenic and suggests a possible mechanism for this leukopenia through the JAK-signal transducer and activator of transcription (STAT) pathway inhibition. In the Automotive Industry (AI), and in all other industries linked to it, the increase in competitiveness and the compliance with environmental requirements are mandatory. Thus, the efforts to increase the lifespan of fabrication equipment and tooling are greater than ever. This work was developed around this real need, seeking to increase the lifespan of electrical terminal crimping punches for the AI. The process to achieve the objective of the practical work began with the collection of samples from the main competitors in the market, to be able to analyze and characterize them. Subsequently, brainstorming sessions were carried out, enabling to choose which alternative samples would be produced, analyzed, and characterized. New crimping punches were then characterized at different stages of production, to study the importance and impact of the finishing stages of the production process. Finally, new samples were produced, put to the test at a real customer and advanced characterization tests were done, which allowed a better understanding of the evolution of tool wear during its lifetime. This work contributed to a better clarification of the lifespan of crimping punches, making it possible to identify potential wear mechanisms and, consequently, propose a solution that maximizes the lifespan of crimping punches. When a soil is subjected to cyclic loading, there are changes in the material's geomechanical behaviour that need to be characterised before safely designing any future projects. In terms of cyclic loading, it is important to characterise not only the failure of the soil but also its behaviour before failure, in particular the yield point and the elastic behaviour of the material. This study examines the effects of the number of loading cycles on the behaviour of a chemically stabilised soft soil with a particular focus on the yield surface. To this end, a series of triaxial tests were performed on specimens, previously or not subjected to a different number of loading cycles (1000–100 000). The results were analysed in terms of the evolution of accumulated permanent axial strain, the yield surface and stress–strain behaviour. It was observed that an increase in the number of loading cycles promoted: an increase in the permanent axial strain, an increase in the undrained resilient modulus, a shrinkage of the yield surface but its shape is maintained, and there is a small increase in the peak strength of the stabilised soil explained by the strain-hardening effect induced by the cyclic loading. Background: Atrioventricular block (AVB) is rare in hyperthyroidism (HTH). Little is known about the true prevalence, clinical course, optimal management, and outcomes of different types of AVBs in patients with HTH. To address these uncertainties, we aimed to conduct a systematic review by combining the available literature to provide more meaningful data regarding AVBs in HTH. Methods: We systematically searched PubMed, Scopus, Embase, and Google Scholar for articles reporting patients who developed AVB in the context of HTH. Data were analysed in STATA 16. The main outcomes included types of AVB, frequency of pacemaker insertion, and resolution of AVB. The systematic review is registered with the International Prospective Register of Systematic Reviews (PROSPERO) with the identification number CRD42022335598. Results: A total of 56 studies (39 case reports, 12 case series, 3 conference abstracts, 1 retrospective study, and 1 prospective observational study) with 87 patients were included in the analysis, with a mean age of 39.1 ± 17.6 years. Females constituted 65.7% (n = 48) of the cohort. Complete heart block (CHB) was the most commonly reported AVB (N = 45, 51.7%), followed by first-degree AVB (16.1%) and second-degree AVB (14.9%). Overall, 21 patients underwent pacing. A permanent pacemaker was inserted in one patient with second-degree AVB and six patients with CHB. Mortality was reported in one patient with CHB. The clinical course and management of HTH and AVBs did not differ in patients with CHB or lower-degree blocks. Apart from lower rates of goitre and more use of carbimazole in those who underwent pacing, no differences were found when compared to the patients managed without pacing. Conclusion: Current data suggest that CHB is the most common type of AVB in patients with HTH. Most patients can be managed with anti-thyroid management alone. Cold brew is a method of coffee extraction that uses low temperature, preserving the volatile compounds of coffee. Freeze-drying allows the preservation of coffee features and nutritional value. The aim of this study was to evaluate the effects of different cryoprotectants in cold brew extracts as a basis for freeze-dried coffee production. Thus, the Coffea arabica extracts and the soluble coffee were characterized concerning caffeine content, antioxidant capacity, total phenolic compounds, and antimicrobial activity to verify the potential of this method. The extracts did not show antimicrobial activity with a high soluble solid content. It was observed that the cold extraction methods were efficient regarding the caffeine content, antioxidant capacity, and total phenolic compounds. Freeze-dried coffees also did not show antimicrobial activity, and they maintained the water and humidity activity standards. In general, cryoprotectants displayed an unfavorable influence on the extract and freeze-dried coffee in the analyses performed. The coffee extract without cryoprotectants had a higher antioxidant capacity (88.12%) and content of phenolic compounds (7.74 mg AG/mL of the coffee extract). Only for the analyses of soluble solids, the cryoprotectants mannitol and fructose showed promising results (14.03 °Brix, 14.40 °Brix, 11.33 °Brix, respectively). Objective: Upper gastrointestinal bleeding (UGIB) has been identified as a potential adverse drug reaction associated with the use of low-dose aspirin (LDA). This study aimed to investigate the relationship between variants in the TBXA2R gene, which is involved in platelet aggregation, and the risk of UGIB in patients with cardiovascular diseases treated with LDA. Methods: A case-control study was conducted at a Brazilian hospital complex. Three groups were defined: (1) case group (n = 50): patients with cardiovascular disease who used LDA and were diagnosed with UGIB of non-variceal etiology, (2) LDA control group (n = 50): patients with cardiovascular disease who used LDA without developing UGIB, and (3) healthy control group (n = 189). Data were collected through face-to-face interviews, and blood samples were collected for the analysis of Helicobacter pylori infection and genotyping of 3 genetic variants [rs2238631 (C > T), rs4807491 (A > G), and rs1131882 (A > G)]. Results: The case group had a significantly higher frequency of carriers of the rs4807491.G allele compared to the control group of LDA users (P -value =.004). No significant difference was observed in the proportion of carriers of the rs2238631.T and 1131882.G variants between the studied groups. Carriers of rs2238631.T (OR: 4.515, 95% CI: 1.37-14.89) and rs4807491.G allele (OR: 3.232, 95% CI: 1.12-9.37) exhibited a higher risk of UGIB. Conclusion: These findings suggest that the presence of the rs2238631 and rs4807491 variant alleles is associates with a 3- to 4-fold increased risk of UGIB in patients with cardiovascular diseases treated with LDA. Future studies with larger sample sizes should confirm these results and to better identify individuals who may benefit from chronic LDA use. [ABSTRACT FROM AUTHOR]

Spinocerebellar ataxias (SCA) are most frequently due to (CAG)n (coding for polyglutamine, polyQ) expansions and, less so, to expansion of other oligonucleotide repeats (non-polyQ) or other type of variants (non-repeat expansion SCA). In this study we compared polyQ and non-repeat expansion SCA, in a cohort of patients with hereditary ataxia followed at a tertiary hospital. From a prospective study, 88 patients (51 families) with SCA were selected, 74 (40 families) of whom genetically diagnosed. Thirty-eight patients (51.4%, 19 families) were confirmed as having a polyQ (no other repeat-expansions were identified) and 36 (48.6%, 21 families) a non-repeat expansion SCA. Median age-at-onset was 39.5 [30.0-45.5] for polyQ and 7.0 years [1.00-21.50] for non-repeat expansion SCA. PolyQ SCA were associated with cerebellar onset, and non-repeat expansion forms with non-cerebellar onset. Time to diagnosis was longer for non-repeat expansion SCA. The most common polyQ SCA were Machado-Joseph disease (MJD/SCA3) (73.7%) and SCA2 (15.8%); whereas in non-repeat expansion SCA ATX-CACNA1A (14.3%), ATP1A3-related ataxia, ATX-ITPR1, ATX/HSP-KCNA2, and ATX-PRKCG (9.5% each) predominated. Disease duration (up to inclusion) was significantly higher in non-repeat expansion SCA, but the difference in SARA score was not statistically significant. Cerebellar peduncles and pons atrophy were more common in polyQ ataxias, as was axonal neuropathy. SCA had a wide range of genetic etiology, age-at-onset and presentation. Proportion of polyQ and non-repeat expansion SCA was similar; the latter had a higher genetic heterogeneity. It is well known that cardiometabolic dysfunction gradually increases after menopause, and the sedentary lifestyle can aggravate this condition. Therefore, we compared the effects of aerobic exercise training during the premenopausal period and after ovariectomy (OVX) on metabolic, hemodynamic, and autonomic parameters in an experimental rat model of menopause. The female rats were divided into four groups: control (C), sedentary OVX (SO), trained OVX (TO), and previously trained OVX (PTO). The PTO group was trained for 4 weeks prior to+8 weeks after OVX, and the TO group trained only after OVX on a motor treadmill. Autonomic modulation was evaluated, white adipose tissue (WAT) was removed and weighed, and lipolysis was assessed. The citrate synthase activity in the soleus muscle was analyzed. The trained groups prevented the impairment of baroreceptor reflex sensitivity in relation to SO; however, only PTO reduced the low-frequency band of the pulse interval compared to SO. PTO reduced the weight of WAT compared to the other groups; lipolysis in PTO was similar to that in C. PTO preserved muscle metabolic injury in all types of fibers analyzed. In conclusion, this study suggests that exercise training should be recommended in a premenopausal model to prevent cardiometabolic and autonomic menopause-induced deleterious effects. Background: Locally advanced rectal cancer (LARC) poses a significantly challenge in clinical management, requiring a multimodal treatment approach. Among innovative strategies, Total Neoadjuvant Therapy (TNT) has emerged, delivering all planned chemotherapy before surgery. Objective: Our aim was to evaluate the real-world application and efficacy of TNT and to compare it with the non-TNT standard strategy. Methods: This retrospective study compared locally advanced rectal adenocarcinoma patients treated with Total Neoadjuvant Therapy (TNT) in 2022 with those who underwent traditional chemoradiotherapy (CRT) in 2020–2021. The primary endpoints were the pathologic complete response rate and the sustained clinical complete response rate in patients under W&W. Results: Among 107 patients (54.2% male, mean age 62.48 years), non-TNT (67 patients) and TNT (40 patients) mean follow-ups were 26.7 and 8.2 months, respectively. No differences in gender(p = 0.163), staging (p = 0.707), or location (p = 0.727) were noted. TNT patients received more short-course radiotherapy (42.5% vs1.5%, p < 0.001). Clinical responses favored TNT (p = 0.030) with no significant differences in pathological responses, recurrence rates, or survival. TNT exhibited higher chemotherapy completion (p = 0.007) and lower adverse events (p < 0.001). Post-surgery events showed no significant differences (p = 0.470). Single center with retrospective design and carries limitations that may restrict the generalizability of the findings and the relatively short follow-up duration are our main limitations. Conclusion: Our data add to the body of literature favoring the TNT treatment strategy for locally advanced rectal cancer, aiming to achieve comparable complete response rates with less adverse events. The rising prevalence of food intolerances and increased health and environmental consciousness has driven the demand for free-from (FF) and organic products. This study aims to analyze consumer needs, motivations, and challenges related to these products in Portugal through an online survey with 2268 eligible responses, with a median age of the participants of 41 years. The sample was predominantly female (76.9%), with male participants comprising 23.1% of the sample. The results show that 97.4% of the respondents were aware of FF products, with 60.0% being regular consumers, particularly of lactose-free milk and gluten-free bread and biscuits. Significant market gaps were identified in the variety of FF bread (62.8%), pastries (49.0%), and cookies (38.4%). The consumers identified high prices (84.9%) and excessive sugar/fat content (52.1%) as the main drawbacks of FF products currently on the market. The women showed greater knowledge and purchase levels compared to the men. The trust in organic certification was low (21.4%), with skepticism higher among the lower-income and male participants. Despite this, 78.1% believed in the health benefits of organic products, and 72.2% agreed that increased demand could lower prices. Alpha cells in the pancreas, traditionally known for their role in secreting glucagon to regulate blood glucose levels, are gaining recognition for their involvement in the pathophysiology of type 1 diabetes (T1D). In T1D, autoimmune destruction of beta cells results in insulin deficiency, which in turn may dysregulate alpha cell function, leading to elevated glucagon levels and impaired glucose homeostasis. This dysfunction is characterized by inappropriate glucagon secretion, augmenting the risk of life-threatening hypoglycemia. Moreover, insulin deficiency and autoimmunity alter alpha cell physiological responses, further exacerbating T1D pathophysiology. Recent studies suggest that alpha cells undergo transdifferentiation and interact with beta cells through mechanisms involving gamma-aminobutyric acid (GABA) signaling. Despite these advances, the exact pathways and interactions remain poorly understood and are often debated. Understanding the precise role of alpha cells in T1D is crucial, as it opens up avenues for developing new therapeutic strategies for T1D. Potential strategies include targeting alpha cells to normalize glucagon secretion, utilizing glucagon receptor antagonists, enhancing GABA signaling, and employing glucagon-like peptide-1 (GLP-1) receptor agonists. These approaches aim to improve glycemic control and reduce the risk of hypoglycemic events in individuals with T1D. Caring for technology-dependent children significantly impacts the family unit, a topic that has not been researched in Portugal. Understanding this impact can help nurses to improve their interventions. This study aimed to describe the lived experiences of parents with technology-dependent children using a qualitative descriptive design. We applied Giorgi's approach to analyze 10 individual semi-structured interviews with parents recruited through a children's hospital in Portugal. Our analysis identified two major themes: (1) Discovering a new parenthood: parent and caregiver, comprising two subthemes: Family reorganization and Learning to use a medical device; (2) Reconciling daily life with the needs of the technology-dependent children, comprising two subthemes: Importance of support systems and Experiencing difficulties. These results indicate that parents experience a wide range of concerns and challenges in managing medical devices, starting from the moment the need for a device is communicated and continuing through the process of learning and providing care. This journey involves significant changes in family dynamics and is marked by sacrifice and adaptation, supported by family, formal support systems, and healthcare professionals. Background/Objectives: The goal of this pilot study is to evaluate adherence to the Mediterranean Food Pattern (MFP) in a self-selected sample of university students, addressing a perspective of food sustainability. In addition, it seeks to relate adherence to MFP with sociodemographic characteristics and nutritional status. Methods: This is a cross-sectional pilot study whose data collection was carried out by an online questionnaire between January and April 2023. Results: Two hundred and forty-eight students participated—most of them were female (78.2%), had a median of 22 (20; 30) years, resided in the central region of Portugal (42.3%), and were pursuing a degree (73.4%) in a public higher education institution (66.5%). The prevalence of overweight (overweight and obesity) found was 33.1%. Females predominantly used olive oil as their main source of fat (95.9%, p = 0.009) and had a higher consumption of sugary drinks (81.4%, p = 0.004) compared to males, who reported usage rates of 85.2% and 63.0%, The median score of the Mediterranean Diet Adherence Screener was 7 points, presented with an interquartile range (Q1: 6, Q3: 8), indicating moderate adherence. The analysis showed no differences between the sexes (p = 0.087). There was also a negative correlation between adherence to the MFP and the body mass index (p = 0.007; r = −0.171). In addition, adherence to the MFP was associated with the area of study and the course attended, with students in health-related fields showing higher adherence. Conclusions: These findings underscore the necessity for targeted interventions aimed at promoting adherence to the MFP among university students, which could contribute to improved health outcomes and enhanced environmental sustainability. This article presents the results of laboratory tests conducted to identify the granulometric stabilization and chemical improvement techniques used in an experimental segment of the unpaved BR-030 highway in the Maraú Peninsula, Bahia. The segment was designed to evaluate the performance of primary coating sections stabilized with sand, clayey gravel, reclaimed asphalt pavement (RAP), and simple graded crushed stone (GCS), as well as chemically improved with Portland cement and hydrated lime. The laboratory campaign focused on mechanical resistance, resilient modulus, and permanent deformation tests. In this research, chemical improvement with the addition of 2% Portland cement presented the most promising results for potential application in the section of the BR-030 highway intended to remain unpaved. Additionally, a life cycle assessment (LCA) revealed that mechanical stabilization of the primary coating has the lowest environmental impacts, making it a suitable and sustainable option among stabilization methods. Lead (Pb) is one the 10 chemicals of major public health concern listed by the World Health Organization. One reason for Pb occurrence in food is agricultural practices. In this context, organic production systems are highlighted for not using pesticides, which may result in less chemical contamination in food. Considering that, this systematic review (SR) and meta-analysis was conducted aiming to investigate if there are differences in Pb levels in organic (OLV) and conventional leafy vegetables (CLV). This SR and its protocol (PROSPERO CRD42022306048) were based on PRISMA. Only analytical, cross-sectional studies which reported data on the analysis of Pb in OLV and CLV were included. Searches were conducted in 12 electronic databases besides manual searches in articles, directory, and archives of thesis and dissertations. No restrictions regarding language or publication period were applied. After the selection stages, 24 studies were included, of which 22 (91.67%) presented low risk of bias, while two (8.33%) presented medium risk. Significant difference (p = 0.008) was observed in the meta-analysis considering results in fresh weight (mean difference = 0.01 mg/kg), indicating an inferior concentration of Pb in OLV, being classified as evidence of 'moderate certainty', which answers our research question. Kallmann syndrome is a rare disorder characterized by hypogonadotropic hypogonadism and an impaired sense of smell (anosmia or hyposmia) caused by congenital defects in the development of the gonadotropin-releasing hormone (GnRH) and olfactory neurons. Mutations in several genes have been associated with Kallmann syndrome. However, genetic testing of this disorder often reveals variants of uncertain significance (VUS) that remain uninterpreted without experimental validation. The aim of this study was to analyze the functional consequences of a heterozygous missense VUS in the CHD7 gene (c.4354G>T, p.Val1452Leu), in a patient with Kallmann syndrome with reversal of hypogonadism. The variant, located in the first nucleotide of exon 19, was analyzed using minigene assays to determine its effect on ribonucleic acid (RNA) splicing. These showed that the variant generates two different transcripts: a full-length transcript with the missense change (p.Val1452Leu), and an abnormally spliced transcript lacking exon 19. The latter results in an in-frame deletion (p.Val1452_Lys1511del) that disrupts the helicase C-terminal domain of the CHD7 protein. The variant was reclassified as likely pathogenic. These findings demonstrate that missense variants can exert more extensive effects beyond simple amino acid substitutions and underscore the critical role of functional analyses in VUS reclassification and genetic diagnosis. While there is a considerable body of data regarding the sources of minerals employed in Bronze Age Egypt, the supply chains to Sudanese Lower Nubia are virtually unknown. This paper presents results of lead isotope analysis of 11 samples of kohl from C-group, Pan-grave and New Kingdom funerary contexts across the Debeira and Ashkeit areas in Sudanese Lower Nubia during the Bronze Age. The samples could be divided in two groups based on their lead isotope ratios. A comparison between the kohl samples from Sudanese Lower Nubia and galena ores from various mining sites on the Red Sea coast of Egypt indicated that some of the galena used in kohl mixtures in Sudanese Lower Nubia came from the Pharaonic mining site of Gebel el-Zeit. The second group of Nubian samples seems to have originated from an alternative galena source yet to be determined. This is the first time that kohl samples from Sudanese Lower Nubia are integrated into known northeast African networks of supply by using Lead isotope analysis. [ABSTRACT FROM AUTHOR]

Background: Limited information is available about functioning and related impairments of children and adolescents with Cerebral Palsy (CP) in low- and middle-income countries (LMIC) like Brazil. The aim of this study is to describe the characteristics, functioning, and impairments of Brazilian children and adolescents with CP. Methods: Cross-sectional preliminary study as part of the PartiCipa Brazil multicentered cohort study. Families of children and adolescents with CP from Brazil, 4 months to 15 years, were enrolled. They responded to an online survey with questions about their child's health condition, impairments, contextual factors, and functioning according to the Gross Motor Function Classification System (GMFCS) and the Manual Ability Classification System (MACS). Data were described as frequencies, percentages, means, and standard deviations, according to age bands. Results: Of the 404 participants (6.5±3.6 years) enrolled in this preliminary analysis, 54.7% are male, 90.4% under 12 years of age, 77.7% have bilateral CP, 49% in GMFCS levels IV and V, and 50.7% in MACS levels II and V. Most participants are from Southeast (63.4%) and Centre-west (19.5%) of Brazil. Regarding the impairments and functioning limitations: 1 in 2 did not talk; 1 in 2 has epilepsy; 2 of 5 reports pain, 1 of 4 has visual impairments, 3 out of 5 did not feed themselves, 1 out of 20 has a hearing impairment and 1 of 4 did not go to school. Conclusion: This first preliminar Brazilian study shows a high prevalence of children at MACS levels II and V and GMFCS levels IV and V, representing almost half of the group, indicating more impairments and limitations than children/adolescents from high-income countries. This study provides a preliminary deeper understanding of the key impairments and limitations in activities among children and adolescents with CP from various Brazilian regions. Introduction: Tissue injury results in the release of inflammatory mediators, including a cascade of algogenic substances, which contribute to the development of hyperalgesia. During this process, endogenous analgesic substances are peripherally released to counterbalance hyperalgesia. The present study aimed to investigate whether inflammatory mediators TNF-α, IL-1β, CXCL1, norepinephrine (NE), and prostaglandin E2 (PGE2) may be involved in the deflagration of peripheral endogenous modulation of inflammatory pain by activation of the cholinergic system. Methods: Male Swiss mice were subjected to paw withdrawal test. All the substances were injected via the intraplantar route. Results: The main findings of this study were as follows: (1) carrageenan (Cg), TNF-α, CXCL-1, IL1-β, NE, and PGE2 induced hyperalgesia; (2) the acetylcholinesterase enzyme inhibitor, neostigmine, reversed the hyperalgesia observed after Cg, TNF-α, CXCL-1, and IL1-β injection; (3) the non-selective muscarinic receptor antagonist, atropine, and the selective muscarinic type 1 receptor (m1AChr) antagonist, telenzepine, potentiated the hyperalgesia induced by Cg and CXCL-1; (4) mecamylamine, a non-selective nicotinic receptor antagonist, potentiated the hyperalgesia induced by Cg, TNF-α, CXCL-1, and IL1-β; (5) Cg, CXCL-1, and PGE2 increased the expression of the m1AChr and nicotinic receptor subunit α4protein. Recent measurements of the Lamb shift of muonic helium-4 ions were used to infer the alpha particle charge radius. The value found is compatible with the radius extracted from the analysis of the electron-helium scattering. Thus, the new spectroscopic data put additional empirical bounds on some free parameters of certain physics theories beyond the Standard Model. In this paper, we analyze the new data in the context of large extra-dimensional theories. Specifically, we calculate the influence of the radion, the scalar degree of freedom of the higher-dimensional gravity, on the energy difference between the 2S and 2P levels of this exotic atom. The radion field is related to fluctuations of the volume of the supplementary space. It should be treated as a phenomenologically independent quantity in relation to the tensorial degrees of freedom of the metric within the braneworld scenario. Based on the spectroscopic data of muonic helium, we find constraints for the effective energy scale of the radion as a function of the alpha particle radius. Then, we discuss the implications of these new constraints on the proton radius puzzle. We also establish a new empirical bound for the radion by examining its influence on the isotopic shift in the 2P-2S transition of muonic hydrogen and muonic deuteron. In connection with this discussion, we study the impact of the radion on the tension observed in measurements of the difference between the squared radii of the helion and alpha particle as extracted from muonic and electronic helium isotopes. Microbiologically contaminated water is a significant source of infections in humans and animals, with Pseudomonas aeruginosa (PSA) being particularly concerning due to its ability to thrive in water environments and its resistance to many disinfectants. Therefore, this study investigates the adhesion potential of PSA strains on various materials used in mineral water extraction wells, focusing on hydrophobic and hydrophilic properties. Mineral water samples were collected from three wells (P-01, P-07, and P-08) within the Guarani Aquifer System and Fractured Aquifer System (SAF) in Brazil. The physicochemical properties of the water, including concentrations of Sr (strontium), Fe (iron), Si (silicon), SO42− (sulfate ions), Cl− (chloride ions), and ORP (oxidation-reduction potential), were analyzed. Results indicated higher PSA adhesion on hydrophobic materials, particularly high-density polyethylene (HDPE) and geomechanically plasticized polyvinyl chloride (PVC). Multiple correlation analyses revealed positive correlations between PSA adhesion on hydrophilic materials and Sr, Fe, Si, SO42−, and Cl− concentrations. Conversely, ORP negatively correlated with bacterial adhesion on PVC surfaces, suggesting higher ORP values reduced PSA attachment. These findings highlight the importance of water composition and material properties in influencing bacterial adhesion and potential biofilm formation in mineral water extraction systems. Purpose: The mine sequencing problem is NP-hard. Therefore, simplifying it is necessary. One way to do this is to employ clusters as input instead of individual blocks. The mining cut clustering problem has been little addressed in the literature, and the solutions used are almost always heuristic. We solve the mining cut clustering problem, which is NP-hard, through single- and multi-objective optimization, finding results that are local optima in acceptable computational time. Design/methodology/approach: We first elaborate an ILP-based model to address the mining cut clustering problem. We employ a mono-objective approach and two multi-objective approaches, solving all these models by constraint programming. To choose the best solutions generated by multi-objective approaches, we employ two multi-criteria decision analysis approaches, considering different weight configurations. We developed a case study using real data. Findings: We verified that the approaches based on multi-objective optimization performed better than the mono-objective approach for the economic return criterion. The weighted-sum multi-objective approach presented the best results considering all objective functions used. Once viable solutions were obtained through multi-objective optimization, multi-criteria decision analysis approaches almost always selected the same solution. We obtained solutions that are local optima in acceptable computational time. Research limitations/implications: This study solves an instance with 80 blocks. Consequently, it is aimed at short-term mine planning. The methodology has not yet been evaluated in large instances related to medium- and long-term mine planning. Originality/value: This is the first time that multi-objective optimization has been employed to solve the mining cut custering problem. Even other problems related to mine planning were, at most, solved by goal programming, so that multi-objective optimization is a knowledge that is not widespread among mining researchers. Objective: To evaluate the combined administration of propylthiouracil (PTU) and levothyroxine (LT4) in managing monocarboxylate transporter 8 (MCT8) deficiency and identify optimal therapeutic dosages. Methods: This multicenter case series involved 12 male patients with MCT8 deficiency whose parents/guardians consented to PTU and LT4 treatment. Data were collected from January 2008 to June 24, 2024. The study focused on treatment safety and outcomes, analyzing baseline and last encounter biochemical, metabolic, and anthropometric parameters. Statistical analyses included Wilcoxon signed ranks tests and generalized estimated equations to assess effects on thyroid and metabolic markers, and receiver operating characteristics curves to predict optimal dose. Results: Patients showed a significant reduction in serum total triiodothyronine (TT3) concentration and TT3/TT4 ratio, with increased serum TT4 and free T4 (fT4) concentrations. The use of PTU effectively reduced TT3 concentration by 25% at an average dose of 6.8 mg/kg/day, while LT4 increased fT4 concentration by 40% from baseline at an average dose of 4.3 µg/kg/day. Thyrotropin concentration was undetectable on treatment. No statistical differences were observed in metabolic and physical parameters between baseline and last encounter overall for the group, but six of eight patients for whom these data were available had an increase in weight (z-score). There were no adverse effects on liver function or granulocyte numbers noted throughout the period of observation. Conclusion: Combined treatment with PTU and LT4 normalized serum T3, fT4, and TT4 in patients with MCT8 deficiency. This work presents the successful production of highly porous 3D nanofibrous hybrid scaffolds of polylactic acid (PLA)/polyethylene glycol (PEG) blends with the incorporation of calcium phosphate (CaP) bioceramics by a facile two-step process using the solution blow spinning (SBS) technique. CaP nanofibers were obtained at two calcium/phosphorus (Ca/P) ratios, 1.67 and 1.1, by SBS and calcination at 1000 °C. They were incorporated in PLA/PEG blends by SBS at 10 and 20 wt% to form 3D hybrid cotton-wool-like scaffolds. Morphological analysis showed that the fibrous scaffolds obtained had a randomly interconnected and highly porous structure. Also, the mean fiber diameter ranged from 408 ± 141 nm to 893 ± 496 nm. Apatite deposited considerably within 14 days in a simulated body fluid (SBF) test for hybrid scaffolds containing a mix of hydroxyapatite (HAp) and tri-calcium phosphate-β (β-TCP) phases. The scaffolds with 20 wt% CaP and a Ca/P ration of 1.1 showed better in vitro bioactivity to induce calcium mineralization for bone regeneration. Cellular tests evidenced that the developed scaffolds can support the osteogenic differentiation and proliferation of pre-osteoblastic MC3T3-E1 cells into mature osteoblasts. This study investigated the effects of a single dose of desvenlafaxine via oral administration on the pharmacokinetic parameters and clinical and laboratory characteristics in healthy volunteers using a pharmacometabolomics approach. In order to optimize desvenlafaxine's therapeutic use and minimize potential adverse effects, this knowledge is essential. Methods: Thirty-five healthy volunteers were enrolled after a health trial and received a single dose of desvenlafaxine (Pristiq®, 100 mg). First, liquid chromatography coupled to tandem mass spectrometry was used to determine the main pharmacokinetic parameters. Next, ultra-performance liquid chromatography–quadrupole time-of-flight mass spectrometry was used to identify plasma metabolites with different relative abundances in the metabolome at pre-dose and when the desvenlafaxine peak plasma concentration was reached (pre-dose vs. post-dose). Results: Correlations were observed between metabolomic profiles, such as tyrosine, sphingosine 1-phosphate, and pharmacokinetic parameters, as well as acetoacetic acid and uridine diphosphate glucose associated with clinical characteristics. Our findings suggest that desvenlafaxine may have a broader effect than previously thought by acting on the proteins responsible for the transport of various molecules at the cellular level, such as the solute carrier SLC and adenosine triphosphate synthase binding cassette ABC transporters. Both of these molecules have been associated with PK parameters and adverse events in our study. Conclusions: This altered transporter activity may be related to the reported side effects of desvenlafaxine, such as changes in blood pressure and liver function.
